3-chloro-1,2-propanediol (3-MCPD) is a member of the group of pollutants known as chloropropanols and is considered a genotoxic carcinogen. Due to the occurrence of 3-MCPD, which cannot be avoided in multiplexed food processes, it is necessary to explore novel agents to reduce or prevent the toxicity of 3-MCPD. Many recent studies on boron compounds reveal their superior biological roles such as antioxidant, anticancer, and antigenotoxic properties. In the current investigation, we have evaluated in vitro cytotoxic, oxidative, and genotoxic damage potential of 3-MCPD on human whole blood cultures and the alleviating effect of boric acid (BA) and borax (BX) for 72 h. In our in vitro experiments, we have treated blood cells with BA and BX (2.5, 5, and 10 mg/L) and 3-MCPD (at IC50 of 11.12 mg/l) for 72 h to determine the cytotoxic damage potential by using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) and lactate dehydrogenase (LDH) release assays. Oxidative damage was assessed using total antioxidant capacity (TAC) and malondialdehyde (MDA) levels. Genotoxicity evaluations were performed using chromosome aberrations (CAs) and 8-hydroxy deoxyguanosine (8-OHdG) assays. The result of our experiments showed that the 3-MCPD compound induced cytotoxicity, oxidative stress, and genotoxicity in a clear concentration-dependent manner. BA and BX reduced cytotoxicity, oxidative stress, and genotoxicity induced by 3-MCPD. In conclusion, BA and BX are safe and non-genotoxic under the in vitro conditions and can alleviate cytotoxic, oxidative, and genetic damage induced by 3-MCPD in the human blood cells. Endometrial cancer is one of the most common malignancies in women. Sphingolipids, a group of lipids, play a key role in cancer biology. Cancer cells often exhibit abnormal redox homeostasis characterized by elevated levels of reactive oxygen species (ROS). Emerging evidence suggests that ceramides are involved in inhibiting proliferation and inducing apoptosis through ROS production. However, there is no data on the relationship between sphingolipid metabolism and oxidative status in endometrial cancer. The present study aims to assess the content of individual sphingolipids and oxidative status in healthy women and those with endometrial cancer. Sphingolipid analysis was performed using mass spectrometry. Total oxidative status (TOS) and total antioxidant capacity (TAC) were assessed colorimetrically. Our results showed a significant increase in the levels of all measured sphingolipids in cancer tissues compared to healthy endometrium. Additionally, a significant decrease in the S1P/ceramide ratio (sphingolipid rheostat) was observed in cancer patients, particularly for C14:0-Cer, C16:0-Cer, C18:1-Cer, C22:0-Cer, and C24:0-Cer. Furthermore, increased TOS and decreased TAC were found in cancer patients compared to healthy women. Significant correlations were observed between the levels of individual sphingolipids and oxidative status, with the strongest correlation noted between C22:0-Cer and TOS (r = 0.64). Live prey used by the aquaculture industry are usually poor in some essential nutrients including long chain polyunsaturated fatty acids (LC-PUFA) and must be enriched to improve their nutritional value prior to larval feeding. Standard enrichment protocols are commonly based on lipid emulsions, being associated to a high oxidative stress condition. The combination of microalgae and lipid emulsion can palliate this situation, where the oxidative stress can be partially compensated by the antioxidant compounds present in microalgae. The maintenance of living microalgae in culture facilities is laborious, and the produced biomass may present fluctuating properties, leading to a serious bottleneck in the cultivation of live prey. Hence, substitutes for live microalgae including pastes or dried formats are receiving increasing research attention due to its nutritional stability, longer shelf-life and easy handling. In this study four different microalgae formats combined with a lipid emulsion are tested as enrichment products for Brachionus plicatilis and Artemia. Thus, fresh, frozen and spray-dried Navicula salinicola (NFRE, NFRO and NSD, respectively), and spray-dried Isochrysis galbana (ISD) were mixed with a commercial oil concentrate (IncromegaTM) or a marine lecithin (LC 60®), and added for 5 h to the rotifer or Artemia culture media. The antioxidant capacity of the microalgae extracts and the live prey activity of antioxidant enzymes, peroxides index (PxI) and thiobarbituric acid reactive substances (TBARS) were evaluated. The lipid profile of microalgae formats and enriched live preys was also determined. Ethyl acetate extract was the most antioxidant active extract of all microalgae formats. In addition, overall, I. galbana seems to be better than any N. salinicola format for a more effective protection against oxidative stress and for live prey lipid enrichment. Both rotifer and Artemia cultured with the mixture of I. galbana and the lipid emulsion generally showed higher DHA/EPA and EPA/ARA ratios. Moreover, the combination of the microalgae with LC 60® lipid emulsion highly favored Artemia´s polar lipid and DHA incorporation. Among microalgae products, both spray-dried formats better enhanced live prey n-3 LC-PUFA content. Agriculture has gained increasing importance in response to the continuous growth of the world population and constant need for food. To avoid production losses, farmers commonly use pesticides. Mancozeb is a fungicide used in agriculture as this compound is effective in combating fungi that harm crops. However, this fungicide may also produce damage to non-target organisms present in soil and water. Therefore, this study aimed to investigate the influence of exposure to mancozeb on survival rate, locomotor activity, behavior, and oxidative status utilizing adult zebrafish (Danio rerio) as a model following exposure to environmentally relevant concentrations of this pesticide. The experimental groups were negative control, positive control, and mancozeb (0.3; 1.02; 3.47; 11.8 or 40 μg/L). Zebrafish were exposed to the respective treatments for 96 hr. Exposure to mancozeb did not markedly alter survival rate and oxidative status of Danio rerio. At a concentration of 11.8 μg/L, the fungicide initiated changes in locomotor pattern of the animals. The results obtained suggest that the presence of mancozeb in the environment might produce locomotor alterations in adult zebrafish, which subsequently disrupt the animals' innate defense mechanisms. Linum album is a rich source of various phenolic compounds, especially lignans. Our previous studies revealed that phenylalanine (Phe), an aromatic amino acid, participates in lignans production. In this study, we tried to characterize how Phe as the precursor feeding affects metabolic fluxes in L. album cells. As expected, the results indicated that an increase in phenylalanine ammonia-lyase (PAL) and tyrosine ammonia-lyase activities (TAL) occurred at 250 μM. Their actions induced phenolic acids production, especially cinnamic acid and salicylic acid (SA). The low dosages of Phe stimulated hydrogen peroxide (H2O2) and nitric oxide (NO) generation, lipid peroxidation, and antioxidant enzymes activation, while the high dosages suppressed the mentioned physiological processes. A depletion in rhamnose/xylose, glucose, and mannose was observed at high concentrations of Phe compared with the control. These molecules altered amino acids profile through supplying carbon skeleton and cell required energy. Our observations also showed that amino acids contents were significantly influenced by Phe treatment. The reduction of endogenous Phe content in the treated cells may closely related to phenolics accumulation such as flavonoids and lignans, as the enhancement of catechin, myricetin, and kaempferol were observed in the treated cells. Likewise, phenylalanine increased lignans, except for lariciresionol (LARI), and the highest amounts of podophyllotoxin (PTOX), and 6-methoxy podophyllotoxin (6MPTOX) were detected at 1000 μM. This study evaluated the