Field survey study was conducted season (2017). Soybeans and weeds were weekly sampled randomly. Thrips adults were identified and counted. Detection of the virus isolate and the natural incidence was determined using; Mechanical transmission, host range, DAS-ELISA, RT-PCR. The natural incidence thrips individuals was detected depending on the SVNV% in thrips individuals and weeds hosts. Ten thrips species were associated with soybean plants in the field. The most abundant species was T. tabaci, average 256.5 average no.of individuals, followed by F. occidentalis (142.5 average no. of individuals), then N. variabilis (86.6/ average no. of individuals). Fourteen thrips species occurred on 5 legumes field crops and 41 weed plant species within soybean field. The highest average number 40.6.of individuals were recorded on Ammi majus. While the lowest one 3.3 average no. of individuals were on Urtica urens. Only 21diagnostic plant species were susceptible to infection with SVNV. G. max and Vigna radiate, were the highest percentage of infection 80% followed by V. unguilata & N. benthamiana, 75%. Egyptian isolate of Soybean vein necrosis virus (SVNV) in this study showed a high degree of similarity and it is closely related to TSWV from Egypt (DQ479968) and TCSV from USA (KY820965) with nucleotide sequence identity of 78%. Four thrips species transmitted SVNV (F. fusca 4.0%, F. schultzei 4.3%, F. tritici 3.3% and N. variabilis 68.0% transmission). Both C. phaseoli and M. sjostedti can acquire the virus but unable to transmit it. The following species; T. tabaci, F. occidentalis, S. dorsallis and T. palmi cannot acquire or transmit SVNV. The incidence of SVNV in the field started by the end of July then increased gradualy from 12.7 to 71.3% by the end of the season. In conclusion, few thrips individuals invaded soybean crops are enough to transmit high rate of SVNV within the crop. Furthermore, several vector species are also abundant on weeds, which are the major sources of soybean viruses transmitted to the crops. This information might be important for control and reduce the incidence of SVNV infection.

Polyploidy is defined as a cell with three or more whole genome sets and enables cell growth across the kingdoms of life. Studies in model organisms have revealed that polyploid cell growth can be required for optimal tissue repair and regeneration. In mammals, polyploid cell growth contributes to repair of many tissues, including the liver, heart, kidney, bladder, and eye, and similar strategies have been identified in Drosophila and zebrafish tissues. This review discusses the heterogeneity and versatility of polyploidy in tissue repair and regeneration. Polyploidy has been shown to restore tissue mass and maintain organ size as well as protect against oncogenic insults and genotoxic stress. Polyploid cells can also serve as a reservoir for new diploid cells in regeneration. The numerous mechanisms to generate polyploid cells provide an unlimited resource for tissues to exploit to undergo repair or regeneration.

The present study was undertaken in the natural temperate Himalayan forests of Himachal Pradesh, India, to assess the tree species composition and diversity. For this purpose, six major forest types (FT) viz., FT1- Upper Himalayan Pinus roxburghii forest, FT2- Quercus leucotrichophora forest, FT3- Low-level P. wallichiana forest, FT4- Moist C. deodara forest, FT5- Western Mixed Coniferous Forest, FT6- Pinus gerardiana forest were selected. Detailed sampling was carried out in these forest types, and the sample plots in each forest type were laid out using the stratified random approach. Tree stem density varied from 191.11 N ha−1 (FT6) to 441.11 N ha−1 (FT2), whereas the tree total basal cover varied from 20.01 m2 ha−1 (FT6) to 47.59 m2 ha−1 (FT5). The diversity indices reflected that a total of 21 tree species (16 genera, 11 families) were identified, with tree species richness ranging from 3 (FT6) to 9 (FT1 & FT5). The forest type FT5 recorded the maximum Shannon index of diversity (2.36), Simpson Index of diversity (0.75), Margalef's Index of richness (1.37), pielou equitability (0.74), menheink index of species richness (0.49), whereas the highest Shannon Index of diversity (0.73) and species heterogeneity (0.85) in FT6 forest type. Furthermore, dominance-diversity (d-d) curves drawn that all the six forest types showed geometric curves reveals that one or two tree species are dominant in a particular forest type. Simultaneously, the research area's species diversity, tree stem density, and tree total basal cover were equivalent to those seen in other sections of Western Himalayas.

Increases in shrub growth and canopy cover are well documented community responses to climate warming in the Arctic. An important consequence of larger deciduous shrubs is shading of prostrate plant species, many of which are important sources of nectar and berries. Here, we present the impact of a shading experiment on two prostrate shrubs, Vaccinium vitis-idaea L. and Arctous alpina L., in northern Alaska over two growing seasons. We implemented three levels of shading (no shade, 40% shade, and 80% shade) in dry heath and moist acidic tundra. Plots were monitored for soil moisture content, surface temperature, normalized difference vegetation index (NDVI), and flowering. Shading was shown to, on average, lower surface temperature (0.7 °C to 5.3 °C) and increase soil moisture content (0.5% to 5.6%) in both communities. Both species- and plot-level NDVI values were delayed in timing of peak values (7 to 13 days) and decreased at the highest shading. Flower abundance of both species was lower in shaded plots and peak flowering was delayed (3 to 8 days) compared with controls. Changes in timing may result in phenological mismatches and can impact other trophic levels in the Arctic as both the flowers and resulting berries are important food sources for animals.

BackgroundIn the context of the SARS-CoV-2 pandemic, reuse of personal protective equipment, specifically that of medical face coverings, has been recommended. The reuse of these typically single-use only items necessitates procedures to inactivate contaminating human respiratory and gastrointestinal pathogens. We previously demonstrated decontamination of surgical masks and respirators contaminated with infectious SARS-CoV-2 and various animal coronaviruses via low concentration- and short exposure methylene blue photochemical treatment (10 µM methylene blue, 30 minutes of 12,500-lux red light or 50,000 lux white light exposure).MethodsHere, we describe the adaptation of this protocol to the decontamination of a more resistant, non-enveloped gastrointestinal virus and demonstrate efficient photodynamic inactivation of murine norovirus, a human norovirus surrogate.ResultsMethylene blue photochemical treatment (100 µM methylene blue, 30 minutes of 12,500-lux red light exposure) of murine norovirus-contaminated masks reduced infectious viral titres by over four orders of magnitude on surgical mask surfaces.Discussion and ConclusionsInactivation of a norovirus, the most difficult to inactivate of the respiratory and gastrointestinal human viruses, can predict the inactivation of any less resistant viral mask contaminant. The protocol developed here thus solidifies the position of methylene blue photochemical decontamination as an important tool in the package of practical pandemic preparedness.

The present study was conducted to evaluate the efficacy of phytogenic extract on growth and health of tilapia. Three experimental diets were designed as the basal diet (Con) and phytogenic extract (Flourishing, F) supplemented diets at 0.5 and 1 g/kg inclusion (F-1 and F-2), which the phytogenic additive was extracted from Eucommia ulmoides, Astragalus membranaceus, Lonicera japonica and Codonopsis pilosula. The three diets were fed to juvenile tilapia (14.0 ± 0.1 g) with 30 fish per cage, and three replicate cages from the three groups were placed in one indoor concrete pool (a total of three pools were used). Understanding the molecular genetic basis of animal magnet reception has been one of the big challenges in biology. Recently it was discovered that the magnetic sense of Drosophila melanogaster is mediated by the ultraviolet (UV)-A/blue light photoreceptor cryptochrome (Cry).Here, using the fruit fly as a magnet receptive model organism, we show that exposure to a specific AC magnetic field during night time affected the health of the fly. AC magnetic field exposure showed lifespan extension under starvation, sleep improvement and prevention of decreased motor function. In contrast, all the health improvement effect was not observed in cryptochrome mutant flies (cryb). We showed that AC magnetic field exposure prevented motor dysfunction in Gaucher’s disease model Drosophila. The data suggests that magnetic field-dependent improvement of sleep quality, lifespan and motor function is mediated through a cry-dependent pathway in animals.

