Background: Marinesco-Sjögren syndrome (MSS) is an autosomal recessive neuromuscular disorder that arises in early childhood and is characterized by congenital cataracts, myopathy associated with muscle weakness, and degeneration of Purkinje neurons leading to ataxia. About 60% of MSS patients have loss-of-function mutations in the SIL1 gene. Sil1 is an endoplasmic reticulum (ER) protein required for the release of ADP from the master chaperone Bip, which in turn will release the folded proteins. The expression of non-functional Sil1 leads to the accumulation of unfolded proteins in the ER and this triggers the unfolded protein response (UPR). A dysfunctional UPR could be a key element in the pathogenesis of MSS, although our knowledge of the molecular pathology of MSS is still incomplete. Methods: RNA-Seq transcriptomics was analysed using the String database and the Ingenuity Pathway Analysis platform. Fluorescence confocal microscopy was used to study the remodelling of the extracellular matrix (ECM). Transmission electron microscopy (TEM) was used to reveal the morphology of the ECM in vitro and in mouse tendon. Results: Our transcriptomic analysis, performed on patient-derived fibroblasts, revealed 664 differentially expressed (DE) transcripts. Enrichment analysis of DE genes confirmed that the patient fibroblasts have a membrane trafficking issue. Furthermore, this analysis indicated that the extracellular space/ECM and the cell adhesion machinery, which together account for around 300 transcripts, could be affected in MSS. Functional assays showed that patient fibroblasts have a reduced capacity of ECM remodelling, reduced motility, and slower spreading during adhesion to Petri dishes. TEM micrographs of negative-stained ECM samples from these fibroblasts show differences of filaments in terms of morphology and size. Finally, structural analysis of the myotendinous junction of the soleus muscle and surrounding regions of the Achilles tendon revealed a disorganization of collagen fibres in the mouse model of MSS (woozy). Conclusions: ECM alterations can affect the proper functioning of several organs, including those damaged in MSS such as the central nervous system, skeletal muscle, bone and lens. On this basis, we propose that aberrant ECM is a key pathological feature of MSS and may help explain most of its clinical manifestations. [ABSTRACT FROM AUTHOR]

The present work aims to develop and evaluate the wound healing potential of bakuchiol nanoemulsion loaded electrospun scaffolds. Since oxidative stress and microbial burden leads the burn wounds to become chronic and fatal to patients, a phytoconstituent, bakuchiol (BAK), was screened on the basis of antioxidant and antimicrobial potential which also defined its dose. Furthermore, BAK was incorporated into a nanoemulsion to enhance its therapeutic efficacy, reduce its dosage frequency, and maximize its stability. The present study is inclined towards the collaborative interaction of natural products and novel drug delivery systems to develop safe and therapeutically efficient systems for burn wound healing. The optimized nanoemulsion showed excellent antioxidant and antimicrobial potential against wound susceptible pathogens, i.e., Candida albicans and Methicillin-resistant Staphylococcus aureus which was further loaded into gelatin based hydrogel and nanofibrous scaffold system. The mesh structure of scaffolds was chosen as a suitable carrier system for wound healing process not only because it offers resemblance to skin's anatomy but is also capable of providing uniform distribution of wound biomarkers across the skin. The prepared nanofibers were assessed for their analgesic, anti-inflammatory, and wound healing potential which was observed to be significantly better than its gel formulation. Introduction: Unintended wounding or bruising during harvest or postharvest handling leads to significant tuber loss and imposes economic burden to potato industry. Therefore, finding effective strategies to mitigate wound-related tuber losses is very important from industry perspectives. Formation of protective barrier through accumulation of suberin polyphenolics (SPP) is a natural and initial response of potato tuber tissues to wounding. Materials and methods: In this study, efficacy of two natural elicitors, such as chitosan oligosaccharide (COS 0.125 g L-1) and cranberry pomace residue (Nutri-Cran 0.125 g L-1) was investigated using a mechanically wounded tuber tissue model and by histological determination of SPP formation in five agronomically relevant and red-skin potato cultivars (Chieftain, Dakota Rose, Dakota Ruby, Red LaSoda, Red Norland). Furthermore, the potential role of stress protective metabolic regulation involving phenolic metabolites, proline, and antioxidant enzymes in tuber WH processes were also investigated during 0-9 days after wounding. Results and discussion: Exogenous treatments of both COS and Nutri-Cran resulted into enhanced SPP formation in wounded surface, but the impact was more rapid with Nutri-Cran treatment in select cultivars. Greater contents of total soluble phenolic, ferulic acid, chlorogenic acid, total antioxidant activity, and superoxide dismutase activity were also observed in elicitor treated tuber tissues at different time points after wounding. Nutri-Cran treatment also reduced the activity of succinate dehydrogenase in Red Norland and Dakota Ruby at 3 d, indicating a suppression in respiration rate. Collectively, these results suggest that Nutri-Cran can be potentially utilized as an effective WH treatment to potato tubers for minimizing wound-related losses. [ABSTRACT FROM AUTHOR]

