Due to pervasive and resilient soil contaminants, heterogeneously contaminated soil poses unpredictable potential threats to ecosystems. In this study, the extension of a previously developed soil algae pipe assay for evaluating heterogeneously contaminated soil under an open system is described. The assay can be used in soil that is heterogeneously contaminated with silver nanoparticles in combination with the examination of morphological changes (e.g., in vivo chlorophyll a, cell granularity, cell size, and mucilaginous sheath) and lipid contents. In addition, we attempted to extend the exposure duration under an open system. We evaluated the applicability of this soil algae pipe assay using green alga Chlamydomonas reinhardtii exposed to heterogeneous and homogeneous polyvinylpyrrolidone capping silver nanoparticles in contaminated soils. The results demonstrated that this method is an applicable bioassay that can be employed to better evaluate soil algal toxicity under an open system, with significant changes in the measured endpoints. In this study, extension of a previously developed algae-soaked disc seeding assay for periodic evaluation of subsoil contamination over time was described. The assay can be used in different contamination configurations of silver nanoparticles in combination with examination of cell morphology, esterase activity, oxidative stress, and membrane permeability. In addition, we periodically attempted to repeat the algae-soaked disc seeding assay every three weeks. We evaluated applicability of this algae-soaked disc seeding assay using alga Chlamydomonas reinhardtii exposed to heterogeneous silver nanoparticle-contaminated soils. The results demonstrated that this assay is applicable for monitoring a change of subsoil contamination by periodic evaluation over time. SUMMARY A unicellular flagellate described as Chlamydomonas monticola NIES‐157 was known to have two unequal flagella and informally stored as Heterochlamydomonas sp. in the culture collection. In this study, the strain NIES‐157 was revisited for clarifying its taxonomic status. In the 18S rDNA tree, this organism was resolved in a distinct position in the clade of Heterochlamydomonas (Reinhardtinia). In this strain, the two basal bodies were arranged in parallel as in H. inaequalis, and naked motile cells (putative gametes) were additionally observed. NIES‐157 was morphologically distinguished from other members of the genus in the nuclear position in motile cells. Based on these data, a transfer of C. monticola NIES‐157 to Heterochlamydomonas as H. monticola comb. nov. is proposed. [ABSTRACT FROM AUTHOR]

Abstract We aimed to develop an ex situ method for evaluating soil quality using soil algae. The soil algae pipe assay involves application and sampling of algae and flow cytometry. As it can be performed in a greenhouse, the assay was effective for pot tests and conducive for soil quality assessment. This novel assay will enable broader toxicity testing. Highlights • We developed a soil algae pipe assay to assess soil quality ex situ. • We validated the pipe assay using C. reinhardtii in pot tests in the greenhouse. • The assay includes 1) application, 2) sampling, and 3) flow cytometry steps. • Chlorophyll- a levels were lower in contaminated than in reference soils. This study evaluated five methods of soil inoculation using the soil alga Chlorococcum infusionum to determine the most efficient and reproducible method for promoting the growth of soil algae for toxicity testing. The five techniques included application of C. infusionum in a circle on top of the soil, to a central spot on top of the soil, to a central spot in the subsoil, to one side on top of the soil, and application divided between a circle and a central spot on top of the soil. Of these, the first method generated the greatest amount of chlorophyll fluorescence and was the method with the best reproducibility. We evaluated the applicability of this method in an assessment of the toxicity of copper and nickel to C. infusionum in two representative standard soils. Copper (20–75 mg/kg for OECD soil and 20–60 mg/kg Lufa 2.2 soil) and nickel (400–500 mg/kg for OECD soil and 60–100 mg/kg Lufa 2.2 soil) reduced the chlorophyll fluorescence of C. infusionum when the inoculation was delivered in a circle on top of both soil types. To our knowledge, this is the first study to assess the suitability of different soil algal inoculation methods for terrestrial toxicity testing. [ABSTRACT FROM AUTHOR]

Arsenic speciation and cycling in the natural environment are highly impacted via biological processes. Since arsenic is ubiquitous in the environment, microorganisms have developed resistance mechanisms and detoxification pathways to overcome the arsenic toxicity. This study has evaluated the toxicity, transformation and accumulation of arsenic in a soil microalga Scenedesmus sp. The alga showed high tolerance to arsenite. The 72-h 50 % growth inhibitory concentrations (IC values) of the alga exposed to arsenite and arsenate in low-phosphate growth medium were 196.5 and 20.6 mg L, respectively. When treated with up to 7.5 mg L arsenite, Scenedesmus sp. oxidised all arsenite to arsenate in solution. However, only 50 % of the total arsenic remained in the solution while the rest was accumulated in the cells. Thus, this alga has accumulated arsenic as much as 606 and 761 μg g dry weight when exposed to 750 μg L arsenite and arsenate, respectively, for 8 days. To our knowledge, this is the first report of biotransformation of arsenic by a soil alga. The ability of this alga to oxidise arsenite and accumulate arsenic could be used in bioremediation of arsenic from contaminated water and soil. [ABSTRACT FROM AUTHOR]
