In degenerative joint disease like osteoarthritis (OA), bioactive compounds like resveratrol, epigallocatechin gallate, curcumin, and other polyphenols often target various signalling pathways, including NFκB, TGFβ, and Wnt/β-catenin by executing epigenetic-modifying activities. Epigenetic modulation can target genes of disease pathophysiology via histone modification, promoter DNA methylation, and non-coding RNA expression, some of which are directly involved in OA but have been less explored. OA patients often seek options that can improve the quality of their life in addition to existing treatment with nonsteroidal anti-inflammatory drugs (NSAIDs). Although bioactive and natural compounds exhibit therapeutic potential against OA, several disadvantages loom, like insolubility and poor bioavailability. Nanoformulated bioactive compounds promise a better way to alleviate OA since they also control systemic events, including metabolic, immunological, and inflammatory responses, by modulating host gut microbiota that can regulate OA pathogenesis. Recent data suggest gut dysbiosis in OA. However, limited evidence is available on the role of bioactive compounds as epigenetic and gut modulators in ameliorating OA. Moreover, it is not known whether the effects of polyphenolic bioactive compounds on gut microbial response are mediated by epigenetic modulatory activities in OA. This narrative review highlights the nanotherapeutic strategies utilizing bioactive compounds, reporting their effects on chondrocyte growth, metabolism, and epigenetic modifications in osteoarthritis amelioration. Nanocurcumin, enhanced through nanotechnology, addresses these limitations, offering potential in oncological applications. This review systematically examines the efficacy, bioavailability, and safety of nanocurcumin in cancer treatment, collating data from in vitro, in vivo, and clinical studies. Methods: A comprehensive systematic search was conducted across four major databases: PubMed (Medline), Scopus, Web of Science, and Embase (up to February 2024). The selection criteria were based on the PICOT structure, and studies were assessed for risk of bias using the Cochrane bias risk tool for clinical studies and related checklists for in vitro and in vivo studies. Statistical analyses were performed in STATA software version 17. Results: In total, 8403 articles were identified and assessed, and then only 61 articles were found eligible to be included. Nanocurcumin formulations, especially with Poly (lactic-co-glycolic acid) (PLGA), displayed superior solubility and therapeutic efficacy. In vitro studies highlighted its enhanced cellular uptake and anti-proliferative effects, particularly against cervical cancer cells. In vivo studies confirmed its chemopreventive efficacy and potential synergy with other cancer therapies. Though in early stages, clinical trials showed promise in reducing side effects and improving efficacy in cancer treatments. Conclusion: Nanocurcumin shows promise as an innovative approach in cancer therapy, potentially offering improved efficacy and reduced side effects compared to traditional treatments. Early clinical trials indicate its potential to enhance the quality of life for cancer patients by mitigating treatment-related toxicities and improving therapeutic outcomes. For nearly 90 years, aluminum (Al) salts have been utilized as vaccination adjuvants. Nevertheless, there is a risk of adverse effects associated with the amount of nanoaluminum used in various national pediatric immunization regimens. This study aimed to investigate the possible genotoxic effects of nanoaluminum incorporated in human vaccines on the brains of newborn albino rats and whether nanocurcumin has a potential protective effect against this toxicity. Fifty newborn albino rats were randomly assigned to 5 groups, with 10 in each group. Groups 1 and 2 received "high" and "low" Al injections corresponding to either the American or Scandinavian pediatric immunization schedules, respectively, as opposed to the control rats (group 5) that received saline injections. Groups 3 and 4 received the same regimens as groups 1 and 2 in addition to oral nanocurcumin. The expression of both the cell breakdown gene tumor protein (P53) and the cell stress gene uncoupling protein 2 (UCP2) was significantly greater in groups 1 and 2 than in group 5. Groups 1 and 2 exhibited severe DNA fragmentation, which was observed as DNA laddering. Nanocurcumin significantly reduced the expression of the P53 and UCP2 genes in groups 3 and 4, with very low or undetectable DNA laddering in both groups. Vaccination with nanoaluminum adjuvants can cause genotoxic effects, which can be mediated by the inflammatory response and oxidative stress, and nanocurcumin can protect against these toxic effects through the modulation of oxidative stress regulators and gene expression. Psychiatric disorders cause long‐lasting disabilities across different age groups. While various medications are available for mental disorders, some patients do not fully benefit from them or experience treatment resistance. The pathogenesis of psychiatric disorders involves multiple mechanisms, including an increase in the inflammatory response. Targeting inflammatory mechanisms has shown promise as a therapeutic approach for these disorders. Curcumin, known for its anti‐inflammatory properties and potential neuroprotective effects, has