Candida albicans is often implicated in nosocomial infections with fatal consequences. Its virulence is contributed to hydrolytic enzymes and biofilm formation. Previous research focused on studying these virulence factors individually. Therefore, this study aimed to investigate the impact of biofilm formation on the hydrolytic activity using an adapted low-cost method. Eleven strains of C. albicans were used. The biofilms were formed on pre-treated silicone discs using 24-well plates and then deposited on the appropriate agar to test each enzyme, while the planktonic cells were conventionally seeded. Biofilms were analysed using Raman spectroscopy, fluorescent and scanning electron microscopy. The adapted method provided an evaluation of hydrolytic enzymes activity in C. albicans biofilm and showed that sessile cells had a higher phospholipase and proteinase activities compared with planktonic cells. These findings were supported by spectroscopic and microscopic analyses, which provided valuable insights into the virulence mechanisms of C. albicans during biofilm formation. Hydraulic fracturing, or fracking, requires large amounts of water to extract fossil fuel from rock formations. As a result of hydraulic fracturing, the briny wastewater, often termed back-produced fracturing or fracking water (FW), is pumped into holding ponds. One of the biggest challenges with produced water management is controlling microbial activity that could reduce the pond water's reusable layer and pose a significant environmental hazard. This study focuses on the characterization of back-produced water that has been hydraulically fractured using chemical and biological analysis and the development of a high-throughput screening method to evaluate and predict the antimicrobial effect of four naturally and commercially available acidic inhibitors (edetic acid, boric acid, tannic acid, and lactic acid) on the growth of the FW microbiome. Liquid cultures and biofilms of two laboratory model strains, the vegetative Escherichia coli MG1655, and the spore-forming Bacillus atrophaeus (also known as Bacillus globigii, BG) bacteria, were used as reference microorganisms. Planktonic bacteria in FW were more sensitive to antimicrobials than sessile bacteria in biofilms. Spore-forming BG bacteria exhibited more sensitivity to acidic inhibitors than the vegetative E. coli cells. Substantial research exists on predation and its ecology. Most research has focused on durophagous fishes, brachyuran crabs, and lobsters. Data are lacking, however, on soft-bodied predators like anemones, and their contribution to overall levels of predation remains largely unevaluated. Here, we compared predation rates of the durophagous predator, the crab C. maenas and the soft-bodied predator, the anemone Actinia equina on 15 intertidal shores around Anglesey, north Wales, UK. We employed a novel approach to assess predation based on measuring faecal output from recently collected individuals and converting it to food consumed using absorption efficiencies (AEs) measured using potential prey species inhabiting the same shores. Anemone mean abundance was 8.21 (± 0.27, s.e.) individuals.m−2, whereas for C. maenas it was 0.23 (± 0.02, s.e.) individuals.m−2. AEs when fed mussel tissue, a polychaete worm, or a shrimp were 92.8–94.0% in C. maenas and 40.5–95.8% in A. equina. This difference in values reflected the different feeding modes of the two predators. Unexpectedly, A. equina consumed 3.5–7 times more prey than C. maenas. The consumption of larger amounts of prey by an anemone than the dominant durophagous predator has important consequences for calculating energy flows in food webs, understanding predation controls in assemblages, and potentially for wider predation trends.

Background/Aims: A full colonoscopy is currently required in children and adolescents with colorectal polyps, because of their potential of neoplastic transformation and complications such as intussusception. We aimed to analyze the associations of polyp characteristics in children and adolescents with colorectal polyps. Here, I describe a new rotifer species within the Ptygura elsteri group collected from Mallorca, Spain (Palearctic). Previously reported from the Nearctic, this form possesses anatomical and ecological characteristics that indicate it to be a separate species. While other morphotypes assigned to the P. elsteri group have cervical hooks, the hooks on this morphotype are clearly different. In this report, I discuss previously published observations of this form, describe its anatomical and ecological details, and discuss its taxonomic position within the genus Ptygura. The name for this new species is Ptygura innominata n. sp.

