The study explored the post-wildfire elemental composition of parts (wood, bark, branch, cone, trunk, litter, twig, needle, sward, fallow, sapling, etc.) and by-products (biomass ashes, partly burnt parts, and char) of different woody species in the Bohemian Switzerland National Park, Czech Republic, and considered their effects on soils. Multi-elemental analysis of the fire by-products of the woody species was determined with inductively coupled plasma-optical emission spectrometry and mass spectrometry and compared with control biomass samples unaffected by wildfire. Most fire by-products were slightly alkaline, with acidic ashes obtained from blueberry wood. The by-products of the wildfire were characterized by varied total contents of macro (P, Ca, K, Mg, and S), micro (Na, Mn, Fe, Cu, and Zn), and other elements (B, Co, Mo, and V) vital to soil fertility and plant growth. The mean content of macro elements in the biomass ashes was up to 4.16 P, 23.5 Ca, 2.48 Mg, 63 K, and 5.57 S g kg−1. These values were comparatively lower than published data for ashes obtained under optimized conditions, e.g., those combusted in power generation facilities. Conversely, partly burnt parts—an indication of incomplete combustion—had higher 9.22 P, 79 Ca, and 5.99 Mg g kg−1 contents in spruce needles than in biomass ashes and the control. Variations in woody species and anthropogenic activities in areas of wildfires produced varied As, Cd, Cr, Ni, and Pb contents above EU fertilizer regulation. Caution in applying biomass ashes from wildfires on fields is required due to risk/toxic elements input from anthropogenic activities. Various woody species are growing in diverse landscapes and form a prominent component of farmland in Ethiopia. Farmers' preferences for woody species also vary in different parts of the country. The diversity and farmers' preferences for woody species have been described earlier. However, a simultaneous analysis of land use diversity and farmers' preferences for woody species is scarcely available. Therefore, to fill the gap, this study was conducted in three selected highlands of Ethiopia to assess the land use diversity and farmers' preferences for woody species. A total of 320 households were randomly selected, and out of these, 105 households were purposely selected for species inventory. Overall, 525 plots were made using a 30 m × 30 m plot size for different land uses. Focus group discussion and key informant interviews were used for farmers' preferences. The results imply that 32 woody species belonging to 16 families were recognized. Fabaceae was the leading family with 25% of species, followed by Rosaceae with 12.5% of species. In terms of species diversity, home garden (2.39) was more diverse than grazing land (1.94), crop land (1.50), boundary (0.92), and woodlot (0.54) in all areas. Among the preferred species, Eucalyptus globulus, Cuppressus lusitanica, Malus domestica, Cytisus proliferus, and Acacia decurrens were the most preferred and planted woody species by farmers. Generally, farmland maintains various tree and shrub species according to landscape practices. Farmers preferred and planted different woody species based on their benefits. This study assesses the diversity, composition, structure and abundance of urban forests in Adama city, Central Ethiopia to provide baseline information useful for developing a conservation strategy. The urban forest of Adama has an estimated 525,200 trees with a tree cover of 20%. The three most common species are Azadirachta Indica (14.8%), Carica papaya (6.8%), and Acacia abyssinica (5%). The i-Tree Eco model was used to organise output. A total of 214 sample plots were generated directly in the i-Tree Eco application using the random plots' generator via the Google Maps function. A 0.1acre circular plot was used. A total of 805 trees were sampled across the 214 plots. Among the 86 species encountered in the sample plots, 68 (79%) were introduced and only 18 (21%) were native species. Most introduced trees originated from Asia (22%). The tree diversity calculated using the Shannon Weiner index (H') was 3.61 and the dominance of Simpson's value was 0.95. The evenness index of the plots sampled was 0.80. The study indicates the crucial role that urban forests play in maintaining and conserving urban trees and enhancing ecosystem services in urban areas. [ABSTRACT FROM AUTHOR]

Somatic embryogenesis (SE) is a process by which somatic cells reprogram, acquire totipotency and embark on embryo formation. Although SE is a valuable tool for micropropagation in many crops, it presents specific challenges in woody species due to some bottlenecks, such as loss of embryogenic competence during subcultures and the often-low somatic embryo conversion into plantlets. Hence, great interest exists in exploring the regulatory networks involved on SE. As transcription modulators, long non-coding RNAs (lncRNAs) have been found essential in a wide range of biological processes. This work aimed to identify lncRNAs related to the embryogenic competence in Solanum betaceum Cav. (tamarillo). Nanopore® long-read sequencing was conducted in cell lines with distinct cell fates and, based on their coding potential, 60 transcripts were selected as lncRNA candidates. Similar expression patterns were found among embryogenic cell lines (EC) and cells that lost their embryogenic potential (herein long-term callus, LTC), whereas non-EC (NEC) showed differentially expressed lncRNAs. Whereas lncRNAs upregulated in EC and LTC were predicted to target embryogenesis-related genes, such as AGAMOUS-like 15 and WUSCHEL-related HOMEOBOX 2, lncRNAs upregulated in NEC were predicted to target mainly genes involved in carbohydrate metabolism, cell wall formation, auxin and ethylene signalling pathways. Altogether, these results show the involvement of lncRNA in the process of somatic embryogenesis in S. betaceum, indicating that EC present a pattern of lncRNA expression that suggests its action on genes that directly modulate the morphogenic response in vitro, while in NEC the modulation through this type of RNAs seems to be predominantly reflected in genes more related to cellular physiology.Key message: LncRNAs upregulated in EC/LTC were predicted to target embryogenesis-related genes, while lncRNAs upregulated in NEC were predicted to target genes involved in auxin and ethylene signalling and carbohydrate metabolism. Floral traits represent a valuable yet underutilized resource for functional ecology. We aimed to examine the variability of six quantitative floral traits (carbon [C] and nitrogen [N] contents, C:N ratio, flower length and width, and dry biomass) among a randomly selected group of 79 temperate woody species at both interspecific and intraspecimen levels. We hypothesized that (1) flower traits are closely related to the evolutionary history of the species, resulting in a strong phylogenetic signal, and (2) flowers collected from the most and the least exposed to sunlight parts of the crown would not differ in trait values. We detected statistically significant phylogenetic signals for all six floral traits under study. We found significant differences between samples from the two light variants only for flower N content and C:N ratio. Given the substantial interspecific and the negligible intra-specimen variability observed, we are convinced that the incorporation of these studied traits, especially those related to flower size (i.e., dry biomass, length, and width), into research on flowering biology and ecology can significantly enrich our comprehension of the plant reproductive processes.

