Objective: A study was planned to investigate the effect of exposure to cigarette smoke and the effects of calcium channel blockers in experimentally induced sciatic nerve injuries. Material-Method: The sciatic nerve was cut on one side in all the groups, and repair was performed in hour 0. After repair, a single daily dose of 1 ml of 0.9% saline was administered to group 1, 90 days, 30 minutes, cigarette smoke to group 2, 1mg/kg calcium channel blocker to group 3, 90 days, 30 minutes, cigarette smoke and 1mg/kg Calcium channel blocker was given to group 4. Results: At the end of the 12th week, gait analysis at and sciatic function index (SFI), the number of myelinated axons, axon areas and myelin diameters were assessed. Aim: Cigarette smoking is a common addictive manner and one of the greatest threats to health. Nicotine is one of the main components of cigarette. The aim of this study was to reveal the effect of cotinine which is an active metabolite of nicotine, on peripheral nerve injury in rats. Material and Methods: We studied 42 male adult albino-Wistar rats that were divided into three groups with simple randomization method. Group 1 were given Cotinine® (C-5923-sigma) intraperitoneally, at a dose of 0.3 mg/kg/day for 21 days. Group 2 were given ethyl alcohol, the solvent of Cotinine in the same way, dose and period. Group 3 were subjected to sciatic nerve compression injury by a clip, which has a closing pressure of 50 gr/cm2. Group 1 and 2 were subjected to the same type of injury at the end of 21 days. Four weeks later after trauma, both three groups were sacrificed and injured sciatic nerve sections are taken for histopathological analysis. Results: It was observed that cotinine aggravated the traumatic degeneration and as privileged caused to fibrosis. In the Schwann cells of thick-myelinated fibers exhibited higher grades of degeneration and mitochondrial augmentation. This situation was estimated as a response to oxidative stress. Conclusion: This study reveals that peripheral nerve regeneration after traumatic injury may be affected negatively in smokers. Bu derlemede yazarlar kök hücre/progenitör hücre transplantasyonunun sinir hasarlarının iyileştirilmesinde kullanımı ile ilgili yakın zamandaki gelişmeler ve bu alandaki başlıca çalışmalar hakkında bilgi vermeyi amaçlamaktadır. Periferik sinirlerin akson rejenerasyonu ve hedef dokularını tekrar innerve etme potansiyeline sahip olmalarına rağmen ağır sinir hasarı durumlarında düzelme sınırlı kalmaktadır. Denervasyona uğramış periferik sinirin Schwann hücrelerinin rejenerasyonda yeteri kadar rol alamayışlarından dolayı denerve olmuş distal sinir dokusu ortamının dış kaynaklı doku hücreleriyle desteklenmesi yaklaşımı gerçekçi görülmüştür. Çeşitli kök hücrelerinin periferik sinir rejenerasyonundaki etkileri çalışılmıştır. Deri, kemik iliği ve yağ doku kökenli kök hücreleri Schwann hücrelerine dönüşme kapasiteleriyle en ümit verici adaylar olarak belirlenmiştir. Pluripotent indüklenmiş kök hücre denemelerine ek olarak, yakın zamanlarda yapılan çalışmalar kök hücrelerinde büyüme faktörlerinin ifadesinin uyarılması da rejenerasyonda önemli potansiyele sahiptir. Kök hücrelerinin etkili kullanılabilecek kaynak olmaları ve sinir rejenerasyonunda ümit verici faktörler olduklarının düşünülmesine rağmen bu hücre tedavi potansiyellerinin klinikte kullanılması için gerekli ideal yöntemler henüz oluşturulamamıştır.