The taxonomy of Cretaceous cirripedes referred to the sessile orders Brachylepadomorpha and Verrucomorpha is revised. New taxa include the brachylepadid genera Crithmumlepas (type species C. hoensis sp. nov., C. aycliffensis sp. nov.) and Calvatilepas (type species C. recurvus sp. nov.). The family Pycnolepadidae nov. is established (constituent genera Pycnolepas, Faxelepas) and Pycnolepas batchelorum sp. nov. is described; a new eoverrucid species, E. barringtonensis sp. nov. is erected. Cladistic analysis of 48 characters of 18 operational taxonomic units, including 16 in-group sessile taxa yielded a consensus tree showing the strongly supported monophyly of Brachylepadomorpha + Verrucomorpha + Balanomorpha. It is recommended that the Order Brachylepadomorpha is abandoned, and its constituent families are identified as, respectively, stem group Verrucomorpha (Pycnolepadidae) and stem group Balanomorpha (Brachylepadidae). The sister-group relationship of Verrucomorpha and Balanomorpha, identified from many molecular studies, is confirmed from shell morphological data. The Neolepadoidea are shown to closely parallel the sessile adaptations of verrucomorphs and balanomorphs. Abandoned and flooded ore mines are examples of hostile environments (cold, dark, oligotrophic, trace metal) with a potential vast diversity of microbial communities rarely characterized. This study aimed to understand the effects of depth, the source of water (surface or groundwater), and abiotic factors on the communities present in the old Forsyth iron mine in Quebec (Canada). Water and biofilm samples from the mine were sampled by a team of technical divers who followed a depth gradient (0 to 183 m deep) to study the planktonic and sessile communities' diversity and structure. We used 16S/18S rRNA amplicon to characterize the taxonomic diversity of Bacteria, Archaea, and Eukaryotes. Our results show that depth was not a significant factor explaining the difference in community composition observed, but lifestyle (planktonic/sessile) was. We discovered a vast diversity of microbial taxa, with taxa involved in carbon- and sulfur-cycling. Sessile communities seem to be centered on C1-cycling with fungi and heterotrophs likely adapted to heavy-metal stress. Planktonic communities were dominated by ultra-small archaeal and bacterial taxa, highlighting harsh conditions in the mine waters. Microbial source tracking indicated sources of communities from surface to deeper layers and vice versa, suggesting the dispersion of organisms in the mine, although water connectivity remains unknown. Research in the field of the evaporation of liquid droplets placed on surfaces with special wetting properties such as biphilic surfaces is of great importance. This paper presents the results of an experimental study of the heat and mass transfer of a water droplet during its evaporation depending on the direction of the gravitational force. A special technique was developed to create unique substrates, which were used to physically simulate the interaction of liquid droplets with the surface at any angle of inclination to the horizontal. It was found that the suspended and sessile droplets exhibited fundamentally different evaporation dynamics. It was shown that the suspended droplets had a higher temperature and, at the same time, evaporated almost 30% faster. [ABSTRACT FROM AUTHOR]

Aims: Legionella pneumophila (Lp), a human pathogen, has been detected in windscreen wiper fluid reservoirs (WWFRs) where commercial screen washes (CSWs) are commonly added. Limited information is available on CSWs against planktonic Lp; however, responses of sessile Lp and planktonic Lp pre‐acclimated in nutrient‐limited water to CSWs remain unknown. This study thus investigates the antibacterial effects of CSWs on sessile and starved planktonic Lp, in comparison with unstarved Lp. Methods and Results: Lp biofilms were produced on glass and WWFR materials of high‐density polyethylene (HDPE) and polypropylene (PP). Planktonic Lp with and without acclimation in tap water were prepared. Log reductions in cell counts averaged 0.4–5.0 for 10 brands of CSWs against sessile Lp and 1.0–3.9 and 0.9–4.9, respectively, against starved and unstarved planktonic Lp for five CSWs. Both biofilm formation and acclimation in tap water enhanced Lp resistance to CSWs. Significantly different log‐reduction values among CSW brands were observed for sessile Lp on HDPE and planktonic Lp regardless of acclimation (p < 0.05). Conclusions: Biofilm formation, starvation acclimation and CSW brand are crucial factors influencing Lp response to CSWs. Aim: Sessile serrated lesions (SSL) have the potential for malignant transformation into colorectal carcinoma (CRC), which develops via the serrated pathway of carcinogenesis. Materials and Methods: Based on colonoscopy and pathology reports, all sessile serrated lesions or polyps in the risk screening group were studied at an urban medical research center between July 2018 and July 2022. We prospectively included consecutive colorectal polyps classified as hyperplastic polyp (HP), sessile serrated lesion (SSL), traditional serrated adenoma (TSA), unclassified serrated adenoma, and synchronous neoplasm or polyp. Results: During the 48-month study period, 167 cases of sessile serrated lesions or polyps were diagnosed. In the study, 97 (58.1%) of the patients were male, 70 were female (41.9%), and the mean age was 59.74 (± 11.1). Hematoxylin-eosin preparations were available for complete analysis for 226 SSL, and 117 had synchronous adenomatous polyps or carcinomas. Approximately 88.1% of all polyps were =10 mm or less in diameter. There was a significant correlation between polyp size (=10 mm) and epithelial polyps (p=0.000). In particular, polyps detected in the sigmoid colon with the rectum were relatively larger than in other locations. The sigmoid colon and rectum were the most common locations for sessile serrated lesions in our study. There was a significant association between epithelial polyps and anatomical locations (p=0.000). The distribution of all epithelial polyps was as follows; HP was the most frequently observed epithelial polyp with 57.7% (194 polyps), SSL was 6.8% (23 polyps), and TSA was 2.1% (7 polyps), unclassified serrated adenoma was 0.6% (2 cases), synchronous conventional was seen in 31% (104 polyps) of SSL or polyps, and carcinoma with polyps was detected in three (0.9%) cases. Conclusion: Hyperplastic polyps are the most common sessile serrated lesions, and synchronous conventional tubular adenomas were most commonly accompanied by hyperplastic polyps. HPs were most commonly observed in the distal colon, particularly in the sigmoid colon and rectum. Future prospective studies using uniform diagnostic criteria and considering clinical outcomes are needed to make accurate decisions regarding surveillance and treatment of patients with serrated lesions. Abstract Background The mural thrombus in the ascending aorta is rare, most of which are associated with aneurysm or atherosclerotic lesions, with high risks of causing catastrophic thrombotic events. A mural thrombus in the non-aneurysmal and non-atherosclerotic ascending aorta is exceptionally uncommon. Case presentation We reported a large mural thrombus in normal ascending aorta of an asymptomatic patient. Preoperative imaging confirmed the presence of the sessile thrombus located at the left anterior wall of ascending aorta. Given that it had the potential to cause fatal thrombotic complications, surgical removal and segment of ascending aorta replacement were executed. The patient had an uneventful recovery and discharged 14 days after surgery. Conclusions Anticoagulant is the therapeutic cornerstone of ascending aortic thrombus, but surgery should be performed aggressively when the thrombus is large or floating to avoid severe embolic complications or recurrence.

