Coconut (Cocos nucifera L.) is an important agricultural commodity with substantial economic and nutritional value, widely used for various products, including coconut water. The sweetness is an important quality trait of coconut water, which is influenced by genetic and environmental factors. In this study, we utilized next-generation sequencing to identify genetic variations in the coconut genome associated with the sweetness of coconut water. Whole-genome resequencing of 49 coconut accessions, including diverse germplasm and an F2 population of 81 individuals, revealed ~27 M SNPs and ~1.5 M InDels. Sugar content measured by °Bx was highly variable across all accessions tested, with dwarf varieties generally sweeter. A comprehensive analysis of the sugar profiles revealed that sucrose was the major sugar contributing to sweetness. Allele mining of the 148 genes involved in sugar metabolism and transport and genotype–phenotype association tests revealed two significant SNPs in the hexose carrier protein (Cnu01G018720) and sucrose synthase (Cnu09G011120) genes associated with the higher sugar content in both the germplasm and F2 populations. This research provides valuable insights into the genetic basis of coconut sweetness and offers molecular markers for breeding programs aimed at improving coconut water quality. The sink strength of developing ovaries in wheat determines the grain weight potential. The period from booting to the grain setting stage is critical for ovary growth and development and potential sink capacity determination. However, the underlying regulatory mechanism during this period by which the wheat plant balances and coordinates the floret number and ovary/grain weight under water stress has not been clarified. Therefore, we designed two irrigation treatments of W0 (no seasonal irrigation) and W1 (additional 75 mm of irrigation at the jointing stage) and analyzed the responses of the ovary/grain weight to water stress at the phenotypic, metabolomic, and transcriptomic levels. The results showed that the W0 irrigation treatment reduced the soil water content, plant height, and green area of the flag leaf, thus reducing grain number, especially for the inferior grains. However, it improved the grain weight of the superior and inferior grains as well as average grain weight at maturity, while the average ovary/grain weight and volume during –3 to 10 days after anthesis (DAA) also increased. Transcriptomic analysis indicated that the genes involved in both sucrose metabolism and phytohormone signal transduction were prominently accelerated by the W0 treatment, accompanied by greater enzymatic activities of soluble acid invertase (SAI) and sucrose synthase (Sus) and elevated abscisic acid (ABA) and indole-3-acetic acid (IAA) levels. Thus, the sucrose content decreased, while the glucose and fructose contents increased. In addition, several TaTPP genes (especially TaTPP-6) were down-regulated and the IAA biosynthesis genes TaTAR1 and TaTAR2 were up-regulated under the W0 treatment before anthesis, which further increased the IAA level. Collectively, water stress reduced the growth of vegetative organs and eliminated most of the inferior grains, but increased the ABA and IAA levels of the surviving ovaries/grains, promoting the enzymatic activity of Sus and degrading sucrose into glucose and fructose. As a result, the strong sucrose utilization ability, the enhanced enzymatic activity of SAI and the ABA- and IAA-mediated signaling jointly increased the weight and volume of the surviving ovaries/grains, and ultimately achieved the trade-off between ovary/grain weight and number in wheat under water stress.

Summary: This study examined the impact of CO2 on the quality of onions stored at 23 °C for 6 weeks, focussing on bioactive compounds, antioxidant capacity, individual anthocyanins, flavonoids and anthocyanin biosynthesis during the storage period. The findings revealed that a 20% CO2 treatment was markedly more effective than the control in maintaining higher levels of phenolics, flavonoids, flavanols and anthocyanins. Moreover, the 20% CO2 treatment preserved significantly higher quantities of individual anthocyanins and flavonoids compared to the control. Throughout storage, the 20% CO2 treatment significantly boosted the activity of enzymes related to anthocyanin biosynthesis and sucrose metabolism relative to the control groups. In summary, the innovation of this study lies in the use of CO2 to maximise anthocyanin and flavonoid content in stored onions, which are usually diminished by extended storage at room temperature. Abstract The SnRK (sucrose non-fermentation-related protein kinase) plays an important role in regulating various signals in plants. However, as an important bamboo shoot and wood species, the response mechanism of PheSnRK in Phyllostachys edulis to hormones, low energy and stress remains unclear. In this paper, we focused on the structure, expression, and response of SnRK to hormones and sugars. In this study, we identified 75 PheSnRK genes from the Moso bamboo genome, which can be divided into three groups according to the evolutionary relationship. Cis-element analysis has shown that the PheSnRK gene can respond to various hormones, light, and stress. The PheSnRK2.9 proteins were localized in the nucleus and cytoplasm. Transgenic experiments showed that overexpression of PheSnRK2.9 inhibited root development, the plants were salt-tolerant and exhibited slowed starch consumption in Arabidopsis in the dark. The results of yeast one-hybrid and dual luciferase assay showed that PheIAAs and PheNACs can regulate PheSnRK2.9 gene expression by binding to the promoter of PheSnRK2.9. This study provided a comprehensive understanding of PheSnRK genes of Moso bamboo, which provides valuable information for further research on energy regulation mechanism and stress response during the growth and development of Moso bamboo.

