Alkalization has been used in aquaculture to improve water quality and increase production. With its intensification, there is a demand for new alkalizing agents or the same agents but with different purities and particle sizes. Therefore, this study aimed to evaluate the effect of alkalizing on the cultivation of juvenile jundiá Rhamdia quelen in a recirculation system. The juveniles were cultured to four alkalization procedures: water without alkalization, water with sodium silicate, water with sodium bicarbonate, and water with calcium carbonate. At the end of the experiment, the biomass and biomass gain of the fish grown in sodium bicarbonate water were higher than those in the environment without alkalizing. There were no differences in other performance parameters such as weight, total length, head length, weight gain, length gain, survival, Fulton, or specific growth rate. The alkalinization doubled the alkalinity and increased the water pH, with the pH of calcium carbonate having the highest value and higher stability over time. The ammonia, nitrite, and nitrate concentrations demonstrated greater nitrification efficiency in waters that received alkalinization, mainly with calcium carbonate and sodium bicarbonate. Calcium carbonate water resulted in the highest hardness due to calcium, which provided the lowest phosphate concentration. The results, associated with the ease of obtaining and the price, suggest that sodium silicate has potential to alkalize and should be further studied. Abstract This study was conducted to determine the best combination of protein sources in diets for jundiá, based on growth, metabolism, and nutrient deposition. Five protein combinations were tested: casein + fish meal (control), casein + gelatin, casein + albumin, casein + albumin + fish meal, and albumin + fish meal, in diets containing 370 g Kg-1 of crude protein and 13.4 MJ Kg-1 of digestible energy. The fish (9.38 ± 0.12 g) were allocated in a water recirculation system at a density of 3.35 g L-1 per experimental unit and fed until apparent satiety for 40 days with the diets. The fish fed with the control diet had the highest final weight, specific growth rate, protein and feed efficiency ratio, protein retention, and best apparent feed conversion. On the other hand, fish that received casein + albumin and albumin + fish meal diets showed worse results in growth and body protein retention, low trypsin and chymotrypsin activity, and high intestinal amylase activity. Therefore, the combination referred to as control (casein + fish meal) conclusively provides the best rhythm for nutrient digestion and metabolism processes, enabling fish to reach greater growth and retention of body protein with low whole-fish fat content.

Oil extracted from fish waste could be a source of valuable fatty acids such as saturated (SFAs), monounsaturated (MUFAs), and polyunsaturated fatty acids (PUFAs). In this study, the solid fat from silver catfish (Pangasianodon hypophthalmus) waste was used to extract oil by centrifugation using different extraction solvent (distilled water, 70% ethanol, 70% acetone and 70% cyclohexane), rotation time (15, 25, 35 & 45 min), rotational speed (2000, 4000, 6000, 8000 & 10000 r.p.m) and rotational temperature (5, 10, 15, 20 & 25 °C). The compositions of fatty acids in the extracted oil were determined by gas chromatography-mass spectrometry (GC-MS) analysis. The highest oil yield of 156.7 ± 16.7 mg/g was achieved by centrifugation at 10000 r.p.m, 25 °C for 15 min using 70% acetone. The extracted oil contains 0.0223 mg/g palmitic acid, 0.0216 mg/g steric acid, and 0.0262 mg/g oleic acid. However, the essential PUFAs such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) could not be detected by GC-MS analysis. The yields of PSC at 24 h and 48 h extraction time were 23.64% and 26.43%, respectively. The chemical composition exhibited significant differences, with PSC extracted at 24 h showing better moisture, protein, fat, and ash content. Cinnamon essential oils (EOs) are widely known for their pharmaceutical properties; however, studies investigating the use of these EOs in aquaculture are scarce. The aims of this study were to evaluate the anesthetic effect of bathing silver catfish (Rhamdia quelen) with Cinnamomum cassia EO (CCEO) and its nanoemulsion (NCCEO); the growth-promoting and antioxidant effects of dietary supplementation with CCEO in silver catfish; and the in vitro antibacterial effect of CCEO, NCCEO, and Cinnamomum zeylanicum EO (CZEO) against bacteria isolated from diseased silver catfish. The two cinnamon EOs showed promising antibacterial activity, which was potentiated by the nanoemulsion. CCEO showed satisfactory anesthetic activity in silver catfish, and its nanoemulsion intensified the sedative activity. Supplementation of 1.0 mL CCEO per kg of diet for 60 days increased weight, length, and weight gain when compared to the control group, evidencing the growth-promoting activity of this EO. Dietary supplementation of CCEO for 30 and 60 days also showed an antioxidant effect, as it decreased levels of thiobarbituric acid reactive species and increased the superoxide dismutase activity in the liver of silver catfish. In this study, the influence of physical environmental factors (rainfall, temperature, water depth, turbidity, transparency and conductivity) on the temporal distribution, abundance and catch of C. nigrodigitatus from the Cross River was investigated. Fish catch and abundance were monitored fortnightly for 12 months. Surface water samples were collected twice monthly and analyzed using standard methods. Seasonal variations in rainfall, water depth, temperature, turbidity, transparency and conductivity were observed across the study period. Correlation analysis showed a positive and significant relationship between rainfall and fish catch (P0.05). On the other hand, water depth and turbidity were positively correlated with fish catch but again both relationships were not statistically significant (P>0.05). These findings suggest that rainfall, temperature, conductivity and other environmental variables play important role(s) in the distribution and abundance of Chrysichthys nigrodigitatus in the Cross River, with implication of climate change impact on the species reproductive ecology and population dynamics.