Deneysel çalışmalarda periferik sinirin akson çapı, miyelinli sinir lifi çapı ve miyelinli sinir için iç çap/dış çap oranı (g-ratio), demiyelinizasyon, remiyelinizasyon ve sinir onarımının değerlendirilmesinde görüntü analiz programlarından yararlanılmaktadır. Bu nedenle, morfometrik değerlendirmeye sıklıkla gereksinim duyulan periferik sinir çalışmaları hedeflenerek, laboratuvarımızda yarı otomatik bir görüntü analiz programı geliştirilmiştir. Çalışmamızda, geliştirdiğimiz görüntü analiz yazılımının performansının, serbest erişimli çok amaçlı diğer bir yazılım ile güvenilirlik yönünden karşılaştırılması amaçlanmıştır. Farelerin genel anestezi altında siyatik sinirlerinden alınan doku örnekleri gluteraldehit solüsyonuna alınmıştır. Rutin elektron mikroskop takip yöntemlerinden geçirilerek epoksi resin (epoxy resin) gömülen dokulardan yarı ince kesitler alınmış ve toluidin mavisi ile boyanmıştır. Kesitler dijital kameralı ışık mikroskopta incelenmiş ve X100’lük objektifte fotoğraflanmıştır. Mikrograflar laboratuvarımızda geliştirilen Tantuna Morfometrik Ölçüm Sistemi (TanTuna Morphometric Measuring System; TT-MMS) yazılımı ile analiz edilmiştir. Seçilen iki adet ışık mikrograftan rastgele seçilen 200 adet miyelinli akson kesitinin ölçümleri program kullanılarak yapılmıştır. Aynı ölçümler ImageJ programı ile tekrarlanmış ve her iki programla elde edilen veriler istatistiksel olarak karşılaştırılmıştır. Ek bir donanıma ihtiyaç duymadan her iki yazılımın da güvenilir olduğu ve birbirleriyle uyumlu ölçümler yaptıkları saptanmıştır (p, Image analysing programs are used in peripheral nerve studies to measure axon diameter, myelinated nerve diameter and the ratio of inner axonal diameter to outer diameter (g-ratio), demyelination, remyelination and nerve repair. So, a semi-automatic, image analysis program was developed in our laboratory by targeting peripheral nerve studies which morphometric evaluation is frequently required. It is aimed to compare the performance and reliability of this program with another free access multipurpose software. Tissue samples from sciatic nerves of mice were taken into gluteraldehyde solution under general anesthesia. After routine electron microscopic processing method, semi-thin sections were taken from epoxy resin embedded blocks and stained with toluidine blue. Sections were examined and photographed under the light microscope at X100 magnification. Micrographs were analyzed using the Tantuna Morphometric Measuring System (TT-MMS) software developed in our laboratory. Measurements of the randomly selected 200 myelinated axon sections from two selected light micrograph were performed using the program. We present a case of a 10 year-old girl with slowly enlarging, painless mass at the volar aspect of her left hand. The lesion had been present since infancy. Diagnosis was lipomatosis of the median nerve which is a very rare, benign tumor involving peripheral nerves. Amaç: Sigara içmek yaygın bir bağımlılık biçimidir ve sağlık için en büyük tehditlerden biridir. Nikotin, sigaranın ana bileşenlerinden biridir. Bu çalışmanın amacı, nikotinin aktif bir metaboliti olan kotininin sıçanlarda periferik sinir hasarına etkisini ortaya koymaktır.Gereç ve Yöntemler: Basit randomizasyon metoduyla üç gruba ayrılan 42 erkek yetişkin albino-Wistar sıçan üzerinde çalıştık. Grup 1'e 21 gün boyunca 0,3 mg/kg/gün dozunda intraperitoneal olarak Cotinine® (C-5923-sigma) verildi. Grup 2'ye aynı şekilde Cotinine çözücüsü olan etil alkol, aynı doz ve sürede verildi. Grup 3, kapanma basıncı 50 gr/cm2 olan klips ile siyatik sinir kompresyonu yaralanmasına maruz bırakıldı. Grup 1 ve 2 ise 21 gün sonunda aynı tipte yaralanmaya maruz bırakıldı. Travmadan dört hafta sonra bütün gruplar sakrifiye edildi ve hasarlı siyatik sinir kesitleri histopatolojik analiz için alındı.Bulgular: Kotininin travmatik dejenerasyonu şiddetlendirdiği gözlenmiştir ve ayrıca kotininin özellikle fibrozun artışına neden olduğu görülmüştür. Kalın miyelinli fiberlerdeki Schwann hücrelerinde daha yüksek derecelerde dejenerasyona sebep olduğu ve ek olarak, bu hücrelerde mitokondriyal artış olduğu görüldü. Objective: The aim of this study was to determine the effects of the arm and leg muscle mass together with demographic factors, on peripheral nerve cross sectional area. Methods: Median and ulnar nerve cross sectional area's were measured at mid-point of forearm and peroneal nerve cross sectional area's above fibular head with ultrasonography in 233 volunteers. Muscle mass was evaluated by the bioelectrical impedance analysis. Results: Ulnar and peroneal nerve cross sectional area's increased with advancing age. BMI was only correlated with ulnar nerve cross sectional area and height with peroneal nerve cross sectional area. Arm muscle mass was significantly correlated with ulnar and median nerve cross sectional area's (p < 0.05); however, leg muscle mass did not correlate with peroneal nerve cross sectional area. Conclusion: The nerve thickness may be influenced by various factors. Muscle mass is one of these factors in healthy nerves. This situation should be taken into account during setting reference values for healthy nerves and assessment of neuropathies by ultrasonography. [ABSTRACT FROM AUTHOR]

