Tetrodotoxin (TTX), which is found in various marine organisms, including pufferfish, shellfish, shrimp, crab, marine gastropods, and gobies, is an effective marine toxin and the cause of many seafood poisoning incidents. Owing to its toxicity and threat to public health, the development of simple, rapid, and efficient analytical methods to detect TTX in various food matrices has garnered increasing interest worldwide. Herein, we reviewed the structure and properties, origin and sources, toxicity and poisoning, and relevant legislative measures of TTX. Additionally, we have mainly reviewed the state-of-the-art progress of analytical methods for TTX detection in the past five years, such as bioassays, immunoassays, instrumental analysis, and biosensors, and summarized their advantages and limitations. Furthermore, this review provides an in-depth discussion of the most advanced biosensors, including cell-based biosensors, immunosensors, and aptasensors. Harmful algal bloom events occur in salt, brackish, and fresh water. In bodies of water such as oceans and estuaries, diatoms or dinoflagellates form "tides" that produce toxins associated with seafood poisoning, including paralytic shellfish poisoning, or respiratory distress from inhalation of aerosolized toxins. Cyanobacteria predominantly bloom in fresh water; they can produce microcystins; cylindrospermopsin; and other toxins that humans or animals might be exposed to through water contact, inhalation, or ingestion. Animals that become ill or die can be sentinels for harmful algal bloom events. In a One Health approach, information about harmful algal bloom exposures and health effects support efforts to detect these events and mitigate and prevent associated illnesses. Cleistanthus collinus is a toxic plant belonging to the Euphorbiaceae family, and all parts of the plant are poisonous. It is used as suicidal, homicidal, cattle and fish poison and for inducing criminal abortion. The leaves and plant parts are consumed in various forms for homicidal and suicidal and it results in symptoms such as chest pain, dyspnea, central nervous system symptoms, and vision disturbances. There is no specific antidote for the C. collinus poisoning. The percentage of mortality with C. collinus poisoning varies from 20% to 50%, and the mortality depends on the amount and mode of consumption of the leaves. The major phytoconstituents of the plant include ellagic acid, diphyllin, collinusin, cleistanthin A, and cleistanthin B. The two major compounds cleistanthin A and cleistanthin B were evaluated for pharmacological activities such as antineoplastic activity, antiviral activity, diuretic activity, neuromuscular blocking property, alpha receptor blocking property, and antioxidant effect. Dinophysistoxin 1 (DTX1, 1) and okadaic acid (OA, 2) , produced by the dinoflagellates Dinophysis spp. and Prorocentrum spp. are primary diarrhetic shellfish toxins (DSTs), which may cause gastric illness in people consuming such as bivalves. Both compounds convert to dinophysistoxin 3 (DTX3, 3 ; generic name for 1 and 2 with fatty acids conjugated at 7-OH) in bivalves. The enzyme okadaic acid O -acyl transferase (OOAT) is a membrane protein found in the microsomes of the digestive glands of bivalves. In this study, we established an in vitro enzymatic conversion reaction using 4-nitro-2,1,3-benzoxadiazole (NBD)-OA (4), an OA derivative conjugated with (R)-(-)-4-nitro-7-(3-aminopyrrolidin-1-yl)-2,1,3-benzoxadiazole (NBD-APy) on 1-CO2H, as a substrate. We detected the enzymatically produced 3 , NBD-7- O -palmitoyl-OA (NBD-Pal-OA), using high-performance liquid chromatography-fluorescence detection. Rabbit fish (Siganus vermiculatus) is one of the biological resources in the sea that is widely consumed by the people in Maluku. Rabbit fish are caught during low tide with nets or by using natural resources to poison the fish, such as tuba root (Derris elliptica), which contains rotenone to make fish easy to catch. This study aimed to determining the concentration of tuba root extract that was effective against damage to the liver tissue of rabbit fish. Rabbit fish and tuba root were collected from Oma Village, Haruku Island, Central Maluku District, Maluku Province. In the study, a laboratory experiment was conducted in which the tuba root was treated with doses of 0.1 g, 0.3 g, 0.5 g and 1 g with an exposure time of 18 to 24 hours. Data were analyzed using two-way analysis of variance (ANOVA) followed by Duncan's post-hoc test. As a result, the lowest level of liver damage in rabbit fish was observed at a dose of 0.1 g, with a total damage of 7 ± 0.00 and 8.5 ± 0.71. Conversely, the highest level of liver damage was observed at a dose of 1 g, with a total damage of 14 ± 0.00 and 15 ± 0.00. The results of the post hoc test showed a subset value of 0.1 g dose (7.75), 0.3 g dose (9.25), 0.5 g dose (12.00), and 1 g dose (14.50). This research indicates that administering tuba root to rabbit fish can result in damage to the liver's structure. The severity of this damage is contingent upon the dose administered, whereas exposure duration does not affect the level of damage to fish organs. [ABSTRACT FROM AUTHOR]

Ciguatoxins (CTXs) are neurotoxic marine biotoxins capable of contaminating marine organisms. Approximately 30 CTX analogues have been described; however, only a few have been documented in ciguatera poisoning (CP) outbreaks. Detecting CTXs from complex matrices at CP-relevant concentrations (<1 µg per kg seafood tissue) is analytically challenging. Analytical standards for CTXs are rare. Even after multi-step sample preparation (including liquid–liquid partition, defatting, and solid-phase extraction (SPE)), extracts can contain undesirable co-eluting matrix components. These limitations can exacerbate discrepancies between results obtained by LC-MS/MS and the N2a-bioassay, which are two common CTX detection methods. Herein, a rapid and simple fractionation method, based on normal phase SPE (silica), is demonstrated. Target CTXs were eluted using solvent mixtures of ascending polarity, passed through the column, and separated into eight fractions. To challenge the method, extracts with eleven naturally incurred CTX analogues among different structural CTX groups (e.g., CTX3C, CTX4A, and C-CTX group) were used. The most complex tissue matrix tested (viscera) was improved the most for extract purity and CTX detection, enhancing the correlation between LC-MS/MS and N2a-bioassay results. This workflow represents an advancement for characterizing CTXs in seafood products and CP outbreaks, irrespective of the responsible CTX analogue and where standards are lacking. Paralytic shellfish toxins (PSTs) are potent neurotoxins produced by certain microalgae, particularly dinoflagellates, and they can accumulate in shellfish in coastal seawater and thus pose significant health risks to humans. To explore the relationship between toxicity and PST profiles in seawater and mussels, the spatiotemporal variations in PST concentrations and profiles were investigated along the southern coast of Korea under peak PST levels during spring. Seawater and mussel samples were collected biweekly from multiple stations, and the toxin concentrations in the samples were measured. Moreover, the dinoflagellate community composition was analyzed using next-generation sequencing to identify potential PST-producing species. The PST concentrations and toxin profiles showed substantial spatiotemporal variability, with GTX1 and GTX4 representing the dominant toxins in both samples, and C1/2 tending to be higher in seawater. Alexandrium species were identified as the primary sources of PSTs. Environmental factors such as water temperature and salinity influenced PST production. This study demonstrates that variability in the amount and composition of PSTs is due to intricate ecological interactions. Saxitoxin (STX), an exceptionally potent marine toxin for which no antidote is currently available, is produced by methanogens and cyanobacteria. This poses a significant threat to both shellfish aquaculture and human health. Consequently, the development of a rapid, highly sensitive STX detection method is of great significance. The objective of this research is to create a novel approach for identifying STX. Therefore, amplified luminescent proximity