use of essential oil of Ocimum gratissimum (EOOG) for anesthesia and in transport of Colossoma macropomum. Experiment 1, Test 1, anesthesia induction and recovery times were determined using different EOOG concentrations (0, 20, 50, 100, 200, 300 mg L−1), with two size classes: Juveniles I (0.86 g) and Juveniles II (11.46 g) (independent tests in a completely randomized design). Based on the results of Test 1, in Test 2 Juveniles II were exposed to EOOG concentrations: 0, 20, 100 mg L−1. Tissue samples were collected immediately after induction and 1 h post-recovery, to assess oxidative status variables. Experiment 2, Juveniles I (0.91 g) and Juveniles II (14.76 g) were submitted to transport in water with different concentrations of EOOG (0, 5, 10 mg L−1) (independent tests in a completely randomized design). The effects on oxidative status variables were evaluated. Concentrations between 50 and 200 mg L−1 EOOG can be indicated for Juveniles I, while concentrations between 50 and 100 mg L−1 EOOG for Juveniles II. The concentration of 100 mg L−1 EOOG was able to prevent oxidative damage in the liver. In Experiment 2, the concentrations of 5 and 10 mg L−1 EOOG added to the transport water caused sedation for both studied size classes of juveniles and did not cause oscillations in water quality variables nor any mortality. The concentration of 10 mg L−1 EOOG improved the oxidative status. It can be concluded that EOOG can be used for anesthesia and transport of C. macropomum. [ABSTRACT FROM AUTHOR]

The Pfaffia glomerata, a plant popularly called Brazilian ginseng, is widely used in Brazil for the treatment of various pathologies, including those associated with the Central Nervous System. 20-hydroxyecdysone (20E), a phytosteroid present in this plant, can promote adaptogenic effects in the organism, providing greater body resistance to stressors. This study aimed to evaluate the phytochemical composition and the anticholinesterase, antioxidant, and antiglycation effects of extracts and fractions of aerial parts and roots of P. glomerata, also analyzing their possible cytotoxic effects. The fractions were obtained by partitioning methanol extracts from the aerial part and roots of P. glomerata with hexane, dichloromethane, ethyl acetate, n-butanol, and water. The samples were initially tested in anticholinesterase, antioxidant, and antiglycation assays, and the most promising samples were submitted for cytotoxicity and chromatographic analyses. Mass spectrometry and chromatography methods revealed that 20E was the main compound in the dichloromethane fractions, there being 35% more 20E in the aerial part (APD) than in the roots (RD). Added to the higher concentration of 20E, the APD fraction also presented more promising results than the RD fraction in anticholinesterase and antioxidant analyses, indicating that their effects may be related to the concentration of 20E. These same fractions showed no hemolytic effects but were cytotoxic in high concentrations. These new findings contribute to scientific information about P. glomerata and open more perspectives for the understanding of its therapeutic properties, allowing the association of biological activity with the presence of 20E. Maternal obesity has been associated with short- and long-term risks of pregnancy-perinatal adverse events, possibly due to alterations of placental mitochondrial bioenergetics. However, several detrimental mechanisms occurring in the placentas of women with obesity still need to be clarified. Here, we analyzed placental mitochondrial features and oxidative environment of 46 pregnancies in relation to pre-pregnancy BMI. Seventeen Caucasian normal-weight (NW) and twenty-nine women who were obese (OB) were enrolled. The protein expression of mitochondrial CypD and electron transfer chain complexes (C) I–V were measured, as well as ATP production and oxygen consumption rates (OCRs). The protein levels of the pro/anti-oxidant enzymes TXNIP, SOD2, and PON2 were also analyzed. Despite no differences in CypD expression, OCRs were significantly lower in OB vs. NW women. Accordingly, ATP synthase (CV) levels and ATP content were decreased in OB women, positively correlating with placental efficiency, suggesting a link between ATP deficiency and placental dysfunction. SOD2 expression negatively correlated with maternal BMI, indicating a possible impairment of antioxidant defenses with increasing BMI. These changes were worsened in 10 OB women presenting with gestational diabetes mellitus. Overall, these results suggest alterations of placental bioenergetics in pregnancies of women with obesity, possibly leading to placental dysfunction and altered fetal development and programming. Highlights: The development of a fermented cherry beverage using Lactobacillus plantarum is a promising solution for improving human health and diet. The cherry-based fermented beverage boosted total antioxidant capacity in rats, increased melatonin and serotonin levels, and aided in the regulation of inflammatory processes. The intake of lactofermented cherry beverage may confer health benefits by modulating the immune system. Cherry is a fruit which contains elevated amounts of antioxidant compounds, such as anthocyanins, pigments, and vitamins. Furthermore, it possesses high water, sugar, mineral, and indolamine contents. The general objective of this study was to characterise a cherry-based fermented beverage (the 'sweetheart' variety) and analyse the effects of its ingestion on (i) circulating serum levels of melatonin and serotonin, (ii) inflammatory response, and iii) serum total antioxidant capacity in rats (Rattus norvegicus). For cherry-based fermented beverage manufacturing, the cherries were washed, the stems and woody endocarps were removed, and ascorbic acid was added (to avoid enzymatic browning). After the homogenisation of the cherry fruit, lactic acid bacteria were inoculated, and the fermentation process was conducted for 36 h. The main bioactive compounds in the cherry beverage were characterised, as well as their total antioxidant capacity. Moreover, an in vivo assay was developed, in which rats ingested the fermented beverage ad libitum for seven days. The inflammatory mediators, the total antioxidant capacity, and the serum levels of melatonin and serotonin were measured. Based on these results, the intake of the cherry-based fermented beverage assayed in this study increased the total antioxidant status of rats, elevated the melatonin and serotonin levels in the serum, and improved the regulation of the inflammatory systemic processes. Abstract The aim of this study was to investigate the associations between cardiorespiratory fitness (CRF), long-term air pollution exposure and biochemical markers of oxidative status and inflammation. This is a cross-sectional investigation focusing on biochemical markers of oxidative status and inflammation. Participants were Caucasian (N = 1188; age 18–65 years) who lived for at least 5 years in a high air-polluted (Moravian-Silesian; MS) or low air-polluted (South Bohemia; SB) region of the Czech Republic. Healthy runners and inactive individuals were recruited. A multiple regression analysis was used to explain the relationship between multiple independent variables (CRF, trunk fat mass, sex, socioeconomic status, and region (MS region vs. SB region) and dependent variables (oxidative status, inflammation). CRF, trunk fat mass, age and sex significantly predicted almost all selected markers of oxidative status and inflammation (except GSSG, GSH/GSSG and BDNF). Participants living in the MS region presented significantly higher GPx (by 3.1%) and lower BDNF values (by 4.5%). All other investigated biochemical markers were not significantly influenced by region. We did not find meaningful interactions between long-term air-pollution exposure versus markers of oxidative status and inflammation. However, we showed various significant interactions with sex, age, CRF and body composition. The significant association of living in the high air polluted MS region with the BDNF level warrants further attention.