A self-suppression mechanism of biofilm development in the cyanobacterium Synechococcus elongatus PCC 7942 was recently reported. These studies required quantification of biofilms formed by mutants impaired in the biofilm-inhibitory process. Here we describe in detail the use of chlorophyll measurements as a proxy for biomass accumulation in sessile and planktonic cells of biofilm-forming strains. These measurements allow quantification of the total biomass as estimated by chlorophyll level and representation of the extent of biofilm formation by depicting the relative fraction of chlorophyll in planktonic cells.

Objectives: Sessile serrated adenomas with dysplasia (SSADs) of the colon are transitional lesions between sessile serrated adenomas (SSAs) and a subset of colorectal adenocarcinomas. We wished to gain insight into the relative percentages and significance of SSAD subtypes.Methods: Retrospective (2007-2012) clinicopathologic review of colorectal polyps initially regarded as having mixed serrated and dysplastic elements. SSADs were subdivided into those with cap-like adenomatous dysplasia (ad1), non-cap-like adenomatous dysplasia (ad2), serrated dysplasia (ser), minimal dysplasia (min), and dysplasia not otherwise specified (nos). MLH1 immunostaining was performed on many.Results: SSADser (7.7%) had a greater propensity for right colon, women, and MLH1 loss vs the entire cohort. SSAad1 (11.6%) had the least female preponderance, was least likely to have MLH1 loss, and was most likely to affect the left colorectum. SSAD with MLH1 loss was associated with an increased burden of SSAs in the background colon (P = .0003) but not tubular adenomas or hyperplastic polyps. Most SSADs (ad2 and nos groups, 80% combined) showed difficult-to-classify dysplasia, intermediate MLH1 loss rates, and intermediate clinical features.Conclusions: While some trends exist, morphologically subclassifying SSADs is probably not justified in routine clinical practice. Growth on solid media as sessile cells is believed to increase the desiccation tolerance of Salmonella enterica. However, the reasons behind increased resistance have not been well explored. In addition, the same effect has not been examined for other foodborne pathogens such as pathogenic Escherichia coli or Listeria monocytogenes. The purpose of this research was twofold: first, to determine the role of oxygenation during growth on the desiccation resistance of S. enterica, E. coli, and L. monocytogenes, and second, to determine the effect of sessile versus planktonic growth on the desiccation resistance of these pathogens. Three different serotypes each of Salmonella, E. coli, and L. monocytogenes were cultured in Trypticase soy broth with 0.6% yeast extract (TSBYE), with (aerobic) shaking or on TSBYE with agar under either aerobic or anaerobic conditions and harvested in the stationary phase. After adding cell suspensions to cellulose filter disks, pathogen survival was determined by enumeration before drying (0 h) and after drying for 24 h. Results showed statistical differences in harvested initial populations before drying. For Salmonella, a correlation was found between high initial population and greater survival on desiccation (P = 0.05). In addition, statistical differences (P 0.05) between survival based on growth type were identified. However, differences found were not the same for the three pathogens, or between their serotypes. In general, Salmonella and E. coli desiccation resistance followed the pattern of aerobic agar media liquid media anaerobic agar media. For L. monocytogenes serotypes, resistance to desiccation was not statistically different based on mode of growth. These results indicate growth on solid media under aerobic conditions is not always necessary for optimal desiccation survival, but may be beneficial when the desiccation resistance of the test serotype is unknown. EPSs also prevent penetration of antimicrobials/antibiotics into the sessile groups of cells. Hence, methods and agents to avoid or remove biofilms are urgently needed. Enzymes play important roles in the removal of biofilm in natural environments and may be promising agents for this purpose. As the major component of the EPS is polysaccharide, amylase has inhibited EPS by preventing the adherence of the microbial cells, thus making amylase a suitable antimicrobial agent. On the other hand, salivary amylase binds to amylase-binding protein of plaque-forming Streptococci and initiates the formation of biofilm. This review investigates the contradictory actions and microbe-associated genes of amylases, with emphasis on their structural and functional characteristics.

PurposeTo assess the association of tumor architecture with cancer recurrence, metastasis, and cancer-specific survival (CSS) in patients treated with radical nephroureterectomy (RNU) for upper urinary tract urothelial carcinoma (UTUC) in Taiwan.Materials and MethodsData were collected from 857 patients treated with RNU between January 2005 and August 2016 in our hospital. Pathologic slides were reviewed by genitourinary pathologists. Propensity score weighting was performed for data analysis.ResultsSessile growth pattern was observed in 212 patients (24.7%). Tumor architecture exhibited a significant association with bladder cancer history, chronic kidney disease (CKD), tumor stage, lymph node status, histological grade, lymphovascular invasion, concomitant carcinoma in situ, and the variant type [standardized mean difference (SMD) > 0.1 for all variables before weighting]. In the propensity score analysis, 424 papillary and sessile tumor architecture were analyzed to balance the baseline characteristics between the groups. Tumor architecture was an independent predictor of metastatic disease and CSS (p = 0.033 and p = 0.002, respectively). However, the associations of tumor architecture with bladder and contralateral recurrence were nonsignificant (p = 0.956 and p = 0.844, respectively).ConclusionsTumor architecture of UTUC after RNU is associated with established features of aggressive disease and predictors of metastasis and CSS. Assessment of tumor architecture may help identify patients who could benefit from close follow-up or early administration of systemic therapy after RNU. Tumor architecture should be included in UTUC staging after further confirmation.

