麝香保心丸对缺氧复氧心肌细胞的保护作用及 凋亡的影响.

Abstract

Objective To study the protective effect of Shexiang Baoxin Pill (SBP) on hypoxia-reoxygenation cardiomyocytes and its effect on apoptosis. Methods The model of hypoxia and reoxygenation of cardiomyocytes was established. The active components of SBP were extracted by water extraction. According to whether different concentrations of SBP active components were given, cells were divided into the control group, the model group, the 0.5 g/L SBP group, the 1 g/L SBP group, the 2 g/L SBP group and the 5 g/L SBP group. The highest survival rate in the SBP intervention group was selected as the group for the follow-up experiments. The cell survival rate of each group was detected by CCK-8. The apoptosis rate of each group was detected by Anexxin-VFITC/PI double staining. Western bolt assay was used to detect the relative expression levels of B cell lymphoma factor 2 (Bcl-2), Bcl-2 associated X protein (Bax) and cysteine protease 3 (Caspase-3) in each group. Results Compared with the model group, survival rates of cardiomyocytes were increased in the 0.5 g/L, 1 g/L, 2 g/L and 5 g/L SBP groups (P<0.01), and the survival rate of cardiomyocytes was the highest in the 2 g/L SBP group. Compared with the control group, the apoptosis rate, relative expression levels of Bax and Caspase-3 were increased in the model group and the 2 g/L SBP group (P<0.05), but there was no significant difference in Bcl-2 between the groups. Compared with the model group, the apoptosis rate, relative expression levels of Bax and Caspase-3 were decreased in the 2 g/L SBP group (P<0.05), while relative expression levels of Bcl-2 and Bcl-2/Bax ratio were increased (P<0.05). Conclusion Shexiang Baoxin Pill has a protective effect on hypoxia-reoxygenation cardiomyocytes, and the inhibition of apoptosis is one of its protective mechanisms. [ABSTRACT FROM AUTHOR]