Background: Various nutritional strategies are increasingly used in sports to reduce oxidative stress and promote recovery. Chokeberry is rich in polyphenols and can reduce oxidative stress. Consequently, chokeberry juices and mixed juices with chokeberry content are increasingly used in sports. However, the data are very limited. Therefore, this study investigates the effects of the short-term supplementation of a red fruit juice drink with chokeberry content or a placebo on muscle damage, oxidative status, and leg strength during a six-day intense endurance protocol. Methods: Eighteen recreational endurance athletes participated in a cross-over high intensity interval training (HIIT) design, receiving either juice or a placebo. Baseline and post-exercise assessments included blood samples, anthropometric data, and leg strength measurements. Results: A significant increase was measured in muscle damage following the endurance protocol in all participants (∆ CK juice: 117.12 ± 191.75 U/L, ∆ CK placebo: 164.35 ± 267.00 U/L; p = 0.001, η2 = 0.17). No group effects were detected in exercise-induced muscle damage (p = 0.371, η2 = 0.010) and oxidative status (p = 0.632, η2 = 0.000). The reduction in strength was stronger in the placebo group, but group effects are missing statistical significance (∆ e1RM juice: 1.34 ± 9.26 kg, ∆ e1RM placebo: −3.33 ± 11.49 kg; p = 0.988, η2 = 0.000). Conclusion: Although a reduction in strength can be interpreted for the placebo treatment, no statistically significant influence of chokeberry could be determined. Background: Oxidative stress plays a major role in the progression of Alzheimer's disease (AD)-related cognitive deficits. Objective: This study was done to determine the protective effects of coenzyme Q10 (CoQ10) and high-intensity interval training (HIIT) alone and in combination for eight continuous weeks, on oxidative status, cognitive functions, and histological changes in the hippocampus in amyloid-β (Aβ)-induced AD rats. Methods: Ninety male Wistar rats were randomly assigned to the sham, control, Q10 (50 mg/kg of CoQ10; P.O.), HIIT (high intensity: 4 min running at 85–90% VO2max, low intensity: 3 min running at 50–60% VO2max), Q10 + HIIT, AD, AD+Q10, AD+HIIT, and AD+Q10 + HIIT groups. Results: The results showed that Aβ injection reduced cognitive functions in the Morris water maze (MWM) test and recognition memory in the novel object recognition test (NORT), which was accompanied by a decrease in total thiol groups, catalase, and glutathione peroxidase activities, an increase in malondialdehyde levels, and neuronal loss in the hippocampus. Interestingly, pretreatment with CoQ10, HIIT, or both, could markedly improve the oxidative status and cognitive decline in the MWM and NOR tests, and hinder neuronal loss in the hippocampus of Aβ-induced AD rats. Conclusion: Therefore, a combination of CoQ10 and HIIT can improve Aβ-related cognitive deficits, probably through an amelioration in hippocampal oxidative status and prevention of neuronal loss. Research indicates that green tea extract (GTE) supplementation is beneficial for a range of conditions, including several forms of cancer, CVD and liver diseases; nevertheless, the existing evidence addressing its effects on body composition, oxidative stress and obesity-related hormones is inconclusive. This systematic review and meta-analysis aimed to investigate the effects of GTE supplementation on body composition (body mass (BM), body fat percentage (BFP), fat mass (FM), BMI, waist circumference (WC)), obesity-related hormones (leptin, adiponectin and ghrelin) and oxidative stress (malondialdehyde (MDA) and total antioxidant capacity (TAC)) markers. We searched proper databases, including PubMed/Medline, Scopus and Web of Science, up to July 2022 to recognise published randomised controlled trials (RCT) that investigated the effects of GTE supplementation on the markers mentioned above. A random effects model was used to carry out a meta-analysis. The heterogeneity among the studies was assessed using the I2 index. Among the initial 11 286 studies identified from an electronic database search, fifty-nine studies involving 3802 participants were eligible to be included in this meta-analysis. Pooled effect sizes indicated that BM, BFP, BMI and MDA significantly reduced following GTE supplementation. In addition, GTE supplementation increased adiponectin and TAC, with no effects on FM, leptin and ghrelin. Certainty of evidence across outcomes ranged from low to high. Our results suggest that GTE supplementation can attenuate oxidative stress, BM, BMI and BFP, which are thought to negatively affect human health. We studied the effects of water-soluble (vitamin C, VC) and fat-soluble antioxidants (oregano essential oil, OEO) on the stress responses and redox status of Astyanax aff. bimaculatus exposed to air. Ten isoproteic and isoenergetic diets were formulated, with different levels of VC (0, 40, 80, 120, and 160 mg kg−1), with or without OEO (1,000 mg kg−1). Fish were fed these diets for 55 days, and then 18 fish from each group were exposed to air. VC at concentrations of 120 and 160 mg kg−1 reduced glycemia. In the gills, VC at 120 and 160 mg kg−1, only in combination with OEO, reduced oxidative damage (malondialdehyde, MDA, and carbonylated proteins, PC) and superoxide dismutase (SOD) activity. In the absence of VC, OEO increased PC and SOD. In the liver, all VC concentrations, without OEO, reduced MDA while in the presence of OEO, only VC at 40 and 160 mg kg−1 reduced MDA. Regardless of OEO, all VC concentrations reduced PC levels and SOD and catalase (CAT) activity. However, OEO increased CAT and glutathione S-transferase (GST) activity regardless of VC level. While VC at a concentration of 40 with 1,000 mg kg−1 OEO reduced oxidative damage to lipids in the gills and liver, a reduction in protein and lipid damage in the gills was only observed with a least VC 120 + OEO. Therefore, we recommend simultaneous supplementation of VC 120 + 1,000 OEO, as their combined use is more effective in reducing oxidative damage in A. aff. bimaculatus exposed to air. Recurrent aphthous stomatitis (RAS) is a chronic and recurrent inflammatory disease of the mouth. It is characterised by the appearance of painful ulcers in the oral mucosa. RAS is believed to be a multifactorial disease with genetic predisposition, environmental factors and alterations in the immune system. Oxidative stress, caused by an imbalance between free radicals and the antioxidant system, also appears to be involved in the pathogenesis of RAS. Several risk factors, such as smoking, iron and vitamin deficiency and anxiety, may contribute to the development of the disease. Understanding the underlying mechanisms may help in the prevention and treatment of RAS. We searched PubMed, Scopus and Web of Science databases for articles on oxidative stress in patients with RAS from 2000 to 2023. Studies analysing oxidant and antioxidant levels in the blood and saliva of RAS patients and healthy controls were selected. Of 170 potentially eligible articles, 24 met the inclusion criteria: 11 studies on blood samples, 6 on salivary samples and 7 on both blood and salivary samples. Multiple oxidative and antioxidant markers were assessed in blood and saliva samples. Overall, statistically significant differences were found between RAS patients and healthy controls for most markers. In addition, increased oxidative DNA damage was observed in patients with RAS. Patients with RAS show elevated levels of oxidative stress compared to healthy controls, with a significant increase in oxidative markers and a significant decrease in antioxidant defences in saliva and blood samples. Simple Summary: Heat stress significantly impacts animal biological processes, including buffaloes, which exhibit signs of heat stress during hot weather. This study investigates the impact of temperature on buffaloes' oxidative stress response over two weather seasons, identifying plasma oxidants and antioxidants. The results showed that high temperatures during the hot season caused distress, while oxidant levels increased from hot to cold season. The oxidative stress index remained unaffected by annual variation but was higher in cold seasons. This study suggests that analyzing both oxidants and antioxidants is necessary to determine the best oxidative stress index for buffalo. Studies in cattle have shown that high temperatures increase the production of reactive oxygen species (ROS) causing an imbalance between ROS and the ability of antioxidant systems to detoxify and remove the reactive intermediates. As such studies remain limited in buffalo, the effect of temperature on oxidative stress was investigated through the oxidative stress index (OSi). Blood samples were collected from 40 buffaloes over 12 time points distributed over two years (2021, 2022). Samples were taken monthly during the hot and cold seasons. Plasma free oxygen radicals were determined using the d-ROMs test (Diacron, Italy), modified for a microplate procedure, and the results were expressed in arbitrary Carratelli Units (U.CARR). Plasma antioxidants were determined by using the BAP test (Diacron) in a dedicated spectrophotometer (Carpe Diem Free, Diacron). The OSi parameter was calculated as d-ROMs/BAP × 100. Temperature and humidity were recorded daily during the trial to calculate the Temperature Humidity Index (THI). For statistical analysis, year and season and their interactions were included in the model. The results of this study showed for the first time the effect of season on the oxidative stress in buffalo. The minimum and maximum THI values for the hot and cold season recorded during the experimental period were 79.27 ± 2.20 and 63.42 ± 3.20, respectively. Levels of d-ROMs and BAP were affected by the seasons (133.0 vs. 145.1 U.CARR, p = 0.0189, and 2489.19 vs. 2392.43 mml/L, p = 0.033, in the hot and cold season, respectively). A significant year × season interaction was found both for d-ROMs and BAP (p = 0.06 and p < 0.0001, respectively). Moreover, OSi was affected by season, showing a growing trend from hot to cold season (5.35 vs. 6.17, p < 0.0001), but, interestingly, it was unaffected by annual variation. Therefore, Osi could be considered a better and independent marker of oxidative status in buffalo, with respect to the evaluation of single determinations of d-ROMs and BAP. Aim: The epidemiological survey was carried out to determine the prevalence of eimeriosis in broiler chickens slaughtered depending the season, to determine the different Eimeria species causing the coccidiosis in poultry farms; and to assess the impact of Eimeria parasite on histomorphological structure and oxidative stress parameters of the intestine. Materials and Methods: The study was conducted from December 2018 to December 2019 in the province of Bejaia, Algeria. The intestines chickens (n = 366) were obtained immediately after slaughter, each cut into different segments (duodenum, jejunum, ileum, and caecum). Microscopic and parasitological examinations were performed according to standard procedures. Histomorphometric measurements of intestine were obtained using Image J software. Oxidative stress parameters were carried out from intestine tissue. Results: Eimeria spp. were detected in 73.77% (95% CI 71.20–76.34) of broiler gut samples. The prevalence varied significantly according to the season, with the lowest rates in winter (42.81%, 95% CI 40.35–45.27) and the highest in autumn (97.92%, 95% CI 97.6–99.4). All seven Eimeria species were identified, most commonly E. necatrix (27.70%), E. brunetti (26.47%), and E. tenella (20.96%). The mean lesion score ranged from 1.51 ± 0.05 to 1.79 ± 0.04. Significant differences in VH/CD ratio of intestinal epithelium (P < 0.001) were observed in different intestinal portions of infested broiler chickens compared to non-infested. The mean MDA concentration of intestinal segments was significantly higher in Eimeria species infested broilers compared to non-infested (P < 0.05). The results show at least one difference in CAT, SOD, and ABTS−+ concentrations (P < 0.05) in both chicken's groups. Conclusion: Our results revealed that coccidiosis is extremely prevalent in slaughtered broilers, with an abundance of pathogenic Eimeria species. Bacillus subtilis is a spore-forming microorganism, recognized as a safe probiotic strain. Its endospores are easily produced at a large scale, can be dehydrated and maintain their characteristics after long-term storage, providing great advantages for their application in aquafeeds. Nonetheless, knowledge on the effects that B. subtilis provokes on fish mucosal immunity is still scarce. In this context, the aim of this work was to explore the effects of the oral administration of B. subtilis spores to rainbow trout (Oncorhynchus mykiss) for 30 days on the intestinal and skin mucus, focusing on a range of immune and enzymatic parameters. Thus, fish supplemented with B. subtilis spores showed increased levels of total immunoglobulin (Ig) in intestinal and skin mucus, as well as IgM levels in both mucus and serum. B. subtilis spores induced an increase of peroxidase activities in mucus from both sources. Although superoxide dismutase (SOD) activity was not affected by the spores, NO production significantly decreased in skin mucus. Finally, the bactericidal activity of the intestinal and skin mucus was significantly higher in fish fed the probiotic spores. The results obtained demonstrate that the dietary supplementation with B. subtilis spores enhances mucosal defense mechanisms by increasing immunological parameters of the intestinal and skin mucus.

