The rapid increase in aquaculture over the last several decades has led to concerns about the environmental impact of fish feeds relying on marine resources for fishmeal (FM). We aim to assess Nannochloropsis sp. QH25 co-product as a viable and sustainable replacement for FM in juvenile rainbow trout, Oncorhynchus mykiss, feeds. We formulated four experimental diets: a reference (FM based), 33N, 66N, and 100N diet (33%, 66%, and 100% co-product replacement). Rainbow trout were randomly assigned to one of 16 tanks and randomly assigned an experimental diet to consume throughout the experiment (64 days total), with four replicate tanks per diet. We compared the phosphorus (P) and nitrogen (N) digestibility, emissions, and growth between diets and, compared six environmental impacts (biotic resource use (BRU), global warming potential (GWP), water use, land use, marine eutrophication potential (MEP), and freshwater eutrophication potential (FEP)) of each diet. Our results indicate that replacing FM with co-product did not significantly alter growth. P digestibility of the experimental and reference diets was comparable. BRU conversion ratio was significantly lower in the experimental diets. However, there were significantly higher water and land use conversion ratios but insignificantly higher results in GWP, MEP, and FEP between the reference and 100N diet.

Recent work has revealed the importance of contemporary evolution in shaping ecological outcomes. In particular, rapid evolutionary divergence between populations has been shown to impact the ecology of populations, communities, and ecosystems. While studies have focused largely on the role of adaptive divergence in generating ecologically important variation among populations, much less is known about the role of gene flow in shaping ecological outcomes. After divergence, populations may continue to interact through gene flow, which may influence evolutionary and ecological processes. Here, we investigate the role of gene flow in shaping the contemporary evolution and ecology of recently diverged populations of anadromous steelhead and resident rainbow trout (Oncorhynchus mykiss). Results show that resident rainbow trout introduced above waterfalls have diverged evolutionarily from downstream anadromous steelhead, which were the source of introductions. However, the movement of fish from above to below the waterfalls has facilitated gene flow, which has reshaped genetic and phenotypic variation in the anadromous source population. In particular, gene flow has led to an increased frequency of residency, which in turn has altered population density, size structure, and sex ratio. This result establishes gene flow as a contemporary evolutionary process that can have important ecological outcomes. From a management perspective, anadromous steelhead are generally regarded as a higher conservation priority than resident rainbow trout, even when found within the same watershed. Our results show that anadromous and resident O. mykiss populations may be connected via gene flow, with important ecological consequences. Such eco-evolutionary processes should be considered when managing recently diverged populations connected by gene flow.

Aquaculture is one of the fastest-growing food sectors, producing almost 50% of fish for human consumption worldwide. However, relying on fish meal and oil for aquaculture diets is not sustainable economically or environmentally. Aquaculture feeds also contain terrestrial plant ingredients with indigestible forms of phosphorus (P), of which 70%–80% can be released into aquatic environments. This P influx contributes to eutrophication of freshwater ecosystems that can lead to anoxic conditions. This study explores a more sustainable diet for salmonids, an important and valuable seafood. Our aim was to test ingredients with highly digestible forms of P in nutritionally balanced portions to support fish growth and reduce P loading. We determined the digestibility of three feeds containing raw, extruded, and enzymatically processed microalgal co-product of Nannochloropsis oculata compared to a conventional diet. We also quantified how much P was retained and excreted. We detected highest growth in trout fed enzymatically processed co-product feed, though it was not statistically different (p = 0.846) from growth of fish fed the reference or other co-product diets. The enzyme-treated, microalgal co-product ingredient and diet had comparable values for P digestibility and solid P excretion to the reference diet, but the lowest average solid P excretion of all test diets. Trout fed the enzyme-treated diet had the highest P retention, while the reference diet had the lowest (p = 0.0429). Trout fed the enzyme-treated diet had the lowest (p = 0.0174) and negative dissolved P excretion, while those fed the reference diet had the highest. Results showed that enzyme-treated N. oculata co-product maintains digestibility, increases P retention, and reduces dissolved P excretion compared to the reference diet in rainbow trout. These findings encourage follow-up research to design and test growth performance of diets containing enzyme-treated microalgal co-product as sustainable trout aquafeed.

Since the first introduction from North America more than a century ago, rainbow trout (Oncorhynchus mykiss) have rapidly established self-sustaining populations in major river basins of Patagonia. Many generations later, only the freshwater resident life history is expressed in the Chubut and Negro rivers of northern Argentinian Patagonia, whereas both the resident and anadromous life histories are found in the Santa Cruz River of southern Argentina. Despite previous studies that have tried to identify the sources of these introduced populations, uncertainty still exists. Here we combined data from many single-nucleotide polymorphisms and microsatellite loci in O. mykiss populations from Argentina and North America to evaluate putative source populations, gene flow between Argentinian river basins, and genetic diversity differences between Argentinian and North American populations. We found that populations from northern and southern Patagonia are highly differentiated and have limited gene flow between them. Phylogeographic analysis also confirmed that they have separate origins, with the northern populations most closely related to the domesticated rainbow trout strains that are raised worldwide and the Santa Cruz River populations most closely related to North American populations from California and Oregon that have an anadromous component. In addition, fish with different life histories in the Santa Cruz River were found to constitute a single interbreeding population. No evidence was found of reduced genetic variation in introduced rainbow trout, suggesting multiple contributing sources. In spite of these advances in understanding, significant questions remain regarding the origins and evolution of the introduced O. mykiss in Patagonia.

Cadmium contamination in aquatic environments poses severe risks to aquatic organisms, particularly fish, where cadmium accumulation in tissues can lead to compromised organ functionality and reproductive issues. The present study aimed to assess the effects of cadmium (Cd) exposure on key biomarkers of oxidative stress, DNA damage, apoptosis, and enzyme activity in the liver and kidney tissues of rainbow trout (Oncorhynchus mykiss). Specifically, the study measured 8-hydroxy-2-deoxyguanosine (8-OHdG) levels, caspase-3 activation, acetylcholinesterase (AChE) activity, and oxidative stress indicators (ONOO−, MDA, GSH, SOD, and CAT) following exposure to three Cd concentrations (1, 3, and 5 mg/L) over three time points (24, 48, and 96 h). Tissue samples were collected post-exposure, and the analysis revealed a significant decrease in MDA levels in both tissues. GSH concentrations declined with prolonged exposure, while SOD activity increased, indicating a response to oxidative stress, contrasted by a reduction in CAT activity. An initial increase in ONOO− levels was observed at 24 h, followed by a subsequent decrease at the 48 and 96 h marks. These results suggest that cadmium induces oxidative stress in the liver and kidney tissues of fish. Cadmium exposure also significantly elevated 8-OHdG levels, signaling DNA damage, and increased caspase-3 activity, indicative of apoptosis, across all doses and time points (p < 0.05). The histological examination of liver and kidney showed tissue injury. Additionally, a negative correlation between AChE activity and exposure duration was noted, with prolonged exposure resulting in substantial AChE inhibition. Given the role of AChE in behavior regulation, these findings underscore the importance of exploring time-dependent, tissue-specific changes in AChE activity to further elucidate the mechanisms underlying cadmium-induced behavioral abnormalities. Our objective was to assess the effects of interactions between the native Percichthys trucha and both Salmo trutta and Oncorhynchus mykiss during the juvenile period. We conducted single and multipredator (conspecific and heterospecific) functional response (FR) experiments with Meridialaris chiloeensis as native prey and estimated the functional response ratio as a predator impact metric. Our results showed that all species have type II FR, with salmonids having a greater impact on prey than native fish. In multipredator experiments, O. mykiss had a greater negative impact on P. trucha than S. trutta. The individual feeding performance of P. trucha was better under conspecific than heterospecific conditions. This study shows that the vulnerability of juvenile P. trucha to salmonid competition could be a real threat in Patagonian lotic environments. [ABSTRACT FROM AUTHOR]