There are several natural and anthropomorphic environments where iron- and/or sulfur-oxidizing bacteria thrive in extremely acidic conditions. These acidophilic chemolithautotrophs play important roles in biogeochemical iron and sulfur cycles, are critical catalysts for industrial metal bioleaching operations, and have underexplored potential in future biotechnological applications. However, their unique growth conditions complicate the development of genetic techniques. Over the past few decades genetic tools have been successfully developed for Acidithiobacillus ferrooxidans, which serves as a model organism that exhibits both iron- and sulfur-oxidizing capabilities. Conjugal transfer of plasmids has enabled gene overexpression, gene knockouts, and some preliminary metabolic engineering. We highlight the development of genetic systems and recent genetic engineering of A. ferrooxidans, and discuss future perspectives.

The state of Paraíba in northeast Brazil contains four of the seven biomes present in the country: Mata Atlântica, Cerrado, Caatinga and Matas Serranas. On the other hand, Amazônia, Pantanaland Pampa were not found in this area. This special situation allows us to analyse changes in the distribution of these four large Brazilian biomes according to bioclimatic conditions, using the methodology of bioclimatic regime types. Based on the analysis of variables from periods of hydric and thermal vegetation stagnation, obtained from hydric and bioclimatic balances, average monthly temperature and rainfall, that methodology enables us to establish a typology of 27 types of bioclimatic regimes and 243 bioclimatic regime subtypes with the 9 Thornthwaite ombrothermal levels. In Paraíba 4 types of bioclimatic regimes are currently identified (mesophyllo, tropophyllo, xerophyllo and eurythermophilous) and 9 subtypes according to ombrothermal levels. In order to analyse the changes, extreme change situations were chosen: a past scenario with the Last Glacial Maximum (40 ky); and an RCP 8.5 climate change scenario for the CMSS 4.0 model for the year 2070. This enabled 3 bioclimatic regime maps of each of the 3 aforementioned situations to be obtained, providing a map of potential distribution of the plant formations of Paraíba state according to the specific field knowledge and bioclimatic mapping obtained for the present. This paper concludes that a retrocession of the Mata Atlântica can be seen from the Last Glacial Maximum up to the present, losing its optimal bioclimatic conditions and therefore remaining in a highly fragile relict situation in the face of anthropic pressure (sugarcane cultivation and urban expansion); an advance toward 2070 of the Caatinga in its shrub form as a predominant formation is indicated by the projection of climate change in 2070 for the analysed situation, specifically resulting from anthropic pressure, in this case due to livestock activities which have affected this biome in Paraíba since the mid-19th century. El estado de Paraiba en el nordeste del país, recoge cuatro de los siete biomas presentes en Brasil: la Mata Atlántica, el Cerrado, la Caatinga y las Matas Serranas.Por otro lado, Amazonia, Pantanal y Pampa no se encontraron en esta área. Esta situación especial nos permite hacer un análisis de los cambios en la distribución de estos cuatro grandes biomas brasileños en función de las condiciones bioclimáticas con el uso de la metodología de tipos regímenes bioclimáticos. Dicha metodología, a partir del análisis de las variables de los periodos de paralización vegetativa hídrica y térmica, obtenidos de los balances hídricos y bioclimáticos, la precipitación y la temperatura media mensual, nos permite establecer una tipología de 27 tipos de regímenes bioclimáticos y 243 subtipos de regímenes bioclimáticos con los 9 niveles ombrotérmicos de Thornthwaite. Se identifican actualmente para Paraiba 4 tipos de regímenes bioclimáticos (mesophyllo, tropophyllo, xerophyllo y euritermophyllo) y 9 subtipos en función de los niveles ombrotérmicos. Para poder hacer un análisis de cambios se han elegido situaciones extremas de cambio: por un lado, un escenario del pasado con el Ultimo Máximo Glaciar (40 ky), y por otro un escenario de cambio climático RCP 8.5 para el modelo CMSS 4.0 para el año 2070. Se obtienen así 3 cartografías de regímenes bioclimáticos de cada una de las tres situaciones citadas y se aporta una cartografía de la distribución potencial de las formaciones vegetales del estado de Paraiba en función de los propios conocimientos de campo y de la cartografía bioclimática que se ha obtenido para la actualidad. Como conclusión de este trabajo se puede ver un retroceso de la Mata Atlántica desde el Ultimo máximo Glaciar hasta la actualidad, perdiendo son condiciones bioclimáticas óptimas y quedando por lo tanto en una situación relicta de alta fragilidad ante la presión antrópica (cultivos de caña de azúcar y expansión urbana) y un avance hacia el 2070 de la caatinga en su forma arbustiva como formación predominante según marca la proyección del cambio climático en el 2070 para el escenario analizado, y como resultado de la propio acción antrópica, en este caso con actividades ganaderas, que se viene ejerciendo sobre este bioma en Paraiba desde mediados del siglo XIX.

Musculoskeletal injuries are common in humans. The cascade of cellular and molecular events following such injuries results either in healing with functional recovery or scar formation. While fibrotic scar tissue serves to bridge between injured planes, it undermines functional integrity. Hence, faithful regeneration is the most desired outcome; however, the potential to regenerate is limited in humans. In contrast, various non-mammalian vertebrates have fascinating capabilities of regenerating even an entire appendage following amputation. Among them, zebrafish is an important and accessible laboratory model organism, sharing striking similarities with mammalian embryonic musculoskeletal development. Moreover, clinically relevant muscle and skeletal injury zebrafish models recapitulate mammalian regeneration. Upon muscle injury, quiescent stem cells - known as satellite cells - become activated, proliferate, differentiate and fuse to form new myofibres, while bone fracture results in a phased response involving hematoma formation, inflammation, fibrocartilaginous callus formation, bony callus formation and remodelling. These models are well suited to testing gene- or pharmaco-therapy for the benefit of conditions like muscle tears and fractures. Insights from further studies on whole body part regeneration, a hallmark of the zebrafish model, have the potential to complement regenerative strategies to achieve faster and desired healing following injuries without any scar formation and, in the longer run, drive progress towards the realisation of large-scale regeneration in mammals. Here, we provide an overview of the basic mechanisms of musculoskeletal regeneration, highlight the key features of zebrafish as a regenerative model and outline the relevant studies that have contributed to the advancement of this field. Aims This study examined the effects of water activity (aw), ammonia and Corynebacterium urealyticum on the survival of Salmonella Typhimurium in sterile poultry litter. Methods and Results Sterile poultry litter inoculated with S. Typhimurium was adjusted to pH 9.0, various aw levels (0.84, 0.92 and 0.96), and total ammonia nitrogen levels were increased either by the addition of ammonium sulphate or C. urealyticum inoculation with 1% urea added. All litter treatments were incubated at 30°C and sampled daily for five days. Similar results were observed at each aw level in both experiments. At 0.84 and 0.92 aw, S. Typhimurium populations in litter fell below 1 log CFU g−1 within 5 days, with no significant differences between the controls and increased ammonia treatments. At 0.96 aw, Salmonella populations treated with increased ammonia levels were significantly lower than control treatments on days 1–5. Conclusions This study showed that C. urealyticum can produce ammonia in litter at higher aw levels with sufficient available urea and that the antimicrobial efficacy of ammonia is dependent on high aw (~0.96) in litter. Significance and Impact of the Study These results provide insights into the production of ammonia in litter, its antimicrobial efficacy in litter and the importance of aw in this interaction.

