Basil (Ocimum basilicum L.) is a widely used culinary herb. In this study, ethanol, dichloromethane, and sunflower oil were used separately as solvents with distinct polarities for the extraction of basil aerial parts to simulate the different polarity conditions in domestic food processing. The oil extract (OE) was re-extracted with acetonitrile, and the chemical composition, antioxidant potential, and antimicrobial activities of the ethanol (EE), dichloromethane (DCME), and acetonitrile (ACNE) extracts were determined. A total of 109 compounds were tentatively identified in EE, DCME, and ACNE by HPLC–DAD/ESI-ToF-MS. Fatty acids were present in all extracts. Phenolic acids and flavonoids dominated in EE. DCME was characterised by triterpenoid acids, while diterpenoids were mainly found in ACNE. The extracts were analysed for their antioxidant capacity using the 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) assay. EE and DCME showed significant radical scavenging potential. Antimicrobial activity was explored in eight bacterial, two yeast, and one fungal species. All extracts exhibited high antifungal activity, comparable to or better than that of the commercial drug nistatin. Antibacterial activities were notable for EE and ACNE, while DCME showed no activity against bacteria in the applied concentration ranges. Sweet basil is very important in the culinary and pharmaceutical sectors because it contains volatile oils that have a variety of biological functions. Considering the economical costs and adverse effects of chemical fertilizers, attention has been turned to the use of ecofriendly and cost effective alternatives. In this regard, eggshells could be used as a natural calcium source for nutring the medicinal and aromatic plants. Therefore, the aim of this trial was to assess the productivity of sweet basil in response to different application methods of eggshells on sandy soil. In this study, eggshells were added to the soil at rates of 1, 2, and 3 g/pot, to foliage at rates of 0.5, 1, and 1.5 g/L, and to irrigation water at rates of 1, 2, and 3 g/L; a rate of 0.5 g/pot of calcium carbonate also used as a reference. Plants exposed to eggshells at 1.5 g/L foliar spray gave the maximum values of fresh and dry weights herb (172.11 and 23.48 g/plant), carbohydrates (36.74%), crude protein (16.56%), inhibition of free radical scavenging (47.61%), total phenols (15.03 mg/g), total flavonoids (2.87 mg/g), and essential oil contents (1.18% or 27.71 g/100 plants). On the other hand, plants subjected to eggshells at 0.5 g/L foliar spray produce the highest amounts of linalool (76.34%) and oxygenated monoterpenes (83.37%). Piercing-sucking insects are notorious devastating insects that cause substantial damage to crops and yield losses due to the direct feeding by both nymphs and adults furthermore through the transmission of viruses and diseases. Field experiments were conducted during the two growing summer seasons of 2021 and 2022 at Zagazig district, Sharkia Governorate, Egypt to assess the seasonal abundance of the major piercing-sucking insects and their associated natural enemies on sweet basil plants. Five piercing-sucking insect species were surveyed; Bemisia tabaci (Genn.), Aphis gossypii Glover, Nezara viridula L., Empoasca decipiens (Paoli), and Thrips tabaci Lind. Also, six predatory species were recorded; Coccinella undecimpunctata L., Chrysoperla carnea (Steph.), Orius sp., Paederus alfierii (Koch), Metasyrphus corollae (Fabr.) and Scymnus sp., and three parasitoid species were observed; Aphidius colemani (Viereck) as a primary parasitoid, Alloxysta sp. as a hyperparasitoid on A. gossypii and Trissolcus megalocephalus (Ashmead) as a parasitoid of N. viridula eggs. Sweet basil (Ocimum basilicum L.) is an important aromatic plant with high edibility and economic value, widely distributed in many regions of the tropics including the south of China. In recent years, environmental problems, especially soil salinization, have seriously restricted the planting and spread of sweet basil. However, the molecular mechanism of the salt stress response in sweet basil is still largely unknown. In this study, seed germination, seedling growth, and chlorophyll synthesis in sweet basil were inhibited under salt stress conditions. Through comparative transcriptome analysis, the gene modules involved in the metabolic processes, oxidative response, phytohormone signaling, cytoskeleton, and photosynthesis were screened out. In addition, the landscape of transcription factors during salt treatment in sweet basil was displayed as well. Moreover, the overexpression of the WRKY transcription factor-encoding gene, ObWRKY16, and the phenylalanine ammonia-lyase-encoding gene, ObPAL2, enhanced the seed germination, seedling growth, and survival rate, respectively, of transgenic Arabidopsis, suggesting that they might be important candidates for the creation of salt-tolerant sweet basil cultivars. In this study, an efficient protocol was developed to obtain multiplicated, healthy sweet basil plantlets with larger leaves and a greener color by culturing them on MS medium containing 1.0 mg/l 2iP. Chitosan and zinc oxide nanoparticles (ZnO NPs) were used as abiotic elicitors to maximize the main secondary metabolites (phenolic and flavonoid compounds and essential oil) in in vitro plantlets of sweet basil. Chitosan with a concentration of 4.0 mg/l recorded the maximum value of caffeic acid (1.189 mg/g DW), and 10 mg/l of ZnO NPs recorded the maximum content of rutin (0.936 mg/g DW). The plantlets cultured with 2.0 mg/l chitosan recorded the maximum percentage of essential oil (90.63%). 