High-mountain lakes (HMLs) are remote, extreme, and sensitive ecosystems recognized as sentinels of global change. Lakes El Sol and La Luna are very close to each other inside the crater of the Nevado de Toluca volcano, but they differ morphometrically and limnologically. This study aimed to identify the seasonal fluctuation of the phytoplankton communities of these two tropical HMLs. El Sol phytoplankton comprised 50 taxa (chlorophytes, diatoms, charophytes) and La Luna 28 taxa (diatoms, euglenoids). The abundance of phytoplankton in El Sol was three times higher than in La Luna, and the biomass in El Sol was five times higher than in La Luna. Tropical seasonality was reflected differently in each lake. In El Sol, the highest phytoplankton abundance occurred in the rainy season, while the highest biomass was recorded in the dry/warm season. Conversely, in La Luna, abundance and biomass were more prominent in the dry/cold season. The study found that no meteorological or limnological factors could explain the seasonal dynamics of the taxonomic richness, abundance, or biomass of the phytoplankton communities in both lakes. The differences between the lakes are likely due to the more extreme conditions of La Luna, such as lower pH, ultra-oligotrophy, and increased exposure to ultraviolet radiation (UVR). No current technology can specifically target grass carp (Ctenopharyngodon idella) for control within aquatic ecosystems. Rotenone and Carbon Dioxide-Carp are currently the only available registered pesticides for grass carp; they are nonselective and typically applied throughout the water, equally exposing target and native species. A more selective control tool or pesticide application could be used by resource managers to support mitigation efforts. Development of delivery systems that exploit carp feeding strategies could increase selectivity of pesticides and minimize effects on native fishes. A pesticide with selective delivery could be less labor intensive and used within an integrative pest management strategy. The present study examined Antimycin A toxicity in juvenile and sub-adult grass carp and rainbow trout (Oncorhynchus mykiss) across two routes of exposure. Water-based toxicity studies were used to calculate the concentration to cause lethality in 50% of treated fish (LC50) at 24-h, while oral gavage toxicity studies were used to calculate the dose to cause lethality in 50% of treated grass carp and rainbow trout (LD50) 24- to 96-h. Although rainbow trout were more sensitive than grass carp to Antimycin A through water-based exposure, oral toxicity was similar between species, even with inherent gastrointestinal morphological differences. Successful delivery of a lethal dose of Antimycin A to grass carp was achieved through an oral route of exposure using the rapeseed bait and shows promise for registration as a control tool and eventual use in pest management plans. Although a lethal dose of Antimycin A could be incorporated into a single bait pellet, more bait was required to achieve desired mortality when fed to fish under laboratory conditions. Astaxanthin is an important aquatic feed additive that enhances the antioxidant capacity, and immune function of rainbow trout (Oncorhynchus mykiss); however, very limited information is available on its underlying molecular mechanisms. Haematococcus pluvialis powder, Phaffia rhodozyma powder, and synthetic astaxanthin were added to the commercial feed (no astaxanthin, NA) to prepare three experimental feeds, referred to as the HPA, PRA, and SA groups, respectively, and their actual astaxanthin contents were 31.25, 32.96, and 31.50 mg.kg-1, respectively. A 16-week feeding trial was conducted on the O. mykiss with an initial body weight of 669.88 ± 36.22 g. Serum and head kidney samples from commercial-sized O. mykiss were collected for metabolomics and transcriptomics analysis, respectively. Metabolomics analysis of the serum revealed a total of 85 differential metabolites between the astaxanthinsupplemented group and the control group. These metabolites were involved in more than 30 metabolic pathways, such as glycerophospholipid metabolism, fatty acid biosynthesis, linoleic acid metabolism, and arginine and proline metabolism. It is speculated that different sources of dietary astaxanthin may regulate antioxidant capacity and immunity mainly by affecting lipid metabolism and amino acid metabolism. Transcriptomic analysis of the head kidney revealed that the differentially expressed genes between the astaxanthin-supplemented group and the control group, such as integrin beta-1 (ITGB1), alpha-2-macroglobulin (A2M), diamine acetyltransferase 1 (SAT1), CCAAT/enhancer-binding protein beta (CEBPB) and DNA damage-inducible protein 45 alpha (GADD45A), which are involved in cell adhesion molecules, the FoxO signaling pathway, phagosomes, and arginine and proline metabolism and play regulatory roles in different stages of the antioxidant and immune response of O. mykiss. Background: Aquaculture, traditionally a form of biotechnology, has evolved to integrate innovative biotechnological applications, such as advanced feed formulations, aimed at improving the growth performance and health of farmed fish species. In the present study, the effects of feeding rainbow trout with novel feed formulations were investigated. Fish growth, gut and liver morphology, the concentration of fatty acids in the fillet, and volatile fatty acids in the gut were assessed. The study also validated scenarios from in vivo experiments using a nutrient-based model called FEEDNETICS™. This globally used model serves as a tool for data interpretation and decision support in the context of precision fish farming. Methods: Alternative protein and oil sources, including poultry by-product meal (PBM) and natural algae oil, were explored as sustainable replacements for fishmeal (FM) and fish oil (FO). A 90-day feeding trial was conducted using rainbow trout, comparing two isoproteic, isolipidic and isoenergetic diets. The control diet contained 15% FM, 5% PBM, and 8% FO, while the test diet replaced FM with 15% PBM and 5% feather meal hydrolysate (FMH), and fully substituted FO with VeraMaris® natural algae oil and rapeseed oil. Results: PBM successfully replaced FM protein without negatively affecting feed intake, growth performance or feed utilization in trout. The combination of PBM and natural algae oil was well tolerated by the trout and showed no negative effects on gut health. A detailed analysis of fatty acids in the fillet revealed that PUFAs of the n3 and n6 series were significantly higher in the PBM group than in the FM group. Values of fatty acid-related health indexes, including atherogenicity index, and thrombogenicity index, confirmed the high nutritional value of trout filet, thus representing a healthy product for human. In addition, the predictions using the FEEDNETICS™ indicated that the tested novel alternative formulations are economically viable. The validation of the model for fish growth resulted in a Mean Absolute Percentage Error (MAPE) of 8%. Conclusions: The FEEDNETICS™ application enhances our ability to optimize feeding strategies and improve production efficiency in the aquaculture industry. Simple Summary: The current state of rainbow trout, Oncorhynchus mykiss, was investigated 50 years after the first introduction of the species to the water bodies of the Balkhash basin. The wild form of the species from Kamchatka and the cultured form from European fish farms have successfully adapted to the local environmental conditions. Unlike in most other regions of the world, rainbow trout have not spread beyond the water bodies of their original introduction and have not significantly impacted native fish diversity. Rainbow trout, or mykiss (Oncorhynchus mykiss), is one of the most popular species used in aquaculture and has been naturalized worldwide, including in the Central Asian Balkhash basin, which has unique aboriginal fish fauna. Both rainbow trout from European farms and wild mykiss from Kamchatka were introduced to some mountain lakes and rivers of the Balkhash basin about 50 years ago. This study investigates the current distribution and life history traits of the alien species and its possible impact on the local fish fauna. This study showed that the rainbow trout occupies various habitats in the Ili River basin: mountain lakes, fast-flowing mountain rivers, and lowland rivers with slow currents and warm water (up to +27 °C). Rainbow trout from European fish farms dominate the mountain Middle Kolsay Lake, while the wild trout from Kamchatka occupies the small Ulken Kokpak River. Both co-occur in the Chilik River. Contrary to that in other regions, the distribution of rainbow trout in the Balkhash basin remained almost the same after their introduction. Broad intrapopulation variability in terms of size, growth rate, and maturation age was revealed, apparently as a result of adaptation to the new environment and intrapopulation competition. In particular, the growth rate has decreased, but life span, surprisingly, has increased as compared to the originally introduced fish. Intrapopulation variation in growth and maturity patterns was also noted. Differences in skin coloration between highland (cold-water) and lowland (warm-water) populations were discovered. The feeding mode of naturalized trout is insectivorous (insect imago), indicating that it occupies its own niche in the local fish communities. The largest population of rainbow trout was recorded in the Lower Kolsay Lake, lowering the population of native fish species, while in other localities, no negative impact on local fish communities was recorded. Simple Summary: Global warming is an important issue in the context of climate change affecting cold-water aquaculture. Heat stress induces oxidative stress in fish, hence the use of a functional diet supplemented with antioxidant additives, such as thymol, can be useful to counteract the impact of global warming on aquaculture. In this study, we used thymol-enriched diets to rear rainbow trout fingerlings over an 8-week period followed by a period of heat stress of 48 h. Although thymol had no significant benefits on the growth and innate immunity of the fish, it improved antioxidant capacity and reduced lipid peroxidation and mortality after heat stress. The appropriate thymol dose for these benefits is 100 mg/kg. Rainbow trout fingerlings were fed, in triplicate, diets supplemented with 0 (CTL), 50 (50 TM), 100 (100 TM), 200 (200 TM), 400 (400 TM) and 800 (800 TM) mg/kg of thymol, followed by 48 h of thermal stress. Growth performance and humoral immunological parameters showed no significant responses to dietary thymol concentrations. Fish fed 50–400 mg/kg thymol diets had significantly higher survival after heat stress. Plasma cortisol, glucose, hepatic glutathione peroxidase, glutathione reductase and erythrocyte catalase significantly increased after thermal stress, whereas total plasma antioxidant capacity, ascorbate, and hepatic/erythrocyte reduced-glutathione significantly decreased. There were significant elevations in plasma ascorbate and hepatic glutathione reductase in the 50 TM, 100 TM and 200 TM groups; plasma total antioxidant capacity in the 100 TM and 200 TM groups; hepatic glutathione peroxidase in the 200 TM group; and hepatic-reduced glutathione in the 100 TM, 200 TM and 400 TM groups, compared to CTL. The highest hepatic superoxide dismutase and lowest hepatic malondialdehyde were observed in the 100 TM group before heat stress. These parameters significantly increased after thermal stress in the treatment groups, except in the 100 TM and 200 TM groups. Hepatic catalase showed no significant difference among the treatment groups before thermal stress. Hepatic catalase significantly increased after heat stress in all treatment groups, except in the 100 TM group. Erythrocyte superoxide dismutase significantly increased in the 100 TM group before heat stress, whereas erythrocyte malondialdehyde significantly decreased in