Background: Conventional sperm selection methods, involving centrifugation, exert a detrimental effect on sperm DNA integrity due to mechanical stress. The recent noninvasive sperm selection device, CA0, based on live sperm sorting technology, facilitates the retrieval of highly motile sperm and minimizes sperm DNA fragmentation (SDF). This study was to investigate the impact of various sperm separation methods, with and without centrifugation, on embryo ploidy status. Methods: The retrospective study comprised 82 intracytoplasmic sperm injection (ICSI) cycles involving preimplantation genetic testing for aneuploidy (PGT-A) cases, with a focus on recruiting egg donation cycles to thoroughly investigate the impact of male factors. Two populations are classified based on semen quality: normozoospermic ([Formula: see text] = 33) and non-normozoospermic ([Formula: see text] = 49). Subjects were allocated to either swim-up (SU) or CA0. Preimplantation genetic testing results were recorded. Results: When comparing male characteristics between subgroups, no significant differences were observed except for a lower normal morphology rate in the CA0 group compared to SU (SU: 3 [3–4] vs. CA0: 2 [2–2.8], [Formula: see text] < 0.0001) in the non-normozoospermic cohort. There were no differences in female factors such as age and mature oocyte count (MII) number between subgroups, indicating that this model is ideal for assessing the impact of male factors on clinical outcomes. In the normozoospermic cohort, euploidy rates were similar between SU and CA0 (SU: 71.9% vs. CA0: 64.2%). However, in the non-normozoospermic cohort, CA0 showed a significantly higher euploidy rate compared to SU (SU: 53.6% vs. CA0: 74.2%) and a lower aneuploidy rate (SU: 37.1% vs. CA0: 25.8%). Additionally, CA0 minimized the incidence of mosaic embryos, whereas a mosaicism rate of 9.3% was observed with SU. This trend highlights CA0’s distinct advantage in optimizing outcomes for non-normozoospermic cases. Conclusions: CA0 is a reliable intervention to optimize paternal genetic quality before assisted insemination, thereafter effectively reducing the incidence of embryo aneuploidy associated with male factors.

While there is long‐standing interest in the role of inter‐ and transgenerational plasticity via the maternal line, it rarely has been studied via the paternal line. Thus, consequences of the paternal environment for parental fitness and for performance of offspring in the environments experienced by either fathers or mothers are not known. We studied the intergenerational plasticity (IGP) of the plant species Paeonia ostii (Paeoniaceae) and tested the hypothesis that exposure of fathers to environmental stress (i) reduces parental fitness and performance of offspring grown under non‐stressful conditions, but (ii) mitigates the negative effects of environmental stress on fitness of parents and performance of offspring.Crosses were made in a greenhouse within six families of P. ostii between parents grown under drought and in a mesic environment, and the offspring of each cross were grown under both dry and mesic conditions. Production and germination of seeds and morphological and physiological traits of offspring were measured as indicators of parental fitness and offspring performance, respectively.Paternal drought decreased seed number per fruit, except when maternal plants also were grown in drought conditions. Offspring drought decreased seedling performance. However, when fathers experienced drought this negative effect on the offspring was partly mitigated, in particular when mothers also had experienced drought. In contrast, offspring grown in mesic conditions had improved seedling performance, especially when either parent (or both) also were grown in mesic conditions. Such statistical differences remained when seed mass was included as a covariate.Overall, paternal pollen of P. ostii mediated IGP to drought almost as well as it did for maternal ovules. IGP was adaptive if environments remained constantly dry across generations but maladaptive if environments changed. Hence, under future climate changes, paternal IGP might be both a blessing and a curse, with the blessing occurring when the focal habitat becomes drier and pollen comes from already‐dry places, while the curse may dominate in predictably moist habitats surrounded by drier habitats. Parental care is a critical determinant of offspring fitness, and parents adjust their care in response to ecological challenges, including predation risk. The experiences of both mothers and fathers can influence phenotypes of future generations (transgenerational plasticity). If it is adaptive for parents to alter parental care in response to predation risk, then we expect F1 and F2 offspring who receive transgenerational cues of predation risk to shift their parental care behaviour if these ancestral cues reliably predict a similarly risky environment as their F0 parents. Here, we used three-spined sticklebacks (Gasterosteus aculeatus) to understand how paternal exposure to predation risk prior to mating alters reproductive traits and parental care behaviour in unexposed F1 sons and F2 grandsons. Sons of predator-exposed fathers took more attempts to mate than sons of control fathers. F1 sons and F2 grandsons with two (maternal and paternal) predator-exposed grandfathers shifted their paternal care (fanning) behaviour in strikingly similar ways: they fanned less initially, but fanned more near egg hatching. This shift in fanning behaviour matches shifts observed in response to direct exposure to predation risk, suggesting a highly conserved response to pre-fertilization predator exposure that persists from the F0 to the F1 and F2 generations. The evolutionary repercussions of parental effects—the impact of the developmental environment provided by parents on offspring—are often discussed as static effects that can have negative influences on offspring fitness that may even persist across generations. However, individuals are not passive recipients and may mitigate the persistence of parental effects through their behaviour. Here, we tested how the burying beetle, Nicrophorus orbicollis , a species with complex parental care, responded to poor parenting. We cross-fostered young and manipulated the duration of parental care received and measured the impact on traits of both F1 and F2 offspring to experimentally extricate the effect of poor parenting from other parental effects. As expected, reducing parental care negatively affected traits that are ecologically important for burying beetles, including F1 offspring development time and body size. However, F1 parents that received reduced care as larvae spent more time feeding F2 offspring than parents that received full care as larvae. As a result, both the number and mass of F2 offspring were unaffected by the developmental experience of their parents. Until recently, clinicians and researchers did not realize paternal exposures could impact child developmental outcomes. Indeed, although there is growing recognition that sperm carry a large amount of non‐genomic information and that paternal stressors influence the health of the next generation, toxicologists are only now beginning to explore the role paternal exposures have in dysgenesis and the incidence of congenital malformations. In this commentary, I will briefly summarize the few studies describing congenital malformations resulting from preconception paternal stressors, argue for the theoretical expansion of teratogenic perspectives into the male preconception