8,9-Dihydro-9-formyl cycloisolongifolene was the major compound (72.44%) and was detected in the plantlets treated with 2.0 mg/l chitosan. The inhibitory activity evaluation of α-amylase revealed that the maximum inhibitory activity was found in the plantlet control and those treated with 4.0 mg/l chitosan of the phenolic and flavonoid fractions. The maximum IC50 (7.36 ± 0.85) was found in the plantlet extract treated with 4.0 mg/l chitosan, followed by the plantlet control, which recorded 11.53 ± 0.86. Using AutoDuck 4.2 and PyMol software, it was revealed that rutin and caffeic acid compounds (the main active components) own hydrogen binding interactions and hydrophobic binding interactions with the active site of alpha-amylase from the porcine pancreas (PPA) enzyme with good binding energy (5.74 and 5.71 kcal/mol, respectively). Inoculation with rhizobacteria and feeding by herbivores, two types of abiotic stress, have been shown to increase the production of secondary metabolites in plants as part of the defense response. This study explored the simultaneous effects of inoculation with Bacillus amyloliquefaciens GB03 (a PGPR species) and herbivory by third-instar Spodoptera frugiperda larvae on essential oil (EO) yield and volatile organic compound (VOC) emissions in Ocimum basilicum plants. The density of glandular trichomes was also examined, given that they are linked to EO production and VOC emission. Herbivory increased EO content, but inoculation on its own did not. When combined, however, the two treatments led to a 10-fold rise in EO content with respect to non-inoculated plants. VOC emissions did not significantly differ between inoculated and non-inoculated plants, but they doubled in plants chewed by the larvae with respect to their undamaged counterparts. Interestingly, no changes were observed in VOC emissions when the treatments were tested together. In short, the two biotic stressors elicited differing plant defense responses, mainly when EO was concerned. PGPR did not stimulate EO production, while herbivory significantly enhanced it and increased VOC emissions. The combined treatment acted synergistically, and in this case, PGPR inoculation may have had a priming effect that amplified plant response to herbivory. Peltate trichome density was higher in inoculated plants, those damaged by larvae, and those subjected to the combination of both treatments. The findings highlight the intricate nature of plant defense mechanisms against various stressors and hint at a potential strategy to produce essential oil through the combined application of the two stressors tested here. Biological control is a natural method that suppresses pests and is a practical, eco-friendly, and cost-effective substitute for insecticide-based approaches. The present investigations were carried out to evaluate the potential impact of three natural enemies; Aphidius colemani Viereck, Chrysoperla carnea (Steph.) on Aphis gossypii Glover under semi-field circumstances and Beauveria bassiana Balsamo (Vuillemin) on Bemisia tabaci (Genn.) under field conditions. The study was done on sweet basil plants during the season of 2022 at Zagazig district, Sharkia Governorate, Egypt. The results revealed that the maximum parasitism percentage of A. colemani was achieved at a rate of 12 parasitoids/cage, whereas the highest percentage of emergence was recorded at a rate of three parasitoids/cage. Data also showed that the reduction percentages of cotton aphid numbers declined as the rate of C. carnea increased. Moreover, B. bassiana caused from 49.19 to 95.20% reduction in the mean numbers of B. tabaci. Therefore, A. colemani, C. carnea, and B. bassiana are being promoted for the management of the major piercing-sucking insects on sweet basil plants under Egyptian field conditions. AbstractOcimum basilicum is an important medicinal plant and culinary herb generally known as sweet basil (SB). These plants are effective radical scavengers, that have been employed in treatment of nervous system disorders, and thus, could be beneficial for the management of neurodegenerative diseases (NDs). Current clinical treatments for NDs present several side effects, therefore, there is need to develop new treatments that can mitigate these deadly diseases. Hence, this study investigated the neuroprotective activities of SB leaf and seed in aluminum chloride (AlCl3)-induced toxicity in Drosophila melanogaster. HPLC characterization of the leaves and seeds were carried out. AlCl3-diet was used to induce neurodegeneration and treated flies received SB leaf and seed extracts-supplemented diet. Survival and locomotor performance activities/levels of oxidative biomarkers [reactive oxygen species (ROS), thiobarbituric acid reactive species (TBARS), total thiol, catalase, superoxide dismutase (SOD) and glutathione-S-transferase (GST)], enzymes linked with neurodegeneration (acetylcholinesterase (AChE) and monoamine oxidase (MAO)) were investigated. SB leaf had significantly (p < 0.05) higher polyphenol contents; gallic acid and P-coumaric acid were the most abundant polyphenol in the leaf and seed respectively. Percentage survival and locomotor rates, level/activities of total thiol, catalase, SOD and GST were significantly (p < 0.05) reduced while ROS, TBARS, AChE and MAO activities were significantly (p < 0.05) increased in AlCl3-diet-fed flies. Treatment with SB leaf and seed diet lessened these observed impairments. However, SB leaf had better neuroprotective activities that could be related to the observed higher phenolic constituents. AbstractOcimum basilicum L. var. basilicum (Sweet Basil) is an aromatic herb belonging to the family Lamiaceae and is known for its medicinal uses. It is commonly used in traditional medicine for its therapeutic value, including anti-allergic, anti-inflammatory, antioxidant, antitumor, and antimicrobial properties. In this study, we generated the complete chloroplast genome sequence of O. basilicum var. basilicum using Illumina paired-end sequencing data. The chloroplast genome was 152,407 bp in length, containing a large single-copy (LSC) region of 83,409 bp and a small single-copy region (SSC) of 17,604 bp, separated by a pair of inverted repeats (IRs) of 25,697 bp. The genome contained 134 genes, including 89 protein-coding, 37 tRNA, and eight rRNA genes. Nine genes had one intron, two genes had two introns, and others did not have any intron. Overall GC content of the chloroplast genome was 38%, while that of LSC, SSC, and IR regions was 35.9%, 31.6%, and 43.1%, respectively. Phylogenetic analysis of the chloroplast genomes revealed that O. basilicum var. basilicum was closely related to Ocimum basilicum from the Ocimum species.

