Species mixture is promoted as a crucial management option to adapt forests to climate change. However, there is little consensus on how tree diversity affects tree water stress, and the underlying mechanisms remain elusive. By using a greenhouse experiment and a soil-plant-atmosphere hydraulic model, we explored whether and why mixing the isohydric Aleppo pine (Pinus halepensis, drought avoidant) and the anisohydric holm oak (Quercus ilex, drought tolerant) affects tree water stress during extreme drought. Our experiment showed that the intimate mixture strongly alleviated Q. ilex water stress while it marginally impacted P. halepensis water stress. Three mechanistic explanations for this pattern are supported by our modeling analysis. First, the difference in stomatal regulation between species allowed Q. ilex trees to benefit from additional soil water in mixture, thereby maintaining higher water potentials and sustaining gas exchange. By contrast, P. halepensis exhibited earlier water stress and stomatal regulation. Second, P. halepensis trees showed stable water potential during drought, although soil water potential strongly decreased, even when grown in a mixture. Model simulations suggested that hydraulic isolation of the root from the soil associated with decreased leaf cuticular conductance was a plausible explanation for this pattern. Third, the higher predawn water potentials for a given soil water potential observed for Q. ilex in mixture can—according to model simulations—be explained by increased soil-to-root conductance, resulting from higher fine root length. This study brings insights into the mechanisms involved in improved drought resistance of mixed species forests.

The intensification of droughts due to climate change is a global concern, and many plant species face increasing water deficits. Understanding the role of phenotypic plasticity in plant adaptation to these changing conditions is crucial. This research focuses on Bromopsis erecta, a dominant perennial grass in European and Mediterranean grasslands, to predict its potential adaptation to climate change. We assessed plants from shallow and deep soils (i.e., with contrasting water reserves) of a Mediterranean rangeland in southern France, and tested the effect of six years of experimentally increased summer drought compared to the ambient conditions on plant traits, survival and abundance. In both field and common garden experiments, we measured water-related traits, including static traits under non-limiting water conditions, and dynamic traits, such as rates of trait variation during drought. Trait plasticity was determined as a reaction norm to increasing soil water stress and was tested against changes in B. erecta abundance over the past decade, including the study period. Trait plasticity was detected only for leaf dry matter content (LDMC), revealing that the resource strategy of B. erecta became more conservative over less than a decade with higher LDMC and leaf thickness according to the plant economic spectrum. No plasticity was found for osmotic potential or specific leaf area. The variability of other traits was ascribed to the possible lagging effect of previous water stress and was associated more with soil depth than with previous summer drought intensity. The abundance decline of B. erecta, which dropped from 20 % to around 5 % in shallow soils, was not associated with the plasticity of LDMC but was positively correlated with variations in leaf base membrane damage, meaning unexpectedly, that plants exposed to the most severe summer drought also had the most sensitive leaf base membranes, a possible sign of maladaptive trait plasticity in the population

Background: Plants have adapted to survive seasonal life-threatening frost and drought. However, the timing and frequency of such events are impacted by climate change, jeopardizing plant survival. Understanding better the strategies of survival to dehydration stress is therefore timely and can be enhanced by the cross-fertilization of research between disciplines (ecology, physiology), models (woody, herbaceous species) and types of stress (drought, frost). Scope: We build upon the 'growth–stress survival' trade-off, which underpins the identification of global plant strategies across environments along a 'fast–slow' economics spectrum. Although phenological adaptations such as dormancy are crucial to survive stress, plant global strategies along the fast–slow economic spectrum rarely integrate growth variations across seasons. We argue that the growth–stress survival trade-off can be a useful framework to identify convergent plant ecophysiological strategies to survive both frost and drought. We review evidence that reduced physiological activity, embolism resistance and dehydration tolerance of meristematic tissues are interdependent strategies that determine thresholds of mortality among plants under severe frost and drought. We show that complete dormancy, i.e. programmed growth cessation, before stress occurrence, minimizes water flows and maximizes dehydration tolerance during seasonal life-threatening stresses. We propose that incomplete dormancy, i.e. the programmed reduction of growth potential during the harshest seasons, could be an overlooked but major adaptation across plants. Quantifying stress survival in a range of non-dormant versus winter- or summer-dormant plants, should reveal to what extent incomplete to complete dormancy could represent a proxy for dehydration tolerance and stress survival. Conclusions: Our review of the strategies involved in dehydration stress survival suggests that winter and summer dormancy are insufficiently acknowledged