the 100 TM and 200 TM groups after thermal stress. Astaxanthin (Ax) determines the flesh redness of a salmonid fish which is the most desirable quality indicator by consumers. Fish cannot synthesize Ax de novo, therefore, the only way to increase flesh redness is to increase dietary input or improve the absorption and retention rate of dietary Ax. As a hydrophobic carotenoid, the absorption of Ax can be modulated by other lipid molecules in the diet. The present study explored the effect of three lipids, cholesterol (CH), phytosterol (PS), and docosahexaenoic acid (DHA) on Ax absorption, transport, and retention in rainbow trout. Dietary CH significantly improved Ax absorption by elevating plasma Ax levels (p < 0.05); however, it had no effect on the whole body Ax or flesh color. Dietary PS appears to inhibit Ax absorption since fish had significantly (p < 0.05) reduced whole body Ax. Dietary DHA appeared to have no effect on Ax absorption or retention. By comparing intestinal transcriptomes, a low density lipoprotein receptor (ldlr) gene was significantly downregulated in fish fed the CH diet as compared to the control diet. Since LDLR protein plays a major role in plasma lipoprotein turnover, we hypothesized that the inhibition of ldlr gene by high dietary CH resulted in higher retention of plasma Ax. The elevation of plasma Ax was not reflected in higher flesh coloration, which suggested other limiting factors governing Ax retention in the muscle. On the other hand, the transcriptomic and proteomic analyses found no changes of genes or proteins involved in Ax absorption, transport, or excretion in fish fed PS or DHA diets as compared to the control diet. In conclusion, this study has suggested that CH promotes Ax absorption by regulating lipoprotein retention and provide evidence for improving Ax absorption via dietary modulation. Background: Recirculating aquaculture systems can cause chronic stress in fish when stocking density is too high. However, this study tested whether low stocking density can cause fish stress. Adult rainbow trout, with an average weight of 1.517 kg (± 0.39), were subjected to low (12 kg/m3 ± 0.94) and moderate (43 kg/m3 ± 2.03) stocking densities for 24 days in a recirculating system maintained at 15 °C. At the end of the experiment, fecal microbiome analysis was carried out using a 16S rRNA amplicon sequencing. Additionally, an untargeted plasma metabolomics analysis was conducted. Results: The moderate stocking density group harboured greater numbers of commensals, such as C. somerae, R. lituseburensis, and L. plantarum. In contrast, detrimental species such as S. putrifacens and P. putida were abundant in the low-stocking density fish. Functional microbiome profiling revealed vitamin B12 salvage and synthesis in moderate stocking densities, which may support intestinal tight junction function. Additionally, vitamin B1 biosynthesis pathways were more abundant in the moderate stocking density group, which may function towards oxidative energy metabolism and protect against oxidative stress. A complementary plasma metabolomics study, although done at slightly different stocking densities and duration, confirmed the presence of blood metabolic stress markers. Elevated levels of L-lactic acid and L-Norvaline, L-Valine, and L-glutamine, indicate low stocking density fish were under stress. Furthermore, a P4HA2 stress gene biomarker confirmed the occurrence of stress in low-density fish. This study suggests that low stocking density can induce stress in fish. Moreover, moderate stocking density leads to a distinct and beneficial fecal microbiome profile. Conclusion: Our study highlights the potential benefits of optimizing the stocking density of fish in recirculating aquaculture systems. ABSTRACT Flavobacterium psychrophilum, the causative agent of bacterial cold water disease (BCWD), is one of the leading pathogens in rainbow trout (Oncorhynchus mykiss) aquaculture. To date, there is little knowledge of the transmission kinetics of F. psychrophilum over the course of infection. In particular, how transmission is affected by host genotype and pathogen exposure dosage are not well studied. In order to fill in these knowledge gaps, we exposed two divergently selected lines of rainbow trout (ARS‐Fp‐R and ARS‐Fp‐S) to a range of dosages of F. psychrophilum (strain CSF117‐10). We then measured mortality and bacterial shedding to estimate transmission risk at multiple time points since initial infection. As dosage increased, the number of fish shedding and the amount of bacteria shed increased ranging from 0% to 100% and 103 to 108 cells fish−1 h−1, respectively. In addition, we found that disease resistance (survival) was not correlated with transmission risk blocking, in that 67% of fish which shed bacteria experienced no clinical disease. In general, fish mortality began on Day 3, peaked between Days 5–7 and was higher in the ARS‐Fp‐R line. Results from this study could be used to develop epidemiological models and improve disease management, particularly in the context of aquaculture and selective breeding. [ABSTRACT FROM AUTHOR]

Introduced species may exhibit variations in their preferred climatic niches between their native and introduced ranges, which can have important implications for the transferability of distribution models. In the Himalayan ecoregion, little is known about the geographic distribution and climatic niche overlap between native and introduced cold‐water species. Here, we used the COUE (centroid shift, overlap, unfilling, and expansion) framework to explore the invasive potential of rainbow (Oncorhynchus mykiss) and brown (Salmo trutta fario) trout and corresponding climatic niche overlap with native snow trout (Schizothorax plagiostomus and Schizothorax richardsonii) in the Indus and Ganges River basins. Although we found more stability in the climatic niche for O. mykiss (93%) than S. trutta (58%), both species do not conserve their climatic niches in this region (p > 0.05). S. trutta has expanded more toward new environmental conditions (42%) compared to O. mykiss (7%). However, there are still available environmental gaps that O. mykiss and S. trutta can potentially occupy in the future. There was a higher overlap in climatic niches between S. plagiostomus and O. mykiss and between S. richardsonii and S. trutta. Observed shifts in climatic niches of these introduced species can negatively affect the transferability of distribution models, which may underestimate the assessments of habitat suitability for introduced trout in the Himalayas. Our study demonstrates that the information on climatic niche dynamics can inform the model‐building process and improve the transferability and predictive performance to better assess vulnerability of sensitive habitats to introduced species in the Himalayas and elsewhere. Oncorhynchus mykiss is a partially migratory salmonid species, and many migratory populations (known as steelhead) have declined in recent decades in the western United States and Canada. Closely related resident populations (known as rainbow trout) may be an effective resource in the recovery of these declining migratory populations. However, the extent to which different populations of resident rainbow trout produce migratory individuals and how likely these individuals are to return as adults to spawn remains unknown. One limitation to answering these questions is the identification of loci that accurately segregate between migratory and resident populations. To address this limitation, we used existing genomic data from a well‐studied population of O. mykiss from Southeastern Alaska (Sashin Creek) to identify loci that segregate between phenotypes. We then utilized Double Mismatch Allele‐Specific qPCR (DMAS‐qPCR) to genotype 233 smolts out‐migrating from Sashin Creek and 99 returning adult steelhead trout across a five‐year period to determine (a) the origin of out‐migrating smolts and returning adults and (b) to quantify the extent to which the resident population contributes to the migratory population. Our results show that 37.3% of out‐migrating smolts were produced from resident parents, whereas 19.3% of returning adults had resident parents. Ultimately, these results demonstrate that resident populations of rainbow trout produce migrant offspring that successfully complete their migration and return to spawn, increasing population sizes and likely improving genetic diversity. Therefore, conservation efforts should consider landlocked resident populations for producing smolts when developing recovery plans for migratory steelhead populations. [ABSTRACT FROM AUTHOR]

Fish meal (FM) is an essential ingredient in fish feeds, but the thermal treatments used during its production might affect its nutrient availability. Therefore, an in vivo digestibility trial was conducted with three different thermal treated FM, at no heat treatment (control), 70, and 140 °C. A total of 216 rainbow trout (230 ± 6.0 g) were randomly assigned to 12 tanks, and feces samples were collected by stripping. FM was produced by lyophilizing mackerel. The lyophilized FM (94% DM) was then divided into three groups: FM Control, which was not exposed to thermal treatments, and FM 70 °C and FM 140 °C, which were exposed for 1 h to the respective temperatures of its descriptions. A conventional FM-based (FM with 64% CP produced at 110 °C) control diet was used as a basal diet. In the experimental groups, 30% of the basal diet was replaced by one of the tested FMs. No differences in apparent total tract digestibility (ATTD) were observed between FM Control and 70 °C. Protein ATTD of FM 140 °C as well as Arg ATTD were lower compared to FM Control and 70 °C. The same effect was observed for His and Asp ATTD. This study aimed to determine the expression and distribution of gangliosides in specific regions of the brains of rainbow trout (Oncorhynchus mykiss Walbaum) and common carp (Cyprinus carpio L.) with regard to seasonal temperature changes. Seasonal changes in ganglioside expression and distribution within the species were expected. The natural ecosystems of these fishes differ significantly due to their distinct habitat preferences, geographic distributions, and environmental requirements. Based on the fact that the common carp is eurythermic and adapts to a wide range of temperatures, while the rainbow trout is stenothermic and thrives in a narrower temperature range, it was expected that these species would exhibit distinct patterns of ganglioside modification as part of their adaptive response to temperature fluctuations. Immunohistochemistry using specific antibodies for the major brain gangliosides (GM1, GD1a, GD1b, GT1b), along with the Svennerholm method for quantifying sialic acid bound to gangliosides, revealed that cold acclimatization led to an increase in polysialylated gangliosides in the common carp brain and an increase in