period, and discuss some of the challenges in this newly emerging branch of toxicology. I argue that we must consider gametes the same as any other malleable precursor cell type and recognize that environmentally‐induced epigenetic changes acquired during the formation of the sperm and oocyte hold equal teratogenic potential as exposures during early development. Here, I propose the term epiteratogen to reference agents acting outside of pregnancy that, through epigenetic mechanisms, induce congenital malformations. Understanding the interactions between the environment, the essential epigenetic processes intrinsic to spermatogenesis, and their cumulative influences on embryo patterning is essential to addressing a significant blind spot in the field of developmental toxicology. The ayu, Plecoglossus altivelis, with an amphidromous life migrating between the sea and streams, is a commercially important fish for the inland fisheries of Japan. Despite intensive transplantation, seedlings originating from non-native population landlocked in the Biwa Lake have been less contributable to stock recovery. In fact, no apparent evidence of introgression from the released fish to the native population has been obtained. We hypothesized that the parental property from the landlocked strain would give some disadvantages to the offspring against saline osmotic pressure during their early life in the sea. The acute performance of newly hatched larvae in 180% artificial seawater at 26°C was evaluated by the parental combinations of amphidromous and landlocked strains. As a result, hatchlings from amphidromous parents lived longer than those from landlocked parents, whereas outbred hatchlings showed no difference in their survival period between landlocked and amphidromous paternities. Hatchlings from amphidromous parents lived longer than outbred half-sibs with landlocked paternity, while hatchlings from landlocked parents lived shorter than outbred half-sibs with amphidromous paternity. Reduced salinity tolerance in the landlocked population was proved to be paternally inherited, which can explain the disappearance of hybridized lineage through the failure in the early survival. Marine heatwave (MHW) events, characterized by periods of anomalous temperatures, are an increasingly prevalent threat to coastal marine ecosystems. Given the seasonal phenology of MHWs, the full extent of their biological consequences may depend on how these thermal stress events align with an organism's reproductive cycle. In organisms with more complex life cycles (e.g., many marine invertebrate species) the alignment of adult and larval environments may be an important factor determining offspring success, setting the stage for MHW events to influence reproduction and development in situ. Here, the influence of MHW-like temperatures on the early development of the California purple sea urchin, Strongylocentrotus purpuratus, were explored within the context of paternal thermal history. Based on temperature data collected during MHW events seen in Southern California from 2014--2020, adult urchins were acclimated to either MHW or non-MHW temperatures for 28 days before their sperm was used to produce embryos that were subsequently raised under varying thermal conditions. Once offspring reached an early larval stage, the impact of paternal and offspring environments were assessed on two aspects of offspring performance: larval size and thermal tolerance. Exposure to elevated temperatures during early development resulted in larger, more thermally tolerant larvae, with further influences of paternal identity and thermal history, respectively. The alignment of paternal and offspring exposure to MHW temperatures had additional positive benefits on larval thermal tolerance, but this tolerance significantly decreased when their thermal experience mismatched. As the highest recorded temperatures within past MHW events have occurred during the gametogenesis of many kelp forest benthic marine invertebrate species, such as the purple sea urchin, such parental mediated impacts may represent important drivers of future recruitment and population composition for these species. Individuals exposed to predation risk can produce offspring with altered phenotypes. Most work on predation-induced parental effects has focused on maternal effects or on generalized parental effects where both parents are exposed to risk. We conducted an experiment to measure and compare maternal and paternal effects on offspring phenotypes and test for interactions in those effects. We exposed 82 snails from 22 lines to control or predator cues and created line dyads with the four possible mating pairings of control and predator cue exposed individuals. We measured the resulting body masses, shell masses, shell shapes, and anti-predator behaviors of the offspring. We found some evidence that offspring were larger and heavier when the mother was exposed to predation cues, but that this effect was negated when the father was also exposed. The mass of offspring shells relative to their total mass was unaffected by parental treatments. Shell shape was marginally affected by maternal treatment, but not paternal treatment. Behavioral responses to cues were not affected by maternal or paternal treatments. Thoroughbred horses have been intensely raised for their athletic potential that is correlated with morphological parameters (i.e., body weight - BW, and withers height - WH). Optimum and consistent body development is aimed, but excessive growth rates may lead to the development of orthopedic diseases. This study aimed to generate growth rate curves and prediction models of Thoroughbred horses by analyzing BW and WH data collected monthly over 16 years of 378 animals (23.6 animals/year). The animals were checked from birth to 18 months (160 colts and 181 fillies) on a farm in the south of Brazil. A prediction performance ARIMA model was developed based on the BW and WH of the foals using a maximum and minimum range of 7320 observations. BW and WH were 54,1 kg and 102,5 cm at birth and 397,8 kg and 150,6 cm at 18 months of age, respectively. No differences were found between sex at any age. Moreover, we have established a nonlinear function for the growth curve and on average, foals were expected to get 7.4 times heavier and 1.5 times taller when animals were fully grown. Males showed greater BW uniformity than females, as females had a lower minimum BW than males. Significant statistical differences (P < 0,05%) were reported for BW and WH of foals between pairs of sires highlighting the kinship (paternal) effect on the animals' development. Seventy-two pairs of stallions showed statistical relevance for BW and 91 for WH. The ARIMA model produced a linear trend of BW and WH for the forecasted years. In conclusion, we recommend that careful sire selection and adequate health (i.e., parasite control, vaccination), and nutrition strategies must be adopted to achieve superior body growth as estimated by the predicting model (positive scenario). Environmental effects on gene expression and offspring development can be mediated by epigenetic modifications. It is well established that maternal diet influences DNA methylation patterns and phenotypes in the offspring; however, the epigenetic effects of paternal diet on developing offspring warrants further investigation. Here, we examined how a prepubertal methionine-enriched paternal diet affected sperm DNA methylation and its subsequent effects on embryo gene expression. Three treatment and three control rams were bred to seven