Dopamine is extremely important for the multiple functions of the brain and spinal cord including locomotor behavior. Extracellular dopamine levels are controlled by the membrane dopamine transporter (DAT), and animals lacking DAT (DAT-KO) are characterized by hyperdopaminergia and several alterations of locomotion including hyperactivity. Neuronal mechanisms of such altered locomotor behavior are still not fully understood. We believe that in hyperdopaminergic animals both the spinal and brain neuronal networks involved in locomotion are modified. Using the c-fos technique, we studied activated neuronal networks of the spinal cord and two brainstem structures related to locomotor control and being under the strong dopaminergic influence, the cuneiform nucleus (CnF) and ventrolateral periaqueductal gray (VLPAG), in wildtype (DAT-WT) and DAT-KO rats. In the spinal cord, most c-fos-positive cells were located in the dorsal laminae II-IV and in the central gray matter (laminae V-VI). No differences were revealed for the central areas. As for the dorsal areas, in the DAT-WT group, labeled cells mostly occupied the lateral region, whereas, in the DAT-KO group, c-fos-positive cells were observed in both medial and lateral regions in some animals or in the medial regions in some animals. In the brainstem of the DAT-WT group, approximately the same number of labeled cells were found in the CnF and VLPAG, but in the DAT-KO group, the VLPAG contained a significantly smaller number of c-fos-positive cells compared to the CnF. Thereby, our work indicates an imbalance in the sensorimotor networks located within the dorsal horns of the spinal cord as well as a disbalance in the activity of brainstem networks in the DAT-deficient animals. This study involved a comparison of 5 kinematic-based algorithms to detect heel strike (HS) and toe-off (TO) events during human locomotion at different speeds. The objective was to assess how different running and walking speeds affect contact event determination during treadmill locomotion. Thirty male runners performed walking at 5 km/h and running at 9,11, and 13 km/h on a treadmill. A kinematic system was used to capture the trajectories of 2 retro-reflective markers placed at the subject's right heel and second metatarsal. A footswitch device was used to determine the "true" times of HS and TO compared with 5 kinematic-based algorithms. The results of the current study illustrated that speed influences the HS error in the vertical position and horizontal velocity algorithms, and the TO error in the vertical position and horizontal velocity algorithms. This difference was found in the transition from walking to running; however, higher running speeds did not affect the error estimation. Higher accuracy was found with combined algorithms, namely, one using vertical acceleration and position and another using horizontal and vertical position with no influence from different locomotion speeds. The use of motorized treadmills as convenient tools for the study of locomotion has been in vogue for many decades. However, despite the widespread presence of these devices in many scientific and clinical environments, a full consensus on their validity to faithfully substitute free overground locomotion is still missing. Specifically, little information is available on whether and how the neural control of movement is affected when humans walk and run on a treadmill as compared to overground. Here, we made use of linear and non-linear analysis tools to extract information from electromyographic recordings during walking and running overground, and on an instrumented treadmill. We extracted synergistic activation patterns from the muscles of the lower limb via non-negative matrix factorization. We then investigated how the motor modules (or time-invariant muscle weightings) were used in the two locomotion environments. Subsequently, we examined the timing of motor primitives (or time-dependent coefficients of muscle synergies) by calculating their duration, the time of main activation, and their Hurst exponent, a non-linear metric derived from fractal analysis. We found that motor modules were not influenced by the locomotion environment, while motor primitives were overall more regular in treadmill than in overground locomotion, with the main activity of the primitive for propulsion shifted earlier in time. Our results suggest that the spatial and sensory constraints imposed by the treadmill environment might have forced the central nervous system to adopt a different neural control strategy than that used for free overground locomotion, a data-driven indication that treadmills could induce perturbations to the neural control of locomotion.