trisialogangliosides in the rainbow trout brain. Immunohistochemical analysis also identified region-specific changes in ganglioside expression, suggesting specific functional roles in neuronal adaptation. These results supported the hypothesis that the composition and distribution of brain gangliosides change in response to seasonal thermal shifts as part of the adaptive response. The results underscore the importance of gangliosides in neuronal function and adaptation to environmental stimuli, with implications for understanding fish resilience to temperature changes. This study offers valuable insights into species' temperature adaptation, with implications for physiological and ecological management and improved aquaculture practices. Future research could expand the species scale, study molecular mechanisms and regulatory pathways in ganglioside metabolism, and examine ganglioside interactions with membrane proteins and lipids for a deeper understanding of thermal adaptation. The current research evaluated the effects of dietary hesperidin (HSP) on growth parameters, digestive enzyme activities, innate immune markers, and antioxidant responses in rainbow trout, Oncorhynchus mykiss after the 30- and 60-day feeding trial. Then, specimens were subjected to Yersinia ruckeri infection for 14 days. For this, six hundred rainbow trout juveniles (initial weight; 25.49±0.40 g) were fed with different levels of HSP including 0 (HSP0; control), 50 (HSP50), 100 (HSP100), 150 (HSP150), and 200 (HSP200) mg/kg feed. After 60 days, dietary HSP100 supplementation significantly improved growth and feed efficiency indices. The optimal dose of HSP based on the regression test for WG and FCR detected 122 and 131 mg/kg, respectively. At the end of the 30th day, dietary HSP150 supplementation markedly boosted serum lysozyme (LYZ), myeloperoxidase (MPO), complement component C3, and immunoglobulin (Ig) levels, but decreased malondialdehyde (MDA) content. In the same period, dietary administration of HSP at the different concentrations markedly increased complement component C4 (HSP150 and HSP200), superoxide dismutase (SOD), skin mucus alkaline phosphatase (ALP) (HSP100–HSP200), skin mucus LYZ and catalase (CAT) (HSP100 and HSP 150), and skin mucus Ig level (HSP50–HSP200). After 60 days, all dietary HSP supplementation significantly improved lipase, serum C3, glutathione peroxidase (GPX), and skin mucus ALP, LYZ, and Ig levels, but decreased MDA, aspartate aminotransferase (AST), and alanine aminotransferase (ALT) values. After 60 days, dietary administration of HSP150 induced a significant discrepancy in the activity of serum LYZ, MPO, respiratory burst (RB), and lactate dehydrogenase (LDH) compared to the control group. In the same period, dietary HSP supplementation at different levels induced a remarkable increase in digestive protease activity (HSP100), serum ACH50 and CAT activities (HSP100 and HSP150), skin mucus peroxidase and serum SOD levels (HSP100–HSP200), and serum C4 value (HSP50, HSP150, and HSP200), but markedly decreased ALP activity (HSP150 and HSP200). In addition, dietary HSP supplementation, especially HSP150, markedly boosted disease resistance against Y. ruckeri infection. The current data demonstrated that the oral administration of 100–150 mg/kg HSP has a high potential to promote growth performance, disease resistance, and faster induction of immune and antioxidant responses. The current study was carried out to explore the effects of lactofeed (LA) as a multi-strain probiotic and succinic acid (SA) on growth variables, gut lactic acid bacteria count, digestive enzymes, innate immune responses, antioxidant capacity, and resistance against yersinia ruckeri in rainbow trout juveniles (Oncorhynchus mykiss). Three hundred sixty healthy rainbow trout juveniles (13.21±0.41 g) were randomly divided into twelve tanks (300 L) as four experimental groups. They were fed with basal diet (Control; BD), FT1: BD + 1 g/kg LA, FT2: BD + 5 g/kg SA, and FT3: BD + 1 g/kg LA + 5 g/kg SA for eight weeks. According to the results, final weight (FW), weight gain (WG), protein efficiency rate (PER), and feed conversion rate (FCR) revealed a remarkable discrepancy compared to the control group. In addition, dietary inclusion of LA (FT1 and FT3) significantly increased the specific growth rate (SGR). Dietary supplementation of LA+SA (FT3) remarkably improved total bacteria count (TBC) and amylase activity compared to the unsupplemented group. Gut lactic acid bacteria (LAB) count and digestive protease activity in all supplemented fish were remarkably higher than in unsupplemented group. Blood immunological factors including white blood cell (WBC) count, total immunoglobulin content (Ig), and the activity of lysozyme (LYZ), alternative complement pathway (ACH50), and acid phosphatase (ACP) were significantly enhanced in the supplemented groups. Neutrophil (NEU) count, skin mucus Ig and hepatic glutathione peroxidase (GPX) increased in SA (FT2 and FT3) treatments. In addition, monocyte (MON) count and skin mucus LYZ activity were significantly elevated following feeding with the FT3 diet. Other immunological parameters of skin mucus including protease activity, alkaline phosphatase (ALP) activity, and ACH50 activity, as well as hepatic superoxide dismutase (SOD) and catalase (CAT) activities increased in fish fed with diets containing LA and/or SA. Malondialdehyde (MDA) value was remarkably decreased in all supplemented rainbow trout compared to the fish fed with BD. Disease resistance against Y. ruckeri in fish fed with supplemented diets significantly improved with respect to the results obtained in the control specimens. Overall, dietary LA+SA supplementation was beneficial to improve growth performance, gut LAB count, digestive enzyme activities, innate immune responses, antioxidant capacity, and disease resistance in rainbow trout. The aim of the present study was to assess the effects of dietary supplementation with Lactobacillus salivarius (LS) ATCC 11741 and lutein (LU) on immunological, antioxidant, and growth responses, and resistance against Yersinia ruckeri infection in rainbow trout (Oncorhynchus mykiss). There were seven experimental diets containing un-supplemented diet (CTL), L. salivarius at 1 × 106 (LS6) and 1 × 108 (LS8) CFU/g, lutein at 50 (LU50) and 150 (LU150) mg/kg, L. salivarius at 1 × 106 CFU/g plus lutein at 50 g/kg (LS6+LU50), and L. salivarius at 1 × 108 CFU/g plus lutein at 150 g/kg (LS8+LU150). These diets were provided to the fish (N= 735; 14.0±0.38 g). After 60 days of feeding, all experimental treatments exhibited significantly higher growth performance, compared to the CTL treatment. The intestinal protease activity and the intestinal lactic acid bacteria population in the probiotic-treated fish were significantly higher than in the CTL fish. The intestinal lipase activity was only higher in the LS6+LU50 treatment, compared to the CTL treatment. Compared to the CTL treatment, the intestinal total bacterial count, and serum superoxide dismutase significantly increased in the LS6+LU50 and LS8+LU150 treatments. The serum catalase significantly increased in LU150, LS6+LU50, and LS8+LU150 treatments, and serum glutathione peroxidase significantly increased in the lutein-treated fish. All experimental treatments, particularly LS6+LU50 and LS8+LU150, had significantly lower serum malondialdehyde levels, compared to the CTL treatment. The levels of serum alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, and lactate dehydrogenase in LU50, LS6+LU50, and LS8+LU150 treatments were significantly lower than the CTL. All experimental treatments showed significantly higher serum lysozyme (LYZ), complement, blood respiratory burst activity (RB), and skin mucus alkaline phosphatase (ALP), compared to the CTL treatment. The highest serum LYZ, C4, total immunoglobulin (Ig), blood RB activity, and skin mucus peroxidase, ALP, and Ig were observed in the LS6+LU50 and LS8+LU150 treatments. All the experimental treatments, especially LS6+LU50, exhibited significantly lower mortality after the bacterial challenge compared to the CTL. In conclusion, dietary supplementation with 1 × 106 CFU/g L. salivarius and 50 mg/kg lutein can maximally improve growth performance, digestive enzymes, antioxidant parameters, immune responses, intestinal lactic acid bacteria, and resistance against yersiniosis in rainbow trout. The aquaculture sector plays a vital role in global food security, yet it grapples with significant challenges posed by infectious diseases. Piscine lactococcosis is one of the significant threats in rainbow trout aquaculture due to its potential to cause severe economic losses through mortalities, reduced growth rates, and increased susceptibility to other pathogens. It poses challenges in disease management strategies, impacting the sustainability and profitability of rainbow trout farming. The current study focuses on the variations in serum blood parameters of farmed rainbow trout Oncorhynchus mykiss during a lactococcosis outbreak caused by Lactococcus garvieae. Blood samples were collected for biochemical analysis, fish were examined for parasites and bacteria, and DNA from bacterial colonies was PCR‐amplified and sequenced for identification. Overall, 13 biochemical parameters, including proteins, enzymes, lipids, chemicals, and minerals, were measured in serum blood samples from both diseased and healthy fish. The results indicate significant alterations in the levels of these parameters during the outbreak, highlighting the impact of infections on the blood profile of farmed rainbow trout. Urea levels were significantly higher in diseased fish compared to controls, and creatinine, phosphorus, and magnesium also showed similar trends. Alanine aminotransferase and total protein levels were higher in control fish. Chloride levels differed significantly between groups. Iron levels were higher in controls and lower in diseased fish. No significant differences were found in other parameters. This study reveals significant changes in serum blood parameters of rainbow trout during a lactococcosis outbreak caused by L. garvieae. These changes highlight the potential of these parameters as tools for monitoring health status, stress, and aquaculture management. Simple Summary: The salmonid industry is susceptible to opportunistic pathogens particularly affecting fish in early developmental stages. Understanding the immunological capacity during these stages is crucial for effective disease control. The receptor for Interleukin 8 (CXCR1), plays a key role in various inflammatory conditions. This study analyzed the expression of IL-8R and IL8 in rainbow trout larvae in response to bacterial lipopolysaccharide (LPS). The larvae 19 day post-hatching (dph) exhibited strong immune responses after 8 h of LPS stimulation, with IL-8 and omCXCR1 protein expression increasing. The use of specific