Basil (Ocimum basilicum L.) contains abundant nutrients and is considered an economically important edible vegetable. The optimal nutrient levels will increase the productivity and basil quality. However, prominent research on basil regarding the diagnostic nutrient deficiency standard and the corresponding nutrient uptake is still scarce. To this end, the basil plants were hydroponically cultured and subjected to one of 14 nutrient solution treatments, corresponding to the omission of a single nutrient element (designated as -N, -P, -K, -Ca, -Mg, -NH4+, -NO3−, -S, -Fe, -Mn, -B, -Zn, -Mo, and -Cu) and a complete nutrient solution (CS) as the control. The most common nutrient deficiency symptoms were chlorosis, stunted roots and growth, and even leaf necrosis and abscission, in particular of -N, -P, -NO3−, and -Fe. We also found that basil is a NH4+-sensitive species. The photosynthetic capacity (photosynthesis pigments, Fv/Fm ratio, and greenness index) was disturbed to varying degrees when a single nutrient was omitted from the nutrient solution. Additionally, the omission of a specific single nutrient confers significant differences in the tissue nutrients, regardless of the macronutrients and micronutrients considered. Concomitantly, multivariate analysis suggested the correlations among certain important nutrients were distinctly different under different treatments (correlation analysis); the influences of different nutrient deficiencies on the tissue nutrient concentrations showed similarity (principal component analysis). Collectively, the growth, physiological, and biochemical changes studied in this trial not only improved our knowledge for diagnosing nutrient deficiency symptoms for practical cultivation but also provided a comprehensive understanding of the internal nutrient associations in basil. Essential oil (EO) from the sweet basil (Ocimum basilicum L.) grown in the Jhum cultivations located in Bangladesh was screened for chemical composition, antioxidant, and antimicrobial activities. EO yield from the Jhumcultivar O. basilicum was 1.55% (v/w). Analysis of EO indicated the presence of several bioactive compounds, among which Geranial (35.5%) and cis-citral (26.2%) are of significant content. The EO showed antioxidant activities inhibiting DPPH radical with a mean value of 45.7% at 2.4 mg mL-1 of EO. The EO has susceptibility against Gram-positive and Gram-negative bacteria (Escherichia coli, Salmonella Typhi, Vibrio cholerae, Staphylococcus aureus, Bacillus cereus, and Micrococcus spp.), with a notable activity against the Gram-positive bacteria. Sweet basil is a perishable green herb that has a short shelf life after harvest. Techniques to extend its shelf life and prolong its storability may be commercially beneficial. In this study, the impact of different ultrasound powers on the postharvest quality and bioactive compounds of sweet basil during storage was investigated. The sampled basil leaves were immersed in an ultrasonic bath (40 kHz) at 200 or 400 W for 3 min at ambient temperature (28 ℃). The control samples were treated with tap water. All of the treatments were packed in a basket covered with a polyethylene sheet and kept at 13 ℃. Changes in the physical and chemical properties were recorded every 3 h to determine the impact of the ultrasound treatment on prolonging the postharvest quality and maintaining the bioactive compounds of the sweet basil leaves. The results showed that ultrasound at 400 W significantly reduced the weight loss and respiration rate of the sweet basil leaves. Additionally, ultrasound at 400 W increased the total phenolics, total flavonoids, antioxidant activity, and vitamin C content, which were related to the activity of phenylalanine ammonium lyase and ascorbate peroxidase, compared with the control. Introduction Drought is one of the most important environmental stressors that adversely affects agricultural products, especially in arid and semi-arid regions. Using Trichoderma fungus along with biopolymers such as chitosan is one of the ways to reduce drought stress. Trichoderma fungus as plant growth-promoting fungus is the most common fungal and soil-modifying species that are able to directly with plant roots in the rhizosphere and improve growth as well as biological control of living stresses such as pathogenic fungi and non-living stresses such as drought, salinity and heavy metals. On the other hand, one of the effective ways to protect the plant in conditions of low irrigation is the use of anti-transpirants, including the biostimulant of chitosan, which markedly limits transpiration from the plant surface. The anti-transipirants action of chitosan can be attributed to the involvement of chitosan in the abscisic acid pathways, which closes the stomata and thus reduces transpiration. Chitosan is readily soluble in water and organic acids. Therefore, it can be used in various methods such as mixing with soil, foliar spraying and impregnation with seeds in agriculture.Material and Methods This research was conducted in a split factorial arrangement based on randomized complete block design. The main plot factor was irrigation interval in three levels (two days as normal irrigation and three and four days as deficit irrigation conditions) and sub-plots were inoculated with T. longibrachiatum at two levels (inoculation and uninoculated control) and chitosan at three levels (0, 0.2 and 0.4 g/L). Each experimental plot consisted of three planting lines two and a half meters long and one meter wide .T. longibrachiatum was obtained from Tabarestan Agricultural Genetics and Biotechnology Research Institute. The first irrigation was done simultaneously with planting basil. Up to one month after sowing the seeds (six to eight leaf stage of plants), the plots were irrigated evenly with tubes and from this stage onwards, irrigation treatments were applied. Pesticides and herbicides were not used during the experiment and weed control was done manually. Chitosan was prepared from Sarina Teb store and prepared in three levels of zero, 0.2 and 0.4 g/l and sprayed in three stages: vegetative, before flowering and 50% flowering.Results and Discussion The results showed that with increasing the irrigation period from two to four days, the morphological traits of basil, such as root length and stem length, leaf dry weight, root, stem and dry matter yield decreased. Also, physiological traits of basil such as carotenoids and chlorophyll meter increased while chlorophyll a, b and total chlorophyll decreased. Application of 0.2 g/L of chitosan inoculated plants increased chlorophyll b content by 68%. The highest percentage and yield of essential oil in both normal and irrigation deficit conditions were obtained when plants inoculated with Trichoderma and foliary sprayed by chitosan. The highest percentage and yield of essential oil were observed with an average of 0.88 and 42.87% in normal irrigation conditions, application of Trichoderma and zero level of chitosan, respectively. According to the results, increasing the irrigation cycle along with chitosan application and fungal inoculation increased the percentage and yield of essential oil. The study aimed to optimize the yield and ultrasonic extraction (UAE) conditions of phenolics from citral genotype basil leaves using response surface methodology (RSM) and to produce a functional beverage using the optimized extract (OE). To develop a functional drink formulation (FDM), OE was enriched with chlorophyll, ascorbic acid, and riboflavin. The model was found to be suitable for accurately predicting experimental results. The optimum conditions were 100% for amplitude, 45 min for extraction time, and 1.06 g/100 mL for plant/solvent ratio. The TPC (mg GAE/g extract), radical scavenging (IC50 mg/mL), and antidiabetic activities (IC50 mg/mL) were 262.40, 0.14, and 2.30 for the OE and, 292,176, 0.156, and 1.67 for FDM, respectively. After the enrichment process, the total chlorophyll of FDM was 3 times higher than OE, whereas total carotenoid content was negligible for both. Additionally, FDM was accepted by panelists in relation to overall acceptability, color, texture, flavor, and aroma. The results of the study revealed that OE and FDM have promising antioxidant and antidiabetic activities due to their rich bioactive and fortified compounds. Sweet basil is a popular culinary herb used in many cuisines around the world and is widely grown commercially for retail as a live potted plant. However, basil is easily damaged by temperatures below 12 °C meaning plants must be transported from the grower to the retailer in a warm transport chain, adding considerable commercial cost in temperate countries. Improvement of chilling tolerance has been demonstrated in post-harvest crops such as tomato fruits and, indeed, fresh cut basil, by manipulation of the red:far red ratio of light provided to plants throughout the photoperiod and for a significant duration of the growing process in controlled environment chambers. We tested the effectiveness of periodic short-duration end-of-production supplementary far red light treatments designed for use with basil plants grown in a large scale commercial glasshouse for the live potted basil market. Mentha crispa L. is one of Vietnam’s most precious folk medicines and is also a spice used in many delicious dishes in Vietnam and around the world. The use of Mentha crispa L. leaf extract as a reducing agent for Ag+ ions to synthesize silver nanoparticles is discussed in this paper. Single silver nanospheres were dispersed in biological media and their sizes were controlled in the range of 20-80 nm. Factors affecting particle size and shape such as extract concentration, AgNO3 concentration, reaction time, and temperature were investigated to determine the optimal parameters for particle synthesis. The optical properties, dispersion, crystal structure, morphology, and sizes of silver nanoparticles were investigated through UV-Vis absorption spectroscopy, dynamic light scattering (DLS) measurements, X-ray diffraction, and transmission electron microscopy (TEM). The functional groups on the L.AgNPs were determined by Fourier-transform infrared (FTIR) transmittance spectra. The synthesised silver nanoparticles were used to treat methylene blue (MB) dye, the main component of dye wastewater, based on their photocatalytic properties. The results show that in the presence of L.AgNPs, the photodegradation efficiency of NaBH4 or H2O2 reducing agents is much higher, up to 95% of illumination.