Motor control and body representation in the central nervous system (CNS) as well as musculoskeletal architecture and physiology are shaped during development by sensorimotor experience and feedback, but the emergence of locomotor disorders during maturation and their persistence over time remain a matter of debate in the absence of brain damage. By using transient immobilization of the hind limbs, we investigated the enduring impact of postnatal sensorimotor restriction (SMR) on gait and posture on treadmill, age‐related changes in locomotion, musculoskeletal histopathology and Hoffmann reflex in adult rats without brain damage. SMR degrades most gait parameters and induces overextended knees and ankles, leading to digitigrade locomotion that resembles equinus. Based on variations in gait parameters, SMR appears to alter age‐dependent plasticity of treadmill locomotion. SMR also leads to small but significantly decreased tibial bone length, chondromalacia, degenerative changes in the knee joint, gastrocnemius myofiber atrophy and muscle hyperreflexia, suggestive of spasticity. We showed that reduced and atypical patterns of motor outputs, and somatosensory inputs and feedback to the immature CNS, even in the absence of perinatal brain damage, play a pivotal role in the emergence of movement disorders and musculoskeletal pathologies, and in their persistence over time. Understanding how atypical sensorimotor development likely contributes to these degradations may guide effective rehabilitation treatments in children with either acquired (ie, with brain damage) or developmental (ie, without brain injury) motor disabilities. Abstract: A number of important health-related outcomes are directly related to a person''s ability to maintain normal gait speed. We hypothesize that cellular telephones may be repurposed to measure this important behavior in a noninvasive, continuous, precise, and inexpensive manner. The purpose of this study was to determine if physical activity (PA) counts collected by cell phone accelerometers could measure treadmill gait speeds. We also assessed how cell phone placement influenced treadmill gait speed measures. Participants included 55 young, middle-aged, and older community-dwelling men and women. We placed cell phones as a pendant around the neck, and on the left and right wrist, hip, and ankle. Subjects then completed an individualized treadmill protocol, alternating 1min rest periods with 5min of walking at different speeds (0.3–11.3km/h; 0.2–7mi/h). No persons were asked to walk at speeds faster than what they would achieve during day-to-day life. PA counts were calculated from all sensor locations. We built linear mixed statistical models of PA counts predicted by treadmill speeds ranging from 0.8 to 6.4km/h (0.5–4mi/h) while accounting for subject age, weight, and gender. We solved linear regression equations for treadmill gait speed, expressed as a function of PA counts, age, weight, and gender. At all locations, cell phone PA counts were strongly associated with treadmill gait speed. Cell phones worn at the hip yielded the best predictive model. We conclude that in both men and women, cell phone derived activity counts strongly correlate with treadmill gait speed over a wide range of subject ages and weights. [Copyright &y& Elsevier]

Abstract: Introduction: Instrumented treadmills are becoming increasingly more common in gait laboratories. Instrumented side-split treadmills allow the collection of forces under each foot during walking. However, there may be a tendency to increase the base of support when walking on these treadmills, influencing other frontal plane mechanics as well. Therefore, the purpose of this study was to examine the effect of walking on a side-split instrumented treadmill on base of gait and frontal plane kinematics of the lower extremity. Methods: Twenty subjects walked on both a split and a single-belt treadmill. Base of gait and frontal plane kinematic angles and variability data were recorded. A one-way ANOVA was used to determine differences between the single and split-belt conditions at baseline and following a 10min accommodation on the split-belt. The relationships between the change in base of gait and change in each kinematic variable were also determined. Results: On average, the base of gait was 3.7cm wider on the split-belt treadmill with a 4mm gap between belts. No significant differences were observed in the mean values of lower extremity kinematics or kinematic variability at baseline or following the 10min accommodation. However, the increase in base of gait was significantly related to a decrease in peak knee and hip adduction angles. Conclusion: The 4mm gap between the treadmill belts significantly increased the mean base of gait in all subjects. This did not alter mean frontal plane kinematics. However, as base of gait increased, the tendency towards hip and knee abduction also increased. [Copyright &y& Elsevier]