antisera allowed localization of both proteins in mucosal tissue cells. This study highlights the effectiveness of LPS immersion in activating the innate immune system of trout larvae using IL-8/CXCR1 as immunological markers. This study focused on the immune response capacity of rainbow trout (Oncorhynchus mykiss) larvae by analyzing IL-8/CXCR1 response to lipopolysaccharide (LPS) from Pseudomonas aeruginosa. Previous research demonstrated that LPS from P. aeruginosa acts as a potent immunostimulant in teleost, enhancing pro-inflammatory cytokines. The methodology included in silico analysis and the synthesis and characterization of an omCXCR1-derived epitope peptide, which was used to produce omCXCR1-specific anti98 serum in mice. The research revealed that rainbow trout larvae 19 days post-hatching (dph) exhibited pronounced immune responses post-stimulation with 1 µg/mL of LPS. This was evidenced by the upregulated protein expression of IL-8 and omCXCR1 in trout larvae 2 and 8 h after LPS challenge, as analyzed by ELISA and immunohistochemistry. Furthermore, fluorescence microscopy successfully revealed the colocalization of IL-8 and its receptor in cells from mucosal tissues after LPS challenge in larvae 19 dph. These findings underscore the efficacy of LPS immersion as a method to activate the innate immune system in trout larvae. Glutamate dehydrogenase (GDH) participates in the energy metabolism of proteins and the synthesis of metabolites important for the organism. In this study, GDH enzyme was purified from the liver of rainbow trout (Oncorhynchus mykiss) by 2',5'‐ADP Sepharose 4B affinity chromatography in one step. As a result of this purification process, GDH enzyme was purified 171‐fold with 5.83 U/mg protein‐specific activity. The characterization experiments presented that the storage stability of the purified GDH enzyme was determined as −80°C; optimum temperature 40°C; it was determined that the optimum ionic strength was 100 mM phosphate buffer and the optimum pH was 8.00. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS‐PAGE) and PAGE studies showed that the natural molar mass of the purified GDH enzyme was 346.74 kDa, and the molar mass of its subunits was 53.71 kDa. Km and Vmax values for substrates and coenzymes of GDH enzyme purified from rainbow trout liver were calculated, and the lowest Km value was found in NAD+ (1.86 mM) and the highest Vmax value in NH4+ (1.79 U/mL). The effects of some metal ions, vitamins, and solvents on the activity of the purified GDH enzyme were investigated and also IC50 values and inhibition types. The metal ion with the lowest IC50 value is Ag+ (8.65 ± 1.68 μM), and the vitamin is B6 (0.77 ± 0.04 mM). ABSTRACT In this study, rainbow trout patties (Oncorhynchus mykiss Walbaum, 1792) were treated under a modified atmosphere (MA) (60/40:CO2/N2) packed with microbial transglutaminase enzyme (MTGase) at varying concentrations (0.5% and 1%) and kept for a full day at 2°C ± 2°C. Denser and more complex intermolecular cross‐linking formations between peptides and gel network structures were observed in the first and last scanning electron microscope images of MTGase‐containing groups. MTGase increased the springiness, hardness, shear force, and work‐of‐shear parameters during the storage period in uncooked and cooked patties compared to the control group. The protein, energy, moisture, and amino acid contents were maintained in the MTGase‐containing groups, but using MTGase at higher concentrations was ineffective. Microbiological analyses have shown that MTGase is partially effective on microbial fauna, and the most significant inhibitory effect was determined on total mesophilic aerobic bacteria (TMAB). In all groups, the total amounts of saturated, monosaturated, and polyunsaturated fatty acids remained unchanged on both production and expiration days (p > 0.05). After mixing the burger patty additives into the ground meat, the amounts of sodium, calcium, potassium, and magnesium increased in all groups (p < 0.05); however, a decrease in calcium was observed in the MTGase‐containing groups (p < 0.05) compared to the control group. The higher proportion of the MTGase‐contained group did not exhibit significant differences compared to the lower proportion of the MTGase‐contained group, except for textural characteristics and the TMAB inhibition effect. Aim: To identify the physiological role of the acid‐base sensing enzyme, soluble adenylyl cyclase (sAC), in red blood cells (RBC) of the model teleost fish, rainbow trout. Methods: We used: (i) super‐resolution microscopy to determine the subcellular location of sAC protein; (ii) live‐cell imaging of RBC intracellular pH (pHi) with specific sAC inhibition (KH7 or LRE1) to determine its role in cellular acid‐base regulation; (iii) spectrophotometric measurements of haemoglobin–oxygen (Hb‐O2) binding in steady‐state conditions; and (iv) during simulated arterial‐venous transit, to determine the role of sAC in systemic O2 transport. Results: Distinct pools of sAC protein were detected in the RBC cytoplasm, at the plasma membrane and within the nucleus. Inhibition of sAC decreased the setpoint for RBC pHi regulation by ~0.25 pH units compared to controls, and slowed the rates of RBC pHi recovery after an acid‐base disturbance. RBC pHi recovery was entirely through the anion exchanger (AE) that was in part regulated by HCO3−‐dependent sAC signaling. Inhibition of sAC decreased Hb‐O2 affinity during a respiratory acidosis compared to controls and reduced the cooperativity of O2 binding. During in vitro simulations of arterial‐venous transit, sAC inhibition decreased the amount of O2 that is unloaded by ~11%. Conclusion: sAC represents a novel acid‐base sensor in the RBCs of rainbow trout, where it participates in the modulation of RBC pHi and blood O2 transport though the regulation of AE activity. Today, due to the high nutritional value of fish, consumers' interest in using ready-to-eat foods based on fish is increasing worldwide. However, due to the presence of high amounts of protein and polyunsaturated fatty acids, aquatic products are highly sensitive to oxidation and the growth of microorganisms. The purpose of this research was to investigate the possibility of delaying oxidative and microbial spoilage and maintaining the quality and safety of rainbow salmon burgers during the storage period in the refrigerator using nano-encapsulated peppermint essential oil. Peppermint essential oil was nanoencapsulated by using a combination of gum arabic and soy protein isolate. Then it was added to the fish burger formulation at the levels of 0, 0.05, 0.1 and 0.15%. The values of pH, total volatile nitrogen bases (TVN), peroxide indices (PV) and thiobarbituric acid (TBA), and total count of bacteria were tested during the 12-day storage period at 4°C. Particle size, PDI index, zeta potential and microencapsulation efficiency of peppermint essential oil nanocapsules were 297.4 nm, 0.328 nm, -32.17 mV and 87.24%, respectively. During the storage period, pH, TVB-N, PV and TBA values of burgers increased. Peppermint essential oil nanocapsules showed significant antioxidant and antimicrobial activity and reduced the rate of fat oxidation and bacterial growth in fish burgers. There was a positive correlation between the concentration of essential oil nanocapsules and its antioxidant and antimicrobial activity. The greatest effect of inhibiting the growth of bacteria and reducing the rate of oxidation of fats was observed in the burger containing 0.15% nanocapsules. The results of this research suggest the use of peppermint essential oil nanocapsules at a concentration of 0.15% in order to maintain quality, reduce the rate of oxidation and microbial spoilage in fish burgers. Objective: To maximize harvest of hatchery steelhead Oncorhynchus mykiss and Pacific salmon Oncorhynchus spp. while reducing impacts to wild conspecifics, fishery managers rely on the ability of anglers to distinguish between the two. Fin erosion, including dorsal fin erosion, is a common result of hatchery rearing of juvenile salmonids. Since the 1980s, managers in the Pacific Northwest have utilized adult steelhead dorsal fin height as a regulatory control rule, requiring anglers to release steelhead with dorsal fins over a height threshold. This approach assumes that all or most hatchery fish have shorter dorsal fins than their wild counterparts. This study was designed to test this assumption. Methods: We compared steelhead origin (hatchery or wild) determined using scale analysis and adipose fin clipping (hatchery n = 127, wild n = 71) with origin estimated using dorsal fin height. Result: Overall, a static dorsal fin height threshold (2.25 in, 5.72 cm) correctly identified 90% of hatchery‐origin steelhead but misidentified nearly half (44%) of wild fish as hatchery. Dorsal fin height of known hatchery fish correlated with fish length, indicating a weakness of using a static dorsal height threshold as a management tool. The accuracy of distinguishing wild fish improved, but only marginally (56–76%), using a classification model where dorsal fin height of known origin fish from this study was a continuous predictor of origin. In comparison, adipose fin status correctly identified 97% of unmarked fish as wild. Conclusion: The low accuracy of the classification model and static dorsal fin height thresholds at distinguishing hatchery and wild steelhead due to a strong correlation between dorsal fin height and fork length and the high fork length variability of hatchery origin fish. Given the low cost and accuracy of adipose fin clipping and the potentially high conservation risk associated with misidentification, we recommend discontinuing the use of dorsal fin height as a harvest control rule for steelhead. Objective: We assessed the relative proportions of wild‐ and hatchery‐origin summer steelhead Oncorhynchus mykiss caught by sport anglers in the Deschutes River, Oregon. Our objective was to evaluate if steelhead of wild or hatchery origin were equally or disproportionately captured by sport anglers relative to the overall run composition. Methods: We conducted access‐based angler creel surveys of the Deschutes River steelhead fishery during 2000–2019 and 2021 to observe reported catch of wild and hatchery steelhead. We collated fishery‐independent hatchery‐wild composition data from three locations (two dams and one fish trap) bounding the fishery area we creel sampled. We then used a series of mixed‐effect logistic regression models fit to compare angler catch of wild‐ and hatchery‐origin steelhead relative to their composition at the fishery‐independent sampling locations. Result: The best‐fit model suggests that anglers were more than twice as likely to observe a wild steelhead in their catch as compared with the chance of the fishery‐dependent sampling sites to observe a wild steelhead. Across all years, wild