The Paramyxoviridae family is a large family of non-segmented negative RNA viruses, some of which are pathogenic to humans or domestic animals. Their small genome comprises six transcription units, including the one coding for the phosphoprotein, which is polycistronic and allows the production of both the phosphoprotein, an essential component for the functioning of the RNA synthesis machinery, and one or more accessory proteins, notably involved in the escape from the immune system. In particular, the genome of most members of the Orthoparamyxovirinae subfamily contains a superimposed gene in the sequence of the gene coding for the phosphoprotein but in a different reading frame. Cells infected with these viruses thus express an accessory protein C by a mechanism of alternative translation initiation. This protein is multi-functional and is involved in regulating viral RNA synthesis, antagonising the innate immune response and budding new viral particles. For some viruses, this protein is a virulence factor. However, we lack information on the structure and mechanisms of action of protein C.The aim of my work was to characterise the structure of the C protein of two paramyxoviruses, alone or in complex with cellular partners, in order to better understand how this protein can act in infected cells.Bioinformatics studies confirmed the conservation of a common architecture to all Orthoparamyxovirinae C proteins and the existence of a disordered N-terminal region and a helix-structured domain in the C-terminal part of the protein. We expressed and purified the C protein of Tupaia paramyxovirus (TPMV) as well as a C-terminal fragment and determined the crystallographic structure of the latter. Despite low sequence similarity, the three-dimensional structure indicates a folding similar to that of the Sendai virus C-terminal domain. Using small-angle X-ray scattering data, we were able to establish an overall model of the entire protein. In collaboration, we have shown that the TPMV C protein inhibits the RNA synthesis machinery of Nipah virus. In particular, it has been shown in the case of measles virus that protein C inhibits RNA synthesis through an interaction with the phosphoprotein. With this in mind, we undertook the structural characterisation of the TPMV phosphoprotein and investigated whether it interacts with the protein C.We used different strategies to express and purify the Nipah virus C protein, alone or in complex with cellular partners. Using a high-throughput screening method, we defined several soluble constructs and used one of these proteins to generate llama nanobodies to use as crystallisation chaperones. In collaboration, we have shown by yeast two-hybrid and TAP-TAG MS methods that NiV C protein binds to the VPS37B (Vacuolar Protein Sorting 37 Homolog B) protein of the ESCRT-I cellular subcomplex of the ESCRT (Endosomal Sorting Complexes Required for Transport) machinery. We co-expressed the protein C with the ESCRT-I subcomplex and showed that the protein C associates with these proteins.In conclusion, my work demonstrates that the C proteins of the Orthoparamyxovirinae subfamily share a common structure and can cross-act with species belonging to different genera suggesting that they have a common origin.; La famille des Paramyxoviridae est une grande famille de virus à ARN négatif non-segmenté dont certains sont pathogènes pour l’homme ou les animaux domestiques. Leur génome de taille réduite comprend six unités de transcription, dont celle codant pour la phosphoprotéine P qui est polycistronique et permet la production, d’une part, de la phosphoprotéine, un composant essentiel pour le fonctionnement de la machine de synthèse d’ARN, et d’autre part, d’une ou de plusieurs protéines accessoires, notamment impliquées dans l’échappement au système immunitaire. En particulier, le génome de la plupart des membres de la sous-famille des Orthoparamyxovirinae contient un gène surimprimé dans la séquence du gène codant pour la phosphoprotéine mais dans un cadre de lecture différent. Les cellules infectées par ces virus expriment ainsi une protéine accessoire C par un mécanisme d’initiation alternative de la traduction. Cette protéine est multi-fonctionnelle, elle est impliquée dans la régulation de la synthèse d’ARN viral, dans l’antagonisme de la réponse immunitaire innée et dans le bourgeonnement de nouvelles particules virales. Pour certains virus, cette protéine est un facteur de virulence. Mais, nous manquons d’information sur la structure et les mécanismes d’action de la protéine C.Mon travail avait pour objectif de caractériser la structure de la protéine C de deux paramyxovirus, seule ou en complexe avec des partenaires cellulaires, afin de mieux comprendre comment cette protéine peut agir dans les cellules infectée.Des études bio-informatiques ont confirmé la conservation d’une architecture commune à l’ensemble des protéines C des Orthoparamyxovirinae et l’existence d’une région N-terminale désordonnée et d’un domaine structuré en hélice- dans la partie C-terminale de la protéine. Nous avons exprimé et purifié la protéine C du paramyxovirus de la toupaye (TPMV) ainsi qu’un fragment C-terminal et nous avons déterminé la structure cristallographique de ce dernier. En dépit d’une faible similarité de séquence, la structure tridimensionnelle indique un repliement similaire à celui du domaine C-terminal du virus Sendai. A partir de données de diffusion de rayons X aux petits angles, nous avons pu établir un modèle d’ensemble de la protéine entière. En collaboration, nous avons montré que la protéine C du TPMV inhibe la machinerie de synthèse d’ARN du virus Nipah. Il a notamment été démontré dans le cas du virus de la rougeole que la protéine C inhibe la synthèse d’ARN par une interaction avec la phosphoprotéine. Dans cette optique, nous avons entrepris la caractérisation structurale de la phosphoprotéine du TPMV et nous avons cherché à déterminer si elle interagit avec la protéine C.Nous avons utilisé différentes stratégies pour exprimer et purifier la protéine C du virus Nipah, seule ou en complexe avec des partenaires cellulaires. Par une méthode de criblage à haut débit, nous avons défini plusieurs constructions solubles et nous avons utilisé une de ces protéines pour générer des nanobodies de lama qui seront utilisés comme chaperons de cristallisation. En collaboration, nous avons montré par les méthodes de double hybride en levure et de TAP-TAG MS que la protéine C du NiV se lie à la protéine VPS37B (Vacuolar Protein Sorting 37 Homolog B) du sous-complexe cellulaire ESCRT-I de la machinerie ESCRT (Endosomal Sorting Complexes Required for Transport I). Nous avons co-exprimé la protéine C avec le sous-complexe ESCRT-I et montré que la protéine C s’associe à ces protéines.En conclusion, mes travaux démontrent que les protéines C des Orthoparamyxovirinae partagent une structure commune et peuvent agir de manière croisée entre espèces appartenant à des genres différents suggérant qu’elles ont une origine commune.