homogeneous assay (AlphaLISA) was established using a direct competition method based on the principles of fluorescence resonance energy transfer and antigen–antibody specific binding. This method is sensitive, rapid, performed without washing, easy to operate, and can detect 8–128 ng/mL of STX in only 10 min. The limit of detection achieved by this method is as low as 4.29 ng/mL with coefficients of variation for the intra-batch and inter-batch analyses ranging from 2.61% to 3.63% and from 7.67% to 8.30%, respectively. In conclusion, our study successfully establishes a simple yet sensitive, rapid, and accurate AlphaLISA method for the detection of STX which holds great potential in advancing research on marine biotoxins. Portable X‐Ray Fluorescence (XRF) has become increasingly popular where traditional laboratory methods are either impractical, time consuming, and/or too costly. While the Limit of Detection (LOD) is generally poorer for XRF compared to laboratory‐based methods, recent advances have improved XRF LODs and increased its potential for field‐based studies. Portable XRF can be used to screen food products for toxic elements such as lead (Pb), cadmium (Cd), mercury (Hg), arsenic (As), manganese, (Mn), zinc (Zn), and strontium (Sr). In this study, 23 seafood samples were analyzed using portable XRF in a home setting. After XRF measurements were completed in each home, the same samples were transferred to the laboratory for re‐analysis using microwave‐assisted digestion and Inductively Coupled Plasma Tandem Mass Spectrometry (ICP‐MS/MS). Four elements (Mn, Sr, As, and Zn) were quantifiable by XRF in most samples, and those results were compared to those obtained by ICP‐MS/MS. Agreement was judged reasonable for Mn, Sr, and As, but not for Zn. Discrepancies could be due to (1) the limited time available to prepare field samples for XRF, (2) the heterogeneous nature of "real samples" analyzed by XRF, and (3) the small beam spot size (~1 mm) of the XRF analyzer. Portable XRF is a cost‐effective screening tool for public health investigations involving exposure to toxic metals. It is important for practitioners untrained in XRF spectrometry to (1) recognize the limitations of portable instrumentation, (2) include validation data for each specific analyte(s) measured, and (3) ensure personnel have some training in sample preparation techniques for field‐based XRF analyses. Lactococcus garvieae has recently been identified and listed as one of the causative agents of hyperacute hemorrhagic sepsis in fish. In intensive recirculating aquaculture systems where there are high fish densities and minimal water changes, not only will it be conducive to the growth of bacteria, but Cryptocaryon irritans as a marine protozoan fish parasite is also prone to appear. This study reports the disease status of Trachinotus ovatus in an aquaculture area in Yangjiang City, Guangdong Province. Through the diagnosis of clinical symptoms of the diseased fish, identification of specific primers, 16s rRNA sequences phylogenetic tree analysis, physiological and biochemical identification, and observation of histopathological sections, the result of the experiment is that the mass death of T. ovatus is caused by a mixture of L. garvieae and C. irritants infections. Subsequently, regression infection experiments were performed to verify Koch's law. It was confirmed that the pathogen had strong virulence to T. ovatus. This is the first time that the co-infection of L. garvieae and C. irritans to T. ovatus was found in South China. The research results of this experiment have certain enlightenment significance for the epidemic trend of fish diseases in relevant sea areas. [ABSTRACT FROM AUTHOR]

Consuming poisoned shellfish can lead to severe health problems and even death. Alexandrium catenella (Group I) and A. pacificum (Group IV) cause paralytic shellfish poisoning (PSP) in Korea, and PSP is detected more in a wider area. However, the association between toxic dinoflagellates and shellfish poisoning is unclear. Therefore, it is necessary to understand the toxicity, detoxification, and compositional differences in PSP in Mytilus edulis exposed to PSP caused by A. catenella and A. pacificum. High-performance liquid chromatography with post-column oxidation was used to analyze PSP toxicity in poisoned M. edulis. PSP in M. edulis increased as the A. catenella and A. pacificum cell density increased. However, the cell density of A. catenella peaked faster than that of A. pacificum, and a high level of toxicity was detected. In the detoxification experiment, PSP in M. edulis decreased rapidly within 24 h in filtered seawater. However, PSP was continuously detected without decreasing below the detection limit until the last day of the experiment. In addition, the carbamate composition (GTX1+4) was detected as the main toxic composition in poisoned M. edulis, unlike in vegetative cells. GTX1+4 can poison shellfish quickly when toxic dinoflagellates appear in the marine environment. However, poisoned shellfish take a long time to be completely detoxified. Moreover, if shellfish continuously feed on poisonous dinoflagellates, their toxicity can increase rapidly due to biotransformation. Morganella morganii is a psychrotolerant organism recognized as a histamine-producing bacteria by decarboxylation of histidine amino acid, it has been involved in cases of histamine fish poisoning reported in different seafood products. In this study, we collected a total of 125 random samples of frozen Mackerel fish stored at -18°C from different fish markets in New Valley Governorate, Egypt. Through isolation from the tissue of fish samples on Chromogenic agar plates, bacteria forming light brown colonies were identified. Confirmatory identification of bacteria was conducted using 16S rRNA as a species-specific gene, revealing the presence of 14 M. morganii strains from samples, representing an incidence of 11.2% of all samples. Biogenic amines (BAs) are Nitrogen-containing organic compounds that are important in aquaculture and food. BAs are formed by decarboxylation through various chemical reactions with bacterial activities in protein-rich fish and other foods. Biogenic amines are more common in sea fish such as tuna, sardines and mackerel. The presence of biogenic amines has also been found in foods such as spoiled fruits and vegetables. Bacteria-induced histamine poisoning in fish causes allergies as well as various disorders such as headache, flushing, abdominal cramps and hypertension. Many cases of fish poisoning occur due to histamines formed in fish such as Mackerel (Scrombridae) consumed in daily life. Biogenic amines, which are objectionable in terms of food safety, are also formed in cheese, dairy products, fruits and vegetables and other foods rich in protein along with fish. Scombroid poisoning (SP) is one of the most common causes of morbidity associated with fish consumption. The condition is caused by elevated levels of histamine in dark-fleshed fish and fish products due to inadequate processing or storage temperatures. This multicenter study was conducted at four hospitals located in two major coastal cities in Türkiye: İstanbul and Trabzon. Patient records were retrospectively evaluated for 108 cases from January 1, 2014, to December 31, 2023. A total of 79 patients were included in the study. Demographics, clinical findings, and discharge outcomes are reported. The onset of complaints was 0-60 minutes in 72 cases (91.1%) and over 1 hour in 7 cases (8.9%). Most patients were admitted to the Emergency Departments in groups of three (36.7%), four (32.9%), and two (20.3%). The most common symptom was skin rash, observed in 78 SP cases (98.7%), followed by abdominal pain in 65 (82.3%), headache in 59 (74.7%), dizziness in 54 (68.4%), heartburn in 47 (59.5%), metallic taste in the mouth in 38 (48.1%), circumoral paresthesias in 34 (43%), nausea/vomiting in 20 (25.3%), and palpitations in 13 (16.5%) patients. This study emphasizes the importance of initial diagnosis based on symptoms and patient history. With accurate diagnosis and antihistamine treatment, the benefits are significant in terms of cost-effectiveness, quality of care, hospital crowding, and patient satisfaction. The significance is discussed from a broad perspective, spanning the