Seedling root traits impact plant establishment under challenging environments. Pearl millet is one of the most heat and drought tolerant cereal crops that provides a vital food source across the sub-Saharan Sahel region. Pearl millet's early root system features a single fast-growing primary root which we hypothesize is an adaptation to the Sahelian climate. Using crop modeling, we demonstrate that early drought stress is an important constraint in agrosystems in the Sahel where pearl millet was domesticated. Furthermore, we show that increased pearl millet primary root growth is correlated with increased early water stress tolerance in field conditions. Genetics including genome-wide association study and quantitative trait loci (QTL) approaches identify genomic regions controlling this key root trait. Combining gene expression data, re-sequencing and re-annotation of one of these genomic regions identified a glutaredoxin-encoding gene PgGRXC9 as the candidate stress resilience root growth regulator. Functional characterization of its closest Arabidopsis homolog AtROXY19 revealed a novel role for this glutaredoxin (GRX) gene clade in regulating cell elongation. In summary, our study suggests a conserved function for GRX genes in conferring root cell elongation and enhancing resilience of pearl millet to its Sahelian environment.

Efficient acquisition and use of available phosphorus from the soil is crucial for plant growth, development, and yield. With an ever-increasing acreage of croplands with suboptimal available soil phosphorus, genetic improvement of sorghum germplasm for enhanced phosphorus acquisition from soil is crucial to increasing agricultural output and reducing inputs, while confronted with a growing world population and uncertain climate. Sorghum bicolor is a globally important commodity for food, fodder, and forage. Known for robust tolerance to heat, drought, and other abiotic stresses, its capacity for optimal phosphorus use efficiency (PUE) is still being investigated for optimized root system architectures (RSA). Whilst a few RSA-influencing genes have been identified in sorghum and other grasses, the epigenetic impact on expression and tissue-specific activation of candidate PUE genes remains elusive. Here, we present transcriptomic, epigenetic, and regulatory network profiling of RSA modulation in the BTx623 sorghum background in response to limiting phosphorus (LP) conditions. We show that during LP, sorghum RSA is remodeled to increase root length and surface area, likely enhancing its ability to acquire P. Global DNA 5-methylcytosine and H3K4 and H3K27 trimethylation levels decrease in response to LP, while H3K4me3 peaks and DNA hypomethylated regions contain recognition motifs of numerous developmental and nutrient responsive transcription factors that display disparate expression patterns between different root tissues (primary root apex, elongation zone, and lateral root apex).

The phenology of tropical forests is tightly related to climate conditions. In the Amazon, the seasonal greening of forests is conditioned by solar radiation and rainfall. Yet, increasing anthropogenic pressures (e.g. logging and wildfires), raise concerns about the impacts of forest degradation on the functioning of forest ecosystems, especially in a climate change context. In this study, we relied on remote sensing data to assess the contribution of solar radiation and precipitation to forest greening in mature and fire degraded forests, with a focus on the 2015 drought event. Our results showed that forest greening is more dependent on water resources in degraded forests than in mature forests. As a consequence, the expected increase in drought episodes and associated fire occurrences under climate change could lead to a long-term drying of tropical forests.