The goal of this study was to examine the possible effect of administering Dexamethasone (Dex), Betamethasone (Bet), and Methylprednisolone (Mps) in promoting nerve regeneration and functional motor recovery after a crush injury to the right sciatic nerve in a rat model would. Sixty-four male Wistar albino rats (weighing between 250 ± 30 g and age 12 to 14 weeks) were randomly divided into eight groups (n=8). The Control (Cont) group received no surgery or treatment, whereas the positive control groups received treatment with Dex, Bet, and Meth at a dose of 2 mg/kg/day for each group. The sciatic nerve was exposed to crush by applying 50 Newton for 60 seconds (sec) in the injury (Inj) or injury-treated groups. In the Inj+Dex group, the animals have received Dex for 10 days. In the Inj+Bet group, the rats were treated by Bet for 14 days. In the Inj+Mps group, the rats were given Mps for 14 days. All the treatments were performed intraperitoneally 24 hours after surgery. The stereological analysis, histological and functional examinations were done to assess the sciatic nerve in each group. 2 Stereological analysis revealed a significant difference in decreasing way in points of total numbers of myelinated axons in the Dex + Inj group compared to the Inj group (p = 0.019). However, the Bet + Inj and Mps + Inj groups revealed no significant differences in the total number of myelinated axons, myelin sheath thickness and axonal area compared to the Inj group (P > 0.05). According to sciatic functional index (SFI) evaluation, Dex+Inj and Mps+Inj groups demonstrated significant differences in increasing way (p = 0.016) and (p = 0.005) respectively. However, compared to the Inj group, there was no significant difference in the EMG assessment in any treated groups (p > 0.05). In conclusion, Dex had a beneficial effect on myelinated axons number and functional improvement, whereas Mps only enhanced functional recovery. Furthermore, microscopic examinations revealed that all the proposed medications had beneficial impacts on the sciatic nerve sections. Bu çalışmanın amacı, sıçan modelinde sağ siyatik sinir hasarından sonra Deksametazon (Dex), Betametazon (Bet) ve Metilprednizolon (Mps)' un sinir rejenerasyonu ve fonksiyonel motor iyileşmeye muhtemel etkilerini incelemektir. Altmış dört erkek Wistar albino sıçan (250 ± 30 g ağırlığında ve 12 - 14 haftalık), rastgele sekiz eşit (n=8) gruba ayrıldı. Kontrol (Cont) grubuna herhangi bir cerrahi işlem ve tedavi uygulaması yapılmazken, pozitif kontrol gruplarına, her grup için 2 mg/kg/gün dozunda Dex, Bet ve Meth verildi. Hasar (Inj) grubu ve Hasar+tedavi gruplarında siyatik sinir, 60 saniye boyunca 50 Newtonluk basınç uygulanarak ezilmeye maruz bırakıldı. Hasar+Dex grubunda, ratlara 10 gün boyunca 2 mg/kg/gün Dex verildi. Hasar+Bet grubunda, ratlara 14 gün boyunca 2mg/kg/gün Bet verildi. Hasar+Mps grubunda ratlara 14 gün boyunca 2 mg/kg/gün Mps verildi. Tüm madde uygulamaları cerrahi işlemden 24 saat sonra intraperitoneal yolla uygulandı. Her grupta siyatik sinir 3 iyileşmesini değerlendirmek için stereolojik analizler, histolojik ve fonksiyonel incelemeler yapıldı. Stereolojik analiz sonuçlarına göre; miyelinli akson sayısı açısından Hasar grubuna kıyasla Hasar+Dex grubunda azalan yönde bir istatistiksel fark (p = 0,019) bulundu. Ancak, Hasar+Bet ve Hasar+Mps grupları ile Hasar grubu arasında miyelinli akson sayısı, miyelin kılıf kalınlığı ve akson alanı parametreleri açısından istatistiksel bir fark bulunmadı (p >0,05). Siyatik fonksiyonel indeks (SFI) verilerine göre, Hasar+Dex ve Hasar+Mps gruplarında Hasar grubuna kıyasla artan yönde anlamlı istatistiksel fark (sırasıyla p = 0,016 ve p = 0,005) tespit edildi. Bununla birlikte, Hasar grubuyla karşılaştırıldığında, hiçbir tedavi grubunda EMG değerlendirmesi açısından anlamlı bir fark görülmedi (p > 0,05). Sonuç olarak; Dex, miyelinli akson sayısı ve fonksiyonel iyileşme açısından olumlu etkiye sahipken, Mps sadece fonksiyonel iyileşmeye olumlu katkı sağlamıştır. Bununla birlikte, mikroskobik değerlendirmeler uygulanan tüm maddelerin siyatik sinir iyileşmesine olumlu etkileri olduğunu göstermektedir.