Mitochondrial antiviral-signaling protein (MAVS) joue un rôle central dans la réponse immunitaire innée, où elle permet la coordination de multiples cascades de signalisation conduisant à la production d'Interférons (IFNs) et de cytokines pro-inflammatoires essentiels à la mise en place d'un état antiviral. MAVS est strictement régulée par différents types de modifications post-traductionnelles (PTMs), telles que la phosphorylation et l’ubiquitination. Les PTMs oxydatives réversibles (ox-PTMs) des thiols des cystéines (Cys) jouent un rôle majeur dans la régulation de la signalisation cellulaire. Notre groupe a observé que l’infection par un virus à ARN, le virus Sendai (SeV), induit des Cys ox-PTMs de MAVS. Notre objectif est d'étudier le rôle des Cys de MAVS potentiellement oxydables dans son activation lors d’une infection par un virus à ARN modèle, le SeV. Pour étudier l'implication des modifications Cys sur la fonction de MAVS, des mutants Cys/Alanine (Ala) et Cys/Phénylalanine (Phe) englobant les Cys contenus dans le domaine CARD N-terminal de MAVS et proche de celui-ci ont été générés. Nous avons constaté que dans les cellules A549 déficientes en MAVS transfectées transitoirement par le mutant de toutes les Cys du domaine CARD (C13/20/33/46A), le mutant C79F proche de celui-ci, les mutants C13/33A, C13/20/33A et C13/33/46A, l’activité du promoteur de l’IFNB est inhibée. Nous avons donc généré des cellules A549 déficientes en MAVS transfectées stablement par ces constructions. Nous avons observé que la signalisation antivirale en aval de MAVS a été totalement abolie pendant l’infection SeV dans les cellules exprimant C13/20/33/46A, C79F, C13/20/33A et C13/33/46A. Cette perte de signalisation se traduit par l’absence de phosphorylation de TBK1 et du facteur de transcription IRF3 ainsi que l’inhibition de la production de la protéine antivirale IFIT1. Ultimement cela résulte en une diminution significative de l’activation du promoteur de l’IFNB. Le double mutant C13/33A présente un phénotype d’activité intermédiaire. Globalement, nous proposons un modèle dans lequel au moins 3 résidus Cys dans le domaine CARD de MAVS et/ou la Cys79, dont l’oxydation est essentielle à l’activité de MAVS en régulant probablement ses interactions, sa polymérisation, sa localisation ou ses autres PTMs. Cette étude servira à caractériser un nouveau mécanisme régulateur de la réponse antivirale contre les virus à ARN et à développer des stratégies antivirales visant ce mécanisme., Mitochondrial antiviral-signaling protein (MAVS) plays a central role in the innate immune response, allowing the coordination of multiple signaling cascades ultimately leading to the production of interferons (IFNs) and pro-inflammatory cytokines that are essential to mount an antiviral state. MAVS is tightly regulated by different types of post-translational modifications (PTMs) such as phosphorylation and ubiquitination. Reversible oxidative PTMs (ox-PTMs) of cysteine thiols (Cys) play a major role in the regulation of cell signaling. Our group observed that infection with an RNA virus, Sendai virus (SeV) induces ox-PTMs of MAVS Cys. Our goal is to investigate the role of the Cys of MAVS that are potentially oxidized in its activity during infection with a model of RNA virus, SeV. In order to determine the involvement of Cys modifications on MAVS function, Cys/Alanine (Ala) and Cys/Phenylalanine (Phe) mutants encompassing Cys contained within and close to the N-terminal CARD domain of MAVS were generated. We found that in MAVS-deficient A549 cells transiently transfected with the mutant of all Cys in the CARD domain (C13/20/33/46A), the C79F mutant close to it, the C13/33A, C13/20/33A and C13/33/46A mutants, IFNB promoter activity is inhibited. We therefore generated MAVS-deficient A549 cells stably transfected with these constructs. We observed that antiviral signaling downstream of MAVS was completely abolished during SeV infection in cells expressing C13/20/33A, C79F, C13/20/33A and C13/33/46A. This dampening of the capacity of signaling include absence of phosphorylation of TBK1 and the transcription factor IRF3 as well as the inhibition of the production of antiviral protein IFIT1. These events ultimately result in a significant decrease of IFNB promoter activation. The C13/33A double mutant exhibits an intermediate phenotype of antiviral activity. Altogether, we propose a model in which at least 3 Cys residues within the CARD domain of MAVS and/or the Cys79, whose potential reversible oxidation is essential for MAVS antiviral response, probably by regulating its interactions, polymerization, localization or its other PTMs. This study will help to characterize a new regulatory mechanism of the antiviral response against RNA viruses and to develop antiviral strategies targeting this mechanism.

Staphylococcus aureus are opportunistic pathogenic bacteria in mammals, responsible for the vast majority of bacterial infections of the skin tissue in humans. The production of a wide variety of virulence factors allows this bacterial species to gain access to the deepest layers of the skin and to cause infections with a wide range of clinical manifestations. These infections represent a real public health issue due to their high frequency and the generalized emergence of S. aureus strains resistant to antibiotherapy. In humans, S. aureus skin infection causes a local inflammatory response characterized by the recruitment of phagocytic cells, such as polymorphonuclear neutrophils (PNN) and monocytes/macrophages (MO/MΦ) at the infection site. These immune cells, along with other cell types in the skin, contribute to the elimination of the pathogen from the infected tissue. However, S. aureus has developed numerous strategies to evade, deactivate, or kill host immune cells. This includes the transition from the planktonic lifestyle to the biofilm lifestyle, the latter corresponding to the growth of microbes in bacterial communities. Biofilms confer to bacteria an increased resistance to antimicrobial agents and to attacks by the host immune system. The work presented in this manuscript has aimed to better understand how bacteria organized in biofilms are able to resist to attacks by the host innate immune system, also called “immune evasion”, in the context of skin infections.An innovative experimental technique using the mouse ear pinna infection model was set up. Using confocal microscopy, this approach made it possible to monitor and quantify by intravital imaging the dynamics of the innate anti-biofilm immune responses, both at the tissue and cellular level. We thus demonstrated a considerable early recruitment of phagocytes in the skin tissue of the ear pinna, with cellular recruitment dynamics specific to the biofilm form. This so-called “biofilm” phenotype was then characterized using the flow cytometry approach and made it possible to show that PNN represent the major population of phagocytes detected in the tissue of the ear tissue. The analysis of the functional properties of these recruited cells made it possible to identify an activation kinetic specific to the biofilm form during the first days of the infection. This was characterised by a state considered as “over-activated” of the PNN at the 48-hour time point after the infection. Finally, an exploratory study of the interactions between dendritic cells and S. aureus carried out in vitro made it possible to highlight the first differences in the responses induced by the planktonic or biofilm form of growth. Ultimately, a better understanding of these first stages of the innate immune response would allow us to contribute to the development of alternative therapeutic strategies to antibiotics such as immunomodulatory therapies, in order to better fight against S. aureus biofilm infections.; Staphylococcus aureus est une bactérie pathogène opportuniste chez les mammifères, et responsable de la grande majorité des infections bactériennes du tissu cutané chez l'Homme. La production d'une grande variété de facteurs de virulence va permettre à cette espèce bactérienne d’atteindre les couches les plus profondes de la peau et de provoquer des infections présentant une large gamme de manifestations cliniques. Ces infections représentent un véritable enjeu de santé publique en raison de leur fréquence élevée et de l'émergence généralisée de souches de S. aureus résistantes à l’antibiothérapie. Chez l’Homme, une infection cutanée à S. aureus est à l’origine d’une réaction inflammatoire locale caractérisée par le recrutement de cellules phagocytaires telles que des polynucléaires neutrophiles (PNN) et des monocytes/macrophages (MO/MΦ) au site d’infection. Ces cellules immunitaires, avec d’autres types cellulaires de la peau, vont contribuer à l’élimination de l’agent pathogène du tissu infecté. Cependant, S. aureus a développé de nombreuses stratégies pour contourner, désactiver ou même tuer les cellules immunitaires de l'hôte, notamment en effectuant la transition du mode de vie planctonique vers le mode de vie biofilm. Ce dernier correspond à la forme bactérienne communautaire. Il confère aux bactéries une résistance accrue aux agents antimicrobiens et aux attaques du système immunitaire de l’hôte. Le travail présenté dans ce manuscrit a eu pour objectif de mieux comprendre comment les bactéries organisées en biofilm sont capables de résister aux attaques du système immunitaire inné de l’hôte, encore appelé « immuno-échappement », dans le contexte d’une infection cutanée.Une approche expérimentale innovante utilisant le modèle d’infection du pavillon de l’oreille de la souris a été mise en place. En utilisant l’approche de microscopie confocale, nous avons pu suivre et quantifier par imagerie intravitale, la dynamique de la réponse immunitaire innée anti-biofilm aussi bien à l’échelle tissulaire que cellulaire. Nous avons ainsi démontré un recrutement précoce important de phagocytes au niveau du tissu cutané de l’oreille, avec une dynamique de recrutement spécifique à la forme biofilm. Ce phénotype dit « biofilm » a ensuite été caractérisé en utilisant l’approche de cytométrie en flux, et a permis de démontrer que les PNN représentent la population majeure de phagocytes recrutés au niveau du tissu de l’oreille. L’analyse des propriétés fonctionnelles de ces cellules a permis d’identifier une cinétique d’activation spécifique à la forme biofilm durant les premières jours de l’infection, avec un état qualifié de « suractivé » des PNN au temps 48h après l’infection. Enfin, une étude exploratoire des interactions entre des cellules dendritiques et S. aureus réalisée in vitro a permis de mettre en évidence des premières différences dans les réponses induites par la forme planctonique ou biofilm. A terme, une meilleure connaissance de ces premières étapes de la réponse immunitaire innée nous permettra de contribuer à la mise au point de stratégies thérapeutiques alternatives à l’antibiothérapie pour lutter contre les infections par le biofilm de S. aureus, notamment en ayant recours à des thérapies immunomodulatrices.