ewes, and blastocysts were flushed for RNA extraction. Semen was collected from all rams and submitted for reduced representation bisulfite sequencing analysis. In total, 166 differentially methylated cytosines were identified in the sperm from treatment versus control rams. Nine genes were found to be differentially expressed in embryos produced from treatment versus control rams, and seven differentially methylated cytosines in the sperm were found to be highly correlated with gene expression in the embryos. When sons and daughters have different fitness costs and benefits, selection may favor deviations from an even offspring sex ratio. Most theories on sex ratio manipulation focus on maternal strategies and sex-biased maternal expenditure. Recent studies report paternal influences on both offspring sex ratio and postpartum sex-biased maternal expenditure. We used long-term data on marked kangaroos to investigate whether and how paternal mass and skeletal size, both determinants of male reproductive success, influenced (a) offspring sex in interaction with maternal mass and (b) postpartum sex-biased maternal expenditure. When mothers were light, the probability of having a son increased with paternal mass. Heavy mothers showed the opposite trend. A similar result emerged when considering paternal size instead of mass. Postpartum maternal sex-specific expenditure was independent of paternal mass or size. Studies of offspring sex manipulation or maternal expenditure would benefit from an explicit consideration of paternal traits, as paternal and maternal effects can modulate each other. Recently, research into non-genomic mechanisms of inheritance has led to the recognition that epigenetic factors carried in sperm also significantly impact the health of future generations. However, although researchers have described a range of potential epigenetic signals transmitted through sperm, we have yet to obtain a mechanistic understanding of how these paternally-inherited factors influence offspring development and modify life-long health. In this endeavor, the emerging influence of the paternal epigenetic program on placental development, patterning, and function may help explain how a diverse range of male exposures induce comparable intergenerational effects on offspring health. During pregnancy, the placenta serves as the dynamic interface between mother and fetus, regulating nutrient, oxygen, and waste exchange and coordinating fetal growth and maturation. Studies examining intrauterine maternal stressors routinely describe alterations in placental growth, histological organization, and glycogen content, which correlate with welldescribed influences on infant health and adult onset of disease. Significantly, the emergence of similar phenotypes in models examining preconception male exposures indicates that paternal stressors transmit an epigenetic memory to their offspring that also negatively impacts placental function. Like maternal models, paternally programmed placental dysfunction exerts life-long consequences on offspring health, particularly metabolic function. Here, focusing primarily on rodent models, we review the literature and discuss the influences of preconception male health and exposure history on placental growth and patterning. We emphasize the emergence of common placental phenotypes shared between models examining preconception male and intrauterine stressors but note that the direction of change frequently differs between maternal and paternal exposures. We posit that alterations in placental growth, histological organization, and glycogen content broadly serve as reliable markers of altered paternal developmental programming, predicting the emergence of structural and metabolic defects in the offspring. Finally, we suggest the existence of an unrecognized developmental axis between the male germline and the extraembryonic lineages that may have evolved to enhance fetal adaptation. Environmental conditions experienced by parent plants can influence offspring performance through parental effects induced by DNA methylation. The offspring can also be influenced by environmental conditions experienced by their parents via soil legacy effects due to plant‐mediated changes in the composition of soil microbes. These two effects are likely to act simultaneously, but empirical evidence for combined effects is limited.We conducted a two‐phase experiment with five genotypes of a clonal plant Hydrocotyle vulgaris. In the first phase, we grew parent plants of each genotype under two light conditions (ambient vs. shade) and two DNA demethylation treatments [treated with water vs. 5‐azacytidine (5‐azaC)]. We then collected soils and clonal offspring for each genotype from each of these four treatments and measured soil (a)biotic properties. In the second phase, we grew the offspring from each of the four treatments in the four different soils, under the two light conditions.When grown under ambient light condition and in soil from ambient parents, offspring produced by ambient parents grew larger than offspring produced by shaded parents when the parents were treated with water. This difference was smaller when the parents were treated with 5‐azaC, and disappeared when the offspring were grown in soil from shaded parents. The growth difference was also observed when the offspring were grown under shaded condition and in soil from shaded parents. However, this difference was greater when the parents were treated with 5‐azaC, and disappeared when the offspring were grown in soil from ambient parents. Moreover, offspring growth was associated with fungal composition and total phosphorus of the soil in which the parents had grown.Our results show, for the first time, that light condition experienced by parents can influence offspring responses to light through both parental effects and soil legacies. The parental effects were mediated by changes in DNA methylation and the soil legacies were due to plant‐mediated changes in a combination of soil biotic and abiotic properties. Embryonic exposure through maternally transferred pollutants can affect embryo vitality, survival, and health. Reptiles face global declines and are sensitive to embryonic pollutant exposure. Yet, they are often neglected in pollution risk assessment and conservation. We analyzed maternal transfer of organic pollutants in reptiles through a systematic extraction, homogenization, and integration of published data on organic pollutants measured in mother-egg pairs into a comprehensive database (DOI:10.5281/zenodo.10900226), complemented with molecular physical-chemical properties of the pollutants. Over four decades, 17 publications provided 19,955 data points shifting from legacy to emerging contaminants although research on newer contaminants lags regulatory and societal demands. Challenges including taxonomic bias, heterogeneity in sampled tissues, and 73% of censored data complicate comparative analyses. However, significant opportunities were identified including the use of the turtle Malachlemys terrapin and snake Enhydris chinensis as flagship species where a large amount of data is available across tissues (allowing investigation into physiological relations) and compounds (allowing insights into maternal transfer across the chemical universe). Data on other freshwater and marine turtles provide the possibility of exploring taxonomic patterns in this subgroup. The analysis, integrated database, and discussion present opportunities for research in an era where science needs to achieve more with limited wildlife data.