Sedum telephium is a succulent plant used in traditional medicine, particularly in Italy, for its efficacy in treating localized inflammation such as burns, warts, and wounds. Fresh leaves or freshly obtained derivatives are directly applied to the injuries for these purposes. However, challenges such as the lack of microbiologically controlled materials and product standardization prompted the exploration of more controlled biotechnological alternatives, utilizing in vitro plant cell cultures of S. telephium. In the present study, we used HPLC-DAD analysis to reveal a characteristic flavonol profile in juices from in vivo leaves and in vitro materials mainly characterized by several kaempferol and quercetin derivatives. The leaf juice exhibited the highest content in total flavonol and kaempferol derivatives, whereas juice from callus grown in medium with hormones and callus suspensions showed elevated levels of quercetin derivatives. The in vitro anti-inflammatory and wound-healing assays evidenced the great potential of callus and suspension cultures in dampening inflammation and fostering wound closure, suggesting quercetin may have a pivotal role in biological activities. Wound healing is a complex biological process crucial for restoring tissue integrity and preventing infections. The development of advanced materials that facilitate and expedite the wound-healing process has been a focal point in biomedical research. In this study, we aimed to enhance the wound-healing potential of hydrogel scaffolds by incorporating graphene oxide and poly (ethylene glycol) methyl ether methacrylate (MEO2MA). Various masses of graphene oxide were added to MEO2MA hydrogels via free radical polymerisation. Comprehensive characterizations, encompassing mechanical properties, and biocompatibility assays, were conducted to evaluate the hydrogels' suitability for wound healing. In vitro experiments demonstrated that the graphene oxide-based hydrogels exhibited a proper swelling degree and tensile strength, responding effectively to moisture conditions and adhesiveness for wound healing. Notably, the tensile strength significantly increased to 626 kPa in the graphene oxide hydrogels. Biocompatibility assessments revealed that the graphene oxide/MEO2MA hydrogels were non-toxic to human dermal fibroblast cell growth, with no significant difference in cell viability observed in the graphene oxide/MEO2MA hydrogel (H-HG) group. In a rat skin experiment, the wound-healing rate of the hydrogel incorporating graphene oxide surpassed that of the pristine hydrogel after a 15-day treatment, achieving over 95% wound closure in the H-HG group. The histopathological analysis further supported the efficacy of the H-HG hydrogel dressing in promoting more effective tissue regeneration. Self-adaptive stage (SAS) is defined as the period between harvest and pre-cooling in this study, which is extremely neglected in the field of postharvest horticultural products. This study aimed to investigate resistant responses and wound-healing activities during the SAS. Broccoli heads that needed cut-harvest were used as experimental materials, and the indices related to storage quality, reactive oxygen species, and phenylpropanoid metabolism were determined during 12 h after the harvest. The results showed that the quality of broccoli heads did not change significantly within 12 h after cut-harvest compared with the normal-growth group. The rapid water loss indicated that measures should be taken for water prevention during SAS. The cut-harvest group showed a significant accumulation of H2O2 after 3 h, which acted as a signal in regulating the antioxidant enzyme system. The activities of phenylalanine ammonia-lyase, cinnamate 4-hydroxylase, and 4-coumarate coenzyme A ligase increased in cut-harvest broccoli stems during 0–6 h, 3–12 h, and 0–9 h, respectively, compared with those in normal-growth broccoli stems. The enhanced enzyme activity involved in phenylpropanoid metabolism and accumulated phenolic acids associated with wound healing at different time points during SAS demonstrated that the response time should be considered when regulating resistance and wound healing. This study focused on revealing the resistance and wound-healing activities of broccoli stems during independent growth after cut-harvest, thus providing a theoretical basis for the subsequent study and regulatory measurement in cut-harvest vegetables. Abstract This study presents the first findings regarding extraction, isolation, enzyme inhibition, and antioxidant activity. The oral mucosal wound‐healing process was investigated using propolis water extract (PWE) incubation with gingival fibroblast cells and concluded that propolis was effective on the oral mucosal wound‐healing pattern compared to untreated controls. Additionally, phenolic compounds (fraxetin, apigenin, galangin, pinobanksin, chrysin, etc.) were isolated from propolis, and their chemical structures were elucidated using comprehensive spectroscopic methods. The antioxidant and anti‐Alzheimer potential activities of PWE and some isolated compounds were screened and revealing their inhibitory effects on acetylcholinesterase (AChE) with IC50 values ranging from 0.45 ± 0.01 to 1.15 ± 0.03 mM, as well as remarkable free‐radical scavenging and metal reduction capacities. The results suggest that these compounds and PWE can be used as therapeutic agents due to their antioxidant properties and inhibitory potential on AChE. It can also be used for therapeutic purposes since its wound‐healing effect is promising.

Abstract Regarding different medical benefits of fungi, using the medical mushroom extracts as wound-healing agents is gaining popularity. This study, evaluated the wound healing characteristics of Trametes versicolor. Anti-oxidant activity addressed by employing the DPPH (2,2-diphenyl-1-picrylhydrazyl) assay resulting 53.7% inhibitory effect. Besides, for anti-microbial ability determination, the MIC (Minimum Inhibitory Concentration) of extract measured which Escherichia coli growth was inhibited at 1.1 mg/ml, and Staphylococcus aureus did not grow at 4.38 mg/ml of extract. The MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) method indicated dose dependence of the extract with 63 ± 3% and 28 ± 3% viability at 1250 μg/ml and 156.25 μg/ml of extract, which higher concentration caused higher cell viability. The outcome of gene expression analysis determined that overall expression of FGF2 (Fibroblast Growth Factor 2), IL-1β (Interleukin-1β), and TGF-β1 (Transforming Growth Factor-β1) was 4 times higher at 48 h than at 24 h in treated cells, suggesting a stimulating effect on cell growth. An in-vivo animal model suggested enhanced wound healing process after treatment with 0.01 g of extract. Furthermore, the number of fibroblasts, epidermal thickness, and collagen fiber was respectively 2, 3, and threefold higher in treated mice when compared to untreated mice. The treated wounds of mice showed 100% and 60% of untreated mice of healing within 14 days. The results of this research show promise for the fungus-based wound healing treatments, which may help with tissue regeneration and the healing of cutaneous wounds.

