4. Childhood maltreatment experiences and child abuse potential: temperamental sensitivity as moderator?

Abstract

This study aimed to examine the relationship between negative experiences in childhood (physical-, sexual-, and emotional abuse and emotional neglect) and the risk for an individual to become a perpetrator of child maltreatment in adulthood. Participants were 337 female college students who completed self-report measures of childhood trauma and temperament. Risk for child abuse was assessed with the Child Abuse Potential Inventory. Results showed experiences of emotional neglect significantly predicted higher child abuse potential. Additionally it was shown that experiences of physical abuse significantly predicted higher child abuse potential but only in those individuals with high temperamental orienting sensitivity. These results underline the potentially damaging long-term effects of emotional neglect in childhood and indicate temperamental sensitivity may moderate the relationship between being abused as a child and being at risk for maltreating one's own offspring. 15. The behavioral phenotype of early life adversity: a 3-level meta-analysis of rodent studies: Supplemental Material

Abstract

Background: Altered cognitive performance has been suggested as an intermediate phenotype mediating the effects of early life adversity (ELA) on later-life development of mental disorders, e.g. depression. Whereas most human studies are limited to correlational conclusions, rodent studies can prospectively investigate how ELA alters cognitive performance in a number of domains. Despite the vast volume of reports, no consensus has yet been reached on the i) behavioral domains being affected by ELA and ii) the extent of these effects. Methods: To test how ELA (here: aberrant maternal care) affects specific behavioral domains, we used a 3-level mixed-effect meta-analysis, a flexible model that accounts for the dependency of observations. We thoroughly explored heterogeneity with MetaForest, a machine-learning data-driven analysis never applied before in preclinical literature. We validated the robustness of our findings with substantial sensitivity analyses and bias assessments. Results: Our results, based on > 400 independent experiments, yielded > 700 comparisons, involving ~8600 animals. Especially in males, ELA promotes memory formation during stressful learning but impairs non-stressful learning. Furthermore, ELA increases anxiety and decreases social behavior. The ELA phenotype was strongest when i) combined with other negative experiences ("hits"); ii) in rats; iii) in ELA models of ~10days duration. Conclusion: Prospective and well-controlled animal studies demonstrate that ELA durably and differentially impacts distinct behavioral domains. All data is now easily accessible with MaBapp (https://osf.io/ra947/), which allows researchers to run tailor-made meta-analyses on the topic, thereby revealing the optimal choice of experimental protocols and study power.

32. Challenging early life environments : Impact on behavioral inhibition and (pro-)social behavior in rats

Abstract

Individuals that have experienced profound stress during childhood can develop deficits in behavioral inhibition and social behavior, two core features of several stress related psychiatric disorders. However, not all children who grew up in challenging environments carry on to develop psychopathology and the mechanism behind this is largely unknown. As part of a Dutch collaborative effort (NWO Consortium on Individual Development) to answer the question ‘Why some children thrive and others don’t’ the research described in this thesis aims to shed light on the factors that contribute to these individual differences. More specifically, I try to answer the question of how challenging early life environments impact behavioral inhibition and (pro-)social behavior. To this end, a rodent model, the rat, was used to study how early life stress, through 24h deprivation of maternal care on the third day after birth, affects adult behavior. I also tested two adolescent interventions to examine if potential negative effects of maternal deprivation could be normalized; i) pharmacological treatment with the GR-antagonist mifepristone and ii) complex housing, in which animals are reared in socially and physically enriched housing that provides a challenging and stimulating environment from early adolescence onwards. Taken together, I show that early life stress has a negative impact on behavioral inhibition and social behavior, but the effects are modest. Because of this, interpretation of results of the two tested interventions is difficult and further research is needed to draw more solid conclusions. Finally, the two final chapters describe two new behavioral tests aimed to study pro-social behavior in rats.