Population-level scaling in ecological systems arises from individual growth and death with competitive constraints. We build on a minimal dynamical model of metabolic growth where the tension between individual growth and mortality determines population size distribution. We then separately include resource competition based on shared capture area. By varying rates of growth, death, and competitive attrition, we connect regular and random spatial patterns across sessile organisms from forests to ants, termites, and fairy circles. Then, we consider transient temporal dynamics in the context of asymmetric competition, such as canopy shading or large colony dominance, whose effects primarily weaken the smaller of two competitors. When such competition couples slow timescales of growth to fast competitive death, it generates population shocks and demographic oscillations similar to those observed in forest data. Our minimal quantitative theory unifies spatiotemporal patterns across sessile organisms through local competition mediated by the laws of metabolic growth, which in turn, are the result of long-term evolutionary dynamics. Suspension feeders are sessile marine organisms dependent on plankton and organic particles transported by the water column for food. This study focused on the distribution of four suspension feeders (Dendropoma maxima, Spirobranchus giganteus, Heteractis magnifica, and Tridacna maxima) in Mo’orea, French Polynesia based on current, suspended particle abundance, and substrate type. Both D. maxima and S. giganteus were more frequently present at the site with highest current flow and suspended particle abundance. The positive correlation between current and suspended particle, or food, abundance suggests that a stronger current transports more organic particles to a set location than a weaker current. In comparison, H. magnifica and T. maxima abundance and occurrence were not related to current. This is because they are less reliant than D. maxima and S. giganteus on the current for suspension feeding. In comparison, all four species studied showed substrate preference. The distribution of the suspension feeders studied showed varying dependence on current and substrate type. Since suspension feeders regulate primary production by consuming plankton, their distribution and abundance greatly affects marine food chains. Understanding coral reef food chains is vital for comprehending the niche suspension feeders’ fill in coral reef ecosystems.

First severe attacks produced by Ceresa bubalus Fabricius, 1794 (Hemiptera: Membracidae) in Romanian forests occurred in autumn 2017 in young sessile oak plantations, in north-eastern Romania (Suceava county), managed by Greengold Est SRL Private Forest District. The attacks (longitudinal incisions on stems and lateral branches) were recorded on a total area of 92.5 ha (66.5 ha in PU I Amelina and 26.0 ha in PU XII Iași South). Heavy infestations occurred on 37.8 ha, the density of incisions was of 58 pieces/dm on stem and 21 pieces/dm on branches, with 3–7 eggs/incision. In order to prevent new attacks on seedlings, winter spraying was done using Confidor Oil SC 004 (dose 7.5 l/ha). No subsequent infestations were recorded after spraying. Gnesiotrochan rotifers display a variety of life styles ranging from taxa with free-swimming larval and sessile adult stages to those with motile adult stages and colonial habits. Several explanations for the C-value enigma posit that genome size is correlated with lifestyle. To investigate this, 13 gnesiotrochan species representing nine genera were measured by flow cytometry. Genome sizes (1C) within Gnesiotrocha ranged from 0.05 pg (Hexarthra mira and Hexarthra fennica) to 0.25 pg (Sinantherina ariprepes). Genome sizes varied within genera and species; e.g., the H. fennica (El Huérfano, Mexico) genome was estimated to be 15% larger than that of H. mira and H. fennica (Keystone Wetland, TX, USA). Gnesiotrochan genome sizes are similar to those reported within Ploima, which range from 0.06 pg (Brachionus rotundiformis, B. dimidiatus) to 0.46 pg (B. asplanchnoidis). Within Gnesiotrocha, genome size was found to be significantly smaller in sessile versus motile species as well as in solitary versus colonial species. To account for phylogenetic background, linear mixed models with hierarchical taxonomic ranks showed that there is a taxonomic component underlying genome size. Rotifers are diverse and abundant aquatic micrometazoans that rely on their ciliated apical end (corona) for locomotion and feeding. In order Collothecaceae, which includes mostly sessile species, larval rotifers go through a complex metamorphosis after settlement wherein they replace their corona with an unusual cup-shaped head that functions exclusively in food capture. This new head, called the infundibulum, consists of morphological elaborations such as lobes or tentacles that develop precociously in the larval stage; in adults they function in ambush and sit-and-wait predation. Here, we provide evidence from brightfield and transmission electron microscopy that the infundibulum of collothecid rotifers is derived from the larval foregut and not the larval corona, suggesting that the adult head of collothecids is a morphological novelty and therefore not homologous with the rotifer corona as classically defined. The wide variety of morphologies of the infundibulum suggests that selection to maximize foraging may have driven the evolution of head form in these sessile species, and that future studies of collothecid rotifers should consider the infundibulum as a unique form of food-collection device that is evolutionarily distinct from the rotifer corona. Small birefringent concretions have been reported in rotifers for over a century and often hypothesized as energy sources. Here, we provide an update on their distribution in superorder Gnesiotrocha and the first data on their ultrastructure. Within Gnesiotrocha, these birefringent bodies (BRB) are known from at least ten species of Collothecaceae and 14 species of Flosculariaceae, both of which include planktonic and sessile species. Among sessile species, the predator Acyclus inquietus contains a single BRB that has been described as starch-like. We examined larvae of A. inquietus with transmission electron microscopy and revealed the BRB to have an irregular, electron-dense margin that surrounds a speckled core. The core appears mostly amorphous, but contains numerous, very small electron-dense spots and thin electron-dense fibers; there is no evidence of any crystalline lattice. The intestinal lumen contains smaller concretions that are probably the result of BRB metabolism. The thin epithelium contains abundant electron-dense granules but relatively few organelles. We hypothesize that the BRB is a unique form of extracellular glycogen that functions as an energy source in larvae for their dispersal and metamorphosis. Prosthetic joint replacement failure has a huge impact on quality of life and hospitalization costs. A leading cause of prosthetic joint infection is bacteria-forming biofilm on the surface of orthopedic devices. Staphylococcus epidermidis is an emergent, low-virulence pathogen implicated in chronic infections, barely indistinguishable from aseptic loosening when embedded in a mature matrix. The literature on the behavior of quiescent S. epidermidis in mature biofilms is scarce. To fill this gap, we performed comparative analysis of the whole proteomic profiles of two methicillin-resistant S. epidermidis strains growing in planktonic and in sessile form to investigate the molecular mechanisms underlying biofilm stability. After 72-h culture of biofilm-forming S. epidermidis, overexpression of proteins involved in the synthesis of nucleoside triphosphate and polysaccharides was observed, whereas planktonic bacteria expressed proteins linked to stress and anaerobic growth. Cytological analysis was performed to determine why planktonic bacteria unexpectedly expressed proteins typical of sessile culture. Images evidenced that prolonged culture under vigorous agitation can create a stressful growing environment that triggers microorganism aggregation in a biofilm-like matrix as a mechanism to survive harsh conditions. The choice of a unique late time point provided an important clue for future investigations into the biofilm-like behavior of planktonic cells. Our preliminary results may inform comparative proteomic strategies in the study of mature bacterial biofilm. Finally, there is an increasing number of studies on the aggregation of free-floating bacteria embedded in an extracellular matrix, prompting the need to gain further insight into this mode of bacterial growth.