disciplines of emergency medicine and public health. The practical clinical suggestions apply to cases on the rise worldwide. Okadaic acid (OA) is one of the most potent marine biotoxins, causing diarrheal shellfish poisoning (DSP). The proliferation of microalgae that produce OA and its analogues is frequent, threatening human health and socioeconomic development. Several methods have been tested to remove this biotoxin from aquatic systems, yet none has proven enough efficacy to solve the problem. In this work, we synthesized and characterized low-cost composites and tested their efficacy for OA adsorption in saltwater. For the synthesis of the composites, the following starting materials were considered: chitosan of low and medium molecular weight (CH-LW and CH-MW, respectively), activated carbon (AC), and montmorillonite (MMT). Characterization by vibrational spectroscopy (FTIR), X-ray diffraction (XRD), and microscopy revealed differences in the mode of interaction of CH-LW and CH-MW with AC and MMT, suggesting that the interaction of CH-MW with MMT has mainly occurred on the surface of the clay particles and no sufficient intercalation of CH-MW into the MMT interlayers took place. Among the composites tested (CH-LW/AC, CH-MW/AC, CH-MW/AC/MMT, and CH-MW/MMT), CH-MW/MMT was the one that revealed lower OA adsorption efficiency, given the findings evidenced by the structural characterization. On the contrary, the CH-MW/AC composite revealed the highest average percentage of OA adsorption (53 ± 11%). Although preliminary, the results obtained in this work open up good perspectives for the use of this type of composite material as an adsorbent in the removal of OA from marine environments. The tapertail anchovy Coilia brachygnathus, a commercially important species mainly distributed along the mid‐lower Chang‐Jiang basin, is by far the most dominant species in Lake Honghu. To figure out its success in this semiclosed lake, some basic biological parameters of this anchovy were analysed based on samples seasonally collected from October 2020 to December 2021. The results demonstrated that the age classes of fished individuals varied from 0.5 to 3.5, with the majority (97.36%) being between 0.5 and 3 years old. The size at 50% maturity of 17.2 cm total length (TL) for females and 19.0 cm TL for males corresponded to 1 and 1.6 years, respectively. Coilia brachygnathus has a short life span, early sexual maturity and a relatively fast growth rate. The flourishing of the fish in the lake is mainly attributed to its short life span, early maturity, fast growth rate, closed fishing, pelagic spawners, the availability of plenty of food and low predation effect on it. Age 3.5 year occurs in an extremely small percentage of the total (<3%), indicating that a large number of larger‐sized or older fish died after spawning, which is probably one of the major sources of water pollution if the closed fishing measure is adopted in Lake Honghu. Thus, individuals older than 2 years or more than 20.0 cm TL should be harvested. These findings have important management implications for the fish resources in Lake Honghu and beyond. Ciguatoxins (CTXs) stand as the primary toxins causing ciguatera fish poisoning (CFP) and are essential compounds distinguished by their characteristic polycyclic ether structure. In a previous report, we identified the structures of product ions generated via homolytic fragmentation by assuming three charge sites in the mass spectrometry (MS)/MS spectrum of ciguatoxin-3C (CTX3C) using LC-MS. This study aims to elucidate the homolytic fragmentation of a ciguatoxin-3C congener. We assigned detailed structures of the product ions in the MS/MS spectrum of a naturally occurring ciguatoxin-3C congener, 51-hydroxyciguatoxin-3C (51-hydoxyCTX3C), employing liquid chromatography/quadrupole time-of-flight mass spectrometry with an atmospheric pressure chemical ionization (APCI) source. The introduction of a hydroxy substituent on C51 induced different fragmentation pathways, including a novel cleavage mechanism of the M ring involving the elimination of 51-OH and the formation of enol ether. Consequently, new cleavage patterns generated product ions at m/z 979 (C55H79O15), 439 (C24H39O7), 149 (C10H13O), 135 (C9H11O), and 115 (C6H11O2). Additionally, characteristic product ions were observed at m/z 509 (C28H45O8), 491 (C28H43O7), 481 (C26H41O8), 463 (C26H39O7), 439 (C24H39O7), 421 (C24H37O6), 171 (C9H15O3), 153 (C9H13O2), 141 (C8H13O2), and 123 (C8H11O). Okadaic acids (OAs) are causative agents of diarrhetic shellfish poisoning, produced by the dinoflagellates Dinophysis spp. and Prorocentrum spp. Microcystins (MCs) are cyclic heptapeptide hepatotoxins produced by some cyanobacteria genera, including Microcystis spp. Traditionally, toxicity detection and quantification of these natural toxins were performed using a mouse bioassay (MBA); however, this is no longer widely employed owing to its lack of accuracy, sensitivity, and with regard to animal welfare. Therefore, alternative toxicity analyses have been developed based on MCs' and OAs' specific inhibition of protein phosphatase 2A (PP2A), using p-nitrophenylphosphate (p-NPP) as a substrate. The assay is simple, inexpensive, ready for use on site, and can be applied to several samples at once. For OA detection, this assay method is appropriate for widespread application as a substitute for MBA, as evidenced by its alignment with the oral toxicity of MBA. In this review, we summarize the structure and function of PP2A, the inhibitory activities of OAs and MCs against PP2A, and the practical applications of the PP2A assay, with the aim of improving understanding of the PP2A assay as an OAs and MCs detection and quantification method, as well as its suitability for screening before confirmatory chemical analysis. The okadaic acid (OA)-group toxins, including OA, dinophysistoxin-1 (DTX1), dinophysistoxin-2 (DTX2), and dinophysistoxin-3 (DTX3), cause diarrheic shellfish poisoning in humans. To manage OA-group toxins more strictly, Korean regulations were recently revised to consider OA, DTX1, DTX2, and DTX3 combined. Thus, our study characterized the occurrence of OA, DTX1, DTX2, and DTX3 in seafood distributed across South Korea, and a risk assessment of seafood consumption was conducted. Two hundred and seventeen samples from 16 bivalve and 7 non-bivalve species collected from three representative coastal areas in 2021 were analyzed via liquid chromatography-tandem mass spectrometry. OA, DTX1, and DTX3 were detected in 2.3%, 4.1%, and 9.2% of the examined samples, with positive mean levels of 11.3, 16.4, and 40.9 µg/kg, respectively. DTX2 was not detected in any of the samples. At least one OA-group toxin was detected in the bivalve samples, including blood clams, pan shells, hard clams, mussels, and scallops, whereas none were detected in non-bivalves. The estimated acute exposure to OA-group toxins through the intake of seafood in the Korean population and consumer groups was low, ranging from 24.7 to 74.5% of the recommended acute reference dose (ARfD) of 0.33 μg OA equivalents/kg body weight. However, for the scallop consumers aged 7–12 years, acute exposure to OA-group toxins exceeded the ARfD, indicating a possible health risk. These results suggest that including DTX3 in the new regulatory limits is appropriate to protect Korean seafood consumers from exposure to OA-group toxins. [ABSTRACT FROM AUTHOR]

