Antarctic benthic ecosystems support high biodiversity but their characterization is limited to a few well-studied areas, due to the extreme environment and remoteness making access and sampling difficult. Our aim was to compare water and sediment as sources of environmental DNA (eDNA) to better characterise Antarctic benthic communities and further develop practical approaches for DNA-based biodiversity assessment in remote environments. We used a cytochrome c oxidase subunit I (COI) metabarcoding approach to characterise metazoan communities in 26 nearshore sites across 12 locations in the Vestfold Hills (East Antarctica) based on DNA extracted from either sediment cores or filtered seawater. We detected a total of 99 metazoan species from 12 phyla across 26 sites, with similar numbers of species detected in sediment and water eDNA samples. However, significantly different communities were detected in the two sample types at sites where both were collected (i.e., where paired samples were available). For example, nematodes and echinoderms were more likely to be detected exclusively in sediment and water eDNA samples, respectively. eDNA from water and sediment core samples are complementary sample types, with epifauna more likely to be detected in water column samples and infauna in sediment. More reference DNA sequences are needed for infauna/meiofauna to increase the proportion of sequences and number of taxa that can be identified. Developing a better understanding of the temporal and spatial dynamics of eDNA at low temperatures would also aid interpretation of eDNA signals from polar environments. Our results provide a preliminary scan of benthic metazoan communities in the Vestfold Hills, with additional markers required to provide a comprehensive biodiversity survey. The Rauer Group and Vestfold Hills are critical components of the East Antarctic Shield and are believed to have been parts of the supercontinents Rodinia and Gondwana. The Rauer Group is characterized by Archean to Pan-African thermal history, while the Pan-African signature in the Vestfold Hills is restricted to its southwestern margin. The correlation between the Rauer Group and Vestfold Hills remains controversial, while the significance of amalgamation between these two terranes remains unclear. Most importantly, the precise nature and timing of the Rauer-Vestfold contact is disputed. In this review, we compare the available structural, metamorphic, geochronological and geophysical data in these two East Antarctic terranes and suggest that (i) the Rauer Group and Vestfold Hills of East Antarctica were amalgamated during the late Mesoproterozoic to early Neoproterozoic (∼1000 Ma), and (ii) the Rauer-Vestfold contact is located beneath the Sørsdal Glacier in the East Antarctic Shield. Finally, we argue that juxtaposition of the Rauer-Vestfold contiguous crustal unit with the Eastern Ghats Province (EGP)-Rayner amalgam took place in the Pan-African time (∼500 Ma). The non-marine diatom flora of the Antarctic continent is currently being revised as recent work within the Maritime and Sub-Antarctic regions has unveiled a number of new endemic species. The under reporting of endemic species is partially due to the historic 'force-fitting' of Antarctic species into European names. Within East Antarctica, the Vestfold Hills are an extensively studied area known for their diversity of lakes spanning a salinity gradient, for which Diatoms of the saline lakes of the Vestfold Hills, Antarctica by Roberts & McMinn (1999) has a primary identification resource for over two decades. To determine if improved taxonomic resolution alters the ecological interpretation of these lakes, 30 prepared lake sediment samples first examined by Roberts & McMinn (1999) were re-analysed using an updated, fine-grained taxonomic approach. Our analyses confirmed the role of salinity and alkalinity in structuring Vestfold Hills diatom communities. Thus, the increased taxonomic resolution did not change the original ecological interpretation. However, the revised flora of the Vestfold Hills features 37 species endemic to the Antarctic Region, including members of Luticola, Sabbea and Halamphora, among others. In total, 183 taxa across 38 genera were observed, in contrast to the 67 taxa reported in the original work. The revised flora, including a larger proportion of endemic species, may improve our understanding of the diatom community in general and has wider implications for both conservation and biodiversity estimates of Antarctica. The Vestfold Hills sector on the coastal fringe of the Princess Elizabeth Land forms part of an Archean to Paleoproterozoic aged cratonic nucleus of the East Antarctic Shield. A charnockite-granite association from the Mossel Gneiss Group in the northern region of the Vestfold Hills is investigated in this work to characterise the metamorphic-magmatic evolution of the area. Conventional thermometry and phase equilibria modelling indicate that the charnockite formed as a result of ultra-high temperature metamorphism at low-intermediate pressure (∼4 kbar). Phase equilibria modelling, reveals that the melt-integrated charnockite composition is a restitic product of a protolith of quartz diorite composition which underwent isobaric heating (peak temperature up to 960 °C) at 4 kbar pressure followed by anatexis. The anatexis of the quartz diorite protolith also resulted in the formation of the associated granitic melt. U–Th-Pb Total ages obtained from monazites of the granite integrated with CHIME ages extracted using the isochron method indicate an emplacement age of ∼2200 Ma and two younger events are recorded at ∼2000 Ma and ∼1700 Ma. The younger ages are ascribed to the resetting during the emplacement of younger basalt dykes that cross cut the charnockite-granite sequence. A 47 cm long sediment core collected from an inland lake of the Vestfold Hills, East Antarctica has been examined to reconstruct the palaeolacustrine environmental changes. The core shows dates at two core intervals of 18-19 cm and 28-29 cm as 5050±98 yrs BP and 5560±96 yrs BP, respectively. The core exhibits a good amount of diatom population throughout the length. Prior to 5560±96 yrs BP, the core shows high abundance of diatom population (>10×107 g-1) along with sufficient salt crystal, which indicates the connection of this lake with the marine environment. From 5560±96 yrs BP to 5050±98 yrs BP the total number diatoms decreased substantially along with the decrease in salt crystal, which indicates the withdrawal of the marine influence from the lake during that period. From 5050±98 yrs BP to Recent, the low number of diatoms and the rare occurrence of salt crystal suggest that the lake remained mostly detached from the sea during the last 5000 yrs.