The association of endophytic fungi with plants was recorded from an early stage of evolution. Endophytic fungi of medicinally important plants are given importance as they mimic the synthesis of bioactive compounds by the host plant. Chromolaena odorata is a wild medicinal plant used in ethnomedicine by different communities. From the root part of C. odorata, an endophytic fungus, Chaetomium sp. R1C1, was isolated, and its biological activities were evaluated. The ITS-rDNA gene sequencing of Chaetomium sp. R1C1 showed 99.21% similarity with Chaetomium globosum. Chaetomium sp. R1C1 was found to secrete protease, amylase, lipase, and cellulose and also produce ammonia. The isolate shows strong antifungal activities against all the nine tested phytopathogens, with the highest inhibition against U. virens (97.45%), followed by A. flavus (94.70%), C. lunata (89.09%), A. brassicicola (78.41%), F. oxysporum (77.71%), R. solani (70.27%), C. capsici (64.41%), A. niger (57.06%), and A. alternata (45.35%). The antibacterial activity evaluated using the agar-well diffusion method and 96-well microtitre method shows that the crude extract was more efficient against gram-negative bacteria, with the highest activity against S. typhi. The DPPH radical scavenging activity shows inhibition of 60.18% and an IC50 value of 85.88 µg/mL, which makes it a strong antioxidant. GC-MS and LC-MS are powerful tools employed to reveal the compounds present in the secondary metabolite of Chaetomium sp. R1C1. This study investigated the characterization, antifungal activity, and biocompatibility of green agar/silver and collagen/silver bionanocomposite films for wound healing and cell growth scaffolds. Silver nanoparticles (AgNPs) are known for their antimicrobial properties, but their toxicity and harsh synthesis limit their applications. To address this, green‐synthesized AgNPs G‐AgNPs were incorporated into agar/collagen suspensions at specific concentrations and three different G‐AgNP‐agar and two different G‐AgNP‐col bionanocomposite films were produced. Nanoparticle homogeneity and film quality were characterized through SEM analysis. Mechanical properties were tested using a uniaxial tensile tester, revealing that the bioplastic control samples exhibited UTS of 3.86 MPa compared to 0.60 MPa for collagen, a 6‐fold improvement. Viable cell metabolic activity derived from MTT assay showed that Col‐4%AgNPs and Bio‐30%AgNPs had a 42.9% and 51.6% increase in net metabolic activity respectively compared to control on day 4. Fluorescence microscopy confirmed enhanced cell adhesion and proliferation in G‐AgNP‐incorporated samples. Antifungal properties were evaluated against Cladosporium spores, able to cause severe diseases when in contact with human skins, following ISO 16869:2008 standards. The demonstrated unique properties and tunability of G‐AgNPs bionanocomposites can be employed in a variety of specialties for wound‐healing applications, to improve rate and quality of healing while reducing the risk of infection. Algae biomass has been recognized as feedstock with diverse application including production of biofuel, biofertilizer, animal feed, wastewater treatment and bioremediation. In addition, algae species are a potential reservoir of metabolites and polymers with potential to be utilized for biomedicine, healthcare and industrial purposes. Carrageenan is one such medicinally and industrially significant polysaccharide which is extracted from red algae species (Kappaphycus alvarezii and Eucheuma denticulatum, among the common species). The extraction process of carrageenan is affected by different environmental factors and the source of biomass, which can vary and significantly impact the yield. Diverse applications of carrageenan include hydrogel beads, bio-composites, pharmacological properties, application in cosmetics, food and related industries. Carrageenan biological activities including antioxidant, anti-inflammatory, antimicrobial, and antitumor activities are significantly influenced by sulfation pattern, yield percentage and molecular weight. In addition to natural biomedical potential of carrageenan, synergetic effect of carrageenan- nanocomposites exhibit potential for further improvisation of biomedical applications. Nanotechnology driven bio-composites of carrageenan remarkably improve the quality of films, food packaging, and drug delivery systems. Such nano bio-composites exhibit enhanced stability, biodegradability, and biocompatibility, making them suitable alternatives for drug delivery, wound-healing, and tissue engineering applications. Introduction: Wound healing is a complex physiological process that involves a series of coordinated events that recover structural and functional tissue integrity. The desire to promote nature-based solutions has led to the screening of various plants and herbs for their wound healing potentials with results showing the enhancement of various aspects of the wound healing process. Dioscorea bulbifera possesses a wide range of applications in folk remedies. In traditional Chinese medicine, it is used in the treatment of mumps, goitre, sore throat, gastric cancer and carcinoma of the rectum alone or in combination with other plants. The present study evaluated the effect of hydromethanolic extract of Dioscorea bulbifera (HEDB) on wound size, percentage of wound contraction and tissue hydroxyproline in male Wistar rat excision wound models. Methods: Twenty-five (25) male Wistar rats (180–200 g) were used for the study. They were divided into five (5) groups comprising five (5) animals per group. A circular excision wound (1.5–2 cm) was experimentally induced in each animal following ketamine anaesthesia. Groups I and II served as negative and positive controls and were treated with topical administration of distilled water and petroleum jelly respectively. Groups III, IV and V served as the experimental groups and were treated with 200, 400 and 800 mg/kg of HEDB topically. The diameter of the wound was measured and the wound area and percentage of wound contraction were calculated. Tissue hydroxyproline was determined using tissue excised from the healed portion of the dorsal surface of the animals via coulometric standard procedures. Results: Data from the study indicates that animals in the experimental groups (III, IV and V) had an earlier wound closure compared to the control. Animals treated with 200 mg/kg HEDB (Group III) showed complete wound closure by day 17, whereas animals treated with 400 mg/kg (Group IV) and 800 mg/kg HEDB (Group V) groups achieved complete wound closure by the 18th and 20th day of the experiment respectively. Similarly, all the animals in the experimental groups had significantly increased percentage of wound contraction compared to negative control (P < 0.05). Tissue hydroxyproline was significantly raised amongst the animals treated with 200 and 800 mg/kgHEDB compared to the negative control (p < 0.05). Discussion: Evidence from the study has shown that HEDB possesses potent wound healing activity as the present study has demonstrated that it reduced wound area, and increased wound contraction and tissue hydroxyproline on experimentally induced excision wound male Wistar rat models. These findings provide scientific evidence that validates the traditional medicinal application of Dioscorea bulbifera in promoting wound healing.

AbstractThe biogenic manufacture of nanoparticles utilising endophytic fungus is an eco-friendly, cost-effective, and secure alternative to constructing chemical methods. The prime focus of the study was to fabricate ZnONPs using the biomass filtrate of endophytic Xylaria arbuscula isolated from Blumea axillaris Linn. and to evaluate their biological properties. The characterisation of the biosynthesized ZnO-NPs was done utilising both spectroscopic and microscopic methods. The bioinspired NPs showed a surface plasmon peak at 370 nm; SEM and TEM micrographs illustrated the hexagonal organisation; XRD spectra proved the crystalline phase as hexagonal wurtzite; EDX analysis confirmed the presence of zinc and oxygen atoms; and the zeta potential analysis proved the stability of ZnONPs. In addition, they also demonstrated significant concentration-dependent inhibition of antimicrobial, antioxidant, anti-inflammatory, and antidiabetic potential in comparison with the reference drugs. In vitro cytotoxicity and wound healing potential of ZnONPs were examined in L929 cell lines, illustrating that they accelerated the wound healing process by roughly 95.37 ± 1.12% after a 24-h exposure to ZnONPs. The photocatalytic activity of the ZnONPs was examined by degrading the methylene blue dye under solar irradiation. In conclusion, our outcomes showed that mycosynthesized ZnONPs possessed potent bioactivity and could be an excellent choice for biomedical applications.