The present study aimed to carry out a diagnosis of good handling practices (GHP), hygienic-sanitary conditions and concentrations of minerals in sururu (Mytella falcata) marketed in the street markets of the municipalities bordered by the lagoons that make up the Mundaú-Manguaba Estuarine-Lagoon Complex (MMELC) in dry and rainy season. A checklist was used to assist in the classification of the markets as to the compliance with the guidelines provided for by RDC No. 216/2004 - ANVISA. To investigate the minerals, samples were collected from all points of sale found, totaling 33 samples in the two research periods. The concentrations of iron (Fe), copper (Cu), manganese (Mn) and zinc (Zn) were analyzed using the atomic absorption spectrophotometry technique, and the concentration of lead (Pb) was analyzed according to specific protocol (SMEWW). With the application of the checklist, it was possible to classify the hygienic-sanitary conditions in all markets visited as unsatisfactory. As for the microminerals, regardless of the collection period, iron had the highest concentrations in all samples, surpassing, in overall means, by more than fifteen times the detected concentrations of copper, manganese and zinc. Regarding the toxic metal lead, the samples were within the limits allowed in bivalve mollusks. The results obtained with this study confirm the importance of sururu for countless families who sell it in street markets and reinforce the need for a joint effort of both vendors and those responsible for managing these spaces to provide the population of these localities with a favorable environment for food marketing. Tetrodotoxin (TTX) can be dangerous to food security or even cause death. But TTX also has beneficial values, for example in clinical applications and drug discovery, it is used to treat arthritis, asthma and as a pain relievers. A clear understanding of the source, ecological function, toxicity, and resistance mechanisms of TTX in animals is beneficial for ecological protection, the prediction of adaptive evolution, and TTX production. Nevertheless, the origin of TTX and how organisms bearing TTX are able to resist its toxicity are still unclear. There are two hypotheses regarding the source of TTX in TTX-containing organisms: exogenous and endogenous sources. Bacteria are involved in both hypotheses. In addition, the presence of TTX-binding proteins and mutations in the gene encoding for voltage-gated sodium channels (Nav) may be responsible for TTX resistance, which remains to be elucidated. Here, we review the ecological interactions between TTX and TTX-bearing organisms. This review provides important information for ecological research on TTX, along with suggestions for scientific prevention of seafood poisoning and the development of TTX detoxification drugs. Recent studies have reported on the occurrence of tetrodotoxin (TTX), a sodium channel-blocking neurotoxin commonly found in pufferfishes, in various marine gastropods in tropical and temperate shallow marine environments, raising concern for potential seafood poisoning as the sea snails are consumed. The intertidal beaches and rocky shores of Jeju Island include numerous subtropical and temperate gastropods including mud snails, which are commonly exploited as seafood. Accordingly, surveillance of TTX-bearing gastropods in intertidal Jeju Island is necessary to ensure seafood safety. In this study, we first screened TTX in 15 gastropod species from the Jeju coast using TTX-specific competitive ELISA. The ELISA indicated that mud snail Nassarius livescens contained a certain level of TTX in their muscle and visceral mass, whereas 14 species of gastropods were free from TTX. Accordingly, the levels of TTX and its analogues were determined in 30 individuals of N. livescens using liquid chromatography–tandem mass spectrometry (LC–MS/MS). The LC–MS/MS applied in this study showed a quantification limit (method detection/quantification limit [MDL/MQL]) of 18–19 µg/kg and 56–57 µg/kg, respectively, indicating that the LC–MS/MS was sensitive enough to detect a small quantity of TTX in the tissues. The TTX levels in the muscle ranged from below the detection limit to 3006.9 µg/kg, and those in the viscera ranged from below the detection limit to 1457.2 µg/kg. The detection frequency of TTX in mud snails was 83.3% in muscle and 70.0% in visceral mass, indicating that most N. livescens contained TTX. Algal toxins pose a serious threat to human and coastal ecosystem health, even if their potential impacts are poorly documented in New Caledonia (NC). In this survey, bivalves and seawater (concentrated through passive samplers) from bays surrounding Noumea, NC, collected during the warm and cold seasons were analyzed for algal toxins using a multi-toxin screening approach. Several groups of marine microalgal toxins were detected for the first time in NC. Okadaic acid (OA), azaspiracid-2 (AZA2), pectenotoxin-2 (PTX2), pinnatoxin-G (PnTX-G), and homo-yessotoxin (homo-YTX) were detected in seawater at higher levels during the summer. A more diversified toxin profile was found in shellfish with brevetoxin-3 (BTX3), gymnodimine-A (GYM-A), and 13-desmethyl spirolide-C (SPX1), being confirmed in addition to the five toxin groups also found in seawater. Diarrhetic and neurotoxic toxins did not exceed regulatory limits, but PnTX-G was present at up to the limit of the threshold recommended by the French Food Safety Authority (ANSES, 23 μg kg−1). In the present study, internationally regulated toxins of the AZA-, BTX-, and OA-groups by the Codex Alimentarius were detected in addition to five emerging toxin groups, indicating that algal toxins pose a potential risk for the consumers in NC or shellfish export. For the purpose of assessing human health exposure, it is necessary to characterize the toxins present in a given area and their potential impact on commercial species. The goal of this research study was: (1) to screen the prevalence and concentrations of lipophilic toxins in nine groups of marine invertebrates in the northwest Iberian Peninsula; (2) to evaluate the validity of wild mussels (Mytilus galloprovincialis) as sentinel organisms for the toxicity in non-bivalve invertebrates from the same area. The screening of multiple lipophilic toxins in 1150 samples has allowed reporting for the first time the presence of 13-desmethyl spirolide C, pinnatoxin G, okadaic acid, and dinophysistoxins 2 in a variety of non-traditional vectors. In general, these two emerging toxins showed the highest prevalence (12.5–75%) in most of the groups studied. Maximum levels for 13-desmethyl spirolide C and pinnatoxin G were found in the bivalves Magallana gigas (21 µg kg−1) and Tellina donacina (63 µg kg−1), respectively. However, mean concentrations for the bivalve group were shallow (2–6 µg kg−1). Okadaic acid and dinophysistoxin 2 with lower prevalence (1.6–44.4%) showed, on the contrary, very high concentration values in specific species of crustaceans and polychaetes (334 and 235 µg kg−−1, respectively), to which special attention should be paid. Statistical data analyses showed that mussels could be considered good biological indicators for the toxicities of certain groups in a particular area, with correlations between 0.710 (for echinoderms) and 0.838 (for crustaceans). The first ciguatera fish poisoning (CFP) in Portugal dates from 2008 when 11 people reported CFP symptoms after consuming a 30 kg amberjack caught around the Selvagens Islands (Madeira Archipelago). Since then, 49 human poisonings have been reported. The emergence of a new threat challenged scientists and regulators, as methods for toxic microalgae analyses and ciguatoxin (CTX) detection were not implemented. To minimise the risk of ciguatera, the Madeira Archipelago authorities interdicted fisheries in Selvagens Islands and banned the capture of amberjacks weighing more than 10 kg in the entire region of Madeira Archipelago. The accurate identification and quantification of the benthic toxin-producing algae species spreading to new areas require efforts in terms of both microscopy and molecular techniques. Two ciguatera-causing dinoflagellates, Gambierdiscus excentricus and Gambierdiscus australes, were identified in the Madeira Island and Selvagens sub-archipelago, respectively. Regarding the CTX analysis (N2a cell-based assay and LC-MS) in fish, the results indicate that the Selvagens Islands are a ciguatera risk area and that fish vectoring CTX are not limited to top predator species. Nevertheless, advances and improvements in screening methods for the fast detection of toxicity in seafood along with certified reference material and sensitive and selective targeted analytical methods for the determination of CTX content are still pending. Rationale: Marine polycyclic ethers have drawn attention owing to their unique chemical structures and involvement in food poisoning and fish killing. To study structural diversity, we performed a structural assignment of product ions produced from a representative ladder‐shaped polycyclic ether, ciguatoxin‐3C, and elucidated the mechanism of generation. Methods: The product ions used for the structural assignment were produced from a precursor