Murray-Darling Freshwater Research CentreMDFRC item.Many floodplain wetlands in south-eastern Australia have become isolated from the main river channel as a consequence of reduced high flows and associated flood events following river regulation. In the Central Murray region of south-eastern Australia, many temporary wetlands would have received water once every five years or so, with large floods maintaining floodplain connectivity every decade, under natural conditions. Now, the River Murray is highly regulated and many of these wetland areas have not been flooded for periods of up to 30 years. Consequently, these wetlands are becoming degraded and the biodiversity of the area is in decline. From 2001–2003, 21 Black Box depression wetlands in the Central Murray region were each watered once. Plant communities in each wetland were monitored for changes in abundance (assessed as percentage cover) before and during the wetting and drying phases. Wetlands were watered during spring or early summer with the length of inundation ranging from 6 to 19 weeks. After watering, the percentage cover of native plant taxa and native plant functional groups in most wetlands increased. In general, there was a decrease in the percentage number of terrestrial plants present and an increase in the percentage cover of aquatic plants. Introduced species were a minor component. Although these wetlands are all located in the Central Murray region, individual wetlands developed plant communities that contained taxa specific to individual wetlands despite initial similarities. These results indicate that wetland plant biodiversity within the landscape can be promoted and maintained by ensuring there is a diversity of wetlands with varying flood regimes within the landscape.

Abiotic stresses negatively impact ecosystems and the yield of crops, and climate change will increase their frequency and intensity. Despite progress in understanding how plants respond to individual stresses, our knowledge of plant acclimatization to combined stresses typically occurring in nature is still lacking. Here, we used a plant with minimal regulatory network redundancy, Marchantia polymorpha, to study how seven abiotic stresses, alone and in 19 pairwise combinations, affect the phenotype, gene expression, and activity of cellular pathways. While the transcriptomic responses show a conserved differential gene expression between Arabidopsis and Marchantia, we also observe a strong functional and transcriptional divergence between the two species. The reconstructed high-confidence gene regulatory network demonstrates that the response to specific stresses dominates those of others by relying on a large ensemble of transcription factors. We also show that a regression model could accurately predict the gene expression under combined stresses, indicating that Marchantia performs arithmetic multiplication to respond to multiple stresses. Lastly, two online resources (https://conekt.plant.tools and http://bar.utoronto.ca/efp_marchantia/cgi-bin/efpWeb.cgi) are provided to facilitate the study of gene expression in Marchantia exposed to abiotic stresses.

Rosmarinus tournefortii is an aromatic evergreen shrub endemic to North Africa, including Algeria. Reports on phytochemicals of this species and related biological activities remain scarce. The essential oils (EOs) and ethanol extracts (EEs) of five populations collected in Algeria are investigated for their antioxidant and antimicrobial properties through different multivariate statistical methods. EOs analysis by GC and GC-MS showed that the main constituents (α-pinene, camphor, camphene and limonene) were almost comparable. The antioxidant capacity was evaluated by the ferric reducing power and the inhibition of free radicals DPPH● and ABTS●+. The free radical scavenging effectiveness of EEs was about 2 times higher than the standard antioxidant (BHT). EOs exhibited weaker antioxidant activity insofar they failed to scavenge 50% radicals. The antimicrobial activity evaluated by determining the minimum inhibitory/bactericidal concentrations towards fifteen multi-drug resistant microorganisms showed that EEs were the most active. The Gram-positive bacteria were the most susceptible. Both EEs and EOs exhibited antifungal and anti-yeast activities.

La mayoría de las especies de plantas introducidas en México no han sido evaluadas para priorizar las acciones de gestión para erradicarlas o contenerlas o para mitigar los daños causados por invasiones de plantas nocivas. Se evaluaron 42 taxa (41 especies y una variedad) de malezas introducidas de la familia Leguminosae usando criterios empíricos y derivados de teoría, en el desarrollo de un modelo centrado en el comportamiento de las especies en el extranjero, su tiempo de residencia y las localidades ocupadas, y la presencia en México de los parientes congenéricos nativos y no nativos. Los datos se obtuvieron a partir de fuentes bibliográficas y de la base de datos “Malezas Introducidas en México” que incluye información de 11 herbarios de México. También desarrollamos un proceso de puntuación para calificar la expansión de las introducidas de acuerdo con tiempo de residencia y número de localidades ocupadas. Clasificamos a las leguminosas introducidas en cuatro categorías de prioridad de atención. Albizia lebbeck, Pueraria phaseoloides, Lablab purpureus, Securigera varia, Delonix regia, Clitoria ternatea y Spartium junceum deben recibir prioridad de atención alta; consideramos que ocho especies requieren una atención prioritaria media, y siete atención prioritaria baja. 20 taxa fueron clasificados como no prioritarios. El modelo de evaluación desarrollado todavía necesita un mayor refinamiento, ya que especies aparentemente inocuas obtuvieron puntajes altos y una potencialmente peligrosa (Cassia fistula) se clasificó en la categoría no prioritaria. Esperamos que el presente modelo de evaluación funcionará como una herramienta y con baja dependencia de expertos, para identificar a las especies introducidas que requieren un análisis de riesgo más completo para priorizar esfuerzos para el manejo de las plantas introducidas nocivas.

The yeast Saccharomyces cerevisiae is widely used as a cell factory and as an important eukaryal model organism for studying cellular physiology related to human health and disease. Yeast was also the first eukaryal organism for which a genome-scale metabolic model (GEM) was developed. In recent years there has been interest in expanding the modeling framework for yeast by incorporating enzymatic parameters and other heterogeneous cellular networks to obtain a more comprehensive description of cellular physiology. We review the latest developments in multiscale models of yeast, and illustrate how a new generation of multiscale models could significantly enhance the predictive performance and expand the applications of classical GEMs in cell factory design and basic studies of yeast physiology.

In mangrove forests, soil salinity is one of the most significant environmental factors determining mangrove forest distribution and productivity as it limits plant water uptake and carbon gain. However, salinity control on mangrove productivity through plant hydraulics has not been investigated by existing mangrove models. Thus, we present a new individual-based model linked with plant hydraulics to incorporate physiological characterization of mangrove growth under salt stress. Plant hydraulics was associated with mangroves nutrient uptake and biomass allocation apart from water flux and carbon gain. The developed model was performed for two-coexisting species of Rhizophora stylosa and Bruguiera gymnorrhiza in a subtropical mangrove forest in Japan. The model predicted that the productivity of both species was affected by soil salinity through downregulation of stomatal conductance, while B. gymnorrhiza trees grow faster and suppress the growth of R. stylosa trees by shading that resulted in a B. gymnorrhiza-dominated forest under low soil salinity conditions (B. gymnorrhiza compared to R. stylosa, leading to an increase in biomass of R. stylosa despite the enhanced salt stress (> 30 ‰). These predicted patterns in forest structures across soil salinity gradient remarkably agreed with field data, highlighting the control of salinity on productivity and tree competition as factors that shape the mangrove forest structures. The model reproducibility of forest structures was also supported by the predicted self-thinning processes, which likewise agreed with field data. In addition, the mangroves morphological adjustment to increasing soil salinity – by decreasing transpiration and increasing hydraulic conductance – was reasonably predicted. Aside from the soil salinity, seasonal dynamics in atmospheric variables (solar radiation and temperature) was highlighted as factors influencing mangrove productivity in a subtropical region. The physiological principle-based improved model has the potential to be extended to other mangrove forests in various environmental settings, thus contributing to a better understanding of mangrove dynamics under future global climate change.