steelhead were 27–31% of the stock composition at the fishery‐independent sampling sites yet averaged 65% of angler catch. Conclusion: Disproportionately high encounter rates of wild‐origin summer steelhead suggest that any adverse impacts associated with catch‐and‐release fishing may be magnified by this effect. We explore some potential mechanisms that could account for this difference and hypothesize that a difference in catchability between wild and hatchery steelhead is the most likely mechanism driving the observed patterns. Regardless of the mechanism, annual catch of wild steelhead is proportionally greater than expectations, given wild steelhead run size compared with that of hatchery steelhead. Changes to ivermectin (IVM [22,23-dihydro avermectin B1a + 22,23-dihydro avermectin B1b]) toxicokinetics (TK) with and without P-glycoprotein (P-gp) inhibition by cyclosporin A (CsA) were examined in rainbow trout (Oncorhynchus mykiss). Rainbow trout were injected with 175 μg/kg 3H-IVM (8.6 μCi/mg IVM) with or without co-administration of 480 μg/kg CsA into the caudal vasculature. Fish were sacrificed at various time points (0.25, 0.5, 1, 3, 24, 48, 96, and 168 h) for organ and tissue sampling (blood, liver, kidney, gill, intestines, brain [5 regions], eye, gonad, and fat) which were analyzed for IVM-derived radioactivity. The IVM concentration decreased over time in blood, liver, kidney, and gill, while concentrations in other tissues remained constant. The highest maximum IVM concentration (Cmax) was found in kidney, followed by liver; the lowest Cmax was found in eye, followed by brain and adipose tissue. The highest % of the administered dose was found in the blood 15 min post-IVM administration, followed by the intestine at 60 min post-IVM administration. P-gp inhibition by CsA did not significantly affect calculated TK parameters (AUC [7.33 ± 0.73 – 11.5 ± 2.5 mg•h/kg], mean residence time [84.7 ± 21 – 125 ± 55 h], T1/2 [58.7 ± 15 – 86.8 ± 38 h], clearance rate [0.0152 ± 0.0033 – 0.0239 ± 0.0024 L/kg•h], or volume of distribution [1.91 ± 0.47 – 2.02 ± 0.33 L/kg]), but resulted in small but significant changes in the % administered dose found in blood and medulla. In recirculating aquaculture systems (RAS), off‐flavours accumulated in fish muscle tissue can be problematic in terms of consumer acceptance and the reputation of farmed fish products. Off‐flavours often give fish earthy, muddy, or other undesirable flavours. Typically, off‐flavours are removed during a depuration period in which fish are fasted and held in clean water. Unfortunately, this causes additional costs and delayed sales, while fish lose weight and show a decrease in lipid content. First, we studied fish growth in a partial RAS (PRAS) where the conditions are very similar to those in depuration with a water exchange rate of 4000 L kg−1 feed, compared to RAS with a 650‐L water kg−1 feed. Rainbow trout (Oncorhynchus mykiss) was reared in both systems. Our aim was to combine the benefits of a higher water exchange rate: the lack of need for biofilters and a lower accumulation of off‐flavours while obtaining stable rearing conditions. Additionally, we studied the effects of moderate feeding and H2O2 addition during depuration. The fish grew faster in a PRAS than in a RAS when fed ad libitum. Thirteen off‐flavour compounds were found in the fish flesh and 11 in the circulating water. The H2O2 addition led to decreased levels of off‐flavours in the tank water and in fish muscle. The results showed no significant differences in off‐flavours between the fed and not‐fed systems, showing that moderate feeding did not prevent a good depuration result. However, the lipid content and the overall fish weight were higher in the fed systems, which suggests more effective depuration. The assessment of the spatiotemporal dynamics of biogenic element accumulation in the bottom sediments of a trout farm located in the northern part of Lake Ladoga was conducted for the first time. Sediment cores were sampled during different hydrological seasons both near the fish cages and at a distance from them. Analyses of total nitrogen and labile phosphorus compounds in the sediments were conducted layer by layer. The dependence of biogenic element content on the organic matter content in the bottom sediments was analyzed. The results showed that the temporal dynamics of biogenic element accumulation in the bottom sediments in the area influenced by the trout farm were closely related to the seasonal feeding intensity of the fish cages. The increase in biogenic element content in the bottom sediments, resulting from the functioning of the fish farm, did not affect the control points. The seasonal formation of an anaerobic reducing environment in the surface layer of sediments may contribute to the increased influx of the studied elements from the bottom sediments into the water column. Ambystoma altamirani is a microendemic amphibian limited to central Mexico, specifically the Bosque de Agua region in the Trans-Mexican Volcanic Belt, renowned for its endemic amphibian species. Anthropogenic activities such as land use change, water pollution, and the introduction of exotic species such as rainbow trout (Oncorhynchus mykiss) have substantially transformed its habitat, creating barriers that fragment it and impeding the mobility of the species and connectivity with other populations. This fragmentation poses challenges, including emerging diseases, inbreeding, limited gene flow, and a loss of genetic diversity, placing Ambystoma altamirani in national and international risk categories. The present study utilized the ENMeval and biomod2 models for environmental niche modeling (ENM) to assess the potential distribution of Ambystoma altamirani in the Bosque de Agua region. The key supporting variables include rivers, lakes, altitude, and a combination of Abies and Pinus forests, while the detrimental factors include urbanization and agriculture. Employing circuit theory (CT) and least-cost path (LCP) methodologies, this research explored functional connectivity, identifying core areas in the central region of Bosque de Agua. As migration distance decreases, the number of corridors facilitating population flow decreases. In the concluding phase, an analysis assessed the coincidence of state and federal Mexican Natural Protected Areas with core areas, revealing a lack of protection. The results of this study could lead to improved knowledge about Ambystoma altamirani, providing valuable tools for helping stakeholders formulate comprehensive strategies for species conservation. ABSTRACT A novel video‐based real‐time system based on AI (artificial intelligence) was developed to detect clinical signs in fish exposed to pathogens. We selected a White Spot Disease model involving rainbow trout as the experimental animal and the parasitic ciliate Ichthyophthirius multifiliis as a pathogen. We compared two identical fish tank systems: one tank was infected by co‐habitation, whereas the other tank was kept non‐infected (sham infection). The two fish tanks were separately video monitored (full top and side view) during the course of infection, during which fish were removed whenever they developed clinical signs (direct visual inspection by the observer). Image analysis (object detection, classification and tracking) was used to track behavioural changes in fish (in every recorded video frame), focusing on movement patterns and spatial localisation. Initially, the two fish groups (infected and non‐infected) exhibited similar behaviour and non‐infected fish did not change behaviour during the 15 d observation period (from 5 d before infection until 10 dpi). At 4, 7, 8, 9 and 10 dpi some infected fish showed clinical signs (equilibrium disturbance, gasping and lethargy) and were removed from the experiment. Anorexia occurred from 5 dpi and a gradual progression of gasping behaviour was noted, whereas the frequency of fish flashing (rubbing/scratching against objects) was low. Equilibrium disturbances and the development of white spots in the skin appeared to be a much later (8–10 dpi at this temperature) indicator of infection. The video analysis showed a general distribution of non‐infected fish in all parts of the fish tank during the entire experiment, whereas infected fish already at 4–5 dpi moved towards higher water currents in the top and bottom positions. This change of fish positioning within the tank appeared as a promising early indicator of infection. The study suggests that continuous monitoring of fish behaviour using AI can potentially optimise the timing of humane endpoints, indicate disease signs earlier and thereby improve animal welfare in both animal experimentation and in aquaculture settings. Systemic inflammation elicits sickness behaviors and fever by engaging a complex neuronal circuitry that begins in the preoptic area of the hypothalamus. Ectotherms such as teleost fish display sickness behaviors in response to infection or inflammation, seeking warmer temperatures to enhance survival via behavioral fever responses. To date, the hypothalamus is the only brain region implicated in sickness behaviors and behavioral fever in teleosts. Yet, the complexity of neurobehavioral manifestations underlying sickness responses in teleosts suggests engagement of higher processing areas of the brain. Using in vivo models of systemic inflammation in rainbow trout, we find canonical pyrogenic cytokine responses in the hypothalamus whereas in the telencephalon and the optic tectum il-1b and tnfa expression is decoupled from il-6 expression. Polyamine metabolism changes, characterized by accumulation of putrescine and decreases in spermine and spermidine, are recorded in the telencephalon but not hypothalamus upon systemic injection of bacteria. While systemic inflammation causes canonical behavioral fever in trout, blockade of bacterial polyamine metabolism prior to injection abrogates behavioral fever, polyamine responses, and telencephalic but not hypothalamic cytokine responses. Combined, our work identifies the telencephalon as a neuronal substrate for brain responses to systemic inflammation in teleosts and uncovers the role of polyamines as critical chemical mediators in sickness behaviors. Triploidy is very useful in both aquaculture and some cultivated plants as the induced sterility helps to enhance growth and product quality, as well as acting as a barrier against the contamination of wild populations by escapees. To use genetic information from triploids for academic or breeding purposes, an efficient and robust method to genotype triploids is needed. We developed such a method for genotype calling from SNP arrays, and we implemented it in the R package named GenoTriplo. Our method requires no prior information on cluster positions and remains unaffected by shifted luminescence signals. The method relies on starting the