ion [M + H]+ using liquid chromatography/quadrupole time‐of‐flight mass spectrometry, by employing an atmospheric pressure chemical ionization source. Results: Three charged sites were considered at both terminals of a molecule. Typical charge‐remote fragmentation was produced at the respective charge sites, yielding a hybrid spectrum. C–C bonds bordering two ethers could cleave and trigger the fission of two other bonds. Prominent ions indicating the serial loss of water molecules resulted from the simultaneous deprivation of ethereal oxygen and hydrogen atoms. The resultant double bonds formed long chains of conjugated polyenes, which stabilized charge via resonance. Conclusions: Three alternative charge sites produce a hybrid spectrum. The simultaneous fission of three bonds was explained. For the first time, intense ions due to serial dehydration were explained by the elimination of ether oxygen atoms and the subsequent conjugation of double bonds. The feeding behavior of P. parvus separately fed on three algae, Prorocentrum minimum, Alexandrium minutum, and Thalassiosira weissflogii, was studied at five different concentrations. The factors characterizing feeding behavior, including the beating frequency (BF), beating time (BT), and rejection behavior, were analyzed. The average BT and BF of P. parvus fed on toxic algae were significantly lower than those of copepods fed on nontoxic algae, indicating that the toxic algae negatively affected their feeding behavior. There were no significant differences in feed rejection among the three algae during the short period of experimentation, indicating that the rejection behavior was insignificant in the early period (within 20 min) of feeding on toxic algae. The feeding behavior was inhibited when the concentration reached 250 cells/mL. The BT was initially affected at increasing concentrations followed by the BF, and P. minimum and A. minutum reduced the BF at concentrations of 250 and 1000 cells/mL, respectively. Analysis of the average BFs revealed that P. parvus was more significantly affected by P. minimum containing diarrheal shellfish poison than by A. minutum containing paralytic shellfish poison. The BF of copepods fed on P. minimum was significantly lower than that of copepods fed on A. minutum at 250–500 cells/mL but was not significantly different from that at 1000 cells/mL. This indicated that the inhibitory effect of P. minimum on the feeding behavior was more significant at concentrations observed at the onset of red tide blooms (0.25–0.5 × 102 cells/mL), but insignificant at concentrations reaching those in advanced red tides (>103 cells/mL). Okadaic acid (OA) and its analogues cause diarrhetic shellfish poisoning (DSP) in humans, and risk assessments of these toxins require toxicity equivalency factors (TEFs), which represent the relative toxicities of analogues. However, no human death by DSP toxin has been reported, and its current TEF value is based on acute lethality. To properly reflect the symptoms of DSP, such as diarrhea without death, the chronic toxicity of DSP toxins at sublethal doses should be considered. In this study, we obtained acute oral LD50 values for OA and dinophysistoxin-1 (DTX-1) (1069 and 897 μg/kg, respectively) to set sublethal doses. Mice were treated with sublethal doses of OA and DTX-1 for 7 days. The mice lost body weight, and the disease activity index and intestinal crypt depths increased. Furthermore, these changes were more severe in OA-treated mice than in the DTX-1-treated mice. Strikingly, ascites was observed, and its severity was greater in mice treated with OA. Our findings suggest that OA is at least as toxic as DTX-1 after repeated oral administration at a low dose. This is the first study to compare repeated oral dosing of DSP toxins. Contamination of the food and especially marine environment with nano/micro-plastic particles has raised serious concern in recent years. Environmental pollution and the resulting seafood contamination with microplastic (MP) pose a potential threat to consumers. The absorption rate of the MP by fish is generally considered low, although the bioavailability depends on the physical and chemical properties of the consumed MP. The available safety studies are inconclusive, although there is an indication that prolonged exposure to high levels of orally administered MP can be hazardous for consumers. This review details novel findings about the occurrence of MP, along with its physical and chemical properties, in the marine environment and seafood. The effect of processing on the content of MP in the final product is also reviewed. Additionally, recent findings regarding the impact of exposure of MP on human health are discussed. Finally, gaps in current knowledge are underlined, and the possibilities for future research are indicated in the review. There is an urgent need for further research on the absorption and bioavailability of consumed MP and in vivo studies on chronic exposure. Perna Viridis has become a common dish in daily life. Consumption of Perna Viridis, which is contaminated with diarrhetic shellfish poisoning (DSP) toxins can cause food poisoning in humans. Therefore, there is an urgent need for a rapid and accurate method to identify Perna Viridis contaminated with DSP toxins. In this study, a non-destructive method based on near-infrared spectroscopy (NIS) is proposed to detect DSP toxin-laden Perna Viridis. Spectral data of Perna Viridis were collected in the range of 950–1700 nm for DSP toxin-laden and non-laden (non-DSP) samples. The second derivative (D2) spectral preprocessing method was used to reduce the influence of noise and light scattering. A support vector machine (SVM) model based on Bayesian optimization (BO-SVM) was constructed as a classifier for the detection of the DSP toxin-laden Perna Viridis. The average accuracy for the D2-BO-SVM classifier in detecting DSP toxin-laden Perna Viridis in 50 random tests reached 99.97%. The recognition performance of the D2-BO-SVM model was analyzed to solve the problems of different ratios of training and test datasets, small sample datasets, and unbalanced class datasets in practical applications. The experimental results show that the D2-BO-SVM model was found to be superior to BO-SVM, D2-Standard SVM, and Standard SVM. These results suggest that the combination of NIS and artificial intelligence models has great potential for the detection of the DSP toxin-laden Perna Viridis. Some dinoflagellates cause harmful algal blooms, releasing toxic secondary metabolites, to the detriment of marine ecosystems and human health. Phosphorus (P) is a limiting macronutrient for dinoflagellate growth in the ocean. Previous studies have been focused on the physiological response of dinoflagellates to ambient P changes. However, the whole-genome's molecular mechanisms are poorly understood. In this study, RNA-Seq was utilized to compare the global gene expression patterns of a marine diarrheic shellfish poisoning (DSP) toxin-producing dinoflagellate, Prorocentrum lima, grown in inorganic P-replete and P-deficient conditions. A total of 148 unigenes were significantly up-regulated, and 30 unigenes were down-regulated under 1/4 P-limited conditions, while 2708 unigenes were significantly up-regulated, and 284 unigenes were down-regulated under 1/16 P-limited conditions. KEGG enrichment analysis of the differentially expressed genes shows that genes related to ribosomal proteins, glycolysis, fatty acid biosynthesis, phagosome formation, and ubiquitin-mediated proteolysis are found to be up-regulated, while most of the genes related to photosynthesis are down-regulated. Further analysis shows that genes encoding P transporters, organic P utilization, and endocytosis are significantly up-regulated in the P-limited cells, indicating a strong ability of P. lima to utilize dissolved inorganic P as well as intracellular organic P. These transcriptomic data are further corroborated by biochemical and physiological analyses, which reveals that under P deficiency, cellular contents of starch, lipid, and toxin increase, while photosynthetic efficiency declines. Our results indicate that has P. lima evolved diverse strategies to acclimatize to low P environments. Ammonia–nitrogen is a common stress factor for aquatic organisms in their habitation