Main conclusion: Upon systemic S. indica colonization in split-root system cyst and root-knot nematodes benefit from endophyte-triggered carbon allocation and altered defense responses what significantly facilitates their development in A. thaliana. Serendipita indica is an endophytic fungus that establishes mutualistic relationships with different plants including Arabidopsis thaliana. It enhances host’s growth and resistance to different abiotic and biotic stresses such as infestation by the cyst nematode Heterodera schachtii (CN). In this work, we show that S. indica also triggers similar direct reduction in development of the root-knot nematode Meloidogyne javanica (RKN) in A. thaliana. Further, to mimick the natural situation occurring frequently in soil where roots are unequally colonized by endophytes we used an in vitro split-root system with one half of A. thaliana root inoculated with S. indica and the other half infected with CN or RKN, respectively. Interestingly, in contrast to direct effects, systemic effects led to an increase in number of both nematodes. To elucidate this phenomenon, we focused on sugar metabolism and defense responses in systemic non-colonized roots of plants colonized by S. indica. We analyzed the expression of several SUSs and INVs as well as defense-related genes and measured sugar pools. The results show a significant downregulation of PDF1.2 as well as slightly increased sucrose levels in the non-colonized half of the root in three-chamber dish. Thus, we speculate that, in contrast to direct effects, both nematode species benefit from endophyte-triggered carbon allocation and altered defense responses in the systemic part of the root, which promotes their development. With this work, we highlight the complexity of this multilayered tripartite relationship and deliver new insights into sugar metabolism and plant defense responses during S. indica–nematode–plant interaction. To accurately evaluate the role of storage temperature in improving the quality of fresh-cut fruits and vegetables, the effects of two storage temperatures (5 °C and 15 °C) on the phenylpropanoid pathway and sucrose metabolism in fresh-cut melon (cv. Yugu) cubes were determined. A higher temperature (15 °C) expedited sucrose decomposition in the melon cubes at the early stage of storage, resulting in higher levels of glucose and fructose. This effect was corroborated by increased activities of acid invertase (AI), neutral invertase (NI), and sucrose synthase cleavage (SS-c), along with higher expressions of CmAI1/2, CmNI1/2, and CmSS1/2 in the melon cubes at 15 °C. Additionally, the higher activity and gene expression of hexokinase in melon cubes at 15 °C led to an increase in the utilization rate of sugars toward downstream metabolic pathways. Moreover, the melon cube storage at 15 °C elevated the activities and gene expressions of phenylalanine ammonia lyase (PAL), cinnamic acid 4-hydroxylase (C4H), and 4-coumaric acid: CoA ligase (4CL), thereby increasing the synthesis of phenolics. Sucrose showed a significant negative correlation with PAL, C4H, and 4CL, as well as with CmPAL5/7 and CmC4H1/3. However, hexokinase displayed a significant positive correlation with PAL, C4H, and 4CL, as well as with CmPAL1, CmPAL3-9, CmC4Hs, and Cm4CLs. These findings demonstrate that a higher-temperature storage of melon cubes can accelerate the phenylpropanoid pathway and sucrose metabolism by regulating the activity and gene expression of related enzymes, thereby inducing phenolic accumulation. These results also indicate that lower-temperature storage is not conducive to the conversion of sugars into phenolics in fresh-cut melon. Advances in carbohydrate metabolism prompted its essential role in defense priming and sweet immunity during plant-pathogen interactions. Nevertheless, upstream responding enzymes in the sucrose metabolic pathway and associated carbohydrate derivatives underlying fungal pathogen challenges remain to be deciphered in Populus, a model tree species. In silico deduction of genomic features, including phylogenies, exon/intron distributions, cis-regulatory elements, and chromosomal localization, identified 59 enzyme genes (11 families) in the Populus genome. Spatiotemporal expression of the transcriptome and the quantitative real-time PCR revealed a minuscule number of isogenes that were predominantly expressed in roots. Upon the pathogenic Fusarium solani (Fs) exposure, dynamic changes in the transcriptomics atlas and experimental evaluation verified Susy (PtSusy2 and 3), CWI (PtCWI3), VI (PtVI2), HK (PtHK6), FK (PtFK6), and UGPase (PtUGP2) families, displaying promotions in their expressions at 48 and 72 h of post-inoculation (hpi). Using the gas chromatography-mass spectrometry (GC–MS)-based non-targeted metabolomics combined with a high-performance ion chromatography system (HPICS), approximately 307 metabolites (13 categories) were annotated that led to the quantification of 46 carbohydrates, showing marked changes between three compared groups. By contrast, some sugars (e.g., sorbitol, L-arabitol, trehalose, and galacturonic acid) exhibited a higher accumulation at 72 hpi than 0 hpi, while levels of α-lactose and glucose decreased, facilitating them as potential signaling molecules. The systematic overview of multi-omics approaches to dissect the effects of Fs infection provides theoretical cues for understanding defense immunity depending on fine-tuned Suc metabolic gene clusters and synergistically linked carbohydrate pools in trees. Northern China faces water scarcity, restricting water usage in place across Inner Mongolia's western region. The integrated irrigation and fertilization model for sugar beet is undergoing rapid development and application in production. However, there is a concerning trend in production where the frequency of irrigation and fertilization is being increased blindly, resulting in the wastage of valuable water and fertilizer resources. Limiting water and fertilizer usage is an effective approach to improve sugar beet production efficiency. Sugar beets are a significant sugar crop in China. A split-plot design was employed to examine the impact of reducing water and fertilizer use on sucrose metabolism in sugar beet root. Our study was performed at the Ulanqab Institute of Agricultural and Forestry Sciences in Inner Mongolia from 2022 to 2023. Three levels of fertilization and irrigation were utilized. We investigated the interactions between irrigation and fertilization on sucrose accumulation in sugar beet root. We examined key enzyme activities involved in sucrose metabolism alongside their gene expression levels. The findings suggested that reducing irrigation by 15%, fertilization by 10%, or both irrigation by 15% and fertilization by 10%, increased sucrose concentrations of sugar beets compared to the control group administered conventional water and fertilizer. Over the two-year period, the average sucrose concentration increased by 0.45, 0.57, and 0.65 degrees, respectively, under each treatment. Subsequent research verified that appropriately reducing water and fertilizer can regulate the expression of enzyme genes, thus influencing enzyme activity. Moreover, due to the higher efficiency of enzyme synthesis compared to decomposition, it contributed to an increase in net enzyme activity. These findings suggest that an appropriate reduction of water and fertilizer can improve sucrose synthesis rates and increase the sucrose concentration in sugar beets, providing a theoretical basis for environmentally friendly generation and enhanced efficiency in sugar beet growth. The oil sunflower is an important oil crop and ornamental plant. Flowering time affects the environmental adaptability and final yield of oil sunflowers. Floral induction is one of the important events that determines subsequent reproductive growth and seed setting, but there has been no systematic study on the regulation of gene expression during the transition from vegetative growth to reproductive growth in oil sunflowers. In this study, an oil sunflower mutant displaying early flowering (ef) was obtained by heavy ion beam irradiation. This mutant had a stable genetic trait, and its flowering time was 15 days earlier than the wild type (WT) in the field. The histology result showed that the ef mutant induced floral meristem at 6-leaf stage earlier than WT. The shoot apical meristems (SAMs) of the ef mutant and WT at 4-leaf, 6-leaf, 8-leaf, 10-leaf and budding periods were collected for RNA sequencing. The results showed that the transition from the leaf meristem to the floral meristem resulted in significant changes in the transcriptional landscape. Overall, 632, 1825, 4549, 5407 and 2164 differentially expressed genes (DEGs) were identified at 4-leaf, 6-leaf, 8-leaf, 10-leaf and budding periods, respectively. These DEGs were mainly enriched in biological pathways, including plant hormone signal transduction, carbon metabolism, protein processing in endoplasmic reticulum, secondary metabolism, and photosynthesis. We also found significant differences in the expression levels of starch and sucrose metabolism-related genes in the ef mutant and WT, indicating that sugar signaling plays an important role in the early flowering of oil sunflowers, especially SUC9 and sugar synthesis and degradation enzyme genes. In addition to hormone and sugar signals, flowering integration genes SOC1, AP1, FUL and LFY were upregulated in the ef mutant, and genes in photoperiod, aging, autonomous and temperature pathways were also involved in the regulation of floral transition. The results showed that plant hormones, sucrose metabolism, and flowering genes synergistically cause the early flowering of oil sunflowers. Summary: Sugar transporter proteins (STPs) play critical roles in regulating plant stress tolerance, growth, and development. However, the role of STPs in regulating crop yield is poorly understood.This study elucidates the mechanism by which knockout of the sugar transporter OsSTP15 enhances grain yield via increasing the tiller number in rice.We found that OsSTP15 is specifically expressed in the shoot base and vascular bundle sheath of seedlings and encodes a plasma membrane‐localized high‐affinity glucose efflux transporter. OsSTP15 knockout enhanced sucrose and trehalose‐6‐phosphate (Tre6P) synthesis in leaves and improved sucrose transport to the shoot base by inducing the expression of sucrose transporters. Higher glucose, sucrose, and Tre6P contents were observed at the shoot base of stp15 plants. Transcriptome and metabolome analyses of the shoot base demonstrated that OsSTP15 knockout upregulated the expression of cytokinin (CK) synthesis‐ and signaling pathway‐related genes and increased CK levels.These findings suggest that OsSTP15 knockout represses glucose export from the cytoplasm and simultaneously enhances sugar transport from source leaves to the shoot base by promoting the synthesis of sucrose and Tre6P in leaves. Main conclusion: ZmSUS1 improved drought tolerance of maize by regulating sucrose metabolism and increasing soluble sugar content, and endowing transgenic maize with higher relative water content and photosynthesis levels. Sucrose synthase (SUS), a key enzyme of sugar metabolism, plays an important role in the regulation of carbon partitioning in plant, and affects important agronomic traits and abiotic responses to adversity. However, the function of ZmSUS1 in plant drought tolerance is still unknown. In this study, the expression patterns of ZmSUS1 in different tissues and under drought stress were analyzed in maize (Zea mays L.). It was found that ZmSUS1 was highly expressed during kernel development but also in leaves and roots of maize, and ZmSUS1 was induced by drought stress. Homozygous transgenic maize lines overexpressing ZmSUS1 increased the content and activity of SUS under drought stress and exhibited higher relative water content, proline and abscisic acid content in leaves. Specifically, the net photosynthetic rate and the soluble sugar contents including sucrose, glucose, fructose and SUS decomposition products including UDP-glucose (UDP-G) and ADP-glucose (ADP-G) in transgenic plants were significantly improved after drought stress. RNA-seq analysis showed that overexpressing of ZmSUS1 mainly affected the expression level of carbon metabolism-related genes. Especially the expression level of sucrose metabolism-related genes including sucrose phosphatase gene (SPP), sucrose phosphate synthase gene (SPS) and invertase gene (INV) were significantly up-regulated in transgenic maize. Overall, these results suggested that ZmSUS1 improved drought tolerance by regulating sucrose metabolism and increasing the soluble sugar content, and endowing transgenic maize with higher relative water content and photosynthesis levels, which can serve as a new gene candidate for cultivating drought-resistant maize varieties. Removing the panicle encourages the growth of buds on the elongated node by getting rid of apical dominance. Strigolactones (SLs) are plant hormones that suppress tillering in rice. The present study employed panicle removal (RP) and external application of synthesized strigolactones (GR) to modulate rice bud growth at node 2. We focused on the full-heading stage to investigate proteomic changes related to bud germination (RP-Co) and suppression (GR-RP). A total of 434 represented differentially abundant proteins (DAPs) were detected, with 272 DAPs explicitly specified in the bud germination process, 106 in the bud suppression process, and 28 in both. DAPs in the germination process were most associated with protein processing in the endoplasmic reticulum and ribosome biogenesis. DAPs were most associated with metabolic pathways and glycolysis/gluconeogenesis in the bud suppression process. Sucrose content and two enzymes of sucrose degradation in buds were also determined. Comparisons of DAPs between the two reversed processes revealed that sucrose metabolism might be a key to modulating rice bud growth. Moreover, sucrose or its metabolites should be a signal downstream of the SLs signal transduction that modulates rice bud outgrowth. Contemplating the result so far, it is possible to open new vistas of research to reveal the interaction between SLs and sucrose signaling in the control of tillering in rice. [ABSTRACT FROM AUTHOR]