Background Fusobacterium species are obligately anaerobic, gram-negative bacilli. Especially, F. nucleatum and F. necrophorum are highly relevant human pathogens. We investigated clinical differences in patients infected with Fusobacterium spp. and determined the antimicrobial susceptibility of Fusobacterium isolates. Methods We collected clinical data of 86 patients from whom Fusobacterium spp. were isolated from clinical specimens at a tertiary-care hospital in Korea between 2003 and 2020. In total, 76 non-duplicated Fusobacterium isolates were selected for antimicrobial susceptibility testing by the agar dilution method, according to the Clinical and Laboratory Standards Institute guidelines (M11-A9). Results F. nucleatum was most frequently isolated from blood cultures and was associated with hematologic malignancy, whereas F. necrophorum was mostly prevalent in head and neck infections. Anti-anaerobic agents were more commonly used to treat F. nucleatum and F. varium infections than to treat F. necrophorum infections. We observed no significant difference in mortality between patients infected with these species. All F. nucleatum and F. necrophorum isolates were susceptible to the antimicrobial agents tested. F. varium was resistant to clindamycin (48%) and moxifloxacin (24%), and F. mortiferum was resistant to penicillin G (22%) and ceftriaxone (67%). β-Lactamase activity was not detected. Conclusions Despite the clinical differences among patients with clinically important Fusobacterium infections, there was no significant difference in the mortality rates. Some Fusobacterium spp. were resistant to penicillin G, ceftriaxone, clindamycin, or moxifloxacin. This study may provide clinically relevant data for implementing empirical treatment against Fusobacterium infections.

Introduction Honey bees (Apis mellifera) play key roles in food production performing complex behaviors, like self-grooming to remove parasites. However, the lipids of their central nervous system have not been examined, even though they likely play a crucial role in the performance of cognitive process to perform intricate behaviors. Lipidomics has greatly advanced our understanding of neuropathologies in mammals and could provide the same for honey bees. Objectives The objectives of this study were to characterize the brain lipidome of adult honey bees and to assess the effect of clothianidin (a neurotoxic insecticide) on the brain lipid composition, gene expression, and performance of self-grooming behavior under controlled conditions (cage experiments). Methods After seven days of exposure to oral sublethal doses of clothianidin, the bees were assessed for self-grooming behavior; their brains were dissected to analyze the lipidome using an untargeted lipidomics approach and to perform a high throughput RNAseq analysis. Results Compared to all other organisms, healthy bee brain lipidomes contain unusually high levels of alkyl-ether linked (plasmanyl) phospholipids (51.42%) and low levels of plasmalogens (plasmenyl phospholipids; 3.46%). This could make it more susceptible to the effects of toxins in the environment. A positive correlation between CL 18:3/18:1/14:0/22:6, TG 6:0/11:2/18:1, LPE 18:0e and intense self-grooming was found. Sublethal doses of a neonicotinoid altered PC 20:3e/15:0, PC 16:0/18:3, PA 18:0/24:1, and TG 18:1/18:1/18/1 levels, and affected gene expression linked to GPI-anchor biosynthesis pathway and energy metabolism that may be partially responsible for the altered lipid composition. Conclusion This study showed that lipidomics can reveal honey bee neuropathologies associated with reduced grooming behavior due to sublethal neonicotinoid exposure. The ease of use, unusual brain lipidome as well as characterized behaviors that are affected by the environment make honey bees a promising model organism for studying the neurolipidome and associations with neurobehavioral disorders.

Background Even though some Dracaena species are used as medicine, their utilization as food is rather unusual. In northern Angola, however, leaves and underground tubers of Dracaena camerooniana Baker are frequently consumed. In particular, the leaves are of increasing economic value in the region. But the anatomy and nutritional aspects of the plant have not been studied so far. Therefore, a detailed anatomic description of the plant was conducted. In a second step the nutritional value of the defined material was analysed, providing a basis for the discussion of a more intensive utilisation of this rainforest shrub. Results The leaf anatomy of Dracaena camerooniana differs from the species of the genus that have been examined so far, in showing adaptations to wet climatic conditions. The stems produce a secondary thickening meristem with amphivasal secondary vascular bundles. During tuber formation, the parenchyma of the pith in the vascular cylinder of the root markedly increases. From a nutritional point of view, leaves do not noteworthy contribute to vitamin intake. The nutritional composition of the tubers is comparable to that of cassava (Manihot esculenta Crantz ) and potatoes (Solanum tuberosum L.). However, a high fructose value indicates inulin as a storage carbohydrate. Conclusion Inspite of its rather moderate nutritional values, Dracaena camerooniana is an indigenous addition to the diet of the Angolan population that could also be cultivated in the future due to the easy vegetative propagation.

According to previous research, the incidence of necrotising enterocolitis (NEC) decreases after supplementation with probiotics. However, few studies have considered the equivalence or otherwise of different strains of probiotics in this respect. Accordingly, this prospective observational study was conducted in a cohort of 245 very-low-birth-weight (VLBW) new-borns to assess the prevalence of NEC after supplementation with the probiotic Inforan® (Berna Biotech, Madrid, Spain) 250 mg capsules containing 109 cfu of Lactobacillus acidophilus (ATCC 4356) and 109 cfu of Bifidobacterium bifidum (ATCC 15696); or with Bivos® (Ferring, Madrid, Spain) containing Lacticaseibacillus (formerly Lactobacillus) rhamnnosus (LGG) (ATCC 53103) (109 cfu); or with no probiotic supplementation. Statistical analysis was performed using multivariant regression for the duration of parenteral nutrition, length of neonatal intensive care unit stay, use of oxygen therapy and presence of chorioamnionitis. Of the VLBW new-borns in the study group, 65 received Infloran, 108 received Bivos and 72 received no probiotic. A significant association was observed between a reduced presence of NEC Stage ≥2 and probiotic supplementation. The odds risk (OR) obtained was 0.174 (95% confidence interval (CI): 0.032-0.936) for Infloran and 0.196 (95%CI: 0.053-0.732) for Bivos. Therefore, both probiotics are associated with a lower prevalence of NEC in VLBW new-borns, with no significant differences.

The Kingdom of Saudi Arabia thrives with great plant diversity, including rare plants of the family Asphodelaceae that have multiple benefits and are still being studied. Aloe shadensis is one of these plants that must be preserved and documented in its natural environment. The most appropriate molecular approach currently approved for documentation is the sequencing of some genomic markers. The current study is the first to use genomic markers to record this rare plant. In this study, the plastid genes matK (Maturase K), rbcL (Ribulose-bisphosphate carboxylase/oxygenase large subunit), and the nuclear region ITS (Internal transcribed spacer) were used to reveal their efficiency in identifying the plant under study. This study is the first to deal with this plant and document it using these genetic markers. The study showed a promising result concerning identifying the sequence of the matK gene and ITS region, while the rbcL gene did not give a good indicator through the used primers. The obtained sequences of the matK gene and the ITS region were determined through two different sets of primers in each case then deposited in GenBank. The evolutionary relatedness of Aloe shadensis was established with the different species of Aloe. The study showed that the closest species is Aloe vera with a similarity of more than 99 %. The study concludes with the possibility of using these genes to correctly identify, distinguish and document the species of Aloe shadensis.

External organic carbon sources are needed to provide electron donors for the denitrification of wastewater with a low COD/NO3−-N (C/N) ratio, increasing the treatment cost. The economic strategy is to enhance the bioactivity and/or biodiversity of denitrifiers to efficiently utilize organic substances in wastewater. In this study, novel zero-valent iron (ZVI) composite carriers were prepared and implemented in a suspended carrier biofilm reactor to enhance the bioactivity and/or biodiversity of denitrifiers. At the influent C/N ratio of 4 (COD was 179.5 ± 5.0 mg/L and TN was 44.2 ± 0.8 mg/L), COD and TN removal efficiencies were 85.1% and 66.4%, respectively, in the reactors filled with 3 wt% ZVI composite carriers. In contrast, COD and TN removal efficiencies were 70.4% and 55.3%, respectively, in the reactor filled with conventional high-density polyethylene (HDPE) biofilm carriers. The biofilm formation on the 3 wt% ZVI composite carriers was optimized due to its higher roughness (surface square roughness increased from 76.0 nm to 93.8 nm) and favorable hydrophilicity (water contact angle dropped to 72.5° ± 1.4° from 94.3° ± 3.2°) compared with the HDPE biofilm carriers. In addition, heterotrophic denitrifiers, Thauera and Dechloromonas, were enriched, whereas autotrophic denitrifiers, Raoultella and Thiobacillus, exhibited high relative abundance in the biofilm of ZVI composite carriers. The coexistence of heterotrophic denitrifiers and autotrophic denitrifiers on the surface of ZVI composite carriers provided mixotrophic metabolism of denitrification (including heterotrophic and iron-based autotrophic), thereby ensuring effective denitrification for wastewater with a low C/N ratio without external organic carbon source addition.