environment, which is enriched in water due to high-density farming and environmental pollution. Ammonia nitrogen can enter fish body through gill, epidermis, digestive tract, and other tissues, causing fish ammonia poisoning. In the present study, juvenile blunt snout bream (average weight, 45 ± 5 g) were exposed to high concentrations of ammonia–nitrogen stress (25.0 ± 0.5 mg/L) for six different treatment times (0, 3, 6, 12, 24, 48, and 72 h); the tissue ultrastructure, mRNA levels of antioxidation system, and apoptosis patterns were studied. The antioxidant systems of malondialdehyde (MDA), catalase (CAT), acid phosphatase (ACP), and reduced glutathione (GSH) in various tissues were highly transcripted at 6 or 12 h (hpt) after treatment under high ammonia–nitrogen, which may play a role in preventing cells from being attacked by highly toxic reactive oxygen species (ROS). After 24 hpt, the antioxidant capacity threshold is breached, followed by the decline of antioxidant enzyme activity. Thus, with the prolonging of high ammonia–nitrogen processing time, ammonia–nitrogen stress caused irreversible damage of organs (gill, liver, and kidney). Furthermore, the expression of caspase-3 apoptotic pathway was highly induced in different tissues, implying the apoptotic system is activated, which causes extensive cell apoptosis in different tissues as shown using TUNEL analysis. Multiple species of the genus Dinophysis produce diarrhetic shellfish toxins (okadaic acid and Dinophysis toxins, OA/DTXs analogs) and/or pectenotoxins (PTXs). Only since 2008 have DSP events (illnesses and/or shellfish harvesting closures) become recognized as a threat to human health in the United States. This study characterized 20 strains representing five species of Dinophysis spp. isolated from three US coastal regions that have experienced DSP events: the Northeast/Mid‐Atlantic, the Gulf of Mexico, and the Pacific Northwest. Using a combination of morphometric and DNA‐based evidence, seven Northeast/Mid‐Atlantic isolates and four Pacific Northwest isolates were classified as D. acuminata, a total of four isolates from two coasts were classified as D. norvegica, two isolates from the Pacific Northwest coast were identified as D. fortii, and three isolates from the Gulf of Mexico were identified as D. ovum and D. caudata. Toxin profiles of D. acuminata and D. norvegica varied by their geographical origin within the United States. Cross‐regional comparison of toxin profiles was not possible with the other three species; however, within each region, distinct species‐conserved profiles for isolates of D. fortii, D. ovum, and D. caudata were observed. Historical and recent data from various State and Tribal monitoring programs were compiled and compared, including maximum recorded cell abundances of Dinophysis spp., maximum concentrations of OA/DTXs recorded in commercial shellfish species, and durations of harvesting closures, to provide perspective regarding potential for DSP impacts to regional public health and shellfish industry. DINOPHYSIS NUTRITIONAL SOURCES I Dinophysis i species are obligate mixotrophs that require ciliate prey (e.g., I Mesodinium i species), light, and nutrients for sustained growth (Hansen et al., [16]; Kim, Kim, et al., [22]). These two issues are dealt with and all available results related to I Dinophysis i and their toxins in the United States are reviewed in: "A survey of I Dinophysis i spp. and their potential to cause diarrhetic shellfish poisoning in coastal waters of the United States" (Ayache et al., [3]). A synoptic view of the current distribution of I Dinophysis i species and reported DSP events (Figure 1) confirms that occurrence of toxic outbreaks is determined by the composition (profile and content) of local strains and the shellfish species affected (Blanco, [4]). Okadaic acid (OA), a marine biotoxin produced by microalgae, poses a significant threat to mariculture, seafood safety, and human health. The establishment of a novel, highly sensitive detection method for OA would have significant practical and scientific implications. Therefore, the purpose of this study was to develop an innovative approach for OA detection. A competitive amplified luminescent proximity homogeneous assay (AlphaLISA) was developed using the principle of specific antigen–antibody binding based on the energy transfer between chemiluminescent microspheres. The method was non-washable, sensitive, and rapid, which could detect 2 × 10−2–200 ng/mL of OA within 15 min, and the detection limit was 4.55 × 10−3 ng/mL. The average intra- and inter-assay coefficients of variation were 2.54% and 6.26%, respectively. Detection of the actual sample results exhibited a good correlation with high-performance liquid chromatography. In conclusion, a simple, rapid, sensitive, and accurate AlphaLISA method was established for detecting OA and is expected to significantly contribute to marine biotoxin research. Although tunas represent a significant part of the global fish economy and a major nutritional resource worldwide, their microbiome still remains poorly documented. Here, we conducted an analysis of the taxonomic composition of the bacterial communities inhabiting the gut, skin, and liver of two most consumed tropical tuna species (skipjack and yellowfin), from individuals caught in the Atlantic and Indian oceans. We hypothesized that each organ harbors a specific microbial assemblage whose composition might vary according to different biotic (sex, species) and/or abiotic (environmental) factors. Our results revealed that the composition of the tuna microbiome was totally independent of fish sex, regardless of the species and ocean considered. Instead, the main determinants of observed diversity were (i) tuna species for the gut and (ii) sampling site for the skin mucus layer and (iii) a combination of both parameters for the liver. Interestingly, 4.5% of all amplicon sequence variants (ASV) were shared by the three organs, highlighting the presence of a core-microbiota whose most abundant representatives belonged to the genera Mycoplasma, Cutibacterium, and Photobacterium. Our study also revealed the presence of a unique and diversified bacterial assemblage within the tuna liver, comprising a substantial proportion of potential histamine-producing bacteria, well known for their pathogenicity and their contribution to fish poisoning cases. Marine biotoxins are produced by certain phytoplankton species and used to accumulate in filter-feeding marine organisms. The occurrence of marine biotoxins in all aquatic environments and latitudes is variable in time and space. Thus, it is an essentially natural phenomenon, but the occurrence of toxigenic phytoplankton cannot be completely avoided or eliminated. A serious concern appears if these substances accumulate at high levels in seafood. If it is consumed by mammals including humans, severe illness of consumers of intoxicated seafood may result. The aim of this study is to assess the presence of marine biotoxins in plankton samples taken in 2021 and to compare the determined levels with a previous period. Plankton samples (n = 21) were collected in 2021 along the whole Bulgarian coastline (Black Sea). The presence of hydrophilic (domoic acid (DA)) and lipophilic toxins (okadaic acid, dinophysis toxin - 1, dinophysis toxin -2, azaspiracid-1, goniodomin A, pectenotoxin-2 (PTX2), yessotoxin, spirolide-1 and gymnodimine A) was investigated via liquid chromatography - tandem mass spectrometry (LC-MS/MS). Results indicated the presence of only DA in three samples and PTX2 in two samples. The positive samples were sporadically distributed throughout the study period. During 2016-2019, LC-MS/MS analysis confirmed the presence of DA, PTX2, YTX, SPX-1 and GDA in plankton net samples collected from the same locations reported here. The matching toxins (DA and PTX2) were at comparable levels in both periods of investigation, thus lower than in other European waters where harmful algal blooms are registered. These results show the persistent appearance of some marine biotoxins in Bulgarian waters. Ciguatera is a major circumtropical poisoning caused by the consumption of marine fish and invertebrates contaminated with ciguatoxins (CTXs): neurotoxins produced by endemic and benthic dinoflagellates which are biotransformed