Rangelands are important for mitigating climate change and rehabilitating land by storing carbon from woody plants and seeds in the soil. However, there is a lack of empirical data on how traditional rangeland management practices (such as enclosure, communal grazing and prescribed fire) affect the carbon storage of woody species and the conservation potential of soil seed banks. This study evaluates the carbon sequestration and soil seed bank conservation potential of traditional rangeland management practices in the Western Guji zone, Ethiopia. A total of 32 sample plots were established using systematic random sampling, with 12 in each management practice, to measure woody biomass and estimate carbon stock using a nondestructive method. Additionally, 72 soil samples were collected for soil seed bank analysis, with 24 samples taken from each type of rangeland management practice. The study found that enclosures had the highest amount of carbon stocks 63.1 tons per hectare (t ha−1), with a corresponding carbon dioxide equivalent (CO2e) sequestration of 231.58 t ha-1, followed by communal grazing areas that stored 27.05 t ha−1 of carbon with a CO2e sequestration of 99.27 t ha−1. Prescribed fire had the lowest carbon stocks at 19.39 t ha−1, with a CO2e sequestration of 71.16 t ha−1. The study also found that different traditional rangeland management practices significantly influenced the diversity, richness, and density of woody species in soil seed banks. The highest Shannon diversity was recorded in enclosures (3.56 ± 0.09), followed by communal grazing (2.87 ± 0.14) and prescribed fire (1.27 ± 0.08). As a result, enclosure promotes the storage of carbon in woody plants and increases the diversity, density, and richness of woody species seeds in the soil, while prescribed fire reduces it. Therefore, it recommend for pastoralists to expand the use of enclosures as it improves the potential for conserving carbon stocks and soil seed banks of woody species.

In developing countries, firewood serves as a primary energy source and plays a role in cultural-religious rituals and festivities. However, the specific patterns of woody species used for the latter remain poorly understood, including the impact of access restrictions to resources and local bans on practices. Therefore, our research focuses on examining how access restrictions to forest resources and bonfire bans due to the coronavirus disease 2019 (COVID-19) impact the cultural-religious tradition of bonfire making during Festas Juninas (June festivities) in northeastern Brazil. Methods Ethnobotanical fieldwork was conducted in two rural populations in northeastern Brazil between 2021 and 2022. Data were collected through semi-structured interviews, observations, and the guided tour technique. In the Shapha sub-watershed, an exclosure was implemented to restore the land and promote vegetation diversity. However, further scientific evidences are necessary to assess the impact of this intervention on woody species diversity and inform sustainable practices. Therefore, this study was designed to i) examine the diversity of woody species, population structure, and regeneration status compared with open grazing land and ii) assess factors affecting exclosure sustainability. The woody species inventory was made in 60 nested quadrants (20 m × 20 m) with eight parallel lines. Systematically, 90 respondents were asked and a focus group discussion was held. Data were analyzed using diversity indices, descriptive statistics, and a logistic regression model. The results reveal that 34 woody species were recorded across the two land-use types. As a result, 32 woody species belonging to 22 families and 29 genera within the exclosure and 11 types of woody plants from eight different families, comprising 10 other genera, were observed in the adjacent grazing area. It is hereby reported that the Shannon diversity indices for exclosure and adjacent open grazing lands were found to be 2.27 and 1.02, respectively. Similarly, the Simpson indices were 0.85 and 0.57, the evenness indices were 0.87 and 0.85, and the species richness values were 0.43 and 3.4, respectively. In the exclosure, woody species displayed a reverse J-shaped pattern in contrast to the J-shaped arrangement observed on the adjacent grazing land. Seven of the nine explanatory variables were crucial for determining the sustainability of the exclosure, with the exception of land tenure and family size. Notably, exclosure has proven to be the most effective for restoring degraded vegetation in the study area because they encourage the growth of new vegetation through natural regeneration.

In addition, as populations remain deeply entrenched in cultural-religious practices amid temporary bans imposed by health crises, there is a pressing need for culturally sensitive environmental policies. The Convention on Biological Diversity seeks to conserve at least 30% of global land and water areas by 2030, which is a challenge but also an opportunity to better preserve biodiversity, including flowering plants (angiosperms). Herein, we compiled a large database on distributions of over 300,000 angiosperm species and the key functional traits of 67,024 species. Using this database, we constructed biodiversity-environment models to predict global patterns of taxonomic, phylogenetic, and functional diversity in terrestrial angiosperms and provide a comprehensive mapping of the three diversity facets. We further evaluated the current protection status of the biodiversity centers of these diversity facets. Our results showed that geographical patterns of the three facets of plant diversity exhibited substantial spatial mismatches and nonoverlapping conservation priorities. Idiosyncratic centers of functional diversity, particularly of herbaceous species, were primarily distributed in temperate regions and under weaker protection compared with other biodiversity centers of taxonomic and phylogenetic facets. Our global assessment of multifaceted biodiversity patterns and centers highlights the insufficiency and unbalanced conservation among the three diversity facets and the two growth forms (woody vs. herbaceous), thus providing directions for guiding the future conservation of global plant diversity. Ethiopia's diverse ecology faces challenges from significant deforestation. Church forests are relatively protected although under pressure from agricultural expansion, grave house construction and climate related factors. The present study was aimed to assess the woody species composition, structure and regeneration status of church forests in the West Gojjam Zone. Twenty-six church forests were selected using stratified random sampling based on agroecology, elevation, forest size, and distance from population centers. A total of 175 main plots of 20 m × 20 m (400 m2) for trees, 875 subplots of 5 m × 25 m (25 m2) for saplings, and 875 subplots of 1 m × 21 m (1 m2) for seedlings were laid down. Diameter at breast height (DBH) was measured for woody plants with a DBH ≥2.5 cm and also height ≥ 2.5 m. Diversity and structural analysis of the woody species were carried out using appropriate tools. The regeneration status of the study forests was evaluated based on the number of seedlings, sapling and matured plants. A total of 111 woody species that belong to 95 genera and 51 families with 72 (64.86 %) trees, 23 (20.72 %) shrubs, 8 (7.2 %) shrubs/trees and 8 (7.2) lianas were identified. In the study sites, the woody plant species consist of 82.88 % indigenous, 15.32 % exotic, and 1.8 % endemic species. Fabaceae was the dominant family in West Gojjam zone church forests. The Shannon diversity ranged from 0.99 to 3.47, with an average of 2.55, and evenness varied from 0.5 to 0.96 (average 0.82). Species richness ranged from 7 to 45 with the average value of 23.81, and a total stem density of 5880 individuals per hectare. Additionally, 5923 saplings and 6136 seedlings per hectare were recorded. The most abundant species include Eucalyptus camaldulensis, Calpurnia aurea, and Juniperus procera. The average basal area was 29.268 m²/ha for the studied church forests, with Ficus vasta having the highest basal area. Juniperus procera and Rhus vulgaris exhibited the highest Importance Value index (IVI), while Premna schimperi, and Myrtus communis had the lowest IVI. The overall regeneration status of west Gojjam church forests was found to be good. Significant differences in woody species composition and diversity were observed between church forests. The study revealed the importance of church forests in the West Gojjam Zone for plant diversity conservation, revealing their resilience and potential for sustainable protection of forests despite the challenges like deforestation and climate change, and their potential for sustainable maintenance. As we have already seen, some of the church's forests were converted to agriculture as a source of income for the church. This practice is dangerous for the survival of the endemic plants in the church forests and leads to deforestation. Therefore, it is recommended to form an alternative source of income for the church.