Rotifers are aquatic microinvertebrates that live in the plankton or in the benthos, which may include a variety of macrophytes. Among these periphytic forms of rotifers, some have taken up a sessile existence and secrete protective tubes around their bodies. One type of tube common to species of Floscularia is made of small round pellets. To date, the building process and some fine structural details are known for Floscularia ringens, but many questions about the composition of the tube and its ultrastructure still remain unanswered. Here, we use transmission electron microscopy and scanning electron microscopy–energy dispersive X‐ray spectroscopy (SEM–EDS) to study the ultrastructure of the pellets and their elemental composition, respectively, in the putative sister species, Floscularia conifera. We revealed several new details that add important information about the physiology of tube‐making in species of Floscularia. First, we note an inner secretory membrane that is thin, electron lucent, and supports the external pellets. The pellets are relatively consistent in size and have a small depression on their inner surface. All pellets are individually wrapped in a secretory membrane that completely encapsulates suspended materials collected from the surrounding water. Elemental signatures of pellets reveal they consist mostly of carbon (C), nitrogen (N), and oxygen (O), with some silicon (Si) content that is likely the result of diatom shells. Other trace elements such as iron (Fe) and sodium (Na) are also present and likely the result of incorporated bacteria and suspended materials. When larval rotifers are cultured in filtered pond water, the pellets consist mostly of C and O, with little N and no Si; Fe is present in smaller amounts. These new discoveries provide a better understanding of the physiology of rotifer tube construction and tube composition, and their future utility in understanding if and how changes in freshwater environments might impact these factors. Analytical microbiology has made substantial progress since its conception, starting from potato slices, through selective agar media, to engineered surfaces modified with capture probes. While the latter represents the dominant approach in designing sensors for bacteria detection, the importance of sensor surface properties is frequently ignored. Herein, we highlight their significant role in the complex process of bacterial transition from planktonic to sessile, representing the first and critical step in bacteria detection. We present the main surface features and discuss their effect on the bio-solid interface and the resulting sensing capabilities for both flat and particulate systems. The concepts of rationally-designed surfaces for enhanced bacterial detection are presented with recent examples of sensors (capture probe-free) relying solely on surface cues. Ticks transmit infectious agents including bacteria, viruses and protozoa. However, their transmission may be compromised by host resistance to repeated tick feeding. Increasing host resistance to repeated tick bites is well known in laboratory animals, including intense inflammation at the bite sites. However, it is not known whether this also occurs in wild rodents such as white-footed mice, Peromyscus leucopus, and other wildlife, or if it occurs at all. According to the “host immune incompetence” hypothesis, if these mice do not have a strong inflammatory response, they would not reject repeated tick bites by Ixodes scapularis. To test this hypothesis, histopathological studies were done comparing dermal inflammation in P. leucopus versus guinea pigs, Cavia porcellus, repeatedly infested with I. scapularis. In P. leucopus, the immune cell composition was like that seen in laboratory mouse models, with some differences. However, there was a broad sessile lesion with intact dermal architecture, likely enabling the ticks to continue feeding unimpeded. In contrast, in C. porcellus, there was a relatively similar mixed cellular profile, but there also was a large, leukocyte-filled cavitary lesion and scab-like hyperkeratotic changes to the epidermal layer, along with itching and apparent pain. Ticks attached to sensitized C. porcellus fed poorly or were dislodged, presumably due to the weakened anchoring of the tick’s mouthparts cemented in the heavily inflamed and disintegrating dermal tissues. This is the first time that the architecture of the skin lesions has been recognized as a major factor in understanding tick–host tolerance versus tick bite rejection. These findings broadly strengthen previous work done on lab animal models but also help explain why I. scapularis can repeatedly parasitize white-footed mice, supporting the “immune evasion theory” but cannot repeatedly parasitize other, non-permissive hosts such as guinea pigs.

