Abstract

Objective: To investigate the change and mechanism of calcium sensing receptor expression in atherosclerosis rat combined with myocardial infarction. Methods: A rat atherosclerosis model combined with myocardial infarction was established, in which rats received a single dose of vitamin D3 and were fed a high-fat and received isoproterenol (ISO) treatment. ISO was used to induce myocardial infarction. The rat myocardial infarction models were prepared using subcutaneous injection of isoprel (200 mg kg-1 d-1.Wistar rats were divided into four groups randomly: l) Control group: 1 ISO group: 2 AS group: 3 AS ISO group. The expressions of CaSR. Bcl-2. Bax and caspase-3 mRNA were analyzed using RT-PCR respectively. Apoptotic cells were measured by TUXEL staining assay. The morphological changes of cardiac tissues were observed under optical microscope. The activity of creatine kinase (CK). were assayed with ultraviolet spectrophotometer. The level of cTnT was detected by electrochemical immunofluorescence. Results: Compared with the control group, the activity of CK and cTnT. apoptosis index, expression of CaSR. Bax and caspase-3 were increased, but the Bcl-2 expression was decreased, the myocardial ultrastructure injury was aggravated in the ISO group. The further increased of above parameters in AS ISO group. Conclusions: The increased expression of CaSR was involved in the pathogeneisi of atherosclerosis combined with myocardial infarction, which might be related with and the increase of apoptosis.
