Seasonal influenza vaccines must be updated annually and suboptimally protect against strains mismatched to the selected vaccine strains. We previously developed a subunit vaccine antigen consisting of a stabilized trimeric influenza A group 1 hemagglutinin (H1) stem protein that elicits broadly neutralizing antibodies. Here, we further optimized the stability and manufacturability of the H1 stem antigen (H1 stem v2, also known as INFLUENZA G1 mHA) and characterized its formulation and potency with different adjuvants in vitro and in animal models. The recombinant H1 stem antigen (50 µg) was administered to influenza-naïve non-human primates either with aluminum hydroxide [Al(OH) 3 ] + NaCl, AS01 B , or SLA-LSQ formulations at week 0, 8 and 34. These SLA-LSQ formulations comprised of varying ratios of the synthetic TLR4 agonist 'second generation synthetic lipid adjuvant' (SLA) with liposomal QS-21 (LSQ). A vaccine formulation with aluminum hydroxide or SLA-LSQ (starting at a 10:25 µg ratio) induced HA-specific antibodies and breadth of neutralization against a panel of influenza A group 1 pseudoviruses, comparable with vaccine formulated with AS01 B , four weeks after the second immunization. A formulation with SLA-LSQ in a 5:2 μg ratio contained larger fused or aggregated liposomes and induced significantly lower humoral responses. Broadly HA stem-binding antibodies were detectable for the entire period after the second vaccine dose up to week 34, after which they were boosted by a third vaccine dose. Since the original outbreak of the SARS-CoV-2 virus, several rapidly spreading SARS-CoV-2 variants of concern (VOC) have emerged. Here, we show that a single dose of Ad26.COV2.S (based on the Wuhan-Hu-1 spike variant) protects against the Gamma and Delta variants in naive hamsters, supporting the observed maintained vaccine efficacy in humans against these VOC. Adapted spike-based booster vaccines targeting Omicron variants have now been authorized in the absence of human efficacy data. We evaluated the immunogenicity and efficacy of Ad26.COV2.S.529 (encoding a stabilized Omicron BA.1 spike) in naive mice and in hamsters with pre-existing immunity to the Wuhan-Hu-1 spike. In naive mice, Ad26.COV2.S.529 elicited higher neutralizing antibody titers against SARS-CoV-2 Omicron BA.1 and BA.2, compared with Ad26.COV2.S. However, neutralizing titers against the SARS-CoV-2 B.1 (D614G) and Delta variants were lower after primary vaccination with Ad26.COV2.S.529 compared with Ad26.COV2.S. In contrast, we found comparable Omicron BA.1 and BA.2 neutralizing titers in hamsters with pre-existing Wuhan-Hu-1 spike immunity after vaccination with Ad26.COV2.S, Ad26.COV2.S.529 or a combination of the two vaccines. Moreover, all three vaccine modalities induced equivalent protection against Omicron BA.2 challenge in these animals. Overall, our data suggest that an Omicron BA.1-based booster in rodents does not improve immunogenicity and efficacy against Omicron BA.2 over an Ad26.COV2.S booster in a setting of pre-existing immunity to SARS-CoV-2., (© 2023. Prior to ~1 million years ago (Ma), variations in global ice volume were dominated by changes in obliquity; however, the role of precession remains unresolved. Using a record of North Atlantic ice rafting spanning the past 1.7 million years, we find that the onset of ice rafting within a given glacial cycle (reflecting ice sheet expansion) consistently occurred during times of decreasing obliquity whereas mass ice wasting (ablation) events were consistently tied to minima in precession. Furthermore, our results suggest that the ubiquitous association between precession-driven mass wasting events and glacial termination is a distinct feature of the mid to late Pleistocene. Before then (increasing), obliquity alone was sufficient to end a glacial cycle, before losing its dominant grip on deglaciation with the southward extension of Northern Hemisphere ice sheets since ~1 Ma.