Sweet basil is a popular culinary herb used in many cuisines around the world and is widely grown commercially for retail as a live potted plant. However, basil is easily damaged by temperatures below 12 °C meaning plants must be transported from the grower to the retailer in a warm transport chain, adding considerable commercial cost in temperate countries. Improvement of chilling tolerance has been demonstrated in post-harvest crops such as tomato fruits and, indeed, fresh cut basil, by manipulation of the red:far red ratio of light provided to plants throughout the photoperiod and for a significant duration of the growing process in controlled environment chambers. We tested the effectiveness of periodic short-duration end-of-production supplementary far red light treatments designed for use with basil plants grown in a large scale commercial glasshouse for the live potted basil market. Ocimum basilicum (sweet basil) is an economically important aromatic herb; in Italy, approximately 1000 ha of "Genovese-type" basil are grown annually in greenhouses and open fields and are subjected to Downy Mildew (DM) disease, caused by Peronospora belbahrii, leading to huge crop losses. Mutation of the Susceptibility (S) gene DMR6 (Downy Mildew Resistant 6) has been proven to confer a broad-spectrum resistance to DM. In this work, an effective Genome Editing (GE) approach mediated by CRISPR/Cas9 in O. basilicum 'Italiko', the élite cultivar used to produce "Pesto Genovese D.O.P", was developed. A highly efficient genetic transformation method mediated by A. tumefaciens has been optimized from cotyledonary nodes, obtaining 82.2% of regenerated shoots, 84.6% of which resulted in Cas9+ plants. Eleven T0 lines presented different type of mutations in ObDMR6; 60% were indel frameshift mutations with knock-out of ObDMR6 of 'FT Italiko'. Analysis of six T1 transgene-free seedlings revealed that the mutations of T0 plants were inherited and segregated. Based on infection trials conducted on T0 plants, clone 22B showed a very low percentage of disease incidence after 14 days post infection. The adoption of suitable irrigation levels (IRL), humic acid doses (HAD) and soil mulching (SM) are important tools for improving the morpho-physiological and biochemical traits of medicinal and aromatic plants. Ocimum basilicum L. cultivated under four IRL: IRL 100 = 100% FC–IRL 75 = 75% FC–IRL 50 = 50% FC–IRL 25 = 25% FC and four HAD: HA 0 = 0.0 Lha−1–HA 10 = 10.0 Lha−1–HA 20 = 20.0 L ha−1–HA 40 = 40.0 L ha−1 were applied in order to evaluate morpho-physiological and biochemical traits under the ecological conditions of Eskişehir in 2016 and 2017. A second trial was conducted with black plastic soil mulch (SM) and compared with the control plots (CP) in 2016. The experiment was arranged in a randomized complete block design with split plots and three replications. The plant height (PH), fresh herb yield (FHY), dry herb yield (DHY), dry leaf yield (DLY), protein ratio (PR), and main essential oil compounds (MEOC) of Ocimum basilicum L. increased and the essential oil ratio (EOR) and essential oil yield (EOY) decreased with increasing IRL (IRL 100 and IRL 75). FHY (7268.3 and 7472.7 kg ha−1) and DLY (635.3 and 637.5 kg ha−1) increased with increasing HAD (HA 20 and HA 40) compared to the values of FHY and DLY at HA 0 (6852.6 and 587.0 respectively). The SM application at IRL 50 increased the PH between 8.8 and 13.5%, FHY 11.7 and 16.7%, DLY 22.5 and 29.2%, and at IRL 75 the EOY between 20.0 and 23.9% compared to CP. In addition, PH, FHY, DLY, and EOY were highest at HA 40 and HA 20. The MEOC (linalool, 1,8-cineole, and (E) – β-bergamotene) under SM were more pronounced at IRL 25 and IRL 50 compared to CP. Aromatic plants are a sustainable supply of flavouring ingredients that apply to the food, fragrance, and medicinal industries. To determine the effects of compost manure fertilization (2, 4, and 6 tons/ha) along with spraying of methyl jasmonate (20 and 30 ppm) and potassium silicate (1000 and 2000 ppm) on the vegetative growth and chemical components of sweet basil (Ocimum basilicum L.) plants, a field experiment was conducted at the Experimental Farm of El-Quassassin Horticultural Research Station, Ismailia Governorate, for two succeeding seasons (2021 and 2022). Four replications of a split-plot design with three treatments of compost manure fertilization in the main plot and three treatments of stimulants in the sub plot. The data collected showed that the foliar treatments of 2000 ppm K-silicate combined with 6.0 tons of compost manure per hectare of sweet basil plants produced themost significant levels of these traits. However, the highest values for plant height were from the application of 6 tons/ha of compost manure combined with 30 ppm of methyl jasmonate. Sweet basil (Ocimum basilicum L.; Fam. Lamiaceae) is an annual herbaceous plant with a high economic value used in folk medicine, pharmacology, and food production. In Italy, most of the varieties are used to produce the famous "pesto" sauce; however, almost all of them are susceptible to basil downy mildew (BDM) disease, strongly decreasing the growth of the fresh leaves and the survival of the whole plant. Nowadays, CRISPR/Cas9 technology is recognized to be a prominent way to enhance basil genetic breeding. In this work, we present an optimized protocol for in vitro direct regeneration of an elite cultivar, which is the major limiting factor for the transformation of O. basilicum. Regeneration has been obtained from different explants (leaves, cotyledons, cotyledonary nodes); the highest frequency has been obtained from cotyledonary nodes of seedlings germinated on MS medium containing TDZ. Background: Sweet basil (Ocimum basilicum) is one of the most significant aromatic plants in Turkiye. Recently, a new pathogen induced symptoms were discovered and identified as basil downy mildew caused by Peronospora belbahrii Thines. The pathogen has been introduced into the country and it has