The objective of this study was to investigate the effects of supplemental vitamin E and a botanical extract in feed or drinking water on growth performance, intestinal health, and oxidative and immune status in growing pigs housed under heat stress conditions. Methods Duplicate experiments were conducted, each using 64 crossbred pigs with an initial body weight of 50.7 ± 3.8 and 43.9 ± 3.6 kg and age of 13-week and 12-week, respectively. Aim: The ongoing research was executed to assess the potential advantages of Bio-Mos® dietary supplementation regarding the growth outcomes, physiological response, oxidative biomarkers and immunity linked gene expression in Nile tilapia (Oreochromis niloticus) fingerlings exposed to bacterial infection with Aeromonas hydrophila. Methods: Four experimental diets were developed using 30% protein baseline diet, with Bio-Mos® added at variable levels; 0.0, 0.5, 1 and 2 g/kg, respectively. 240 healthy Nile tilapia fingerlings were split into four groups at random and assigned into 12 glass aquarium (three replicates of 20 fish/ treatment). Diets were admitted at 3% rate of fish biomass/aquarium for 8 weeks. Following the feeding trial, fish from every treatment were intraperitoneally injected with pathogenic Aeromonas hydrophila, and then observed for 15 days to record the survival rate post challenge. Background: Organic selenium (Sel-Plex®) supplementation holds a considerable promise for improving the effectiveness of fish production. Aim: This experiment was accomplished to judge the potential benefits of Sel-Plex® nutritional additive on growth outcomes, physiological response, oxidative status, and immunity-linked gene expression in Nile tilapia (Oreochromis niloticus) fingerlings exposed to bacterial infection with Aeromonas hydrophila. Methods: Utilizing a basal diet of 30% protein, four experimental diets were prepared, each of which contained Sel-Plex® at concentrations of 0.0, 0.5, 1, and 2 mg/kg, respectively. Three replicates of 20 fish/treatment were used using 240 healthy Nile tilapia fingerlings. Fish were placed in twelve glass aquariums and separated into four groups at random. For the entire span of 8 weeks, diets were admitted to fish at a 3% rate of fish biomass/aquarium. After the feeding trial, pathogenic Aeromonas hydrophila was intraperitoneally injected into fish of each treatment, and fish were observed for fifteen days to track the survival rate after the challenge. Results: Growth performance, physiological response, immunological parameters (Phagocytic activity, Phagocytic index, and Lysozyme), and antioxidant parameters (Catalase, Superoxide dismutase, Malondialdehyde, and Glutathione Peroxidase) were noticeably improved in Sel-Plex® treated groups. Moreover, Sel-Plex® increased gene expression linked with the immune system in the liver (TNF-α, and IL-1β), to growth (IGF-1, and GHr), and antioxidants (SOD, and GSH-Px). Under pathogen-challenge conditions, the employed dietary Sel-Plex® supplementation could successfully lower fish oxidative stress, offering a potential preventive additive for Nile tilapia instead of antibiotics. On the other hand, Sel-Plex® significantly enhanced each of three intestinal morphological measurements (villus width, villus length, and crypt depth), demonstrating the greatest influence on the improvement of intestinal structure overall. In the Nile tilapia control group, the infection with A. hydrophila caused noticeable degenerative alterations in the gut, hepatopancreas, spleen, and posterior kidney. The severity of the lesion was significantly reduced and significantly improved with higher Sel-Plex® concentrations. Sel-Plex® supplemented groups had 100% survival rates among the A. hydrophila-challenged groups. Conclusion: It could be advised to enrich the diets of Nile tilapia fingerlings with 1-2 mg.kg−1 of Sel-Plex® to enhance growth rate, physiological response, immunological reaction, and intestinal absorptive capacity. The goal of this study was to investigate the effect of supplementing Spirulina, either alone or in a mix with live yeast, on blood biochemical constituents and oxidative status in goats and their kids. Eighteen pregnant multiparous goats were equally distributed into three groups. The first group (control) received normal feeding without any additions. The second group (SP) received the normal diet plus 5 grams per head per day of Spirulina platensis, while the third group (SPSC) received the same amount of SP mixed with 3 grams per head per day of Saccharomyces cerevisiae for 30 days before parturition and continued for 45 days of lactation period. Results revealed that applying both additives caused (P≤0.01) decrease in serum cholesterol, urea, alanine aminotransferase, lipid peroxidase, and glutathione peroxidase. Serum insulin and triiodothyronine levels increased (P≤0.05) in goats fed diets supplemented with SP alone or mixed with SC compared to the control group. Concentrations of calcium and phosphorous were higher (P≤0.05) in both treated groups than in the control group. Birth weights for kids born from goats supplemented with SP alone or mixed with SC were higher (P≤0.05) compared to kids in the control group. Serum cholesterol concentration decreased (P≤0.05) in the SPSC kids group, while serum phosphorus level increased (P≤0.05) in SP kids group. It could be concluded that supplementation with SP alone or mixed with SC improved health and antioxidant status in both Damascus female goats and their newborns.

This study investigated the effect of Phyllanthus emblica leaves and branches mixture (PE) on growth performance, oxidative status and intestinal characteristics in broilers, as well as the in vitro antioxidant potential of PE extract (PEE). As the result of in vitro evaluation, the PEE had a total phenolic compound content of 95.5 ± 2.75 mg of Gallic acid equivalent/g dry weight (DW), and a total flavonoid content of 3.04 ± 0.06 mg of quercetin equivalent/g DW. The PEE exhibited significant antioxidant activity through various mechanisms, including chelation of ferrous iron, reducing power, scavenging 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals, inhibition of liposome peroxidation and trolox equivalent antioxidant capacity (TEAC). A total of 320 Ross 308 broilers were randomly allocated into four groups: control, basal diet supplemented with 0.5%, 1%, 2% PE. Each group was fed for 35 d. The results showed that the group fed with 0.5% PE had significantly better weight gain, and improved feed conversion ratio (FCR) compared to the control group. Moreover, each PE group significantly reduced coliform and Clostridium perfringens in the ileum and caecum. Additionally, PE group with 0.5% concentration had significantly higher villus height in the jejunum compared to the other group. Furthermore, compared to the control group, each PE group had significantly lower levels of malondialdehyde (MDA) and higher levels of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) activity in the serum. In the peripheral blood mononuclear cells (PBMCs) of chickens, each PE group showed significantly higher mRNA levels of antioxidant-regulated genes, including Nrf2, GST, GCLC, SOD1 and HO-1. In contrast, the expression levels of NOX1 and ROMO1 genes were significantly lower. In conclusion, the inclusion of 0.5% PE in the basal diet could have a positive impact on the growth performance of broilers by modulating intestinal characteristics and oxidative status.