Antisera with Fiji disease virus (FDV) protein-specific and double-stranded (ds) RNA-specific antibodies were prepared by injecting rabbits with highly purified preparations of FDV cores. Electron microscopic examination of negatively stained core preparations disclosed the presence of icosahedral particles, about 54 nm in diameter, with projections (B spikes) at their vertices as previously reported. However, observations on similar preparations by electron microscopy of specimens shadowed at a low angle, by immune electron microscopy and by examination of thin sections of pelleted material, established that the particles consisted of cores to which some ds RNA strands were attached externally. The relationship of FDV core structure to its immunogenic properties is discussed.

Ultraviolet (UV) light is an important environmental mutagen and one of the major factors in the etiology of skin cancer in man. In this study we have addressed the question whether UV light can activate in vitro the N-ras proto-oncogene into an active oncogene, and whether pyrimidine cyclobutane dimers or (6-4) photoproducts are the premutagenic lesion. To that end, the cloned human N-ras proto-oncogene (pN-ras) was irradiated with UV light (254 nm) which results in the formation of cyclobutane dimers and (6-4) photoproducts and the irradiated DNA was transfected into Rat-2 cells. Whereas untreated pN-ras never gave foci upon transfection into Rat-2 cells, UV-irradiated pN-ras (100-300 J.m-2) yielded 1-2 foci/micrograms DNA. The DNAs of 49 primary foci were subjected to a second round of transfection. 13 of these were positive. The DNAs that did not give rise to secondary foci invariably contained high copy numbers of pN-ras. The DNAs from the secondary foci had only a low copy number of pN-ras. Using synthetic oligonucleotide probes we have found that all N-ras genes that gave rise to secondary foci contained a mutation at positions in codon 12 or 61. Only one primary transformant, negative in the secondary transformation assay, was shown to harbor a mutated N-ras gene. From these results we conclude that UV light can activate N-ras genes. Mutations were preferentially found at the 61st codon (11), harboring a TT doublet, while only 3 mutations were found at the 12th codon and none at the 13th codon, both harboring CC doublets. Of the 14 point mutations analysed 6 represented transitions and 8 transversions. No specific base-substitution could be regarded as predominant. Furthermore, treatment of the irradiated N-ras plasmids with photoreactivating enzyme prior to transfection, which specifically monomerizes cyclobutane dimers but not other photoproducts, reduced the transformation frequency several fold. None of the 36 DNAs from the primary foci obtained after transfection of photoreactivating enzyme-treated N-ras gave rise to secondary foci. These DNAs all contained high copy numbers of unmutated pN-ras. From this result we conclude that cyclobutane dimers, and not (6-4) photoproducts, are the major premutagenic lesions, responsible for the activation of N-ras. The possible role of ras genes as targets for UV-induced carcinogenesis is discussed.

Fusion of SV40-transformed rat (BRKSV) cells which do not spontaneously produce infectious virus, with permissive monkey cells resulted in a low level of production of infectious virus in the heterokaryons. UV-Irradiation of the BRKSV cells prior to fusion did not result in increased virus production, but irradiation of the monkey cells prior to fusion did result in enhanced induction (EI) of SV40, as compared to control experiments in which neither cell type was irradiated. This indicated that rat cells lack the ability to initiate replication of integrated SV40 upon UV-irradiation and do not contain "permissiveness" factors that are required to support SV40 replication. In contrast, monkey cells do contain such permissiveness factors which seem to be temporally enhanced by UV-irradiation, and thus may be responsible for the EI phenomenon. Expression of EI was dose-dependent and reached maximum values approximately 24 h after UV-irradiation. The kinetics of EI resembled that of EI previously established for SV40 induction in semi-permissive cells, and of enhanced reactivation (ER) and enhanced mutagenesis (EM) of SV40 in monkey cells. Similar kinetics of EI were obtained when human diploid fibroblasts were used for fusion with BRKSV cells. Similar levels of EI were found with normal human cells and repair-deficient xeroderma pigmentosum (XP) cells of complementation groups A and C, and XP variant cells. This suggests that expression of EI is not related to excision repair. Since EI is also normally expressed in XP cells which display an abnormal ER of HSV and in XP variant cells which show a delayed EM of HSV, we conclude that EI may occur independently of ER and EM. Finally it was shown that treatment of human cells with N-ethyl-N-nitrosourea results in similar induction of EI as irradiation with UV-light, and that addition of TPA in fusion experiments has no effect on EI.