Drought is a major constraint on plant productivity globally. Sorghum [Sorghum bicolor (L.) Moench] landraces have evolved in drought-prone regions, but the genetics of their adaptation is poorly understood. Here we sought to identify novel drought-tolerance loci and test hypotheses on the role of known loci including those underlying stay-green (Stg) postflowering drought tolerance. We phenotyped 590 diverse sorghum accessions from West Africa in 10 environments, under field-based managed drought stress [preflowering water stress (WS1), postflowering water stress (WS2), and well-watered (WW)] and rainfed (RF) conditions over 4 yr. Days to 50% flowering (DFLo), aboveground dry biomass (DBM), plant height (PH), and plant grain yield components (including grain weight [GrW], panicle weight [PW] and grain number [GrN] per plant, and 1000-grain weight [TGrW]) were measured, and genome-wide association studies (GWAS) was conducted. Broad-sense heritability for biomass and plant grain yield was high (33–92%) across environments. There was a significant correlation between stress tolerance index (STI) for GrW per plant across WS1 and WS2. Genome-wide association studies revealed that SbZfl1 and SbCN12, orthologs of maize (Zea mays L.) flowering genes, likely underlie flowering time variation under these conditions. Genome-wide association studies further identified associations (n = 134; common between two GWAS models) for STI and drought effects on plant yield components including 16 putative pleiotropic associations. Thirty of the associations colocalized with Stg1, Stg2, Stg3, and Stg4 loci and had large effects. Seven lead associations, including some within Stg1, overlapped with positive selection outliers. Our findings reveal previously undescribed natural genetic variation for drought-tolerance-related traits and suggest a broad role of Stg loci in drought adaptation of sorghum.

Currently, drought stress is a major issue for crop productivity, and future climate models predict a rise in frequency and severity of drought episodes. Polyploidy has been related to improved tolerance of plants to environmental stresses. In Citrus breeding programs, the use of triploidy is an effective way to produce seedless fruits, one of the greatest consumer expectations. The current study used physiological and biochemical parameters to assess the differential responses to moderate water deficit of 3x genotypes compared to 2x genotypes belonging to the same hybridization. Both parents, the mandarin Fortune and Ellendale tangor, were also included in the experimental design, while the 2x common clementine tree was used as reference. Water deficit affects leaf water status, as well as physiological and detoxification processes. Triploid genotypes showed a better ability to maintain water status through increased proline content and photosynthetic capacity. Moreover, less oxidative damage was associated with stronger antioxidant defenses in triploid genotypes. We also found that triploidy improved the recovery capacity after a water deficit episode.

Cocoa is an important crop in Cameroon, where it is cultivated in different areas, including marginal areas, characterized by a rather low level of annual rainfall and marked dry seasons. In order to release cocoa varieties with a good level of adaptation to these marginal conditions, nine full-sib progenies, already released to farmers in other producing areas of the country, were assessed on twelve on farm cocoa plots, set up in 2006, in Mbam et Inoubou county, which is a forest–savannah transition zone. The traits assessed were mortality rate, yield and yield stability. Mortality rate and yield vary widely among trial plots and among progenies. Four out of the nine assessed progenies present a yield level significantly higher than the five others. The lowest level of yield stability (estimated by the contribution to total ecovalence) was observed in both the highest and the lowest yielding progenies. Recommendation for the large-scale release of these progenies to farmers of the county, and to other cocoa producing forest–savannah transition areas, are made, based on the results obtained from this study.

The triploidy has proved to be a powerful approach breeding programs, especially in Citrus since seedlessness is one of the main consumer expectations. Citrus plants face numerous abiotic stresses including water deficit, which negatively impact growth and crop yield. In this study, we evaluated the physiological and biochemical responses to water deficit and recovery capacity of new triploid hybrids, in comparison with diploid hybrids, their parents (“Fortune” mandarin and “Ellendale” tangor) and one clementine tree used as reference. The water deficit significantly decreased the relative water content (RWC) and leaf gas exchange (Pnet and gs) and it increased the levels of oxidative markers (H2O2 and MDA) and antioxidants. Compared to diploid varieties, triploid hybrids limited water loss by osmotic adjustment as reflected by higher RWC, intrinsic water use efficiency (iWUE Pnet/gs) iWUE and leaf proline levels. These had been associated with an effective thermal dissipation of excess energy (NPQ) and lower oxidative damage. Our results showed that triploidy in citrus enhances the recovery capacity after a water deficit in comparison with diploids due to better carboxylation efficiency, restored water-related parameters and efficient antioxidant system.