In vitro cytotoxicity and wound healing potential of ZnONPs were examined in L929 cell lines, illustrating that they accelerated the wound healing process by roughly 95.37 ± 1.12% after a 24-h exposure to ZnONPs. The photocatalytic activity of the ZnONPs was examined by degrading the methylene blue dye under solar irradiation. Between 2013 and 2018, there has been a 71% increase in the number of patients who have required wound care in the NHS and such large numbers has placed a significant burden on healthcare systems. However, there is currently no evidence as to whether medical students are equipped with the necessary skills to deal with an increasing number of wound care related issues that patients present with. A total of 323 medical students across 18 UK medical schools completed an anonymous questionnaire evaluating the wound education received at their medical school, encompassing the volume, content, format and efficacy of teaching. 68.4% (221/323) of respondents had received some form of wound education during their undergraduate studies. On average students received 2.25 h of structured, preclinical teaching and only 1 h of clinical based teaching in total. All students that received wound education reported undertaking teaching on the physiology of, and factors affecting wound healing, with only 32.2% (n = 104) of students receiving clinically based wound education There was very weak correlation and no significant association in student's ability to assess wounds (R2 = 0.190, p = 0.013), manage wounds (R2 = 0.060, p = 0.37), and prescribe wound care products (R2 = 0.093, p = 0.18) with their stage of training. Students strongly agreed that wound education is an important part of the undergraduate curriculum and post graduate practice, and do not feel their learning needs have been met. This is the first study to assess the provision of wound education in the United Kingdom, demonstrating a clear deficit in the provision of wound education compared to expectation of junior doctors. Wound education is largely overlooked in the medical curriculum, lacks a clinical focus and does not prepare junior doctors with the necessary clinical abilities to deal with wound related pathology. Expert opinion to direct changes to future curriculum and further evaluation of teaching methodology is required to address this deficit and ensure students have the necessary clinical skills to excel as newly graduated doctors. Background: Lower limb oedema is a common co-morbidity in those with diabetes and foot ulceration and is linked with increased amputation risk. There is no current guidance for the treatment of concurrent diabetic foot ulcers and lower limb oedema, leading to uncertainty around the safety and efficacy of combination approaches incorporating offloading and compression therapies. To determine indications and contraindications for such strategies and identify any other supplementary treatment approaches, a scoping review was undertaken to map the evidence relating to off-loading and compression therapy strategies to treat both diabetic foot ulcers and lower limb oedema in combination. Methods: Following the Joanna Briggs Institute (JBI) and PRISMA – Scoping Review (ScR) guidance, this review included published and unpublished literature from inception to April 2022. Literature was sourced using electronic databases including Cochrane Library, PubMed, CINAHL, AMED; websites; professional journals and reference lists of included literature. Eligible literature discussed the management of both diabetic foot ulceration and lower limb oedema and included at least one of the treatment strategies of interest. Data extraction involved recording any suggested off-loading, compression therapy or supplementary treatment strategies and any suggested indications, contraindications and cautions for their use. Results: Five hundred twenty-two publications were found relating to the management of diabetic foot ulcers with an off-loading strategy or the management of lower limb oedema with compression therapy. 51 publications were eligible for inclusion in the review. The majority of the excluded publications did not discuss the situation where diabetic foot ulceration and lower limb oedema present concurrently. Conclusions: Most literature, focused on oedema management with compression therapy to conclude that compression therapy should be avoided in the presence of severe peripheral arterial disease. Less literature was found regarding off-loading strategies, but it was recommended that knee-high devices should be used with caution when off-loading diabetic foot ulcers in those with lower limb oedema. Treatment options to manage both conditions concurrently was identified as a research gap. Integrated working between specialist healthcare teams, was the supplementary strategy most frequently recommended. In the absence of a definitive treatment solution, clinicians are encouraged to use clinical reasoning along with support from specialist peers to establish the best, individualised treatment approach for their patients. Ethnopharmacological relevance: Astragalus polysaccharide (APS), the most biologically active ingredient of Astragali Radix, is used to treat diabetes mellitus (DM)-related chronic wounds in traditional Chinese medicine for several decades. This herb possesses an anti-inflammatory effect. Our study proved that APS can reduce excessive inflammation at the late phase of wound-healing in diabetic ulcers. Aim of the study: To clarify the molecular mechanism of APS in promoting wound-healing via reducing excessive inflammation in diabetic ulcers during the late stages of wound-healing. Methods and materials: The rat model of the diabetic ulcers was established via intraperitoneal injection of streptozocin (60 mg/kg). We detected the regulation of APS on diabetic ulcers by measuring wound-healing rates. Bioinformatics was used to predict the target genes of APS, and autodocking was used to predict the combination of APS and target genes. Immunohistochemistry, Enzyme-linked immunosorbent assay, Western blot, immunofluorescence staining, flow cytometry, and flow cytometric sorting were investigated. Results: The results demonstrated that APS promoted wound-healing and inhibited excessive inflammation at the late phase of wound-healing in diabetic rats. Mechanistic findings showed that APS promoted the expression of β-catenin and Rspo3 while inhibiting the expression of NF-KB and GSK-3β, which leads to the transformation of M1-type macrophages into M2-type macrophages and thus reducing excessive inflammation at the late phase of wound-healing in diabetic ulcers. Conclusion: We found an interesting finding that APS promoted the polarization of macrophages towards M2-type through the β-catenin/NF-κB axis to reduce excessive inflammation at the late phase of wound-healing. Therefore, APS may be a promising drug for treating diabetic ulcers in clinic.

The aim of the research is to create a medical bandage utilizing an eco-friendly herb extract that is both antibacterial and wound-healing without the use of external dressing materials. The goal was accomplished by solvent extraction of the coriander herb (Coriandrum sativum). Ten layers of extracted solution were combined with one cotton bandage to create it using the pad-dry-cure technique. Following the same process, a second sample was created with a single layer using a ten-fold more concentrated coriander extraction solution. The K/S value measurement, SEM analysis, ATR analysis, and antimicrobial test were performed on treated materials. Implementations of the samples were made on a rabbit's wound site to demonstrate their ability to recover. The second sample's K/S value was remarkably similar to that of the first sample's tenth layer which was 0.89. The second sample's SEM picture showed that a significant amount of coriander extract had been deposited on the fibre surface. The presence of coriander extract's phytochemicals in the treated sample was confirmed by ATR analysis. Samples that had been padded with coriander extract had zones of inhibition (ZOI) against Grampositive and Gram-negative bacteria that were 14 mm and 12 mm, respectively, in terms of antibacterial activity. Healing of the wound showed good results when the bandages were removed after seven days. The wound had fully recovered. Environmentally friendly products are in high demand in the medical industry. Therefore, these modified samples are marketable for use as medical bandages.

Sedum telephium is a succulent plant used in traditional medicine, particularly in Italy, for its efficacy in treating localized inflammation such as burns, warts, and wounds. Fresh leaves or freshly obtained derivatives are directly applied to the injuries for these purposes. However, challenges such as the lack of microbiologically controlled materials and product standardization prompted the exploration of more controlled biotechnological alternatives, utilizing in vitro plant cell cultures of S. telephium. In the present study, we used HPLC-DAD analysis to reveal a characteristic flavonol profile in juices from in vivo leaves and in vitro materials mainly characterized by several kaempferol and quercetin derivatives. The leaf juice exhibited the highest content in total flavonol and kaempferol derivatives, whereas juice from callus grown in medium with hormones and callus suspensions showed elevated levels of quercetin derivatives. The in vitro anti-inflammatory and wound-healing assays evidenced the great potential of callus and suspension cultures in dampening inflammation and fostering wound closure, suggesting quercetin may have a pivotal role in biological activities.