Saffron (Crocus sativus L.) is the low yielding plant of medicinal and economic importance. Therefore, it is of interest to report the draft genome sequence of C. sativus. The draft genome of C. sativus has been assembled using Illumina sequencing and is 3.01 Gb long covering 84.24% of genome. C. sativus genome annotation identified 53,546 functional genes (including 5726 transcription factors), 862,275 repeats and 964,231 SSR markers. The genes involved in the apocarotenoids biosynthesis pathway (crocin, crocetin, picrocrocin, and safranal) were found in the draft genome analysis.

Some insects cause high losses of the common bean during storage, namely, Acanthoscelides obtectus Say. Chemical insecticides are commonly used to control insects, but their overuse adversely affects the environment and consumers' health. There is, therefore, the need for an alternative control method. This study was aimed to evaluate the chemical characterization and insecticidal property of Moringa oleifera seed extract on the developmental stages of A. obtectus in stored beans. Three types of M. oleifera seed extracts, namely methanol, ethanol and acetone extracts, were used at doses of 12.5, 25 and 50 µl/ml each. HPLC-MS was used to characterize these extracts. The toxicity of extracts against the adults, the number of eggs and the viability rate of laid or emerged eggs of A. obtectus were evaluated. The results show that M. oleifera seed extract is rich in Hesperidin, Quinic acid, gallic acid, protocatechuic acid, 4-OH benzoic acid, cynaroside, isoquercitrin, cosmosiin, quercitrin, luteolin, naringenin, salicylic acid, Apigenin and Fumaric acid. The acetone extract, from day 2, showed a 100% mortality rate in all doses compared to about only 14% in the control treatment. The number of eggs laid in control (236.67) was higher than the null (0) in acetone treatment at 50 µl/ml. M. oleifera seed extracts contain chemical molecules. They significantly reduced the damage caused by A. obtectus on stored P. vulgaris grains. Therefore, they can be used as an alternative to chemicals for the protection of stored foodstuffs.

l -ascorbic acid 2-O-α- d -glucoside (AA-2G), a highly stabilized vitamin C derivative, has been widely applied in cosmetics, foods, and pharmaceuticals. Sucrose phosphorylase (SPase) was recently reported to be effective for producing AA-2G. In this study, SPase from Bifidobacterium breve (BbrSPase) was identified as a new AA-glycosylating tool with high activity and good thermostability. The optimal conditions for AA glycosylation are pH 5.5 and 55 °C. The K m and k c a t when sucrose was applied as a glycosyl donor were 2.72 ± 0.49 mM and 9.70 ± 0.10 s−1, respectively. The transglycosylation activity was seriously inhibited by fructose, which caused a sharp decline in the glycosylation rate but did not necessarily stop the yield of AA-2G due to the high affinity to sucrose. Therefore, to attenuate the inhibition of fructose, a new sucrose batch-feeding strategy was developed for the effective production of AA-2G. The maximum titer was 185 g/L within 72 h. The molar conversion rate of l -AA reached 50.2 %, which is the highest conversion rate to our knowledge.

Pathogens can affect physiological and immunological reactions in immunocompromised animals and genetically engineered mice. Specifically, murine norovirus (MNV), Helicobacter, and intestinal protozoa are prevalent in rodent laboratory facilities worldwide. In this study, microbiological test results of the soiled bedding of sentinel mice showed the prevalence of MNV (50.9%, 28/55), Helicobacter hepaticus (29.1%, 16/55), Trichomonas spp. (14.5%, 8/55), and Entamoeba spp. (32.7%, 18/55). No single infections were detected as all cases were confirmed to have complex infections with two or four pathogens. In previous studies, the success rate of the cross-fostering method was not perfect; therefore, in this study, the entire mouse strain of the SPF rodent facility was rederived using embryo transfer. For up to three years, we confirmed that the results were negative with regular health surveillance tests. Embryo transfer was, thus, determined to be an effective method for the rederivation of specific pathogen free (SPF) barrier mouse facilities. This is the report for the effectiveness of embryo transfer as an example of successful microbiological clean-up of a mouse colony with multiple infections in an entire SPF mouse facility and embryo transfer may be useful for rederiving.

Background There are limited data regarding the risk factors for hyperglycemia in extremely low birth weight infants (ELBWIs). The aim of this observational study was to investigate the incidence of hyperglycemia among ELBWIs during the first 14 days of life and identify independent risk factors for hyperglycemia development. Methods We retrospectively evaluated 55 ELBWIs (32 male infants) between January 2015 and March 2020. Hyperglycemia was diagnosed when the glucose level was ≥180 mg/dL. Demographic and clinical data were extracted from the patients’ medical records. The risk factors associated with the onset of hyperglycemia were identified by Cox proportional hazards regression analysis with variables that had previously been identified as risk factors for hyperglycemia. Results Hyperglycemia developed in 23 patients (41.8%) within the first 14 days of life. Gestational age, chorioamnionitis, postnatal intravenous glucocorticoids, and probiotic type were included in the analysis. The results indicated that hyperglycemia was significantly associated with gestational age (hazard ratio [HR], 0.65; 95% confidence interval [CI], 0.48–0.87; P = 0.004). Further, Bifidobacterium breve (B. breve M-16V) use was related to hyperglycemia in ELBWIs (HR, 2.95; 95% CI, 1.10–7.87; P = 0.031). Conclusion Hyperglycemia was strongly associated with lower gestational age and B. breve M-16V use in our study population. Although probiotic supplementation may be beneficial for preterm infants to reduce the incidence of necrotizing enterocolitis, the dextrin used as an excipient in B. breve M-16V may lead to an undesirable carbohydrate load in ELBWIs.

OBJECTIVE To compare the bacteriome of the oral cavity in healthy dogs and dogs with various stages of periodontal disease. ANIMALS Dogs without periodontal disease (n = 12) or with mild (10), moderate (19), or severe (10) periodontal disease. PROCEDURES The maxillary arcade of each dog was sampled with a sterile swab, and swabs were submitted for next-generation DNA sequencing targeting the V1–V3 region of the 16S rRNA gene. RESULTS 714 bacterial species from 177 families were identified. The 3 most frequently found bacterial species were Actinomyces sp (48/51 samples), Porphyromonas cangingivalis (47/51 samples), and a Campylobacter sp (48/51 samples). The most abundant species were P cangingivalis, Porphyromonas gulae, and an undefined Porphyromonas sp. Porphyromonas cangingivalis and Campylobacter sp were part of the core microbiome shared among the 4 groups, and P gulae, which was significantly enriched in dogs with severe periodontal disease, was part of the core microbiome shared between all groups except dogs without periodontal disease. Christensenellaceae sp, Bacteroidales sp, Family XIII sp, Methanobrevibacter oralis, Peptostreptococcus canis, and Tannerella sp formed a unique core microbiome in dogs with severe periodontal disease. CONCLUSIONS AND CLINICAL RELEVANCE Results highlighted that in dogs, potential pathogens can be common members of the oral cavity bacteriome in the absence of disease, and changes in the relative abundance of certain members of the bacteriome can be associated with severity of periodontal disease. Future studies may aim to determine whether these changes are the cause or result of periodontal disease or the host immune response.

