We report the development of an efficient and reproducible genetic transformation system for the recalcitrant Spanish elite rice paella genotype, Bomba. Preconditioned embryos derived from dry seeds were bombarded with gold particles carrying a plasmid containing a screenable and a selectable marker. We confirmed integration and expression of hpt and gusA in the rice genome. Transformation frequency was ca: 10% in several independent experiments. We show Mendelian inheritance of the input transgenes and zygosity determination of the transgenic lines in the T1 generation. A unique and critical step for the regeneration of plants from transformed tissue was shading during the early stages of regeneration, combined with a specific cytokinin:auxin ration at the onset of shifting callus to regeneration media.

Twin registers are wonderful research resources for applications in epidemiology, molecular genetics, and other areas of research. New registers continue to be launched all over the world as researchers from different disciplines recognize their potential. In this chapter, we discuss multiple aspects that need to be considered when initiating a register. This encompasses aspects related to the strategic planning and key elements of research designs, promotion, and management of a twin register, including recruitment and retaining of twins and family members, phenotyping, database organization, and collaborations between registers. Information on questions unique to twin registers and twin-biobanks, such as the assessment of zygosity by genotype arrays, the design of (biomarker) studies involving related participants and the analyses of clustered data, is presented. Altogether, we provide a number of basic guidelines and recommendations for reflection when planning a twin register.

Accurate zygosity determination is a fundamental step in twin research. Although DNA-based testing is the gold standard for determining zygosity, collecting biological samples is not feasible in all research settings or all families. Previous work has demonstrated the feasibility of zygosity estimation based on questionnaire (physical similarity) data in older twins, but the extent to which this is also a reliable approach in infancy is less well established. Here, we report the accuracy of different questionnaire-based zygosity determination approaches (traditional and machine learning) in 5.5 month-old twins. The participant cohort comprised 284 infant twin pairs (128 dizygotic and 156 monozygotic) who participated in the Babytwins Study Sweden (BATSS). Manual scoring based on an established technique validated in older twins accurately predicted 90.49% of the zygosities with a sensitivity of 91.65% and specificity of 89.06%. The machine learning approach improved the prediction accuracy to 93.10%, with a sensitivity of 91.30% and specificity of 94.29%. Additionally, we quantified the systematic impact of zygosity misclassification on estimates of genetic and environmental influences using simulation-based sensitivity analysis on a separate data set to show the implication of our machine learning accuracy gain. In conclusion, our study demonstrates the feasibility of determining zygosity in very young infant twins using a questionnaire with four items and builds a scalable machine learning model with better metrics, thus a viable alternative to DNA tests in large-scale infant twin studies.

This study evaluated the applicability of droplet digital PCR (ddPCR) as a tool for maize zygosity determination using quantitative real-time PCR (qPCR) as a reference technology. Quantitative real-time PCR is commonly used to determine transgene copy number or GMO zygosity characterization. However, its effectiveness is based on identical reaction efficiencies for the transgene and the endogenous reference gene. Additionally, a calibrator sample should be utilized for accuracy. Droplet digital PCR is a DNA molecule counting technique that directly counts the absolute number of target and reference DNA molecules in a sample, independent of assay efficiency or external calibrators. The zygosity of the transgene can be easily determined using the ratio of the quantity of the target gene to the reference single copy endogenous gene. In this study, both the qPCR and ddPCR methods were used to determine insect-resistant transgenic maize IE034 zygosity. Both methods performed well, but the ddPCR method was more convenient because of its absolute quantification property.

