Abstract

Item does not contain fulltext, Translation studies increasingly foreground the significance of local actors as agentic translators. Drawing on a brokerage work perspective, this article seeks to advance our understanding of managerial agents as translators by examining how and why these may vary in their role as intermediary or ‘strategic third’, and how these roles are associated with different patterns of translation. Examining qualitative data from a study of individuals tasked with implementing Lean in hospital contexts, we identify three brokerage modes of translation these actors may engage in (stretching, shielding and synthesizing), their main conditions, and the specific translation tactics they use within these modes (positioning, labeling and channeling). Our study extends our understanding of micro-level translation and reveals that purposeful ‘misalignment’ may be a significant and under-theorized part of the translation process.

Item does not contain fulltext, The widespread promotion of management ideas, their regular inclusion in textbooks and business school curricula and their use in organizational change programs has engendered debates about the impact of these ideas on management and organizational practice. Based on analyses of managerial audience members' activities and related meaning-making prior to, during and after guru events with leading management thinkers, this book sheds new light on how management practitioners come to use management ideas in the different relevant contexts of their working lives. The authors argue that a broader, more differentiated and more dynamic view of managerial audiences is essential in understanding the impact of management ideas as well as the nature of contemporary managerial work. For scholars and students in organisation studies, knowledge management and management consultancy, as well as reflective management practitioners.

8. Translating Management Concepts: Towards a Typology of Alternative Approaches

Abstract

Translation has been established as an important theoretical perspective for studying the flow of management concepts. Yet, despite its potential, we find limited reflection on the various ways in which the perspective is understood and used. As the under-theorized and fragmented discourse may hamper the progress of translation research as an academic field, it is in need of closer examination. The purpose of this paper is to explore the different conceptualizations of translation, in terms of their key foci and base assumptions, and to review the work that has accumulated into different sub-streams. Based on a systematic literature review of 150 publications, we identify two theoretically relevant dimensions that mark important differences between these different streams of research: (1) the source of variation; and (2) the object of variation. With these dimensions, we develop a typology of four alternative approaches to translation and show how these are associated with institutional, rational, dramaturgical and political perspectives. We draw on these broader theoretical lenses to contextualize and deepen our understanding of the specific possibilities and limitations of alternative translation approaches, and we highlight the potential for further connections and integration between them.

Abstract

Purpose: The loss of olfactory function is known to occur in patients suffering from (behavioral variant) frontotemporal dementia ((bv)FTD) and Alzheimer's disease (AD), although different pathophysiological mechanisms underpin this clinical symptom in both disorders. This study assessed whether brain metabolism of the olfactory circuit as assessed by positron emission tomography (PET) imaging with 2-[fluorine-18]fluoro-2-deoxy-d-glucose ([ 18 F]-FDG) can distinguish these entities in different subsets of patients., Methods: Patients presenting with cognitive decline were included from a prospectively kept database: (1) bvFTD patients, (2) AD patients and (3) patients with logopenic primary progressive aphasia (PPA). Metabolic rates were calculated for different regions of the olfactory circuit for each subgroup and compared with a cohort of subjects with normal brain metabolism. Additionally, in patients with a logopenic PPA pattern on PET-imaging, statistical parametric mapping (SPM) analysis was performed., Results: The metabolism of subdivisions of the olfactory circuit as assessed by [ 18 F]-FDG PET brain imaging to bvFTD and AD from control subjects resulted in sensitivity/specificity rates of 95/87.5% and 80/83.3%, respectively. A sensitivity/specificity rate of 100/87.5% was achieved when used to differentiate AD from bvFTD. In patients with the PPA pattern on imaging, the underlying cause (either FTD or AD) could be determined with a sensitivity/specificity rate of 88/82%. SPM analysis concurred that different regions of the olfactory circuit were affected in patients suffering from AD PPA or bvFTD PPA., Conclusion: Metabolic dysfunction in the olfactory circuit is different in various neurodegenerative disorders. Abstract