clustering algorithm with an initial higher number of groups than expected from the ploidy level of the samples, followed by merging groups that are too close to each other to be considered as distinct genotypes. Accurate classification of SNPs is achieved through multiple thresholds of quality controls. We compared the performance of GenoTriplo with that of fitPoly, the only published method for triploid SNP genotyping with a free software access. This was assessed by comparing the genotypes generated by both methods for a dataset of 1232 triploid rainbow trout genotyped for 38,033 SNPs. The two methods were consistent for 89% of the genotypes, but for 26% of the SNPs, they exhibited a discrepancy in the number of different genotypes identified. For these SNPs, GenoTriplo had >95% concordance with fitPoly when fitPoly genotyped better. On the contrary, when GenoTriplo genotyped better, fitPoly had less than 50% concordance with GenoTriplo. GenoTriplo was more robust with less genotyping errors. It is also efficient at identifying low-frequency genotypes in the sample set. Finally, we assessed parentage assignment based on GenoTriplo genotyping and observed significant differences in mismatch rates between the best and second-best couples, indicating high confidence in the results. GenoTriplo could also be used to genotype diploids as well as individuals with higher ploidy level by adjusting a few input parameters. Author summary: To cultivate plants, fish and shellfish more profitable for both farmers and consumers, one can utilize individuals with three chromosome sets instead of the two found in fertile populations that are diploids. These individuals, called triploids, are generally sterile and then often exhibit higher growth and quality of products, such as seedless fruits or better flesh quality for fish and shellfish. To be able to improve performances of the sterile triploids by selective breeding, it is important to know the versions of the genes present in the three chromosome sets of triploids. Until now, few methods existed to identify these three versions, and none have been demonstrated as sufficiently effective. It is the reason why we developed the GenoTriplo software. We demonstrate in this paper the possibility to accurately genotype triploids, as well as how it can be used to reconstruct pedigree information of triploid progeny. Ultimately, we expect that it can help select for reproduction the parents that have the best triploid progeny for the traits of interest such as growth, vigour or product quality. Background: The salmonid pathogen Flavobacterium psychrophilum poses a significant economic threat to global aquaculture, yet our understanding of its genetic and phenotypic diversity remains incomplete across much of its geographic range. In this study, we characterise the genetic and phenotypic diversity of 70 isolates collected from rainbow trout (Oncorhynchus mykiss) and brown trout (Salmo trutta m. fario) from fish farms in the Czech Republic between 2012 and 2019 to compare their genomic content with all draft or complete genomes present in the NCBI database (n = 187). Results: The Czech isolates underwent comprehensive evaluation, including multiplex PCR-based serotyping, genetic analysis, antimicrobial resistance testing, and assessment of selected virulence factors. Multiplex PCR serotyping revealed 43 isolates as Type 1, 23 as Type 2, with sporadic cases of Types 3 and 4. Multi-locus sequence typing unveiled 12 sequence types (ST), including seven newly described ones. Notably, 24 isolates were identified as ST329, a novel sequence type, while 22 were classified as the globally-distributed ST2. Phylogenetic analysis demonstrated clonal distribution of ST329 in the Czech Republic, with these isolates lacking a phage sequence in their genomes. Antimicrobial susceptibility testing revealed a high proportion of isolates classified as non-wild type with reduced susceptibility to oxolinic acid, oxytetracycline, flumequine, and enrofloxacin, while most isolates were classified as wild type for florfenicol, sulfamethoxazole-trimethoprim, and erythromycin. However, 31 isolates classified as wild type for florfenicol exhibited minimum inhibitory concentrations at the susceptibility breakpoint. Conclusion: The prevalence of the Czech F. psychrophilum serotypes has evolved over time, likely influenced by the introduction of new isolates through international trade. Thus, it is crucial to monitor F. psychrophilum clones within and across countries using advanced methods such as MLST, serotyping, and genome sequencing. Understanding the environmental impact of nanoparticle (NP) mixtures is essential to accurately assess the risk they represent for aquatic ecosystems. However, although the toxicity of individual NPs has been extensively studied, information regarding the toxicity of combined NPs is still comparatively rather scarce. Hence, this research aimed to investigate the individual and combined toxicity mechanisms of two widely consumed nanoparticles, zinc oxide (ZnO NPs) and titanium dioxide (TiO2 NPs), using an in vitro model, the RTgill-W1 rainbow trout gill epithelial cell line. Sublethal concentrations of ZnO NPs (0.1 µg mL−1) and TiO2 (30 µg mL−1) and a lethal concentration of ZnO NPs causing 10% mortality (EC10, 3 µg mL−1) were selected based on cytotoxicity assays. Cells were then exposed to the NPs at the selected concentrations alone and to their combination. Cytotoxicity assays, oxidative stress markers, and targeted gene expression analyses were employed to assess the NP cellular toxicity mechanisms and their effects on the gill cells. The cytotoxicity of the mixture was identical to the one of ZnO NPs alone. Enzymatic and gene expression (nrf2, gpx, sod) analyses suggest that none of the tested conditions induced a strong redox imbalance. Metal detoxification mechanisms (mtb) and zinc transportation (znt1) were affected only in cells exposed to ZnO NPs, while tight junction proteins (zo1 and cldn1), and apoptosis protein p53 were overexpressed only in cells exposed to the mixture. Osmoregulation (Na + /K + ATPase gene expression) was not affected by the tested conditions. The overall results suggest that the toxic effects of ZnO and TiO2 NPs in the mixture were significantly enhanced and could result in the disruption of the gill epithelium integrity. This study provides new insights into the combined effects of commonly used nanoparticles, emphasizing the importance of further investigating how their toxicity may be influenced in mixtures. ABSTRACT Flunixin's pharmacokinetics, bioavailability, and plasma protein binding were examined in rainbow trout. The experiment involved 252 rainbow trout (Oncorhynchus mykiss) maintained at 12 ± 0.6°C. Flunixin was administered to rainbow trout via intravascular (IV), intramuscular (IM), and oral routes at a dosage of 2.2 mg/kg. Plasma samples were collected at times 0 (control), 0.25, 0.5, 1, 2, 4, 6, 8, 10, 12, 24, 48, 72, and 96 h. High‐pressure liquid chromatography‐ultraviolet was employed to quantify flunixin concentrations. The elimination half‐life (t1/2ʎz) for flunixin was 8.37 h for IV, 8.68 h for IM, and 8.76 h for oral. The t1/2ʎz was similar between administration groups. The volume of distribution at a steady state and total body clearance were 55.81 mL/kg and 6.83 mL/h/kg, respectively, after IV administration. The mean peak plasma concentration was 6.24 ± 0.41 μg/mL at 4 h for oral administration and 13.98 ± 0.86 μg/mL at 2 h for IM administration. The in vitro protein binding ratio of flunixin in rainbow trout plasma was 96.34 ± 2.29%. The bioavailability of flunixin after oral (25.74%) administration was lower than that after IM (66.70%) administration. Purpose: The purpose of this paper is to assess the microbiological quality of rainbow trout (Oncorhynchus mykiss) and brook trout (Salvelinus fontinalis) from aquaculture farms located within the "Dolina Słupi" (Slupia Valley) Landscape Park, with a focus on regional sustainability practices. The research aims to evaluate water hygiene, the microbiological quality of feed, the prevalence of antibiotic-resistant bacteria, and their impact on the final quality of the farmed trout. This study seeks to contribute to the broader discourse on sustainable aquaculture by highlighting the importance of quality and risk management practices in maintaining food safety and environmental stewardship. Design/methodology/approach: The research employed a comprehensive microbiological analysis of samples collected from trout farms, including external and internal parts of trout, viscera, water, and feed. The study focused on identifying the presence of specific pathogens, such as Staphylococcus aureus, Escherichia coli, Salmonella spp., methicillin-resistant S. aureus, and vancomycin-resistant Enterococcus spp. The approach combined field sampling with laboratory testing to assess the hygienic conditions of the aquaculture environment and the quality of the final fish products. The research focuses on evaluating the microbiological quality of trout from aquaculture farms, with particular attention to local practices and their alignment with sustainability goals. Findings: The study revealed that the trout from the examined farms generally met good microbiological quality standards. Specifically, no methicillin-resistant S. aureus or vancomycin-resistant E. faecium and E. faecalis were detected, indicating effective risk management. However, Salmonella spp. were present in 30.5% of water samples, yet their presence did not significantly affect the contamination levels in fish samples. The highest S. aureus count was found on the skin of rainbow trout (1.5±101 cfu/g), while E. coli was detected on brook trout skin and viscera (11%). Enterococcus spp. were found in 17% of feed samples, but at low concentrations (<10 cfu/g). These results suggest that while overall microbiological quality is satisfactory, ongoing monitoring and adherence to good management practices are crucial for maintaining safety and quality in aquaculture. Research limitations/implications: One limitation of the research is the geographical focus on aquaculture farms within the "Dolina Słupi" Landscape Park, which may limit the generalizability of the findings to other regions or types of aquaculture systems. Future research could expand the scope to include a broader range of aquaculture environments and examine the long-term impact of sustainability practices on fish quality. Various biological traits support the invasive success of different organisms. The osmoregulatory capacity and food preferences of the signal crayfish Pacifastacus leniusculus were experimentally tested to determine if they contribute to its invasive success. The osmotic concentrations of haemolymph were determined after acclimation of the crustaceans to seven salinities from 0 to 20 PSU. Food preferences were