Background: The study of twins is a well-known and a unique method that is used to investigate the genetic contributions to physical features and diseases in human. Aim: To estimate the relative contributions of genetic and environmental influence to observed variations in different dental features or traits. Methodology: Thirty twins in the age group of 15–30 years were selected for the study, and the dental caries experience scores, dental anomalies, and morphometric analysis of monozygotic (MZ) and dizygotic (DZ) twins were compared. Results: MZ twins unlike DZ twins showed within-pair resemblance in decayed, decayed missing and filled teeth (DMFT), and molar relation. No morphometric analysis parameters and dental anomalies had significant mean difference between the groups. Decay, filling, DMFT, spacing, mandibular irregularity, open bite, and mesiodistal width of the right and left canine showed a higher positive correlation in MZ than DZ twins. Heritability estimate was low for morphometric analysis parameters. There is genetic dominance in decay, DMFT, spacing, molar relation, and diastema. Pearson's correlation revealed a positive and statistically significant correlation among all morphometric analysis parameters for MZ twins, but not DZ twins. Dental caries has a higher concordance rate (87.5%), correlation (71.6%), and heritability (87.8%). Conclusion: Most of the dental parameters are significantly heritable with up to 85% of the variance attributable to genetic factors. Environmental factors play a major role in developing malocclusion and causing dental caries. Morphology of canines can be used as a supplementary tool to determine zygosity.

The aim of the report was to present a method of zygosity determination in multiple pregnancy. The study wascarried out on same-sex neonates born as a result of spontaneous quadruplet pregnancy. Zygosity was determinedby DNA profiling. The pregnancy was confirmed to be polyzygotic. DNA profiling may be used as a method ofzygosity determination in multiple pregnancy.

All same-sex dizygotic (DZ) twins and approximately one-third of monozygotic (MZ) twin pairs have separate placentas, making it impossible to use the number of placentas to determine zygosity. Zygosity determination is further complicated because incorrect assumptions are often made, such as that only DZ pairs have two placentas and that all MZ pairs are phenotypically identical. These assumptions, by twins, their families and health professionals, along with the lack of universal zygosity testing for same-sex twins, has led to confusion within the twin community, yet little research has been conducted with twins about their understanding and assumptions about zygosity. We aimed to explore and quantify understanding and assumptions about zygosity using twins attending an Australian twin festival. We recruited 91 twin pairs younger than 18 years of age and their parents, and 30 adult twin pairs who were all uncertain of their zygosity, to complete one pen and paper questionnaire and one online questionnaire about their assumed zygosity, reasons for their assumptions and the importance of accurate zygosity knowledge. Responses were compared with their true zygosity measured using a genetic test. We found a substantial proportion of parents and twins had been misinformed by their own parents or medical professionals, and that knowledge of their true zygosity status provided peace of mind and positive emotional responses. For these reasons we propose universal zygosity testing of same-sex twins as early in life as possible and an increase in education of medical professionals, twins and families of twins about zygosity issues.

Twins reared apart provide a fascinating experiment to distinguish genetic from environmental influences. However, there is as yet no broad report on distribution of twins reared apart, especially in the Chinese population. In this study, information on 18,295 volunteer twin pairs of all age groups was compiled in nine provinces or cities of China, and questionnaires were used for zygosity determination. It was discovered that twins reared apart from 0 to 10 years of age accounted for 2.2% of all twin interviewees, with the proportion of this 0–10 group separated before 1, 2, and 5 years old, accounting for 65.3%, 76.1%, and 91.3%, respectively. The proportion of twins reared apart is not significantly related to zygosity or gender, but it is related to region and twin age. As the age of twins lowers, the proportion of those reared apart gradually decreases. Twins reared apart will become rarer in the future and therefore should be cherished as a resource.

Zygosity determination using similarity ratings is frequently applied in twin studies. A correct determination of zygosity is essential for the estimation of heritability and environmental influences on phenotypes. Therefore, the present study examined the validity of two similarity questionnaires used in the German TwinLife study, in which data from 4,097 twin pairs and their families were assessed: twin children’s zygosities were determined with the Zygosity Questionnaire for Young Twins, which was administered in parent-report form. For adolescent twins, the Self Report Zygosity Questionnaire was used. For the present validation analyses, DNA samples of N = 328 twin pairs were collected via buccal swabs. In this DNA subsample, questionnaires were filled out by parents for n = 212 (aged 4 to 12 years) twin pairs while self-reports were collected from n = 116 adolescent twins (16 to 23 years of age). Using DNA-based zygosity as criteria, correct classification rates of 97% for parent- and 92% for self-reports were established and cross-validated. Additionally, classification rates based on a single item and variants of questionnaire based zygosity determination used in other twin studies were calculated and compared. Implications of incorrectly classified zygosity on genetic and environmental estimates in twin studies are discussed.

