Abstract

[Objective] The present paper aimed to explore the biological characteristics of a wild Lentinula edodes and the fermentation of different additives on the antioxidant activity of lentinan polysaccharide in mycelium, and to lay the foundation for the preparation of high vitality strains and the development of functional products. [Method] Colony diameter and dry weight of mycelium were used as main indexes, the biological traits and growth of mycelium of tested strains under different carbon and nitrogen sources, culture temperature and initial pH were studied by solid and liquid culture. Meanwhile, the scavenging effects of Lentinan on DPPH,ABTS and hydroxyl radicals were further determined by adding fermented liquid Huangzhu grass (HLNT) and Chinese medicine residue (ZLNT). [Result] The best carbon and nitrogen sources in the solid medium were glucose and ammonium sulfate, and the optimum culture temperature was 25-30 °C, initial pH 4. 0 - 6. 0. When sucrose was used as carbon source, ammonium sulfate or peptone as nitrogen source, mycelium biomass was accumulated in liquid medium. The culture temperature 25 °C, inoculation amount 3% (v/v) and pH 6. 0 were suitable conditions. ZLNT had significant scavenging effect on DPPH and hydroxyl radicals, as the concentration was 5 mg/ mL, the scavenging rate was 86. 7% and 73. 5%, respectively. The scavenging rate of HLNT with 5 mg/mL concentration to ABTS radical was 75. 5%. [Conclusion] The nutritional requirements and cultivation conditions suitable for mycelia growth of the wild L. edodes were preliminarily screened, and the strain showed better suitability for byproducts, which antioxidant activity of the liquid fermentation could be promoted by adding the byproduct immersion solution.
