草莓4种主要病毒检测及SVBV贵州分离物基因组测定及分析.

Abstract

[Objective] This paper aims to investigate incidence of four strawberry viruses in Guizhou Province and the evolutionary relationship between strawberry vein banding virus SVBV-GZ and other SVBV isolates. [Method]77 strawberry plants and the tissue culture seedlings were collected from greenhouses located in Guiyang City, Kaili City and Qiandongnan Vocational & Technical College for Nationalities.Strawberry plants were collected for detection of four strawberry viruses. The full genome sequences and CP nucleotide sequences of SVBV were cloned to the vector for sequencing and analysis. [Result] Both SVBV and strawberry mild yellow edge virus(SMYEV) were detected by RT-PCR, and the rate of virus carrying SVBV and SMYEV were 19.5 % and 2.6%, respectively. However strawberry mottle virus(SMoV) and strawbeny crinkle virus(SCV) viruses were not detected among these samples. The complete genome sequence of SVBV-GZ(GenBank accession No.:M H894295) is 7859 bp, and the genome structure might be composed of seven open reading frames.Phylogenetic analysis showed that the highest homology was observed between SVBV-GZ and SCBV-BJ(GenBank accession No.:KR080547) nucleotide sequences. [Conclusion] The nuclear sequence analysis of SVBV CP showed that the SVBV isolates in Guizhou are the most related to different isolates from different area in China. This may be caused by importing the strawberry seedlings from different area. [ABSTRACT FROM AUTHOR]