To verify the efficacy of citral in inducing sedation and anesthesia in silver catfish (Rhamdia quelen) and grass carp (Ctenopharyngodon idella) and to assess the safety of essential oil (EO) of Aloysia citriodora and citral in inducing and maintaining anesthesia in silver catfish. Clinical study, randomized, parallel, multi-arm with control group in target species. A total of 96 juvenile and 72 adult silver catfish and 80 juvenile grass carp were used. Silver catfish and grass carp were exposed to different concentrations of citral, 15–675 and 15–600 μL L–1, respectively, during the maximum period of 30 minutes to verify sedation and anesthesia induction and recovery times. In addition, for anesthetic induction, silver catfish were exposed to the EO of A. citriodora and citral at 225 μL L–1 for 3.5 minutes. Then, fish were transferred to an anesthesia maintenance solution at 50 μL L–1 for 10 minutes to assess hematologic and biochemical variables at 60 minutes, 2 and 6 days after treatment. Citral only induced sedation from 15, 25 and 40 μLL–1 in both species. Anesthesia without mortality was induced in silver catfish at 50–600 μL L–1 and grass carp at 75–450 μL L–1. At 675 and 600 μL L–1, mortality was recorded in silver catfish and grass carp, respectively. The EO of A. citriodora and citral were safe in inducing and maintaining anesthesia in silver catfish, with mean corpuscular hemoglobin concentration being the only variable that varied in relation to time and treatments. Citral was effective in inducing sedation and anesthesia in both species. In addition, A. citriodora EO and citral were safe in inducing and maintaining anesthesia in silver catfish. Background: The cost of feed in fish farming can exceed 70% of the total costs. Thus, the feed industry have sought to formulate balanced diets including low-cost by-products. Objective: To evaluate sunflower meal as a partial substitute (0, 10, and 20%) for soybean meal, with or without phytase supplementation (1,500 FTU/kg) in isoproteic (27% crude protein (CP)) and isoenergetic diets (3,400 kcal/ED/kg of feed) for silver catfish (Rhamdia quelen) juveniles. Methods: A total of 360 fish (weight: 15.77 ± 0.56 g) were randomly distributed in 24 tanks in a completely randomized design following a factorial scheme (3x2) with 4 replicates. The fish were evaluated on the following variables after 90 days: productive performance, hematological variables, histology of intestinal tissue, bromatological composition of carcass, and composition of bone mineral matter. Results: 20% dietary inclusion of sunflower meal improved weight gain and protein efficiency. No difference was observed in the other treatments and no interactions were observed between the other factors. Conclusion: 20% sunflower meal can be included in the feed of silver catfish as a substitute for soybean meal. Phytase inclusion (1,500 FTU/kg) did not improve the nutritional value of the treatments.

Maclura tinctoria is a tree species native from Brazil and rich in phenolic compounds. Since plant antibacterial activity is highly associated with phenolic compound concentration, we aim to evaluate the in vitro antimicrobial activity of different extracts against fish pathogenic bacteria. In addition, some phenolic compounds have central depressant effects and can be useful in aquaculture due to possible sedative and/or anesthetic effects. Four M. tinctoria extracts were extracted separately with ethanol; leaves (LE), bark (BE), heartwood (HE), and the sapwood (SE). In vitro antimicrobial activity was tested against Aeromonas strains at concentrations of 6400 to 3.125 μg/mL. The sedative effect was evaluated for 24 h with 30 and 100 mg/L concentrations. Chemical composition was analyzed by HPLC-DAD-MS. The HE extract had the best MIC (400 μg/mL) and MBC (800 μg/mL) compared to the LE, BE, and SE extracts. LE extract induced deep sedation and the BE, SE, and HE extracts induced light sedation. Additionally, BE, SE, and HE induced a normal behavior without side effects. Polyphenolic compounds with antimicrobial activity and sedative effects were identified mainly in HE. Thus, HE extract is safe and can be used as a sedative for silver catfish. This study was conducted to evaluate the prebiotic action of distinct linseed fibers on diets for silver catfish (Rhamdia quelen) juvenile. For this, soluble and insoluble fractions of linseed fiber were concentrated separately and combined in four ratios (1:0.5, 1:1, 1:2, 1:4), which were added in diets and evaluated along with a control diet. After 45 days receiving the experimental diets, the animals were submitted to biometry for data collection and samples. The experimental design was completely randomized, data were submitted to analysis of variance, and the means were compared by Tukey's test (P < 0.05). Fish performance was higher for the group that received the 1:2 and 1:4 diets. The production of intestinal acetic acid was higher in fish fed on the 1:2 diet, and butyric acid was higher with 1:4 diet and the propionic acid with the control diet. The control diet led to lower counts of goblet cells. Total proteins and globulins in plasma, mucoprotein, total immunoglobulins, and cutaneous mucus pH were higher in