Pregnancy losses negatively affect the cattle industry, impacting economic indices and consequently the entire production chain. Early embryonic failure has been an important challenge in the embryo industry because proper identification of embryo death at the beginning of gestation is difficult. This review aimed to provide a better understanding on reproductive failure and the relationship between early embryonic loss and different reproductive biotechniques. This review also considers insights and possible strategies for reducing early embryonic loss. In many species, parental age at reproduction can influence offspring performance and life span, but the direction of these effects and the traits affected vary among studies. Data on parental age effects are still scarce in noncaptive populations, especially insects, despite species such as fruit flies being models in laboratory-based aging research. We performed a biologically relevant experimental manipulation of maternal and paternal age at reproduction of antler flies (Protopiophila litigata) in the laboratory and tracked the adult life span and reproductive success of their male offspring released in the wild. Increased paternal, but not maternal, age somewhat increased sons' adult life span, while parental ages did not influence sons' mating rate or reproductive senescence. Our results indicate that while parental age effects do exist in an insect in the field, they may be beneficial in such a short-lived animal, in contrast to results from most wild vertebrates and laboratory invertebrates. The use of different sires influences in vitro embryo production (IVP) outcome. Paternal effects are observed from the first cleavages until after embryonic genome activation (EGA). Little is known about the mechanisms that promote in vitro fertility differences, even less about the consequences on embryo development. Therefore, this study aimed to evaluate the paternal effect at fertilization, embryo developmental kinetics, gene expression and quality from high and low in vitro fertility bulls. A retrospective analysis for bull selection was performed using the In vitro Brazil company database from 2012 to 2015. The dataset was edited employing cleavage and blastocyst rates ranking a total of 140 bulls. Subsequently, the dataset was restricted by embryo development rate (blastocyst/cleaved rate) and ten bulls were selected as high (HF; n = 5) and low (LF; n = 5) in vitro fertility groups. IVP embryos derived from high and low fertility bulls were classified according to their stage of development (2 cells, 3–4 cells, 6 cells, 8–16 cells), at 24, 36, 48, 60, 72 hpi, respectively, to evaluate embryo kinetics. Pronuclei formation (24 hpi), cleavage rate (Day 3), development rate, and blastocyst morphology (Grade I and II – Day 7) were also assessed, as well as the abundance of 96 transcripts at 8–16 cell stage and blastocysts. There was no difference in early embryo kinetics (P > 0.05), and cleavage rate (HF = 86.7%; LF = 84.9%; P = 0.25). Nevertheless, the fertilization rate was higher on HF (72%) than LF (62%) and the polyspermy rate was lower on HF compared to LF (HF:16.2% LF:29.2%). As expected, blastocyst rate (HF = 29.4%; LF = 16.0%; P < 0.0001) and development rate (HF = 33.9% LF = 18.9%; P < 0.0001) were higher in HF than LF. At the 8–16 cell stage, 22 transcripts were differentially represented (P ≤ 0.05) between the two groups. Only PGK1 and TFAM levels were higher in HF while transcripts related to stress (6/22, ∼27%), cell proliferation (6/22, ∼27%), lipid metabolism genes (5/22, ∼23%), and other cellular functions (5/22, ∼23%) were higher on LF embryos. Blastocysts had 9 differentially represented transcripts (P ≤ 0.05); being only ACSL3 and ELOV1 higher in the HF group. Lipid metabolism genes (3/9, 33%) and other cellular functions (6/9, 67%) were higher in the LF group. In conclusion, the timing of the first cleavages is not affected by in vitro bull fertility. However, low in vitro fertility bulls presented higher polyspermy rates and produced 8–16 cells embryos with higher levels of transcripts related to apoptosis and cell damage pathways compared to high in vitro fertility ones. The aim of this study was to examine the effects of long-term dietary supplementation of young Nellore bulls with rumen-protected polyunsaturated fatty acids (PUFAs) and of the inclusion of catalase in the semen extender on semen quality, in vitro sperm fertilizing ability, and intracytoplasmic lipid content in the resulting embryos. Twelve Nellore bulls were supplemented with rumen-protected PUFAs or with a basal diet from 14 to 24 months of age. The semen was collected at the end of supplementation. For cryopreservation, the ejaculate was divided into two equal volumes and catalase was added to the extender in one of the fractions. Thus, the experimental design consisted of a 2 × 2 factorial scheme with two diets (control and PUFA) and two extenders (Cat+ and Cat-). Total motility and the percentage of rapid cells in fresh semen were negatively affected by dietary supplementation with PUFAs (P < 0.05), but these effects did not persist after freezing. The frozen/thawed semen of animals fed PUFAs exhibited an increase in the percentages of damaged plasma and acrosomal membranes, as well as an increase in the proportion of lipids ions at m/z 578 and m/z 757 detected by MALDI-MS. Nevertheless, there was no effect of the treatments on in