The main aim of this study was to develop and optimize a questionnaire-based zygosity determination method in Chinese adolescent twins. Participants were 471 pairs of same-sex twins (345 monozygotic, 126 dizygotic) with a mean age of 14.56 years (SD = 2.62). A second sample was recruited for cross-validation, including 382 pairs of same-sex twins (261 monozygotic, 121 dizygotic) with a mean age of 12.53 years (SD = 2.22). The questionnaire consisted of 12 questions dealing with co-twin similarity or frequency of confusion. Two means were put forward to improve the predictive accuracy of the questionnaire — adding parent-reports to the analysis, and using a 2-point rather than 3-point response format. DNA genotyping was performed on nine short tandem repeat loci, with an estimated zygosity classification accuracy very close to 100%. The validity of all questionnaires was assessed by being compared to the results of DNA analysis. Results of stepwise logistic regression analysis showed that the predictive accuracy of the 3-point self-reported questionnaire is 83.8%. Using parent-reports and 2-point scale led to 3.9% and 4.6% increase in predictive accuracy, respectively. When using the parent-reports and children's self-reports jointly, the predictive accuracy was enhanced to 90.6%. For the cross-validation, the equations and cut-offs derived from the first sample led to an acceptable accuracy (91.3%) in the second sample. In conclusion, the method we developed can be used in future studies among Chinese adolescent twins. Multiple-rater and 2-point response format were suggested for all twin studies for enhancing the predictive accuracy of questionnaires.

The use of DNA fingerprinting with synthetic oligonucleotides is illustrated for practical applications familiar in clinical diagnostics: pre- and postnatal zygosity determination and the monitoring of bone marrow transplantation. A simple method for non-radioactive detection is described which may be interesting for many diagnostic laboratories.

Summary. Objective. To assess concordance of tooth size and occlusal morphology in monozygotic (identical) and dizygotic (non-identical) twins compared to unrelated controls in order to determine how much these dental traits are related to inheritance. Methods. Mesiodistal and bucco-lingual dimensions of tooth size in 34 pairs of twins were measured with dial callipers. In addition, the occlusal morphology of nine pairs of teeth was assessed using a co-ordinate measuring machine with a non-contact laser probe. Results. The results showed greater concordance in tooth size for both mesiodistal and bucco-lingual dimensions within twin pairs compared to twin and unrelated controls. Furthermore, monozygotic (MZ) twins showed greater concordance than dizygotic (DZ) twins for all permanent teeth with statistically significant variance for the mesiodistal dimension (P = 0·01) but not bucco-lingual dimension. Superimposed computer models of the digitized occlusal surfaces showed a high degree of similarity for MZ twins compared to DZ twins. The mean intrapair deviation was 147 µm for MZ, 209 µm for DZ and 258 µm for unrelated controls. Conclusions. The remarkable similarity in the tooth size and morphology of monozygotic twins suggests a strong inheritability factor to tooth size and shape and that these may be useful as additional tools for zygosity determination along with other dental traits.

This study examined cross-validation and test-retest reliability of questions and questionnaire indices commonly used for twin zygosity classification. Mothers of 58 monozygotic (MZ) and 52 dizygotic (DZ) same sex twin pairs were interviewed by telephone to answer questions regarding the similarity of their twins (mean age = 14.6 ± 2.8 years). A logistic regression equation correctly classified 91% of both MZ and DZ twin pairs in our sample using 7 of the 12 zygosity questions. The internal consistency for the total questionnaire (Cronbach's alpha) was 0.88. The median two month temporal stability estimate for the individual questions was r = .56 and r = .79 for the test total. For the cross-validation, zygosity classification indices taken from 9 previous studies were applied to our sample and compared to classification according to DNA microsatellite analyses (agreement range = 44 to 100%). The accuracy of the classification indices was significantly lower than the original studies for 62% of the comparisons. If zygosity determination with DNA markers or blood group typing for all subjects is not feasible, rather than using classification indices based on other studies, an optimal classification scheme can be achieved by using a zygosity questionnaire of which the reliability and validity of the questions is established in a random subsample of the same twin cohort.