The Bacillus Cereus (B cereus) group consists of Gram-positive bacteria, commonly isolated from the environment. Members of the B. cereus group are spore-forming bacteria commonly associated with food poisoning and intestinal infections and represent the third cause of food poisoning in Europe and the second in France. The most-studied members of this group are B. anthracis, B. cereus sensu stricto (Bc), and B. thuringiensis (Bt) that are well known for their pathogenic activity. Bt is characterized by the production, during its sporulation, of a crystal containing insecticidal Cry toxins. Thanks to this entomopathogenic activity, bioinsecticides based on spores and crystals of Bt are used to specifically kill lepidopteran pests. Bt bioinsecticides represent more than 50% of the global market share of biopesticides. Upon ingestion of Bt bioinsecticides (spores and toxin crystals), Cry toxins are released from the crystal and destroy the insect gut epithelium allowing spores to invade the internal milieu. Then spores’ germination in the hemolymph lead to the pest death by septicemia in less than 3 days. However, the increasing environmental dispersion of Bt products raises the question about their potential risks for non-target animals.Using Drosophila melanogaster (a non-target organism), it has been previously shown that Bt vegetative cells are rapidly cleared from the intestine. This elimination is due to the activation of the local innate immune response. However, few studies suggest that Bt spores can persist in human and rodent intestines. To understand why and how Bt/Bc spores persist in the gut of non-target organisms and the implication of the innate immune response in fighting spores, I have used two powerful models: Drosophila melanogaster and mice. First, I have shown that Bt/Bc spores can persist, after acute ingestion, up to 10 days in the Drosophila midguts and at least 5 days in mice small intestines., Then, using an innovative tool, generated in my lab, consisting of red fluorescent spores that switch in green fluorescent once germinated (corresponding to vegetative bacteria), I have shown that spores accumulate mainly in the posterior midgut of Drosophila and the posterior small intestine of mice where they can further germinate. Then, I confirmed these data using quantitative monitoring of spores vs. vegetative bacteria accumulation in vivo in the intestines of Drosophila and mice. Altogether, my data suggest that, in the posterior regions of the Drosophila midgut (and the small intestine of mice), the innate immune response is inefficient to promptly eliminate spores and germinated bacteriaIn Drosophila midgut, the local innate immune response (i.e. the production of Reactive Oxygen Species (ROS) and Anti-Microbial Peptides (AMPs)) is mainly mounted in the anterior regions. I have shown that, unlike vegetative cells, spores do not trigger ROS and AMPs production. Strikingly, I have even observed that spores inhibit AMPs production. Interestingly, I have found a transcriptional induction of negative regulators (e.g. PGRP-SC1, -SC2, and -LB) of the immune signaling pathways in the posterior region of the midgut in response to spore ingestion. Then, using combinations of mutants of the two immune pathways, Imd and Toll, I have been the first to demonstrate that both immune pathways coexist in the Drosophila midgut, and may contribute either additively or synergistically to fight spore infections., Le groupe Bacillus cereus (B. cereus) regroupe des bactéries Gram positives issues de l’environnement. Ces bactéries sont capables de sporuler quand les conditions sont défavorables. Les membres de ce groupe peuvent être impliqués dans des intoxications alimentaires représentant ainsi la troisième cause d'intoxication alimentaire en Europe et la deuxième en France. L’espèce B. thuringiensis (Bt) appartient au groupe B. cereus et se caractérise par la production de cristaux parasporaux contenant des toxines Cry insecticides avec une forte spécificité d’action. Ainsi, les spores et les cristaux de Bt sont utilisés comme bioinsecticides pour lutter spécifiquement contre les larves de lépidoptères ravageurs. Les produits à base de Bt représentent plus de 50 % de marché mondiale des biopesticides. Une fois les spores et les cristaux ingérés par un organisme cible (les larves de lépidoptères), les toxines Cry contenues dans le cristal sont libérées et détruisent l'épithélium intestinal. Ensuite, le passage et la germination des spores dans l'hémolymphe entraînent une mort rapide du ravageur par septicémie. L’utilisation accrue de ces bioinsecticides Bt pose la problématique des risques potentiels engendrés sur les organismes non ciblés.En utilisant le modèle Drosophila melanogaster (un organisme non-cible de Bt), il avait été précédemment montré que les formes végétatives de Bt sont rapidement éliminées de l'intestin. Cette élimination est corrélée avec l'activation de la réponse immunitaire innée locale. Cependant, des études disparates suggéraient que les spores de Bt pouvaient persister dans les intestins des mammifères. Afin d’étudier le comportement de ces spores dans l'intestin des organismes non ciblés et l’implication de la réponse immunitaire innée, j’ai utilisé deux modèles de laboratoire : Drosophila melanogaster et la souris. En premier lieu, j'ai pu montrer qu’après une ingestion ponctuelle, les spores persistent jusqu'à 10 jours dans l'intestin moyen des drosophiles et au moins 5 jours dans l’intestin grêle des souris. Ce résultat m’a conduit à étudier le devenir de ces spores dans l’intestin. Pour cela, j'ai utilisé un outil généré au laboratoire où les spores fluorescent en rouge qui une fois germées en cellules végétatives acquièrent une fluorescence verte. En utilisant ces spores marquées, j'ai montré que ces dernières s'accumulent et germent préférentiellement dans les parties postérieures de l’intestin moyen de drosophile et de l’intestin grêle de souris. J’ai ensuite confirmé ces résultats qualitatifs par une approche quantitative en dénombrant spores et bactéries végétatives dans les différentes régions de l’intestin. Ainsi, toutes mes données suggèrent que la réponse immunitaire innée au niveau de la région postérieure de l’intestin moyen/grêle est inefficace pour éliminer rapidement les spores et les bactéries germées.Ensuite, je me suis donc focalisée sur la réponse immunitaire innée locale. Dans l'intestin moyen de la drosophile, cette réponse est localisée principalement dans les régions antérieures et se manifeste par une production d’espèces réactives de l’oxygène (ROS) et de peptides antimicrobiens (AMP). Mes résultats montrent que contrairement aux cellules végétatives, les spores n’induisent pas la production de ROS et d’AMP par l’épithélium intestinale. Etonnamment, j’ai même observé que la production d’AMP est réprimée. J’ai ensuite démontré que l’ingestion de spores induit l’expression de gènes codant pour des régulateurs négatifs (PGRP-SC1, -SC2 et -LB) de la réponse immunitaire innées dans la région postérieure de l’intestin moyen. Enfin, en utilisant des combinaisons de mutants des deux voies de signalisation immunitaires, Imd et Toll, j’ai pu caractériser pour la première fois leur coexistence dans l'intestin moyen de la drosophile et leur contribution de manière additive ou synergique aux infections par des spores du groupe B. cereus.