Maintaining forest biodiversity and carbon sequestration potential of forest resources enhances the provision capacity of forest ecosystem services. Experience from around the world and in Ethiopia has shown that shifting forest management from state-centred to community-centred arrangement can result in a better forest stock. Therefore, this study examined the status of woody species diversity, regeneration and total living biomass carbon stock of forests under participatory forest management (PFM) and the adjacent state managed non-participatory forest management (Non-PFM) in South eastern Ethiopia and implications to REDD+. Data were collected from 89 (44 PFM and 45 Non-PFM) nested circular plots from four PFM and three Non-PFM selected forest sites with transects laid systematically. Tree DBH and height were measured, the number of saplings, seedlings, mature trees were counted and species names were recorded. Woody species diversity was estimated using shannon, simpson, and evenness diversity indices. A total of 29 and 23 woody species were recorded in PFM and Non-PFM forests, respectively. Woody species diversity did not show significant difference between PFM and Non-PFM forests but it was relatively higher in PFM forest. The density per hectare of seedlings, sapling and mature trees were significantly greater in PFM forest than in Non-PFM forest. The mean aboveground biomass carbon stock of PFM forest (225.50±26.54) was significantly greater than that of the Non-PFM (156.24±15.72) forest. Hence, managing forests through participatory approaches contributes to the enhancement of sustainable management and climate change mitigation potentials through reducing emission from deforestation and forest degradation.

The present survey was conducted to investigate the ecological distribution range of woody species and the most important factors affecting their distribution in the Zagros forests of Ilam province. physical and chemical characteristics of soil, as well as topographical factors, were assessed in 109 circular sampling plots with an area of 500 m² n four forest areas with an altitude range of 800-2400 m. The vertical distance of the plots on the slope aspect was 100 m, and the distance of the sampling routes varied from 500-1000 m. ecological distribution range index was calculated based on the most important factors affecting species distribution. Based on the Principal Component Analysis (PCA), the most significant factors include: the percentage of theclay, Potassium, nitrogen, carbon, sand, phosphorus, and altitude. species such as Amygdalus orientalis, Lonicera nummularifolia, Daphne mucronata and Cerasus mahaleb and Acer monspessulsnum were found at hight altitudes with the highest levels of carbon, nitrogen, potassium and phosphorus. In addition, Quercus brantii along with Acer monspessulanum and Daphne mucronata showed the highest ecological distribution range while Pistacia atlantica, Amygdalus arabica and Amygdalus orientalis species respectively has the lowest values. Aims: Within root systems, framework pioneer roots mainly function in soil exploration and resource transport, while fibrous roots and their associated fungi extend the foraging soil space for resource absorption. However, the plasticity of pioneer and fibrous roots in response to elevated nutrient availability has received relatively little attention. Methods: For three hardwood species, newly formed root branches were harvested in order to compare pioneer and fibrous roots for variation in morphology, mycorrhizal colonization and anatomy, as well as their trait shifts in response to fertilization. Results: Compared with the fibrous roots, pioneer roots were generally larger in diameter, stele and cortex size, root tissue density, and hydraulic conductivity, but lower in specific root length (SRL) and mycorrhizal colonization rate (MCR). Fertilization generally decreased SRL and MCR in fibrous roots, and increased conduit diameters in pioneer roots. Additionally, fertilization enhanced hydraulic conductivity for all three species: 162, 45 and 263% in pioneer roots, and 46, 13 and 172% in fibrous roots for Juglans mandshurica Maxim., Fraxinus mandshurica Rupr. and Phellodendron amurense Rupr., respectively. Conclusions: Pioneer and fibrous roots responded differently to fertilization. The resulting enhanced resource transportation capacity may serve to mitigate the toxic effect in fibrous roots and avoid pioneer roots becoming a bottleneck for absorbed water and nutrient in an expanding root system, suggesting tightly associated traits and functional equilibrium within an individual root and even the root branches. The observed trait shifts and coordination between pioneer and fibrous roots would facilitate resource acquisition in changing soil conditions. [ABSTRACT FROM AUTHOR]