Endometrial cancer is one of the most common malignancies in women. Sphingolipids, a group of lipids, play a key role in cancer biology. Cancer cells often exhibit abnormal redox homeostasis characterized by elevated levels of reactive oxygen species (ROS). Emerging evidence suggests that ceramides are involved in inhibiting proliferation and inducing apoptosis through ROS production. However, there is no data on the relationship between sphingolipid metabolism and oxidative status in endometrial cancer. The present study aims to assess the content of individual sphingolipids and oxidative status in healthy women and those with endometrial cancer. Sphingolipid analysis was performed using mass spectrometry. Total oxidative status (TOS) and total antioxidant capacity (TAC) were assessed colorimetrically. Our results showed a significant increase in the levels of all measured sphingolipids in cancer tissues compared to healthy endometrium. Additionally, a significant decrease in the S1P/ceramide ratio (sphingolipid rheostat) was observed in cancer patients, particularly for C14:0-Cer, C16:0-Cer, C18:1-Cer, C22:0-Cer, and C24:0-Cer. Furthermore, increased TOS and decreased TAC were found in cancer patients compared to healthy women. Significant correlations were observed between the levels of individual sphingolipids and oxidative status, with the strongest correlation noted between C22:0-Cer and TOS (r = 0.64). We conclude that endometrial cancer is characterized by profound changes in sphingolipid metabolism, contributing to oxidative dysregulation and tumor progression.

Serum malondialdehyde was reduced with heat stress on d 2, but increased on d 28 (interaction, P < 0.001), and was greater (P < 0.05) for antioxidant supplementation compared to control. Cellular proliferation was reduced (P = 0.037) in the jejunum under heat stress, but increased in the ileum when vitamin E was supplemented in feed and water under heat stress (interaction, P = 0.04). Tumor necrosis factor-α in jejunum and ileum mucosa decreased by heat stress (P < 0.05) and was reduced by vitamin E supplementations under heat stress (interaction, P < 0.001). In physiological concentrations, reactive oxygen species play a vital role in regulating cell signaling and gene expression. Nevertheless, oxidative stress is implicated in the pathogenesis of numerous diseases and can inflict damage on diverse cell types and tissues. Thus, understanding the factors that mitigate the deleterious effects of oxidative stress is imperative for identifying new therapeutic targets. In light of the absence of direct treatment recommendations for reducing oxidative stress, there is a continuing need for fundamental research that utilizes innovative therapeutic approaches. Metformin, known for its multifaceted beneficial properties, is acknowledged for its ability to counteract the adverse effects of increased oxidative stress at both molecular and cellular levels. In this review, we delve into recent insights regarding metformin's antioxidant attributes, aiming to expand its clinical applicability. Our review proposes that metformin holds promise as a potential adjunctive therapy for various diseases, given its modulation of oxidative stress characteristics and regulation of diverse metabolic pathways. These pathways include lipid metabolism, hormone synthesis, and immunological responses, all of which may experience dysregulation in disease states, contributing to increased oxidative stress. Furthermore, our review introduces potential novel metformin-based interventions that may merit consideration in future research. Nevertheless, the necessity for clinical trials involving this drug remains imperative, as they are essential for establishing therapeutic dosages and addressing challenges associated with dosedependent effects. To assess the impact of supplementing betaine (BT) under heat stress (HS) conditions on broiler performance and intestinal health from 21 to 42 days of age, a total of 150 male Ross 308 broilers were indiscriminately allotted to 3 treatments with 10 replications of 5 birds each. The control (CON) group was given a basal ration and accommodated at a thermoneutral condition (22 ± 1 °C), whereas the HS and HS + BT groups were raised under cyclic HS (33 ± 1 °C for 8 h and 22 ± 1 °C for 16 h per day) and received the basal ration without or with 1000 mg/kg BT, respectively. The HS reduced average daily gain (ADG); average daily feed intake; villus height (VH); VH to crypt depth (CD) ratio (VCR); activities of trypsin, lipase, glutathione peroxidase (GPX), and catalase; and enumeration of Lactobacillus and Bifidobacterium (P < 0.05) and augmented feed conversion ratio (FCR), CD, malondialdehyde (MDA) accumulation, and enumeration of Escherichia coli, Clostridium, and coliforms (P < 0.05). Conversely, BT supplementation heightened ADG, VH, VCR, trypsin activity, GPX activity, and populations of Lactobacillus and Bifidobacterium (P < 0.05) and lowered FCR, MDA accumulation, and Clostridium population (P < 0.05). Furthermore, the FCR value, trypsin and GPX activities, MDA content, and Bifidobacterium and Clostridium populations in the HS + BT group were nearly equivalent to those in the CON group. To conclude, feeding BT under HS conditions could improve broiler performance through improving intestinal health by specifically mitigating oxidative damage and enhancing the colonization of beneficial bacteria. Our study aimed to investigate the impact of nano-zinc oxide (nZnO), a widely used pollutant in industry, pharmaceuticals, and personal care products, on the behavior and oxidative stress of freshwater mussels (Potomida littoralis) an indicator species and also a model non-target organism in ecotoxicology. To this end mussels were exposed to nZnO (50 and 100 µg/L) and Zn2+ from ZnSO4 (50 and 100 µg/L) for 7 days. ZnSO4 was used for comparison purposes and to determine if the toxicity of nZnO was due to the release of ions into the aquatic environment. We evaluated changes in oxidative stress markers, including catalase (CAT), glutathione-S-transferase (GST), acetylcholinesterase (AChE), and malondialdehyde (MDA) levels, on the mussel gills and digestive glands. Additionally, the effect of nZnO on the filtration rate of bivalves was studied. The findings showed that the mussel tissue's different parameters were significantly affected by exposure to various concentrations of nZnO, causing changes in their behavior that led to a decrease in filtration rate. Additionally, noteworthy increments were observed in CAT activity, AChE activity, and MDA levels, whereas GST activity displayed a decreasing trend, implying that oxidative stress contributes to the toxicity of nZnO. Efforts are being intensively channeled to uncover strategies that could mitigate the adverse health outcomes associated with TCDD exposure. In the realm of counteractive agents, boron compounds are emerging as potential candidates. These compounds, which have found applications in a spectrum of industries ranging from agriculture to pharmaceutical and cosmetic manufacturing, are known to modulate several cellular processes and enzymatic pathways. However, the dose–response relationships and protective potentials of commercially prevalent boron compounds, such as boric acid (BA), ulexite (UX), and borax (BX), have not been comprehensively studied. In our detailed investigation, when peripheral blood mononuclear cells (PBMCs) were subjected to TCDD exposure, they manifested significant cellular disruptions. This was evidenced by compromised membrane integrity, a marked reduction in antioxidant defense mechanisms, and a surge in the malondialdehyde (MDA) levels, a recognized marker for oxidative stress. On the genomic front, increased 8-OH-dG levels and chromosomal aberration (CA) frequency suggested that TCDD had the potential to cause DNA damage. Notably, our experiments have revealed that boron compounds could act as protective agents against these disruptions. They exhibited a pronounced ability to diminish the cytotoxic, genotoxic, and oxidative stress outcomes instigated by TCDD. Thus, our findings shed light on the promising role of boron compounds. Results: Results revealed significant improvement in growth performance, physiological response, immunological parameters (phagocytic index, phagocytic activity, and lysozyme), and antioxidant parameters [catalase, malondialdehyde, glutathione peroxidase (GSH-Px), and superoxide dismutase (SOD)] among Bio-Mos® treated groups. Moreover, Bio-Mos® increased the expression of tumor necrosis factor alpha and Interleukin 1β, genes linked to the liver immune system. In fish subjected to pathogens, dietary MOS supplementation could significantly lower oxidative stress, showing promise as a preventative supplement for Nile tilapia in place of antibiotics. On the other hand, Bio-Mos® considerably improved each of the three intestinal morphological measures (villus width, villus length, and crypt depth), showing the best overall intestinal structure-improving impact. The