Dendritic cells (DCs) are a heterogeneous population that includes pDC, cDC1, cDC2 and the recently described Axl+ DC in the human blood. However, it remains to be evaluated whether Axl+ DCs possess specific functions after exposure to a pathogen. They constitutively express Siglec-1, involved in HIV-1 capture, which prompted us to study their relationship with the virus. We showed that only Axl+ DC were susceptible to infection in a Siglec-1-dependent manner. They produce infectious viruses that accumulate in compartments similar to those observed in macrophages. Moreover, after activation, Axl+ DC are able to capture HIV-1 via Siglec-1 and transmit the virus to CD4+ T cells, in absence of viral replication. Moreover, Axl+ DC exhibit functional specificities in HIV-1 sensing and response. We have shown that they are more efficient than cDC2 in inducing an interferon response and an inflammatory response. After exposure to HIV-1, these responses are engaged in different Axl+ DC. In contrast, Axl+ DCs that produce new viruses show simultaneous activation of both responses. Our results suggest that this functional diversity is due to different virus detection mechanisms involving either endosomal TLR or the STING pathway.Our work describes for the first time a function of Axl+ DC in an infectious context. It illustrates the complexity of the relationships between DC subtypes and HIV-1, which must be analyzed to better understand the activation processes of the immune response., Les cellules dendritiques (DC) constituent une population hétérogène qui comporte les pDC, les cDC1, les cDC2 et les Axl+ DC récemment décrites dans le sang. Cependant, il reste à évaluer si les Axl+ DC possèdent des fonctions spécifiques après exposition à un pathogène. Elles expriment constitutivement Siglec-1, impliqué dans la capture du VIH-1, ce qui nous a incité à étudier leurs relations avec le VIH-1. Nous avons montré que seules les Axl+ DC étaient susceptibles à l’infection de manière dépendante de Siglec-1. Elles produisent des virus infectieux qui s’accumulent dans des compartiments semblables à ceux observés chez les macrophages. De plus, après activation, les Axl+ DC sont capables de capturer le VIH-1 via Siglec-1 et de le transmettre aux lymphocytes T CD4+, sans réplication virale. Les Axl+ DC possèdent également des spécificités fonctionnelles dans la détection et la réponse au VIH-1. Nous avons montré qu’elles étaient plus efficaces que les cDC2 à induire une réponse interféron et une réponse inflammatoire. Après exposition au VIH-1, ces réponses sont engagées dans des Axl+ DC différentes. En revanche, les Axl+ DC qui produisent de nouveaux virus présentent une activation simultanée de ces deux réponses. Nos résultats suggèrent que cette diversité fonctionnelle serait due à des mécanismes de détection du virus différents impliquant soit les TLR endosomaux, soit la voie STING. Nos travaux décrivent pour la première fois une fonction des Axl+ DC dans un contexte infectieux. Ils illustrent la complexité des relations entre les sous-types de DC et le VIH-1 dont l’analyse est nécessaire pour mieux comprendre les processus d’activation de la réponse immunitaire.

The integrated innate immune features of the calcareous egg and its contents are a critical underpinning of the remarkable evolutionary success of the Aves clade. Beginning at the time of laying, the initial protective structures of the egg, i.e., the biomineralized eggshell, egg-white antimicrobial peptides, and vitelline membrane, are rapidly and dramatically altered during embryonic development. The embryo-generated extra-embryonic tissues (chorioallantoic/amniotic membranes, yolk sac, and associated chambers) are all critical to counteract degradation of primary egg defenses during development. With a focus on the chick embryo (Gallus gallus domesticus), this review describes the progressive transformation of egg innate immunity by embryo-generated structures and mechanisms over the 21-day course of egg incubation, and also discusses the critical interplay between autonomous development and maternal anticipation.

Avian pathogenic Escherichia coli (APEC) cause severe respiratory and systemic disease in chickens, commonly termed colibacillosis. Early immune responses after initial infection are highly important for the outcome of the infection. In this study, the early interactions between GFP-expressing APEC strains of serotypes O1:K1:H7 and O2:K1:H5 and phagocytic cells in the lung of CSF1R-reporter transgenic chickens were investigated. CSF1R-reporter transgenic chickens express fluorescent protein under the control of elements of the CSF1R promoter and enhancer, such that cells of the myeloid lineage can be visualized in situ and sorted. Chickens were separately inoculated with APEC strains expressing GFP and culled 6 h post-infection. Flow cytometric analysis wasperformed to phenotype and sort the cells that harbored bacteria in the lung, and the response of the sorted cells was defined by transcriptomic analysis. Both APEC strains were mainly detected in CSF1R-transgeneneg (CSF1R-tgneg) and CSF1R-tglow MHC IIneg MRC1L-Bneg cells and low numbers of APEC were detected in CSF1R-tghigh MHC IIpos MRC1L-Bpos cells. Transcriptomic and flow cytometric analysis identified the APECpos CSF1R-tgneg and CSF1R-tglow cells as heterophils and the APECpos CSF1R-tghigh cells as macrophages and dendritic cells. Both APEC strains induced strong inflammatoryresponses, however in both CSF1R-tgneg/low and CSF1R-tghigh cells, many immune related pathways were repressed to a greater extent or less activated in birds inoculated with APEC O2-GFP compared to APEC O1-GFP inoculated birds. Comparison of the immune pathways revealed the aryl hydrocarbon receptor (AhR) pathway, IL17 and STAT3 signaling, heterophil recruitment pathways and the acute phase response, are modulated particularly post-APEC O2-GFP inoculation. In contrast to in vivo data, APEC O2-GFP was more invasive in CSF1R-tghigh cells in vitro than APEC O1-GFP and had higher survival rates for up to 6 h post-infection. Our data indicate significant differences in the responses induced by APEC strains of prevalent serotypes, with Q14 important implications for the design and interpretation of future studies. Moreover, we show that bacterial invasion and survival in phagocyte populations in vitro is not predictive of events in the chicken lung.

La pasteurellose est la première cause de mortalité des femelles en élevage cunicole. Lors du projet RELAPA, 953 lapins ont été inoculés à l’âge de 6 semaines avec une souche de Pasteurella multocida (Pm) pyogène et suivis pendant 14 jours. La réponse des animaux était très variable, avec 7% de lapins résistants et 11% d’animaux très sensibles. Des numérations formules sanguines ont été réalisées 14 jours après l’inoculation sur 574 lapins inoculés et 28 témoins. Des différences significatives de composition du sang en globules blancs, rouges et plaquettes ont été observées selon la note de résistance attribuée à l’animal. On constate chez les animaux sensibles un nombre de globules rouges inférieur, résultant probablement de l’activité hémolytique et hémorragique de Pm. Les animaux sensibles ont une proportion plus importante de monocytes, neutrophiles, éosinophiles impliqués dans la réponse immunitaire innée et la réponse inflammatoire, et une proportion inférieure de lymphocytes (impliqués dans la réponse immunitaire adaptative) comparés aux animaux très résistants et aux témoins. Ces animaux sensibles semblent donc ne pas réussir à monter une réponse immunitaire leur permettant de contrôler l’infection. L’effet significatif du père sur le nombre des différents types de leucocytes suggère une variabilité génétique dans la réponse hématologique à l’infection.

Mycobacterium leprae, the causative agent of leprosy, is unique amongst human pathogens in its capacity to produce the virulence factor phenolic glycolipid (PGL)-I. In addition to mediating bacterial tropism for neurons, PGL-I interacts with Complement Receptor (CR)3 on macrophages (MPs) to promote infection. We demonstrate here that PGL-I binding to CR3 also enhances bacterial invasion of both polymorphonuclear neutrophils (PMNs) and dendritic cells (DCs). Moreover, in all cell types CR3 engagement by PGL-I activates the Syk tyrosine kinase, inducing calcineurin-dependent nuclear translocation of the transcription factor NFATc. This selectively augments the production of IL-2 by DCs, IL-10 by PMNs and IL-1β by MPs. In intranasally-infected mice PGL-I binding to CR3 heightens mycobacterial phagocytosis by lung PMNs and MPs, and stimulates NFATc-controlled production of Syk-dependent cytokines. Our study thus identifies the CR3-Syk-NFATc axis as a novel signaling pathway activated by PGL-I in innate immune cells, rewiring host cytokine responses to M. leprae.