The benefits of green areas mostly depend on the trees' dimension, which in the urban environment is not as predictable as it is in woodland. Growth models based on tree height and tree diameter at breast height have been largely employed to predict tree growth also in urbanised contexts. However, city environments differ from natural ones, and in addition to geographical influence, they present high variability in growth conditions within a few metres. Different urban conditions may influence tree vertical and diametral growth. The present study aims to jointly consider the effect of urban environment, maintenance practices and available space on the size growth of some tree species. Dendrometrical, space, management and status parameters of the sampled trees were collected during field surveys 14 years apart. Tree height and diameter at breast height data interpolations were realised and charted by an open-source graphing calculator to build three different growth equations (linear, logarithmic and logistic) for each of the five species considered regardless of tree growth conditions. The comparison between the comprehensive best-fitting models realised within this study and a USA inventory-based study highlighted quite similar tree growth trends, probably due to the consideration of both constrained and unconstrained trees. Furthermore, a study on the curves of same-growth condition trees of the same species was conducted. Generally, heavy pruning techniques such as control and pollard pruning, characterised by the removal of a consistent part of the tree crown, stand out as the main growth-limiting factor for the different species. Little information is available about the vegetation impact on soil, comparing the effects of different woody plant species on topsoil properties in subalpine degraded grasslands. Therefore, we studied the effects of three native woody plants with different structural canopies (evergreen needle-leaved vs. deciduous broad-leaved and cushion vs. free canopy) on the nutrient topsoil properties. We selected 20 sites as 20 replications in which all four woody species were found closed to each other in each site. Then soil samples were collected under the canopy of each woody species in June 2019 and transported to the laboratory for physico-chemical analyses. The results showed that generally, woody species differed in their effects on physico-chemical topsoil properties. So that, the needle-leaved evergreen Juniperus sabina exerted the most influence on soil properties compared to Berberis integerrima and Onobrychis cornuta (p < 0.05). Topsoil total organic carbon content was significantly highest under J. sabina and B. integerrima (3.30% and 3.07% respectively), while lowest under O. cornuta and herbaceous vegetation (2.61% and 2.23%, respectively). In addition, soil total nitrogen content was significantly highest under J. sabina and B. integerrima (0.18% and 0.17%, respectively), while lowest under O. cornuta and herbaceous vegetation (0.16% and 0.15%, respectively). The results of this study indicated that in sub-alpine grasslands, the presence and maintaining of woody species is important due to its positive role on substratum soil, considering in the rangeland improvement projects. Climatic change related extreme events such as droughts negatively affect local communities in the semi-arid savanna ecosystems. This study mainly records and analyses local knowledge on the use of edible indigenous woody plant species by local communities during drought periods, as a coping strategy, in southeast lowveld of Zimbabwe. Secondary data on utilization of edible indigenous woody plants were gathered from literature sources focusing mainly on the southeast lowveld parts of Zimbabwe and covering the period 2000–2019. Quantitative ethnobotanical data analysis involved computing the frequency of citation (FC), relative frequency of citation (RFC) and family importance value (FIV) to determine the local significance of indigenous woody plant species. A total of 23 species from 12 families were recorded as being used during drought periods with key species including baobab (Adansonia digitata), bird plum (Tamarindus indica), corky-monkey orange (Strychnos cocculoides) and black monkey orange (Strychnos madagascariensis). Major use categories were food, medicine, and livestock feed. The study findings points to the need for embracing indigenous woody plants as a buffer against drought in semi-arid parts of the savanna. Future projects should focus on developing innovative strategies such as value addition and promoting sustainable use and restoration of non-wood forest products as part of livelihood diversification under drought situations.

Biodiversity in agroforestry systems contributes to diversified socio-economic and cultural dimensions and to managing biodiversity. Agroforestry is one of Ethiopia's highest conservation priorities to promote forest-based ecosystem services and environmental incentive programs that minimize deforestation. This study was carried out across intensively cultivated areas of Midakegn district to assess the woody species diversity potential of agroforestry systems managed by farmers. Out of 114 plots laid down on transect lines, 33 (30 × 30 m), 44 (50 × 50 m), and 9 (10 × 10 m) plots belonged to grazing land, cropland, and woodlots, respectively, and 28 representative homegardens were systematically selected. For each sample of data encountered, the diameter and height of woody species were measured. Micropropagation has enormous potential in the forestry industry for the mass production of elite genotypes. A limiting factor that hinders a wider adoption of this technique is its high associated cost, dissuading small producers and laboratories from its use. Thus, it is important to develop low-cost protocols to increase the competitiveness of micropropagation. The goal of the present study was to elaborate a low-cost culture medium that would allow plant growth without diminishing plant quality and production. The replacement of laboratory sucrose with commercial sugar as a carbon source, and laboratory agar with locally produced agar as a gelling agent was tested. To validate the effects of the medium for woody species micropropagation, two relevant forest species, chestnut and birch, were used and several phenotypic characteristics were recorded. Our data indicated that the substitutions made in the media composition did not alter growth or rooting parameters. Substitution of laboratory sucrose and Bacto agar by table sugar and local commercial agar provides a valuable alternative for forest species micropropagation and for increasing their potential profitability.

Radial increment objectively reflects the quality of the assimilation apparatus of a woody plant. Its features indicate the influence of various anthropogenic, biotic and abiotic factors as well as the stability of the plant under stress. The aim of this study was to survey the long-term dynamics of the radial growth of six tree species: Betula pendula, Ulmus glabra, Quercus robur, Tilia cordata, Picea abies and Pinus silvestris, depending on the impact of creating a reservoir and the fluctuations of the water level in it. Additionally, another aim was to determine whether there is a dependence between the annual radial increment of these tree species and the change in the temperature factor during the growing season. We studied cores of trees growing in a park on the coast of the Sheksna river, the level of which is regulated by the water level in the Rybinsk Reservoir and changes along with it. This research presents the dynamics of the annual radial growth of Betula pendula over 59 years, Ulmus glabra over 50 years, Quercus robur over 59 years, Tilia cordata over 82 years, Picea abies over 128 years and Pinus silvestris over 125 years. The average annual radial growth decreases in the series Ulmus glabra > Betula pendula, Quercus robur > Tilia cordata > Picea abies > Pinus silvestris. The radial increase does not correlate with the annual water level in the Rybinsk Reservoir. However, years with extreme minimum and maximum reservoir filling affect the radial growth of Quercus robur (r = 0.56) and Tilia cordata (r = 0.52). It was found that after the creation of the reservoir, the radial increment of Picea abies was significantly reduced by 1.10 mm. The clearest dependence of ring width index was obtained for Picea abies on temperature in May (r = −0.30 at p ≤ 0.05) and for Pinus sylvestris on temperature in July (r = −0.25 at p ≤ 0.05). Similarly, a weak correlation of ring width index with temperature in some months was noted for deciduous trees. Picea abies is notable for its particular display of moderate inverse correlation to the radial growths of other trees.

