Abstract

[Objective] The purpose of this study was to investigate the polymorphism of the pig CKM gene and its mRNA expression level. By detecting the polymorphism in the 3 kb region upstream of the start codon of the CKM gene, single nucleotide polymorphism sites C-1185T and C-1324T, and this site complies with Hardy-Weinberger's law ( P < 0. 05 ) . [Method] Real-time quantitative PCR was used to detect the mRNA expression of CKM gene in different tissues of Tibetan pigs and Yorks hire pigs. [ Result] It was found that in the same breed of pig breeds, the mRNA expression level of CKM gene in order from the highest to the lowest was the longest back muscle > back fat> liver tissue, CKM genes in different pig breeds were differentially expressed in longissimus dorsi muscle, back fat and liver tissues. In longissimus dorsi muscle, Tibetan pig (TP) was significantly higher than Yorkshire pig (YY) ( P <0.01), in back fat, Yorkshire pigs (YY) were significantly higher than Tibetan pigs ( TP) ( P < 0. 05), and in liver tissue, Tibetan pigs ( TP) were extremely significantly higher than Yorkshire pigs ( YY) ( P <0. 01 ) . [Conclusion] It is speculated that there is a negative correlation between the CKM gene and the growth and development of skeletal muscle.