Background: Expansion of OKF6/TERT-2 oral epithelial cells in vitro is important for studying the molecular biology of disease and pathology affecting the oral cavity. Keratinocyte serum-free medium (KSFM) is the medium of choice for this cell line. This study compares three media for OKF6/TERT-2 cultures: KSFM, Dulbecco’s Modified Eagle Medium/Nutrient Mixture of Hams F-12 (DMEM/F12), and a composite medium comprised of DMEM/F-12 and KSFM (1:1 v/v), referred to as DFK. The toxicological effects of electronic cigarette liquids (e-liquids) on OKF6/TERT-2 cells cultured in these media were also compared. Methods: Cells were cultured in KSFM, DMEM/F12, or DFK, and cellular morphology, growth, wound healing and the gene expression of mucins and tight junctions were evaluated. Additionally, cytotoxicity was determined after e-liquid exposures. Results: Switching from KSFM to DMEM/F12 or DFK 24 h post-seeding leads to typical cellular morphologies, and these cultures reach confluency faster than those in KSFM. Wound-healing recovery occurred fastest in DFK. Except for claudin-1, there is no difference in expression of the other genes tested. Additionally, e-liquid cytotoxicity appears to be amplified in DFK cultures. Conclusions: DMEM/F12 and DFK are alternative media for OKF6/TERT-2 cell culture to study the molecular biology of disease and pathology, provided cells are initially seeded in KSFM.

This study evaluated the wound-healing activity of a polymer, Poly(3,4-ethylenedioxythiophene):poly-(styrene sulfonate) (PEDOT: PSS), and determined its mechanism based on angiogenic activity in a full-thickness excision wound model in Spraque Dawley (SD) rats. Administering PEDOT: PSS (1.6) 1.5 ppm at a dose of 50 mg/kg/day significantly improved wound healing in the SD rats on the eleventh day after the incision was created. PEDOT: PSS-treated animals presented no anti-inflammatory skin effects; however, there was an increase in angiogenic behavior. VEGF was found to be significantly elevated in the PEDOT: PSS-treated groups seven days post-incision. However, only a higher concentration of PEDOT: PSS increased TGF-β1 expression within the same time frame. Our results showed that PEDOT: PSS enhances wound healing activity, mainly in terms of its angiogenic effects. In this paper, we describe the highly conductive macromolecular material PEDOT: PSS, which demonstrated accelerated wound-healing activity in the animal incision model. Thymbra spicata subsp. spicata L. is a member of the Lamiaceae family, which is used in Türkiye against various health issues among other uses. The main components of T. spicata essential oil (TE) were identified as carvacrol (52.3%) and p-cymene (21.1%) using GC-FID and GC/MS. The in vitro effects of TE on Miapaca-2 and HUVEC cell lines were reported for the first time. The initial results showed that TE applied to Miapaca-2 cell lines at concentrations of ≥62.18 g/mL for 48 hours could reduce cell growth and induce apoptosis. The application of relatively high concentrations of TE (≥82.91 g/mL) significantly (P<0.001) suppressed cell growth. Administration of relatively high TE concentrations (≥82.91 g/mL) significantly (P<0.001) suppressed cell growth. Concentrations of TE lower than 20.73 g/mL did not affect the viability of the HUVEC cell line in 48 hours. The IC50 value for Miapaca-2 cells was 62.18 µg/mL, and HUVEC cells' IC50 value was 263.97 µg/mL for 48h. The number of apoptotic cells in Miapaca-2 (55±5%) and HUVEC (38.33±6%) were significantly higher (P<0.001). Aim: Oxidative stress has been a significant factor in wound-healing pathophysiology for a long time. Antioxidants, especially natural compounds, have recently been emphasized in instructions for wound healing treatments. Jaceosidin (JACE), a flavone derived from Artemisia princeps, is a potent antioxidant. This study aims to investigate JACE's anti-inflammatory and antioxidant properties and its capacity to improve the effects of in vitro wound healing. Methods: Wound healing activities have been tested using cell proliferation and migration in vitro assays in the mouse fibroblast cell line L929. The concentration of hydrogen peroxide (H2O2-0.5 mM) has been used to induce the oxidative stress model. Tumor necrosis factor-alpha (TNF-α) and nuclear factor (NF-δβ) have been investigated as inflammatory indicators. Antioxidant activity has been checked using total antioxidant status (TAS) and total oxidant status (TOS) tests. Results: JACE has significantly increased the proliferation of fibroblasts dose-dependent manner. It has enhanced the cell migration rate of fibroblasts compared with the H2O2 group. JACE at a concentration of 50 and 100 µM has significantly decreased TOS and oxidative stress index (OSI) levels and increased TAS levels. The anti-inflammatory mechanism of JACE has involved down-regulation of the mRNA expressions of the NF-δβ and TNF-α in a dose-dependent manner. Conclusions: JACE has beneficial impacts on fibroblast viability and migration qualities through antioxidative actions and down-regulating proinflammatory cytokines through anti-inflammatory effects to promote wound healing. The development of biomaterial platforms for dispensing reagents of interest such as antioxidants, growth factors or antibiotics based on functional hydrogels represents a biotechnological solution for many challenges that the biomedicine field is facing. In this context, in situ dosing of therapeutic components for dermatological injuries such as diabetic foot ulcers is a relatively novel strategy to improve the wound healing process. Hydrogels have shown more comfort for the treatment of wounds due to their smooth surface and moisture, as well as their structural affinity with tissues in comparison to hyperbaric oxygen therapy, ultrasound, and electromagnetic therapies, negative pressure wound therapy or skin grafts. Macrophages, one of the most important cells of the innate immune system, have been described as the key not only in relation to the host immune defense, but also in the progress of wound healing. Macrophage dysfunction in chronic wounds of diabetic patients leads to a perpetuating inflammatory environment and impairs tissue repair. Modulating the macrophage phenotype from pro-inflammatory (M1) to anti-inflammatory (M2) could be a strategy for helping to improve chronic wound healing. In this regard, a new paradigm is found in the development of advanced biomaterials capable of inducing in situ macrophage polarization to offer an approach to wound care. Such an approach opens a new direction for the development of multifunctional materials in regenerative medicine. This paper surveys emerging hydrogel materials and bioactive compounds being investigated to induce the immunomodulation of macrophages. We propose four potential functional biomaterials for wound healing applications based on novel biomaterial/bioactive compound combination that are expected to show synergistic beneficial outcomes for the local differentiation of macrophages (M1–M2) as a therapeutic strategy for chronic wound healing improvement. Although inflammation is a vital defence response to infection, if left uncontrolled, it can lead to pathology. Macrophages are critical players both in driving the inflammatory response and in the subsequent events required for restoring tissue homeostasis. Extracellular vesicles (EVs) are membrane‐enclosed structures released by cells that mediate intercellular communication and are present in all biological fluids, including blood. Herein, we show that extracellular vesicles from plasma (pEVs) play a relevant role in the control of inflammation by counteracting PAMP‐induced macrophage activation. Indeed, pEV‐treatment of macrophages simultaneously with or prior to PAMP exposure reduced the secretion of pro‐inflammatory IL‐6 and TNF‐α and increased IL‐10 response. This anti‐inflammatory activity was associated with the promotion of tissue‐repair functions in macrophages, characterized by augmented efferocytosis and pro‐angiogenic capacity, and increased expression of VEGFa, CD300e, RGS2 and CD93, genes involved in cell growth and tissue remodelling. We also show that simultaneous stimulation of macrophages with a PAMP and pEVs promoted COX2 expression and CREB phosphorylation as well as the accumulation of higher concentrations of PGE2 in cell culture supernatants. Remarkably, the anti‐inflammatory activity of pEVs was abolished if cells were treated with a pharmacological inhibitor of COX2, indicating that pEV‐mediated induction of COX2 is critical for the pEV‐mediated inhibition of inflammation. Finally, we show that pEVs added to monocytes prior to their M‐CSF‐induced differentiation to macrophages increased efferocytosis and diminished pro‐inflammatory cytokine responses to PAMP stimulation. In conclusion, our results suggest that pEVs are endogenous homeostatic modulators of macrophages, activating the PGE2/CREB pathway, decreasing the production of inflammatory cytokines and promoting tissue repair functions. Context As the interest on the research of plant derived bioactive peptides (BPs) for nutraceutical, cosmeceutical and medical applications is increasing, in this work, the application of peptide derived from broccoli to keratinocytes was studied. Objective We focussed on the characterization of different peptides hydrolysates from broccoli stems [extracted from total protein (E) and from membrane protein (MF)], and their activity when applied to human keratinocytes. Materials and methods Peptide mixtures from broccoli stems (E and MF) were characterized by proteomics. They were applied to HaCaT cells in order to study cytotoxicity in a concentration range between 20 and 0.15625 µg of protein/mL and wound healing was studied after 24 and 48 h of treatment application. Also, proteomic and gene expression of keratinocytes were analysed. Results Depending on the source, proteins varied in peptide and amino acid composition. An increased proliferation of keratinocytes was shown after the application of the E peptides mixtures, reaching 190% with the lowest concentrations, but enhanced wound healing repair with E and MF appeared, reaching 59% of wound closure after 48 h. At the gene expression and protein levels of keratinocytes, the upregulation of anti-oncogene p53 and keratinization factors were observed. Discussion These results suggest that peptide mixtures obtained from broccoli augmented cell proliferation and prevented the carcinogenic, uncontrolled growth of the cells, with different mechanisms depending on the protein source. Conclusions The results encourage the opening of new lines of research involving the use of Brassica peptides for pharmaceutic or cosmetic use.