Smallholder farmers in the Wondo Genet were forced to switch from long-standing, diverse traditional home gardens to monoculture khat production due to increasing population pressure-induced farmland constraints. The composition of woody species and the biomass carbon stock are thought to drop as homegardens transition from polyculture to monoculture; however there is little quantitative evidence to support this claim. This study was started to assess the effects on woody species, enset, and biomass carbon of converting traditional homegardens to a fast spreading perennial monocropping of khat (Catha edulis Forskal). In 10 m × 10 m (100m2) plots from 43 farms with neighboring land use patterns for each homegarden and khat, woody species and enset were inventoried, the total number of tree, shrub, and enset species counted, as well as the height and diameter of each species measured. To determine the biomass carbon stock of each land use type, both general and species-specific allometric equations are used. Simpson's diversity index, Shannon-Wiener, and Shannon equitability were used to evaluate the diversity of enset and woody species. There were 27 different types of woody species identified, with trees making up 67 % of the total and shrubs accounting for 33 %. Shannon, Simpson, and the richness of woody species all decreased by 46 %, 51 %, and 38 %, respectively, in comparison to residential gardens. For homegardens and khat, respectively, the mean Evenness values were 0.876 and 0.539. In homegardens, Coffea arabica was the most valuable woody species, followed by Cordia africana Lam, Persea americana, Eucalyptus camaldulensis, and Grevillea Robusta. In contrast, Catha edulis was the most valuable woody species in the Khat land use type, followed by Coffea arabica, Croton macrostachyus Del, and Cordia africana. In comparison to homegardens, the above-ground, below-ground, and total biomass carbon reported in khat land use types were reduced by 18 %, 63 %, and 42 %, respectively. Grevillea and Eucalyptus species made up 51 % of the total biomass carbon stock in the homegardens, which suggests that khat and quickly expanding fast-growing plants have displaced native woody species.Understanding the long-term effects of agro-biodiversity loss requires greater research on the implications of the decline in woody species diversity and biomass carbon stock on soil fertility and sustainable farming. This is due to the numerous functions that woody species and enset play.

Abstract The internal structure of seeds is relevant to germination, seedling establishment, and early plant growth. The objective of this work was to evaluate whether internal seed structure can influence germination and initial growth in four native woody species of the Cerrado. The species chosen for this study were Anadenanthera falcata, Stryphnodendron adstringens, Stryphnodendron polyphyllum, and Tabebuia aurea. Seeds were dissected to evaluate: the lengths of the hypocotyl-radicular axis, the epicotyl and the embryonic axis, mass of the cotyledons and the embryonic axis, and eophyll number (preformed organs). Another group of seeds was germinated in a greenhouse for sixty days to verify the emergence of the epicotyl, eophylls, metaphylls, and to measure seedling biomass. Only A. falcata presented a plumule with differentiated eophylls. Anadenanthera falcata germinated before the other species in the greenhouse and had faster epicotyl and leaf emergence. Possibly, the presence of eophylls inside the seed enabled A. falcata seedlings to carry out photosynthesis, grow faster, and accumulate more aerial biomass than the other species soon after emergence. The presence of eophylls in a differentiated embryo (as in A. falcata) may hasten germination and favour seedling establishment.

Mountains represent 'experiments by nature' that permit testing ecological theory. Using herbarium samples of 92 Rhododendron species collected between 800 and 4500 m a.sl. in the Himalaya-Hengduan Mountains region, SW China, we explored congeneric elevational trends in key plant traits at the among- and at the within-species level. We aimed at identifying the contributing factors to phenotypic trait expression across a climatic gradient, distinguishing effects in response to elevation from those due to phylogenetic relatedness, non-phylogenetic species effect, and within-species trait variability. We found that a substantial fraction of the trait variation (up to 88%) is deeply rooted in phylogeny, with species elevation also revealing a strong phylogenetic signal. While accounting for these phylogenetic influences, we still found consistent associations with elevation of anatomical (increasing leaf thickness and decreasing specific leaf area), morphological (decreasing plant and leaf size, annual length increment), physiological (less negative foliage δ13C signal), and reproductive traits (flower size largely conserved, while leaf size declines, hence relative investment in flowers increases). Our findings support a combined action of phylogenetic inertia and phenotypic adjustments of plants to life conditions at high elevation. Given that our samples come from one genus and a natural climatic gradient (rather than botanical gardens) with temperature the dominant variable, such patterns represent a robust and representative signature of the multiple causes of trait-environment associations in woody species. Ancient trees hold considerable aesthetic and ecological value within urban environments. The examination of ancient trees and their overall health and condition is an integral part of a comprehensive assessment aimed at preserving these trees and conserving the natural environment within urban spaces. The purpose of the study was to examine the experience of preserving valuable trees growing in urban space. General scientific theoretical methods – analysis and synthesis – were used to work with lists of tree inventory. Heritage trees grow in all districts in Vienna. It was established that ancient trees were given the status of natural monuments since 1936, the dynamics of bequests were traced, and the decades of Vienna’s most active nature conservation activities were determined. The most famous heritage trees in the city were described. The species diversity of monumental trees was analysed, and their taxonomic affiliation was summarised. The heritage trees in Vienna are represented by 49 genera, most of which belong to the phylum of Magnoliophyta. The most common species among protected trees are Platanus orientalis L., Pinus nigra J.F. Arnold, Aesculus hippocastanum L., Quercus robur L., Taxus baccata L., Tilia platyphyllos Scop. Most of heritage trees (60%) are solitary trees. It was evaluated that the general sanitary condition of protected trees is excellent; almost all trees have high vitality and play a leading role in shaping urban landscapes. Every heritage tree possesses a commendable and elevated aesthetic rating. Analysis of the existing inventory of ancient trees in Vienna, their sanitary condition and aesthetic condition will contribute to the environmental awareness of communities and the improvement of management measures for such trees. The obtained results can be used to share the experience of researchers from different countries regarding preserving ancient trees and forming European databases of heritage trees