Pathogens that attack invasive plants can positively affect the integrity and functioning of ecosystems. Stem-tip dieback and extensive wilting followed by sudden death have been observed in Chrysanthemoides monilifera subsp. rotundata (bitou bush), one of Australia’s worst invasive shrubs. Metabarcoding and culturing methods were used to investigate whether fungi are implicated in this syndrome. Metabarcoding results revealed significantly different endophytic fungal communities within healthy and diseased bitou bush, and colocated native plants. There was no difference in fungal communities between soil sampled in the root zone of healthy and diseased bitou bush at the same site. Two Diaporthe sp. operational taxonomic units (OTUs), dominant at sites with extensive wilting explained 30% of the similarity between diseased bitou bush across all sites. Two other OTUs, Austropleospora osteospermi and Coprinellus sp., explained 20 and 40% of the similarity between diseased plants, respectively, and were only dominant at sites with dead or stunted, partially defoliated but not wilted bitou bush. A Penicillium sp. OTU explained 90% of the similarity between healthy bitou bush plants. Various Diaporthe spp. dominated isolations from diseased bitou bush. Manipulative experiments confirmed Diaporthe spp. pathogenicity on bitou bush excised and in situ stems. In another experiment, Diaporthe masirevicii infected flowers and, from there, colonized stems endophytically; however, wilting and sudden death of bitou bush did not occur within the experimental timeframe. Our study provides circumstantial evidence that bitou bush sudden death syndrome is the result of a shift in the composition of its endophytic fungal community from mutualist to pathogenic species. Intracellular components (including organelles and biomolecules) at the submicron level are typically analyzed in situ by special preparation or expensive setups. Here, a label-free and cost-effective approach of screening microalgal single-cells at a subcellular resolution is available based on impedance cytometry. To the best of our knowledge, it is the first time to show the relationships between impedance signals and submicron intracellular organelles and biomolecules. Experiments were performed on Euglena gracilis (E. gracilis) cells incubated under different incubation conditions (i.e., aerobic and anaerobic) and 15 μm polystyrene beads (reference) at two distinct stimulation frequencies (i.e., 500 kHz and 6 MHz). Based on the impedance detection of tens of thousands of samples at a throughput of 900 cells/s, three metrics were used to track the changes in biophyscial properties of samples. As a result, the electrical diameters of cells showed a clear shrinkage in cell volume and intracellular components, as observed under microscope. The morphology metric of impedance pulses (i.e., tilt index) successfully characterized the changes in cell shape and intracellular composition distribution. Besides, the electrical opacity showed the stable ratio of the intracellular components to cell volume under the cellular self-regulation. Additionally, simulations were used to support these findings and to elucidate how submicron intracellular components and cell morphology affect impedance signals, providing a basis for future improvements. This work opens up a label-free and high-throughput way to analyze single-cell intracellular components by impedance cytometry.

The invasive shrub Scotch broom (Cytisus scoparius (L.) Link) is a pervasive threat to regenerating Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco var. menziesii) stands in the Pacific Northwest, USA. Field observations indicate that the susceptibility of areas to Scotch broom invasion and dominance can vary by site. We selected 10 sites throughout the western Pacific Northwest that spanned a gradient of soil textures and other factors to test the site-specific susceptibility of Douglas-fir to overtopping by Scotch broom. We expected to find that the ability of Scotch broom to dominate a site was mediated by site-level factors, particularly those influencing soil water - the most limiting factor to growth in the region. We found Scotch broom and Douglas-fir were inversely affected by site-level factors. In general, Douglas-fir absolute height growth rates were more competitive with those of Scotch broom on fine-textured soils than on more coarsely textured soils. We also found Douglas-fir to have a more dramatic response to increasing down woody material than Scotch broom. Scotch broom height growth approached an asymptote at 3 m. Sites with fast-growing Douglas-fir were able to surpass this height 6-7 years after planting and appear likely to avoid suppression by Scotch broom. Forest fires are important sources of polycyclic aromatic hydrocarbons (PAHs) in soils. However, factors controlling PAH production in soils subjected to fires in different sites are poorly understood. Here, we analyzed 143 sets of previously published data to evaluate the concentrations and composition profiles of PAHs in ash and soils associated with forest fires and to assess the impacts of soil depth, fire intensity, post-fire duration, and vegetation type on their occurrence. Compared to unburned soils, the total PAH concentrations increased by 205% (95% confidential interval of 152-269%; n=136) in soils associated with fires. This increase surpassed that of PAH toxic equivalents (73%) because fires produce dominantly low-ring PAHs with relatively low toxicity. PAH concentrations in fire-impacted sites increased by 684%, 258%, and 155% in the ash, 0–5-cm soil depth interval, and > 5-cm soil depth interval, respectively. The increases in PAH concentrations associated with mild-intensity fires (412%) exceeded those associated with moderate-intensity (163%) and high-intensity (168%) fires, which is possibly due to pyromineralization or volatilization of organic matters at high burning temperatures. These increases were highest within a month after the fire (280%), gradually decreasing over time, and showed no significant difference compared to the reference sites after 24 months. The concentration increases exhibit no major difference between various vegetation types (broad-leaved forest vs. coniferous forest vs. shrub). Assessments reveal that exposure to the post-fire soil PAHs involves no serious human health risk. However, potential adverse effects of the soil PAHs on the other organisms (e.g., microbes and plants) and the ecosystems should be further examined. The present study highlights the strong impacts of soil depth, fire intensity, and post-fire duration, and the relatively weak impact of the vegetation type on the PAH concentrations in soils associated with fires in different areas.

Duckweeds (Lemnaceae) are representative producers in fresh aquatic ecosystems and also yield sustainable biomass for animal feeds, human foods, and biofuels, and contribute toward effective wastewater treatment; thus, enhancing duckweed productivity is a critical challenge. Plant-growth-promoting bacteria (PGPB) can improve the productivity of terrestrial plants; however, duckweed–PGPB interactions remain unclear and no previous study has investigated the molecular mechanisms underlying duckweed–PGPB interaction. Herein, a PGPB, Ensifer sp. strain SP4, was newly isolated from giant duckweed (Spirodela polyrhiza), and the interactions between S. polyrhiza and SP4 were investigated through physiological, biochemical, and metabolomic analyses. In S. polyrhiza and SP4 coculture, SP4 increased the nitrogen (N), chlorophyll, and ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO) contents and the photosynthesis rate of S. polyrhiza by 2.5-, 2.5-, 2.7-, and 2.4-fold, respectively. Elevated photosynthesis increased the relative growth rate and biomass productivity of S. polyrhiza by 1.5- and 2.7-fold, respectively. Strain SP4 significantly altered the metabolomic profile of S. polyrhiza, especially its amino acid profile. N stable isotope analysis revealed that organic N compounds were transferred from SP4 to S. polyrhiza. These N compounds, particularly glutamic acid, possibly triggered the increase in photosynthetic and growth activities. Accordingly, we propose a new model for the molecular mechanism underlying S. polyrhiza growth promotion by its associated bacteria Ensifer sp. SP4, which occurs through enhanced N compound metabolism and photosynthesis. Our findings show that Ensifer sp. SP4 is a promising PGPB for increasing biomass yield, wastewater purification activity, and CO2 capture of S. polyrhiza. [Formula: see text] Copyright © 2021 The Author(s). Screw tree is a shrub from family Malvaceae that has many benefits in pharmacy. Screw tree seed has very hard seed coat, so the seed must be treated before germination. The aim of this research was to assess the characteristic of fruit and seeds, effect of seed breaking dormancy, and effect of NPK fertilizer dosage on the growth of screw tree (Helicteres isora) seedlings. The results indicated that screw tree fruits belong to dry fruits with capsule form and has 5 follicles that has different direction. Screw tree seed is orthodox type that has 9 ̶ 36 seeds in one follicle with triangle and square shape. Germination technique in laboratory with soaking the seeds in hot water 100oC until the water cool for 48 hours treatment produced the best germination amounting to 65.0%. Germination technique in green house with soaking the seeds in hot water 100oC until the water cool for 24 hours treatment was the best germination amounting to 77.0%. Seedlings fertilized with 1 g NPK doses increased screw tree growth in diameter parameter of 6.35 mm and shoot-root ratio parameter of 3.9 compared to control.

