Microbiology Centennial Symposium 2017 - Exploring Microbes for the Quality of Life (Book of Abstracts), Marine anaerobes play an important role in the global carbon cycle. This includes the degradation of organic matter by hydrolytic and fermentative marine anaerobes, of which ecologically relevant isolates are lacking. This lack could stem from the use of monomeric substrates in anaerobic cultivation rather than the recalcitrant polymeric substrates available to marine anaerobes in situ. We tested this hypothesis by using environmentally representative cultivation conditions with polysaccharides as substrates. Enrichment cultures were set up with mineral marine medium and low concentrations (2.5 mM monomer equivalent) of one of several polysaccharides. The cultures were inoculated with anoxic Black Sea sediment from the abyssal plain at 2100 m depth and incubated at 15°C (in situ T = 8°C). Various microbial clades without cultured representatives were enriched, indicating a potential hydrolytic and/or fermentative metabolism. We isolated hydrolytic strains of Kiritimatiellaeota (formerly Verrucomicrobia subdivision five) clade R76-B128, which is ubiquitous in anoxic marine waters, including the Black Sea. The only isolate of this clade is Kiritimatiella glycovorans, which is a halophilic bacterium that does not grow on polysaccharides. The 16S rRNA genes of our isolates form two phylogenetic clusters that share 94% identity with each other, and 83-84% identity with the 16S rRNA gene of K. glycovorans. According to taxonomic threshold values, this is strong evidence that our isolates represent two novel genera within a novel family of the Kiritimatiellaeota. In 2008 Europe introduced the CEN standards for Asphalt Concrete. The Netherlands adopted the approach of functional requirements, rather than empirical, recipe based requirement. The experiences since 2008 showed that, although this approach allows for a better, more fundamental understanding of Asphalt Concrete, the current understanding is far from complete. Especially the effect of higher percentages (60-70%) of reclaimed asphalt was surprising, since it appeared to improve all functional requirements without the typical interrelation where an increase in stiffness corresponds with a decrease in fatigue resistance. These experiences led to the initiation of a program using the Dutch road network as a living laboratory (NL-LAB). In combination with actual laboratory research on mixes used in pavement construction projects, this program aims first of all to assess the effects of mixing and compaction on functional properties. Secondly, it tries to establish the predictive quality of lab determined functional properties for field performance. This paper describes the NL-LAB program and the results for the first four projects that are analysed. Considering the variation in the results, it is clear that more results are needed to arrive at any definite conclusions. However, the available data do show that the stiffness can be determined quite well and the values found are consistent. For the resistance against fatigue the results are fairly reproducible, but the trends are not consistent over the different projects. The resistance to moisture damage appears to be predicted fairly well from the lab data, but the variation in the individual indirect tensile tests is such that that actual value of the indirect tensile strength ratio remains to be determined. Finally, the cyclic triaxial test results vary considerably over the first two projects, but they are very consistent over the second two projects. This could be related to the mixing and compaction used in the different projects.

In an ever faster changing world, innovation is a crucial tool to maintain the quality of road networks. In order to safely use innovative materials and techniques, validation procedures are needed. Because of the variable and non-standard nature of innovations, it is hard to standardize this kind of validation. Also, ensuring that innovations address the needs of road authorities can be quite a challenge. The Dutch highway authority, Rijkswaterstaat, currently uses a three tier approach to the validation of innovations. These consists of solicited innovations to address specific agency challenges and two systems to address unsolicited innovations. This paper describes the need for innovations, the most urgent topics that require solutions in the Dutch situation, the current approach towards innovation validation including examples of all three types of validation and the further developments foreseen for that system.