quickly become the most damaging disease in basil cultivation. The purpose of this study was to investigate the molecular and morphological properties of the causal organism of downy mildew observed on sweet basil and determine the disease incidence and prevalence in Antalya province. Methods and results: According to morphological characteristics (conidia, conidiophores) disease was determined as downy mildew caused by P. belbahrii. Pathogenicity tests were performed by spraying with a sporangial suspension of P. belbahrii (1 × 105 sporangia/mL). After 1 week, all inoculated plants exhibited characteristic downy mildew symptoms on their leaves, whereas non-inoculated control plants remained disease-free. All molecular analyses involving the internal transcribed spacer region were amplified using Nested PCR with primer pairs ITS4 and ITS6 for the first round and ITS4 and DC6 for the second round. Resulting sequences of all the nested PCR products had 99% similarity with P. belbahrii isolates. Disease incidence was 22.4–70.2% of sweet basil cultivation area in Antalya province. Conclusions: Based on the molecular analysis, morphological characteristics and pathogenicity tests the pathogen was identified as P. belbahrii. The current research work focuses on the characterization and biological activities (antibacterial, antioxidant and hemolytic) of essential oils isolated from the flowers of Sweet basil (Ocimum basilicum L.) from three different regions of Punjab, Pakistan. The yield of hydro distilled sweet basil flower essential oil (SBFEO) ranged from 0.40 to 0.54 % (w/w) across the regions. The SBFEOs showed considerable antibacterial activity against S. aureus, E. coli and K. oxytoca. The results of DPPH (59.33-71.31%), TPC (13.46 to 57.75 GAE mg/100g) and TFC assays (3.99 to 21.72 CE mg/100g) also showed that SBFEOs have good antioxidant activity. Similarly, lower cytotoxic/ hemolytic effect (1.85 to 3.90%) of the tested SBFEOs also supported their safer nature as ingredient in food/medicinal products. GC/MS analyses of the oils reveal that estragole (56.57%), α-linalool (46.25%) and τ-cadinol (52.54%) are present as major components. The findings of this research work depicted significant variations in the yield, composition and biological activities of SBFEOs that can be linked to varying morphological features of the plants harvested from different agro-climatic regions. Hydroponic sweet basil (O. basilicum L.) farming uses a recirculating nutrient solution that may spread waterborne microbial contamination including algae. Plasma, the fourth state of matter, generates antimicrobial reactive oxygen and nitrogen species when exposed to water. The objective of this work was to study the effect of plasma-treated water-based nutrient solution on plant growth and in reduction of algae. Basil plants were grown in isolated ebb and flow hydroponic systems (under monitored environmental conditions) using nutrient solution (NS) and plasma-activated nutrient solution (PANS) with two separate treatments: the same irrigation solutions were used in the growth cycle (Treatment 1: NST1 and PANST1 once at the beginning growth cycle) and new irrigation solutions at every week of the growth cycle (Treatment 2: NST2 and PANST2). The plant growth parameters (height, fresh and dry weight, number of branches and nodes, root length, leaf index), quality parameters (color, texture, aroma, and tissue nutrients concentration), and algae concentrations were measured. Compared to NST1, plants grown on PANST1 were significantly taller (up to 12%), had a higher fresh mass (up to 29%) and dry mass (up to 45%), and had a higher greenness value (up to 28%). Algae growth was significantly reduced in the PANST2 reservoir (up to 24%) compared to the NST2 reservoir. It was confirmed that Treatment 1 significantly improved the yield, morphology, and quality of sweet basil plants, while Treatment 2 was best suited to decreasing algae concentration in the hydroponic environment. This preliminary study indicated that PANS could improve the quality and growth of sweet basil in hydroponic farming while controlling the algae growth in the growing environment. Basil (Ocimum basilicum L.) contains abundant nutrients and is considered an economically important edible vegetable. The optimal nutrient levels will increase the productivity and basil quality. However, prominent research on basil regarding the diagnostic nutrient deficiency standard and the corresponding nutrient uptake is still scarce. To this end, the basil plants were hydroponically cultured and subjected to one of 14 nutrient solution treatments, corresponding to the omission of a single nutrient element (designated as -N, -P, -K, -Ca, -Mg, -NH4+, -NO3−, -S, -Fe, -Mn, -B, -Zn, -Mo, and -Cu) and a complete nutrient solution (CS) as the control. The most common nutrient deficiency symptoms were chlorosis, stunted roots and growth, and even leaf necrosis and abscission, in particular of -N, -P, -NO3−, and -Fe. We also found that basil is a NH4+-sensitive species. The photosynthetic capacity (photosynthesis pigments, Fv/Fm ratio, and greenness index) was disturbed to varying degrees when a single nutrient was omitted from the nutrient solution. Additionally, the omission of a specific single nutrient confers significant differences in the tissue nutrients, regardless of the macronutrients and micronutrients considered. Concomitantly, multivariate analysis suggested the correlations among certain important nutrients were distinctly different under different treatments (correlation analysis); the influences of different nutrient deficiencies on the tissue nutrient concentrations showed similarity (principal component analysis). Collectively, the growth, physiological, and biochemical changes studied in this trial not only improved our knowledge for diagnosing nutrient deficiency symptoms for practical cultivation but also provided a comprehensive understanding of the internal nutrient associations in basil.