Abstract Background Induced pluripotent stem cells (iPSCs) have the potential to promote wound healing; however, their adhesion to the extracellular matrix (ECM) might decrease iPSC migration, thereby limiting their therapeutic potential. Integrin β1 (Itgb1) is the major integrin subunit that mediates iPSC-ECM adhesion, suggesting that knocking out Itgb1 might be an effective method for enhancing the therapeutic efficacy of iPSCs. Methods We knocked out Itgb1 in mouse iPSCs and evaluated its effects on the therapeutic potential of topically applied iPSCs, as well as their underlying in vivo and in vitro mechanisms. Results The Itgb1-knockout (Itgb1-KO) did not change iPSC pluripotency, function, or survival in the absence of embedding in an ECM gel but did accelerate wound healing, angiogenesis, blood perfusion, and survival in skin-wound lesions. However, embedding in an ECM gel inhibited the in vivo effects of wild-type iPSCs but not those of Itgb1-knockout iPSCs. Additionally, in vitro results showed that Itgb1-knockout decreased iPSC-ECM adhesion while increasing ECM-crossing migration. Moreover, ECM coating on the culture surface did not change cell survival, regardless of Itgb1 status; however, the in vivo and in vitro functions of both Itgb1-knockout and wild-type iPSCs were not affected by the presence of agarose gel, which does not contain integrin-binding sites. Knockout of Integrin α4 (Itga4) did not change the above-mentioned cellular and therapeutic functions of iPSCs. Conclusions Itgb1-knockout increased iPSCs migration and the wound-healing-promoting effect of topically applied iPSCs. These findings suggest the inhibition of Itgb1 expression is a possible strategy for increasing the efficacy of iPSC therapies.

Cord blood-platelet lysate (CB-PL), containing growth factors such as a platelet-derived growth factor, has a similar efficacy to peripheral blood-platelet lysate (PB-PL) in initiating cell growth and differentiation, which makes it a unique alternative to be implemented into oral ulceration healing. This research study aimed to compare the effectiveness of CB-PL and PB-PL in promoting oral wound closure in vitro. Alamar blue assay was used to determine the optimal concentration of CB-PL and PB-PL in enhancing the proliferation of human oral mucosal fibroblasts (HOMF). The percentage of wound closure was measured using the wound-healing assay for CB-PL and PB-PL at the optimal concentration of 1.25% and 0.3125%, respectively. The gene expressions of cell phenotypic makers (Col. I, Col. III, elastin and fibronectin) were determined via qRT-PCR. The concentrations of PDGF-BB were quantified using ELISA. We found that CB-PL was as effective as PB-PL in promoting wound-healing and both PL were more effective compared to the control (CTRL) group in accelerating the cell migration in the wound-healing assay. The gene expressions of Col. III and fibronectin were significantly higher in PB-PL compared to CB-PL. The PDGF-BB concentration of PB-PL was the highest and it decreased after the wound closed on day 3. Therefore, we concluded that PL from both sources can be a beneficial treatment for wound-healing, but PB-PL showed the most promising wound-healing properties in this study. [ABSTRACT FROM AUTHOR]

Carduus adpressus has been used for its anti-hair loss effect in traditional folk medicine. The plant species is mainly distributed in Türkiye, Bulgaria, and the Western Caucasus. The studies on this specific plant in the genus Carduus is limited which remarks the significance of the current study. This study aims to investigate the antibacterial, antioxidant, and wound-healing properties of the methanolic extract of Carduus adpressus. Extract was obtained by maceration. The broth microdilution assay was performed on Staphylococcus aureus, Enterococcus faecalis, Pseudomonas aeruginosa, and Escherichia coli. DPPH and ABTS radical scavenging assays were performed to detect the antioxidant capacity. Wound-healing activity was tested using alloxan-induced diabetic BALB-c mice. 4 groups, control, vehicle, CAE and Carduus adpressus groups were treated with the relative agent for 10 days. Extract demonstrated 62.5 μg/mL MIC against S. aureus and E. faecalis, and 125 μg/mL MIC against P. aeruginosa and E. coli. ABTS assay showed higher antioxidant activity compared to the DPPH assay. Carduus adpressus group demonstrated strong regeneration, epithelisation, and angiogenesis compared to the control group on day 10. Additionally, expression of PDGF, VEGF, and collagen formation was increased in the Carduus adpressus group compared to the control group on day 10. Extract demonstrated strong antibacterial, antioxidant, and wound-healing activities which indicate that it could be a source in developing wound-healing agents. [ABSTRACT FROM AUTHOR]