Physiotherapy and rehabilitation is always the first choice in the management of Thoracic Outlet Syndrome (TOS). Due to difficulty to make objective diagnosis or making the diagnosis behind time despite obvious clinical features, physiotherapy approaches should include causative factors and protective approaches than symptomatic treatments. Creating approaches oriented causative factors for TOS is more important while postural modifications, patient education, strengthening and stretching exercises seem as basic approaches. The basic aim in conservative management is to remove extrinsic compression with decreasing the irritation of peripheral nerves. Moreover, to offer methods to treat TOS as well as decreasing compression in the neurovascular structures. Assessment should include sensory and motor symptoms which induced by neural and vascular compressions. Posture assessment, muscle imbalance, muscle shortness and flexibility, respiratory assessment and pain assessments are performed in this manner. Conservative treatment approaches can be ranged to gain the diaphragmatic breathing pattern, to decrease inflammation, to relieve the soft tissues with tension, to provide muscle balance, to maintain neural excursion, to improve posture and body mechanics and to provide the spinal stabilization. In accordance with these basic aims, physiotherapy and rehabilitation can be planned as 3 stages: Preventative phase, restorative phase and rehabilitation phase. [ABSTRACT FROM AUTHOR]

20. Assessment of Regeneration in Peripheral Nerve Lesion Compared With Conventional Suture Technique and Using Cyanoacrylate (Animal Experiment).

In this study, it was aimed to assess effect of classical micro-suturing techniques combined with cyanoacrylate, used in nerve repair, on nerve regeneration in histopathological manner and to evaluate whether it could be safely used in peripheral nerve surgery. Adult Wistar-Albino female rats were used in this study. Animals were assigned into 4 groups for surgical processes as follows: Group 1: Control group (n=7); External circumferential neurolysis Group 2: Experiment group (n=7); Repair with 4 epineural sutures. Group 3: Experiment group (n=7); Repair with cyanoacrylate Group 4: Experiment group (n=7); combined group Histopathologically, it was seen that there was no vacuolar degeneration, foreign body type tissue reaction or fibrosis in the nerves of subjects in the control group. Moreover, it was found that there were findings of vacuolar degeneration, foreign body type tissue reaction and fibrosis in all experiments group in a similar manner. However, it was found that there was a statistically significant difference regarding grade results of vacuolar degeneration, fibrosis and foreign body type tissue reaction between control group and classical suturing, cyanoacrylate or combined groups (p<0.05). In conclusion, it has been thought that cyanoacrylate is a feasible method for peripheral nerve surgery; as it can be easily applied at conditions where surgical suturing is challenging; it reduces the trauma exposed by nerve endings; it provides easy identification of nerve endings during secondary repair, which are contaminated and could not be primarily repaired; it shortens the procedure time; it is cost-effective and; it easy to learn. Objective: Benign schwannomas are the most common tumour of the peripheral nerves. Symptomatic schwannomas are treated by surgical excision, but new neurological deficits may develop. We performeda retrospective review of cases of schwannomas in the extremities and reviewed the relevant literature.Methods: We retrospectively reviewed the demographic characteristics of 11 patients with schwannomas treated at our institution. We also reviewed the clinical characteristics and postoperative results of these cases, determined the possible risk factors influencing the development of complications and compared the risk factors with those reported in the literature.Results: There were five males and six females with a mean age of 37.6 (range: 17–62) years. The mean postoperative follow-up was 54.6 (range: 26–88) months. Three tumours were located in the