Abstract: The Henares-Jarama-Tajo river system is the largest drainage basin in the Province of Madrid, Spain. This area is characterized by the presence of intensive urban and industrial activities influenced by a continental Mediterranean climate with rainfalls presenting substantial fluctuations along the different seasons. This research aimed to monitor seasonal variations in concentrations of 22 pharmaceutically active compounds (PhACs) in this river system and to establish the potential risk of sublethal effects on aquatic organisms. A total of 10 sampling sites were selected along the river system with samples collected in each of the four seasons during a year-round schedule. Most of the PhACs detected were present in sampling sites downstream in the vicinity of the most populated cities (i.e. Madrid, Guadalajara and Alcalá de Henares). Only two PhACs, fluoxetine and paraxantine, were detected in all sites regardless of the season, and showed median (± interquartile range) concentrations of 21.4 (±31.2) ngL-1 and 8.5 (±5.3) ngL-1, respectively. Other PhACs were detected with a frequency >80% and included, caffeine, diphenylhydantoin, hydrochlorotiazide, ibuprofen, ketoprofen, diclofenac, sulfamethoxazole, atenolol, naproxen, carbamazepine and propanolol. Seasonal variations were observed with the highest concentrations in December and the lowest in September. By combining measured environmental concentrations with toxicity data (either publicly available or obtained experimentally in our laboratory), and by calculating an Maximum Risk Index (MaxRI) that each combination of PhACs should have for non exceeding the risk threshold, a high potential for long-term risk (MaxRI<10) was estimated for most of the sampling sites and sampling dates. This research allowed the characterization of the potential risk for each of the PhACs to exert sublethal effects on aquatic organisms using acute screening methods, justifying the need for chronic data in order to refine the risk of these compounds to aquatic organisms. A procedure involving bead-injection concept and sequential determination of copper and mercury ions in river-water samples is proposed. The method is based on the solid-phase extraction of both metal ions on the same beads surface (Chelex 100 resin) and in their subsequent reaction with the colorimetric reagents (APDC and Dithizone for copper and mercury ions, respectively). For this task, a resin mini-column is established in the optical path by the selection, introduction and trapping of a defined volume of the Chelex-100 resin beads suspension in the flow system. The passage of the sample solution through the resin mini-column promotes the sorption of Cu(II) ions and, making the APDC colorimetric reagent flows through the beads, the formation of the coloured complex on the solid phase surface occurs. The absorbance of the formed APDC–Cu complex is then monitored at 436 nm and the spent beads are discarded. Packing another resin mini-column in the flow cell and repeating the concentration step it is possible to carried out the mercury determination by using Dithizone as reagent. The absorbance of the Dithizone–Hg complex is monitored at 500 nm. After each measurement, the spent beads are wasted and a new portion of fresh one is trapped in the system, letting it ready for the next measurement. The bead injection system is versatile and can be used to concentrate different sample volumes, which permits the determination of a wide range of copper and mercury ions concentrations. When the sample-selected volumes are 100 and 1000 μl the analytical ranges were 5.0 up to 500.0 μg l-1 and 2.5 up to 30.0 μg l-1 for Cu(II) and Hg(II) ions, respectively. Under these conditions, the detection limit was estimated as 0.63 and 0.25 μg l-1 for copper and mercury ions determination. The system consumes 2 mg of Chelex 100 resin beads, 0.20 mg of APDC or 1.25 mg of Dithizone per determination and the traditional organic solvent extraction methodology, normally used in connection with APDC and Dithizone reagents, is not used here which permits to classify the present method as green. Certain chemicals possess the potential to modulate endocrine systems, and thereby interfere with reproduction and developmental processes in the wild. We analyzed endocrine disrupters nonylphenol (NP) and bisphenol A (BPA) levels at various sites in Okinawa and Ishigaki Islands, Japan. River-water samples showed undetectable to low concentrations of NP and BPA at most of the sites investigated. However, an appreciable amount of BPA was detected in sediments at one coral reef site. In addition, significant numbers of river sediment samples showed appreciable amounts of NP and BPA. Most of the sampling sites for this study are located within a distance of 1 km from the coral reefs, which are under influence of river-waters to a variable extent. Therefore, influence of endocrine disrupters may have already begun on adjacent coral reefs. Both endocrine disrupters were positively correlated with human population densities, but not with the contents of red soil generated by farm land reformation. Therefore, it is concluded that NP and BPA pollution is a consequence of human waste discharge, both domestic and industrial, and not by agricultural activities. [Copyright &y& Elsevier]
