Humane Papillomviren (HPV) infizieren Keratinozyten der Haut- und Schleimhaut. Die Hochrisiko-Typen (HR) der alpha-Gattung verursachen dadurch anogenitalen- und oropharyngalen-Krebs. Vorherrschend dabei ist das Zervixkarzinom, welches zu 70 % von HPV16 und HPV18 ausgelöst wird. Im Gegensatz dazu sind die onkogenen Eigenschaften der beta-HPV Gattung weniger gut erforscht. Bei Patienten mit der seltenen Erbkrankheit Epidermodysplasia Verruciformis (EV) und Organtransplantatempfängern (OTRs) wird ein Zusammenhang zwischen beta-HPV-Infektionen und der Entstehung von kutanen Plattenepithelkarzinomen vermutet. Bisher wurden die Eigenschaften von beta-HPV vor allem durch retrovirale Expression der E6 und E7 Onkogene in Keratinozyten, durch Transfektion der Osteosarkomzelllinie U2OS mit beta-HPV Genomen und in transgenen Mausmodellen untersucht, aber nicht mit vollständigen viralen Genomen in normalen humanen Keratinozyten (NHK), den natürlichen Zielzellen. Im Rahmen meiner Dissertation konnte ich zeigen, dass HPV8-, 38- und 49-Genome in humanen Keratinozyten mindestens neun Tage lang transkriptionell aktiv sind und replizieren. Durch Inaktivierung des viralen E8^E2 Repressors (E8-/E8^E2-) erhält das HPV49 Genom die Fähigkeit NHK zu immortalisieren. Die immortalisierten HPV49 E8- Zelllinien enthalten große Mengen an episomalen Virusgenomen und viralen Transkripten und behalten diese in Kultur über einen langen Zeitraum. Nicht nur der Verlust der E6 und E7 Onkogene, sondern auch eine Inaktivierung der E1 oder E2 Replikationsgene verhindern die Immortalisierung. Die E8-/E1- und E8-/E2- Genome zeigen deutlich niedrigere E6 und E7 Transkriptmengen in transienten Experimenten. Dies legt nahe, dass für die Immortalisierung eine starke E6 und E7 Expression von extrachromosomalen Virusgenomen erforderlich ist. Die Notwendigkeit für eine Inaktivierung von E8 im Kontext von intakten E1 und E2 Genen für die Immortalisierung von NHK zeigt, dass E8^E2 eine wichtige Rolle bei der Kontrolle der Onkogenität von beta-HPV spielen könnte, und weist auf grundlegende Unterschiede zwischen HPV49 und HR-HPV Genomen hin. Durch RNA-Sequenzierung der HPV49 E8- positiven Zelllinien konnten bekannte Spleißverknüpfungen bestätigt, das frühe Polyadenylierungssignal lokalisiert und Hinweise auf unterschiedliche virale Promotoren erhalten werden. Außerdem wurden neue Spleißverbindungen kartiert und ein neuer Spleißdonor im E6 Gen funktionell untersucht. Dieser beeinflusst die Menge an E6 Protein und vermutlich dadurch die Immortalisierung durch das HPV49 E8- Genom. Bei HPV8 und 38 konnten auch durch Inaktivierung von E8^E2 keine immortalisierten Zelllinien erzeugt werden, obwohl ähnliche Mengen an Transkripten für die E6 und E7 Onkogene wie bei HPV49 E8- Genomen transkribiert wurden. Dies legt nahe, dass beta-HPV Genome unterschiedliche onkogene Eigenschaften besitzen. Die Inaktivierung der Replikationsgene E1 und E2 reduzierte die Transkription aller untersuchten beta-HPV in NHK, was nahelegt, dass auch WT Genome aktiv replizieren. Die Kultivierung von HPV8- oder 38-transfizierten NHKs in organotypischen Modellen, die die Analyse des produktiven Replikationszyklus von HR-HPV ermöglichen, induziert Transkripte für das L1 Kapsidgen, was nahelegt, dass der produktive Lebenszyklus eingeleitet wurde. Außerdem ähnelt das HPV8-Transkriptionsmuster in organotypischen Kulturen dem einer HPV8-positiven EV-Läsion. Zusammengefasst bedeutet dies, dass NHK ein physiologisch relevantes System sind, um die Replikation und onkogenen Eigenschaften von beta-HPV zu untersuchen. Human papillomaviruses (HPV) infect keratinocytes of the skin and mucosal membranes. The high-risk types (HR) of the alpha genus thus cause anogenital and oropharyngeal cancers. Predominant among these is cervical carcinoma, 70% of which is caused by HPV16 and HPV18. In contrast, the oncogenic properties of the beta-HPV genus are less well understood. In patients with the rare hereditary disease epidermodysplasia verruciformis (EV) and organ transplant recipients (OTRs), a link between beta-HPV infections and the development of cutaneous squamous cell carcinoma is