Nematodes and drought are major constraints in tropical agriculture and often occur simultaneously. Plant responses to these stresses are complex and require crosstalk between biotic and abiotic signaling pathways. In this study, we explored the transcriptome data of wild Arachis species subjected to drought (A-metaDEG) and the root-knot nematode Meloidogyne arenaria (B-metaDEG) via meta-analysis, to identify core-stress responsive genes to each individual and concurrent stresses in these species. Transcriptome analysis of a nematode/drought bioassay (cross-stress) showed that the set of stress responsive DEGs to concurrent stress is distinct from those resulting from overlapping A- and B-metaDEGs, indicating a specialized and unique response to combined stresses in wild Arachis. Whilst individual biotic and abiotic stresses elicit hormone-responsive genes, most notably in the jasmonic and abscisic acid pathways, combined stresses seem to trigger mainly the ethylene hormone pathway. The overexpression of a cross-stress tolerance candidate gene identified here, an endochitinase-encoding gene (AsECHI) from Arachis stenosperma, reduced up to 30% of M. incognita infection and increased post-drought recovery in Arabidopsis plants submitted to both stresses. The elucidation of the network of cross-stress responsive genes in Arachis contributes to better understanding the complex regulation of biotic and abiotic responses in plants facilitating more adequate crop breeding for combined stress tolerance.

Meeting food demand for the growing population will require an increase to crop production despite climate changes and, more particularly, severe drought episodes. Sorghum is one of the cereals most adapted to drought that feed millions of people around the world. Valorizing its genetic diversity for crop improvement can benefit from extensive phenotyping. The current methods to evaluate plant biomass, leaves area and plants height involve destructive sampling and are not practical in breeding. Phenotyping relying on drone based imagery is a powerful approach in this context. The objective of this study was to develop and validate a high throughput field phenotyping method of sorghum growth traits under contrasted water conditions relying on drone based imagery. Experiments were conducted in Bambey (Senegal) in 2018 and 2019, to test the ability of multi-spectral sensing technologies on-board a UAV platform to calculate various vegetation indices to estimate plants characteristics. In total, ten (10) contrasted varieties of West African sorghum collection were selected and arranged in a randomized complete block design with three (3) replicates and two (2) water treatments (well-watered and drought stress). This study focused on plant biomass, leaf area index (LAI) and the plant height that were measured weekly from emergence to maturity. Drone flights were performed just before each destructive sampling and images were taken by multi-spectral and visible cameras. UAV-derived vegetation indices exhibited their capacity of estimating LAI and biomass in the 2018 calibration data set, in particular: normalized difference vegetative index (NDVI), corrected transformed vegetation index (CTVI), seconded modified soil-adjusted vegetation index (MSAVI2), green normalize difference vegetation index (GNDVI), and simple ratio (SR) (r2 of 0.8 and 0.6 for LAI and biomass, respectively). Taro (Colocasia esculenta (L.) Schott) and sweet potato (Ipomoea batatas (L.) Lam.) are important food crops worldwide, whose productivity is threatened by climatic constraints, namely drought. Data calibration, validation, and model development of high-precision near-infrared spectroscopy (NIRS) involving multivariate analyses are needed for the fast prediction of the quality of tubers and shoots impacted by drought stress. The main objective of this study was to generate accurate NIRS models for quality assessment of taro and sweet potato accessions (acc.) subjected to water scarcity conditions. Seven taro and eight sweet potato acc. from diverse geographical origins were evaluated for nitrogen (N), protein (Pt), starch (St), total mineral (M), calcium oxalate (CaOx), carbon isotope discrimination (∆13C), and nitrogen isotopic composition (δ15N). Models were developed separately for both crops underground and aboveground organs. N, Pt, St, and M models could be used as quality control constituents, with a determination coefficient of prediction (r²pred) between 0.856 and 0.995. δ13C, δ15N, and CaOx, with r²pred between 0.178 and 0.788, could be used as an informative germplasm screening tool. The approach used in the present study demonstrates NIRS's potential for further research on crop quality under drought.