Sucrose synthase (SUS) and sucrose phosphate synthase (SPS) are essential in plant sucrose metabolism. The potato is an important crop worldwide, but systematic analyses of the StSUS and StSPS gene families in potatoes are still lacking. Ten sucrose metabolism-related genes were identified in this study. The SUSs and SPSs could each be split into three subgroups through phylogenetic analysis. StSUSIc was the most highly expressed gene in different developmental tissues. Ka/Ks analysis showed that StSUSIb and StSUSIc were subjected to more-significant homozygous selection pressure. Our cis-acting element analysis of the StSUS and StSPS promoter sequences showed four elements: defense- and stress-responsive, hormone-responsive, light-responsive, and transcription factor elements. The expression of StSUS and StSPS genes was found to be regulated by circadian rhythm. In the treatments of 1% to 5% sucrose, glucose, and fructose, the expression of StSUS and StSPS family genes was enhanced by sucrose, but inhibited at high-glucose and fructose concentrations. This study identified six StSUS and four StSPS genes and analyzed their gene structure, conserved motifs, chromosome position, promoter elements, phylogenetic tree, and tissue-specific expression patterns. The effect of drought stress on carbon metabolism in the leaves and roots of bermudagrass was investigated. Plants established in PVC tubes suffered from three water treatments for 10 days. C138 and Tifway (drought-tolerant) were found to have lower relative electrical conductivity and higher water use efficiency than C32 (drought-sensitive) under moderate drought by increasing carotenoid and soluble sugar content and rapidly decreasing leaf starch content. The sucrose synthase activity of leaves and roots, acid invertase and neutral invertase activity of C32 roots substantially decreased under severe drought, resulting in a slow sucrose decomposition rate and significantly lower fructose and glucose contents than C138 and Tifway. The activities of four carbon metabolism enzymes and sucrose content in the leaves were greater than those in the roots, while the fructose and glucose contents were on the contrary, indicating that bermudagrass transported fructose and glucose obtained from sucrose decomposition from leaves to roots under drought to reduce roots damage. The path analysis indicated that leaves neutral invertase activity, and roots soluble sugar content might be the key parameter of carbon metabolism in bermudagrass under drought.

Saline and alkali stress affects the growth and development, survival rate, and final yield of rice, while new nano materials can have a positive effect on rice growth. In order to investing the effects of carboxymethyl multi walled carbon nanotubes (MWCNTs) on the growth and development of rice seedlings under salt alkali stress, rice seedlings were cultured using rice variety “Songjing 3” using nutrient solution water culture method. The effects of MWCNTs on water absorption capacity, leaf photosynthesis, and sucrose metabolism of rice seedlings under 50 mmol/L saline-alkali stress (1NaCl: 9Na2SO4: 9NaHCO3: 1Na2CO3) conditions were investigated. The results showed that MWCNTs can improve the water use ability of roots and leaves, especially the water absorption ability of roots, which provides a guarantee for the improvement of rice biomass and the enhancement of leaf photosynthetic capacity under adverse conditions. After treatment with MWCNTs, the photosynthetic rate (Pn), stomatal conductance (gs), and transpiration rate (Tr) of leaves increased significantly, and the photochemical quenching value (qP), photochemical quantum efficiency value (Fv/Fm), and electron transfer rate value (ETR) of chlorophyll fluorescence parameters increased significantly, which is beneficial to the improvement of the PSII photosynthetic system. MWCNTs treatment promoted the increase of photosynthetic pigment content in leaves under salt and alkali stress, improved the ratio of Chla and Chlb parameters, increased the activities of key photosynthetic enzymes (RUBPCase and PEPCase) in leaves, increased the value of total lutein cycle pool (VAZ), and significantly enhanced the deepoxidation effect of lutein cycle (DEPS), which can effectively alleviate the stomatal and non stomatal constraints on leaf photosynthesis caused by salt and alkali stress. MWCNTs treatment significantly enhanced the activities of sucrose phosphate synthase (SPS) and sucrose synthase (SS) under salt and alkali stress, and decreased the activities of soluble acid invertase (SAInv) and alkaline/neutral invertase (A/N-Inv), indicating that MWCNTs promoted sucrose synthesis while inhibiting sucrose decomposition, thereby promoting sucrose accumulation in rice leaves. This study can provide theoretical and experimental basis for the application of MWCNTs to the production of rice under salt and alkali stress, and can find a new way for rice production in saline and alkaline lands.

In a recent Review Article on Gregor Mendel’s (1822–1884) work with pea (Pisum sativum)-plants, it was proposed that this crop species should be re-vitalized as a model organism for the study of cell- and organ growth. Here, we describe the effect of exogenous gibberellic acid (GA3) on the growth of the second internode in 4-day-old light-grown pea seedlings (Pisum sativum, large var. “Senator”). lnjection of glucose into the internode caused a growth-promoting effect similar to that of the hormone GA3. Imbibition of dry pea seeds in GA3, or water as control, resulted in a drastic enhancement in organ development in this tall variety. Similar results were reported for dwarf peas. These “classical” experimental protocols are suitable to study the elusive effect of gibberellins (which act in coordination with auxin) on the regulation of plant development at the biochemical and molecular levels.