vitro embryo development. However, embryos derived from bulls supplemented with PUFAs exhibited higher lipid accumulation compared to control (P < 0.05). The reproductive traits are lowly heritable but can be improved through direct genetic selection. Interestingly, the breeding values for GESS on insemination success traits (CR and NRR56) are unfavorably and significantly correlated with some production, health, and type breeding values (ranging from −0.449 to 0.274), while the GESS values on calving traits (CE, SB, and GL) are usually favorably associated with those traits (ranging from −0.493 to 0.313). One hundred sixty-two significant single-nucleotide polymorphisms (SNPs) and their surrounding protein-coding genes were identified as significantly associated with GESS and GED, respectively. This study explored the dynamic effects between fathers and mothers' education levels on the language outcomes of Mandarin-speaking preschoolers with hearing loss who had received early intervention in Taiwan. Data was collected using the revised preschooler language assessment from 113 children with hearing loss; the children in this study were aged between 37–72 months. The dynamic relationships among parental education levels, audio logical histories, and language performance with respect to early intervention were investigated using path analysis. The effect of maternal education level on the outcome of language comprehension was fully mediated by paternal education level, but the effect of parental education level was not revealed in relation to oral expression. Furthermore, the effect of the degree of hearing loss on language performance outcomes was partially mediated by the length of time the child was enrolled in early intervention, implying that the degree of effects of parental education level or difficulties of children could be remedied through early intervention. Imprinting is a phenomenon that alters the expression of genes according to the parental origin of their alleles. A quantitative form to evaluate the imprinting effect is known as parent-of-origin effect. Our aim with this work is to identify parent-of-origin effects that influence the number of oocytes and embryos in Gir dairy cattle. A dataset with 17,526 Ovum Pick Up observations from 1641 Gir donors was used to estimate parent-of-origin effects for the traits number of total oocytes (TO), number of viable oocytes (VO) and number of embryos (EM). To identify parent-of-origin effects, dam and sire gametic effects were included, individually or together, in an animal model for TO, VO and EM traits. For TO, inclusion of paternal origin effects in the model was significant (P < 0.05), and explained 6 % of the total phenotypic variance. For VO and EM no significant parent-of-origin effects were found for either parental line. In conclusion, paternal effects appear to influence the total oocyte production in the Gir cattle breed. Transgenerational plasticity, which occurs when the environment experienced by parents changes the phenotype of offspring, is widespread in animal and plant species. Both maternal and paternal environments can underlie transgenerational plasticity, but experimental studies unraveling how their effects interact together and with the personal (both developmental and immediate) environments are still rare. Yet unraveling these interactions is fundamental to understanding how offspring integrate past and present environmental cues to produce adaptive phenotype. Using the hermaphroditic and freshwater snail Physa acuta, we tested how predator cues experienced by offspring, mothers and fathers interact to shape offspring anti-predator behavior. We raised a first generation of snails in the laboratory with or without chemical predator cues and realized full-factorial crosses to disentangle maternal and paternal cues. We then raised the second generation of snails with or without predator cues and assessed, when adults, their escape behavior in two immediate environments (with or without predator cues) and activity in the immediate environment without predator cues. We found that personal, maternal, and paternal predator cues interacted to shape offspring escape behavior and activity. Firstly, for escape behavior, snails integrated the cues from developmental and parental environments only when exposed to predator cues in their immediate environment, suggesting that personal immediate experience must corroborate the risky parental environment to reveal transgenerational plasticity. For activity, this same hypothesis helps explain why no clear pattern of transgenerational plasticity was revealed, as activity was only measured without predator cues in the immediate environment. Secondly, a single maternal exposure to predator cues decreased offspring escape behavior while a single paternal exposure had no effect, surprisingly demonstrating sex-specific transgenerational plasticity for a simultaneous hermaphroditic species. Thirdly, when both mother and father were exposed, paternal cues were integrated by offspring according to their own developmental environment. The paternal exposure then mitigated the reduction in escape behavior due to the maternal exposure only when offspring developed in control condition. Overall, our study highlighted complex patterns of sex-specific transgenerational plasticity resulting from non-additive interactions between parental, developmental and immediate experiences.