fish fed on diets 1:2 and 1:4. Cortisol levels and intestinal pH were lower in these treatments. The linseed fiber intake increased total plasma immunoglobulins and plasma alkaline phosphatase activity and in mucus. It was concluded that the use of 1:2 and 1:4 soluble fiber/insoluble linseed in the diet beneficially modulates the production of short-chain fatty acids in the digesta, with reflection on growth performance, number of goblet cells in the intestinal epithelium, and improvement in plasma and skin mucus parameters of fish. Plants are sources of secondary metabolites, and some are rich in essential oils (EOs), which possess a bioactive pharmacological importance as sedatives and anaesthetics that have been experimentally demonstrated in fish. In search of products with lower residual effects and optimization of the production based on the local flora, the properties of the EO of Aloysia triphylla (EOAT) have been investigated. The samples of two years were analysed for the percentage variability of the chemical composition and the totality of the major components according to the season. Limonene, citral isomers (α and β), caryophyllene, caryophyllene oxide and spathulenol were the main constituents of the EOAT throughout these two years. In the second year, caryophyllene predominates over its oxide in all seasons; however, in the first year, it occurs only in autumn and winter. Seasonal pools of EOs were made for the biennium obtaining four samples, and silver catfish (Rhamdia quelen) juveniles were subjected to a concentration range of 20–800 μl/L EOAT in order to assess induction and recovery times of anaesthesia. The relationships between percentages of individual components in EOAT and time to induce deep anaesthesia indicate that its activity may be attributed to the association of several of its components. ABSTRACT: Anesthetics are effective to reduce or minimize stress in handling operations related to fish cultures. Aiming to avoid residual effects of synthetic drugs, several natural agents, such as essential oils (EOs), have been tested. The aim of this study was to determine the optimal concentration of the EO of Ocimum micranthum for induction of anesthesia in juveniles of silver catfish (Rhamdia quelen) and grass carp (Ctenopharyngodon idella). Results demonstrated that the most suitable concentration to sedate silver catfish and carp is 25μL/L, while to anesthetize 200μL/L is more suitable for silver catfish and 100μL/L for grass carp, because these concentrations induce deep anesthesia in less than three min and recovery in less than five min. Analysis of the EO constituents showed methyl chavicol, a compound with carcinogenic potential, as the major component (58.2%). Therefore, the EO of O. micranthum is not recommended for fish anesthesia if the fish are to be used as food.

The immune modulating activity of β-glucan on aquatic species has been a matter of intense investigation. Here, we aimed to investigate the effect of β-glucan on wound healing of silver catfish, a Neotropical South American scale-free fish. Small sections of skin and muscle (3 mm in diameter) were removed and fish were bathed daily with β-glucan (0.1% and 0.5%) up to 28 days when cicatrization was complete. A group of fish similarly injured and non-exposed to β-glucan was used as control. Wound closure and healing was monitored visually and by histopathological analysis. In fish bathed with 0.5% β-glucan we found reduced blood cortisol levels at day one post-wounding and, by day 7 post wounding, the deposition of granulation tissue was higher compared to non-exposed fish. In addition, from day 7 forward, wound size was significantly lower in fish bathed with 0.5% β-glucan. Histopathological analysis of the wounded site indicated a thin layer of immature epidermal cells at day one post wounding. A discrete inflammation with mixed inflammatory cell infiltrate was observed on wounded muscle and was lower by day 7 post wounding on fish bathed with 0.5% β-glucan. By day 14 post wounding, the deposition of collagen fibers and the presence of fibroblast and new muscle fibers were higher in fish exposed to 0.5% β-glucan, and dermis restoration was complete. Mycotoxins are secondary metabolites produced by varieties of fungi that contaminate food and feed resources and are capable of inducing a wide range of toxicity. This problem is extensively aggravated due to the increasing replacement of fish meal by plant-derived proteins. Among the mycotoxins, aflatoxins have received a great deal of attention owing to their great prevalence in plant feedstuffs and to the detrimental effects on animals. The objective of this study was to evaluate whether dietary supplementation with tea tree (Melaleuca alternifolia) oil (TTO) would avoid or minimize the negative impacts on silver catfish (Rhamdia quelen) fed with aflatoxins-contaminated diets. Four treatments were tested: control (fish fed with a control diet); AFB (fish fed with a mycotoxin-contaminated diet – 1893 μg kg−1 of AFB 1 and 52.2 μg kg−1 AFB 2); TTO (fish fed with a control diet + 1 mL kg−1 of TTO), and TTO + AFB (fish fed with a mycotoxin contaminated diet - 2324 μg kg−1 of AFB 1 and 43.5 μg kg−1 AFB 2 + 1 mL kg−1 of