Abstract: Early brain injury including white matter damage (WMD) appears strongly correlated to perinatal hypoxia-ischemia and adverse neurological outcomes in preterm survivors. Indeed, WMD has been widely associated with subtle to major motor disturbances, sensory, behavioral and cognitive impairments in preterm infants who afterward develop cerebral palsy (CP). Prenatal ischemia (PI) has been shown to reproduce the main features of WMD observed in preterm infants. The present study was aimed at determining in adult rats the impact of PI on brain axons, musculoskeletal histology and locomotor activity. PI was induced by unilateral intrauterine artery ligation at E17 in pregnant rats. We found axonal degeneration and reactive astrogliosis in several white matter regions of adult PI rats. We found mild myopathic and secondary joint changes, including increased variability in myofiber size in several hind limb muscles, decreased myofibers numbers but increased Pax 7 cells and myofiber size in the gastrocnemius, and mild knee and ankle chondromalacia. Although treadmill locomotion appeared normal, several kinematic parameters, such as stride length, amplitude, velocity and leg joint angles were altered in adult PI rats compared to shams. Using intra- and inter-group variability of kinematic parameters, PI seemed to impair the maturation of locomotion on the treadmill. In addition, PI rats exhibited spontaneous hyperactivity in open-field test. Musculoskeletal changes appeared concomitant with mild impairments in gait and posture. Our rodent model of WMD based on PI reproduces the mild motor deficits and musculoskeletal changes observed in many preterm infants with a perinatal history of hypoxia-ischemia, and contributes towards a better understanding of the interplay between brain injury, musculoskeletal histopathology and gait disturbances encountered subsequently. Virtual Reality has known exponential development in the recent years, its advantages extending over a large number of other domains, like Medicine and Automation Industry. The common treadmill is taken to another level by simulating locomotion in virtual environments, extending its usage. The purpose of this paper is to present an adapted treadmill capable of compensating the user's walking speed and direction with use in different domains, particularly in Virtual Reality. Automatic adjustments of muscle activity throughout the body are required for the maintenance of balance during human walking. One mechanism that is likely to contribute to this control is the heteronymous spinal excitation between human ankle dorsiflexors and knee extensors ( CPQ-reflex). Here, we investigated the CPQ-reflex at different walking speeds (1–6 km/h) and stride frequencies (0.6–1.3 Hz) in healthy human subjects to provide further evidence of its modulation, and its role in ensuring postural stability during walking. The CPQ-reflex was small or absent at walking speeds below 2–3 km/h, then increased with walking speeds about 3–4 km/h, and reached a plateau without any further change at walking speeds from 4 to 6 km/h. The reflex showed no modulation when the stride cycle was varied at constant speed (4 km/h; short steps versus long steps). These changes were unlikely to be only caused by changes in the background EMG activity and modifications in peripheral input, and likely reflected central modulation of transmission in the involved reflex pathways as well. It is suggested that the purpose of the reflex is to ensure knee stability at moderate-to-high walking speeds. [ABSTRACT FROM AUTHOR]

Abstract: The purpose of this paper was to describe a technique that enables three-dimensional (3D) gait kinematics to be obtained using an electromagnetic tracking system, and to report the intra-trial, intra-day/inter-tester and inter-day/intra-tester repeatability of kinematic gait data obtained using this technique. Ten able-bodied adults underwent four gait assessments; the same two testers tested each subject independently on two different days. Gait assessments were conducted on a custom-built long-bed treadmill with no metal components between the rollers. Each gait assessment involved familiarisation to treadmill walking, subject anatomical and functional calibration, and a period of steady-state treadmill walking at a self-selected speed. Following data collection, 3D joint kinematics were calculated using the joint coordinate system approach. 3D joint angle waveforms for 10 left and right strides were extracted and temporally normalised for each trial. Intra-trial, intra-day/inter-tester and inter-day/intra-tester repeatability of the temporally normalised kinematic waveforms were quantified using the coefficient of multiple determination (CMD). CMDs for joint kinematics averaged 0.942 intra-trial, 0.849 intra-day/inter-tester and 0.773 inter-day/intra-tester. In general, sagittal plane kinematics were more repeatable than frontal or transverse plane kinematics, and kinematics at the hip were more repeatable than at the knee or ankle. The level of repeatability of kinematic gait data obtained during treadmill walking using this protocol was equal or superior to that reported previously for overground walking using image-based protocols. The article presents a study which aims to find out whether the gait transitions during human locomotion occur abruptly, and if so, to determine the instant during a stride at which a transition occurs. The study concludes that gait transitions are initiated at about midstance of the transition stride. It revealed that gait transitions during human locomotion are not abrupt events.