The process of delayed wound-healing and unhealed wounds can cause serious problems to the patients. In this context it is revealed that the scientists are looking for herbal treatment of wounds, as the remedy is known for having less or minimum undesired effects. Schima wallichii is a lesser known traditional medicinal plant used by certain section of people in Assam, India as a healing agent; however, record on scientific validation of its wound-healing potential is not available. Therefore, an in vivo study was undertaken to evaluate the wound-healing efficacy of the leaves of the plant using excision wound model in Swiss albino mice. The animals were divided into three groups after creating the wounds, namely control, positive control and treated groups. On application of methanolic crude extract of the leaves of Schima wallichii, it was observed that the epithelialization period was lesser and the rate of closure of wounds was faster in the extract treated group when compared with the control group indicating that the application of the extract has accelerated the healing process in the extract treated group. The concentration of protein, Schima wallichii deoxyribonucleic acid and hydroxyproline were also found to be augmented in the treated group than the control suggesting faster rate of healing as deoxyribonucleic acid and protein content in the healing tissues serve as indicator of protein synthesis and cellular proliferation. The above results were supported by the histological and ultra-structural observations as scanning electron microscopy revealed smoother surface topography, H and E staining revealed thicker epithelial layer in the treated animals. Van Gieson's staining showed higher level of collagen in the extract treated group validating biochemical observation where higher concentration of hydroxyproline was found in treated group compared to the control. Amphiphilic polysaccharides can be used as wall materials and applied to encapsulate hydrophobic active chemicals; moreover, there is significant demand for novel medical high-molecular-weight materials with various functions. In order to prepare amphiphilic schizophyllan (SPG), octenyl succinic anhydride (OSA) was chosen to synthesize OSA-modified schizophyllan (OSSPG) using an esterified reaction. The modification of OSSPG was demonstrated through FT-IR and thermal analysis. Moreover, it was found that OSSPG has a better capacity for loading curcumin, and the loading amount was 20 μg/mg, which was 2.6 times higher than that of SPG. In addition, a hydrogel made up of PVA, borax, and C-OSSPG (OSSPG loaded with curcumin) was prepared by means of the one-pot method, based on the biological effects of curcumin and the immune-activating properties of SPG. The mechanical properties and biological activity of the hydrogel were investigated. The experimental results show that the dynamic cross-linking of PVA and borax provided the C-OSSPG/BP hydrogel dressing with exceptional self-healing properties, and it was discovered that the C-OSSPG content increased the hydrogel's swelling and moisturizing properties. In fibroblast cell tests, the cells treated with hydrogel had survival rates of 80% or above. Furthermore, a hydrogel containing C-OSSPG could effectively promote cell migration. Due to the excellent anti-inflammatory properties of curcumin, the hydrogel also significantly reduces the generation of inflammatory factors, such as TNF-α and IL-6, and thus has a potential application as a wound dressing medicinal material. Lymphedema causes tissue swelling due to the accumulation of lymphatic fluid in the tissue, which delays the process of wound-healing. Developing effective treatment options of lymphedema is still an urgent issue. In this study, we aim to fabricate tissue-engineered moist wound dressings with adipose stem cells (ASCs) and decellularized Wharton's jelly (dWJ) from the human umbilical cord in order to ameliorate lymphedema. Rat ASCs were proliferated and an apparent layer was observed on dWJ at day 7 and 14. A rat tail lymphedema model was developed to evaluate the efficacy of the treatment. Approximately 1 cm of skin near the base of the rat tail was circularly excised. The wounds were treated by secondary healing (control) (n = 5), decellularized Wharton's jelly (n = 5) and ASC-seeded dWJ (n = 5). The wound-healing rate and the tail volume were recorded once a week from week one to week five. Angiogenesis and lymphangiogenesis were assessed by immunochemistry staining with anti-CD31 and anti-LYVE1. The results showed that the wound-healing rate was faster and the tail volume was lesser in the ASC-seeded dWJ group than in the control group. More CD31+ and LYVE-1+ cells were observed at the wound-healing area in the ASC-seeded dWJ group than in the control group. This proves that tissue-engineered moist wound dressings can accelerate wound-healing and reduce lymphedema by promoting angiogenesis and lymphangiogenesis. [ABSTRACT FROM AUTHOR]