TTO). Diets were tested in three replications and analyzed at days 5 and 10 of dietary intake. Significantly reduced antioxidant enzymes (SOD, GPx, and GST) and increased lipid peroxidation (LOOH) and protein carbonyl (PC) content in plasma and liver, with 16.6% mortality occurrence, were observed in the group fed aflatoxin-contaminated diet. Furthermore, aflatoxins also significantly increased plasmatic and hepatic ROS levels and decreased hepatic antioxidant capacity against peroxyl radical (ACAP) levels. Plasma cortisol levels were not altered by aflatoxicosis, but the intoxication induced hepatose. Notwithstanding, addition of TTO to the groups receiving aflatoxins showed a protective effect, avoiding the increase of ROS, LOOH, and PC levels in plasma and liver. Moreover, TTO treatment ameliorated the aflatoxin-associated liver damage. Thus, TTO supplementation at concentration of 1 mL kg−1 in feed may be used in fish to increase antioxidant status and reduce the negative effects caused by aflatoxins toxicity. The use of natural products, such as essential oils (EOs), is a potential novel approach to treat fish bacterial infections with a lower risk of developing resistance. There has been a number of studies reporting the activity of EOs as those obtained from the species Achyrocline satureioides, Aniba parviflora, Aniba rosaeodora, Anthemis nobilis, Conobea scoparioides, Cupressus sempervirens, Illicium verum, Lippia origanoides, and Melaleuca alternifolia against bacteria. However, there are few studies investigating the effect of these EOs against fish bacteria. Therefore, the aim of this study was to evaluate the in vitro antibacterial activity of EOs against the following fish bacteria, Aeromonas hydrophila, Citrobacter freundii, and Raoultella ornithinolytica. Additionally, the in vivo antibacterial activity of the EO L. origanoides was evaluated against experimentally induced A. hydrophila infection of silver catfish (Rhamdia quelen). The EO of L. origanoides was chosen as it showed the highest in vitro antibacterial activity, with minimum inhibitory concentrations ranging from 0.2 to 0.8 mg mL-1. This EO also presented a therapeutic success of 58.33%, on a 30 day A. hydrophila infection. Abstract The aim of this study was to evaluate the resistance of Rhamdia quelen (silver catfish) to Aeromonas hydrophila infection after treatment with pure and nanoencapsulated forms either terpinen-4-ol, thymol, or carvacrol and the effects of these treatments on fish metabolic responses. After A. hydrophila inoculation, fish were treated with 30 min daily baths for 6 consecutive days with terpinen-4-ol, thymol, or carvacrol in their pure or nanoencapsulated forms at concentrations of 5, 10, 15 or 25 mg L−1. A positive control group, negative control group and saline group were also included. Survival was evaluated at the end of treatment for six consecutive days. Muscle and liver were collected to determine glucose and lactate levels. The fish treated with the nanoencapsulated form of the compounds had a high survival rate, similar to saline group and negative control groups. The carvacrol, thymol and terpinen-4-ol nanoencapsulated forms improved survival of silver catfish infected with A. hydrophila. Muscle and liver glucose and lactate levels are not indicated as biomarkers because they did not present any correlation between the metabolic state of the fish and the bacterial infection. The aims of this study are to isolate and characterize acid soluble collagen (ASC) and pepsin soluble collagen (PSC) extracted from silver catfish (Pangasius sp.) skin. Isolated ASC and PSC collagen were characterized in terms of chemical composition (moisture, protein, fat and ash content), protein concentration, functional group, solubility, and morphological properties as compared to commercial collagen. Yields of ASC and PSC were 4.27% and 2.27%, respectively. The chemical compositions of raw skin were 34.64%, 2.81%, 3.68%, and 0.31%, while the chemical compositions of ASC and PSC were 94.21%, 3.48%, 0.81%, 59.15%, and 88.25%, 3.46%, 0.92%, and 29.24%, for moisture, protein, fat, and ash, respectively. ASC and PSC had protein concentrations of 2.27 mg/mL and 2.70 mg/mL, respectively. Functional group analysis revealed that both isolated collagens exhibited Amide A, II and III as a fingerprint for collagen structure. The highest solubility was found at pH 4 for ASC, pH 1 for PSC, and pH 5 for commercial collagen. The morphology of the isolated collagens was porous and they contained fibril. In conclusion, the characteristics of the isolated ASC and PSC from silver catfish (Pangasius sp.) skin indicate that value-added collagen can be produced from the alternative source of freshwater fish. Abstract This study evaluated the antibacterial activity of Origanum majorana essential oil (EOM) and nanocapsules of this oil (NOM) in silver catfish, Rhamdia quelen , infected with Aeromonas hydrophila , and addressed their effects on silver catfish hematological and metabolic parameters. Fish were inoculated with A. hydrophila (360 μL, at a concentration of 1.5 × 109 CFU mL−1) and submitted to 1 h daily baths with EOM (0 (control), 20 or 30 μL L−1), NOM (0 (control), 5 or 10 μL L−1) or a positive