International audience; Lipopolysaccharides (LPS) are potent activator of the innate immune response through the binding to the myeloid differentiation protein-2 (MD-2)/toll-like receptor 4 (TLR4) receptor complexes. Although a variety of LPSs have been characterized so far, a detailed molecular description of the structure-activity relationship of the lipid A part has yet to be clarified. Photosynthetic Bradyrhizobium strains, symbiont of Aeschynomene legumes, express distinctive LPSs bearing very long-chain fatty acids with a hopanoid moiety covalently linked to the lipid A region. Here, we investigated the immunological properties of LPSs isolated from Bradyrhizobium strains on both murine and human immune systems. We found that they exhibit a weak agonistic activity and, more interestingly, a potent inhibitory effect on MD-2/TLR4 activation exerted by toxic enterobacterial LPSs. By applying computational modeling techniques, we also furnished a plausible explanation for the Bradyrhizobium LPS inhibitory activity at atomic level, revealing that its uncommon lipid A chemical features could impair the proper formation of the receptorial complex, and/or has a destabilizing effect on the pre-assembled complex itself.

Escherichia coli is one of the major pathogens causing mastitis in dairy cattle. Yet, the factors which mediate the ability for E. coli to develop in the bovine mammary gland remain poorly elucidated. In a mouse model, infections induced by the reference mastitis E. coli P4 showed a strong colonisation of the mammary gland, while this strain had a low stimulating power on cells of the PS bovine mammary epithelial cell line. In order to understand if such a reduced response contributes to the severity of infection, a library of random mutants of P4 strain was screened to identify mutants inducing stronger response of PS cells. Among hyper-stimulating mutants, six were altered in genes involved in biosynthesis of lipopolysaccharide (LPS) and had lost their O-polysaccharide region, suggesting that the presence of O-antigen impairs the response of PS cells to LPS. Using purified smooth (S) and rough (R) fractions of LPS, we showed that the R-LPS fraction induced a stronger response from PS cells than the smooth LPS fraction. Biological activity of the S-LPS fraction could be restored by the addition of recombinant bovine CD14 (rbCD14), indicating a crucial role of CD14 in the recognition of S-LPS by Mammary Epithelial Cells (MEC). When S-LPS and R-LPS were injected in udder quarters of healthy lactating cows, an inflammation developed in all infused quarters, but the S-LPS induced a more intense pro-inflammatory response, possibly in relation to sizeable concentrations of CD14 in milk. Altogether, our results demonstrate that the O-antigen modulates the pro-inflammatory response of MEC to LPS, that S-LPS and R-LPS trigger different responses of MEC and that these responses depend on the presence of CD14.

Chronic hepatitis B virus (HBV) infection is a major cause of liver disease and cancer worldwide. The mechanisms of viral genome sensing and the evasion of innate immune responses by HBV infection are still poorly understood. Recently, the cyclic GMP-AMP synthase (cGAS) was identified as a DNA sensor. In this PhD work, we aimed to investigate the functional role of cGAS in sensing of HBV infection and elucidate the mechanisms of viral evasion. We performed functional studies including loss- and gain-of-function experiments combined with cGAS effector gene expression profiling in an HBV infection-susceptible cell culture model. Collectively, our data show that (1) the cGAS-STING pathway exhibits robust antiviral activity against HBV infection including reduction of viral cccDNA levels; (2) naked HBV genomic rcDNA is sensed in a cGAS-dependent manner whereas packaging of the viral genome during infection abolishes host cell recognition of viral nucleic acids; (3) HBV infection down-regulates the cGAS/STING pathway actors as well as innate immune effector gene expression in vitro and vivo. Overall, this work led to describing new aspects of the complex interaction between HBV and the DNA sensor cGAS in hepatocytes.; Le virus de l’hépatite B (HBV) est l’agent étiologique de l’hépatite B. Ce virus est responsable d’hépatite chronique B, de cirrhose et de cancer du foie au niveau mondial. L’absence d’activation de la voie Interféron (IFN) suite à l’infection par HBV est encore mal comprise. Récemment, le senseur cellulaire cytosolic GMP-AMP synthase (cGAS) a été décrit comme un senseur efficace de DNA double brin possédant également une activité antivirale envers des virus à ADN et à ARN. Le but de mes travaux de thèse a été de contribuer à la compréhension des relations existants entre le HBV et cGAS, à des stades précoces et tardifs de l’infection HBV en utilisant des expériences de perte- et gain- de function ainsi que du profilage génomique des génes apparentés à cGAS dans un modéle cellulaire permissif au HBV. Mes travaux ont démontré (1) que cGAS exerce une forte activité antivirale envers le HBV incluant une réduction de la forme nucléaire du génome, le cccDNA; (2) alors que le rcDNA génomique nu est reconnu par la voie cGAS/STING et induit une réponse IFN efficace, la nucléocapside virale protège le DNA génomique viral et l’empêche d’être détecté par la réponse immunitaire innée; et (3) que l’infection par HBV diminue l’expression des acteurs de la voie cGAS-STING et des gènes impliqués dans la réponse immunitaire innée in vitro et in vivo. Ce dernier point met en lumière le rôle de cGAS dans un nouveau mécanisme d’échappement du HBV au système immunitaire inné dans les cellules hépatocytaires et dans ce mécanisme.

Toll-like receptors (TLRs) are major players of the innate immune system. Once activated, they trigger a signalling cascade that leads to NF-κB translocation from the cytoplasm to the nucleus. Single cell analysis shows that NF-κB signalling dynamics are a critical determinant of transcriptional regulation. Moreover, the outcome of innate immune response is also affected by the cross-talk between TLRs and estrogen signalling. Here, we characterized the dynamics of TLR5 signalling, responsible for the recognition of flagellated bacteria, and those changes induced by estradiol in its signalling at the single cell level. TLR5 activation in MCF7 cells induced a single and sustained NF-κB translocation into the nucleus that resulted in high NF-κB transcription activity. The overall magnitude of NF-κB transcription activity was not influenced by the duration of the stimulus. No significant changes are observed in the dynamics of NF-κB translocation to the nucleus when MCF7 cells are incubated with estradiol. However, estradiol significantly decreased NF-κB transcriptional activity while increasing TLR5-mediated AP-1 transcription. The effect of estradiol on transcriptional activity was dependent on the estrogen receptor activated. This fine tuning seems to occur mainly in the nucleus at the transcription level rather than affecting the translocation of the NF-κB transcription factor.

Vaccination is considered as one of the best therapeutic intervention to fight against infectious diseases. However, we still fail to develop protective vaccines for diseases such as AIDS or B hepatitis, which highlights our lack of knowledge of immune processes.For this thesis, we relied on a non-human primate model associated wih a vaccinia virus living attenuated vaccine derived called MVA (Modified Vaccinia virus Ankara). We deciphered innate, but also adaptive immune response using this model.MVA induces an important inflammatory reaction at the injection site and in the blood after intramuscular, subcutaneous, and intradermal administration. However, the magnitude of this immune reaction, as well as cellular and molecular effectorsin varies depending on the administration route. The consequences are important and lead to different adaptive immune profiles, with Th2 oriented response after subcutaneous route, and Th1 oriented response after intramuscular and intradermal route. It allowed us to highlight several cellular and molecular signatures that might modulate immune response in our model., La vaccination est considérée aujourd’hui comme une des méthodes les plus efficaces de se prémunir des maladies infectieuses. Respiratory syncytial virus (RSV) causes severe bronchiolitis in infants worldwide. The immunological factors responsible for RSV susceptibility in infants are poorly understood. Here, we used the BALB/c mouse model of neonatal RSV infection to study the mechanisms leading to severe disease upon reexposure to the virus when adults. Two major deficiencies in neonatal lung innate responses were found: a poor DCs mobilization, and a weak engagement of the IFNI pathway. The administration of Flt3 ligand (Flt3-L), a growth factor that stimulates the proliferation of hematopoietic cells, to neonates before RSV-infection, resulted in increased lung DC number, and reconditioned the IFNI pathway upon RSV neonatal infection. Besides, neonates treated with Flt3-L were protected against exacerbated airway disease upon adult reexposure to RSV. This was associated with a reorientation of RSV-specific responses toward Th1-mediated immunity. Thus, the poor lung DCs and IFNI responses to RSV in neonates may be partly responsible for the deleterious long-term consequences revealed upon adult reexposure to RSV, which could be prevented by Flt3-L treatment. These results open new perspectives for developing neonatal immuno-modulating strategies to reduce the burden of bronchiolitis.