Eater is an EGF-like repeat transmembrane receptor of the Nimrod family and is expressed in Drosophila hemocytes. Eater was initially identified for its role in phagocytosis of both Gram-positive and Gram-negative bacteria. We have deleted eater and show that it appears to be required for efficient phagocytosis of Gram-positive but not Gram-negative bacteria. However, the most striking phenotype of eater deficient larvae is the near absence of sessile hemocytes, both plasmatocyte and crystal cell types. The eater deletion is the first loss of function mutation identified that causes absence of the sessile hemocyte state. Our study shows that Eater is required cell-autonomously in plasmatocytes for sessility. However, the presence of crystal cells in the sessile compartment requires Eater in plasmatocytes. We also show that eater deficient hemocytes exhibit a cell adhesion defect. Collectively, our data uncovers a new requirement of Eater in enabling hemocyte attachment at the sessile compartment and points to a possible role of Nimrod family members in hemocyte adhesion.

Infestation of the White Mango Scale (WMS) ( Aulacaspis tubercularis Newstead (Hemiptera: Diaspididae) on mango was known in Kenya for a longer time, yet the severity of its impact has only been reported frequently in Ethiopia where the first record of the pest was only about a decade. This study investigated the WMS statuses in Kenya and Ethiopia. A total of four fields, two from each country, were addressed to determine the population abundances of the pest at different developmental stages. Five blocks, one at the center and the rest at each corner of the study orchards were set in Ethiopia. In Kenya, Randomized Complete Block Design (RCBD) was employed to determine the levels of susceptibility of different mango varieties to the pest. The abundance of WMS was significantly different (p < 0.05) between Arjo-Gudetu and Bako-Tibe sites in Ethiopia. Sessile mango scales in hundreds and eggs and crawlers each in thousands per leaf were recorded in Ethiopia. In contrast, very low WMS populations were recorded from the Kenyan orchards, with most leaves bearing no mango scale. However, the sessile stage populations observed were significantly different (p < 0.05) between Kimani-Sofia and Samuru-Kandara orchards. In each of these orchards, the levels of pest infestation varied significantly (p < 0.05) among the mango varieties, Kent and Vandyke being the most infested at Kimani-Sofia and Samuru-Kandara, respectively. In conclusion, these studies revealed the presence of a very high population abundance of mango scale in Ethiopia, whereas in Kenya it was very low. This variation may pertain mainly to the differences in mango production and pest management practices between the two countries. Clostridium acetobutylicum has traditionally been used for production of acetone, butanol, and ethanol (ABE). Butanol is a commodity chemical due in part to its suitability as a biofuel; however, the current yield of this product from biological systems is not economically feasible as an alternative fuel source. Understanding solvent phase physiology, solvent tolerance, and their genetic underpinning is key for future strain optimization of the bacterium. This study shows the importance of a [NiFe]-hydrogenase in solvent phase physiology. C. acetobutylicum genes ca_c0810 and ca_c0811, annotated as a HypF and HypD maturation factor, were found to be required for [NiFe]-hydrogenase activity. They were shown to be part of a polycistronic operon with other hyp genes. Hydrogenase activity assays of the DhypF/hypD mutant showed an almost complete inactivation of the [NiFe]-hydrogenase. Metabolic studies comparing DhypF/hypD and wild type (WT) strains in planktonic and sessile conditions indicated the hydrogenase was important for solvent phase metabolism. For the mutant, reabsorption of acetate and butyrate was inhibited during solventogenesis in planktonic cultures, and less ABE was produced. During sessile growth, the DhypF/hypD mutant had higher initial acetone: butanol ratios, which is consistent with the inability to obtain reduced cofactors viaH2 uptake. In sessile conditions, the DhypF/hypD mutant was inhibited in early solventogenesis, but it appeared to remodel its metabolism and produced mainly butanol in late solventogenesis without the uptake of acids. Energy filtered transmission electron microscopy (EFTEM) mapped Pd(II) reduction via [NiFe]-hydrogenase induced H2 oxidation at the extracelluar side of the membrane on WT cells. A decrease of Pd(0) deposits on DhypF/hypD comparatively to WT indicates that the [NiFe]-hydrogenase contributed to the Pd(II) reduction. Calculations of reaction potentials during acidogenesis and solventogenesis predict the [NiFe]-hydrogenase can couple NAD+ reduction with membrane transport of electrons. Extracellular oxidation of H2 combined with the potential for electron transport across the membrane indicate that the [NiFe}-hydrogenase contributes to proton motive force maintenance via hydrogen cycling. Rotifers are common aquatic microscopic invertebrates. Most rotifers are planktonic but several gnesiotrochan species are sessile and produce tubular sheaths around their bodies. These tubes have a variable morphology and may be produced by different glands. Here, we applied scanning electron microscopy (SEM) and transmission electron microscopy (TEM) to study the ultrastructure of the tube and its potential origin of secretion in Limnias melicerta. Results from SEM confirm earlier observations that juvenile rotifers first secrete a segment-less tube and