Transgenerational plasticity (TGP) occurs when the environment encountered by one generation (F0) alters the phenotypes of one or more future generations (e.g. F1 and F2). Sex selective TGP, via specific lineages or to only male or female descendants, has been underexplored in natural systems, and may be adaptive if it allows past generations to fine‐tune the phenotypes of future generations in response to sex‐specific life‐history strategies.We sought to understand if exposing males to predation risk can influence grandoffspring via sperm in three‐spined stickleback Gasterosteus aculeatus. We specifically tested the hypothesis that grandparental effects are transmitted in a sex‐specific way down the male lineage, from paternal grandfathers to F2 males.We reared F1 offspring of unexposed and predator‐exposed F0 males under 'control' conditions and used them to generate F2s with control grandfathers, a predator‐exposed maternal grandfather (i.e. predator‐exposed F0 males to F1 daughters to F2s), a predator‐exposed paternal grandfather (i.e. predator‐exposed F0 males to F1 sons to F2s) or two predator‐exposed grandfathers. We then assayed male and female F2s for a variety of traits related to antipredator defence.We found little evidence that transgenerational effects were mediated to only male descendants via the paternal lineage. Instead, grandpaternal effects depended on lineage and were mediated largely across sexes, from F1 males to F2 females and from F1 females to F2 males. When their paternal grandfather was exposed to predation risk, female F2s were heavier and showed a reduced change in behaviour in response to a simulated predator attack relative to grandoffspring of control, unexposed grandparents. In contrast, male F2s showed reduced antipredator behaviour when their maternal grandfather was exposed to predation risk. However, these patterns were only evident when one grandfather, but not both grandfathers, was exposed to predation risk, suggesting the potential for non‐additive interactions across lineages.If sex‐specific and lineage effects are common, then grandparental effects are likely underestimated in the literature. These results draw attention to the importance of sex‐selective inheritance of environmental effects and raise new questions about the proximate and ultimate causes of selective transmission across generations. Background: Parent's history of atopic traits increases the risk of the same traits in their children, but mother's history may confer an increased risk compared to father's history. Objective: To investigate parent‐specific effects on risk of developing allergic sensitization and asthma in childhood. Methods: We included 685 parent‐child trios from the Copenhagen Prospective Studies on Asthma in Childhood 2010 (COPSAC2010) cohort. Parent's asthma was assessed by structured interviews and child's asthma was diagnosed prospectively at regular visits to the COPSAC clinic until age 6. Specific IgE and total IgE levels were measured in parents and children by age 0.5, 1.5 and 6 years. Associations between parent and child disease traits were analyzed using general estimating equations model adjusted for breastfeeding and maternal smoking during 3rd trimester. Results: Maternal compared to paternal elevated specific IgE increased the child's risk of elevated specific IgE from 0‐6 years: adjusted odds ratio (aOR)mother = 1.49 [1.09‐2.03], P =.01 and aORfather = 1.32 [0.96‐1.82], P =.08. Maternal elevated total IgE also increased the child's risk of elevated total IgE: adjusted relative risk (aOR)mother = 4.32 [1.51‐10.8], P <.01, while a trend was observed for paternal total IgE: aORfather = 2.01 [0.76‐4.82], P =.13. Individual time point analyses showed that the maternal effect was strongest in early life, whereas the parental effects were comparable by age 6. A similar parent‐specific pattern was observed for the child's risk of asthma. Conclusions and Clinical Relevance: The effect of mother's history of atopic traits on the child's risk of developing the same traits in early childhood was stronger than the effect from father's history, which was not evident before age 6. In a large majority of animal species, the only contribution of males to the next generation has been assumed to be their genes (sperm). However, along with sperm, seminal plasma contains a wide array of extracellular factors that have many important functions in reproduction. Yet, the potential intergenerational effects of these factors are virtually unknown. We investigated these effects in European whitefish (Coregonus lavaretus) by experimentally manipulating the presence and identity of seminal plasma and by fertilizing the eggs of multiple females with the manipulated and unmanipulated semen of several males in a full‐factorial breeding design. The presence of both own seminal plasma and foreign seminal plasma inhibited sperm motility, and the removal of own seminal plasma decreased embryo survival. Embryos hatched significantly earlier after both semen manipulations than in control fertilizations; foreign seminal plasma also increased offspring aerobic swimming performance. Given that our experimental design allowed us to control potentially confounding sperm‐mediated (sire) effects and maternal effects, our results indicate that seminal plasma may have direct intergenerational consequences for offspring phenotype and performance. This novel source of offspring phenotypic variance may provide new insights into the evolution of polyandry and mechanisms that maintain heritable variation in fitness and associated female mating preferences. Parental experience alters survival‐related phenotypes of offspring in both adaptive and nonadaptive ways, yielding rapid inter‐ and transgenerational fitness effects. Yet, fitness comprises survival and reproduction, and parental effects on mating decisions could alter the strength and direction of sexual selection, affecting long‐term evolutionary trajectories. We used a full factorial design in which threespine stickleback (Gasterosteus aculeatus) mothers, fathers, both, or neither were exposed to a model predator at developmentally appropriate times to test for predator‐induced maternal, paternal, and joint parental effects on daughters' mating behavior. We tested the responsiveness, preferences, and mate choices of adult daughters in no‐choice trials with wild‐caught males who had varied sexual signals. Maternal and paternal predator exposure independently yielded daughters who preferred males who were intermediate in conspicuousness (with duller nuptial coloration and who courted less vigorously), relaxing the typical preference for the most conspicuous males. The combined effects of maternal and paternal predator exposure were not cumulative; when both parents were predator exposed, single‐parent effects on mate preferences were reversed. Thus, we cannot assume that maternal and paternal effects additively combine to produce "parental" effects. Further, joint parental predator exposure yielded daughters who were three times less likely to mate at all. Stress‐induced intergenerational parental effects on reproductive decisions such as those observed here may potentiate rapid transgenerational responses to novel and changing mating environments. Intergenerational fitness effects on offspring due to the early life of the parent are well studied from the standpoint of the maternal environment, but intergenerational effects owing to the paternal early life environment are often overlooked. Nonetheless, recent laboratory studies in mammals and ecologically relevant studies in invertebrates predict that paternal effects can have a major impact on the offspring's phenotype. These nongenetic, environment‐dependent paternal effects provide a mechanism for fathers to transmit