It often happens that an efficient algorithm can be designed by considering some special properties of the problem. In this short paper we describe a problem in twin zygosity determination that involves a convolution of probability distributions. Our solution to this problem was to develop an algorithm for counting in the base-3 or “ternary” number system.

It is generally held that there are genetic factors for dizygotic (DZ) twinning, but the pattern of inheritance is far from clear.We report on a rare pedigree with three sets of spontaneous DZ female twin births in three successive generations. To our knowledge no similar pedigrees have been reported in literature.Zygosity determination in three pairs of twins was performed with ABO, MN and Rh blood groups from erythrocytes and with STR analysys from samples of saliva.In our pedigree, the direct trasmission of the DZ twinning through the maternal line seems show an autosomal dominant pattern of inheritance. We believe that the inheritance of DZ twinning appears to be recessive in most case and autosomal dominant only in rare instances. It is plausible with the hypothesis that the expression of the dizygotic twinning trait in a mother shows a genetic heterogeneity.

Discordance for Turner syndrome in monozygotic (MZ) twins, which is known as heterokaryotypia, is very rare in MZ pregnancies. The combined effect of idiochromosome loss due to an anaphase lag and the relocation of discordant blastomeres may trigger the twinning procedure and discordance of Turner syndrome. We present 2 cases of MZ twins discordant for Turner syndrome that were diagnosed prenatally by ultrasound and cytogenetic studies (one fetus was 45,X and the other 46,XX). Both cases, which involved monochorionic (MC) diamniotic twins, underwent selective feticide and had favorable outcomes for the remaining twin. Ultrasound, amniocentesis of both sacs (dual amniocentesis) and zygosity determination are indispensable in diagnosing heterokaryotypia. Selective feticide is a treatment option in cases of heterokaryotypic MC diamniotic twins, and in our cases, it resulted in favorable outcomes for the remaining twin.

Precision of zygosity determination in twins can be improved by the use of modern methods of DNA analysis. The clinical application of 4 single- (SLS) and 2 multi-locus (MLS), and 6 PCR (polymerase chain reaction) systems for zygosity determination in 12 twin pairs with oral clefts was compared with regard to the quality and quantity of sample material required and the probability of error in monozygosity determination. PCR systems proved to be superior to SLS or MLS, as DNA sampling is much more convenient, while its level of accuracy still fulfils clinical requirements. For this reason, PCR systems should be considered a basic method in modern clinical twin research.

The Swedish Twin Registry (STR) today contains more than 194,000 twins and more than 75,000 pairs have zygosity determined by an intra-pair similarity algorithm, DNA, or by being of opposite sex. Of these, approximately 20,000, 25,000, and 30,000 pairs are monozygotic, same-sex dizygotic, and opposite-sex dizygotic pairs, respectively. Since its establishment in the late 1950s, the STR has been an important epidemiological resource for the study of genetic and environmental influences on a multitude of traits, behaviors, and diseases. Following large investments in the collection of biological specimens in the past 10 years we have now established a Swedish twin biobank with DNA from 45,000 twins and blood serum from 15,000 twins, which effectively has also transformed the registry into a powerful resource for molecular studies. We here describe the main projects within which the new collections of both biological samples as well as phenotypic measures have been collected. Coverage by year of birth, zygosity determination, ethnic heterogeneity, and influences of in vitro fertilization are also described.

In twin pregnancy studies, molecular genetic techniques have rarely been used to determine zygosity, despite their known precision and accuracy. The present work aimed to assess the power of discrimination in zygosity assessment, using a set of microsatellite markers that were routinely used for aneuploidy screening by multiplex-PCR in a prenatal context. Rapid aneuploidy screening using a group of 20 microsatellite markers (STRs) located on chromosomes 13, 18, 21 and X has been performed in our lab for over 10 years, with a total of approximately 1,500 samples studied to date. A retrospective analysis of the 257 prenatal samples from multiple pregnancies was carried out. A subset of 14 cases presenting theoretical monozygosity were re-evaluated by the use of biostatistics tools accessed via the ZygProb website. Further monozygosity determination relative to dizygosity was calculated, given an estimated overall error value of 0.093%. The results show that monozygosity had been correctly determined in all our previously studied twins. This work demonstrates that accurate zygosity assessment can be achieved with the same STRs applied in aneuploidy screening with a high power of discrimination and a matching probability of over 99.999999%.