in the fish food-web. We provide a history of ciguatera research conducted over the past 70 years on ciguatoxins from the Pacific Ocean (P-CTXs) and Caribbean Sea (C-CTXs) and describe their main chemical, biochemical, and toxicological properties. Currently, there is no official method for the extraction and quantification of ciguatoxins, regardless their origin, mainly due to limited CTX-certified reference materials. In this review, the extraction and purification procedures of C-CTXs are investigated, considering specific objectives such as isolating reference materials, analysing fish toxin profiles, or ensuring food safety control. Certain in vitro assays may provide sufficient sensitivity to detect C-CTXs at sub-ppb levels in fish, but they do not allow for individual identification of CTXs. Recent advances in analysis using liquid chromatography coupled with low- or high-resolution mass spectrometry provide new opportunities to identify known C-CTXs, to gain structural insights into new analogues, and to quantify C-CTXs. Together, these methods reveal that ciguatera arises from a multiplicity of CTXs, although one major form (C-CTX-1) seems to dominate. However, questions arise regarding the abundance and instability of certain C-CTXs, which are further complicated by the wide array of CTX-producing dinoflagellates and fish vectors. The marine thecate dinoflagellate Azadinium includes species known to produce the diarrhetic shellfish toxins known as azaspiracids (AZAs). In this study, the morphology of a thecate dinoflagellate isolated from Mutsu Bay, Japan, was examined by LM and SEM, and its phylogenetic position was inferred from ITS and LSU rDNA sequences. The production of AZAs was examined by liquid chromatography-triple quadrupole mass spectrometer (LC-MS/MS). Cells were elliptical, 15.9–22.4 µm long and 11.1–17.1 µm wide. A chloroplast was positioned peripherally, with a pyrenoid near the cingulum, and the nucleus was placed in the hyposome. The hyposome was smaller than the episome, with an antapical spine on plate 2''''. Thecal tabulation was Po, cp, X, 4', 3a, 6'', 6C, 5S, 6''', 2''''. The apical pore complex consisted of a symmetrical apical pore plate (Po), a cover plate (cp) and a small canal plate (X), and was surrounded by a prominent collar formed by the apical plates. A ventral pore was located on the mid-anterior edge of plate 1' and contacted with the X-plate and/or the ventral left of plate Po. Plate 1'' was in contact with plate 1a. AZA was not detected in the cultures. Molecular phylogeny showed that the species was related to Azadinium species that possess the ventral pore on the left side of plate 1', i.e. A. cuneatum, A. dalianense, A. obesum, A. poporum, A. spinosum and A. trinitatum, but that it was not included in these species. The new species Azadinium anteroporum sp. nov. differs in the ventral pore position (mid-anterior edge of plate 1') from these related species (which have the ventral pore on the left side of plate 1'), and from A. polongum (with ventral pore on the posterior left side of 1') and all other Azadinium species (with ventral pore on the right side of 1'). The Clibadium spp. is a shrub of occurrence in the Amazon, popularly known as Cunambi. The compounds in the leaves demonstrate ichthyotoxic properties, and its major substance, cunaniol, is a powerful central nervous system stimulant with proconvulsant activity. Few current studies relate behavioral changes to the electrophysiological profile of fish poisoning. This study aimed to describe the behavioral, electromyographic, electroencephalographic, electrocardiographic, and seizure control characteristics of anticonvulsant drugs in Colossoma macropomum submitted to cunaniol intoxication during bathing containing 0.3 μg/L cunaniol. The behavioral test showed rapid evolution presenting excitability and spasms, which were confirmed by the analysis of Electroencephalogram (EEG), Electromyogram (EMG), and changes in cardiac function detected in the ECG. Cunaniol-induced excitability control was evaluated using three anticonvulsant agents: Phenytoin, Phenobarbital, and Diazepam. While phenytoin was not effective in seizure control, diazepam proved to be the most efficient. These results demonstrate the susceptibility of Colossoma macropomum to cunaniol poisoning, given that the central nervous system and electrocardiographic changes were considered severe. Species within the dinoflagellate genus Dinophysis can produce okadiac acid and dinophysistoxins leading to diarrhetic shellfish poisoning. Since the first report of D. ovum from the Gulf of Mexico in 2008, reports of other Dinophysis species across US have increased. Members of the D. cf. acuminata complex (D. acuminata, D. acuta, D. ovum, D. sacculus) are difficult to differentiate due to their morphological similarities. Dinophysis feeds on and steals the chloroplasts from the ciliate, Mesodinium rubrum, which in turn has fed on and captured the chloroplasts of its prey, the cryptophyte Teleaulax amphioxeia. The objective of this study was to generate de novo transcriptomes for new isolates of these mixotrophic organisms. The transcriptomes obtained will serve as a reference for future experiments to assess the effect of different abiotic and biotic conditions and will also provide a useful resource for screening potential marker genes to differentiate among the closely related species within the D. cf. acuminata-complex. Okadaic acid (OA), a lipophilic phycotoxin distributed worldwide, causes diarrheic shellfish poisoning and even leads to tumor formation. Currently, the consumption of contaminated seafood is the most likely cause of chronic OA exposure, but there is a serious lack of relevant data. Here, the Sprague-Dawley rats were exposure to OA by oral administration at 100 µg/kg body weight, and the tissues were collected and analyzed to assess the effect of subchronic OA exposure. The results showed that subchronic OA administration disturbed colonic mucosal integrity and induced colitis. The colonic tight junction proteins were disrupted and the cell cycle of colonic epithelial cells was accelerated. It is inferred that disruption of the colonic tight junction proteins might be related to the development of chronic diarrhea by affecting water and ion transport. Moreover, the accelerated proliferation of colonic epithelial cells indicated that subchronic OA exposure might promote the restitution process of gut barrier or induce tumor promoter activity in rat colon. The successful cultivation of Dinophysis norvegica Claparède & Lachmann, 1859, isolated from Japanese coastal waters, is presented in this study, which also includes an examination of its toxin content and production for the first time. Maintaining the strains at a high abundance (>2000 cells per mL−1) for more than 20 months was achieved by feeding them with the ciliate Mesodinium rubrum Lohmann, 1908, along with the addition of the cryptophyte Teleaulax amphioxeia (W.Conrad) D.R.A.Hill, 1992. Toxin production was examined using seven established strains. At the end of the one-month incubation period, the total amounts of pectenotoxin-2 (PTX2) and dinophysistoxin-1 (DTX1) ranged between 132.0 and 375.0 ng per mL−1 (n = 7), and 0.7 and 3.6 ng per mL−1 (n = 3), respectively. Furthermore, only one strain was found to contain a trace level of okadaic acid (OA). Similarly, the cell quota of pectenotoxin-2 (PTX2) and dinophysistoxin-1 (DTX1) ranged from 60.6 to 152.4 pg per cell−1 (n = 7) and 0.5 to 1.2 pg per cell−1 (n = 3), respectively. The results of this study indicate that toxin production in this species is subject to variation depending on the strain. According to the growth experiment, D. norvegica exhibited a long lag phase, as suggested by the slow growth observed during the first 12 days. In the growth experiment, D. norvegica grew very slowly for the first 12 days, suggesting they had a long lag phase. However, after that, they grew exponentially, with a maximum growth rate of 0.56 divisions per day (during Days 24–27), reaching a maximum concentration of 3000 cells per mL−1 at the end of the incubation (Day 36). In the toxin production study, the concentration of DTX1 and PTX2 increased following their vegetative growth, but the toxin production still increased exponentially on Day 36 (1.3 ng per mL−1 and 154.7 ng per mL−1 of DTX1 and PTX2, respectively). The concentration of OA remained below detectable levels (≤0.010 ng per mL−1) during the 36-day incubation period, with the exception of Day 6. Background: There are inconsistent results of cohort studies analyzing the association between fish intake and mortality. Objective: This study was performed to explore the association of oily fish consumption and nonoily fish consumption with all-cause mortality and cause-specific mortality. Methods: A total of 431,062 participants from the UK Biobank who were without cancer or cardiovascular disease (CVD) at baseline between 2006 and 2010 were included in this study, and they were followed up through 2021. We constructed Cox proportional hazard models to calculate the hazard ratio (HR) and 95% confidence interval (CI) to assess the correlation of oily fish and nonoily fish intakes with mortality. Then, we performed subgroup analyses, and sensitivity analyses were developed and performed to examine the robustness of this study. Results: Among the participants, 383,248 (88.9%) and 410,499 (95.2%) consumed oily fish and nonoily fish, respectively. Compared with the participants who did not consume oily fish, the adjusted HRs for the association of oily fish consumption (1 serving/week) with all-cause mortality and CVD mortality were 0.93 (0.87 to 0.98; p < 0.05) and 0.85 (0.74 to 0.98; p < 0.05), respectively. The multivariable-adjusted HRs of all-cause mortality for those who reported consuming < 1 serving/week of oily fish were 0.92 (0.86 to 0.98; p < 0.05). Conclusion: Compared with participants who reported never consuming oily fish, the consumption of oily fish with 1 serving/week was more beneficial for all-cause and CVD mortality. Background. The diarrhoeal shellfish poisoning (DSP) toxin is a powerful marine biological toxin. Eating DSP toxin-contaminated mussels will lead to serious gastrointestinal diseases. To this end, a method for the detection of DSP toxins using near-infrared reflectance spectroscopy combined with pattern recognition is proposed. Material and methods. In the range from 950−1700 nm, spectral data of healthy mussels and DSP-contaminated mussels were obtained. To select the optimal band subsets, a band selection algorithm based on model cluster analysis was applied. As distinguishing DSP toxin-contaminated mussels from healthy mussels is a classification problem of imbalanced data, an improved fuzzy support vector machine-based recognition method was proposed. The influence of the parameters of the band selection algorithm and the fuzzy support vector machine on the model performance was analyzed. Results. Compared with the traditional support vector machine, the proposed model has better performance in detecting DSP toxins and is not affected by the imbalance ratio. Its geometric mean value can reach 0.9886 and the detection accuracy can reach 98.83%. Conclusion. Fish poisoning (FP) affects human health, trade and livelihood in Fiji, where management has depended mainly on traditional ecological knowledge (TEK). This paper investigated and documented this TEK through a 2-day stakeholder workshop, group consultation, in-depth interviews, field observations, and analyses of survey data from the Ministry of Fisheries, Fiji. Six TEK topics were identified and classified as preventative and treatment options. The preventive approach involves identifying toxic reef fishes, the spawning season of edible seaworms, hotspot areas of toxic fishes, folk tests, and locating and removing toxic organs. For example, 34 reef fish species were identified as toxic. The FP season was associated with the spawning of balolo (edible seaworm) and the warmer months of October to April (cyclone seasons). Two well-known toxic hotspots associated with an abundance of bulewa (soft coral) were identified. Folk tests and locating and removing toxic fish organs are also practised for moray eels and pufferfish. At the same time, various locally available herbal plants are used to treat FP as the second line of defence. The TEK collated in this work can help local authorities better identify the sources of toxicity, and applying TEK preventive measures could stem the tide of fish poisoning in Fiji. The more frequent occurrence of marine harmful algal blooms (HABs) and recent problems with newly-described toxins in Puget Sound have increased the risk for illness and have negatively impacted sustainable access to shellfish in Washington State. Marine toxins that affect safe shellfish harvest because of their impact on human health are the saxitoxins that cause paralytic shellfish poisoning (PSP), domoic acid that causes amnesic shellfish poisoning (ASP), diarrhetic shellfish toxins that cause diarrhetic shellfish poisoning (DSP) and the recent measurement of azaspiracids, known to cause azaspiracid poisoning (AZP), at low concentrations in Puget Sound shellfish. The flagellate, Heterosigma akashiwo, impacts the health and harvestability of aquacultured and wild salmon in Puget Sound. The more recently described flagellates that cause the illness or death of cultivated and wild shellfish, include Protoceratium reticulatum, known to produce yessotoxins, Akashiwo sanguinea and Phaeocystis globosa. This increased incidence of HABs, especially dinoflagellate HABs that are expected in increase with enhanced stratification linked to climate change, has necessitated the partnership of state regulatory programs with SoundToxins, the research, monitoring and early warning program for HABs in Puget Sound, that allows shellfish growers, Native tribes, environmental learning centers and citizens, to be the "eyes on the coast". This partnership enables safe harvest of wholesome seafood for consumption in the region and helps to describe unusual events that impact the health of oceans, wildlife and humans. Liquid chromatography – tandem mass spectrometry (LC-MS/MS) is routinely applied for the determination of toxins associated with diarrhetic shellfish poisoning (DSP) as a part of the Japanese official method. For scallops, not only the edible part but also the midgut gland may be used for analysis. In this study, the matrix effects in the LC-MS/MS-based determination of okadaic acid (OA) and dinophysistoxin-1 (DTX1) in the scallop midgut gland were evaluated to assess the impact on the accuracy of the results. The analyzed sample in an interlaboratory comparison study was subjected to the Japanese official method. The solutions obtained with and without a cleanup process (combination of hexane washing and solid-phase extraction using a C18 cartridge) were diluted to varying ratios and used for evaluation. For all prepared solutions, the ionization of OA and DTX1 was enhanced compared to the samples in methanol; the cleanup process did not sufficiently reduce the matrix effects. Although the dilution of the sample solution reduced the matrix effect, the improvement was limited. Okadaic acid (OA) is a marine biotoxin that is produced by algae and accumulates in filter-feeding shellfish, through which it enters the human food chain, leading to diarrheic shellfish poisoning (DSP) after ingestion. Furthermore, additional effects of OA have been observed, such as cytotoxicity. Additionally, a strong downregulation of the expression of xenobiotic-metabolizing enzymes in the liver can be observed. The underlying mechanisms of this, however, remain to be examined. In this study, we investigated a possible underlying mechanism of the downregulation of cytochrome P450 (CYP) enzymes and the nuclear receptors pregnane X receptor (PXR) and retinoid-X-receptor alpha (RXRα) by OA through NF-κB and subsequent JAK/STAT activation in human HepaRG hepatocarcinoma cells. Our data suggest an activation of NF-κB signaling and subsequent expression and release of interleukins, which then activate JAK-dependent signaling and thus STAT3. Moreover, using the NF-κB inhibitors JSH-23 and Methysticin and the JAK inhibitors Decernotinib and Tofacitinib, we were also able to demonstrate a connection between OA-induced NF-κB and JAK signaling and the downregulation of CYP enzymes. Overall, we provide clear evidence that the effect of OA on the expression of CYP enzymes in HepaRG cells is regulated through NF-κB and subsequent JAK signaling. [ABSTRACT FROM AUTHOR]