To accurately evaluate the role of storage temperature in improving the quality of fresh-cut fruits and vegetables, the effects of two storage temperatures (5 °C and 15 °C) on the phenylpropanoid pathway and sucrose metabolism in fresh-cut melon (cv. Yugu) cubes were determined. A higher temperature (15 °C) expedited sucrose decomposition in the melon cubes at the early stage of storage, resulting in higher levels of glucose and fructose. This effect was corroborated by increased activities of acid invertase (AI), neutral invertase (NI), and sucrose synthase cleavage (SS-c), along with higher expressions of CmAI1/2, CmNI1/2, and CmSS1/2 in the melon cubes at 15 °C. Additionally, the higher activity and gene expression of hexokinase in melon cubes at 15 °C led to an increase in the utilization rate of sugars toward downstream metabolic pathways. Moreover, the melon cube storage at 15 °C elevated the activities and gene expressions of phenylalanine ammonia lyase (PAL), cinnamic acid 4-hydroxylase (C4H), and 4-coumaric acid: CoA ligase (4CL), thereby increasing the synthesis of phenolics. Sucrose showed a significant negative correlation with PAL, C4H, and 4CL, as well as with CmPAL5/7 and CmC4H1/3. However, hexokinase displayed a significant positive correlation with PAL, C4H, and 4CL, as well as with CmPAL1, CmPAL3-9, CmC4Hs, and Cm4CLs. These findings demonstrate that a higher-temperature storage of melon cubes can accelerate the phenylpropanoid pathway and sucrose metabolism by regulating the activity and gene expression of related enzymes, thereby inducing phenolic accumulation. These results also indicate that lower-temperature storage is not conducive to the conversion of sugars into phenolics in fresh-cut melon. Therefore, the temperature can be appropriately and briefly raised in the production and preservation process of fresh-cut melon to obtain higher levels of phenolics.

The oil sunflower is an important oil crop and ornamental plant. Flowering time affects the environmental adaptability and final yield of oil sunflowers. Floral induction is one of the important events that determines subsequent reproductive growth and seed setting, but there has been no systematic study on the regulation of gene expression during the transition from vegetative growth to reproductive growth in oil sunflowers. In this study, an oil sunflower mutant displaying early flowering (ef) was obtained by heavy ion beam irradiation. This mutant had a stable genetic trait, and its flowering time was 15 days earlier than the wild type (WT) in the field. The histology result showed that the ef mutant induced floral meristem at 6-leaf stage earlier than WT. The shoot apical meristems (SAMs) of the ef mutant and WT at 4-leaf, 6-leaf, 8-leaf, 10-leaf and budding periods were collected for RNA sequencing. The results showed that the transition from the leaf meristem to the floral meristem resulted in significant changes in the transcriptional landscape. Overall, 632, 1825, 4549, 5407 and 2164 differentially expressed genes (DEGs) were identified at 4-leaf, 6-leaf, 8-leaf, 10-leaf and budding periods, respectively. These DEGs were mainly enriched in biological pathways, including plant hormone signal transduction, carbon metabolism, protein processing in endoplasmic reticulum, secondary metabolism, and photosynthesis. We also found significant differences in the expression levels of starch and sucrose metabolism-related genes in the ef mutant and WT, indicating that sugar signaling plays an important role in the early flowering of oil sunflowers, especially SUC9 and sugar synthesis and degradation enzyme genes. In addition to hormone and sugar signals, flowering integration genes SOC1, AP1, FUL and LFY were upregulated in the ef mutant, and genes in photoperiod, aging, autonomous and temperature pathways were also involved in the regulation of floral transition. The results showed that plant hormones, sucrose metabolism, and flowering genes synergistically cause the early flowering of oil sunflowers. Our study provided important information for understanding flowering and is helpful for the genetic improvement of sunflowers.

Northern China faces water scarcity, restricting water usage in place across Inner Mongolia’s western region. The integrated irrigation and fertilization model for sugar beet is undergoing rapid development and application in production. However, there is a concerning trend in production where the frequency of irrigation and fertilization is being increased blindly, resulting in the wastage of valuable water and fertilizer resources. Limiting water and fertilizer usage is an effective approach to improve sugar beet production efficiency. Sugar beets are a significant sugar crop in China. A split-plot design was employed to examine the impact of reducing water and fertilizer use on sucrose metabolism in sugar beet root. Our study was performed at the Ulanqab Institute of Agricultural and Forestry Sciences in Inner Mongolia from 2022 to 2023. Three levels of fertilization and irrigation were utilized. We investigated the interactions between irrigation and fertilization on sucrose accumulation in sugar beet root. We examined key enzyme activities involved in sucrose metabolism alongside their gene expression levels. The findings suggested that reducing irrigation by 15%, fertilization by 10%, or both irrigation by 15% and fertilization by 10%, increased sucrose concentrations of sugar beets compared to the control group administered conventional water and fertilizer. Over the two-year period, the average sucrose concentration increased by 0.45, 0.57, and 0.65 degrees, respectively, under each treatment. Subsequent research verified that appropriately reducing water and fertilizer can regulate the expression of enzyme genes, thus influencing enzyme activity. Moreover, due to the higher efficiency of enzyme synthesis compared to decomposition, it contributed to an increase in net enzyme activity. These findings suggest that an appropriate reduction of water and fertilizer can improve sucrose synthesis rates and increase the sucrose concentration in sugar beets, providing a theoretical basis for environmentally friendly generation and enhanced efficiency in sugar beet growth.