Background: Reproductive-stage drought stress is a major impediment to rice production in rainfed areas. Conventional and marker-assisted breeding strategies for developing drought-tolerant rice varieties are being optimized by mining and exploiting adaptive traits, genetic diversity; identifying the alleles, and understanding their interactions with genetic backgrounds for their increased contribution to drought tolerance. Field experiments were conducted in this study to identify marker-trait associations (MTAs) involved in response to yield under reproductive-stage (RS) drought. A diverse set of 280 indica-aus accessions was phenotyped for ten agronomic traits including yield and yield-related traits under normal irrigated condition and under two managed reproductive-stage drought environments. The accessions were genotyped with 215,250 single nucleotide polymorphism markers. Results: The study identified a total of 219 significant MTAs for 10 traits and candidate gene analysis within a 200 kb window centred from GWAS identified SNP peaks detected these MTAs within/ in close proximity to 38 genes, 4 earlier reported major grain yield QTLs and 6 novel QTLs for 7 traits out of the 10. The significant MTAs were mainly located on chromosomes 1, 2, 5, 6, 9, 11 and 12 and the percent phenotypic variance captured for these traits ranged from 5 to 88%. The significant positive correlation of grain yield with yield-related and other agronomic traits except for flowering time, observed under different environments point towards their contribution in improving rice yield under drought. Seven promising accessions were identified for use in future genomics-assisted breeding programs targeting grain yield improvement under drought. Conclusion: These results provide a promising insight into the complex genetic architecture of grain yield under reproductive-stage drought in different environments. Water deficiency affects the growth and development of many crops. Citrus is a major global fruit crop and its production is reduced by water paucity. The use of polyploid citrus rootstocks has been proposed as a strategy to improve water-deficit resilience. Although tetraploid rootstocks have been found to be more waterdeficit tolerant, the mechanisms of these dynamics are unknown, including how they respond to rapid vs. slow onset of water deficts. In this study, diploid (2x) and tetraploid (4x) Volkamer lemon (Citrus volkameriana Tan. and Pasq.) plants were evaluated under fast and slow onset water-deficit conditions to observe their water use pattern and antioxidant defence machinery. The 2x plants showed more decline in pot water and chlorophyll fluorescence Fv/Fm (pre-dawn) as compared to the 4x. Decreases in these variables were greater in fast the water deficit condition relative to the to slow water deficit condition. Leaf relative water content also decreased in relation with decrease in pot water. The decrease in leaf relative water content was greater in 2x as compared to 4x. Malondialdehyde content in leaves was also greater in 2x plants as compared to 4x, which indicated that 2x have more lipid peroxidation, and thus more oxidative stress, than 4x. Catalase activity and proline content in leaves were greater in 4x plants than 2x in fast and slow treatments. We conclude that Volkamer lemon 4x tolerate more in fast and slow water deficit condition by maintaining their water status protect against oxidative stress with high intrinsic antioxidant machinery.

