Abstract

Objective To investigate the effect of cancer suppressor gene NM23-H2 on migration ability of colorectal adenocarcinoma and its correlation with Janus kinase-Signal transducer and activator of transcription (JAK-STAT) signaling pathway. Methods A total of 80 pathological wax blocks were collected from the first people’s hospital of Xiangtan city in from January 2018 to January 2021,including 35 cases of adenocarcinoma tissue,20 cases of para-carcinoma tissue, and 25 cases of lymph node metastasis. Immunohistochemical methods was used to staining, and the expressions of P-STAT3 and NM23-H2 in the three wax blocks were statistically analyze. The recombinant plasmid with NM23-H2 gene was transfected into human colorectal adenocarcinoma SW620 cells. Transwell assay was used to detect the effect of NM23-H2 on the migration ability of SW620 cells. Western blotting was used to detect the expression of P-STAT3 in SW620 cells of stable transfection group and non-transfection group. Results There were significant differences in the expressions of P-STAT3 and NM23-H2 among the three wax blocks (P<0.05);NM23-H2 could inhibit the migration ability of SW620 cells and down-regulate the expression of P-STAT3 in SW620 cells. Conclusion The inhibition of NM23-H2 on the occurrence, development and migration of colorectal adenocarcinoma may be related to the down-regulation of JAK-STAT signaling pathway activity.