Bivalves collected in Vestfold Hills during the 55th Russian Antarctic Expedition (RAE, 2009−2010) are represented by five species. Four of them ( Laternula elliptica, Thracia meridionalis, Adamussium colbecki, and Philobrya sublaevis) were collected in the sediments that filled the coastal lake terraces during the Holocene; these species are still abundant on the Antarctic shelf at present. Bivalves were found in eight samples, with L. elliptica shells and fragments thereof found in seven of those samples. The sample collected near Deep Lake had the most diverse species composition, as it contained all four species named above. Shells of named species widely occurring in present-day Antarctica were found high above sea level in the marine sediments of the oasis. This indicate to the similarity of the oasis habitats in the past and the recent marine conditions. Fragments of shells of the fifth (now extinct) species Ruthipecten tuftsensis were found in glacial-marine sediments of the Marine Plain dating back to the late Pleistocene. An Archaean continent ‘SIWA’, an acronym for South India–Western Australia, comprising the Bastar–Dharwar craton, the Yilgarn craton, the Napier Complex, and the Vestfold Hills has been identified from palaeomagnetic and spatio-temporal data. This assembly was dispersed in three phases with the development of the proto-Indian ocean. The first and second events ~2350 and ~2000 Ma were related to the separation of the Yilgarn craton and the Napier Complex, respectively, to form a proto-Indo-Antarctic ocean and the Cuddapah basin. The proto-ocean was closed ~1650 Ma by the collision of the Lambert Terrane of East Antarctica and the Bastar–Dharwar craton. This collision, associated with ultra-high temperature (UHT) granulite facies metamorphism, is identified in the southern domain of the Eastern Ghats and the Oygardens domain of East Antarctica. The third extensional event between 1500 and 1200 Ma was associated with the separation of the Vestfold Hills block and a second phase of opening of the proto-Indian ocean, and the development of a series of basins on the western side of the Eastern Ghats (the Chhatisgarh, Khariar, Ampani, Indravati, and Sabari basins). The closing of this ocean basin during the Eastern Ghats–Rayner orogeny at ~950 Ma was related to the amalgamation of India and East Antarctica to form the supercontinent Rodinia. During the Neoproterozoic, this part of Rodinia was involved in orogenic collapse/extension and deposition of the Sodruzhesvo Group. An integrated analysis of biological and geoscientific data collected from the nearshore marine environment of the Vestfold Hills was used to identify benthic habitats and associated communities and examine relationships between benthic community composition and environmental characteristics. A 48 km 2 area was surveyed using a multibeam echosounder system (MBES) to produce high-resolution bathymetry and backscatter intensity maps of the seabed. Epibenthic community data and in situ observations of substrate composition and seafloor bedforms and features were obtained from towed underwater video. A comparison of top-down and bottom-up approaches to defining benthic habitats was used to improve understanding of the applicability of mapping methodologies. On a broad scale, both approaches produced habitat classes distinguished largely by geomorphic features, with substrate and depth identified as the main controls of benthic community composition, however, the relationship between benthic community composition and environmental characteristics is complex with many variables contributing to differences in community composition. The top-down approach was based on geomorphic units defined using abiotic characteristics and the assemblages identified within the geomorphic were very broad with only weak distinction between assemblages. Conversely, the bottom-up approach generated additional habitat classes, identified clear defining taxa for each class, greater distinction between the benthic communities, and allowed identification of additional environmental factors (i.e. sea ice cover) that influence benthic community distribution that are not discernible from geomorphic information alone. The habitat types identified and mapped using the bottom-up approach include shallow boulder fields and exposed bedrock which are dominated by dense macroalgae communities, and steep slopes, muddy basins and sandy plains which are dominated by invertebrate communities. The results indicate that a bottom-up approach is preferable for benthic habitat mapping, however, where detailed information is not available, geomorphic information provides a reasonable indication of the distribution of benthic habitats and communities. This study highlights the utility of multibeam sonar for interpretation of seafloor morphology and substrate and the multibeam data provide a physical framework for understanding benthic habitats and the distribution of benthic