forearm and the rest were localized in the lower extremity. No recurrences were observed during the follow-up period. New motor and sensory deficits were observed in only one patient.Conclusion: Schwannomas in the extremities can be excised with acceptable risk of neurological deficits. Meticulous dissection is required during surgery., Amaç: Benign shcwannom periferik sinirlerin en yaygın tümörüdür. Semptomatik schwannomun tedavisi cerrahi eksizyondur. Cerrahlar yeni nörolojik defisitlerin gelişme olasılığına hazırlıklı olmalıdır. Biz literatürle birlikte ekstremite schwannomlarını içeren olguların geriye dönük bir analizi yaptık. Çalışma planı: Hastanemizde tedavi edilen 11 schawannomlu hastanın verilerini geriye dönük olarak inceledik. Klinik özellikleri ve ameliyat sonrası sonuçları gözden geçirdik ve komplikasyonların gelişimini etkileyen olası risk faktörlerini belirledik ve bunları literatürle karşılaştırdık. Bulgular: Ortalama yaşın 37.6 (17–62) olduğu beş erkek ve altı kadın vardı. Ameliyat sonrası ortalama takip süresi 54.6 (26–88) aydı. Önkolda üç tümör tespit edildi ve diğer tümörler bacak alt kısmındaydı. Takip periyodunda nüks gözlenmedi. Bir hastada yeni motor ve duysal defisit gözlendi. Çıkarımlar: Ekstremite schwannomaları kabul edilebilir nörolojik defisit riski ile eksize edilebilir. Cerrahi sırasında titiz bir disseksiyon gereklidir.

AIM: The aim of this study was to investigate the dose dependence of citicoline's previously-reported effects on recovery of peripheral nerve injury. MATERIAL and METHODS: Right sciatic nerves of sixty adult female Wistar Albino rats were incised and primary anastomosis was performed. Rats were then divided into four groups: Control group received 2 ml of saline intraperitoneally, while rats in C-300, C-600 and C-900 groups received 300 mu mol/kg, 600 mu mol/kg and 900 mu mol/kg citicoline dissolved in 2 ml saline, respectively. Rats were tested for sciatic functional index (SFI) on the 4th, 8th and 12th weeks and electrophysiological recordings were obtained on the 12th week. Rats were then sacrificed to investigate nerve adhesions and perform histomorphological examinations. Periferik sinir hasarları; kazalar, kesici delici alet yaralanmaları gibi travmalar sonrasında sık görülen ve özellikle tam kesiler, gecikmiş ve yetersiz onarımlar sonucunda ilerleyici kas atrofisi ve yetersiz fonksiyonel geri dönüş ile sonuçlanan dramatik tablolardır. Son zamanlarda, literatürde periferik sinir onarımları için farklı cerrahi teknikler dışında alternatif yöntemler de yer almaya başlamıştır. Kök hücre uygulamaları bu bağlamda, son zamanlarda tüm tıp camiasının üzerinde durduğu ve her yıl yüzlerce yeni çalışmanın yapıldığı bir konu haline gelmiştir. Pluripotent ve sınırsız yenilenme kapasiteleri nedeniyle kök hücrelerin, doku mühendisliğinde gelecek vadetmesi ve farklı rekonstrüktif prosedürler için önemli gelişmelere izin vermesi beklenir. Çalışmamızda, deneysel siyatik sinir yaralanmasında adipöz kaynaklı yeşil fluoresan proteini (YFP) işaretli mezenkimal kök hücre (ADSCs) uygulamasının, fonksiyonel analizler, histomorfometrik ve biyokimyasal incelemelerle periferik sinir rejenerasyonuna etkisinin saptanması amaçlandı. Deneysel 4 Wistar grubu (n:8) tasarlandı: Ratların sol siyatik sinirinde oluşturulan 1 cm’lik defektler, 1.grup dışında oluşturulan epinöral kılıf içerisine ADSCs uygulamasıyla rekonstrukte edildi. Fonksiyonel geri dönüş Walking Track analizi ile değerlendirildi. Siyatik sinir ve gastroknemius kasının histomorfometrik çalışmaları, postoperatif 4.ayında yapıldı. Çalışmamızın sonunda tüm klinik, biyokimyasal ve histomorfometrik analizler, ADSCs uygulamasının periferik sinir rejenerasyonuna, kök hücrenin nöral diferansiyasyonu, yeni akson üretimi ve myelin formasyonunu destekler şekilde olumlu etkisi olduğunu gösterdi. Bu etki muhtemelen ADSCs hücrelerin nöronal differansiyasyonu ve ortama salınan nörotopik faktörlere bağlıdır. ADSCs uygulamalarının periferik sinir rejenerasyonu üzerine olan etkilerinin klinik uygulamalara uyarlanabilmesi için ek çalışmalara ihtiyaç duyulmaktadır. Peripheral nerve damages are seen commonly after accidents, sharp objects