Background: l‐Glutamate is involved in many important chemical reactions in horticultural products and improves postharvest disease resistance. Quality decline of apple fruit caused by senescence and fungus invasion often leads to tremendous losses during logistics. This study was performed to evaluate the variations of quality attributes, carotenoid, sorbitol and sucrose metabolisms in apples (cv. Qiujin) after l‐glutamate dipping treatment. RESUITS: l‐Glutamate immersion maintained high values of L*, a* and b*, flesh firmness, titratable acidity, as well as the total soluble solids, soluble sugar, reducing sugar and ascorbic acid contents in apples. l‐Glutamate also decreased mass loss, respiratory rate and ethylene release, enhanced sucrose synthase‐cleavage, acid invertase and neutral invertase activities, whereas reduced sorbitol dehydrogenase, sucrose phosphate synthase, sucrose synthase synthesis and sorbitol oxidase activities in apples. Moreover, l‐glutamate inhibited lutein, β‐carotene and lycopene accumulation, and down‐regulated phytoene synthase, lycopene β‐cyclase, ζ‐carotene desaturase, phytoene desaturase, carotenoid isomerase, ζ‐carotene isomerase and carotenoids cleavage dioxygenase gene expressions, but up‐regulated 9‐cis‐epoxycarotenoid dioxygenase gene expression in apples. Low temperature (LT) disrupted the osmotic regulation system and sucrose and nitrogen metabolism of Bitter gourd (Momordica charantia L.). The osmotic regulation system and the sucrose and nitrogen metabolism in bitter gourd were investigated in ungrafted (M), self-grafted (MS), and grafted onto rootstocks of pumpkin (Cucurbita moschata Duch., MC) and loofah (Luffa cylindrica Roem., ML) exposed to LT (8 °C) for 24 h and then allowed to recover growth (RG; 24/18 °C) for 24 h. MC and ML maintained better osmotic adjustment ability. Gas exchange parameters of MC and ML are higher than M and MS under both LT and RG. However, chloroplast fluorescence parameters varied considerably for the different groups under LT and RG. The soluble sugar and sucrose contents and sucrose phosphatase and sucrose synthase synthesis activities of MC leaves under LT and RG were higher than those of M and MS, however, the changes of these indexes in ML are not as obvious as that of MC. The activities of the sucrose-decomposing enzymes neutral invertase and acid invertase in ML and MC were lower than those of M and MS. After the induction of LT and RG, the nitrogen metabolites and enzyme activities of M and MS leaves were significantly reduced, while the MC and ML seedlings were on the contrary and the ammonium content in the leaves was significantly reduced. Low-temperature stress limits the growth and development of foxtail millet. Freezing stress caused by sudden temperature drops, such as late-spring coldness, often occurs in the seedling stage of foxtail millet. However, the ability and coping strategies of foxtail millet to cope with such stress are not clear. In the present study, we analyzed the self-regulatory mechanisms of freezing stress in foxtail millet. We conducted a physiological study on foxtail millet leaves at −4 °C for seven different durations (0, 2, 4, 6, 8, 10, and 12 h). Longer freezing time increased cell-membrane damage, relative conductance, and malondialdehyde content. This led to osmotic stress in the leaves, which triggered an increase in free proline, soluble sugar, and soluble protein contents. The increases in these substances helped to reduce the damage caused by stress. The activities of superoxide dismutase, peroxidase, and catalase increased reactive oxygen species (ROS) content. The optimal time point for the response to freezing stress was 8 h after exposure. The transcriptome analysis of samples held for 8 h at −4 °C revealed 6862 differentially expressed genes (DEGs), among which the majority are implicated in various pathways, including the starch and sucrose metabolic pathways, antioxidant enzyme pathways, brassinolide (BR) signaling pathway, and transcription factors, according to Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment. We investigated possible crosstalk between BR signals and other pathways and found that BR signaling molecules were induced in response to freezing stress. The beta-amylase (BAM) starch hydrolase signal was enhanced by the BR signal, resulting in the accelerated degradation of starch and the formation of sugars, which served as emerging ROS scavengers and osmoregulators to resist freezing stress. In conclusion, crosstalk between BR signal transduction, and both starch and sucrose metabolism under freezing stress provides a new perspective for improving freezing resistance in foxtail millet. Abstract Background Cassava (Manihot esculenta Crantz) is an important multiuse crop grown for economic and energy purposes. Its vegetative organs are storage roots, in which the main storage material is starch. The accumulation characteristics of starch in cassava roots can directly affect the yield, starch content and maturation of cassava storage roots. In this study, we used a cassava sexual tetraploid (ST), which showed early maturation heterosis in previous work, as the main test material. We analyzed the sucrose metabolism and starch accumulation characteristics of the ST and its parents from the leaf “source” to the storage root “sink” during different developmental stages and explored the regulatory mechanisms of ST storage root early maturation by combining the transcriptome data of the storage roots during the expansion period. Results The results showed that the trends in sucrose, glucose and fructose contents in the ST leaves were similar to those of the two parents during different stages of development, but the trends in the ST storage roots were significantly different from those of their parents, which showed high sucrose utilization rates during the early stage of development and decreased utilization capacity in the late developmental stage. Transcriptome data showed that the genes that were expressed differentially between ST and its parents were mainly involved in the degradation and utilization of sucrose in the storage roots, and four key enzyme genes were significantly upregulated (Invertase MeNINV8/MeVINV3, Sucrose synthase MeSuSy2, Hexokinase MeHXK2), while the expressions of key enzyme genes involved in starch synthesis were not significantly different. Conclusions The results revealed that the pattern of sucrose degradation and utilization in the cassava ST was different from that of its parents and promoted early maturation in its tuberous roots. Starch accumulation in the ST from sucrose mainly occurred during the early expansion stage of the storage roots, and the starch content during this period was higher than that of both parents, mainly due to the regulation of invertase and hexokinase activities during sucrose metabolism. This study provides a basis for further genetic improvements to cassava traits and for breeding varieties that mature early and are adapted well to provide starch supply requirements.

AbstractSalt stress is a major abiotic stress with detrimental effects on plants. Sucrose, the main product of photosynthesis in plants, is used as a source of energy for the development of Chinese rose (Rosa chinensis). Hua Xianzi, a variety of Chinese rose, was investigated in this study. Phenotypic traits such as digital biomass, greenness average, leaf angle and leaf area, as well as physiological characters such as proline (Pro), malondialdehyde (MDA), H2O2, O2- and antioxidant enzymes such as SOD (Superoxide Dismutase), POD (Peroxidase), CAT (Catalase) and APX (Ascorbate Peroxidase), changed significantly in response to salt stress (150 mmol/L NaCl for 72 h). The physiological changes were examined based on photosynthetic and fluorescence indicators. Salt stress increased the concentration of glucose, fructose and sucrose, upregulated the expression of Rc-SS1, Rc-SS2, Rc-SPS1, Rc-SPS2, Rc-αA1, Rc-αA2, Rc-αA3, Rc-βA1 and Rc-βA3 genes. Finally, it decreased the starch content in leaves. This study revealed that salt stress disrupted the equilibrium of sucrose metabolism in leaves. This finding provides the basis for further research into the mechanism of salt tolerance adaption.

Starch is an important reserve of sugar, and starch-sugar conversion in plants plays an important role in the response of plants to various abiotic stresses. Nicosulfuron is a post-emergence herbicide commonly applied to maize fields. However, it is unclear how sucrose and starch in sweet corn are converted to accommodate nicosulfuron stress. Field and pot experiments were conducted to study the effects of nicosulfuron on the sugar metabolism enzymes, starch metabolism enzymes, non-enzyme substances, and expression of key enzyme genes in leaves and roots of sweet maize seedlings. Accordingly, this research compared the responses of the sister lines HK301 and HK320, which are nicosulfuron tolerant and sensitive, respectively. Under nicosulfuron stress, compared with HK301 seedlings, the accumulation of stem and root dry matter of HK320 seedlings was significantly reduced, resulting in a lower root-to-shoot ratio. Compared with HK320 seedlings, nicosulfuron stress significantly increased the sucrose, soluble sugar, and starch contents in HK301 leaves and roots. This may be related to the enhanced carbohydrate metabolism under nicosulfuron stress, including significant changes in sugar metabolism enzyme activity and the levels of SPS and SuSys expression. Further, under nicosulfuron stress, sucrose transporter genes (SUC 1, SUC 2, SWEET 13a, and SWEET 13b) in the leaves and roots of HK301 seedlings were significantly upregulated. Sucrose synthase (SUS) and sucrose phosphate synthase (SPS) are essential in plant sucrose metabolism. The potato is an important crop worldwide, but systematic analyses of the StSUS and StSPS gene families in potatoes are still lacking. Ten sucrose metabolism-related genes were identified in this study. The SUSs and SPSs could each be split into three subgroups through phylogenetic analysis. StSUSIc was the most highly expressed gene in different developmental tissues. Ka/Ks analysis showed that StSUSIb and StSUSIc were subjected to more-significant homozygous selection pressure. Our cis-acting element analysis of the StSUS and StSPS promoter sequences showed four elements: defense- and stress-responsive, hormone-responsive, light-responsive, and transcription factor elements. The expression of StSUS and StSPS genes was found to be regulated by circadian rhythm. In the treatments of 1% to 5% sucrose, glucose, and fructose, the expression of StSUS and StSPS family genes was enhanced by sucrose, but inhibited at high-glucose and fructose concentrations. This study identified six StSUS and four StSPS genes and analyzed their gene structure, conserved motifs, chromosome position, promoter elements, phylogenetic tree, and tissue-specific expression patterns. Our results will motivate more research into the biological process underlying the genes of sucrose metabolism in potatoes.

