Abstract

Background: C. delpyi, C. decalvatus and C. caudatus are in the Euphorbiaceae family. The aerial parts, such as twigs, leaves and barks of these plants are used as traditional medicine such as anti-inflammatory, cytotoxicity, and antifungal properties. Objectives: The aims of this work were 1) to study the chemical composition of C. delpyi, C. decalvatus, and C. caudatus 2) to test the antibacterial, anti-HIV-1 RT, and cytotoxicity activities of crude extracts and pure compounds from these plants. Methods: Extraction, separation and purification of three plants were performed following chromatographic method. The biological activities, including antibacterial, anti-HIV-1 RT and cytotoxicity assay of three plants, were evaluated by the standard methods. Results: Phytochemical investigation of C. delpyi highlighted a new clerodane diterpenoid; crotondelpyitin A (1). The five known compounds were found, such as acetyl aleuritolic acid (2), 5- hydroxy-7,4′-dimethoxyflavone (3), and pilloin (4) in C. decalvatus and 3α-benzoyloxy-D while Afriedo- oleanan-27,16α-lactone (5), and bergenin (6) in C. caudatus. Compound 3 showed the most effective antibacterial activities with MIC in range 5.0 mg/mL. The six compounds were inactive with anti-HIV-1 RT. In addition, compound 4 was active for cytotoxic activities on FaDu and KKU-M213 at Conclusion: The present study reveals that the Croton species are sources of diterpenoid-type compounds and a significant guide for further research of chemical constituents from these plants as potential medicines.

Abstract

Pseuduvaria phuyensis (Annonaceae), was collected from Kanchanaburi Province, western Thailand. The study aimed to evaluate the antibacterial, anti-HIV-1RT, anti-syncytium (MC99+1A2), and cytotoxic potentials of the crude hexane, ethyl-acetate, and methanol extracts of P. phuyensis leaf-twigs and stems, all in comparison with standard drugs. Also, the isolation and characterization of a compound from one of the active extracts as well as its antibacterial and cytotoxicity activities were carried out. The MIC and MBC values of selected bacterial strains ranged from 3.125 - 200 and 6.25 - 200 mg/mL, respectively for all the extracts, while the isolated compound was not active. The anti-HIV-1 RT and the anti-syncytium assay of the hexane and ethyl acetate extracts revealed they were very active, with % inhibitions of 90.50 and 89.36, respectively. The best inhibitory concentration at 50% (IC50) value was 44.33 µM as exhibited by the hexane extract of the stems of the reverse transcriptase assay, while, the best effective concentration at 50% (EC50) value was 10.4 µM (TI>4.83) as displayed by the ethyl acetate extract of the stems. The cytotoxicity study showed that the hexane extract of the stems displayed high cytotoxicity with ED50 value at 13.07 µg/mL against the FaDu cancer cell line, whil that of the isolated compound was not active. The structure of the isolated compound was characterized and elucidated as trans, trans-cyclohexane-1, 2, 4, 5-tetrol monohydrate using spectroscopic technique and X-ray crystallography analysis. Background: Although the chemical constituents and biological activities of a large number of plants in the Croton genus have been studied, there are still recently discovered plants to be investigated. Objective: 1. To investigate the anti-bacterial, anti-HIV1-RT, and cytotoxicity activities of crude extracts from these plants. 2. To investigate the chemical constituents of Croton fluviatilis, Croton acutifolius, and Croton thorelii. Method: The anti-bacterial, anti-HIV1-RT, and cytotoxicity of the three plants were evaluated by standard techniques. Extraction, separation, and purification of extracts from the three plants were undertaken. Results: The ethyl acetate extract of C. fluviatilis showed low anti-bacterial activity against E. aerogenes, E. coli 0157: H7, and P. mirabilis, together with the ethyl acetate extract of C. acutifolius displayed low anti-bacterial activity against E. aerogenes, while all the crude extracts of C. thorelii were inactive. The ethyl acetate extracts of C. thorelii, and C. fluviatilis showed strong inhibited HIV1-RT, whereas the ethyl acetate extract of C. acutifolius, and the hexane extract of C. fluviatilis displayed moderate inhibited HIV1-RT. Cytotoxic properties of three Croton plants were specific to KKU-M213, MDA-MB-231, A-549, and MMNK-1. Especially, the ethyl acetate extract of C. acutifolius exhibited strong cytotoxic activities against MDA-MB-231, A-549, and MMNK-1. Furthermore, the ethyl acetate extract of C. thorelii showed high cytotoxic activities against KKU-M213, and MDA-MB-231. Compounds 1, and 4 were found in C. fluviatilis. Compounds 2 and 4 were also found in C. acutifolius. Moreover, compound 3 was only found in C. thorelii. Conclusion: The present study revealed that the three Croton species are good sources of flavonoid compounds and further investigation of the chemical constituents from these plants may prove to be fruitful to discover more active compounds to be tested as potential medicines.