Most wastes generated during food production in the world are released to nature. However, the target and non-target species take these wastes into their organisms through nutrition although their bioavailability is unknown. In this study, the aim was to determine the bioavailability of wasted pumpkin seed membrane (PSM) for species in nature through its effects on the Drosophila obesity model along with the increased fat intake. For this purpose, either fat (20%) and PSM (0-2 g) were added to the Drosophila melanogaster diet. Survival-development, sex ratio, and adult lifespan were evaluated. As a result, the survival rate decreased and development time increased in insects depending on the amount of PSM added to the food. In the study, fed with PSM and fat intake negatively affected the insect in terms of survival-development and lifespan.

In the context of global climate change, the frequency and duration of harmful algal blooms (HABs) due to eutrophic coastal waters have increased. HABs can cause marine ecological imbalance, massive socio-economic damage, and potential public health problems. Therefore, accurate and specific techniques that can rapidly detect harmful algae are critical to expedite early monitoring and prediction of HABs. Although some methods based on amplification have previously been described, limitations associated with limited popularity and high costs still exist. In this study, the partial sequence of nuclear ribosomal DNA from Amphinidium carterae was selected as the target region. The biotin-labeled recombinase-aided amplification (RAA) products and the 6-carboxyfluorescein-labeled probe were specifically conjugated, which formed double-labeled RAA products analyzed by lateral flow dipstick (LFD). A rapid RAA–LFD assay aimed at visually detecting A. carterae was established and optimized. The specificity and sensitivity of RAA–LFD were determined. The nucleic acid test with other control algal species yielded negative results, which indicated that RAA–LFD assay can specifically detect A. carterae. The detection limit of RAA–LFD assay for A. carterae was 8.49 ng μL−1 genomic DNA and 1 cell mL−1. The whole detection process of RAA–LFD could be completed within 50 min. In conclusion, the proposed RAA–LFD assay may be promising for the efficient, specific, and rapid detection of A. carterae in monitoring efforts.

Rhizosphere bacterial communities (‘rhizobacteria’) can mediate plant-soil feedbacks that enhance plant drought tolerance, but the composition and ecological characteristics of these populations are poorly understood. We investigated the impact of drought on rhizobacteria communities and the effect of propagating drought-impacted communities on the drought tolerance of naive plants in a two-phase experiment. We tested whether propagating rhizosphere soils from drought-stressed shrub willow (Salix purpurea), or continuously watered controls, affected the performance of a subsequent generation of droughted plants. The impacts of drought on rhizobacteria and plant fitness were evaluated using 16S rRNA gene sequencing and measures of plant growth (root mass and stem length etc.) and stress (abscisic acid, osmolality). Uninoculated reference plants were used to isolate the effects of inoculation. Drought had a significant and lasting impact on the structure of rhizobacterial communities, characterized by increased populations of Actinobacteria (Acidimicrobiia, Thermoleophilia and Micrococcaceae). Foliar osmolality was significantly higher in plants receiving drought-selected communities relative to uninoculated controls, but no significant differences in plant growth were observed. Inoculation with rhizosphere soil from watered controls significantly reduced plant growth. The rhizosphere of watered control plants was dominated by Proteobacteria and Bacteroidetes. Our findings demonstrate that plants are affected by the legacy effects of drought on the rhizosphere microbiome. This drought legacy was propagated and persisted throughout nine weeks of plant growth, independent of prevailing water stress. Drought-impacted rhizospheres had larger populations of desiccation-tolerant (ex. Arthrobacter) and putatively endophytic taxa (ex. Rhizobium) with established plant growth promoting capabilities.

Summary The purpose of this study is to assess the repellent activity of lemongrass (Cymbopogon citratus) essential oil (EO), grown in Serbia under greenhouse conditions, against four prevalent stored product pests: Plodia interpunctella (larvae), Sitophilus oryzae, Acanthoscelides obtectus and Tribolium castaneum (adults). The lemongrass EO repellency was tested using filter paper in Petri dishes and a Y-tube olfactometer. According to the repellency index (RI), the lemongrass EO repellency was divided into 5 classes. Prior to biotesting, the chemical characterization of lemongrass EO was performed and the following main compounds were detected: myrcene (31.0%), geranial (30.0%), and neral (23.6%). The C. citratus EO considered was found to exhibit the Class III repellent activity against P. interpunctella larvae only at the highest concentration (namely 0.5%). This is the very first report on the C. citratus EO repellent activity against this pest. The lemongrass EO examined showed strong repellency (Class IV) against S. oryzae (0.2% and 0.5% of EO), A. obtectus (0.1% and 0.2%), and T. castaneum (0.05-0.1%). Moreover, higher lemongrass EO concentrations (0.5%) were found to exhibit extreme repellency (Class V) against A. obtectus and T. castaneum. The results obtained were confirmed in the bioassays performed, indicating the great potential of lemongrass EO as a bio-repellent when applied in higher concentrations to all the insects considered, regardless of the exposure period.

Tetrastigma angustifolia (Roxb.) Deb is an evergreen shrub belonging to the family Vitaceae. This plant species has been used for thousand years in Ayurvedic medicine. Ethnomedicinal study also documents the use of T. angustifolia leaves in the management of diabetes. On the other hand, Oxalis debilis Kunth. is a tristylous plant commonly known as pink woodsorrel belong to the family Oxalidaceae. This plant has been used traditionally for the treatment of dysentery and diarrhea. Ethnobotanical study also reports the use of leaf decoction of O. debilis in the treatment of diabetes. In our earlier work, the antidiabetic activity of hydro-alcoholic leaf extracts of T angustifolia and O. debilis have been reported. In spite of ethnomedicinal implications and several scientific studies in the recent past, phytochemical investigations in support of the antidiabetic potential of these plant species are yet to be explored. Therefore, the present study was aimed at the isolation of bioactive phytoconstituents as antidiabetic principle( s) from T angustifolia and O. debilis leaves. In this paper, two bioactive compounds, namely apigenin derivative (AGD) and stigmasterol (STM) were isolated from the methanolic leaf extracts of T. angustifolia and O. debilis, respectively by column chromatographic technique. The structures of the isolated compounds were established by spectroscopic/ spectrometric techniques including FT-IR, H-1 NMR and C-13 NMR and Mass. The isolated flavonoids were identified as 8-hydroxyapigenin 7-O-beta-D glucopyranoside, a derivative of apigenin (AGD) and stigmasta-5,22-dien-3,beta-ol i.e., stigmasterol (STM). The antidiabetic potential of AGD and STM was evaluated by in vitro alpha-glucosidase and alpha-amylase inhibitory assays. To validate the antidiabetic efficacy, molecular docking and dynamics studies were performed using AutoDock Vina and GROMACS software. In vitro assays revealed the antidiabetic potential of AGD and STM with alpha-glucosidase and alpha-amylase inhibitory activities. From docking and MD simulation studies, promising binding affinity of AGD and STM for human lysosomal alpha-glucosidase (5NN8) and human pancreatic alpha-amylase enzymes (1B2Y) with favorable binding modes, stable protein-ligand complexes and well defined protein-ligand interactions were observed. Based on in vitro and in silico studies, our study reports the antidiabetic potential of the isolated apigenin derivative, AGD (a new flavonoid molecule) and stigmasterol, STM (an existing plant sterol) with alpha-glucosidase and alpha-amylase inhibitory activities. However, the flavonoid molecule, AGD possesses better antidiabetic profile than the steroid molecule, STM particularly against human alpha-amylase enzyme. Our present investigation successfully validates the traditional as well as ethnomedicinal claims of T. angustifolia and O. debilis as antidiabetic medicines. (C) 2021 SAAB. Model organisms such as mice are important for basic research and serve as valuable tools in preclinical translational studies. A challenge with translating findings from mice to humans is identifying and separating evolutionarily conserved mechanisms in the immune system from those diverging between species. A significant emphasis has been placed on defining conserved gene regulation principles, with divergent mechanisms often overlooked. We put forward the perspective that both conserved and divergent mechanisms that regulate gene expression programs are of equal importance. With recent advances and availability of datasets, immunologists should take a closer look at the role for genetic diversity in altering gene expression programs between mouse and human immune cells.