environmental information to their offspring and could allow rapid adaptation. We used the bank vole Myodes glareolus, a wild rodent species with no paternal care, to test the hypothesis that a high population density environment in the early life of fathers can affect traits associated with offspring fitness. We show that the protein content in the diet and/or social environment experienced during the father's early life (prenatal and weaning) influence the phenotype and survival of his offspring and may indicate adaptation to density‐dependent costs. Furthermore, we show that experiencing multiple environmental factors during the paternal early life can lead to a different outcome on the offspring phenotype than stimulated by experience of a single environmental factor, highlighting the need to study developmental experiences in tandem rather than independent of each other. Intake of milk fat in human nutrition is important because of unsaturated and especially essential fatty acids (FAs), linoleic and α-linolenic acid, and conjugated linoleic acid (CLA), which is found only in meat and milk of ruminants. The objective of our study was to investigate the effect of paternal bulls on fatty acids composition in milk fat of dairy cows of different breeds. The milk samples were taken in total from 299 dairy cows from 11 dairy farms. In experiment Holstein (H, n = 105), Red Holstein (R, n = 120) and Pinzgau (P, n = 74) breeds originated from different bulls were used. Individual milk samples were analyzed for fatty acids in milk fat using gas chromatography (apparatus GC Varian 3800, Techtron, USA), using FID detector in capillary column Omegawax 530; 30 m. In the chromatography records there were identified 54 fatty acids inclusive of particular isomers. Their relative proportions were expressed in percent's (%). Among the studied breeds, the highest content of conjugated linoleic acid (CLA) - 0.67%, essential FAs (EFA) - 2.98%, monounsaturated FAs (MUFA) - 25.84% and the lowest atherogenic index (AI) - 3.10 was at breed P. Within this breed there was high variability and daughters of bull COS1 achieved significant above-average values of CLA content 1.07%, EFA 3.71%, MUFA 29.93% and under breed average AI = 2.40. The group of daughters of NOB3 was significant lower in CLA, 0.50% as compared with an average of P breed. . From the breed H bull MTY2 showed significantly higher value of 0.62% CLA, EFA 3.42%, 34.29% MUFA and lower value of AI, 1.9 as compared to H breed average. Statistically significantly lower levels of CLA 0.29% and 21.46% MUFA and higher AI 3.72 in milk fat of his daughters, bull STY3 may be considered as potential worser of these properties. At the breed R bull MOR506 showed in compar to the breed average significantly higher value of the EFA 3.80% and also the higher content of CLA 0.50% and MUFA 25.09%, resulting in statistically significant lower AI = 2.91. Bull MOR506 could be considered as potential improver of milk fat composition. The above described variability in the composition of milk fat of dairy cows and the subsequent relationships between these values suggest that the selection of the bull according to the fatty acid composition of milk fat may be considered.

In humans, pronounced age differences between parents have deleterious fitness consequences. In particular, the number of children is lower when mothers are much older than fathers. However, previous analyses failed to disentangle the influence of differential parental age per se from a direct age effect of each parent. In this study, we analyse the fitness consequences of both parental age and parental age differences on litter size and offspring survival in two closely related species of lemurs living in captivity. As captive lemurs do not choose their reproductive partner, we were able to measure litter size and offspring survival across breeding pairs showing a wide range of parental age differences. However, we demonstrated that the effect of the parental age difference on litter size was fully accounted for by female reproductive senescence because females mating with much younger males were old females. On the other hand, both parental age difference and female reproductive senescence influenced offspring survival. Our results emphasize the importance of teasing apart the effect of parental reproductive senescence when investigating the health and fitness consequences of parental age differences and also provide new insights for conservation programmes of endangered species. Abstract: While it has been recognized for many years that parental condition can influence offspring performance, recent research on adaptive parental effects has focused primarily on anticipatory effects, whereby parents adjust the phenotype of their offspring for the anticipated environment. Here, we make the case that condition transfer is a widespread and important type of adaptive parental effect, and endeavour to clarify how such effects should be interpreted and studied. Some authors have suggested that condition‐transfer effects result simply from resource limitation constraints (passive condition transfer, or transmissive effects). However, condition transfer can also reflect evolved parental investment strategies (active condition‐transfer effects). In some species, such strategies can involve cryptic mechanisms such as epigenetic inheritance. As recently shown in this journal by Engqvist and Reinhold, condition‐transfer effects can be obscured by anticipatory effects and interactions between the effects of parental and offspring environments. Nonetheless, we argue that these complications can be largely overcome by examining a broad range of ecologically relevant environments in both parental and offspring generations. This can be accomplished by adapting a powerful methodology from the nutritional sciences—the geometric framework—to research on parental effects. [ABSTRACT FROM AUTHOR]

Differential allocation (DA) is the adaptive adjustment of reproductive investment (up or down) according to partner quality. A lack of theoretical treatments has led to some confusion in the interpretation of DA in the empirical literature. We present a formal framework for DA that highlights the nature of reproductive benefits versus costs for females mated to males of different quality. Contrary to popular belief, analytical and stochastic dynamic models both show that additive benefits of male quality on offspring fitness have no effect on optimal levels of female investment and thus cannot produce DA. Instead, if offspring fitness is affected multiplicatively by male quality, or male quality affects the female cost function, DA is expected because of changes in the marginal benefits or costs of extra investment. Additive male quality effects on the female cost function can cause a novel form of weak DA, because reduced costs can slightly favor current over future reproduction. Combinations of these distinct effects in more realistic model scenarios can explain various patterns of positive and negative DA reported for different species and mating systems. The modification of pre- and postnatal development conferred by immunogenic stimulation of mothers provides a population-level adaptation mechanism for non-genetic transfer of maternal experiences to progeny. However little is known about the transmission of paternal immune experiences to offspring. Here, we show that immune priming of males 3-9 days before mating affects the growth and humoral environment of developing embryos of outbred (ICR) and inbred (C57BL and BALB/c) mice. Antigenic stimulation of fathers caused a significant increase in embryonic bodyweight as measured on Day 16 of pregnancy and altered other gestation parameters, such as feto-placental ratio. Pregnant females mated with immunised males were