This study aimed to compare the chemical composition of the essential oils obtained from the leaves and flowers of three Ocimum species cultivated in north-western Poland. The content of macro- and microelements as well as dry matter in the leaves and flowers of studied basils were also determined. Essential oils were isolated by hydrodistillation in a Clevenger-type apparatus from Ocimum x citriodorum Vis. (lemon basil), Ocimum americanum L. (lime basil) and Ocimum basilicum L. (cv. Sweet Dani Lemon) and analyzed by Gas Chromatography -- Mass Spectrometry (GC/MS). The principle components of Lemon basil were linalool (35.34% in leaves and 39.54% in flowers), geranial (17.37% in leaves and 8.06% in flowers) and neral (13.56% in leaves and 6.49% in flowers). Geranial (27.67% in leaves and 18.43% in flowers), neral (21.70% in leaves and 14.70% in flowers), caryophyllene oxide (8.83% in leaves and 13.35% in flowers) and linalool (9.68% in leaves and 12.11% in flowers) dominated in Lime basil. Similarly, the Sweet Dani Lemon cultivar was rich in geranial (27.14% in leaves and 11.55% in flowers), neral (20.97% in leaves and 9.42% in flowers) and caryophyllene oxide (8.20% in leaves and 7.12% in flowers). Contents of macroelements in three Ocimum species were in the range: P (0.54 - 0.83 g kg-1), K (108.37 - 170.50 g kg-1), Ca (15.38 - 233.53 g kg-1), Mg (1.25 - 15.89 g kg-1) and Na (1.96 - 2.09 g kg-1). The concentration of microelements in the examined samples decreased as follows: Fe > Zn > Mn > Cu. The highest amounts of iron were determined in the leaves and flowers of O. americanum L. (500.37 mg kg-1 and 543.73 mg kg-1, respectively), while the highest content of zinc was determined in the flowers of O. basilicum L. (244.47 mg kg-1) and O. americanum L. (227.33 mg kg-1). The leaves of O. basilicum L. were the richest in manganese (117.3 mg kg-1), while flowers accumulated the greatest amounts of copper (68.55 mg kg-1). [ABSTRACT FROM AUTHOR]

In India, numerous cultivars and varieties of O. basilicum have been released for commercial cultivation, which are rich in specific essential oils. In this study, the effects of phenological stages and plant density on the productivity of high-quality essential oil of CIM-Sukhda cultivar of O. basilicum and the effects of drying methods on essential oil quality were investigated. In the present study, four plant spacing (45cm × 30cm, 45cm × 40cm, 45cm × 45cm, 45cm × 60cm), four growth stages (vegetative, semi-flowering, full flowering and seed formation) and drying up to one month were compared. A total of 20 compounds were identified by GC and GC-MS, with linalool (66.9-84.2%), geranial (1.1-9.0%), neral (0.8-7.0%) and 1,8-cineole (0.5-3.4%) as the main constituents. The results showed that both plant density and growth stages affected herb and essential oil yield. The essential oil yield of CIM-Sukhda plants is higher at the time of full flowering (106.2 kg ha−1) with > 75.0% linalool content in the essential oil. [ABSTRACT FROM AUTHOR]

The use of phytotherapy in aquaculture is rapidly increasing for more environmentally sustainable measures. The present work aimed to investigate the effects of different dietary levels of sweet basil, Ocimum basilicum, leaves ethanol (BEE) or aqueous (BAE) extracts (0, 200, 300, and 500 mg/kg) on Oreochromis niloticus growth, digestive enzyme activities, hemato-biochemical profile, antioxidant status, immune responses, and resistance against Streptococcus agalactiae. Oreochromis niloticus fingerlings (40.00 ± 1.00 g/fish, n = 210) were randomly divided into seven triplicated groups (control, BEE200, BEE300, BEE500, BAE200, BAE300, and BAE500) and fed the experimental diets for 8 weeks. The results revealed that dietary inclusion of BEE and BAE significantly improved final weight, weight gain, survival, and digestive enzyme activities. The growth response revealed a dose-dependent in favor of fish fed BEE. The hemato-biochemical biomarkers showed a significant improvement in RBCs, WBCs, hemoglobin, and lymphocyte, and a significant decrease in aminotransferases, creatinine, and cholesterol levels with dietary basil extracts. The cellular and humoral immune responses (phagocytic activity, phagocytic index, lysozyme activity, immunoglobulin) were significantly improved with increasing BEE and BAE in a dose-dependent manner. The expression of IL-1β and TNF-α genes were increased, while TGF-β was decreased in a dose-dependent manner and BEE500 have the highest expression. The antioxidant balance was improved with increasing basil extracts supplementation, and the BEE500 group showed the best antioxidant status. Dietary BEE and BAE increased Nile tilapia resistance to S. agalactiae. In conclusion, the dietary supplementation of both ethanolic and aqueous extracts could improve the growth performance and physiological, and immune-antioxidant status of Nile tilapia.