tested using Canadian pondweed Elodea canadensis, and rainbow trout Oncorhynchus mykiss. The results showed that the signal crayfish exhibits a hyper-hypoosmotic regulation pattern in the salinity range from 0 to 20 PSU, enabling them to inhabit both freshwater and brackish environments. Furthermore, the study found signal crayfish to have non-specific food preferences, although fish muscle tissue is more beneficial as a source of energy. Both features, osmoregulatory ability and food preferences, can increase the invasive success of this species as it expands into new areas. The ability to survive in higher salinities compared to the coastal waters of the Baltic Sea along the Polish coastline should be considered in targeted management strategies to control the spread of this invasive species. The study investigated the effectiveness of three extruded feeds in the feed conversion index for the rainbow trout (Oncorhynchus mykiss Walbaum, 1792) reared at the Ain Aghbal fish farming station in Morocco. While the biochemical composition (moisture, protein, lipid, and carbohydrate percentages) and energy content (digestible and raw) of feeds A, B, and C are known, these factors alone are insufficient for selecting the optimal feed based on conversion index. The test was conducted over 222 days in tanks supplied with water from Oum Er-Rbia, flowing at 11.04m³/ h, with a water change every half an hour. Initial rearing conditions included a density of 1.58kg/ m³ and a volume-to-flow ratio of 1.05m³/ h. Fish were fed two to three times daily, depending on their growth stage. Kynurenic acid (KYNA) is a metabolite of tryptophan formed on the kynurenine pathway. Its pharmacological effects are relatively well characterized in mammals, whereas its role in fish is poorly understood. Therefore, the aim of the study was to expand the knowledge of KYNA's presence inside a fish's body and its impact on fish development and function. The study was performed on zebrafish larvae and adult rainbow trout. We provide evidence that KYNA is present in the embryo, larva and mature fish and that its distribution in organs varies considerably. A study of KYNA's effect on early larval development suggests that it can accelerate larval maturation, especially under conditions that are suboptimal for fish growth. Moreover, KYNA in concentrations over 1 mM caused morphological impairment and death of larvae. However, long-lasting exposure of larvae to subtoxic concentrations of KYNA does not affect the behavior of 5-day-old larvae kept under standard optimal conditions. We also show that ingestion of KYNA-supplemented feed can lead to KYNA accumulation, particularly in the pyloric caeca of mature trout. Fish are exposed to increased water temperatures and aquatic pollutants, including endocrine-disrupting compounds (EDCs). Although each stressor can disturb fish liver metabolism independently, combined effects may exist. To unveil the molecular mechanisms behind the effects of EDCs and temperature, fish liver cell lines are potential models needing better characterisation. Accordingly, we exposed the rainbow trout RTL-W1 cells (72 h), at 18 °C and 21 °C, to ethynylestradiol (EE2), levonorgestrel (LNG), and a mixture of both hormones (MIX) at 10 µM. The gene expression of a selection of targets related to detoxification (CYP1A, CYP3A27, GST, UGT, CAT, and MRP2), estrogen exposure (ERα, VtgA), lipid metabolism (FAS, FABP1, FATP1), and temperature stress (HSP70b) was analysed by RT-qPCR. GST expression was higher after LNG exposure at 21 °C than at 18 °C. LNG further enhanced the expression of CAT, while both LNG and MIX increased the expressions of CYP3A27 and MRP2. In contrast, FAS expression only increased in MIX, compared to the control. ERα, VtgA, UGT, CYP1A, HSP70b, FABP1, and FATP1 expressions were not influenced by the temperature or the tested EDCs. The RTL-W1 model was unresponsive to EE2 alone, sensitive to LNG (in detoxification pathway genes), and mainly insensitive to the temperature range but had the potential to unveil specific interactions. Aggregation is a fundamental feature of parasite distribution in the host population, but the biological implications of the aggregation indices used to describe the relationships between the populations of parasites and hosts are not evident. It is speculated that the form of distribution in each case is predicated on the host’s varying resistance to the infection, which is hard to control, making it difficult to adequately interpret the index values. This paper examines several cases from trout farms in Russian Karelia to explore the monogenean Gyrodactylus spp. infection in rainbow trout of varying ages. The genetic homogeneity of cage-reared fish and the direct life cycle of the helminths make the relationship between the species more lucid than in natural host-parasite systems. The results give no ground to speak of any specific patterns: as well as in the natural systems, the infection rates in trout vary widely, i.e., the helminth distribution has not become more uniform; the observed distributions in all cases are adequately approximated by the negative binomial model; the positive abundance-occupancy relationships (AORs) and abundance-variance relationships (AVRs) common for parasitic systems apply to the basic infection parameters. The form of the negative binomial distribution is shaped by two parameters—k and θ, the former being a metric of the infection variability, which depends on the host’s individual resistance, and the latter representing the parasites’ reproduction and establishment success rates. A rise in the parameter k indicates increased aggregation and a higher parameter θ points to a more uniform frequency distribution. This work aimed to make a smart biofilm based on tapioca starch/bovine gelatin and roselle calyx extract (RCE)/nanofiller-ZnO. Then the utilization of the biofilm was studied to monitor the rainbow trout fillets. The biofilms were fabricated by casting the starch/gelatin solution with sorbitol/glycerol (3.2g) on casting plates and were dried at 25°C. RCE was added into biofilms (0.5 g for TP/RCE/ZnO-N and 2g for TP/RCE), and the biofilm characteristics, such as thickness, color, and antioxidant properties were investigated. The films containing nanofiller-ZnO/RCE and RCE showed significantly (p<0.05) more antioxidant activity than the neat film which respective values of 18.31 11.42 and 3.1. The thickness of the smart films significantly increased (p<0.05) from 0.08 to 0.13 by the addition of RCE/nanofiller ZnO into the solution. According to the results, a* value was significantly higher (p<0.05) in smart film with nanofiller-ZnO/RCE and RCE. The lowest L* value was obtained in film with nanofiller-ZnO/RCE. At the end of the conservation, the TVB-N value reached 35.7 mg N/100 and with the TVB-N significantly increase (p<0.05), the color properties of the smart films also changed. The color of the biofilm was purple, but the a* value significantly decreased. Therefore, the film based tapioca starch/bovine gelatin/nanofiller ZnO/RCE can be applied as a natural indicator of rainbow trout fillets. [ABSTRACT FROM AUTHOR]

With the rapid and significant cost reduction of next-generation sequencing, low-coverage whole-genome sequencing (lcWGS), followed by genotype imputation, is becoming a cost-effective alternative to single-nucleotide polymorphism (SNP)-array genotyping. The objectives of this study were 2-fold: (1) construct a haplotype reference panel for genotype imputation from lcWGS data in rainbow trout (Oncorhynchus mykiss); and (2) evaluate the concordance between imputed genotypes and SNP-array genotypes in 2 breeding populations. Medium-coverage (12×) whole-genome sequences were obtained from a total of 410 fish representing 5 breeding populations with various spawning dates. The short-read sequences were mapped to the rainbow trout reference genome, and genetic variants were identified using GATK. After data filtering, 20,434,612 biallelic SNPs were retained. The reference panel was phased with SHAPEIT5 and was used as a reference to impute genotypes from lcWGS data employing GLIMPSE2. A total of 90 fish from the Troutlodge November breeding population were sequenced with an average coverage of 1.3×, and these fish were also genotyped with the Axiom 57K rainbow trout SNP array. The concordance between array-based genotypes and imputed genotypes was 99.1%. After downsampling the coverage to 0.5×, 0.2×, and 0.1×, the concordance between array-based genotypes and imputed genotypes was 98.7, 97.8, and 96.7%, respectively. In the USDA odd-year breeding population, the concordance between array-based genotypes and imputed genotypes was 97.8% for 109 fish downsampled to 0.5× coverage. Therefore, the reference haplotype panel reported in this study can be used to accurately impute genotypes from lcWGS data in rainbow trout breeding populations. [ABSTRACT FROM AUTHOR]

Simple Summary: Aquaculture is presented as a viable solution to overfishing and the growing demand for marine products. This study investigates the rearing of rainbow trout (Oncorhynchus mykiss) in a recirculating system at 3000 m above sea level, in Copaquilla, northern Chile. A total of 5000 juvenile trout were transported from the Rio Blanco fish farm over 2100 km. For 20 months, growth parameters were evaluated, including specific growth rate, weight gain, feed conversion, survival, and Fulton's condition factor. The results indicated normal growth and good quality, confirming the feasibility of trout aquaculture at a high altitude. This suggests new opportunities for aquaculture in the Andean region of northern Chile. Given the overexploitation of fisheries and the growing consumption of sea products, aquaculture is emerging as an alternative to meet the demand for protein at regional, national, and global levels. In northern Chile, the foothills of the Andes offer an opportunity for sustainable economic diversification. In this study, results of a rainbow trout (Oncorhynchus mykiss) culture in a recirculation system are presented, analyzing its growth and performance under altitude conditions. The research was carried out in Copaquilla, a small area in the foothills of northern Chile, 3000 m above sea level. Five thousand 15 g juvenile trout were acquired and transported by land from the Rio Blanco fish farm, successfully traveling more than 2100 km. During the 20-month-long culture, several growth parameters were evaluated, including specific growth rate, percentage of weight growth, feed conversion factor, survival, and Fulton's condition factor. All these parameters were within the normal growth range for trout, evidencing good-quality growth of the organisms. The results offer evidence that rainbow trout farming at 3000 m above sea level is viable and presents a viable performance, opening new opportunities for aquaculture in northern Chile.