also characterised by changes in humoral conditions as shown by measurements of blood and amniotic progesterone, testosterone and granulocyte-macrophage colony-stimulating factor (GM-CSF) cytokine concentrations. Although the ecological and evolutionary importance of environmentally induced parental effects is now widely recognized, such effects are still typically studied by contrasting just two environments in a single parental sex. The importance of trans-generational effects in shaping an individuals' phenotype and fitness, and consequently even impacting population dynamics is increasingly apparent. Most of the research on trans-generational effects still focuses on plants, mammals, and birds. In the past few years, however, increasing number of studies, especially on maternal effects, have highlighted their importance also in many insect systems. Lepidoptera, specifically butterflies, have been used as model systems for studying the role of phenotypic plasticity within generations. As ectotherms, they are highly sensitive to environmental variation, and indeed many butterflies show adaptive phenotypic plasticity in response to environmental conditions. Here, we synthesize what is known about trans-generational effects in Lepidoptera, compile evidence for different environmental cues that are important drivers of trans-generational effects, and point out which offspring traits are mainly impacted. Finally, we emphasize directions for future research that are needed for better understanding of the adaptive nature of trans-generational effects in Lepidoptera in particular, but potentially also in other organisms. Telomere-capping complexes (TCCs) protect the ends of linear chromosomes from illegitimate repair and end-to-end fusions and are required for genome stability. The identity and assembly of TCC components have been extensively studied, but whether TCCs require active maintenance in nondividing cells remains an open question. Here we show that Drosophila melanogaster requires Deadbeat (Ddbt), a sperm nuclear basic protein (SNBP) that is recruited to the telomere by the TCC and is required for TCC maintenance during genome-wide chromatin remodeling, which transforms spermatids to mature sperm. Ddbt-deficient males produce sperm lacking TCCs. Their offspring delay the initiation of anaphase as early as cycle 1 but progress through the first two cycles. Persistence of uncapped paternal chromosomes induces arrest at or around cycle 3. This early arrest can be rescued by selective elimination of paternal chromosomes and production of gynogenetic haploid or haploid mosaics. Progression past cycle 3 can also occur if embryos have reduced levels of the maternally provided checkpoint kinase Chk2. The findings provide insights into how telomere integrity affects the regulation of the earliest embryonic cell cycles. PREMISE OF the STUDY: Competition among pollen grains from a single donor is expected to increase the quality of the offspring produced because of the recessive deleterious alleles expressed during pollen-tube growth. However, evidence for such an effect is inconclusive; a large number of studies suffer from confounding variation in pollen competition with variation in pollen load. METHODS: In this study, we tested the effect of pollen competition on offspring performance independently of pollen-load variation. We compared seed mass and early seedling performance in Dalechampia scandens (Euphorbiaceae) between crosses in which variation in pollen competition was achieved, without variation in pollen load, by manipulating the dispersion of pollen grains on the stigmas. KEY RESULTS: Despite a large sample size (211 crosses on 20 maternal plants), we failed to find an effect of pollen competition on seed characteristics or early seedling performance. Paternal effects were always limited, and pollen competition never reduced the within-father (residual) variance. conclusion: These results suggest that limited within-donor variation in genetic quality of pollen grains reduces the potential benefits of pollen competition in the study population. The lack of paternal effects on early sporophyte performance further suggests that benefits of pollen competition among pollen from multiple donors should be limited as well, and it raises questions about the significance of pollen competition as a mechanism of sexual selection. Purpose: Various time-lapse studies have postulated embryo selection criteria based on early morphokinetic markers. However, late paternal effects are mostly not visible before embryonic genome activation. The primary objective of this retrospective study was to investigate whether those early morphokinetic algorithms investigated by time-lapse imaging are reliable enough to allow for the accurate selection of those embryos that develop into blastocysts, while of course taking into account the correlation with the type of injected spermatozoa. Methods: During a period of 18 months, a total of 461 MII oocytes from 43 couples with severe male factor infertility and previous 'external' IVF failures after cleavage-stage embryo transfer (ET) were fertilized by intracytoplasmic morphologically selected sperm injection (IMSI). Thereof, 373 embryos were monitored in a time-lapse incubator until ET on day 5. Blastocyst outcome in combination with three previously postulated MKc (cc2: t3-t2, 5-12 h; t3, 35-40 h; t5, 48-56 h) and the morphology of the selected sperm were analyzed. Results: A significant increase in the rate of blastocysts (54.0 vs. 36.3 %; P < 0.01) and top blastocysts (25.3 vs. 10.8 %; P < 0.001) was observed in the group of those meeting all three morphokinetic criteria (MKc3). However, MKc3 were only met in 23.3 % of all embryos. Moreover, TBR was influenced by the type of injected spermatozoa. In both groups, TBR decreased dramatically (MKc3, 35.0 vs. 17.0 %; MKc < 3, 14.2 vs. 8.4 %) when class II/III sperm instead of class I were injected. Conclusion: Early morphokinetic parameters might give some predictive information but fail to serve as a feasible selective tool for the prediction of blastocyst development given the influence of the type of spermatozoa injected. Insect polyandry often selects for increased male investment in ejaculates as it generates both sperm competition and the potential for cryptic (postcopulatory) female choice, a process males will seek to influence. Among the Coccinellidae, allomonal factors in male ejaculates are known to stimulate female fecundity and fertility and confer positive effects on the developmental phenotypes of progeny. We used the subtropical species Eriopis connexa (Germar) to test the hypothesis that male epigenetic factors will be diminished by sexual activity. We subjected virgin females to either a single mating, or 10 daily matings with the same male or different ones, either virgin or previously mated. There was no effect of male mating status or male novelty on female fecundity or fertility, but multiple matings with different virgin males resulted in faster production of clutches during the first 10 d of oviposition. When females were permitted only a single mating, their mean clutch size was diminished by males that had been previously mated two or more times and they took longer to produce 10 clutches, although egg viability was unaffected. Paternal effects did not vary with paternal mating history and progeny development was similar between first and tenth clutches, indicating an absence of the age-specific maternal effects observed in certain other species. The habitats occupied by this subtropical species provide prey for reproduction throughout the year and lack the strong seasonal patterns of prey availability that have been hypothesized to drive the evolution of age-specific maternal effects observed in some temperate species.