injuries and particularly as a result of complete transection, delayed and incomplete surgery; they are dramatic views result in progressive muscle atrophy and ineducate functional recovery. Recently, new methods have started to take place besides the different surgery techniques for peripheral nerve repairing in literature. In this context, Stem cells applies have become one of the most popular topics of medicine recently, and hundreds of studies are being published every year. Stem cells, because of their pluripotentiality and unlimited capacity for self-renewal, project great promise for tissue engineering and are expected to allow significant advances for distinct reconstructive procedures. In our study, the effects of the green fluorescent protein (GFP) marked adipose derived mesenchymal stem cells (ADSCs) application were aimed research by functional analysis, histomorphometric and biochemical evaluation on experimental sciatic nerve injury to peripheral nerve regeneration. Experimental 4 Wistar groups (n:8) was designed: A nerve defect of 1 cm was constructed in the left sciatic nerve of the rats was reconstructed with ADSCs application in epineural cover built out of first group. Functional recovery was evaluated by walking track analysis. Histomorphometric studies of the sciatic nerves and gastrocnemius muscles were also performed in postoperative fourth month. In conclusion; All clinic, biochemical and histomorphometric analysis showed that ADSCs application has a possitive effect on supporting peripheral nerve regeneration, stem cells neural differantiation, axone production and myeline formation. This effect is probably due to the nörotrophic factors that are released from the ADSCs during activation. Further study will help to clarify the relative advantages of ADSCs applications on peripheral nerve regeneration. 25. Streptozotosinle Oluşturulan Diabetin Sıçan Periferik Sinirleri Üzerine Etkisinin Elektron Mikroskobik İncelenmesi

Bu çalışmada sıçanlarda streptozotosinle (STZ) oluşturulan diabetin akut etkisinin periferik sinir üzerinde oluşturduğu yapısal değişiklikler elektron mikroskop düzeyinde incelenmiştir. Çalışmada denekler, 24 erkek Wistar albino sıçan, kontrol grubu, Sham grubu, 15 ve 30 günlük diabet grubu olacak şekilde dört grup olarak ayrılmıştır. Kontrol grubuna hiçbir şey uygulanmazken, Sham grubuna STZ’nin çözündüğü sodyumsitrat, diabetik gruba ise sodyumsitrat içinde çözünmüş 45mg/kg STZ uygulanmıştır. Enjeksiyonu takiben 15 ve 30. günlerde sıçanlar yüksek doz anestezi altında sakrifiye edildi ve siyatik sinirlerinin medial bölgeleri çıkarılarak rutin elektron mikroskop takibine alındı. Doku kesitleri incelendiğinde hem 15 hem de 30 günlük diabetik grupta perinöryum ile sinir lifleri arasında belirgin bir açıklık meydana gelmiş ve miyelinli sinir liflerinde miyelin kılıf dejenerasyonları 30 günlük grupta daha belirgin bulunmuştur. Buna karşın, Schwann hücreleri ve miyelinsiz sinir lifleri normal yapıda gözlenmiştir. Sonuç olarak bu çalışmada sıçanlarda STZ ile oluşturulan diabet, n.ischiadicus’ta dejeneratanif değişiklikler oluşturmuş ve bu değişiklikler 30 günlük grupta daha belirgin bulunmuştur. In this study, structural changes caused by acute effects of STZ induced diabetes on peripheral nerves were investigated with electron microscope. Twenty four male Wistar Albino rats were divided into 4 groups: control, sham, 15 days and 30 days diabetic groups. Nothing applied to control group; sodium citrate, as STZ solvent, to Sham group and 45 mg/kg STZ solved in sodium citrate to diabetic group. 15 and 30 days after injections, rats were sacrified under high dose anesthesia, and sciatic nerves were extracted for electron microscopic examination. A evident gap between perineurium and nerve fibres were observed in both 15 and 30 days diabetic groups. And in 30 days group, myelin sheath degenerations were more obvious in myelinated nerve fibres. On the other hand, Schwann cells and unmyelinated nerve fibres appeared normal. Consequently, STZ induced diabetes were seen to cause degenerative changes in n. ischiadicus and those changes were more evident in 30 days diabetic group.