One new clerodane-type diterpenoid, together with one known, was isolated from the leaves and twigs of C. krabas. The structures of these compounds were elucidated as krabasinolide A (1) and taraxerol (2) by spectroscopic methods (UV, IR, HRESIMS, 1 D, and 2 D NMR), and the relative stereochemistry was confirmed by X-ray diffraction analysis with graphite monochromated Mo-Kα (λ = 0.71073 Å) radiation at 296(2) K. 23. Identification and antiproliferative activity evaluation of a series of triterpenoids isolated from flueggea virosa (Roxb. ex Willd.)

Abstract

Problem statement: Medicinal plants derived anticancer were now being subjects of many research groups especially, the secondary metabolite triterpenoids trees which had enormous potential to inspire and influence modern antiproliferative research. The study aimed to investigate the chemical constitution and their potential use as antiproliferative activity of purified compounds derived from F. virosa. Approach: The F. virosa was selected and percolated with hexane, ethyl acetate, acetone and methanol. The extracts were purified and elucidated chemical structures. Furthermore, the isolated compounds were tested for biological activity. The bioassays were performed on two cancer cell lines, adriamycin-sensitive erythroleukemia cells (K562) and adriamycin-resistant erythroleukemia cells (K562/Adr) which overexpressed P-glycoprotein (MDR1/ABCB1). Results: Friedelin (1), epifriedelanol (3), stigmasterol (4) and betulinic acid (5) were isolated from the leaves and twigs of F. virosa. The molecular structures of these compounds were determined using several spectroscopic methods. The compounds i.e., 1, a chemically modified compound 1 heptanolide (2), 3 and 4 showed a limited cytotoxic activity towards human cancer cell lines mainly due to a low aqueous solubility which prevented their use in cell viability assays. Interestingly, compound 5 exhibited a high cytotoxicity characterized by an effective concentration value ([IC.sub.50]) equal to 9.7[+ or -]2.1 [micro][g.mL.sup.-1] (21.2[+ or -]4.6 [micro]M) and 7.1[+ or -]0.7 [micro][g.mL.sup.-1] (15.5[+ or -]1.5 [micro]M) for K562 and K562/Adr, respectively. Moreover, the antiproliferative activity of compound 5 was independent of the multidrug resistance phenotype exhibited by the K562/Adr cell line suggesting that compound 5 was not the effluxes out of the K562/Adr cells by MDR1 (ABCB1). Conclusion: The results clearly showed that the betulinic acid of the four isolated compounds from F. virosa could be considered as high potential source of cytotoxic activity. Abstract