control containing florfenicol (30 μL L−1) called group Maxflor® for five consecutive days. All treatments improved the survival rate of the infected fish, but we suggest the treatment of A. hydrophila infections through daily baths with 20 μL L−1 EOM or 5 μL L−1 NOM for five consecutive days as these were the lowest effective concentrations tested. Silver catfish treated with EOM and NOM had higher lymphocyte levels, indicating stimulation of the immune system in these fish. The lowest liver glucose level was found in the group treated with the lowest concentration of NOM, and the lactate values in the liver and muscle of all groups were within the normal values reported for this species. In addition, nanocapsules required much less EOM to elicit effective antibacterial treatment. Thiamethoxam is a neonicotinoid pesticide utilized on a worldwide scale, it has been reported in freshwater ecosystems, and detected in fishery products. Nevertheless, there is a lack of information about thiamethoxam sublethal effects on the gills of freshwater fish, principally linked to energetic metabolism. In this context, creatine kinase (CK) is an enzyme of the phosphoryl transfer network that provides a temporal and spatial energy buffer to maintain cellular energy homeostasis in tissues with high energy requirements, such as gills. Based on this evidence, the aim of this study was to evaluate whether exposure to thiamethoxam impairs the cytosolic and mitochondrial CK activities in gills of Rhamdia quelen , and the involvement of oxidative stress in the energetic imbalance. Branchial CK (cytosolic and mitochondrial) activity and sodium‑potassium pump (Na + , K + -ATPase) were inhibited, and adenosine triphosphate (ATP) levels decreased after 96 h exposure to 1.125 and 3.75 μg/L thiamethoxam compared to the control group. Moreover, levels of branchial thiobarbituric acid reactive substances (TBARS) and protein carbonylation increased at 3.75 μg/L thiamethoxam after 96 h of exposure compared to the control group, while the non-protein thiol (NPSH) content did not differ between groups. It is important to emphasize that all evaluated parameters did not recover after 48 h in clean water. To summarize, the data presented here clearly demonstrated that thiamethoxan exposure severely impairs cytosolic and mitochondrial CK activities, a key enzyme for gill energy buffering to maintain cellular energy homeostasis, and this effect appears to be mediated by oxidation of lipid and protein molecules, which consequently thereby induces oxidative stress. In aquaculture, nutrition and supplemented diets have been shown to affect broodstock reproductive performance. In this study, we investigated the effects of dietary supplementation with Cymbopogon flexuosus essential oil (CFEO) microcapsules on reproductive-related parameters in silver catfish (Rhamdia quelen) male broodfish. Adult male broodstocks were separated into three groups according to the concentrations of supplemented CFEO (0.0 = control; 1.0 or 3.0 mL per kg of diet). After 20 days under experimental conditions, the animals were euthanized and the gonads were harvested for gonadosomatic index, sperm analysis, oxidative stress, and histopathology; testosterone levels were measured in the plasma; gene expression of prl, smtl, pomca, and pomcb was assessed in the pituitary gland by real-time PCR. The results showed no alterations on reproductive parameters in R. quelen males treated with Cymbopogon flexuosus essential oil compared to the control-diet animals. In conclusion, CFEO microcapsules supplied for 20 days in the concentrations of 1.00 or 3.00 mL per kilogram of diet did not affect the reproduction criteria evaluated in this study in male silver catfish. The use of multiple biomarkers has been shown to be an efficient method for evaluating environmental contamination. In this work, we evaluate neurotoxic effects and the antioxidant system responses of the R. branneri collected in two streams of lower Iguazu River basin, relating them with different percentage of vegetation coverture, presence of pesticides and fall and winter seasons. The biological samples were collected in March and August of 2015, from two streams that belong to the lower Iguazu River basin (Brazil): the Manoel Gomes River and the Arquimedes Stream. Soil analyses were performed, and the results showed the presence of the following organophosphates in the Manoel Gomes River and the Arquimedes Stream: disulfoton, methyl parathion, and ronnel. The present study detected inhibition of cholinesterase activity in the brain and muscle of fish samples during the fall from the Manoel Gomes River and the Arquimedes Stream. In the Manoel Gomes River, elevated lipoperoxidation was also observed during the fall. It was observed that the increase or decrease of biomarkers was related to temporal variation and, possibly, to the exposure of animals to agrochemicals. Although the Manoel Gomes River and the Arquimedes Stream are located in regions with large areas of vegetation, the soil analyses show that agrochemical residues are able to reach these locations, which suggests that the fauna are in contact with oxidant and anti-cholinesterase agents during the fall, in addition