Abstract Background Abiotic factors exert different impacts on the abundance of individual tree species in the forest but little has been known about the impact of abiotic factors on the individual plant, particularly, in a tropical forest. This study identified the impact of abiotic factors on the abundances of Podocarpus falcatus, Croton macrostachyus, Celtis africana, Syzygium guineense, Olea capensis, Diospyros abyssinica, Feliucium decipenses, and Coffea arabica. A systematic sample design was used in the Harana forest, where 1122 plots were established to collect the abundance of species. Random forest (RF), artificial neural network (ANN), and generalized linear model (GLM) models were used to examine the impacts of topographic, climatic, and edaphic factors on the log abundances of woody species. The RF model was used to predict the spatial distribution maps of the log abundances of each species. Results The RF model achieved a better prediction accuracy with R 2 = 71% and a mean squared error (MSE) of 0.28 for Feliucium decipenses. The RF model differentiated elevation, temperature, precipitation, clay, and potassium were the top variables that influenced the abundance of species. The ANN model showed that elevation induced a negative impact on the log abundances of all woody species. The GLM model reaffirmed the negative impact of elevation on all woody species except the log abundances of Syzygium guineense and Olea capensis. The ANN model indicated that soil organic matter (SOM) could positively affect the log abundances of all woody species. Logging activities, climate change and variation are threatening the sustainability of the entire forest ecosystem. Maintenance and periodic assessment of diverse ecosystems are, therefore, crucial for the long-term survival of human beings. However, baseline data on the resulting depletion of forest ecosystems in Nigeria are insufficient or lacking. The aim of the present study was to assess the phytosociological relationship between species composition and the diversity of woody species at Omo Biosphere Reserve. Three lines of transect were cut apart 200 m far from each other. Each transect has 4 plots of 50 x 50m in an alternate manner at 100m intervals. Trees with e" 10 cm diameter at breast height (DBH) in each plot were measured for height, DBH. The data were analyzed for species composition, richness, diversity, relative density, and relative dominance, Species Importance Value Index (IVI), Species diversity was computed using Shannon's Weaner and Simpson's diversity indices. The Data were analyzed using PAST software Version.3 and Microsoft Excel 2010. The Carl-Pearson correlation coefficient was calculated between various phytosociological parameters. A total of 427 trees representing 57 from 22 families were encountered in Omo Biosphere Reserve. The stocking density of dominant tree species/ha ranges from 0.33 trees/ha to 26.33 trees/ha. For Simpson's similarity between the plots, it varied from 0 % to 77 % for all the plots. The species diversity (Simpson's diversity indices) between the plots varies from 64 % to 93 % for all plots. Abandoned crop fields in central Kansas, USA, are soon colonized by several non-native and native woody species. This study compared subsequent vegetation change in control (unmanipulated) old field successional plots and plots planted with Quercus macrocarpa (bur oak) seedlings following cessation of row crop agriculture. The site, a former corn field, was subject to invasion primarily by Ulmus pumila (Siberian elm), Lonicera tartarica (Amur honeysuckle), and Juniperus virginiana (eastern redcedar). I established eight 20 m × 32 m plots in April 2007, four as controls and four each containing 40 bur oak seedlings planted 4 m apart. I monitored permanent 4 m × 4 m quadrats (12 quadrats per plot) for 16 years for the presence of naturally establishing herbaceous and woody species. I sampled a total of 94 herbaceous and 27 woody species, with L. maackii , U. pumila , and J. virginiana occurring in 95 to 100% of quadrats by 2022. Total species richness, total woody species richness, native woody species richness, and richness of animal-dispersed woody species were higher in the oak plots than in the controls. Dissimilarity between the treatments steadily increased with time. The results suggest that an enrichment planting of bur oak seedlings at the outset can influence the early successional trajectory of old fields, and indicate that such plantings can drive restorations away from invasive non-native species and toward more desirable native species in later years. Land use is known to influence the diversity of vascular plants in the Miombo woodlands. However, little is known about the interaction between soil and land use in herbaceous and woody species. We compared the diversity of vascular plants at the plot level (20 m × 50 m) and site level for three sites in the Miombo woodlands of western Zambia subject to different levels of intensity classes of diffuse land use (e.g., livestock herbivory and selective timber harvesting). For each of the sites, twenty plots were randomly selected for assessment of species composition of vascular plant species, indicators of land-use intensity, and soil chemistry per plot. We hypothesized that the site with the lowest human impact would have the highest richness and diversity of woody and herbaceous species. At the site level, we found that richness and diversity of woody species were unaffected by land-use intensity, whereas herbaceous species richness was higher for the protected site (28 species on average per 1000 m2) than the two other sites (23 and 21 species on average per 1000 m2). At the plot level, herbaceous species richness was positively associated with woodcutting and soil pH. We interpret the positive effect of woodcutting on herbaceous species richness as the effect of lower competition by the woody component for resources such as water, nutrients, and light. With regard to the absence of any effect of land-use intensity on the richness of woody species, we conclude that in our study areas selective timber harvesting may be at a sustainable level and might even have a positive effect on the diversity of the herbaceous layer. Phosphogypsum (PG) is a by-product of phosphorus fertilizer that is typically stacked near production sites. Phosphogypsum contains trace elements and naturally occurring radioactive materials which may be hazardous to the surrounding environment. Phosphogypsum stack reclamation typically involves placing a soil cap and seeding grass to create a barrier for reducing environmental impacts; using woody species is uncommon. This study used three soil treatments with grass and woody species to determine whether mixing PG with soil affects soil chemical properties, and metal and radionuclide concentrations in tissue. None of the elements in soil was above Canadian guidelines for industrial land use. Aluminum, beryllium, chromium, copper, iron, magnesium, manganese, nickel, and vanadium were significantly higher in both study and reference sites than in pure PG; cadmium, calcium, fluoride, and strontium were significantly higher in pure PG. There was a poor correlation between soil and plant concentrations for most elements indicating trace elements were not in a bioavailable form. Trace elemental concentrations in plant tissue generally differed significantly with vegetation type but not within similar species. Trace elements and isotopes in PG were not high enough to affect plant growth. Among the isotopes, 222Ra emissions differed significantly with vegetation covers; activity of 226Ra in pure PG was above Canadian guidelines, but lower in vegetation tissue. This study suggests 15 cm soil mixed with PG can be used for PG stack revegetation when fast-growing Salix and Populus species are used in reclamation. [ABSTRACT FROM AUTHOR]

Premise: The isolation of RNA from trees is challenging due to the interference of polyphenols and polysaccharides with downstream processes. Furthermore, many RNA extraction protocols are time consuming and involve hazardous chemicals. To address these issues, we aimed to develop a safe protocol for high‐quality RNA extraction from diverse Metrosideros taxa representing a broad range of leaf toughness, pubescence, and secondary metabolites. Methods and Results: We tested popular RNA isolation kits and protocols that were effective on other recalcitrant trees, including a broad range of optimization and purification steps. We optimized a protocol involving two silica‐membrane column‐based kits that yielded high‐quantity RNA with an RNA integrity number >7 and without DNA contamination. All RNA samples were used successfully in a follow‐on RNA‐Seq experiment. Conclusions: We present an optimized high‐throughput RNA extraction protocol that yielded high‐quality and high‐quantity RNA from three contrasting leaf phenotypes within a hyperdiverse woody species complex. Mountainous ecosystems provide social, economic, and environmental services at different scales. Nonetheless, currently, they have been exposed to environmental degradation risks. This study was conducted to investigate woody plant species diversity and threats to the study forest. Systematic sampling was employed to collect vegetation data from fifty 400 m2 sample plots along five transect lines. Vegetation and the environmental variables were recorded from each plot. Eighty randomly sampled households were selected for socioeconomic data. Descriptive statistics and ranking exercises were employed. The vegetation classification was performed using the R program version 2.15.2. The species diversity, richness, and evenness were computed. The result showed that five plant communities were recognized. A total of 99 woody plants belonging to 87 genera and 50 families were identified, of which 13 % were endemic. The total basal area of the study forest was 3.40 m2ha-1, and the forest was characterized by a bell-shaped population structure identified by poor regeneration and recruitment. With increasing altitude, there was a significant decrease in human impacts, grazing, and the number of plant species. About 90 % of the informants disclosed deforestation as the major threat to forest cover change, whereas 84 % of them recommended forest restoration as a tangible measure. There was a high deforestation rate of the selected woody species (e.g., Arundinaria alpina) and high expansion of agriculture at the fringes of the forest. This requires promoting forest land rehabilitation activities, nature tourism, establishing partnerships with communities on the forest management, consolidating village-level institutions and developing livelihood alternatives for communities at the local governments and the community level to restore the degraded forest.