International audience; In addition to important regulatory roles in gene expression through RNA interference, it has recently been shown that microRNAs display immune stimulatory effects through direct interaction with receptors of innate immunity of the Toll-like receptor family, aggravating neuronal damage and tumour growth. Yet no evidence exists on consequences of microRNA immune stimulatory actions in the context of an autoimmune disease. Using microRNA analogues, we here show that pancreatic beta cell-derived microRNA sequences induce pro-inflammatory (TNFa, IFNa, IL-12, IL-6) or suppressive (IL-10) cytokine secretion by primary mouse dendritic cells in a sequence-dependent manner. For miR-29b, immune stimulation in RAW264.7 macrophages involved the endosomal Toll-like receptor-7, independently of the canonical RNA interference pathway. In vivo, the systemic delivery of miR-29b activates CD11b+B2202 myeloid and CD11b-B220+ plasmacytoid dendritic cells and induces IFNa, TNFa and IL-6 production in the serum of recipient mice. Strikingly, in a murine model of adoptive transfer of autoimmune diabetes, miR-29b reduces the cytolytic activity of transferred effector CD8+ T-cells, insulitis and disease incidence in a single standalone intervention. Endogenous miR-29b, spontaneously released from beta- cells within exosomes, stimulates TNFa secretion from spleen cells isolated from diabetes-prone NOD mice in vitro. Hence, microRNA sequences modulate innate and ongoing adaptive immune responses raising the question of their potential role in the breakdown of tolerance and opening up new applications for microRNA-based immune therapy.

Streptococcus pneumoniae is a major human pathogen, which causes pneumonia, meningitis and septicemia. To insure its survival and dissemination, the pneumococcus deploys an array of virulence factors promoting invasion of tissues and evasion from the immune system. A particular class of proteins not associated with any known export system, the moonlighting proteins, is found at the pneumococcal surface. Moonlighting proteins are conserved cytoplasmic metabolic enzymes or molecular chaperones localized in various cellular compartments and exhibiting additional functions. The glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is found at the surface of numerous eukaryotic and prokaryotic cells, and display diverse roles in the virulence processes of pathogenic organisms. The pneumococcal surface GAPDH acts as a virulence factor by binding to host plasminogen/plasmin, which facilitates the bacterial invasion through the extracellular matrix and the endothelial and epithelial cell barriers. However, the mechanisms leading to the GAPDH export and binding to the bacterial surface had not been deciphered yet. This work demonstrates that the GAPDH is released upon cell lysis and associates with the peptidoglycan. C1q, a key component of the classical complement pathway, is a major player in the response to microbial infection and has been shown to detect noxious altered-self substances such as apoptotic cells. The use of complementary experimental approaches allowed the identification of the GAPDH as a C1q partner when exposed at the surface of S. pneumoniae and human apoptotic cells. However, and rather unexpectedly, the pneumococcal GAPDH activates the complement cascade unlike the human one. Those results encourage further studies in order to understand how C1q recognition of two closely related proteins can lead to such striking differences on its complement activation properties., Streptococcus pneumoniae est un pathogène humain majeur causant des pneumonies, méningites et septicémies. Pour assurer sa survie et sa dissémination le pneumocoque déploie un ensemble de facteurs de virulence favorisant l'invasion des tissus et l'évasion du système immunitaire. Une classe particulière de protéines dites « moonlighting », ne sont associées à aucun système d'export connu et pourtant se trouvent localisées en surface du pneumocoque. Les protéines « moonlighting » sont des protéines cytoplasmiques conservées, localisées dans divers compartiments cellulaires et présentant des fonctions additionnelles. La glycéraldéhyde-3-phosphate déshydrogénase (GAPDH) est retrouvée en surface de nombreuses cellules et exerce divers rôles dans les processus de virulence d'organismes pathogènes. La GAPDH de surface du pneumocoque agit comme un facteur de virulence en recrutant le plasminogène / la plasmine de l'hôte, ce qui facilite l'invasion bactérienne à travers la matrice extracellulaire et les barrières endothéliales et épithéliales. Cependant, les mécanismes permettant l'export et la fixation de la GAPDH à la surface de la bactérie n'avaient pas encore été découverts. Ce travail démontre que la GAPDH est relarguée par lyse cellulaire et s'associe au peptidoglycane. C1q, un composant clé de la voie classique du complément, est un acteur majeur dans la réponse aux infections microbiennes et peut également détecter des éléments nocifs du soi-altéré comme les cellules apoptotiques. L'usage d'approches expérimentales complémentaires a permis d'identifier la GAPDH comme un partenaire de C1q quand elle est exposée en surface de S. pneumoniae et de cellules apoptotiques humaines. Néanmoins et de manière plutôt inattendue, seule la GAPDH du pneumocoque active la cascade du complément à la différence de la protéine homologue humaine. Ces résultats encouragent la poursuite d'études afin de comprendre comment la reconnaissance par C1q de deux protéines très proches peut conduire à de telles différences sur ses propriétés d'activation du complément.

T; International audience; Background: Implantation is a complex process that requires a delicate cooperation between the immune and reproductive system. Any interference in the fine balance could result in embryo loss and infertility. We have recently shown that Toll-like receptor 5 activation results in a decrease of trophoblast cells binding to endometrial cells in an in vitro model of human implantation. However, little is known about the downstream signalling leading to the observed failure in implantation and the factors that modulate this immune response. Methods and Principal Findings: An in vitro model of embryo implantation was used to evaluate the effect of trophoblasts and flagellin on the activation of NF-kappa B in endometrial cells and whether TLR5-related in vitro implantation failure is signalled through NF-kappa B. We generated two different NF-kappa B reporting cell lines by transfecting either an immortalized endometrial epithelial cell line (hTERT-EECs) or a human endometrial carcinoma cell line (Ishikawa 3-H-12) with a plasmid containing the secreted alkaline phosphatase (SEAP) under the control of five NF-kappa B sites. The presence of trophoblast cells as well as flagellin increased NF-kappa B activity when compared to controls. The NF-kappa B activation induced by flagellin was further increased by the addition of trophoblast cells. Moreover, blocking NF-kappa B signalling with a specific inhibitor (BAY11-7082) was able to restore the binding ability of our trophoblast cell line to the endometrial monolayer. Conclusions: These are the first results showing a local effect of the trophoblasts on the innate immune response of the endometrial epithelium. Moreover, we show that implantation failure caused by intrauterine infections could be associated with abnormal levels of NF-kappa B activation. Further studies are needed to evaluate the target genes through which NF-kappa B activation after TLR5 stimulation lead to failure in implantation and the effect of the embryo on those genes. Understanding these pathways could help in the diagnosis and treatment of implantation failure cases.

After LR delivery of an AAV8 vector expressing GFP in non-human primates, we showed detection of transgene expression until at least 8 months post-injection in contrast to IM where GFP was lost at 3 months post-injection. Interestingly, GFP persistence after LR was observed despite the detection of anti-GFP IgG antibodies, even at higher titers than IM route, and the presence of T cells responding to GFP peptides in the spleen. Surprisingly, GFP-restimulated T cells secreted IFNγ but not IL2 after LR as compared to IM route where both cytokines were detected.