The term moonlighting proteins refers to those proteins that present alternative functions performed by a single polypeptide chain acquired throughout evolution (called canonical and moonlighting, respectively). Over 78% of moonlighting proteins are involved in human diseases, 48% are targeted by current drugs, and over 25% of them are involved in the virulence of pathogenic microorganisms. These facts encouraged us to study the link between the functions of moonlighting proteins and disease. We found a large number of moonlighting functions activated by pathological conditions that are highly involved in disease development and progression. The factors that activate some moonlighting functions take place only in pathological conditions, such as specific cellular translocations or changes in protein structure. Some moonlighting functions are involved in disease promotion while others are involved in curbing it. The disease-impairing moonlighting functions attempt to restore the homeostasis, or to reduce the damage linked to the imbalance caused by the disease. The disease-promoting moonlighting functions primarily involve the immune system, mesenchyme cross-talk, or excessive tissue proliferation. We often find moonlighting functions linked to the canonical function in a pathological context. Moonlighting functions are especially coordinated in inflammation and cancer. Wound healing and epithelial to mesenchymal transition are very representative. They involve multiple moonlighting proteins with a different role in each phase of the process, contributing to the current-phase phenotype or promoting a phase switch, mitigating the damage or intensifying the remodeling. All of this implies a new level of complexity in the study of pathology genesis, progression, and treatment. Wound healing continues to pose a challenge in clinical settings. Moreover, wound management must be performed properly and efficiently. Acute wound healing involves multiple cell divisions, a new extracellular matrix, and the process of formation, such as growth factors and cytokines, which are released at the site of the wound to regulate the process. Any changes that disrupt the healing process could cause tissue damage and prolong the healing process. Various factors, such as microbial infection, oxidation, and inflammation, can delay wound healing. In order to counter these problems, utilizing natural products with wound-healing effects has been reported to promote this process. Several natural products have been associated with wound healing, most of which are from medicinal plants. However, secondary microbial metabolites have not been extensively studied for their wound-healing properties. Further, investigations on the wound-healing control of natural microbial products are required due to a lack of studies. This review discussed the in vivo and in vitro research on the wound healing activities of natural microbial products, which may assist in the development of better wound treatments in the future. Introduction: Polysaccharide and alcohol extracts of Anoectochilus formosanus Hayata have attracted great attention as they exhibit noteworthy properties such as prebiotic and anti-hyperglycemic effects. However, the antioxidant and wound-healing activities of the polysaccharide extract as well as the antibacterial and cytotoxic effects of the ethanol extracts have not been thoroughly uncovered. Therefore, our study investigated these bioactivities of the two extracts prepared from Anoectochilus formosanus to broaden understandings of medical benefits of the plant. Methods: The monosaccharide composition was analyzed by HPAEC-PAD. The antioxidant and wound-healing activities of the polysaccharide extract were evaluated by ABTS and scratch assays, respectively. The broth dilution method was used to determine the antibacterial ability of the ethanol extract. Additionally, the cytotoxic and mechanistic effects of this extract against hepatocellular carcinoma HUH-7 cells was assessed by MTT assay, qRT-PCR and Western blotting methods. Results: The polysaccharide extract possessed an effective free radical scavenging ability in an ABTS assay (IC50 = 44.92 μg/ml). The extract also ameliorated wound recovery in a fibroblast scratch assay. Meanwhile, the ethanol extract was able to inhibit the growth of Staphylococcus aureus (MIC = 2500 μg/ml), Bacillus cereus (MIC = 2500 μg/ml), Escherichia coli (MIC = 2500 μg/ml), and Pseudomonas aeruginosa (MIC = 1250 μg/ml). Additionally, it repressed the viability of HUH-7 cells (IC50 = 53.44 μg/ml), possibly through upregulating the expression of caspase 3 (CASP3), CASP8, and CASP9 at both mRNA and protein levels. Conclusion: The polysaccharide extract of A. formosanus exhibited the antioxidant and wound-healing properties whereas the ethanol extract showed the antibacterial activity and cytotoxicity against HUH-7 cells. These findings specify notable biological effects of the two extracts which could be of potential use in human healthcare.

Abstract Background Lower limb oedema is a common co‐morbidity in those with diabetes and foot ulceration and is linked with increased amputation risk. There is no current guidance for the treatment of concurrent diabetic foot ulcers and lower limb oedema, leading to uncertainty around the safety and efficacy of combination approaches incorporating offloading and compression therapies. To determine indications and contraindications for such strategies and identify any other supplementary treatment approaches, a scoping review was undertaken to map the evidence relating to off‐loading and compression therapy strategies to treat both diabetic foot ulcers and lower limb oedema in combination. Methods Following the Joanna Briggs Institute (JBI) and PRISMA – Scoping Review (ScR) guidance, this review included published and unpublished literature from inception to April 2022. Literature was sourced using electronic databases including Cochrane Library, PubMed, CINAHL, AMED; websites; professional journals and reference lists of included literature. Eligible literature discussed the management of both diabetic foot ulceration and lower limb oedema and included at least one of the treatment strategies of interest. Data extraction involved recording any suggested off‐loading, compression therapy or supplementary treatment strategies and any suggested indications, contraindications and cautions for their use. Results Five hundred twenty‐two publications were found relating to the management of diabetic foot ulcers with an off‐loading strategy or the management of lower limb oedema with compression therapy. 51 publications were eligible for inclusion in the review. The majority of the excluded publications did not discuss the situation where diabetic foot ulceration and lower limb oedema present concurrently. Conclusions Most literature, focused on oedema management with compression therapy to conclude that compression therapy should be avoided in the presence of severe peripheral arterial disease. Less literature was found regarding off‐loading strategies, but it was recommended that knee‐high devices should be used with caution when off‐loading diabetic foot ulcers in those with lower limb oedema. Treatment options to manage both conditions concurrently was identified as a research gap. Integrated working between specialist healthcare teams, was the supplementary strategy most frequently recommended. In the absence of a definitive treatment solution, clinicians are encouraged to use clinical reasoning along with support from specialist peers to establish the best, individualised treatment approach for their patients. Context: As the interest on the research of plant derived bioactive peptides (BPs) for nutraceutical, cosmeceutical and medical applications is increasing, in this work, the application of peptide derived from broccoli to keratinocytes was studied. Objective: We focussed on the characterization of different peptides hydrolysates from broccoli stems [extracted from total protein (E) and from membrane protein (MF)], and their activity when applied to human keratinocytes. Materials and methods: Peptide mixtures from broccoli stems (E and MF) were characterized by proteomics. They were applied to HaCaT cells in order to study cytotoxicity in a concentration range between 20 and 0.15625 mg of protein/mL and wound healing was studied after 24 and 48 h of treatment application. Also, proteomic and gene expression of keratinocytes were analysed. Results: Depending on the source, proteins varied in peptide and amino acid composition. An increased proliferation of keratinocytes was shown after the application of the E peptides mixtures, reaching 190% with the lowest concentrations, but enhanced wound healing repair with E and MF appeared, reaching 59% of wound closure after 48 h. At the gene expression and protein levels of keratinocytes, the upregulation of anti-oncogene p53 and keratinization factors were observed. Discussion: These results suggest that peptide mixtures obtained from broccoli augmented cell proliferation and prevented the carcinogenic, uncontrolled growth of the cells, with different mechanisms depending on the protein source. Chitosan-based hydrogels as wound dressings are expected to improve the efficiency of the wound-healing process. Fabrication of the composite structure of Aloe vera and biopolymeric hydrogels has been demonstrated to promote the wound-healing process through protection against a wide spectrum of microbes, and enhanced cell adhesion and differentiation. Therefore, the present study investigates to development of improved CHO/Aloe hydrogel for improving the wound-healing process in an animal model. CHO hydrogel with Aloe was prepared, and its properties were evaluated in terms of viscosity, antibacterial activity, and ints In-vivo wound-healing efficiency in full-thickness wounds of rat models. Physical examination of wound-healing efficiency of CHO/Aleo hydrogel was evaluated by analyzing total wound closure, recovery percentage, and the epiderm thickness of wounds. Histological evaluation was performed using hematoxylin and eosin staining to evaluate the re-epithelialization, inflammatory response, granulation tissue formation, and fibrotic tissue formation. The results showed a significantly higher wound-healing rate of the CHO/Aleo group compared to other groups at 3,7,14 days (p < 0.05). After 14 days of treatment, the best healing effect was observed in the CHO/Aleo gel with the highest tissue tension compared with other groups (p < 0.05). Histological findings indicated a significant difference in inflammatory response between control and treatment groups after three days of treatment (p < 0.05). Epidermal thickness was also significantly thicker in the CHO/Aleo gel group than others (p < 0.05).