then add segments as they grow. Tubes consist of two distinct secretions: an inner mucus-like layer that extends from the base to the foot region of the adult, and an external layer that is secreted by “cement cells” sensu Wright. The external layer consists of a series of thickened rings and elongated girdles, both of which are somewhat fibrous in appearance and occasionally show differences in electron density. The ultrastructure of the “cement cells” indicates that these secretory regions are not cellular but rather a modified region of the syncytial integument that forms a belt-like gland around the animal. This gland is highly papillated due to localized folding of the intracytoplasmic lamina of the integument. The ultrastructure of the gland shows a voluminous swelling of the syncytium with abundant endoplasmic reticulum and secretion vesicles. At least three types of membrane-bound secretion vesicles are present based on electron density. Mesenteric cysts are uncommon intra-abdominal benign masses that appear in childhood with varying degrees of clinical manifestations, ranging from being asymptomatic to presenting as an acute abdomen. The diagnosis is made incidentally during the work-up for other abdominal pathologies such as acute appendicitis, bowel obstruction, etc. The treatment is mostly surgical and varies depending on the clinical type of the lesion. A 26-month-old child was referred to our teaching hospital's pediatric surgery department with an abdominal mass. The patient had previously complained of constipation and been treated symptomatically with laxatives. Ultrasonography reported ovarian cysts confirmed by computerized tomography scanning (as a misdiagnosis report of an ovarian cyst instead of a mesenteric cyst), done outside the hospital in a private diagnostic center. The patient was prepared for operation and during the surgical procedure, she was found to have a duplex cyst, which was mostly incorporated in the mesentery of the distal 1/3 of the transverse mesocolon. The cyst was enucleated through a lower midline laparotomy incision without bowel resection and anastomosis. The histopathological analysis of the specimen confirmed a mesenteric cyst. Mesenteric cysts are rare lesions in children and should be considered when approaching any intra-abdominal mass. Except for the rare cases where intestinal resection and/or partial cyst excision are required, all mesenteric cysts can be excised while preserving intestinal integrity and vascular supply. • Mesenteric cyst is a rare intra-abdominal condition in children. • The common location is the small bowel mesentery, but it may occur anywhere from the duodenum to the rectum. • The condition is asymptomatic, but sometimes it is accompanied by gastrointestinal signs and symptoms, therefore, mesenteric cysts should always be considered in the differential diagnosis of acute abdomen in children. • Complete enucleation of the cyst with or without bowel resection and anastomosis is the treatment of choice. Background: Pressure therapy is one of many remedies that have been used for the management of keloid, which lesions have been classified as sessile or pedunculated. This study aims to investigate the response of sessile and pedunculated keloid lesions to pressure therapy applied using tight-fitting garments worn daily by African women. Design: Prospective, non-invasive intervention study. Methods: Pressure therapy was applied to lesions on the back using elastic brassiere straps; tight-fitting cycling shorts for lower abdominal lesions; and auricular lesions were managed using head gears which pressed them against the skull. Results: The study involved 18 keloid lesions in 14 women. The mean percentage reduction in volume among pedunculated lesions was 66.8; 100% was achieved among all sessile lesions. Conclusion: All sessile keloid lesions were found to flatten completely over time on regular application of pressure. No pedunculated lesion was found to flatten completely following the same therapy. Extirpative techniques are required to completely flatten pedunculated lesions. [ABSTRACT FROM AUTHOR]

Ecosystem engineers can influence biodiversity by enhancing complexity, and modifying the availability of resources. Understanding the mechanisms by which ecosystem engineers shape biodiversity is central to the concept of ‘ecological engineering’ of anthropogenic structures to enhance biodiversity. Here the presence and complexity of artificial turf was manipulated on an artificial structure to test the hypothesis that the colonisation of sessile invertebrates and mobile epibiota would vary with habitat complexity. Both sessile and mobile assemblage compositions differed according to the presence of artificial turf, and its complexity. Sessile invertebrates occupied greater proportions of available space on topographically simple ‘blank’ surfaces or low complexity artificial turf than those with high complexity turf, whereas mobile taxa were generally more abundant on the turf. However, the mobile assemblage was unrelated to the sessile assemblage when examined within each level of initial substrata complexity. Contrary to the increasing number of studies demonstrating nested hierarchical relationships between co-occurring ecosystem engineers, this study provides an example of an ecosystem engineer mimic (artificial turf) leading to the formation of habitat mosaics at small scales. The introduction of complex substrata to otherwise topographically simple artificial structures is a promising means of actively influencing assemblage composition. [ABSTRACT FROM AUTHOR]