L’objectif de cette étude est d’évaluer l’influence de stimulations microbiennes de poules sur le système de défense innée du blanc d’oeuf en utilisant deux modèles expérimentaux. Le premier est basé sur la comparaison de l’activité antibactérienne des blancs d’oeufs de poules axéniques élevées en conditions stériles (Ax), de poules exemptes d’organismes pathogènes spécifiques (EOPS) et de poules conventionnelles (C). Le second modèle compare des oeufs de poules contrôles ou immunostimulées par injection de lipopolysaccharide bactérien (LPS). Les résultats ont montré que les blancs d’oeufs des groupes C et EOPS ont une activité inhibitrice renforcée vis-à-vis de Staphylococcus aureus comparée à ceux du groupe Ax. De même, une meilleure activité antimicrobienne contre ce pathogène est observée pour les blancs d’oeufs du groupe LPS par rapport à ceux du groupe contrôle. En revanche, l’activité anti-Escherichia coli n’est pas modifiée ni par le degré de contamination du milieu des poules (Ax vs EOPS vs C) ni par l’injection de lipopolysaccharide (LPS vs contrôle). L’analyse de l’expression de gènes codants pour des protéines/peptides antimicrobiens de l’oeuf et les mesures des activités du lysozyme et d’antiprotéases dans le blanc n’ont pas permis d’expliquer les stimulations de l’activité antimicrobienne du blanc d’oeuf. Ces résultats suggèrent que les poules peuvent renforcer modérément l’activité antimicrobienne du blanc d’oeuf en réponse à des stimuli microbiens de leur milieu et anticiper les besoins de l’embryon en termes de protection. Les mécanismes moléculaires à l’origine de cette modulation demeurent néanmoins inconnus à ce jour.

L’asthme chez l’homme et le souffle chez le cheval sont des maladies inflammatoires chroniques des voies respiratoires partageant plusieurs caractéristiques physiopathologiques dont la bronchoconstriction réversible, l’inflammation des voies respiratoires inférieures, l’hyperréactivité bronchique et le remodelage tissulaire. Les phénotypes cliniques d’asthme se caractérisent en partie selon le type d’inflammation affectant les voies respiratoires et la présence ou non d’allergie. Le souffle chez le cheval s’avère être un modèle adapté pour l’étude des mécanismes impliqués dans l’asthme neutrophilique, lesquels demeurent particulièrement mal compris, en contraste avec ceux associés avec l’asthme éosinophilique. La réponse immunologique sous-jacente au souffle implique entre autres l’expression de cytokines de type Th2, suggestives d’une réponse allergique (immunité acquise). La poussière environnementale qui provoque les symptômes du souffle contient également des agents non-spécifiques dérivés de bactéries, champignons et moisissures, susceptibles d’activer des mécanismes immunitaires innés chez les chevaux atteints du souffle. Nous avons étudié le rôle des neutrophiles dans l’inflammation associée à la réponse innée et acquise chez le cheval atteint du souffle. Dans un premier temps, l’effet de produits dérivés de bactéries sur l’activation des neutrophiles sanguins provenant de chevaux normaux et atteints de souffle a été étudié dans le but d’évaluer la contribution de la réponse innée dans la physiopathologie du souffle. Nous avons évalué l’effet de l’IL-4, une cytokine de type Th2, sur les neutrophiles des deux groupes de chevaux afin d’évaluer de quelle manière le neutrophile peut participer à la réponse acquise associée à la réponse allergique. Finalement, nous avons étudié l’expression des isoformes de l'arginase par les neutrophiles équins car cette enzyme métabolise la L-arginine et est potentiellement impliquée dans le bronchospasme et le remodelage tissulaire associés à l’asthme. Nos résultats suggèrent que les neutrophiles et les mononucléaires sanguins isolés des chevaux atteints du souffle possèdent une réponse inflammatoire exagérée en réponse aux lipopolyssacharides et peptides formylés et surexpriment les cytokines pro-inflammatoires IL-1β, TNF et IL-8. Cette réponse innée aberrante est associée à une inflammation systémique caractérisée par des concentrations sériques élevées de TNF chez les chevaux atteints du souffle en période de rémission clinique. De plus, nos résultats montrent que l’IL-4 active le neutrophile équin et favorise son chimiotactisme de manière autocrine. L’IL-4 induit un phénotype d’activation typique dans le neutrophile équin, caractérisé par l’expression accrue des cytokines pro-inflammatoires (IL-8 et TNF) ainsi que de récepteurs potentiellement impliqués dans la réponse allergique (IL-4Rα et CD23). Enfin, nous montrons que que l’arginase 1 n’est pas un marqueur de l’activation des neutrophiles équins par l’IL-4, mais que ces cellules expriment constitutivement l’isoforme 2 fonctionnelle de l’arginase. La surrégulation des deux isoformes au niveau des poumons périphériques semble être associée à la pathologie du souffle, ce qui est en accord avec les modèles d’asthme chez la souris, le rat et le cobaye. L'ensemble de ces travaux suggère que les neutrophiles sont des cellules effectrices importantes de la réponse innée et acquise dans la pathophysiologie du souffle, un modèle naturel d’asthme neutrophilique., Human asthma and equine heaves are chronic pulmonary diseases sharing several pathophysiological properties including lower airway inflammation, reversible bronchoconstriction, bronchial hyperresponsiveness, and tissue remodeling. Clinical phenotypes of asthma are characterized in part by the inflammatory cell populations infiltrating the airways, and the presence or absence of allergy. Heaves is a suitable animal model for the study of the poorly defined pathophysiological processes leading to airway neutrophilia. The immune response in heaves involves Th2 cytokine expression, which is, among other features, associated to allergic inflammation (acquired immunity). Environmental dust exposure leading to clinical exacerbation of heaves contains non-specific agents derived from bacteria, molds or fungi which could also activate innate immune responses in heaves affected horses. We studied the role of neutrophils in innate and acquired immune responses in heaves affected-horse. First, innate immune responses of neutrophils isolated from normal and heaves-affected horses to bacterial-derived products were studied. We also assessed the effect of IL-4, a Th2 cytokine, on equine neutrophils isolated from both groups of horses. Finally, we evaluated the arginase isoforms expressed by equine neutrophils as this enzyme that takes part to the L-arginine metabolism and is thought to contribute to bronchospasm and tissue remodeling associated with asthma. Our results suggest that both neutrophils and mononuclear cells from heaves-affected horses, when compared to healthy horses, have an excessive inflammatory response to lipopolyssacharides and formylated peptides characterized by increased IL-1β, IL-8 and TNF expression. This altered innate response was associated with systemic inflammation in asymptomatic susceptible horses as high serum TNF concentrations were detected. Furthermore, we found that equine neutrophils are activated by IL-4 and release neutrophil chemotactic factors in response to this cytokine. IL-4 also induces a distinctive activation phenotype in neutrophils that is characterized by increased expression of the pro-inflammatory cytokines (IL-8 and TNF) and receptors (IL-4Rα and CD23) potentially involved in the allergic response. Finally, we showed that arginase 1 is not a marker of IL-4-activated equine neutrophils although they constitutively express a functionally active isoform 2 of the enzyme. The up-regulation of arginase isoforms in the peripheral lungs of horses with heaves suggests a role for arginase in this model, as it is described in the mouse, rat and guinea pig models. Cancer represents the second most common cause of death in industrialized countries. Epidemiological and biological studies have now conclusively proved that a variety of infectious agents constitute one of the main causes of cancer worldwide. It has been pointed out that more than 20% of cancers are from infectious origin. HPV high-risk mucosal types are associated to 98% of all cervical cancer cases. Regarding EBV, over 90% of the world’s population is infected and can give rise to malignancies such as Burkitt lymphoma or Hodgkin disease.(Young and Rickinson 2004) Keys features for oncoviruses to induce cancer are firstly to per¬sist by dampening host immune responses and to induce genomic instability in the host by altering the regulation of the cell cycle leading the infected cells to an uncontrolled proliferation. The purpose of this thesis was to find new mechanisms by which EBV and HPV can promote carcinogenesis. We have shown that EBV can alter the regulation and expression of TLRs, the key effectors molecules of the innate immune response. EBV infection of human primary B cells resulted in the inhibition of TLR9 functionality. Our study described a mechanism used by EBV to suppress the host immune response by deregulating the TLR9 transcript through LMP1-mediated NF-κB activation. As TLR was found deregulated in many cancers, we hypothesized that TLR9 may also a direct role in the process of cell cycle control and that loss of its expression may lead to transformation of the cell. Our overall objective here was to study the role of TLR9 in suppressing the events that initiates transformation of epithelial cells in the setting of cervical cancer (virus-associated) and in head and neck cancer (non–virus-associated). A third project dealt with the mechanism cell cycle deregulation by the oncoprotein E6 which expressed during infection with HPV16. We reported that HPV16E6 targets the cellular factor p150Sal2, which positively regulates p21 transcription. HPV16E6 associates with p150Sal2, inducing its functional inhibition by preventing its binding to cis elements on the p21 promoter. These data described a novel mechanism by which HPV16E6 induces cell cycle deregulation with a p53-independent pathway preventing G1/S arrest and allowing cellular proliferation and efficient viral DNA replication, Le cancer représente la deuxième cause de mortalité dans les pays industrialisés. Il a été démontré que 20% des cancers sont d'origine infectieuse. Nous nous sommes intéressés à deux oncovirus HPV (virus du papillome humain) et EBV (Epstein-Barr Virus) responsable du cancer de l'utérus et de divers lymphome B réciproquement. Les événements clés pour le développement d'un cancer viro-induit sont la persistance du virus via la dérégulation des réponses immunitaires et l'induction d'une instabilité gé¬nomique via une dérégulation du cycle cellulaire. Nous avons donc cherché si EBV était capable d'altérer la réponse immunitaire innée. Nous avons montré que EBV était capable d'inhiber TLR9 un acteur clef de la réponse immunitaire innée. Comme TLR9 est inhibé dans un certain nombre de cancers, nous nous sommes demandé si ce récepteur pouvait également, avoir un rôle dans l'oncogenèse. Nous avons montré que la réexpression de TLR9 induisait un ralentissement transitoire de la prolifération cellulaire. Nous nous sommes par la suite intéressés aux mécanismes de dérégulation du cycle cellulaire induits par E6 une oncoprotéine de HPV16. Nous avons trouvé un nouveau mécanisme d'inhibition de l'inhibiteur du cycle cellulaire, p21. HPV16E6 se lie et inhibe les fonctions de du facteur de transcription p150Sal2, ce qui induit une inhibition de p21 dans un contexte p53 indépendant