communities. This research provides the scientific context and spatial framework for managing the Vestfold Hills nearshore marine environment and provides a baseline for assessing environmental change. We represent Paleoenvironmetal condition of Vestfold Hills region, based on the relative abundance of a single diatom species Fragilariopsis curta, a dominating sea-ice taxon. Increase in relative abundance of F. curta in a core from Vestfold Hills representing Holocene climatic history before 3820 years BP and 5560 year BP indicates the prevalence of colder climatic conditions, as this species is known to prefer the icier condition to thrive. Abstract: The Vestfold Hills terrane in East Antarctica is a granulite facies terrane that preserves a record of sedimentation, magmatism and metamorphism that differs significantly from the other Archaean terranes exposed in East Antarctica. Sensitive High Resolution Ion Mass Spectrometry (SHRIMP) U–Pb isotopic data from detrital zircon cores, metamorphic rims and metamorphic zircon and monazite from the granulite facies Chelnok and Taynaya Paragneisses that demonstrate Archaean ages that are incompatible with the established ages of other East Antarctic source regions. The original sediments to the Chelnok and Taynaya Paragneisses were deposited between c. 2575 and c. 2520Ma and underwent a protracted period of high-grade granulite-facies metamorphism from c. 2520 to c. 2450Ma that is synchronous with the emplacement of the tonalitic Mossel Gneiss and the granitic to gabbroic rocks of the Crooked Lake Gneiss Group. The distinctive detrital zircon age populations and the period metamorphism recorded by the Vestfold Hills paragneisses lead us to speculate that: (1) these rocks were sourced from, and deposited on the margin of the crystalline basement to the North China Craton at the end of the Archaean, and (2) they underwent a period of high-grade metamorphism related to orogenesis driven by the collision of the North China Craton with the Archaean nucleus of South India during the late-Archaean to early-Paleoproterozoic. Viruses are abundant ubiquitous members of microbial communities and in the marine environment affect population structure and nutrient cycling by infecting and lysing primary producers. Antarctic lakes are microbially dominated ecosystems supporting truncated food webs in which viruses exert a major influence on the microbial loop. Here we report the discovery of a virophage (relative of the recently described Sputnik virophage) that preys on phycodnaviruses that infect prasinophytes (phototrophic algae). By performing metaproteogenomic analysis on samples from Organic Lake, a hypersaline meromictic lake in Antarctica, complete virophage and near-complete phycodnavirus genomes were obtained. By:introducing the virophage as an additional predator of a predator-prey dynamic model we determined that the virophage stimulates secondary production through the microbial loop by reducing overall mortality of the host and increasing the frequency of blooms during polar summer light periods. Virophages remained abundant in the lake 2 y later and were represented by populations with a high level of major capsid protein sequence variation (25-100% identity). Virophage signatures were also found in neighboring Ace Lake (in abundance) and in two tropical lakes (hypersaline and fresh), an estuary, and an ocean upwelling site. These findings indicate that virophages regulate host-virus interactions, influence overall carbon flux in Organic Lake, and play previously unrecognized roles in diverse aquatic ecosystems. Evidence is provided from a sediment core from saline Abraxas Lake, Vestfold Hills, that indicates that the lake existed through the Last Glacial Maximum. It can therefore be concluded that at least part of the Vestfold Hills also remained ice-free through the Last Glacial Maximum, or at most was covered by a thin, non-erosive cold-based ice sheet. The evidence for the continued existence of Abraxas Lake includes a 14C date that significantly predates the Last Glacial Maximum (though this cannot be considered direct proof of the existence of the lake prior to the Last Glacial Maximum); the presence of saline porewater throughout the core, including in compacted sediments deposited during the glacial period, which implies that the lake obtained its salt prior to any Holocene marine highstand; and the occurrence of marine-derived fauna from the onset of significant biological activity late in the Pleistocene. The occurrence of ice-free land in the Vestfold Hills and similar oases suggests that the margin of the polar ice cap did not reach far beyond its current position at the Last Glacial Maximum, at least in regions now occupied by these oases. Antarctic lakes with simple plankton ecosystems are believed to be sensitive biological indicators of climate change. Models of the physical environment, in particular the ice layer, support understanding of how the