to respond differently during each season. It is known that the cytotoxic effects of aflatoxin B 1 (AFB 1 ) in endothelial cells of the blood–brain barrier (BBB) are associated with behavioral dysfunction. However, the effects of a diet contaminated with AFB 1 on the behavior of silver catfish remain unknown. Thus, the aim of this study was to evaluate whether an AFB 1 -contaminated diet (1177 ppb kg feed −1 ) impaired silver catfish behavior, as well as whether disruption of the BBB and alteration of neurotransmitters in brain synaptosomes are involved. Fish fed a diet contaminated with AFB 1 presented a behavioral impairment linked with hyperlocomotion on days 14 and 21 compared with the control group (basal diet). Neurotransmitter levels were also affected on days 14 and 21. The permeability of the BBB to Evans blue dye increased in the intoxicated animals compared with the control group, which suggests that the BBB was disrupted. Moreover, acetylcholinesterase (AChE) activity in brain synaptosomes was increased in fish fed a diet contaminated with AFB 1 , while activity of the sodium-potassium pump (Na + , K + -ATPase) was decreased. Based on this evidence, the present study shows that silver catfish fed a diet containing AFB 1 exhibit behavioral impairments related to hyperlocomotion. This diet caused a disruption of the BBB and brain lesions, which may contribute to the behavioral changes. Also, the alterations in the activities of AChE and Na + , K + -ATPase in brain synaptosomes may directly contribute to this behavior, since they may promote synapse dysfunction. Abstract: Rhamdia quelen is the most produced native freshwater fish in fish farms in South Brazil. Until recently, Rhamdia branneri and Rhamdia voulezi were synonyms of R. quelen, and all the species are commercialized as silver catfish (locally called jundiá or bagre sapo) by the aquaculture industry. In fact, because these species are morphologically very similar, interspecific crosses easily might occur in fish farming. We employed standard DNA barcoding to identify jundiá molecular operational taxonomic units (MOTUs) in fish cultivated and commercialized in the industry and in possible escapees in the natural environment in southern Brazil. We analysed 48 individuals from six fish farms and 48 individuals from three rivers (Uruguay, Benedito Novo and Itapocu Rivers) using the mitochondrial cytochrome oxidase subunit I (COI). Four MOTUs were identified based on the estimated optimum threshold (OT = 0.77), and these MOTUs were concordant with Bayesian Inferece (BI) and Neighbour‐Joining (NJ) trees. Our results support the existence of at least three species in our dataset: R. branneri, R. voulezi, and R. quelen 1 and R. quelen 2. The interspecific genetic divergence ranged from 1.1% to 5.1% (mean = 3.5%), and the intraspecific distance ranged from 0% to 1.4% (mean = 0.24%). The presence of cultivated fish in the Uruguay and Benedito Novo Rivers provides evidence of genetic contamination in native populations. These results show the need to regulate aquaculture activities and to characterize the species and commercial lineages of silver catfish that are cultivated in South Brazil. [ABSTRACT FROM AUTHOR]

Abstract The fish may experience periods of food deprivation or starvation which produce metabolic changes. In this study, adult Rhamdia quelen males were subjected to fasting periods of 1, 7, 14, and 21 days and of refeeding 2, 4, 6, and 12 days. The results demonstrated that liver protein was depleted after 1 day of fasting, but recovered after 6 days of refeeding. After 14 days of fasting, mobilization in the lipids of the muscular tissue took place, and these reserves began to re-establish themselves after 4 days of refeeding. Plasmatic triglycerides increased after 1 day of fasting, and decreased following 2 days of refeeding. The glycerol in the plasma oscillated constantly during the different periods of fasting and refeeding. Changes in the metabolism of both protein and lipids during these periods can be considered as survival strategies used by R. quelen. The difference in the metabolic profile of the tissues, the influence of the period of fasting, and the type of reserves mobilized were all in evidence.

ABSTRACT This study evaluated anesthetic efficacy and possible effects of the essential oils (EOs) of Cunila galioides (EOC) and Origanum majorana (EOO) on ventilatory rate (VR) and ionoregulation in Rhamdia quelen. In the anesthesia assessments, 50, 100, 200 and 300 μL L-1 EOC and 50, 100, 200, 300, 400 and 500 μL L-1 EOO were tested, and time for induction to sedation and anesthesia stages, as well as recovery, were taken. A second trial employed lower concentrations of both EOs, 10, 25, 50 and 100 μL L-1, in order to verify VR and Na+, K+ and Cl- whole body net fluxes. Sedation was achieved with both oils at 100 µL L-1, and anesthesia at ≥ 200 µL L-1. There was no significant difference between control and EO-treated groups regarding VR, but all fish subjected to 100 µL L-1 EOC died within 2 h of exposure. Overall, ionic loss declined in the presence of the EOs. The EOC at 200 - 300 μL L-1 and EOO at 400 - 500 μL L-1 present the potential to promote fast anesthesia in R. quelen.