Aims: A diabetic eye screening programme has huge value in reducing avoidable sight loss by identifying diabetic retinopathy at a stage when it can be treated. Artificial intelligence automated systems can be used for diabetic eye screening but are not employed in the national English Diabetic Eye Screening Programme. The aim was to report the performance of a commercially available deep-learning artificial intelligence software in a large English population., Methods: 9817 anonymised image sets from 10,000 consecutive diabetic eye screening episodes were presented to an artificial intelligence software. The sensitivity and specificity of the artificial intelligence system for detecting diabetic retinopathy were determined using the diabetic eye screening programme manual grade according to national protocols as the reference standard., Results: For no diabetic retinopathy versus any diabetic retinopathy, the sensitivity of the artificial intelligence grading system was 69.7% and specificity 92.2%. The performance of the artificial intelligence system was superior for no or mild diabetic retinopathy versus significant or referrable diabetic retinopathy with a sensitivity of 95.4% and specificity of 92.0%. No cases were identified in which the artificial intelligence grade had missed significant diabetic retinopathy., Conclusion: The performance of a commercially available deep-learning artificial intelligence system for identifying diabetic retinopathy in an English national Diabetic Eye Screening Programme is presented. 18. Simultaneous screening and classification of diabetic retinopathy and age-related macular degeneration based on fundus photos-a prospective analysis of the RetCAD system.

Abstract

Aim: To assess the accuracy of an artificial intelligence (AI) based software (RetCAD, Thirona, The Netherlands) to identify and grade age-related macular degeneration (AMD) and diabetic retinopathy (DR) simultaneously based on fundus photos., Methods: This prospective study included 1245 eyes of 630 patients attending an ophthalmology day-care clinic. Fundus photos were acquired and parallel graded by the RetCAD AI software and by an expert reference examiner for image quality, and staging of AMD and DR. Adjudication was provided by a second expert examiner in case of disagreement between the AI software and the reference examiner. Statistical analysis was performed on eye-level and on patient-level, by summarizing the individual image level-gradings into and eye-level or patient-level score, respectively. The performance of the RetCAD system was measured using receiver operating characteristics (ROC) analysis and sensitivity and specificity for both AMD and DR were reported., Results: The RetCAD achieved an area under the ROC (Az) of 0.926 with a sensitivity of 84.6% at a specificity of 84.0% for image quality. On image level, the RetCAD software achieved Az values of 0.964 and 0.961 with sensitivity/specificity pairs of 98.2%/79.1% and 83.9%/93.3% for AMD and DR, respectively. On patient level, the RetCAD software achieved Az values of 0.960 and 0.948 with sensitivity/specificity pairs of 97.3%/73.3% and 80.0%/90.1% for AMD and DR, respectively. After adjudication by the second expert examiner sensitivity/specificity increases on patient-level to 98.6%/78.3% and 100.0%/92.3% for AMD and DR, respectively., Conclusion: The RetCAD offers very good sensitivity and specificity compared to manual grading by experts and is in line with that obtained by similar automated grading systems. The RetCAD AI software enables simultaneous grading of both AMD and DR based on the same fundus photos. Its sensitivity may be adjusted according to the desired acceptable sensitivity and specificity. Abstract