As environmental pollution and the global population increase, and the COVID-19 pandemic becomes more severe, demands for indoor farming, especially home food gardening, have also increased. However, most research thus far has focused on large-scale food production, with very few studies having been conducted at the household scale. Also, the devices cultivating household crops with control systems in a continuous way, which minimize fluctuations of environmental conditions, have been rarely developed. Therefore, this study aimed to design a household cultivation system for sweet basil that is automatically and continuously controlled by fuzzy logic with a Raspberry Pi4. Three inputs (temperature, humidity, and growth stage) and seven outputs (fan, humidifier, heater 1, heater 2, LED red, green, and blue) were used with six rules, ensuring that three lights operated independently upon three growth stages. Simulation and actual operation were carried out, resulting in an appropriately controlled system that operated with few defects. In the case of an operation of the input variable, temperature and humidity were maintained at an average of 21.24 °C and 75.58%, respectively, and the LED operation for the growth stage was confirmed to be flawless. For verification of the designed fuzzy system, a comparison between the simulation and actual operation was performed to examine differences and identify problems. To this end, Pearson's correlation coefficients were used, and the direction of correction of the fuzzy logic system was proposed. Through these results, the feasibility of a home cultivation system using fuzzy logic was demonstrated, and it is expected that further studies applying it will be conducted in the future. Background: Mild cognitive decline, a common issue in aging, affects memory, learning, and attention. Nutrition can influence cognition, and research indicates that Ocimum sp. (holy basil and sweet basil) leaf extracts may enhance cognition in rodents and humans. However, these studies do not address whether these benefits extend to fresh or dry leaves consumed in typical human diets, along with physiological aging. Aim: To investigate the effects of sweet basil supplementation on cognition in mature and aged female mice. Methods: Female C57bl mice were divided into four groups: 8-month-old mature adults and 18-month-old aged adults, each receiving either a control or supplemented diet. The supplemented diet included a mix of standard chow and fresh basil leaves, administered for 2-8 months. Cognitive and behavioral assessments were conducted using the novel object recognition (NOR), Morris water maze (MWM), and elevated plus maze (EPM) tasks, focusing on memory, learning, and anxiety. Results: No cognitive improvement was observed in mature mice. However, aged mice receiving long-term basil supplementation showed enhanced discrimination in NOR and stayed closer to the absent platform in MWM compared to nonsupplemented controls. While aging mice exhibited reduced anxiety-like behavior in EPM, basil supplementation prevented this reduction. Conclusion: Basil supplementation appears beneficial in elderly mice, potentially preventing age-related cognitive decline and behavioral changes. These findings support the benefits of basil consumption in cognition and underscore its potential role in promoting healthy aging. The mechanism through which gravity influences the biosynthesis of essential oils in herbs is an important issue for plant and space biology. Sweet basil (Ocimum basilicum L.) seedlings were cultivated under centrifugal hypergravity conditions at 100 g in the light, and the growth of cotyledons, development of glandular hairs, and biosynthesis of essential oils were analyzed. The area and fresh weight of the cotyledons increased by similar amounts irrespective of the gravitational conditions. On the abaxial surface of the cotyledons, glandular hairs, where essential oils are synthesized and stored, developed from those with single-cell heads to those with four-cell heads; however, hypergravity did not affect this development. The main components, methyl eugenol and 1,8-cineole, in the essential oils of cotyledons were lower in cotyledons grown under hypergravity conditions. The gene expression of enzymes in the phenylpropanoid pathway involved in the synthesis of methyl eugenol, such as phenylalanine ammonia lyase (PAL) and eugenol O-methyltransferase (EOMT), was downregulated by hypergravity. Hypergravity also decreased the gene expression of enzymes in the 2C-methyl-d-erythritol 4-phosphate (MEP) pathway involved in the synthesis of 1,8-cineole, such as 1-deoxy-d-xylulose-5-phosphate synthase (DXS) and 1,8-cineole synthase (CINS). The aim of this study was to assess the effect of inoculated endophytic bacteria on the concentrations of vitamins E (tocopherols), K (phylloquinone), B1 (thiamine), B2 (riboflavin), C (ascorbic acid) and the peroxyl radical scavenging capacity of hydroponically grown sweet basil. Endophytic strains were all isolated from hydrocarbon-stressed herbaceous plants or from basil showing superior growth. Plants inoculated with the endophytes displayed up to 40% increase (p < 0.05) in the concentration of the reduced form of vitamin C relative to control [0.56 mg/g fresh weight (FW)] which indicated less oxidative stress in the presence of endophytes. In the case of γ-tocopherol, the highest content [25.8 μg/g of fresh weight (FW)] in inoculated basils was significantly higher compared to control plants (18.5 ± 1.2 μg/g FW) (p < 0.05). Antioxidant activity (ROO∙ radicals scavenging) was as high as 94 ± 4 μM Trolox equivalents (TE)/g FW vs. 53 ± 5 μM TE/g FW for the control basil. Concentrations of vitamins C, B1, and B2 were not affected by most strains. The results showed that endophytic bacteria have the capacity to alter free radical quenching capacity and vitamin concentrations in basil plants and, that their effect is strain and nutrient dependent.

Ocimum basilicum (sweet basil) is an economically important aromatic herb; in Italy, approximately 1000 ha of “Genovese-type” basil are grown annually in greenhouses and open fields and are subjected to Downy Mildew (DM) disease, caused by Peronospora belbahrii, leading to huge crop losses. Mutation of the Susceptibility (S) gene DMR6 (Downy Mildew Resistant 6) has been proven to confer a broad-spectrum resistance to DM. In this work, an effective Genome Editing (GE) approach mediated by CRISPR/Cas9 in O. basilicum ‘Italiko’, the élite cultivar used to produce “Pesto Genovese D.O.P”, was developed. A highly efficient genetic transformation method mediated by A. tumefaciens has been optimized from cotyledonary nodes, obtaining 82.2% of regenerated shoots, 84.6% of which resulted in Cas9+ plants. Eleven T0 lines presented different type of mutations in ObDMR6; 60% were indel frameshift mutations with knock-out of ObDMR6 of ‘FT Italiko’. Analysis of six T1 transgene-free seedlings revealed that the mutations of T0 plants were inherited and segregated. Based on infection trials conducted on T0 plants, clone 22B showed a very low percentage of disease incidence after 14 days post infection. The aromatic profile of all in vitro edited plants was also reported; all of them showed oxygenated monoterpenes as the major fraction.