The citral (EO-C) and linalool (EO-L) chemotypes were identified, and both presented similar anesthetic effects for silver catfish. Fish were exposed to two concentrations of each EO, which induced slow and fast anesthesia (100 and 300 µL L-1, respectively). Blood ions did not change at any time of anesthesia induction and recovery and, therefore, the electrolyte balance was not altered. Blood gases oscillated through all exposure and recovery times, but there was an increase in pO2 after 10 min recovery in fish anesthetized with EO-C. Glucose increased in fish exposed to both EOs when compared with the control group. Overall, exposure to both EOs (except 100 µL L-1 EO-L at most times) reduced plasma cortisol levels compared to the control and/or ethanol groups. However, as plasma creatinine levels in fish anesthetized with EO-C were higher than control fish, the use of EO-L is preferable.

The aim of this work was to investigate the effects of glyphosate on the antioxidant system, as well as the neurotoxic effects on the larvae of Rhamdia quelen . A completely randomized design was implemented with the eggs of silver catfish distributed in 48 containers with 300 mL of water, which were subdivided randomly into two groups: control and treated with 6.5 mg L of glyphosate. These groups were evaluated at four time points (12 h, 24 h, 48 h, and 72 h), each with six replications. The survival rate of eggs/larvae (%) was evaluated, and samples were collected for antioxidant system analysis (catalase – CAT, glutathione transferase – GST, glutathione reductase – GR, and lipoperoxidation – LPO), and neurotoxic evaluation (cholinesterase – ChE). Throughout the 72 h of experimentation, there was a higher survival rate among the animals treated with glyphosate. The highest value of integrated biomarkers response (IBR = 1.26) was at 12 h, presenting induction of the cholinesterase (ChE) enzyme and GR. At 24 h, the value of IBR was −2.56, with inhibition of ChE and induction of GR. At 48 h, the value was −0.76, with induction of LPO. The lowest value of IBR was at 72 h (−4.65), with induction of GST and inhibition of all other biomarkers. Finally, it was possible to detect an acute effect of glyphosate throughout the early development of R. quelen , with a decrease in the antioxidant system control and neurotoxic effects. The objective of this study was to evaluate the effects of garlic (Allium sativum) essential oil on the rates of eggs fertilization and hatching and on the normal development of larvae of artificially incubated silver catfish (Rhamdia quelen). The experiment was carried out using a completely randomized experimental design with four treatments and five replicates. The treatments consisted of introducing garlic essential oil into the incubators, which used a closed system of water recirculation, at concentrations of 0, 1, 3, and 5mg L-1. Rates of eggs fertilization and larval hatching were determined at 10 and 24h after the beginning of eggs hydration. Morphology of larvae was assessed with the aid of a stereo microscope to determine the rate of normal development. The concentration of 5mg L-1 promoted better rates of eggs fertilization and larval hatching. Concentrations of garlic essential oil had a linear effect on eggs fertilization rates and larval hatching. Garlic oil did not influence the morphology of the larvae (P>0.05). It is recommended to use 5mg L-1 of garlic essential oil to promote better rates of eggs fertilization and larval hatching of artificially incubated silver catfish. [ABSTRACT FROM AUTHOR]

Concentrations of heavy metals, aluminium (Al), chromium (Cr), copper (Cu), iron (Fe), nickel (Ni), vanadium (V) and zinc (Zn) were analysed in the tissues of two estuarine fish species; Tilapia ( Sarotherodon melanotheron) and silver catfish ( Chrysichthys nigrodigitatus) and also in the surface sediments from Okrika estuary in the Niger Delta area of Nigeria. The sampling was conveyed in two seasons, October/November, 2015 and January/February, 2016, representing the climax of both wet and dry seasons, respectively. The levels of the metals in the fish species and surface sediments were analysed by inductively coupled plasma-atomic emission spectroscopy (ICP-AES). The sequence of the metal accumulations in Tilapia was; Fe > Al > Zn > Cu > Cr > V > Ni, in silver catfish; Fe > Al > Zn > Cu > Cr > Ni > V and for sediment; Al > Fe > Zn > Cr > V > Ni > Cu. Levels of the metals in the fish species and sediment were higher than the international permissible limits based on the criteria or application of relevant pollution indices and standards. This indicates that the two fish species were not fit for human consumption. A two-way analysis of variance was used to test the difference in metal concentrations in the fish tissues and sediment. Mean differences were separated by the use of t test. Although, some degree of pollution was found in the reference or control site because Niger-Delta is strongly localised and congested with oil-refining and other industrial activities thereby making it difficult to get a clean estuary. In comparison to the metal accumulations in the tissues of the fish species from oil-impacted creeks (Ekerekana, Okari and Ogoloma) and less oil-impacted counterpart (control site), higher levels ( p < 0.05) of the metals were observed in the impacted than the less impacted creek. For most of the metals, S. melanotheron had higher concentrations than the C. nigrodigitatus. Seasonal variations reflected that there was higher