基于“异病同治”理论的宁泌泰胶囊治疗慢性前列腺炎和良性前列腺增生症的网络药理学研究.

Abstract

Objective To investigate the mechanism of Ningmitai Capsule in the treatment of chronic prostatitis(CP)and benign prostatic hyperplasia(BPH) using network pharmacology. Methods The ingredients and action targets of Ningmitai Capsule were retrieved through TCMSP and BATMAN-TCM database, the action targets of CP and BPH were retrieved through gene cards, OMIM and Disgenet database, the common targets of CP and BPH were retrieved through Venn diagram tool and the effective targets of Ningmitai Capsule in the treatment of CP and BPH were retrieved, and the consensus gene to construct a protein interaction(PPI) network by string database, The core targets were screened using Cytoscape software to construct a "drug-component-target" network. Go and KEGG enrichment analyses were performed using the metascape database. Molecular docking validation of core targets and constituents was performed using autodock4.2.6. Results 47 active ingredients of Ningmitai Capsule with 280 targets and 1445 disease targets were obtained after screening,while 168 potential therapeutic targets were obtained after intersection, the core ingredients including quercetin, kaempferolβ-Sitosterol, among others, and core targets include AR, ESR1, TNF, IL-1B, among others. Go analysis showed that the therapeutic targets were associated with processes such as response to inorganic substances, apoptotic signaling pathway,response to lipopolysaccharide, mainly involving PI3K/Akt, MAPK and other signaling pathways. Conclusion Ningmitai Capsule may achieve the effects of "Treat Different Diseases with the Same Method" for CP and BPH by inhibiting the synthesis of proinflammatory cytokines, inhibiting signal transduction, regulating cell growth in prostate tissue, regulating androgen levels, and other mechanisms.
