脂肪组织中纳米脂肪与基质血管成分的对比研究.

Abstract

Objective To compare the cell content, components, activity of nano-fat and SVF. Methods Abdominal lipoaspirates were obtained from 12 healthy women with no systemic diseases, and were prepared into 12 nano-fat and SVF suspension. The cells of two group were examined for surface marker expression and counted for cell numbers, then cultured in vitro. Cultured cells were examined for surface marker expression and histology on the sixth and thirteenth days. Results The results of 12 nano-fat and SVF suspension: mean cell number was (1.85±0.73)×106/ml and (5.57±2.63)×106/ml respectively(P<0.05); Mean cell number of adipose-derived stromal cells(CD34+/CD31-/CD45-)was(1.92±0.57)×105/ml and(3.07±0.87)×105/ml respectively(P<0.05); Mean cell number of endothelial cells(CD34+/CD31+/CD45-)was (5.09±1.06)×105/ml and (0.19±0.06)×105/ml respectively(P<0.05);On the sixth days after culture, mean cell number of adipose-derived stromal cells(CD73+/CD90+/CD31-/CD45-) was(4.23±1.63)×105/ml and(5.11± 1.33)×105/ml respectively(P>0.05); On the thirteenth days, mean cell number was(7.48±1.57)×105/ml and(4.67±0.87)×105/ml respectively(P<0.05), and oil red o stain show that lipid aggregate in cells in two groups on the same day. Conclusion The study finds that nano-fat suspension have fewer cell content and adipose-derived stromal cells than SVF suspension, but have more endothelial cells. The adipose-derived stromal cells of two group both can proliferation and adipogenesis in vitro. This study can provide reference for clinical application of nano-fat in Cell-Assisted Lipotransfer.