accumulation of the metals in the two fish species in the rainy season than the dry season. For sediment, there was significant difference for most of the metals between these two creeks except for Al. The multivariate tools of principal component analysis, Pearson correlation analysis and the use of vanadium/nickel ratio were used to confirm the possible source of the metals in the environmental matrices from oil pollution. Therefore, it is mandatory and expedient to ensure regular monitoring of the trace element loadings in these creeks and to take appropriate measures to alleviate the incident to safeguard the health of the public. Among all organotin compounds , triphenyltin hydroxide (TPhTH) is widely used as fungicide and moluscicide in Brazil. However, the effects of TPhTH on the biochemical parameters of non-target organisms, such as fish, are little known. The aim of the present study is to assess the possible toxic effects of different concentrations of waterborne TPhTH on silver catfish belonging to species Rhamdia quelen. The fish were exposed to two different concentrations of TPhTH (1.08 and 1.70 µg/L as Sn) for 15 days and then compared to the control group (triplicate, n = 3). The antioxidant profile (catalase (CAT) and the glutathione S-transferase (GST)) and the oxidative stress parameters (TBARS-thiobarbituric acid-reactive substances and protein carbonyl (PC)) were set after the exposure to TPhTH. The TBARS level and the PC content increased in several organs of the Rhamdia quelen (brain, liver, muscle and gills) under the two concentrations of TPhTH in comparison to the control group. The CAT activity in the liver and gills has enhanced in all tested TPhTH concentrations. The G ST activity increased in the brain, liver and muscle tissues under all the TPhTH concentrations. The significant changes in the biomarkers indicated that the investigated pesticide could have harmful effect on fish, in the field. Appropriate control of immune responses is a critical determinant of fish health. The cholinergic and adenosinergic systems have important roles in the immune response, since acetylcholine (ACh) and adenosine (Ado) possess important anti-inflammatory properties and modulate the immune response during infectious diseases. Thus, the aim of this study was to evaluate whether splenic and lymphocytic adenosinergic and cholinergic systems are able to modulate the immune response during Pseudomonas aeruginosa infection. Acetylcholinesterase (AChE) and adenosine deaminase (ADA) activities in spleen tissue and splenic lymphocytes increased in infected animals compared to uninfected animals. Moreover, plasmatic and splenic metabolites of nitric oxide (NOx) increased in infected animals compared to uninfected animals. Therefore, our results support the hypothesis that the cholinergic and adenosinergic systems are not able to modulate the immune response against P. aeruginosa . This study evaluated sequential double-temperature shock (heat and cold) to induce triploidy in Rhamdia quelen. Fertilized eggs were heat shocked at 37, 39 and 41°C at 2 min post-fertilization and then cold shocked at 1°C for 20 min. Intact eggs were used as controls. Egg viability in control groups was higher (65.5 ± 37.0%) than that at 37°C (58.2 ± 37.7%), 39°C (1.8 ± 0.3%) and 41°C (0%). The survival of control hatchery (64.6 ± 36.8%) was higher compared to treatments at 37°C (24.4 ± 15.5%) and 39°C (0.6 ± 0.07%). Triploid percentages were 98.5% at 37°C and 100% at 39°C. Therefore, while double-temperature shock induced 100% triploidy in R. quelen, it also significantly decreased embryo survival and increased the deformity larvae. [ABSTRACT FROM AUTHOR]

Fish reared in heterogeneous batches and fed on food with a very small particle size can practice cannibalism, because it is energetically more feasible. Jundiá is an omnivorous fish that exhibits heterogeneous growth and high cannibalism rate (CR) in the early stages of its life. Thus, to increase the survival of jundiá, regular size grading has become a necessity to regularly sort into narrow size ranges. Therefore, the purpose of this study was to evaluate the changes in the cannibal behavior of jundiá fry obtained from a wild pair and fed on food with different particle sizes. The behavior of 20 groups of 12 fry composed of five small-sized prey (13.51 ± 1.75 mm), five medium-sized prey (20.76 ± 2.66 mm), and two predators (34.47 ± 3.36 mm) was observed for 42 h. Commercial food with five different particle sizes were given to these groups in four replicates referred to as S = <200 μm, M = 200–400 μm, L = 400–600 μm, and SL = S + L or ML = M + L. Behavioral interactions were observed, and at the end of the study, the total length (TL), mouth gape of the predators, and TL and head height of the prey were measured. It was found that CR was unaffected by the different food particle sizes ( P > 0.05) and that 82.71%–100% of the prey with head height smaller than or equal to the predator’s mouth gape were ingested. The swallowed prey weighed around 11.63% ± 4.30% of the predator weight, and 62.72% of cannibalism occurred during the night. Moreover, it was verified that if the difference in the TL of the individuals of the batch is >13.7 mm, then the smaller fish may be eaten by the larger ones. Thus, it is concluded that cannibalism persists despite the supply of more energetically favorable rations and that size grading remains the only practical method to reduce cannibalism in the hatchery of jundiá fry obtained from a wild pair.