Vegetable grafting has been applied to fruiting crops, yet only to a limited extent in leafy greens and herbs which may also benefit from grafting. In this study, we examined the effect of reciprocal grafting two sweet basil (Ocimum basilicum L.) cultivars of differing vigor on plant growth and leaf mineral nutrient concentration to test whether differences in growth exist due to grafting and whether such differences are due to altered plant nutrient status in two trials. Two cultivars 'Nufar' (NU), a high vigor cultivar, and 'Dolce Fresca' (DF), a cultivar developed for compact growth, were selected. Four grafted treatments (scion/rootstock) were created by self-grafting (NU/NU and DF/DF) and reciprocal inter-cultivar grafting (DF/NU and NU/DF). Un-grafted plants (ug-NU and ug-DF) served as controls. Following grafting, plants were grown for 26 or 21 days in a greenhouse. DF rootstocks decreased NU shoot dry mass (19-29%) and stem length (12%) compared to ug-NU in both trials, while dry root mass was reduced (28%) in the second trial. In contrast, NU rootstocks did not affect DF growth in the first trial but significantly decreased dry shoot (18%) and root (31%) mass, compared to ug-DF in the second. Concentration of most inorganic nutrients examined was affected by both rootstock and scion genotype. For NU scions, DF rootstocks resulted in significantly higher (5-29%) levels of nitrogen, phosphorus, potassium, calcium, magnesium, sulfur, copper, and zinc in leaf tissue than ug-NU. For DF scions, NU rootstocks resulted in significantly higher (7-9%) levels of potassium and phosphorus but significantly lower (11-23%) levels of magnesium, sulfur, boron, copper, and zinc when compared to ug-DF. Results of this study show that intercultivar grafting sweet basil using a more vigorous cultivar as a rootstock did not enhance the growth of a less vigorous scion and reduced concentrations of certain nutrients. However, grafting a vigorous scion to a less vigorous rootstock reduced shoot growth but generally increased nutrient concentrations. This suggests that changes in growth in grafted basil are not due to altered nutrient status. Ocimum basilicum, commonly known as sweet basil, was found naturally infected showing mosaic mottling and witches' broom symptoms. Semi-nested PCR using specific primers revealed the presence of a mixed infection of TMV and Candidatus phytoplasma. This study aims to evaluate and compare the phytochemical composition of essential oils from aerial parts produced in sweet basil. The comparison covered uninfected, mixed infected, TMV infected, and phytoplasma infected sweet basil. Thirty different essential oils were isolated and evaluated using gas chromatography-mass spectrometry (GC-MS) analysis. The essential oils produced were grouped into 5 main classes: monoterpenes hydrocarbons, oxygenated monoterpenes, sesquiterpenes hydrocarbons, oxygenated sesquiterpenes and ketones. All classes were severely altered due to TMV and Candidatus phytoplasma infection compared to the control. The pathological stress caused by the mixed infection was significantly higher than each pathogen separately. The monoterpenes hydrocarbons, oxygenated monoterpenes and ketones showed a significant increase in the essential oil percentages induced in the mixed infection than that of single infections. Yet, a significant decrease in sesquiterpenes hydrocarbons and oxygenated sesquiterpenes was reported in the infected plants. Furthermore, α-bergamotene, (+)spathulenol, α bergamotene, δ-guaiene, cubenol, bornyl acetate and 1,3-bis(cinnamoyloxymethyl)adamantane, were found only in healthy plants, while α-pinene, camphene, sabinene, β-pinene, β-myrcene, trans-sabinene hydrate, eugenol and trans-caryophyllene were detected only in infected plants. An experiment was conducted to study the effect of integrated nutrient management practices on the microbial community dynamics of soils under sweet basil (Ocimum basilicum L.) at ICAR - Indian Institute of Horticultural Research, Bengaluru during the Kharif season of 2015 and 2016. There were nine treatments replicated thrice in randomized complete block design. The results indicated that integrated application of FYM (10 t/ha) + 100% recommended N through FYM + bio-fertilizer i.e., T2 recorded the highest population of heterotrophic free-living N2 fixers (40.66 and 63.33 CFU ×103/ g), phosphate solubilizing bacteria (5.6 and 6.6 CFU ×103/ g) and fungal (6.4 and 5.33 CFU ×103/ g) while T9 with the application of NPK (160:80:80 kg /ha) + FYM (10 t/ha) recorded the highest population of actinomycetes (29.93 and 44.56 CFU ×103/ g) in the soil during 2015 and 2016, respectively. Application of recommended dose of FYM (10 t/ha) in T7 resulted in a reduction in the population of heterotrophic free-living N2 fixers (26.13 and 34 CFU ×103/ g) and actinomycetes (20 and 30.5 CFU ×103/ g) whereas, the application of a recommended dose of chemical fertilizer in T8 recorded the lowest population of phosphate solubilizing bacteria (3.9 CFU ×103/ g) and fungal (3.6 and 2.5 CFU ×103/ g) during 2015 and 2016, respectively. The highest organic carbon (0.63 and 0.66 %) content in the post-harvest soil samples was recorded with the application of NPK (160:80:80 kg /ha) + FYM (10 t/ha) while, the lowest organic carbon value (0.52 and 0.53%) was recorded in T8 during 2015 and 2016, respectively. Application of recommended FYM (10 t/ha) along with recommended NPK (160:80:80 kg/ha) in T9 recorded maximum herbage yield in the main crop (41.59 and 38.31 t/ha) and ratoon (20.97 and 17.77 t/ha) during 2015 and 2016, respectively. The results obtained from this study clearly demonstrated that integrated nutrient management can maximize soil microbial community dynamics which is considered as the driving force behind regulating soil processes that support sustainable sweet basil cultivation.

Plant stress diagnosis is essential for efficient crop management and productivity increase. Under stress, plants undergo physiological and compositional changes. Vegetation indices obtained from leaf reflectance spectra and bioimpedance spectroscopy provide information about the external and internal aspects of plant responses, respectively. In this study, bioimpedance and vegetation indices were noninvasively acquired from sweet basil (Ocimum basilicum L.) leaves exposed to three types of stress (drought, salinity, and chilling). Integrating the vegetation index, a novel approach, contains information about the surface of plants and bioimpedance data, which indicates the internal changes of plants. The fusion of these two datasets was examined to classify the types and severity of stress. Among the eight supervised machine learning models (three linear and five non-linear), the support vector machine (SVM) exhibited the highest accuracy in classifying stress types. Bioimpedance spectroscopy alone exhibited an accuracy of 0.86 and improved to 0.90 when fused with vegetation indices. Additionally, for drought and salinity stresses, it was possible to classify the early stage of stress with accuracies of 0.95 and 0.93, respectively. This study will allow us to classify the different types and severity of plant stress, prescribe appropriate treatment methods for efficient cost and time management of crop production, and potentially apply them to low-cost field measurement systems.
