Objective: To evaluate the immunological response elicited by an inactivated bacterial vector carrying the K39 antigen of Leishmania infantum, and a purified antigen. Methods: Mice were subjected to the following treatments: (1) Purified recombinant K39 (rK39) protein at a 20 μg dose with complete Freund’s adjuvant; (2) Inactivated Escherichia coli (BL21 DE3) carrying the K39 protein at an equivalent total protein content of 200 μg; (3) Inactivated bacteria lacking the K39 protein; (4) Non-immunized control animals. Serological monitoring was performed. All groups were challenged by intraperitoneal injection of 107 Leishmania infantum promastigotes. After euthanasia, the liver and spleen were collected to analyze the levels of TNF, IFN-γ, IL-12, IL-4, and IL-10. Results: Mice immunized with purified rK39 or the inactivated bacterial vector carrying the K39 antigen of Leishmania infantum showed a long-lasting immune response with high levels of polyclonal antibodies specifically recognizing the recombinant proteins. The IgG1 subclass was the predominant immunoglobulin; however, the induction of IgG2a and the profile of cytokines produced were indicative of the induction of a mixed-type response. Conclusions: The inactivated bacterial vector carrying the K39 antigen, as well as the purified antigen can induce a long-lasting immune response in immunized mice, predominantly favouring a Th2 profile response.

Summary: The aim of this study was to characterize the protein profile of ovarian follicular fluid (FF) of brown brocket deer (Mazama gouazoubira). Five adult females received an ovarian stimulation treatment and the FF was collected by laparoscopy from small/medium (≤3.5 mm) and large (>3.5 mm) follicles. Concentrations of soluble proteins in FF samples were measured and proteins were analyzed by 1-D SDS-PAGE followed by tryptic digestion and tandem mass spectrometry. Data from protein list defined after a Mascot database search were analyzed using the STRAP software tool. For the protein concentration, no significant difference (P > 0.05) was observed between small/medium and large follicles: 49.2 ± 22.8 and 56.7 ± 27.4 μg/μl, respectively. Mass spectrometry analysis identified 13 major proteins, but with no significant difference (P > 0.05) between follicle size class. Zika (ZIKV) and Dengue (DENV) viruses are clinically significant due to their severe neurological and hemorrhagic complications. Rapid diagnostics often rely on nonstructural proteins to generate specific antibodies. This study aimed to produce IgG antibodies from the recombinant ZIKV protein and plant-expressed NS2B protein for arbovirus detection in serum and urine samples. The NS2B protein was expressed in Nicotiana benthamiana and purified chromatographically. Validation of recombinant NS2B as an antigen in indirect immunoassays demonstrated 95% sensitivity and 100% specificity in IgM/IgG ELISA tests, enabling effective detection of ZIKV and DENV. Notably, r-ZIKV-NS2B IgG identified positive ZIKV and DENV cases in urine but failed to detect negatives, suggesting limitations in specificity for urine diagnostics. Using urine as a diagnostic medium offers a less invasive and more practical approach, broadening the test applicability. This study utilized patient-derived positive urine samples and healthy samples spiked with an exogenous virus. The present study was conducted to characterize the metabolome of accessory gland fluid (AGF) of locally adapted Morada Nova rams, raised in the Brazilian Northeast. AGF was collected by an artificial vagina from five vasectomized rams. Metabolites were identified by gas chromatography-mass spectrometry (GC/MS) and high-performance liquid chromatography-mass spectrometry (LC/MS), with the support of Human Metabolome Database, PubChem, LIPID Metabolites, Pathways Strategy databases, and MetaboAnalyst platforms. There were 182 and 190 metabolites detected by GC/MS and LC/MS, respectively, with an overlap of one molecule. Lipids and lipid-like molecules were the most abundant class of metabolites in the ram AGF (127 compounds), followed by amino acids, peptides, and analogs(103 metabolites). Considering all GC/MS and LC/MS, fructose, glycerol, citric acid, d-mannitol, d-glucose, and l-(+)-lactic acid were the most abundant single metabolites present in the ram AGF. Meaningful pathways associated with AGF metabolites included glycine, serine and threonine metabolism; pantothenate and CoA biosynthesis; galactose metabolism; glutamate metabolism and phenylalanine metabolism, and so forth. In conclusion, the combined use of LC/MS and GC/MS was essential for getting a holistic view of the compounds embedded in the ram AGF.
