The diversity of physiological effects and pharmacological actions of glucocorticoids determines their use in the treatment of diseases and pathological conditions in rheumatology, nephrology, allergology, hematology, transplantology, hepatology, neurology, intensive care etc. Experimental and clinical research in recent decades has revealed new aspects of pharmacological and pleiotropic effects based on elucidation of the genomic and non-genomic mechanism; influence on different ways of transmitting and regulating the intracellular signal; variability of cellular and tissue responses depending on physiological or pathological condition; the diversity of pharmacological or adverse effects depending on the dose and dosing regimen, purpose of treatment; pharmacokinetic characteristics depending on the structure and physicochemical properties. The pharmacodynamic and pharmacokinetic aspects of glucocorticoids allow the practitioner to rationally select the glucocorticoid in order to perform an effective and harmless therapy.