ecosystems respond to meteorological variables. This paper describes how data from a previously reported automatic measuring probe and meteorological data from Davis station were used to develop a detailed thermodynamic model of the ice layer on Crooked Lake, one of the largest and deepest freshwater lakes in Antarctica. The general model structure is similar to a previously reported model of sea ice but with modifications specific to the Antarctic freshwater lake case informed by the data. The model inputs are atmospheric variables as well as water temperature, ice albedo and the radiation extinction coefficient for the ice. Heat and radiation fluxes at the ice–air and ice–water boundaries are calculated using equations chosen for their suitability for the Antarctic. In the case of shortwave radiation, equations were fitted to data from the automatic probe. Using the heat fluxes to establish boundary conditions, and incorporating the known thermodynamic properties of ice, the temperature profile within the ice and the resulting growth and melt of the ice can be calculated. The model uses a largely mechanistic approach, with most equations taken from established thermodynamic theories or empirical studies and only one adjustable parameter related to the sensible heat flux from the water, which is not easily calculated from the available data. It was found to accurately reproduce ice temperature and ice thickness data for the year 2003, with r 2 =0.89, n =2005. Finally, the model was simplified to run with air temperature as the only input variable and was shown to perform well—this suggests that freshwater lake ice is affected more by air temperature than any other variable, and is therefore a useful indicator of climate change in its own right. Abstract: The sediment record of the fauna of Ace Lake, a saline meromictic lake in the Vestfold Hills, Antarctica, consists of copepod eggs, spermatophores and exoskeletal fragments, rotifer and tintinnid loricae, and foraminiferal and folliculinid tests. The relative abundance of these remains, along with other characteristics of the core, allows the development of a coherent picture of the progress of Ace Lake from a species-poor, freshwater lake early in the Holocene to a biodiverse marine basin following a marine transgression. Subsequent sea level fall reformed Ace Lake as a saline lake and productivity initially increased after isolation. After a major event, possibly associated with overturn of the meromictic lake, biodiversity and productivity decreased, and have continued to do so until the present. [Copyright &y& Elsevier]

Abstract: We present a relative sea-level (RSL) history, constrained by AMS radiocarbon-dated marine–freshwater transitions in isolation basins from a site adjacent to the Lambert Glacier, East Antarctica. The RSL data suggest an initial ice retreat between c. 15,370 and 12,660 cal yr B.P.. Within this period, meltwater pulse IA (mwp IA, between c. 14,600–14,200 and 14,100–13,700 cal yr B.P.) occurred; an exceptionally large ice melting event, inferred from far-field sea-level records. The RSL curve shows a pronounced highstand of approximately 8 m between c. 7570–7270 and 7250–6950 cal yr B.P. that is consistent with the timing of the RSL highstand in the nearby Vestfold Hills. This is followed by a fall in RSL to the present. In contrast to previous findings, the isolation of the lakes in the Larsemann Hills postdates the isolation of lakes with similar sill heights in the Vestfold Hills. An increase in RSL fall during the late Holocene may record a decline in the rate of isostatic uplift in the Larsemann Hills between c. 7250–6950 and 2847–2509 cal yr B.P., that occurred in response to a documented mid-Holocene glacier readvance followed by a late-Holocene retreat. The water chemistry of lake systems on the edge of the Antarctic continent responds quickly to changes in the moisture balance. This is expressed as increasing salinity and decreasing lake water level during dry periods, and the opposite during wet periods. The diatom composition of the lakes also changes with these fluctuations in salinity and lake water depth. This is important, as their siliceous remains become incorporated into lake sediments and can provide long-term records of past salinity using transfer functions. In order to develop transfer functions, diatoms and water chemistry data were inter-calibrated from five different East Antarctic oases, namely the Larsemann Hills, the Bølingen Islands, the Vestfold Hills, the Rauer Islands and the Windmill Islands. Results indicate that salinity is the most important environmental variable explaining the variance in the diatom flora in East Antarctic lakes. In oligo- saline lakes the variance is mainly explained by lake water depth. This dataset was used to construct a weighted averaging transfer function for salinity in order to infer historical changes in the moisture balance. This model has a jack-knifed r2 of 0.83 and a RMSEP