Purpose: To provide proof-of-concept data to support use of Doxil-liposomal topotecan (Topophore C) combinations to treat ovarian cancer., Experimental Design: ES-2, OVCAR-3, and SKOV-3 ovarian cancer cell lines were treated with doxorubicin-topotecan combinations by exposing the cells to drugs from 1 to 72 hours. Pharmacokinetic analysis was conducted following administration of liposomal formulations of these drugs alone and in combination. Efficacy assessments were completed in ES-2 and SKOV-3 ovarian cancer models., Results: On the basis of drug doses capable of achieving 50% reduction in cell viability over 72 hours, doxorubicin-topotecan combinations were additive in SKOV-3 but highly synergistic in ES-2 and OVCAR-3 cells. Favorable drug-drug interactions increased with increased drug exposure time. Topophore C pharmacokinetic remained unaffected when co-administered with Doxil. In the ES-2 model, Doxil at maximum tolerated dose (MTD 7.5 mg/kg) in combination with free topotecan (MTD 15 mg/kg) did not enhance median survival time (MST) over that achieved with topotecan alone. In contrast, MST was increased to 52 days with combination of Topophore C (MTD 2.5 mg/kg) and Doxil (7.5 mg/kg) compared with untreated animals (MST 18 days) or those treated with Topophore C alone (MTD 5 mg/kg, MST 40 days). In the SKOV-3 model, combination treatments showed better therapeutic efficacy than the individual drugs., Conclusions: Topotecan-doxorubicin combinations produced additive or synergistic effects which were best achieved when the tumor cells were exposed to drugs over extended time. Abstract

We hypothesized that the spatial distribution of groundwater inflows through river bottom sediments is a critical factor associated with the distribution of coaster brook trout (a life history variant of Salvelinus fontinalis) spawning redds. An 80-m reach of the Salmon Trout River, in the Huron Mountains of the upper peninsula of Michigan, was selected to test the hypothesis based on long-term documentation of coaster brook trout spawning at this site. A monitoring well system consisting of 22 wells was installed in the riverbed to measure surface and subsurface temperatures over a 13-month period. The array of monitoring wells was positioned to span areas where spawning has and has not been observed. Over 200,000 total temperature measurements were collected from five depths within each monitoring well. Temperatures in the substrate beneath the spawning area were generally less variable than river temperatures, whereas temperatures under the nonspawning area were generally more variable and closely tracked temporal variations in river temperatures. Temperature data were inverted to obtain subsurface groundwater velocities using a numerical approximation of the heat transfer equation. Approximately 45,000 estimates of groundwater velocities were obtained. Estimated groundwater velocities in the spawning area were primarily in the upward direction and were generally greater in magnitude than velocities in the nonspawning area. Both the temperature and velocity results confirm the hypothesis that spawning sites correspond to areas of significant groundwater flux into the river bed., (© 2011, The Author(s). Abstract

Transgenic tobacco plants have been obtained expressing nucleoprotein (N) gene sequences of three different tospoviruses known to affect vegetable crops: tomato spotted wilt virus (TSWV), tomato chlorotic spot virus (TCSV), and groundnut ringspot virus (GRSV). The chimeric plant transformation vector used comprised the three viral N gene sequences, each with a copy of the CaMV 35S promoter and the nos terminator. Despite the high levels of homology between the different N gene sequences (74-82%) and the presence of repeated promoter and terminator sequences in this construct, unrearranged copies of this triple N gene construct were stably maintained in both Escherichia coli and Agrobacterium tumefaciens plasmids used during the cloning process, as well as in several generations of transgenic tobacco plants. A transgenic tobacco line was obtained that exhibited high levels of resistance to all three tospoviruses, showing the possibility of producing transgenic plants with a broad resistance to tospoviruses by introducing tandemly cloned viral N gene sequences. DNA analysis of this transgenic plant line shows that the multivirus resistance trait is confined to a single genetic locus, which is very convenient for further breeding purposes.

Abstract

Recently high levels of protection against tomato spotted wilt virus (TSWV), a negative-strand RNA virus infecting plants, have been obtained by transforming tobacco with viral nucleoprotein (N) gene sequences. Here we demonstrate that this protection is primarily due to the presence of N gene transcripts in the cells of transgenic plants, and hence appears to be RNA-mediated. Further, transgenic tobacco plants are only protected to isolates and strains of TSWV and not to other tospoviruses that share considerable nucleotide sequence homology in their N genes to TSWV. In addition to being protected after mechanical inoculation, the transgenic tobacco plants are also resistant to inoculation using viruliferous thrips, i.e. Frankliniella occidentalis (Perg.), one of the most important natural vector species.