Abstract Exclosure becomes popular as a naming of the practice of excluding degrading agents from degraded lands for natural rehabilitation. However, its role on woody species regeneration in the Loma Bosa District of the southwestern Ethiopia has not been investigated. Therefore, this study examines the role of exclosure on woody species regeneration by comparing exclosure, open woodland, and degraded land areas. A systematic transect sampling method was employed to collect vegetation data in sampling quadrats, each with a size of 20 × 20 m, evenly distributed along parallel transect lines. All the woody plant species in each plot were identified and measured for DBH and height. Twenty-six woody species, representing 16 plant families, were recorded at the study area, of which only eight were recorded all in the exclosure, open woodland and open degraded land. Species Diversity Index (H′) was 2.62, 2.38, and 1.56 for woody species in exclosure, open woodland area, and open degraded land area. Wood species density were 2225 ha−1, 1642 ha−1, and 297 ha−1 for exclosure, open woodland area, and open degraded land area, respectively. The distribution of the height and DBH of the recorded species in exclosure exhibited an inverted “J” shape pattern suggesting a healthy regeneration status of the important species, while others revealed irregular and less interpretable pattern. Overall results from this study indicated that exclosure is important for improvement of woody species regeneration in degraded lands in the study area.

Effects of exclosures on restoring degraded lands may vary with soil type, exclosure age, and conditions before the establishment of exclosures. Yet, studies investigating the effectiveness of exclosures in restoring degraded lands under different environmental conditions are lacking. This study aims at investigating the changes in woody species richness and diversity, and ecosystem carbon stocks after implementing exclosures in the Central Rift Valley, Ethiopia. Vegetation and soil data were gathered from 120 nested plots established in exclosures of eight and 30‐years‐old and adjacent grazing lands. Results showed that exclosures contained a higher number of economically important woody species compared to their respective adjacent grazing lands. However, the exclosures and respective adjacent grazing lands did not differ significantly in the diversity of tree and shrub species, and both the exclosures and adjacent grazing lands were dominated by few tree and shrub species. The older exclosure (30 years old) displayed significantly (p < 0.01) higher soil organic carbon and soil total nitrogen content and stocks than the adjacent grazing land, whereas the youngest exclosure (8 years old) did not show a significant difference in these variables. The results suggest that a longer time (e.g., ≥10 years) is needed to detect significant differences in soil organic carbon and total soil nitrogen. However, exclosures could bring considerable changes in woody species density in a relatively shorter period (e.g., ≤10 years) and support to restore degraded native woody species. Fire use in protected savannas of Africa is a common practice. Fires in these savannas create many environmental benefits, such as reducing grass, brush and trees that can fuel large and severe wildfires and improving wildlife habitat. However, wrong timing of fire can threaten plants, animals and habitats. This study investigated the effects of time of burning on woody plant composition, diversity and density in the Mole National Park, Ghana. A total of twelve 300 m2 plots were systematically sampled in a 200 m × 200 m treatment plot established by Park Management each for early burn, late burn and no-burn plots. Twenty-seven different woody species belonging to fourteen families were recorded in all the treatments. Most of the species identified belonged to the families Fabaceae and Combretaceae. Vitellaria paradoxa (Shea), Terminalia avicennioides, Combretum adenogonium and Combretum molle were the most common and abundant in all treatments. A TWINSPAN on sites and species revealed four species groups based on affinity to burning time. A follow-up DCA showed a strong association between burning time and species composition, with the first two axes explaining 65% of variation. The late burn and no-burn treatments recorded the lowest diversity amongst the three treatments. Stem density was highest in no-burn treatment which had lowest species richness and diversity compared to early and late burn treatments. Early burn treatment had the highest diversity and the lowest density of woody species. The study revealed that the different times of prescribed burning influenced vegetation differently. Prescribed early dry season burning could contribute to the management of indigenous woody species in protected fire-prone savannas, because it can promote the diversity of species, as found in the Mole National Park in the Guinea savanna of Ghana. Increased access to genome‐wide data provides new opportunities for plant conservation. However, information on neutral genetic diversity in a small number of marker loci can still be valuable because genomic data are not available to most rare plant species. In the hope of bridging the gap between conservation science and practice, we outline how conservation practitioners can more efficiently employ population genetic information in plant conservation. We first review the current knowledge about neutral genetic variation (NGV) and adaptive genetic variation (AGV) in seed plants, regarding both within‐population and among‐population components. We then introduce the estimates of among‐population genetic differentiation in quantitative traits (QST) and neutral markers (FST) to plant biology and summarize conservation applications derived from QST–FST comparisons, particularly on how to capture most AGV and NGV on both in‐situ and ex‐situ programs. Based on a review of published studies, we found that, on average, two and four populations would be needed for woody perennials (n = 18) to capture 99% of NGV and AGV, respectively, whereas four populations would be needed in case of herbaceous perennials (n = 14). On average, QST is about 3.6, 1.5, and 1.1 times greater than FST in woody plants, annuals, and herbaceous perennials, respectively. Hence, conservation and management policies or suggestions based solely on inference on FST could be misleading, particularly in woody species. To maximize the preservation of the maximum levels of both AGV and NGV, we suggest using maximum QST rather than average QST. An accurate profile of gene expression at a cellular level can contribute to a better understanding of biological processes and complexities involved in regulatory mechanism of woody plants. Laser microdissection is one technique that allows isolation of specific, target cells or tissue from a heterogeneous cell population. This technique entails microscopic visualization of the selected tissue and use a laser beam to separate the desired cells from surrounding tissue. Initial identification of these cells is made based on morphology and/or histological staining. Some works have been made in several tissues and plant models. However, there are few studies of laser microdissection application in woody species, particularly, lignified and suberized cells. Moreover, the presence of high level of suberin in cell walls can be a big challenge for the application of this approach. In our