challenge with A. hydrophila caused marked degenerative alterations in the intestine, hepatopancreas, spleen, and posterior kidney of Nile tilapia, in the control group. However, lesion severity was greatly decreased and showed marked amelioration with an increased concentration of Bio-Mos®. The A. hydrophila-challenged groups revealed a 100% SR% mainly among the Bio-Mos® supplemented groups. Conclusion: It is recommended to enrich the Nile tilapia fingerlings diets with 2 g.kg−1 of MOS for better results on the growth rate, physiological response, immunological response, and intestinal absorptive capacity. Background: Organic selenium (Sel-Plex®) supplementation holds considerable promise for improving the effectiveness of fish production. Aim:This experiment was accomplished to judge the potential benefits of Sel-Plex® nutritional additive on growth outcomes, physiological response, oxidative status, and immunity-linked gene expression in Nile tilapia (Oreochromis niloticus) fingerlings exposed to bacterial infection with Aeromonas hydrophila. Methods: Utilizing a basal diet of 30% protein, four experimental diets were prepared, each of which contained Sel- Plex® at concentrations of 0.0, 0.5, 1, and 2 mg/kg, respectively. Three replicates of 20 fish/treatment were used using 240 healthy Nile tilapia fingerlings. Fish were placed in 12 glass aquariums and separated into 4 groups at random. For the entire span of 8 weeks, diets were admitted to fish at a 3% rate of fish biomass/aquarium. After the feeding trial, pathogenic A. hydrophila was intraperitoneally injected into fish of each treatment, and fish were observed for 15 days to track the survival rate (SR) after the challenge. Results: Growth performance, physiological response, immunological parameters (phagocytic activity, phagocytic index, and lysozyme), and antioxidant parameters [catalase, superoxide dismutase (SOD), malondialdehyde, and glutathione peroxidase (GPx)] were noticeably improved in Sel-Plex® treated groups. Moreover, Sel-Plex® increased gene expression linked with the immune system in the liver (tumor necrosis factor-alpha and interleukin 1β), to growth (insulin-like growth factor 1 and growth hormone receptor), and antioxidants (SOD and GPx). Under pathogen-challenge conditions, the employed dietary Sel-Plex® supplementation could successfully lower fish oxidative stress, offering a potential preventive additive for Nile tilapia instead of antibiotics. A study was designed to evaluate the effect of Moringa oleifera, Phyllanthus amarus and Viscum album leaf meal as herbal inclusions to alleviate the detrimental outcomes of heat stress in weaned female rabbits. Forty (40) weaned rabbit does (527.99 ± 10.35 g; 28 days old) were randomly allotted to four dietary groups consisting of Diet 1(control diet; without leaf meal), Diets 2 (supplemented with 10% V. album); 3 (supplemented with 10% M. oleifera) and 4 (supplemented with 10% P. amarus) in an 84 days trial at the peak of heat stress in Southwest Nigeria. At the end of the trial, blood samples were collected to assess physiological responses and oxidative status of the rabbit does. The results obtained revealed that rabbit does were exposed to heat stress; rabbit does fed control diet had higher leucocyte and neutrophil/lymphocyte ratio compared to rabbit does fed on herbal inclusions. The herbal inclusions enhanced oxidative stability of rabbit does by lowering lipid peroxidation and enhancing antioxidant activities during heat stress conditions. Rabbit does fed control‐based diet had significantly higher heat shock protein 70, leptin and adiponectin compared to rabbit does on M. oleifera, P. amarus and V. album supplemented diets. The herbal inclusions tend to suppress proinflammatory cytokines in rabbit does during heat stress condition. Background: The cerebellar cortex is responsible for coordinating movement, adapting to special conditions, and is involved in storing memories. This cortex undergoes age-related pathological changes in the form of declined cortical thickness, neuronal loss particularly Purkinje cells, hypertrophy, and hyperplasia of the astrocytes as well as alterations in oxidative status. These changes are responsible for various senile disorders. The aim of work is to evaluate histological changes in cerebellar Purkinje cells and astrocytes and to determine the alteration in Malonaldehyde (MDA) and Glutathione (GSH) in relation to age in albino rats, and to find a probable correlation between the cellular changes and the oxidative status. Methods: Two age groups of albino rats (3-6 months & 22-26 months) were sacrificed, and their cerebella were excised and divided into three parts. 1st part was sectioned and stained with Haematoxylin and Eosin, Silver and Cresyl Violet stains. 2nd part was sectioned and prepared for immunohistochemical study using Glial Fibrillary Acid Protein (GFAP) antibodies then examined by light microscope and morphometric measurements were performed. 3rd part was biochemically processed to measure the MDA and GSH levels. All data were statistically analysed. Results: The obtained data showed that the most pronounced age-related changes were the decreased thickness of cerebellar cortex, decreased Purkinje cell number with increased degenerated cells, significant increase in astrocytes immune reactivity to GFAP and insignificant change in MDA and GSH levels. Conclusions: Cerebellar cortex of senile rats showed pathological changes in Purkinje cells and astrocytes. These changes were not solely relevant to the oxidative status. The coqui frog (Eleutherodactylus coqui) was introduced to the island of Hawai'i in the 1980s, and has spread across much of the island. There is concern they will invade higher elevation areas where negative impacts on native species are expected. It is not known if coqui change behavior and baseline physiology in ways that allow them to invade higher elevations. We investigated where coqui are found across the island and whether that includes recent invasion into higher elevations. We also investigated whether elevation is related to coqui's microhabitat use, including substrate use and height off the forest floor, and physiological metrics, including plasma osmolality, oxidative status, glucose, free glycerol, and triglycerides, that might be associated with invading higher elevations. We found coqui have increased the area they occupy along roads from 31% to 50% and have moved into more high‐elevation locations (16% vs. 1%) compared to where they were found 14 years ago. We also found frogs at high elevation on different substrates and closer to the forest floor than frogs at lower elevations—perhaps in response to air temperatures which tended to be warmer close to the forest floor. We observed that blood glucose and triglycerides increase in frogs with elevation. An increase in glucose is likely an acclimation response to cold temperatures while triglycerides may also help frogs cope with the energetic demands of suboptimal temperatures. Finally, we found that female coqui have higher plasma osmolality, reactive oxygen metabolites (dROMs), free glycerol, and triglycerides than males. Our study suggests coqui behavior and physiology in Hawai'i may be influenced by elevation in ways that allow them to cope with lower temperatures and invade higher elevations. The goal of this study was to investigate the effect of supplementing Spirulina, either alone or in a mix with live yeast, on blood biochemical constituents and oxidative status in goats and their kids. Eighteen pregnant multiparous goats were equally distributed into three groups. The first group (control) received normal feeding without any additions. The second group (SP) received the normal diet plus 5 grams per head per day of Spirulina platensis, while the third group (SPSC) received the same amount of SP mixed with 3 grams per head per day of Saccharomyces cerevisiae for 30 days before parturition and continued for 45 days of lactation period. Results revealed that applying both additives caused (P=0.01) decrease in serum cholesterol, urea, alanine aminotransferase, lipid peroxidase, and glutathione peroxidase. Serum insulin and triiodothyronine levels increased (P=0.05) in goats fed diets supplemented with SP alone or mixed with SC compared to the control group. Concentrations of calcium and phosphorous were higher (P=0.05) in both treated groups than in the control group. Birth weights for kids born from goats supplemented with SP alone or mixed with SC were higher (P=0.05) compared to kids in the control group. Serum cholesterol concentration decreased (P=0.05) in the SPSC kids group, while serum phosphorus level increased (P=0.05) in SP kids group. Since that use of bioactive mineral components of dental composites have been accompanied with various toxicities, including neurotoxicity, the aim of the study was to examine the effect of chronic application of hydroxyapatite, tricalcium phosphate and amorphous calcium phosphate in nanoparticles (nHA, nTCP, nACP) to parameters of