Many mammalian cells exhibit damage-inducible phenomena that resemble the bacterial SOS functions. However, whereas RecA plays a prominent role in the prokaryotic SOS response, in mammalian cells so far no enhanced recombination as a result of treatment with DNA-damaging agents of the cells, rather than of infecting viruses, has been found. In order to study recombination as a UV-inducible cellular phenomenon we infected UV-irradiated normal and repair-deficient human fibroblasts with a mixed population of adenovirus 5 (Ad5) mutants that carried a deletion in the E1A or the E2A gene. Wild-type recombinant progeny viruses were readily obtained, but no enhanced recombination was observed at any UV dose given to the cells, nor at any time point between -6 h and +4 days between irradiation and infection. Control experiments, in which we infected unirradiated cells with UV-irradiated Ad5 deletion mutants (a test for recombination targeted at UV-damaged DNA) showed a strong increase in wild-type recombinant viruses when both deletion mutants had been irradiated compared to the additive effect of irradiation of either one of the mutants alone. Therefore, this study shows that UV irradiation results in an enhanced recombination activity in cells that is specifically targeted to damaged DNA, but it does not cause a general (untargeted) recombinational response (enhanced recombination) in the cell.

UV irradiation of simian virus 40 (SV40)-transformed human and hamster cells induced them both to express a mutator phenotype and to produce SV40. The mutator could also be activated indirectly by transfecting unirradiated cells with UV-damaged calf thymus DNA. In contrast, UV-damaged exogenous DNA failed to rescue SV40 from unirradiated transformed cells. These results suggest that the expression of transforming viruses and of cellular mutator functions is regulated by at least partially independent mechanisms. Unlike the activation of a cellular mutator phenotype, the rescue of SV40 from virus-transformed mammalian cells by UV light might require that the integrated viral DNA and/or specific cellular sequences are directly damaged.

DNA-damaging agents such as ultraviolet (UV) light are known to cause stimulation of virus replication in SV40-transformed hamster and human cells. The dose-response curves of UV-induced SV40 replication in transformed hamster cells resemble that obtained for UV-enhanced reactivation (ER) and UV-enhanced mutagenesis (EM) of SV40 or herpes viruses in mammalian cells. We have investigated whether UV-enhanced production of SV40 from transformed hamster (THK) and human (NB-E) cells belongs to the same category of conditional responses as ER and EM. To answer this question we have made use of the phenomenon that fusion of the SV40-transformed cells with monkey cells that are permissive to SV40 results in a considerable increase in the production of SV40 virus. When THK or NB-E cells were fused with UV-irradiated CV-1 cells at various times after irradiation, induction of SV40 was further increased and reached a maximum value of 2--3-fold when fusion was delayed for 24-48 h after irradiation. The kinetics of enhanced SV40 induction resembled that of ER and EM, suggesting that the UV-stimulated part of the induction represents one of the pleiotropic responses that are transiently induced in mammalian cells by DNA-damaging agents. Evidence is presented, showing that this transient effect can be induced only in cells that are permissive to SV40 replication. This suggests that the enhanced induction observed after fusion with irradiated monkey cells may be attributed to a transient increase in the activity of "permissiveness' factors. No enhanced induction was found when the THK or NB-E cells were fused with irradiated rodent cells, that are not or only slightly permissive to SV40 replication.

Phenotypic wild-type revertants from a UV-irradiated temperature-sensitive late mutant (ts BC245) of simian virus 40 (SV40) were isolated after replication in monkey cells at the nonpermissive temperature. The mutations occurring in 7 revertants were identified by DNA sequence analysis of the entire gene involved. All 10 mutations identified constituted single base substitutions, 7 of which occurred opposite pyrimidine doublets. Transitions were 3 times more abundant than transversions. Three base changes did not occur opposite pyrimidine-pyrimidine sequences. Exchange of a DNA fragment harbouring the altered base from a revertant with the corresponding fragment from the parental virus, showed that the base substitution was indeed responsible for the reversions to the wild-type phenotype (growth at the restrictive temperature). The data suggest that most base substitutions in highly UV-irradiated simian virus 40 are targeted at sites comprising two adjacent pyrimidines.