Members of the sessile rotifer species Cupelopagis vorax are unusual ambush predators that live permanently attached to submerged freshwater plants. Previous light microscopical research has revealed several uncommon features in this species including a stellate-patterned integument and an expansive foregut region called the proventriculus. In this study, we apply transmission electron microscopy to explore the ultrastructure of both the integument and foregut to determine how they differ from other rotifers. Our results reveal that the integument is covered by a thick glycocalyx and is patterned with tubercles that originate from the intracytoplasmic lamina ( ICL) within the syncytial epidermis. The ICL forms an apical layer within the syncytium, is electron dense and mostly amorphous, and forms tubercles up to 2.3 μm; these tubercles probably account for the patterned appearance of the integument and are similar to what has been found in other gnesiotrochan rotifers. The basal cytoplasm is highly granular and contains two types of membrane-bound vesicles: large ovoid vesicles (320-411 nm) with amorphous, opaque contents, and secretory bulbs (110-264 nm) with electron-lucent cores and occasionally electron-dense contents. Only the secretory bulbs were observed to form connections to the apical plasmalemma, and so are probably exocytotic. Internally, the proventriculus is a large distensible sack that connects the anterior pharyngeal tube to the posterior mastax. The proventricular epithelium is a thin syncytium mostly covered with a dense glycocalyx and a strong brush border of microvilli underlain by a thin terminal web. The cytoplasm contains few organelles and there is no evidence that it is either secretory or has features (e.g., ICL) that might aid in maceration. We hypothesize that the thick glycocalyx might serve a protective function against the movements of live prey and/or against enzymes released from the rotifer's gastric glands that become regurgitated during feeding. Many studies have described the response of the facultative anaerobe, Escherichia coli, to anaerobic conditions, yet they all investigated free-living (planktonic) cells because attempts to cultivate anaerobic E. coli biofilm were mostly unsuccessful. We challenged these findings and cultivated E. coli strain MG1655 biofilm under both aerobic and anaerobic conditions, characterizing the mature biofilm architecture and global gene expression profile. We used RNA sequencing technology to compare stationary phase planktonic cells with mature biofilm, cultured with and without oxygen. Our results suggest that gene expression patterns significantly differ between biofilm and planktonic cultures cultivated under the same oxygenic conditions. The anaerobic E. coli biofilms were slow growing and patchy compared to aerobic biofilms, yet some features were unchanged like the production of extracellular polymeric substances. A closer inspection of the mRNA data revealed that essential cell processes were attenuated in anaerobic biofilms, including protein synthesis, information transfer, cell structure, regulation and transport. Our results suggest that lack of oxygen imposes severe stress on mature biofilms thus limiting the cells’ activity. We further propose that E. coli does not favor growing in anaerobic biofilms and when forced to do so, the cells prevail by attenuating their activity in order to survive. [ABSTRACT FROM AUTHOR]

Background/aims: A full colonoscopy is currently required in children and adolescents with colorectal polyps, because of their potential of neoplastic transformation and complications such as intussusception. We aimed to analyze the associations of polyp characteristics in children and adolescents with colorectal polyps. Based on these findings, we also aimed to reevaluate the necessity of conducting a full colonoscopy., Methods: Pediatric patients <18 years of age who had undergone a colonoscopic polypectomy and those with <5 colorectal polyps were included in this multicenter, retrospective study. Baseline clinicodemographics, colonoscopic and histologic findings were investigated., Results: A total of 91 patients were included. Multivariate logistic regression analysis showed that polyp size was the only factor associated with the presence of any polyps located proximal to the splenic flexure (odds ratio [OR], 2.25; 95% confidence interval [CI], 1.28 to 4.28; p=0.007). Furthermore, polyp location proximal to the splenic flexure and sessile morphology were associated with the presence of any adenomatous polyp (OR, 8.51; 95% CI, 1.43 to 68.65; p=0.023; OR, 18.41; 95% CI, 3.45 to 173.81; p=0.002, respectively)., Conclusions: In children and adolescents presenting with <5 colorectal polyps, polyp size and the presence of any adenomatous polyp were positively associated with polyp location proximal to the splenic flexure. This finding supports the necessity of a full colonoscopic exam in pediatric patients with colorectal polyps for the detection of polyps before the occurrence of complications such as intussusception or neoplastic transformation.

Polymer stabilized emulsions are commonplace in industries ranging from cosmetics and foods to pharmaceuticals. Understanding the physical properties of emulsions is of critical importance to the rapid advancement of industrial applications. In this work, we use a sessile drop geometry to examine the effects of viscosity changes of the surrounding glycerine/water solution on polystyrene-b-polyethylene oxide (PS-PEO) covered toluene droplets. In the experiment, emulsion drops are driven by the buoyant force into a smooth mica surface. The drops buckle as they approach the mica, trapping some of the outer fluid which slowly drains out over time. The characteristic time of the drainage process as well as the surface tension was measured as a function of glycerine/water concentration. The surface tension is found to have a minimum at a glycerine concentration of approximately 50% (by weight to water) and the drainage rate is shown to be well described by a recent model. The simple experiment not only shows how critical features of emulsion stability can be easily and reliably measured, but also identifies important new features of the drainage process.