The genus Goniothalamus belongs to the Annonaceae family. The pure compounds from Goniothalamus rongklanus and Goniothalamus latestigma were isolated by repeated column chromatography. The structures were recognized by NMR spectral methods. The chemical compounds of ethyl acetate extract (EtOAc) from the stems of G. rongklanus were identified as goniotriol (1) and stigmasterol glucoside (2). The EtOAc leaves extract of G. latestigma yielded a compound, which was identified as pinocembrin (3). The ethyl acetate (EtOAc) and methanol (MeOH) extract were tested for anti-HIV-1 RT and cytotoxic activities against P-388, KB, HT 29, MCF-7, A 549, ASK and CL cell lines. The EtOAc and MeOH extract of G. rongklanus showed evidence of anti-HIV-1 RT inhibition at 76.44 and 88.48 %, respectively. The EtOAc extract of G. rongklanus and G. latestigma showed cytotoxic activities on KB and HT 29 with an ED50 at <4 μg/mL. In addition, antibacterial study on extracts and isolated compounds was also performed. Antibacterial study was evaluated using nine strains (Staphylococcus aureus, Enterobacter aerogenes, Escherichia coli 0157: H7, Escherichia coli (ETEC), Escherichia coli (EPEC), Proteus mirabilis, Salmonella typhimuriam, Shigella flexneri and Vibrio cholera) by Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) method. Goniotriol was the most effective antibacterial with a MIC in the range <0.16 - 0.6 mg/mL and MBC in the range 0.3 - >5 mg/mL. The antibacterial activity of goniotriol has been reported for the first time.

Abstract

A new ent-kaurane-type diterpenoid, crotonmekongenin A (1) together with two known compounds, epifriedelanol (2) and sitosterol glucoside (3) from the leaves and twigs of Croton mekongensis were isolated. The structure of these compounds was determined by spectroscopic analysis. The structure of the new ent-kaurane-type diterpenoid (1) was elucidated using spectroscopic technique and X-ray crystallography analysis. Moreover, the cytotoxicity of all extract and isolated compounds was tested for cytotoxicity against KKU-M213, FaDu, HT-29, MDA-MB-231, A-549, SH-SY5Y, and CL cell lines. Abstract

Three new diterpenoids, boesenmaxanes A–C (1–3), with an unprecedented core skeleton consisting of an unusual C–C bond between C-12 and an exo-cyclic methylene C-13, were isolated from the rhizome extracts of Boesenbergia maxwellii. The structures were elucidated by analysis of spectroscopic and X-ray diffraction data. Electronic circular dichroism spectra were used to determine the absolute configuration. All the isolates were evaluated for their cytotoxic effects, anti-HIV activity, and antimicrobial activity. Boesenmaxanes A and C (1and 3) showed significant inhibitory activity in the syncytium reduction assay, with EC50values of 55.2 and 27.5 μM, respectively.

Abstract

Background: C. delpyi, C. decalvatus and C. caudatus are in the Euphorbiaceae family. The aerial parts, such as twigs, leaves and barks of these plants are used as traditional medicine such as anti-inflammatory, cytotoxicity, and antifungal properties. Objectives: The aims of this work were 1) to study the chemical composition of C. delpyi, C. decalvatus, and C. caudatus 2) to test the antibacterial, anti-HIV-1 RT, and cytotoxicity activities of crude extracts and pure compounds from these plants. Methods: Extraction, separation and purification of three plants were performed following chromatographic method. The biological activities, including antibacterial, anti-HIV-1 RT and cytotoxicity assay of three plants, were evaluated by the standard methods. Results: Phytochemical investigation of C. delpyi highlighted a new clerodane diterpenoid; crotondelpyitin A (1). The five known compounds were found, such as acetyl aleuritolic acid (2), 5- hydroxy-7,4′-dimethoxyflavone (3), and pilloin (4) in C. decalvatus and 3α-benzoyloxy-D while Afriedo- oleanan-27,16α-lactone (5), and bergenin (6) in C. caudatus. Compound 3 showed the most effective antibacterial activities with MIC in range <0.16 -1.25 mg/mL, and MBC in range 0.6 - >5.0 mg/mL. The six compounds were inactive with anti-HIV-1 RT. In addition, compound 4 was active for cytotoxic activities on FaDu and KKU-M213 at <4 μg/mL. Conclusion: The present study reveals that the Croton species are sources of diterpenoid-type compounds and a significant guide for further research of chemical constituents from these plants as potential medicines.