The study of twins is a well-established method for evaluating the relative roles of heredity and environmental factors in the etiology of diseases. Conclusions depend on zygosity determination and on the classification of minor forms of diseases. This paper reports on ten (5 mono- and 5 dizygotic) out of thirteen twin pairs among 1039 patients with cleft lip and palate (n = 677) or cleft palate (n = 362). Zygosity was determined using “DNA fingerprinting” on blood samples In all 10 pairs and on cleft-associated tissue in one pair. Including minor forms of clefting, two of five pairs of monozygotic and two of five pairs of dizygotic twins of the same sex showed concordance. “DNA fingerprinting” should be established as a definitive method for zygosity determination, and the calculation of concordance rates should always include minor forms of diseases.

The National Heart, Lung and Blood Institute (NHLBI) twin study is a collaborative, longitudinal study of the role of genetic risk factors on subsequent cardiovascular disease processes in 514 pairs of white, male World War II and Korean veteran twins born between 1917 and 1927. This paper describes the sampling procedures and zygosity determination at the initial examination, participation by the cohort members at later examinations, and a summary of the variables provided for Genetic Analysis Workshop 8 (GAW8).

Although numerous genetic and anthropological markers are available for determining zygosity of twins, there is still a need for a more practical and informative method in zygosity diagnosis. Dinucleotide repeats or other short repeats (microsatellites) are highly variable between individuals and offer a simple, fast, cheap, and exact approach for zygosity determination. The feasibility of a set of microsatellites to be used for this purpose is demonstrated.

This study reports on the inter- and intrarater reliability of physical features observation. Study subjects were 176 Chinese adult persons, consisting of 89 males and 87 females. Three trained research assistants responded simultaneously and respectively to 12 items regarding the subject's physical features including ‘hair’, ‘Mongoloid folds’, left and right ‘ear lobes’, ‘earwax’, ‘nostril shape’, ‘tongue rolling’, left and right ‘hitchhiker's thumb’, ‘mid-digital hair’ and left and right ‘simian crease’ at the moment of interview. And 14 days later, these subjects received the same observation once again. The results showed that the inter- and intra-observer agreements of ‘hair’, ‘earwax’, ‘tongue rolling’, ‘mid-digital hair’ and ‘simian crease’ were almost perfect with most kappa (κ) coefficients ≥ .80, while ‘Mongoloid fold’ and ‘nostril shape’ showed poor inter-observer agreement and ‘nostril shape’ showed poor intra-observer agreement (κ < .40). Two other physical features, ‘hitchhiker's thumb’ and ‘ear lobes’ showed moderate inter-observer agreement and three features, ‘hitchhiker's thumb’, ‘ear lobes’ and ‘Mongoloid fold’, showed moderate intra-observer agreement (.40≤ κ

Monozygotic twins (MZ) are rarely absolutely "identical." This review discusses the types of genetic/epigenetic and prenatal environmental post-zygotic mechanisms that cause discordance within such twin pairs. Some of these mechanisms--ranging from heterokaryotypia to skewed X-chromosome inactivation--may cause extreme discordance, but these extremes are merely the more emphatic examples of discordance that, to some degree, underlies the majority of MZ twin pairs. Because of the entrenched misconception that MZ twins are necessarily identical, many MZ twin pairs are mistakenly designated as dizygotic (DZ). Clinical benefits to accurate zygosity determination include correct solid organ transplantation matching, if one twin requires donation for a non-genetically mediated disease; the opportunity of preventive management for disorders that do not manifest synchronously; and better counseling to parents regarding their individually unique, and often psychologically puzzling, twin offspring. In twin pairs with complex and confusing biological origins, more detailed zygosity testing may be required. For example, intermediate trigametic and tetragametic chimeric dizygotic twins are reviewed, some of whom are, nevertheless, monochorionic (MC). Because of inter-fetal vascular anastomoses in MC twins, genetic results from blood samples may not accurately reflect discordance in solid organs. Previously, it was thought that MZ twinning was some sort of embryological fluke. However, familial monozygotic twinning is more common than suggested by the literature. Seven new families are presented in an accompanying paper. Despite the difficulties and dangers of twin pregnancy (especially so for MC twins), human twinning persists, and continues to both challenge and fascinate parents, clinicians and geneticists.

