Abstract

The experiment was to investigate the feasibility of producing trifolium flavone from Cymbidium trifolium cell suspension culture. The callus was induced by leaves and stems of wild Cymbidium trifolium as explants, and the proliferation, single cell culture and total flavonoid content were measured. The suspension cultured cells were cultured with different liquid medium formulations, light mode, shaking table speed, inoculum amount, pH value and culture days. The total flavonoids were extracted by reflux with ethanol as solvent, and NaNO2-Al(NO3)3 was used for color development. Total flavonoid content was determined by UV spectrophotometer at 500 nm. The results show that the optimal medium for Cymbidium trifolium was MS + 1.5 mg/L 6-BA + 2.5 mg/L 2-4D + 3 mg/L NAA + 0.4 mg/L 6-KT, 8 h/d illumination, shaking speed 110 r/min, inoculation dosage 25 mL/L, pH value 5.8, incubation time was 13 d. The results show that flavonoids could be produced by Cymbidium trifolium cell culture, and the accumulation of flavonoids in the suspension culture system of Cymbidium trifolium cell could reach 20.524 mg/g under optimal culture conditions.