Removing the panicle encourages the growth of buds on the elongated node by getting rid of apical dominance. Strigolactones (SLs) are plant hormones that suppress tillering in rice. The present study employed panicle removal (RP) and external application of synthesized strigolactones (GR) to modulate rice bud growth at node 2. We focused on the full-heading stage to investigate proteomic changes related to bud germination (RP-Co) and suppression (GR-RP). A total of 434 represented differentially abundant proteins (DAPs) were detected, with 272 DAPs explicitly specified in the bud germination process, 106 in the bud suppression process, and 28 in both. DAPs in the germination process were most associated with protein processing in the endoplasmic reticulum and ribosome biogenesis. DAPs were most associated with metabolic pathways and glycolysis/gluconeogenesis in the bud suppression process. Sucrose content and two enzymes of sucrose degradation in buds were also determined. Comparisons of DAPs between the two reversed processes revealed that sucrose metabolism might be a key to modulating rice bud growth. Moreover, sucrose or its metabolites should be a signal downstream of the SLs signal transduction that modulates rice bud outgrowth. Contemplating the result so far, it is possible to open new vistas of research to reveal the interaction between SLs and sucrose signaling in the control of tillering in rice.

After harvest, the metabolism of Gynura bicolor DC (G. bicolor) is vigorous, resulting in sugar scarcity and reactive oxygen species (ROS) accumulation, thus aggravating the quality deterioration. 1-Methylcyclopropene (1-MCP) shows crucial effect in alleviating the postharvest metabolism of vegetables and fruits. This research aimed to evaluate the effect of 1-MCP on ROS scavenging and sucrose metabolism in G. bicolor. In this research, G. bicolor was treated with 10 μL L−1 1-MCP for 12 h, followed by storage at 20 ± 2 °C and 90 ± 5% relative humidity in darkness for 7 days. During storage, the increases in the respiration rate, electrolytic leakage, weight loss rate, ROS levels, and membrane lipid oxidation were effectively inhibited by 1-MCP. Moreover, starch and hexose degradation was decreased in the 1-MCP group, as were sucrose synthesis and catabolism. Correlation analysis indicated that sugar starvation was associated with respiration, activities regulation of CAT, SOD, and enzymes involved in sucrose metabolism were associated with the levels of hydrogen peroxide at the early storage. In conclusion, 1-MCP delayed postharvest quality deterioration of G. bicolor by alleviating respiration, inducing oxidative stress to enhance ROS scavenging, and inhibiting sucrose metabolism. • 1-MCP improved postharvest metabolism by inhibiting increase in respiration. • 1-MCP enhanced ROS scavenging and inhibited sucrose metabolism. • Sugar scarcity was alleviated by 1-MCP-induced inhibition of sucrose metabolism. • Increased H 2 O 2 concentration at an early stage may modulate sucrose metabolism. Carbon reserves in cotton roots can be remobilized to support reproductive growth, thus potentially affecting cotton yield. However, the regulation of carbon remobilization in cotton roots and its relationship with cotton yield are still poorly understood. Plant population density (PPD) and mepiquat chloride (MC) have been hypothesized to affect the dynamics of nonstructural carbohydrate content and the resulting carbon remobilization in cotton roots through the regulation of carbohydrate metabolism enzyme activities. A mid-maturation cotton line 4003–6 was field-grown in 2019 and 2020. Three different levels of PPD (D1: 2.25 plants m−2, D2: 4.5 plants m−2, and D3: 6.75 plants m−2) and two levels of MC dosage (M0: 0 g hm−2, M1: 82.5 g hm−2) were combined to create six populations differing in terms of the source–sink relationship. The changes in the hexose, sucrose, and starch contents and the key carbon metabolic enzyme activities in cotton roots were examined during peak squaring (PS) to late boll opening (LB). The combination of the PPD of 6.75 plants m−2 and MC application (M1D3) exhibited the greatest cotton yield and reproductive biomass–to–leaf area ratio from peak flowering (PF) onwards. M1D3 presented the greatest total nonstructural carbohydrate (TNC) remobilization amount of 2.96 and 3.80 g m−2, the highest efficiency of 39.11% and 48.39%, and the largest gross contribution to seed cotton yield of 0.66% and 0.79% in 2019 and 2020, respectively. The three parameters were positively correlated with the seed cotton yield except for the remobilization rate in 2019. Unlike the other treatments, the greater carbohydrate content per unit ground area in M1D3 prior to the PF stage was attributed to the higher sucrose phosphate synthase (SPS) and ADP-glucose pyrophosphorylase (AGPase) activities during the PS to first flowering (FF) stages. Conversely, the greater α-amylase and β-amylase activities in M1D3 at the PF stage accounted for the lower starch content at the EB stage, and the smaller vacuolar invertase (VIN) and cell wall invertase (CWIN) activities at the EB stage could be responsible for the lower hexose concentration at that time. The TNC remobilization amount had a positive association with the AGPase and SPS activities at the FF stage and with β-amylase activity at the PF stage in cotton tap roots in 2019 and 2020. Melatonin plays key roles in improving fruit quality and yield by regulating various aspects of plant growth. However, the effects of how melatonin regulates primary and secondary metabolites during fruit growth and development are poorly understood. In this study, the surfaces of tomato fruit were sprayed with different concentrations of melatonin (0, 50, and 100 µmol·L−1) on the 20th day after anthesis; we used high-performance liquid chromatography (HPLC) and liquid chromatography/mass spectrometry (LC/MS) to determine the changes in primary and secondary metabolite contents during fruit development and measured the activity of sucrose metabolizing enzymes during fruit development. Our results showed that 100 µmol·L−1 melatonin significantly promoted the accumulation of soluble sugar in tomato fruit by increasing the activities of sucrose synthase (SS), sucrose phosphate synthase (SPS), and acid convertase (AI). The application of 100 µmol·L−1 melatonin also increased the contents of ten amino acids in tomato fruit as well as decreased the contents of organic acids. In addition, 100 µmol·L−1 melatonin application also increased the accumulation of some secondary metabolites, such as six phenolic acids, three flavonoids, and volatile substances (including alcohols, aldehydes, and ketones). In conclusion, melatonin application improves the internal nutritional and flavor quality of tomato fruit by regulating the accumulation of primary and secondary metabolites during tomato fruit ripening. The global atmospheric CO2 concentration ([CO2]) has sharply risen due to human activities, and it has reached 410 μmol mol–1 at present. It is expected that the atmospheric [CO2] will reach approximately 550 μmol mol–1 by the middle of the 21st century. The increase in the [CO2] has a certain "fertilizing effect" on plants. On the other hand, climate warming and precipitation reductions caused by the greenhouse effect will lead to expanding arid areas and aggravated drought, which will restrict plant growth and yield. Therefore, studying the potential physiological mechanism and synergistic effects of an enriched [CO2] combined with environmental stress factors has great significance for understanding plant physiological and ecological processes. We investigated the effects of an enriched [CO2] (800 ± 40 μmol mol–1) on the antioxidant capacity and sucrose metabolism of cucumber (Cucumis sativus L. cv. 'Jinyou No. 35') seedlings under drought stress (5 or 10% PEG 6000 in nutrient solutions). The results showed that (1) the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were enhanced by the enriched [CO2]; the contents of malondialdehyde (MDA) and hydrogen peroxide (H2O2), as well as the superoxide anion radical () production rate, in the leaves were reduced; and lipid peroxidation and cytomembrane permeability decreased under drought stress. (2) The enriched [CO2] significantly improved sucrose synthase (SS) activity but decreased sucrose phosphate synthase (SPS) activity under drought stress, resulting in the accumulation of hexose through accelerated sucrose decomposition and inhibited synthesis. Thus, the osmotic potential of cells was reduced, and the water absorption capacity was enhanced. In conclusion, the enriched [CO2] relieved the detrimental effects of drought stress and enhanced the resistance of cucumber plant growth during drought stress through enhanced cellular membrane stability and antioxidative enzyme activities, thus promoting the accumulation of soluble sugar, while the MDA content and accumulation of reactive oxygen species (ROS) were reduced. Sugarcane, the world's largest source of sugar and biofuel, still carries an unassembled ~ 10 GB polyaneuploid genome, which is a major bottleneck in studying this crop at molecular level. Sugar is mainly derived from sugarcane; hence, the sucrose metabolism pathway is one of the most intriguing hotspot for sugar crop researchers. Four main gene families have been reported to be involved in sucrose metabolism including the SuSy (sucrose synthase) gene family. Sucrose synthase in plants degrades sucrose to UDP-glucose and fructose to ensure their availability for growth and developmental purposes. Here, we attempted to get deep insights into sugarcane SuSy gene family and carried out bioinformatics and expression analyses to get a high-resolution holistic snapshot. Multiple stress-responsive cis motifs, abscisic acid-responsive element (ABRE), anaerobic induction-responsive element (ARE), auxin-responsive element (AuxRE), low-temperature-responsive (LTR), wounding response element (WUN), WRKY transcription factors binding site (W-motif), predicted in the regulatory region showed their involvement in the stress signaling pathways, in addition to sucrose metabolism. Similarly, the protein interaction network analysis predicted an array of proteins of diverse range of functions. Moreover, the expression pattern of SuSy gene family in two varieties CPF251 (higher sugar level) and CPF252 (lower sugar level) was compared in leaf and internodes (top and bottom). qRT-PCR indicated the differential expression pattern of the SuSy genes in these two varieties. The expression of SoSuSy2 was high in leaf and top internodes, while low in bottom internodes indicating its activity in this tissue. In CPF251, both SoSuSy2 and SoSuSy4 displayed higher expression level; however, only SuSy2 had higher expression in CPF252. In contrast, SoSuSy6 and SoSuSy7 were the least expressed genes followed by SoSuSy1. Sugar signal mediated by Cell wall invertase (CWIN) plays a central role in seed development. In higher plants, invertase inhibitors (INHs) suppress CWIN activities at a post-translational level. In Litchi chinensis cultivar 'Nuomici', impaired CWIN expression is associated with seed abortion. Here, the expression of LcINH1 was significantly higher in the funicle of seed-aborting cultivar 'Nuomici' than big-seeded cultivar 'Heiye'. Promoter analyses found LcINH1 contained a 404 bp repeat fragment with an endosperm regulatory element of Skn-1_motif. LcINH1 and LcCWIN2/5 were located in plasma membrane. LcINH1 was able to interact with LcCWIN5, but not with LcCWIN2. In vitro enzyme activity assay demonstrated that LcINH1 could inhibit CWIN activity. Silencing LcINH1 in 'Nuomici' resulted in normal seed development, paralleled increased CWIN activities and glucose levels. Transcriptome analysis identified 1079 differentially expressed genes (DEGs) in LcINH1 -silenced fruits. KEGG analysis showed significant enrichment of DEGs in pathways related to transporters and plant hormone signal transduction. Weighted gene co-expression network analysis indicated that the turquoise module was highly correlated with fructose content, and LcSWEET3b was closely associated with early seed development. These findings suggest that LcINH1 regulate LcCWIN5 activity at the post-translational level to alter sucrose metabolism, thereby affecting early seed development in litchi. [ABSTRACT FROM AUTHOR]