Background The development of mucosal vaccines is crucial to efficiently control infectious agents for which mucosae are the primary site of entry. Major drawbacks of these protective strategies are the lack of effective mucosal adjuvant. Synthetic oligodeoxynucleotides that contain several unmethylated cytosine-guanine dinucleotide (CpG-ODN) motifs are now recognized as promising adjuvants displaying mucosal adjuvant activity through direct activation of TLR9-expressing cells. However, little is known about the efficacy of these molecules in stimulating the intestinal immune system in neonates. Methodology/Principal Findings First, newborn mice received CpG-ODN orally, and the intestinal cytokine and chemokine response was measured. We observed that oral administration of CpG-ODN induces CXC and CC chemokine responses and a cellular infiltration in the intestine of neonates as detected by immunohistochemistry. We next compared the efficiency of the oral route to intraperitoneal administration in stimulating the intestinal immune responses of both adults and neonates. Neonates were more responsive to TLR9-stimulation than adults whatever the CpG-ODN administration route. Their intestinal epithelial cells (IECs) indirectly responded to TLR9 stimulation and contributed to the CXC chemokine response, whereas other TLR9-bearing cells of the lamina-propria produced CC chemokines and Th1-type cytokines. Moreover, we showed that the intestine of adult exhibited a significantly higher level of IL10 at homeostasis than neonates, which might be responsible for the unresponsiveness to TLR9-stimulation, as confirmed by our findings in IL10-deficient mice. Conclusions/Significance This is the first report that deciphers the role played by CpG-ODN in the intestine of neonates. This work clearly demonstrates that an intraperitoneal administration of CpG-ODN is more efficient in neonates than in adults to stimulate an intestinal chemokine response due to their lower IL-10 intestinal level. In addition we report the efficiency of the oral route at inducing intestinal chemokine responses in neonate that might be taken into consideration for further vaccine development against neonatal diseases.

The type III exoenzyme S toxin of Pseudomonas aeruginosa possesses a GAP domain (GTPase Activating Protein) (ExosGAP) inhibiting the Rho GTPases (Rho, Rac and Cdc42) and phagocytosis in Mammal cell lines. We have used a transgenesis approach in Drosophila melanogaster using a tissue-specific inducible expression system (UAS-Gal4) to express ExoSGAP. We have demonstrated that ExoSGAP targeted in vivo the Rho GTPases Rho, Rac1, Rac2 and Cdc42. The ExoSGAP expression affects fly resistance to infection by inhibiting phagocytosis of bacteria by plasmatocytes, a macrophage cell line, but has no effect on NF-kB signalling pathways. A genetic approach has allowed the identification of new ExoSGAP targets by looking for genes whose misexpression modifies the eye or wing phenotype induced by the toxin expression. We have identified several genes which can be implicated in JNK and NF-kB signalling pathways. All these results validate a new strategy to study type III toxins by transgenesis in Drosophila.In parallel, I have demonstrated GTPase Rac2 specificity in Drosophila resistance to infection. Rac2 participates notably in phagocytosis.The works of Dr. H. Tricoire led to the identification of 180 genes whose misexpression modifies fly response to oxidative stress. We have tested 105 of these lines for their resistance to infections to study a possible correlation between the oxidative and infectious stress response. Mastitis caused by Escherichia coli and Staphylococcus aureus is a major pathology of dairy cows. To better understand the differential response of the mammary gland to these two pathogens, we stimulated bovine mammary epithelial cells (bMEC) with either E. coli crude lipopolysaccharide (LPS) or with S. aureus culture supernatant (SaS) to compare the transcriptomic profiles of the initial bMEC response. By using HEK 293 reporter cells for pattern recognition receptors, the LPS preparation was found to stimulate TLR2 and TLR4 but not TLR5, Nod1 or Nod2, whereas SaS stimulated TLR2. Biochemical analysis revealed that lipoteichoic acid, protein A and α-hemolysin were all present in SaS, and bMEC were found to be responsive to each of these molecules. Transcriptome profiling revealed a core innate immune response partly shared by LPS and SaS. However, LPS induced expression of a significant higher number of genes and the fold changes were of greater magnitude than those induced by SaS. Microarray data analysis suggests that the activation pathways and the early chemokine and cytokine production preceded the defense and stress responses. A major differential response was the activation of the type I IFN pathway by LPS but not by SaS. The higher upregulation of chemokines (Cxcl10, Ccl2, Ccl5 and Ccl20) that target mononuclear leucocytes by LPS than by SaS is likely to be related to the differential activation of the type I IFN pathway, and could induce a different profile of the initial recruitment of leucocytes. The MEC responses to the two stimuli were different, as LPS was associated with NF-κB and Fas signaling pathways, whereas SaS was associated with AP-1 and IL-17A signaling pathways. It is noteworthy that at the protein level secretion of TNF-α and IL-1β was not induced by either stimulus. These results suggest that the response of MEC to diffusible stimuli from E. coli and S. aureus contributes to the onset of the response with differential leucocyte recruitment and distinct inflammatory and innate immune reactions of the mammary gland to infection.