This paper described a number of critical assumptions and methodological issues that influence the interpretation of data from family, twin and adoption studies of substance abuse. Also, using data from a recent study, it examined the role of one methodological issue (method of zygosity determination) and one critical assumption (representativeness of twins as research subjects) to determine the validity of data in twin studies of substance abuse. The accuracy of the questionnaire methods for zygosity determination in alcoholic twins was similar to that previously reported for nonalcoholic twins. Also, alcoholic twins were found to be representative of both nonalcoholic twins and alcoholics in general.

Comparisons of physical features within identical (monozygous) and non-identical (dizygous) twin pairs have provided valuable insights into the relative contributions of genetic and environmental influences to observed variability. The special nature of the twinning process itself also provides an opportunity to learn more about early human development, including how body symmetry is determined. The mechanisms of twinning, mortality and morbidity in twins, determination of body symmetry including the phenomenon of mirror-imaging, postnatal growth and development of twins, and zygosity determination are discussed. Twin studies with direct relevance to clinical dentistry are reviewed and illustrated by examples from an ongoing investigation of dentofacial morphology in South Australian twins.

Simple heritability estimators of continuous as well as discrete traits from twin data are known to overestimate the degree of genetic determination of the measured traits for several reasons. Errors of zygosity determination will, however, underestimate the true heritability. The bias due to wrong assignment of dizygous twin pairs into monozygous type is evaluated here, and the results indicate that this negative bias has a compensatory effect on the estimate of the degree of genetic determination when other factors of similarity between twin pairs are taken into account. It is shown that when an estimate of zygosity error is available, the bias due to this factor can be evaluated quantitatively, and hence the adjustment for zygosity error can be incorporated in the estimation of the degree of genetic determination of a trait.

Many studies of embryogenesis and fate of twin pregnancies are invalidated because zygosity is not determined definitively, or is assumed on the basis of inadequate criteria. This paper briefly reviews methods of zygosity determination. It reports published results and a new series of twins in which zygosity was determined by DNA fingerprinting. Implications for methods of prenatal diagnosis of zygosity are discussed in the context of the occasional need for intervention in twin transfusion syndrome or in twins discordant for major malformations. Definitive zygosity and placental anatomy (number of chorions and amnions) is discussed as the firm substrate for studies of normal and abnormal twin development.

Establishment and maintenance of transgenic mouse strains require being able to distinguish homozygous from heterozygous animals. To date, the developed real-time quantitative PCR techniques are often complicated, time-consuming and expensive. Here, we propose a very easy and rapid method with a simple data analysis to determine zygosity in transgenic mice. We show that the real-time quantitative PCR using SYBR® Green fluorescent dye can be applied to discriminate two-fold differences in copy numbers of the transgene. Our procedure has to fit only three simple requirements: (1) to design primers capable of detecting one Ct difference for two-fold differences in DNA amounts (2) to measure genomic DNA concentrations accurately and (3) to have a reference animal of known zygosity in each run. Then, if the Ct values for the control gene are similar in all samples, we are able to compare directly the Ct values for the transgene in every sample, and so, to deduce the zygosity status of each mouse relative to the reference animal. This method is really simple and reliable, and it may be valuable as a rapid screening tool for zygosity status in transgenic animals.