Taro (Colocasia esculenta (L.) Schott) is a substantial staple food in most of the tropical regions. Prolonged exposure to drought impairs crop production worldwide. Tolerant crops have the best capability to cope and avoid drought, through phenotypic flexibility mechanisms. The water use efficiency (WUE) is well known in taro crops, but very scarce information is available relating to their nutrient efficiency (NER) in drought conditions. Our work provided pertinent information about the physiological variation of seven taro accessions subjected to seven months of drought, by recording the differences for nutrient allocation, chlorophyll canopy, biomass loss, and stress intensity. Significant relationships between control and drought treatments on WUE (+85%), total plant biomass (TPB, -26.8%), chlorophyll content index (CCI, +1.8%), and nutrient harvest index (NHI, +0.2%) were detected. Drought led to a generalized loss of TPB as drought avoidance strategy, although distinct phenotypic flexibility was observed through the root:shoot ratio (R:S) and stress index (SI) from the corm and shoot organs. The nutrient allocation from the corms to shoots, with NER increase registered in drought conditions, can be a valuable tool to complement the TPB and WUE productivity traits, to be used in taro breeding programs.

Background: The benefits of leaf stay-green for maintaining grain filling in sorghum under drought was largely demonstrated. However, its role in the stability of a dual production (grain, stem sugar) in tall sweet sorghum remains to be elucidated. This study aimed to analyze the impact of a post-anthesis drought on sugar accumulation along stem internodes in sweet and tall West-African so:rghum with variable leaf stay-green and grain yield abilities. Methods Four accessions with similar phenology were studied in two consecutive years in the field at the National Agronomic Research Centre (CNRA) of Bambey (Senegal, West Africa) under two post-anthesis water treatments (irrigated, non-irrigated). Plant morphology, stem sugar related traits, grain production, and plant leaf area (PLA) variation were assessed. Carbohydrate contents (sucrose, hexoses, starch) were determined during grain filling in the whole stem juice and at three internode levels: bottom, median, top. Analysis of variance was performed to test post-anthesis water treatment, accession, organ, year effects and their interactions on the studied traits. Results: Panicle dry weight (PDW) was not affected by drought, but strongly varied among years and accessions. The PDW/PLA ratio was negatively correlated with the variation of sucrose and hexoses at the three internodes levels. This carbohydrates reduction was mainly influenced by the PDW. The bigger the panicle the higher the carbohydrates remobilization from the stem to panicle for grain filling. This was mainly shown on accessions G3 and G11 which exhibited low stay green ability. However, G10 with low PDW/PLA ratio and showing higher stay green ability, exhibited a low reduction of total soluble sugars and sucrose and inversely higher increase of hexoses mainly at the median internode. Conclusions: This ability to better maintain green leaf area and high hexoses in the stem under post-flowering drought could be an osmoregulation mechanism to adapt to

An increasing number of candidate genes related to abiotic stress tolerance are being discovered and proposed to improve the existing cultivars of the high oil-bearing crop sesame (Sesamum indicum L.). However, the in planta functional validation of these genes is remarkably lacking. In this study, we cloned a novel sesame R2-R3 MYB gene SiMYB75 which is strongly induced by drought, sodium chloride (NaCl), abscisic acid (ABA) and mannitol. SiMYB75 is expressed in various sesame tissues, especially in root and its protein is predicted to be located in the nucleus. Ectopic over-expression of SiMYB75 in Arabidopsis notably promoted root growth and improved plant tolerance to drought, NaCl and mannitol treatments. Furthermore, SiMYB75 over-expressing lines accumulated higher content of ABA than wild-type plants under stresses and also increased sensitivity to ABA. Physiological analyses revealed that SiMYB75 confers abiotic stress tolerance by promoting stomatal closure to reduce water loss; inducing a strong reactive oxygen species scavenging activity to alleviate cell damage and apoptosis; and also, up-regulating the expression levels of various stress-marker genes in the ABA-dependent pathways. Our data suggested that SiMYB75 positively modulates drought, salt and osmotic stresses responses through ABA-mediated pathways. Thus, SiMYB75 could be a promising candidate gene for the improvement of abiotic stress tolerance in crop species including sesame.