of 0.31. The disadvantage of this transfer function is that salinity changes in oligo-saline lakes are reconstructed inaccurately due to the ‘edge effect’ and due to the low species turnover along the salinity gradient at its lower end. In order to infer changes in the moisture balance in these lakes, a second transfer function using weighted averaging partial least squares (with two components) for depth was constructed. This model has a jack-knifed r2 of 0.76 and a RMSEP of 0.22. Both transfer functions can be used to infer climate driven changes in the moisture balance in lake sediment cores from oligo-, hypo-, meso- and hyper-saline lakes in East Antarctic oases between 102–75°E. The transfer function for lake water depth is promising to track trends in the moisture balance of small freshwater lakes, where changes in shallow and deep-water sediments are readily reflected in changing diatom composition. Antarctic polar deserts can be of considerable scientific interest, but can also exhibit great environmental sensitivity. A variety of factors, including Australia’s legal obligations under the Madrid Protocol, public expectations, certain research opportunities and ethical considerations, demand a very high standard of environmental protection. A survey outside the Davis Station limits in the Vestfold Hills, East Antarctica, identified 66 sites at which past human activities had left long-term impacts on the physical environment. Nearly half of all observed impacts were the result of geoscientific research that had left old pit and trench sites, vehicular tracks, partly excavated palaeontological material, rock sampling and drilling sites, localised slope instability caused by disturbance, and discarded equipment and markers. Comparisons between rehabilitated sites and others where little, if any, rehabilitation appears to have been attempted suggest natural processes alone are generally insufficient to heal the damage, but that effective rehabilitation is often possible if undertaken immediately after the initial disturbance. An analysis of diatom assemblages, Total Organic Carbon (TOC), carbon-nitrogen ratio, d 13 C, biogenic silica and carbonate in two marine sediment cores from Ellis Fjord, eastern Antarctica, reveals episodes of climatic change at 3600, 2500, 2000 and 1000 yr BP. Using the diatom data, principal component analysis, the fast-ice index (FI), benthic index (BI) and snow index (SI) (which are derived from the diatom data), a period of reduced temporal sea-ice cover is identified prior to 2500 BP. There was a large increase in sea-ice extent between 2500 and 2000 BP followed by a smaller decline. Temporal sea-ice extent after 2000 BP was still significantly greater than prior to 2500 BP. Both cores were taken from anoxic basins within the fjord. It was found that the better preservation allowed the recognition of taxa such as Chaetoceros vegetative cells, which are not usually preserved. This enabled the distinction between ice-edge blooms, dominated by Fragilariopsis curta, and open-water blooms dominated by Chaetoceros. A high-resolution record of evaporation for the last |650 years was derived from the diatomsalinity signal preserved in a sediment core taken from Ace Lake, Vestfold Hills (68° 289S, 78° 119E), Antarctica. The seasonal oxygen isotope signal preserved in an ice core from Law Dome (66° 449S, 112° 509E), Antarctica, revealed a high-resolution summer temperature record for the same time period. The two proxies show highly correlated behaviour despite having significantly different climatic response mechanisms and a large geographic separation. The correlation observed between proxies based on such differing processes and analytical method-ologies provides not only a climate record for the past |650 years that is both robust and regionally representative of coastal East Antarctica but also confirmation of the utility of reconstructions using these methods. A comprehensive diatom stratigraphy is used to calculate a palaeosalinity history for an Antarctic lake via an established diatom-salinity transfer function for the Vestfold Hills, Antarctica. A sediment core taken from Ace Lake in 1995 shows three distinct changes in diatom assemblage constituents: initial benthic hyposaline – freshwater taxa are replaced by marine planktonic and sea-ice taxa with these taxa in turn replaced by the benthic hypersaline taxa dominant in the lake today. These changes in assemblage composition enable the lakewater salinity of each stage to be determined, and the Holocene evolution of the lake to be refined. Deglaciation of the Vestfold Hills at the beginning of the Holocene exposed Ace Lake basin; following this, fresh lacustrine diatoms were deposited from | 11 380 to | 8110 corrected 14 C yr BP. Relative sea-level rise after this time led to the progressive marine inundation of the lake and the deposition of marine diatom taxa. Marine taxa were dominant in the sediment for more than 6000 years.