Spring cold stress poses a great threat to wheat reproductive growth, leading to compromised spike development and grain yield. There are two types of cold stress i.e. chilling stress (CS, above zero) and freezing stress (FS, below zero). However, it is unclear whether there is a difference in the mechanism of CS and FS in regulating spikelet development. This study aimed to unravel the underlying regulation in determining the difference for wheat spikelet exposed to CS at 2 °C and FS at −2°C by integrative analyses of transcriptome, metabolome, and physiology. Delayed floret development and shrunken cellular morphology in both CS and FS were observed, even malformation and degradation of anther cells occurred in FS. Kyoto Encyclopedia of gene and genomes (KEGG) analyses revealed that the most abundantly enriched pathways are phytohormone biosynthesis, starch and sucrose metabolism, and phenylpropanoid biosynthesis. Further physiological assays related to the identified pathways were performed. Compared to CS, the signal of abscisic acid (ABA), salicylic acid (SA) and jasmonic acid (JA) was more pronounced, and the signal of auxin (IAA) and gibberellin (GA) was inhibited further in FS. In addition, the contents of glucose, fructose and trehalose were elevated in CS, owing to greater activities of cell wall invertase and sucrose synthase, while the hexose content was decreased owing to lower activities of such enzymes in FS, concomitantly, flavonoid barely changed in CS, but it dramatically amounted in FS. Taken together, the glucose and trehalose pathway, along with induced ABA and SA signal were intensified in CS to maintain growth, while greater flavonoid and promoted JA synthesis were induced in FS for cold survival. Abstract Strigolactone is a newly discovered type of plant hormone that has multiple roles in modulating plant responses to abiotic stress. Herein, we aimed to investigate the effects of exogenous GR24 (a synthetic analogue of strigolactone) on plant growth, photosynthetic characteristics, carbohydrate levels, endogenous strigolactone content and antioxidant metabolism in cucumber seedlings under low light stress. The results showed that the application of 10 μM GR24 can increase the photosynthetic efficiency and plant biomass of low light-stressed cucumber seedlings. GR24 increased the accumulation of carbohydrates and the synthesis of sucrose-related enzyme activities, enhanced antioxidant enzyme activities and antioxidant substance contents, and reduced the levels of H2O2 and MDA in cucumber seedlings under low light stress. These results indicate that exogenous GR24 might alleviate low light stress-induced growth inhibition by regulating the assimilation of carbon and antioxidants and endogenous strigolactone contents, thereby enhancing the tolerance of cucumber seedlings to low light stress.

Biofuels and other biologically manufactured sustainable goods are growing in popularity and demand. Carbohydrate feedstocks required for industrial fermentation processes have traditionally been supplied by plant biomass, but the large quantities required to produce replacement commodity products may prevent the long-term feasibility of this approach without alternative strategies to produce sugar feedstocks. Cyanobacteria are under consideration as potential candidates for sustainable production of carbohydrate feedstocks, with potentially lower land and water requirements relative to plants. Several cyanobacterial strains have been genetically engineered to export significant quantities of sugars, especially sucrose. Sucrose is not only naturally synthesized and accumulated by cyanobacteria as a compatible solute to tolerate high salt environments, but also an easily fermentable disaccharide used by many heterotrophic bacteria as a carbon source. In this review, we provide a comprehensive summary of the current knowledge of the endogenous cyanobacterial sucrose synthesis and degradation pathways. We also summarize genetic modifications that have been found to increase sucrose production and secretion. Finally, we consider the current state of synthetic microbial consortia that rely on sugar-secreting cyanobacterial strains, which are co-cultivated alongside heterotrophic microbes able to directly convert the sugars into higher-value compounds (e.g., polyhydroxybutyrates, 3-hydroxypropionic acid, or dyes) in a single-pot reaction. We summarize recent advances reported in such cyanobacteria/heterotroph co-cultivation strategies and provide a perspective on future developments that are likely required to realize their bioindustrial potential.