suspected. So far, the properties of beta-HPV have been investigated mainly by retroviral expression of the E6 and E7 oncogenes in keratinocytes, by transfection of the osteosarcoma cell line U2OS with beta-HPV genomes and in transgenic mouse models, but not with complete viral genomes in normal human keratinocytes (NHK), the natural target cells. As part of my dissertation, I was able to show that HPV8, 38 and 49 genomes are transcriptionally active and replicate in human keratinocytes for at least nine days. By inactivating the viral E8^E2 repressor (E8-/E8^E2-), the HPV49 genome acquires the ability to immortalise NHK. The immortalised HPV49 E8- cell lines contain large amounts of episomal viral genomes and viral transcripts and retain them in culture for a long time period. Not only the loss of the E6 and E7 oncogenes, but also inactivation of the E1 or E2 replication genes prevent immortalisation. The E8-/E1- and E8-/E2- genomes show significantly lower E6 and E7 transcript levels in transient experiments. This suggests that strong E6 and E7 expression of extrachromosomal viral genomes is required for immortalisation. The requirement for E8 inactivation in the context of intact E1 and E2 genes for NHK immortalisation indicates that E8^E2 may play an important role in controlling the oncogenicity of beta-HPV, and points to fundamental differences between HPV49 and HR-HPV genomes. RNA sequencing of HPV49 E8-positive cell lines confirmed known splice junctions, localised the early polyadenylation signal and provided evidence for distinct viral promoters. In addition, new splice junctions were mapped and a new splice donor in the E6 gene was functionally investigated. This influences the amount of E6 protein and presumably thereby the immortalisation by the HPV49 E8- genome. For HPV8 and 38, immortalised cell lines could not be generated even by inactivating E8^E2, although similar amounts of transcripts for the E6 and E7 oncogenes were transcribed as in HPV49 E8- genomes. This suggests that beta-HPV genomes have different oncogenic properties. Inactivation of the replication genes E1 and E2 reduced transcription of all beta-HPVs examined in NHK, suggesting that wildtype genomes also actively replicate. Culturing HPV8- or 38-transfected NHKs in organotypic models that allow analysis of the productive replication cycle of HR-HPV induces transcripts for the L1 capsid gene, suggesting that the productive life cycle has been initiated. Furthermore, the HPV8 transcriptional pattern in organotypic cultures resembles that of an HPV8-positive EV-lesion. In summary, this means that NHK are a physiologically relevant system to study the replication and oncogenic properties of beta-HPV.

Coinfection with hepatitis C virus (HCV) influences HIV reservoir size. However, it is unknown whether this coinfection also induces a higher provirus transcription. Viral transcription is promoted by synergy between cellular factors such as NF-&kappa, B and the viral regulator Tat. The impact of HCV coinfection on HIV provirus transcription was analyzed in resting (r)CD4 T+ cells (CD3+CD4+CD25-CD69-HLADR-) and rCD4 T cells-depleted PBMCs (rCD4 T- PBMCs) from a multicenter cross-sectional study of 115 cART-treated HIV patients: 42 HIV+/HCV+ coinfected individuals (HIV+/HCV+), 34 HIV+ patients with HCV spontaneous clearance (HIV+/HCV&minus, ) and 39 HIV patients (HIV+). Viral transcription was assessed in total RNA through the quantification of unspliced, single spliced, and multiple spliced viral mRNAs by qPCR. Linear correlations between viral reservoir size and viral splicing were determined. A 3-fold increase of multiple spliced transcripts in rCD4 T+ cells of HIV+/HCV+ patients was found compared to HIV+ individuals (p &lt, 0.05). As Tat is synthesized by multiple splicing, the levels of Tat were also quantified in these patients. Significant differences in single and multiple spliced transcripts were also observed in rCD4 T- PBMCs. Levels of multiple spliced mRNAs were increased in rCD4 T+ cells isolated from HIV+/HCV+ subjects, which could indicate a higher Tat activity in these cells despite their resting state.