BACKGROUND: Knowledge about paternal RHD hemi- or homozygosity is of clinical interest in alloimmunized pregnant women. D negativity in white persons is usually caused by deletion of the RHD gene. Recently, the physical structure of the RH locus and the mechanism causing the deletion of the RHD gene have been explored, enabling RHD zygosity determination in white persons by specific detection of a hybrid Rhesus box characteristic for the RHD– locus. STUDY DESIGN AND METHODS:RHD zygosity was determined in 402 samples from five different ethnic groups by polymerase chain reaction (PCR)-restriction fragment length polymorphism and by a newly developed real-time quantitative PCR. The Rhesus boxes of samples showing discrepancies between both tests were cycle sequenced. RESULTS: In nonwhite persons, several mutated Rhesus boxes exist that hamper zygosity determination by detection of the RHD– locus. Such mutated Rhesus boxes in D+RHD homozygous black persons have a frequency of 0.22. In white persons, no mutated Rhesus boxes were encountered so far. CONCLUSIONS: Owing to the high frequency of the mutated Rhesus boxes, zygosity determination by detection of the RHD– locus is not feasible in nonwhite persons. The cosegregation of variant RHD genes (RHDψ and (C)cdes) with specific mutated Rhesus boxes yields more insight into the evolutionary events concerning variant RHD genes and mutated Rhesus boxes.

The true incidence of monozygosity after assisted reproduction is unknown. Proxy estimations, such as counting the number of monochorionic twins, using Weinberg's differential rule, and counting cases where the number of fetuses exceeds the number of transferred embryos, are less accurate than zygosity assessment in all twins. These methods commonly underestimate the true frequency of zygotic splitting because they disregard like-sexed monozygotic-dichorionic twins, consider twin births rather than twin pregnancies, and do not count the number of higher-order multiples with a monozygotic pair. Because zygotic splitting following assisted reproduction is of biological interest as well as of clinical significance, efforts should be directed to increase the accuracy of zygosity determination. This will improve understanding of the zygotic splitting phenomenon and its relation to pathologic processes.

The only known twin pair evidently discordantly affected for the BDLS (Brachmann-de Lange syndrome) and who had been considered monozygotic (MZ) based on blood analysis remained a problem because biological zygosity determination needed further typing. In this report we review the clinical findings of this pair of twins at the age of 20. The use of DNA fingerprinting with three multilocus probes, F10, DNF24, and 33.6, allowed us to present evidence of monozygosity with a high degree of certainty. The significance of this confirmation of discordance in determining the cause of BDLS is discussed. Intensive comparative genomic studies of the discordant twin sisters may be useful to unravel the molecular genetics of this enigmatic pattern of malformation.

STR typing becomes more and more valuable for different approaches of science. It revolutionized determining of zygosity in twin research and it is very often the only possibility for discrimination in forensic trace analysis. In this study 55 twin pairs from Denmark were genetically investigated to determine their zygosity. For analysis two multiplex PCR kits, the AmpFlSTR Identifiler kit, which comprises 15 loci plus the amelogenin gender determination and the Powerplex ES kit with eight different loci were employed. For a forensic approach every twin pair was regarded as being a forensic trace analysis, i.e. suspect or victim/biological trace and then we determined the security and precision with that a match of genetic patterns or an exclusion could be observed. Sixty percent of the twin pairs were di- and 40% monozygotic. There were no differences in zygosity determination between the two multiplex kits. The lowest number of exclusions by determining dizygosity was four loci for the Identifiler and three for Powerplex ES kit, the highest was 13 (Identifiler) and eight (Powerplex). It could be shown that the highly discriminative multiplex PCR kits gave matching probabilities of over 99.999999% (calculation based on data for unrelated individuals) even when only five or six STRs could be determined (assuming a trace analysis with some non-detectable STRs and therefore an incomplete genetic pattern). No questionable results regarding zygosity of the twin pairs were obtained even when only eight loci (using the Powerplex ES kit) were investigated. The simulated forensic results showed that matching probabilities should always be handled with care to not possibly come to wrong conclusions concerning the origin of the biological trace.

The usefulness of PCR systems (D1S80, D17S30, ApoB, TC11, VWA, and SE33) is discussed. The statistical evaluation shows that these systems – together with other systems – are well suited for zygosity testing.
