The manipulation of follicular waves through hormonal treatments, such as estrogen administration, plays a crucial role in optimizing in-vivo oocyte collection for assisted reproductive technologies. The present study aimed to evaluate the effect of two specific types of estrogen on follicular wave dynamics and their impact on in-vivo oocyte collection in Brown Swiss cows. Fourteen cows, in their first lactation, weighing approximately 340 kg were randomly assigned to one of two treatments including T1 (estradiol cypionate) and T2 (estradiol benzoate). Both treatments were administered at 1.2 mg of estrogen, at day 0 of the experiment. All Brown Swiss cows were provided with a diet entirely consisting of alfalfa grazing. On day 7, follicular wave dynamics were assessed using a DP-50 vet ultrasound device equipped with a 7.5 MHz transducer for transvaginal follicular aspiration guidance. Follicle counts were categorized into three size ranges including 2-4 mm (small), 4-8 mm (medium), and greater than 8 mm (large). Additionally, the quantity and quality (viable oocytes) of the collected oocytes were evaluated by the Ovum Pick Up (OPU) team for oocyte viability on day 7. The study assessed the follicular dynamics (number of follicles) and efficiency of oocyte collection (viable oocytes) in cows treated with Estradiol Cypionate (T1) and Estradiol Benzoate (T2). The average number of small, medium-sized, and large follicles size were 6.048 ± 6.037, 3.16 ± 2.01, and 0.53 ± 0.67 respectively. The total number of follicles was 9.59 ± 3.56. The mean number of viable oocytes recovered was 3.024 ± 1.66, while the mean number of non-viable oocytes was 1.47 ± 1.01. The results indicated no significant differences between treatments in the size of small, medium, and large follicles, nor in the total number of follicles and viable oocytes recovered. However, a significant difference was observed in the number of non-viable oocytes recovered, with a higher mean in T2 (1.86) compared to T1 (1.09). The results indicated an adequate follicular response and viable oocyte recovery in both treatment groups (estradiol cypionate and estradiol benzoate). Objective: We sought to identify patients at risk of incomplete transvaginal oocyte retrieval, develop a risk assessment formula to identify patients who would benefit from a transabdominal approach, and compare complication and pregnancy rates between these two approaches. Materials and Methods: In this retrospective case control study in a private in vitro fertilization center, 95 cases of women undergoing transabdominal follicular aspiration for oocyte retrieval (15 transabdominal only and 80 transabdominal and vaginal combined) were compared with 278 controls of women undergoing the transvaginal aspiration only. Transabdominal oocyte retrieval was performed when one or more ovaries could not be retrieved via the transvaginal approach. Main study outcomes included need for transabdominal retrieval, pregnancy rates, and complications. Results: A risk assessment scoring system was developed as follows: difficulty seeing ovaries on ultrasound (+4), history of pelvic surgery (+3), and body mass index of 30 kg/m2 or greater (+2). With a cutoff score of 4 or greater, the overall sensitivity is 75%, specificity is 80%, positive predictive value is 57%, and negative predictive value is 90%. No statistically significant differences were found for pregnancy rates or complications. Conclusion: The transabdominal approach is an alternative option that would increase the total number of oocytes retrieved with no statistical difference in complication or pregnancy rates. We also developed a scoring system that can serve as a useful screening tool for identifying women at increased risk of transabdominal oocyte retrieval.

This study was carried out to observe the influence of ovarian side and initiation time of first aspiration during OPU programme in HF X Sahiwal cows. Eight cows were randomly divided in to two groups (A and B) each consisting of 4 donors. Follicular aspiration was initiated on day 4 (first follicular wave) and day 13 (second follicular wave) of estrous cycle, respectively. From group-A donors, total 200 follicles were aspirated in eight sessions with a mean of 6.25 ± 2.05 per animal per session of which 52.50 (105/200) and 47.50 (95/200) per cent follicles were from left and right ovary, respectively. The mean numbers of small, medium and large follicles aspirated were 2.06 ± 0.87, 2.65 ± 0.88 and 1.53 ± 0.91 with the recovery rate of 33.0 (66/200) (3–5 mm), 42.50 (85/200) (6–9 mm) and 24.50 (49/200) (=10 mm) per cent, respectively. From group-B donors, total 179 follicles were aspirated of which 48.60 (87/179) and 51.40 (92/179) per cent follicles were from left and right ovary, respectively. The mean number of aspirated follicles per cow per session from group-B donor cows was 5.59 ± 1.03 and the mean numbers of small, medium and large follicles aspirated 1.87 ± 0.96, 2.5 ± 0.66 and 1.25 ± 0.64 with the recovery rate of 34.07 (61/179), 44.94 (80/179) and 21.22 (38/179) per cent, respectively. The number of follicles from left and right ovaries of different categories among group A and B donor cows did not differ significantly (P>0.05).