sensitivity to thermal pain stimuli. Although the systemic toxicity of those compounds is frequently attributed to an oxidative damage, we also decided to examine the potential effects of Filipendula ulmaria extract on nociception alterations induced by the nano-sized mineral components of dental composites. Forty-two Wistar albino rats were divided into control and six experimental (equal) groups that orally received either nHA, nTCP, nACP alone, or simultaneously with FU extract for 30 days. Nociceptive alterations were quantified in the hot plate and tail flick test. The chronic administration of nHA and nACP resulted in significant increase in reactivity to thermal stimulus, with no significant change observed in nTCP group when compared to the control in the hot plate test, while simultaneous application of FU extract prevented any significant alteration of time to respond. The reaction time in the tail flick test for all three groups that received only nano calcium phosphates was reduced, with no changes in the groups treated with FU extract. The results of this study confirmed that calcium phosphates of mineral components of dental composites produced hyperalgesic effects, and this side effect were significantly attenuated by antioxidant supplementation. This study investigated the effect of Phyllanthus emblica leaves and branches mixture (PE) on growth performance, oxidative status and intestinal characteristics in broilers, as well as the in vitro antioxidant potential of PE extract (PEE). As the result of in vitro evaluation, the PEE had a total phenolic compound content of 95.5 ± 2.75 mg of Gallic acid equivalent/g dry weight (DW), and a total flavonoid content of 3.04 ± 0.06 mg of quercetin equivalent/g DW. The PEE exhibited significant antioxidant activity through various mechanisms, including chelation of ferrous iron, reducing power, scavenging 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals, inhibition of liposome peroxidation and trolox equivalent antioxidant capacity (TEAC). A total of 320 Ross 308 broilers were randomly allocated into four groups: control, basal diet supplemented with 0.5%, 1%, 2% PE. Each group was fed for 35 d. The results showed that the group fed with 0.5% PE had significantly better weight gain, and improved feed conversion ratio (FCR) compared to the control group. Moreover, each PE group significantly reduced coliform and Clostridium perfringens in the ileum and caecum. Additionally, PE group with 0.5% concentration had significantly higher villus height in the jejunum compared to the other group. Furthermore, compared to the control group, each PE group had significantly lower levels of malondialdehyde (MDA) and higher levels of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) activity in the serum. In the peripheral blood mononuclear cells (PBMCs) of chickens, each PE group showed significantly higher mRNA levels of antioxidant-regulated genes, including Nrf2, GST, GCLC, SOD1 and HO-1. In contrast, the expression levels of NOX1 and ROMO1 genes were significantly lower. In conclusion, the inclusion of 0.5% PE in the basal diet could have a positive impact on the growth performance of broilers by modulating intestinal characteristics and oxidative status. The administration of yeast products as feed additives has been proven to beneficially affect animal productivity through energy, oxidative, and immune status improvement. This study evaluated a combination of Saccharomyces cerevisiae live yeast (LY) with yeast postbiotics (rich in mannan-oligosaccharides (MOS) and beta-glucans) and selenium (Se)-enriched yeast on ewes' milk performance and milk quality, energy and oxidative status, and gene expression related to their immune system during the peripartum period. Ewes were fed a basal diet (BD; F:C = 58:42 prepartum and 41:59 postpartum) including inorganic Se (CON; n = 27), the BD supplemented with a LY product, and inorganic Se (AC; n = 29), as well as the combination of the LY, a product of yeast fraction rich in MOS and beta-glucans, and organic-Se-enriched yeast (ACMAN; n = 26) from 6 weeks prepartum to 6 weeks postpartum. The β-hydroxybutyric acid concentration in the blood of AC and ACMAN ewes was lower (compared to the CON) in both pre- and postpartum periods (p < 0.010). Postpartum, milk yield was increased in the AC and ACMAN Lacaune ewes (p = 0.001). In addition, the activity of superoxide dismutase (p = 0.037) and total antioxidant capacity (p = 0.034) measured via the 2,2-Azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) method was increased in the blood plasma of the ACMAN postpartum. Higher ABTS values were also found (p = 0.021), while protein carbonyls were reduced (p = 0.023) in the milk of the treated groups. The relative transcript levels of CCL5 and IL6 were downregulated in the monocytes (p = 0.007 and p = 0.026 respectively), and those of NFKB were downregulated in the neutrophils of the ACMAN-fed ewes postpartum (p = 0.020). The dietary supplementation of ewes with yeast postbiotics rich in MOS and beta-glucans, and organic Se, improved energy status, milk yield and some milk constituents, and oxidative status, with simultaneous suppression of mRNA levels of proinflammatory genes during the peripartum period. Purpose: Babesiosis is a tick-borne disease caused by protozoon species in the Babesia genus of the Babesiadae family. The systemic inflammatory response induced by infection is considered to be an important feature of the pathophysiology of ovine babesiosis. In this study, it was aimed to determine serum oxidative status, levels of some cytokines, acute phase proteins and heart damage markers in babesiosis infection. Materials and Methods: A sample of 40 sheep was used for this purpose, of which 20 were healthy and 20 were infected with Babesia ovis. Babesia infection was diagnosed from Giemsa-stained peripheral blood smears. Infection was also confirmed by the polymerase chain reaction (PCR). Sera from blood samples was tested for oxidative stress parameters (malondialdehyde [MDA], total antioxidant status [TAS], superoxide dismutase [SOD], catalase [CAT] and glutathione peroxidase [GPx]), cytokines (interleukins IL-6, IL-1β, IL-10, tumour necrosis factor α (TNF-α) and interferon-ϒ [IFN-ϒ]), acute-phase proteins (C-reactive protein [CRP], serum amyloid A [SAA] and haptoglobin [Hp]) and specific (troponin I [cTnI], creatine kinase–MB [CK–MB]) and nonspecific (lactate dehydrogenase [LDH], aspartate transaminase [AST]) cardiac damage markers. Results: MDA, SOD, CAT, Hp, TAS, IL-6, IL-10, TNF-α, IL-1β, INF-γ, AST, LDH, CK–MB mass and troponin I values were higher in the patient group than in the healthy group (P < 0.05). However, there was not found to be a statistical difference between the healthy and patient groups in terms of GPx, SAA and CRP values (P > 0.05). Conclusions: It can be stated that serum levels of oxidative stress, some acute phase proteins and cardiac damage markers may increase in naturally infected sheep with babesiosis. Abstract Coccidiosis is the most prevalent disease-causing widespread economic loss among farm and domestic animals. Currently, several drugs are available for the control of this disease but resistance has been confirmed for all of them. There is an urgent need, therefore, for the identification of new sources as alternative treatments to control coccidiosis. The present work aimed to study the effect of the Persea americana extract (PAE) as an anti-coccidial, anti-oxidant, and anti-apoptotic modulator during murine intestinal Eimeria papillata infection. A total of 25 male mice were divided into five groups, as follows: Group1: Non-infected-non-treated (negative control), Group2: Non-infected-treated group with PAE (500 mg/kg b.w). Group3: Infected-non-treated (positive control), Group4: Infected-treated group with PAE (500 mg/kg b.w.), and Group5: Infected-treated group with Amprolium (120 mg/kg b.w.). Groups (3–5) were orally inoculated with 1 × 103 sporulated E. papillata oocysts. After 60 min of infection, groups (4 and 5) were treated for 5 consecutive days with the recommended doses of PAE and amprolium. The fact that PAE has an anti-coccidial efficacy against intestinal E. papillata infection in mice has been clarified by the reduction of fecal oocyst output on the 5th day post-infection by about 85.41%. Moreover, there is a significant reduction in the size of each parasite stage in the jejunal tissues of the infected-treated group with PAE. PAE counteracted the E. papillata-induced loss of glutathione peroxidase (GPx), superoxide dismutase (SOD), and total antioxidant capacity (TCA). E. papillata infection also induced an increase in the apoptotic cells expressed by caspase-3 which modulated after PAE treatment. Moreover, the mRNA expression of the goblet cell response gene, mucin (MUC2), was upregulated from 0.50 to 1.20-fold after treatment with PAE. Based on our results, PAE is a promising medicinal plant with anti-coccidial, anti-oxidant, and anti-apoptotic activities and could be used as a food additive.