study it was developed a technique for tissue isolation, using laser microdissection of four different plant cell types (phellogen, lenticels, cortex and xylem) from woody tissues of cork oak (Quercus suber), followed by RNA extraction and RNA-Seq. We tested several methodologies regarding laser microdissection, cryostat equipments, fixation treatments, duration of single-cells collection and number of isolated cells by laser microdissection and RNA extraction procedures. A simple and efficient protocol for tissue isolation by laser microdissection and RNA purification was obtained, with a final method validation of RNA-Seq analysis. The optimized methodology combining RNA-Seq for expression analysis will contribute to elucidate the molecular pathways associated with different development processes of the xylem and phellem in oaks, including the lenticular channels formation. Background: Abiotic factors exert different impacts on the abundance of individual tree species in the forest but little has been known about the impact of abiotic factors on the individual plant, particularly, in a tropical forest. This study identified the impact of abiotic factors on the abundances of Podocarpus falcatus, Croton macrostachyus, Celtis africana, Syzygium guineense, Olea capensis, Diospyros abyssinica, Feliucium decipenses, and Coffea arabica. A systematic sample design was used in the Harana forest, where 1122 plots were established to collect the abundance of species. Random forest (RF), artificial neural network (ANN), and generalized linear model (GLM) models were used to examine the impacts of topographic, climatic, and edaphic factors on the log abundances of woody species. The RF model was used to predict the spatial distribution maps of the log abundances of each species. Results: The RF model achieved a better prediction accuracy with R2 = 71% and a mean squared error (MSE) of 0.28 for Feliucium decipenses. The RF model differentiated elevation, temperature, precipitation, clay, and potassium were the top variables that influenced the abundance of species. The ANN model showed that elevation induced a negative impact on the log abundances of all woody species. The GLM model reaffirmed the negative impact of elevation on all woody species except the log abundances of Syzygium guineense and Olea capensis. The ANN model indicated that soil organic matter (SOM) could positively affect the log abundances of all woody species. The GLM showed a similar positive impact of SOM, except for a negative impact on the log abundance of Celtis africana at p < 0.05. The spatial distributions of the log abundances of Coffee arabica, Filicium decipenses, and Celtis africana were confined to the eastern parts, while the log abundance of Olea capensis was limited to the western parts. Conclusions: The impacts of abiotic factors on the abundance of woody species may vary with species. This ecological understanding could guide the restoration activity of individual species. Understanding how species turnover responds to environmental change may provide insights into the ecological factors influencing biogeographical patterns. Here, I examined geographic patterns in taxonomic and functional turnover of tree assemblages in Europe and compared the influence of environmental factors on turnover. I conducted a principal component analysis with nine above‐ and below‐ground plant traits for 210 tree species. I used the resulting four principal components (82% of the variance) to create five functional dendrograms considering all trait dimensions together and individually. Further, I used species composition and the functional dendrograms to calculate pairwise taxonomic and functional turnover between tree assemblages in 100 × 100 km grid cells across Europe. To assess the influence of temperature, precipitation, precipitation seasonality, soil pH and geographic distance on taxonomic and functional turnover, I conducted multiple regression on distance matrices (MRM). I also compared the slope of the relationship between functional turnover and environmental distance among trait dimensions to detect what ecological strategies may be more sensitive to environmental changes. I found that mean taxonomic and functional turnover was particularly high in lowland areas of the Mediterranean Basin. Geographic patterns of individual trait dimensions largely reproduced those considering all trait dimensions together, additionally revealing some regional differences. MRM explained a similar fraction of the variation in taxonomic and functional turnover. The influence of environmental distance was stronger for trait dimensions related to tree size and woodiness than for trait dimensions related to the leaf and root economics. I conclude that geographic patterns in the turnover of tree assemblages in Europe coincide with major biome transitions. Trees have been used for phytoremediation and as biomonitors of air pollution. However, the mechanisms by which trees mitigate nanoparticle pollution in the environment are still unclear. We investigated whether two important tree species, European beech (Fagus sylvatica L.) and Scots pine (Pinus sylvestris L.), are able to take up and transport differently charged gold nanoparticles (Au-NPs) into their stem by comparing leaf-to-root and root-to-leaf pathways. Au-NPs were taken up by roots and leaves, and a small fraction was transported to the stem in both species. Au-NPs were transported from leaves to roots but not vice versa. Leaf Au uptake was higher in beech than in pine, probably because of the higher stomatal density and wood characteristics of beech. Confocal (3D) analysis confirmed the presence of Au-NPs in trichomes and leaf blade, about 20–30 μm below the leaf surface in beech. Most Au-NPs likely penetrated into the stomatal openings through diffusion of Au-NPs as suggested by the 3D XRF scanning analysis. However, trichomes were probably involved in the uptake and internal immobilization of NPs, besides their ability to retain them on the leaf surface. The surface charge of Au-NPs may have played a role in their adhesion and uptake, but not in their transport to different tree compartments. Stomatal conductance did not influence the uptake of Au-NPs. This is the first study that shows nanoparticle uptake and transport in beech and pine, contributing to a better understanding of the interactions of NPs with different tree species. Abstract Premise The isolation of RNA from trees is challenging due to the interference of polyphenols and polysaccharides with downstream processes. Furthermore, many RNA extraction protocols are time consuming and involve hazardous chemicals. To address these issues, we aimed to develop a safe protocol for high‐quality RNA extraction from diverse Metrosideros taxa representing a broad range of leaf toughness, pubescence, and secondary metabolites. Methods and Results We tested popular RNA isolation kits and protocols that were effective on other recalcitrant trees, including a broad range of optimization and purification steps. We optimized a protocol involving two silica‐membrane column‐based kits that yielded high‐quantity RNA with an RNA integrity number >7 and without DNA contamination. All RNA samples were used successfully in a follow‐on RNA‐Seq experiment. Conclusions We present an optimized high‐throughput RNA extraction protocol that yielded high‐quality and high‐quantity RNA from three contrasting leaf phenotypes within a hyperdiverse woody species complex.