Het zelfherstellend vermogen, of de healingscapaciteit, van asfaltmengsels wordt in de meeste ontwerpmethoden voor wegverhardingen meegenomen. De kennis over dit zelfherstellend vermogen is gebaseerd op ervaringen en onderzoeken uit het verleden en de werkelijke mechanismen erachter zijn niet bekend. Hierdoor is het moeilijk om voor nieuwe materialen een realistische healingscapaciteit te bepalen. Een te hoge waarde leidt tot onverwachte, dure, schade aan wegen en een te lage waarde benadeelt een nieuw product. Het is, zeker met de huidige druk op grondstoffen en de daarmee samenhangende snelle veranderingen in de gebruikte materialen van groot belang de healingscapaciteit objectief te kunnen bepalen. Uit eerder onderzoek aan de TU Delft (Pronk et al, 2009) kwam naar voren dat de combinatie van de in Nederland gebruikelijk vierpuntsbuig vermoeiingsproeven met rustperioden en het partial healing model het mogelijk maakten om op een enkel proefstuk een indicatie van healing te bepalen. Dat de bepaling op een proefstuk gendaan kan worden is van belang omdat de verschillen tussen proefstukken van een mengsel in vermoeiingsonderzoek zodanig zijn dat dit de bepaling van een healingswaarde lastig maakt. In dit onderzoek wordt gekeken of de benadering met proeven en modellen voor twee volumetrisch gelijke mengsels met bitumen van verschillende hardheden (penetratie 10/20 en 70/100) de verwachte resultaten geeft. Die verwachtingen zijn gebaseerd op empirie: zachte bitumen geeft meer healing dan harde. Indien dit met deze benadering ook gevonden zou worden, kan de benadering ook op andere materialen worden toegepast. Naast het partial healing model werd ook een eenvoudiger model gebruikt om te bepalen of dit voordelen zou bieden in robuustheid. De toepassing van de modellen bleek helaas niet te werken omdat deze te gevoelig waren voor de vorm van de proefsignalen om robuuste voorspellingen te geven. Er is daarom op een meer pragmatische manier naar de data gekeken, waarbij er nog steeds een healingswaarde per proefstuk werd verkregen. Zonder de modellen waren deze waarden noodgedwongen gekoppeld aan een andere stijfheidsafname dan de 50% stijfheidsreductie die als einde levensduur gebruikt wordt in de vermoeiingsproef en dat maakt de koppeling van de gemeten data aan een levensduurverlenging en daarmee het bepalen van een healingscapaciteit lastig. Het verloop van de healingscapaciteit over de stijfheid voor beide onderzochte mengsels verschilde sterk, wat een goede basis leek voor een healingsindicator. Helaas was ook het verschil in stijfheidsverloop in de vermoeiingsproef heel verschillend en als beide effecten gecombineerd worden om een verloop van healing over de levensduur te vinden, valt het verschil vrijwel weg. Hierdoor werd er nauwelijks verschil gevonden tussen beide mengsels en blijkt deze benadering niet geschikt voor het bepalen van de healingscapaciteit van asfaltmengsels. Doordat er bij proefstukproductie een fout is gemaakt, was het verschil tussen beide bitumina niet zo groot als bij de opzet werd beoogd. Voor de volledigheid zouden proeven met het beoogde 10/20 bitumen gedaan moeten worden. Aangezien de healing altijd groter dan 1 moet zijn en de voor het zachte mengsels gevonden waarden al weinig boven de 1 liggen, zullen deze proeven niet leiden tot nieuwe inzichten over de bruikbaarheid van de benadering. In die uitwerking van de resultaten is gekeken naar het gebruik van de (verandering) in energie dissipatie gedurende de proef als een alternatief schade criterium voor de halvering van de stijfheid. Hieruit lijkt te komen dat het afwijken van de gedissipeerde energie van de quasi-lineaire ontwikkeling in fase II van de vermoeiingsproef een bruikbare indicator zou zijn. De ratio (RDEC) heeft geen meerwaarde voor de hier geanalyseerde materialen en test condities. Het gebruik van de afwijking van de de gedissipeerde energie van de steady state als indicator van de vermoeiingslevensduur kan achteraf makkelijk worden toegepast, mits de data doorloopt tot in fase III. Het implementeren in de proefsoftware heeft meer voeten in aarde, omdat gedurende de proef de indicator voor steady state verloop bepaald moet worden, waarbij het de vraag is hoe groot het interval voor de bepaling moet zijn en hoe groot de afwijking moet worden om te stellen dat fase III is begonnen.

Vanwege het grote aantal variabelen dat een rol speelt bij het gedrag van asfaltverhardingen, is het cruciaal om bij het volgen van het gedrag in de tijd gedetailleerde gegevens te hebben over de samenstelling, aanlegcondities en het gedrag en verloop in eigenschappen in de tijd. Op basis van ervaringen uit het verleden, kan men lessen leren over hoe het gedrag van materialen het beste gemonitord kan worden. Puntsgewijs incidenteel of structureel meerjarig bemonsteren? Het kan tot verrassende conclusies leiden. Deze paper gaat in op de ervaring die is opgedaan met de opzet en uitvoering van het monitoren van de veroudering van de ZEBRA vakken bij het Innovatie Programma Geluid.

This study evaluated the long-term subjective outcomes of 19 patients (13 females, six males), following surgical resection and proximal transposition of lower-extremity neuromas. The patient mean age was 49 years (S.D. : 18), with a mean post-injury time of 8 years and post-surgery follow-up of 4 years. Thirteen patients reported symptomatic improvement in pain, and six reported no improvement. Often subjects who were unemployed prior to surgery, two returned to work and four retired postoperatively. Preoperatively, 13 patients used analgesic medication and postoperatively, eight reported taking less medication ; four had completely stopped taking pain medication. Symptomatic pain relief apparently can be achieved with surgical management of lower-extremity neuromas.

This working paper details modifications carried out to the congested assignment traffic network model SATURN in order to represent mad user charging. The work was undertaken as part of an SERC funded study to investigate the network effects of a series of alternative road user charging systems upon an existing road network. Four road user charging systems which were tested during the study have been represented in SATURN. These are toll cordons, time-based charging, congestion charging and distance-based charging.

The aim of this research has been to investigate the modelled effects of alternative road user charging systems upon an existing road network using the congested assignment models SATURN and CONTRAM applied to the city of Cambridge. Four road user charging systems which are being considered for practical application have been tested. These are toll cordons, time-based charging, a congestion charging system similar to that proposed in Cambridge and distance-based charging. Tests have been conducted using current morning peak travel demand patterns both with a fixed trip matrix, to isolate rerouteing issues and using the SATURN elastic assignment program, SATEASY, in order to address the effects of charging upon the frequency, timing and distribution of trips. Network impacts have been assessed using a series global indicators, in particular effects on vehicle-km, vehicle-hours and the resulting average network speeds. In addition, results have been obtained for total delay times, cordon crossing flows and revenues generated from charging. These results are presented and their implications discussed.