Septins, a conserved family of GTP-binding proteins, are widely recognized as an essential cytoskeletal component, playing important roles in a variety of biological processes, including division, polarity, and membrane remodeling, in different eukaryotes. Although the roles played by septins were identified in the model organism Saccharomyces cerevisiae, their importance in other fungi, especially pathogenic fungi, have recently been determined. In this review, we summarize the functions of septins in pathogenic fungi in the cell cycle, autophagy, endocytosis and invasion host-microbe interactions that were reported in the last two years in the field of septin cell biology. These new discoveries may be expanded to investigate the functions of septin proteins in fungal pathogenesis and may be of wide interest to the readers of Microbiology and Molecular Pathology.

Saffron is one of the highly prized spices in the world. This spice is dark-red owing to the accumulation of crocins. Crocins are a group of carotenoid derivatives termed apocarotenoids with various health-promoting properties, especially anticancer and antidepressant ones. The present study aims to investigate the changes in the total crocin content in Crocus sativus stigma under salt stress. Additionally, plant resistance was evaluated by measuring the amounts of proline, soluble and insoluble sugars, potassium, and sodium in leaves. The corms about 80 g were planted in planting boxes under 6 levels of salinity (0, 2, 4, 6, 10 dS/m) in a completely random design with four replications. Results showed that after a relative decrease under electrical conductivities (EC) from 2 to 4 dS/m (49.62 mg/g DW), a sudden increase occurred in the total crocin content at the EC of 6 dS/m (63.07 mg/g DW). The amount of proline significantly increased at the EC of 4 dS/m (18.1 µg/g DW). With increasing levels of salinity, the amount of glucose, calcium, potassium, and sodium gradually increased in the leaves. Therefore, one of the advantages of saffron cultivation in saline areas or with saline water can be an increase in crocin. This not only brings huge economic benefits but also has a beneficial effect on human health.

Acid rain is a serious environmental problem and has obvious impacts on the growth, reproduction, and photosynthesis of terrestrial plants. Ulva prolifera, a main blooming species of green tides, was studied on its physiological response to acid rain. The photosynthetic parameters were determined under different conditions (salinity: 1, 10, 30‰; pH: 3.0, 3.5, 4.5; duration: 0.5, 1.0, 2.0 h) followed by 24-h recovering under natural conditions. Results showed 1-h treatment with pH 3.5 caused 50-70% reduction in the maximal quantum yield of PSII photochemistry (Fv/Fm) and effective quantum yield of PSII photochemistry (ФPSII) at normal salinity but when the low pH was combined with a salinity of 10‰ or lower, PSII activity was almost completely inhibited. Moreover, the low salinity (1‰ and 10‰) reduced the degree of photoprotection under low pH (3.5) conditions. Finally, we speculated if the pH of acid rain ≤ 3.5, with 1‰ salinity and 2-h rainfall time, the amount of U. prolifera and the scale of green tides would decrease.

Changes in the microbiota are associated with alterations in nervous system structure-function and behavior and have been implicated in the etiology of neuropsychiatric and neurodegenerative disorders. Most of these studies have centered on mammalian models due to their phylogenetic proximity to humans. Indeed, the germ-free mouse has been a particularly useful model organism for investigating microbiota-brain interactions. However, microbiota-brain axis research on simpler genetic model organisms with a vast and diverse scientific toolkit (zebrafish, Drosophila melanogaster, and Caenorhabditis elegans) is now also coming of age. In this review, we summarize the current state of microbiota-brain axis research in rodents and humans, and then we elaborate and discuss recent research on the neurobiological and behavioral effects of the microbiota in the model systems of fish, flies, and worms. We propose that a cross-species, holistic and mechanistic approach to unravel the microbiota-brain communication is an essential step toward rational microbiota-based therapeutics to combat brain disorders.

Sound management of native forests used for cattle grazing requires understanding the dynamics of forage productivity in the openings. Despite their importance, forage productivity drivers in highly heterogeneous forested landscapes, or their variability over the year, are still unclear. The aim of this work is to find predictors of Normalized Difference Vegetation Index (NDVI) variation in the openings of native temperate forests and to evaluate how these predictors change within the growing season. We used high spatial resolution remote sensing imagery from NW Patagonia to separate forest openings from tree dense canopy. We obtained data of each opening related with herbaceous and shrub forage productivity and calculated landscape metrics. We estimated a multiple linear regression model for predicting NDVI in each season. Beyond known variables related with forage productivity (altitude, precipitation, etc.), the shape of forest’ openings appeared as relevant in predicting NDVI. Higher values of forest opening perimeters were related with a decrease in NDVI in spring when soil water content is not limiting and conversely with an increase in NDVI in summer when water is limiting growth. These results suggest that environmental drivers such as temperature and soil moisture inside the opening, and competition or facilitation process between trees and grasses are mediated by the shape of the opening. Management of heterogeneous native forests for cattle raising requires considering the shape of the openings to maximize forage productivity.

The floristic life-form spectrum of any area unveils the prevailing phytoclimatic conditions, helps in understanding the species-environment relationship and can be applied to compare the vegetation pattern of one eco-zone to another. We scrutinized the pattern of species richness (SR) and floristic spectrum along the elevation gradient in a biodiversity-rich eco-zone of western Himalaya (western Ramganga valley, Uttarakhand, India) between 1200 m (river bank) to 3100 m a.s.l. (summit). The elevation range was classified into six elevation bands (i.e. 2700 m), to prepare the presence (1)/ absence (0) species data matrix using local distribution (elevation range) of each species to estimate the SR of various growth-forms and life-forms for the particular elevational band. The findings suggest that the correlation of SR with elevation is not linear as the species richness peaks at the middle elevation (humped-back shape relationship). The forest cover is large and continues up to the summit; thus, the correlation of tree and shrub SR with elevation is not clear; though, a partly positive correlation was observed between elevation and herb SR while negative correlation between climber SR and elevation. The percentage of phanerophytes and therophytes in total life-forms decreased significantly with increase in elevation while inverse trend was shown by hemicryptophytes and cryptophytes. The pattern of phytoclimate in the area varied from phanero-therophytic at the lower elevation to phanero-hemicryptophytic at the middle and hemicrypto-phanerophytic at the upper elevation. The present contribution is an attempt to fill the knowledge gap in pattern of growth-form, life-form and phytoclimate within the forest-covered montane zone of western Himalaya. It will be helpful in understanding the vegetation dynamics in mountain ecosystems and serve as a ready reference to planners in any future conservation strategy for the area.

Puparia of Pealius kufriensis Dubey sp. nov. found infesting a native and highly invasive plant, Rubus ellipticus Sm. (Rosaceae) are described. The host shrub was found highly infested with the new whitefly on the underside of leaves. Puparial characteristics of the genus Pealius Quaintance and Baker shared with Aleyrodes Latreille and Bemisia Quaintance and Baker are discussed.