Background and purpose: Acrylamide is a chemical compound that is widely used in the production of paper, paint, and other industrial products. Food and cigarette smoke are primary sources of exposure to acrylamide. Betaine is a native compound with antioxidant properties and has also been reported as a protective agent against tissue damages induced by some toxicants. This study aimed to evaluate the possible effectiveness of betaine against the damage caused by acrylamide in brain. Materials and methods: In this experimental study, 28 male rats were randomly divided into four groups. The first group was considered as the control group. Group 2 received acrylamide (50 mg/kg;ip). Group 3 received the diet containing 2% betaine in addition to acrylamide. Rats in group 4 received diet containing 2% betaine. At the end of the experimental period (the eleventh day), brain tissue was sampled for biochemical and histopathological evaluation. ANOVA and Bonferroni tests were utilized to compare the means of biochemical data, and Kruskal-Wallis and Mann-Whitney tests were employed to investigate histopathological data. Heavy metal contamination has been regarded as an environmental variable that affects the efficiency of pest biological control, but the parasitic fitness of parasitoids under heavy metal stress is poorly understood. Herein, the effect of Cd exposure through the host pupa of Hyphantria cunea on the parasitic fitness of Chouioia cunea was investigated, and the mechanism by which Cd exposure affects the interaction between H. cunea and C. cunea from the perspective of innate immunity in host insect and the oxidative status in the parasitoid offspring was explored. Our results indicated that Cd can be transferred from the H. cunea pupae to the parasitoid offspring, and the transfer coefficient reflected biological amplification. There were no significant differences in the rates of parasitism success and offspring emergence between the untreated and Cd-treated groups. However, after parasitizing Cd-accumulated pupae, the parasitic fitness of offspring wasps (e.g., the number, individual size and life span) decreased significantly. Under Cd exposure, the cellular and humoral immunity of H. cunea pupae decreased significantly. Compared with the untreated group, the H2O2 content of parasitoid offspring in the Cd-treated group was significantly increased. Cd exposure significantly inhibited superoxide dismutase activity in parasitoid offspring, but the contents of ascorbic acid and glutathione were significantly increased by Cd stress. Taken together, these results indicate that Cd exposure reduces the cyclic utilization efficiency of C. cunea on H. cunea pupae. The oxidative status of parasitoid offspring triggered by Cd exposure could be responsible for the reduced parasitic fitness of C. cunea on Cd-accumulated H. cunea pupae.

Maternal obesity has been associated with short- and long-term risks of pregnancy-perinatal adverse events, possibly due to alterations of placental mitochondrial bioenergetics. However, several detrimental mechanisms occurring in the placentas of women with obesity still need to be clarified. Here, we analyzed placental mitochondrial features and oxidative environment of 46 pregnancies in relation to pre-pregnancy BMI. Seventeen Caucasian normal-weight (NW) and twenty-nine women who were obese (OB) were enrolled. The protein expression of mitochondrial CypD and electron transfer chain complexes (C) I–V were measured, as well as ATP production and oxygen consumption rates (OCRs). The protein levels of the pro/anti-oxidant enzymes TXNIP, SOD2, and PON2 were also analyzed. Despite no differences in CypD expression, OCRs were significantly lower in OB vs. NW women. Accordingly, ATP synthase (CV) levels and ATP content were decreased in OB women, positively correlating with placental efficiency, suggesting a link between ATP deficiency and placental dysfunction. SOD2 expression negatively correlated with maternal BMI, indicating a possible impairment of antioxidant defenses with increasing BMI. These changes were worsened in 10 OB women presenting with gestational diabetes mellitus. Overall, these results suggest alterations of placental bioenergetics in pregnancies of women with obesity, possibly leading to placental dysfunction and altered fetal development and programming.

The surgical site infiltration of a local anesthetic is defined as the direct injection of a drug. This study aimed to compare the effects of surgical site infiltration with 4 mg kg−1 lidocaine using a Comfort-in device and traditional syringe on oxidative status and intra- and postoperative pain in dogs undergoing regional mastectomy. Sixty adult female dogs divided into C (Comfort-in device), S (traditional syringe), and CTR (control) groups received 2 µg kg−1 dexmedetomidine and 4 mg kg−1 tramadol IM, 5 mg kg−1 tiletamine/zolazepam IV, and isoflurane. The physiological and anesthesiological parameters were measured. The assessment of intra- and postoperative responses to the surgical stimulus was performed using a cumulative pain scale (CPS score of 0–4) and the Colorado Pain Scale (CSU-CAPS score of 0–4). The hematological and biochemical parameters and inflammatory oxidative status were measured. The CPS scores showed no significant differences between the C and S groups (p = 0.236), while the comparison between the CTR, C, and S groups, respectively, showed a significant difference (p < 0.001). The postoperative analgesia scores were significantly lower in the C group compared to those of the S and CTR groups (p < 0.001). In the C group, no subject received rescue analgesia during the intra- and postoperative periods. The level of oxidative inflammatory stress was lower in group C than those in S and CTR groups, and no side effects were observed in all the groups.

ABSTRACT: Metabolic and oxidative stress have been characterized as risk factors during the transition period from pregnancy to lactation. Although mutual relations between both types of stress have been suggested, they rarely have been studied concomitantly. For this, a total of 99 individual transition dairy cows (117 cases, 18 cows sampled during 2 consecutive lactations) were included in this experiment. Blood samples were taken at −7, 3, 6, 9, and 21 d relative to calving and concentrations of metabolic parameters (glucose, β-hydroxybutyric acid (BHBA), nonesterified fatty acids, insulin, insulin-like growth factor 1, and fructosamine) were determined. In the blood samples of d 21, biochemical profiles related to liver function and parameters related to oxidative status were determined. First, cases were allocated to 2 different BHBA groups (ketotic vs. nonketotic, N:n = 20:33) consisting of animals with an average postpartum BHBA concentration and at least 2 out of 4 postpartum sampling points exceeding 1.2 mmol/L or remaining below 0.8 mmol/L, respectively. Second, oxidative parameters [proportion of oxidized glutathione to total glutathione in red blood cells (%)], activity of glutathione peroxidase, and of superoxide dismutase, concentrations of malondialdehyde and oxygen radical absorbance capacity were used to perform a fuzzy C-means clustering. From this, 2 groups were obtained [i.e., lower antioxidant ability (LAA80%, n = 31) and higher antioxidant ability (HAA80%, n = 19)], with 80% referring to the cutoff value for cluster membership. Increased concentrations of malondialdehyde, decreased superoxide dismutase activity, and impaired oxygen radical absorbance capacity were observed in the ketotic group compared with the nonketotic group, and inversely, the LAA80% group showed increased concentrations of BHBA. In addition, the concentration of aspartate transaminase was higher in the LAA80% group compared with the HAA80% group. Both the ketotic and LAA80% groups showed lower dry matter intake. However, a lower milk yield was observed in the LAA80% group but not in the ketotic group. Only 1 out of 19 (5.3%) and 3 out of 31 (9.7%) cases from the HAA80% and LAA80% clusters belong to the ketotic and nonketotic group, respectively. These findings suggested that dairy cows vary in oxidative status at the beginning of the lactation, and fuzzy C-means clustering allows to classify observations with distinctive oxidative status. Dairy cows with higher antioxidant capacity in early lactation rarely develop ketosis.

Abstract Objective To investigate the relation between interleukin-6 concentration and oxidative status of HIV infected patients with or at risk of Kaposi’s disease in Yaoundé. Methods We conducted a two-months cross-sectional study on 87 consenting HIV infected patients followed at the Day Hospital of the Yaoundé Central Hospital. Serum/plasma obtained after centrifugation of blood collected in dry/EDTA tubes was used for the determination of Human Herpes Virus-8 antigen (HHV-8) and Interleukin-6 (IL-6) by the ELISA technique, and that of oxidative stress markers: Malondialdehyde (MDA) reduced Glutathione (GSH) and total antioxidant capacity by spectrophotometry. Results Subjects belonging to the [40–50[year-old age group were mainly represented in our study population with 43.7%. The average age was 44.6 ± 10.4 years with extremes ranging from 26 to 72 years. The sex ratio was 0.24. Our population was mainly represented by people infected with HIV type I (90.8%) and 3.4% had developed clinical signs of Kaposi’s disease. The prevalence of the HHV-8 antigen was 57.5%. Immune and oxidative parameters did not vary with age, sex and therapeutic line.