Despite well-established pathways and metabolites involved in grapevine-Plasmopara viticola interaction, information on the molecules involved in the first moments of pathogen contact with the leaf surface and their specific location is still missing. To understand and localise these molecules, we analysed grapevine leaf discs infected with P. viticola with MSI. Plant material preparation was optimised, and different matrices and solvents were tested. Our data shows that trichomes hamper matrix deposition and the ion signal. Results show that putatively identified sucrose presents a higher accumulation and a non-homogeneous distribution in the infected leaf discs in comparison with the controls. This accumulation was mainly on the veins, leading to the hypothesis that sucrose metabolism is being manipulated by the development structures of P. viticola. Up to our knowledge this is the first time that the localisation of a putatively identified sucrose metabolite was shown to be associated to P. viticola infection sites. [ABSTRACT FROM AUTHOR]

Cell wall invertase (CWI) is as an essential coordinator in carbohydrate partitioning and sink strength determination, thereby playing key roles in plant development. Emerging evidence revealed that the subtle regulation of CWI activity considerably depends on the post-translational mechanism by their inhibitors (INHs). In our previous research, two putative INHs (StInvInh1 and StInvInh3) were expected as targets of CWI in potato (Solanum tubersum), a model species of tuberous plants. Here, transcript analysis revealed that StInvInh1 showed an overall higher expression than StInhInh3 in all tested organs. Then, StInvInh1 was further selected to study. In accordance with this, the activity of StInvInh1 promoter increased with the development of leaves in plantlets but decreased with the development of microtubers in vitro and mainly appeared in vascular bundle. The recombinant protein StInvInh1 displayed inhibitory activities on the extracted CWI in vitro and StInvInh1 interacted with a CWI StcwINV2 in vivo by bimolecular fluorescence complementation. Furthermore, silencing StInvInh1 in potato dramatically increased the CWI activity without changing activities of vacuolar and cytoplasmic invertase, indicating that StInvInh1 functions as a typical INH of CWI. Releasing CWI activity in StInvInh1 RNA interference transgenic potato led to improvements in potato microtuber size in coordination with higher accumulations of dry matter in vitro. Taken together, these findings demonstrate that StInvInh1 encodes an INH of CWI and regulates the microtuber development process through fine-tuning apoplastic sucrose metabolism, which may provide new insights into tuber development. Different maize varieties respond differentially to cadmium (Cd) stress. However, the physiological mechanisms that determine the response are not well defined. Antioxidant systems and sucrose metabolism help plants to cope with abiotic stresses, including Cd stress. The relationship of these two systems in the response to Cd stress is unclear. Seed is sensitive to Cd stress during germination. In this study, we investigated changes in the antioxidant system, sucrose metabolism, and abscisic acid and gibberellin concentrations in two maize varieties with low (FY9) or high (SY33) sensitivities to Cd under exposure to CdCl2 (20 mg L−1) at different stages of germination (3, 6, and 9 days).The seed germination and seedling growth were inhibited under Cd stress. The superoxide, malondialdehyde, and proline concentrations, and the superoxide dismutase, peroxidase, catalase, and lipoxygenase activities increased compared with those of the control (CK; without Cd). The expression levels of three genes (ZmOPR2, ZmOPR5, and ZmPP2C6) responsive to oxidative stress increased differentially in the two varieties under Cd stress. The activity of the antioxidant system and the transcript levels of oxidative stress–responsive genes were higher in the Cd-tolerant variety, FY9, than in the sensitive variety, SY33. Sucrose metabolism was increased under Cd stress compared with that of the CK and was more active in the Cd-sensitive variety, SY33. These results suggest that the antioxidant system is the first response to Cd stress in maize, and that sucrose metabolism is cooperative and complementary under exposure to Cd. Summary: Invertase (INV)‐mediated sucrose (Suc) hydrolysis, leading to the irreversible production of glucose (Glc) and fructose (Frc), plays an essential role in abiotic stress tolerance of plants. However, the regulatory network associated with the Suc catabolism in response to cold environment remains largely elusive.Herein, the cold‐induced alkaline/neutral INV gene PtrA/NINV7 of trifoliate orange (Poncirus trifoliata (L.) Raf.) was shown to function in cold tolerance via mediating the Suc hydrolysis. Meanwhile, a nuclear matrix‐associated region containing A/T‐rich sequences within its promoter was indispensable for the cold induction of PtrA/NINV7.Two AT‐Hook Motif Containing Nuclear Localized (AHL) proteins, PtrAHL14 and PtrAHL17, were identified as upstream transcriptional activators of PtrA/NINV7 by interacting with the A/T‐rich motifs. PtrAHL14 and PtrAHL17 function positively in the cold tolerance by modulating PtrA/NINV7‐mediated Suc catabolism. Furthermore, both PtrAHL14 and PtrAHL17 could form homo‑ and heterodimers between each other, and interacted with two histone acetyltransferases (HATs), GCN5 and TAF1, leading to elevated histone3 acetylation level under the cold stress.Taken together, our findings unraveled a new cold‐responsive signaling module (AHL14/17‐HATs‐A/NINV7) for orchestration of Suc catabolism and cold tolerance, which shed light on the molecular mechanisms underlying Suc catabolism catalyzed by A/NINVs under cold stress. In order to better understand the changes in fruit quality and soluble sugar components in wild Actinidia eriantha at the soft ripening stage, this study explored the fruit quality indexes, soluble sugar components, sucrose metabolism-related enzyme activities and the expression of sucrose metabolism-related enzyme genes in wild A. eriantha germplasm resources. The results showed that the fruit quality of wild A. eriantha at the soft ripening stage was quite different, and the coefficient of variation of fructose content was the largest, followed by sucrose and glucose. Principal component analysis and systematic clustering analysis showed that the comprehensive performance of fruit quality indexes of M28 and M10 was the most prominent. The accumulation and composition of soluble sugar components in different wild A. eriantha varieties (lines) were not consistent. The activities of sucrose metabolism enzymes among wild A. eriantha varieties (lines) were different to some extent. The sucrose metabolism-related enzyme genes among wild A. eriantha germplasm resources had different expression patterns. The results will contribute to understanding the fruit quality changes and the mechanism of sugar metabolism in wild A. eriantha at the soft ripening stage, and lay a foundation for the protection and utilization of wild A. eriantha germplasm resources. The Rhizobium rhizogene-transformed root culture from Physalis peruviana L. (P. peruviana) may be a promising and novel source of valuable phenolics, including caffeoylputrescine (CP), which is known for antioxidant, antidiabetic, insect-resistant, disease-resistant, and neuroprotective properties. In this study, to improve the production efficiency of phytochemical components in P. peruviana hairy root cultures, we optimized various culture conditions, including the inoculum size, liquid volume, culture media type, carbon source, sucrose concentration, initial pH, and application of elicitors, to enhance the total phenolic content and CP yield in these hairy root cultures. The findings indicate that the use of sucrose as carbon source resulted in the highest biomass (13.28 g DW/L), total phenolic content (6.26 mg/g), and CP yield (2.40 mg/L). The White medium excelled in enhancing the total phenolic content (9.35 mg/g), whereas the B5 medium was most effective for the biomass (13.38 g DW/L) and CP yield (6.30 mg/L). A sucrose concentration of 5% was best for the biomass (18.40 g DW/L), whereas a sucrose concentration of 4% was ideal for the CP yield. Optimal culture conditions were as follows: an inoculum size of 0.5 g/100 mL, a liquid volume of 100 mL in a 250-mL flask, B5 medium, 4% sucrose, and a pH of 5.5. Among the tested elicitors, methyl jasmonate (MeJA) at 100 µM significantly increased the biomass (21.3 g/L), total phenolic content (23.34 mg/g), and CP yield (141.10 mg/L), which represent 0.96-, 2.12-, and 13.04-fold increases, respectively, over the control after 8 days.