been the subject of studies investigating its potential as a treatment option for psychiatric disorders. This review comprehensively examines the potential therapeutic role of curcumin and its nanoformulations in psychiatric conditions, including major depressive disorder (MDD), bipolar disorder, schizophrenia, and anxiety disorders. There is lack of robust clinical trials across all the studied psychiatric disorders, particularly bipolar disorder and schizophrenia. More studies have focused on MDD. Studies on depression indicate that curcumin may be effective as an antidepressant agent, either alone or as an adjunct therapy. However, inconsistencies exist among study findings, highlighting the need for further research with improved blinding, optimized dosages, and treatment durations. Limited evidence supports the use of curcumin for bipolar disorder, making its therapeutic application challenging. Well‐designed clinical trials are warranted to explore its potential therapeutic benefits. Exploring various formulations and delivery strategies, such as utilizing liposomes and nanoparticles, presents intriguing avenues for future research. The objective of this study is the development of innovative nanocurcumin-based formulations designed for the treatment and prevention of oxidative stress and diabetes. Nanocurcumin was obtained through a micronization process and subsequently encapsulated within biopolymers derived from corn starch and fenugreek mucilage, achieving encapsulation rates of 75% and 85%, respectively. Subsequently, the encapsulated nanocurcumin was utilized in the formulation of sugar-free syrups based on Stevia rebaudiana Bertoni. The stability of the resulting formulations was assessed by monitoring particle size distribution and zeta potential over a 25-day period. Dynamic light scattering (DLS) revealed a particle size of 119.9 nm for the fenugreek mucilage-based syrup (CURF) and 117 nm for the corn starch-based syrup (CURA), with polydispersity indices PDIs of 0.509 and 0.495, respectively. The dissolution rates of the encapsulated nanocurcumin were significantly enhanced, showing a 67% improvement in CURA and a 70% enhancement in CURF compared with crude curcumin (12.82%). Both formulations demonstrated excellent antioxidant activity, as evidenced by polyphenol quantification using the 2.2-diphenyl 1-pycrilhydrazyl (DPPH) assay. In the evaluation of antidiabetic activity conducted on Wistar rats, a substantial reduction in fasting blood sugar levels from 392 to 187 mg/mL was observed. Alzheimer's disease (ad) is a neurological condition that worsens over time and is characterized by the buildup of amyloid (Aβ) plaques in the brain parenchyma. Neuroprotection and cholinesterase inhibition have been the two primary techniques used in the creation of medications to date. In ad , a novel sort of programmed cell death known as ferroptosis takes place along with iron buildup, lipid peroxidation, and glutathione deficiency. The objective of the current investigation was to examine the neuroprotective and anti-ferroptotic role of nanocurcumin and Donepezil against model of aluminum chloride AlCl3 and D-galactose induced ad. The experiment was performed on 70 rats divided into (G1: control, G2: NCMN, G3: Donepezil, G4: ad -model, G5: Donepezil co-treatment, G6: NCMN co-treatment and G7: NCMN+Donepezil co-treatment). Hematological parameters and biochemical investigations as oxidative stress, liver function, kidney function, iron profile and plasma fibrinogen were evaluated. Treatment with Nanocurcumin alone or in combination with Donepezil improved oxidative stress, liver functions, and kidney functions, improve iron profile and decreased plasma fibrinogen. Introduction: Due to its biological and antibacterial qualities, many plants, including curcumin, are used as phytomedicines in dentistry. They are primarily used as intracanal medication in endodontics to prevent probable chemical side effects and also to address antimicrobial resistance. Curcumin nanoformulations have improved antibacterial activity and improved dispersion, making them the superior form of curcumin. The purpose of this study was to assess curcumin and nanocurcumin's antibacterial properties. As a gutta-percha coating, they are to be tested against Escherichia coli. Materials and Methods: The study employs the standard strain of E. coli, ATCC 25922. The antibacterial activity of gutta-percha cones against E. coli is assessed after coating them with suspensions of curcumin and nanocurcumin. Scanning electron microscopy is utilized to evaluate the coatings' continuity. Results: The gutta-percha cones that are untreated, coated with curcumin, and coated with nanocurcumin exhibit significantly different levels of antibacterial activity. There is statistically significant variation in their antibacterial activity. Conclusion: (1) Compared to curcumin-coated and untreated gutta-percha cones, those coated with nanocurcumin exhibit a stronger antibacterial activity. A total 375 one-day-old male broiler chickens were allotted into five groups including: 0, 25, 50, 100 and 200 mg curcumin nanoparticles) CurNPs). Final body weight increased and feed conversion ratio improved by CurNPs supplementation. Antioxidant enzymes activities were increased and malondialdehyde was reduced by CurNPs. The white blood cells (except heterophil) increased and red blood cells and hematocrit decreased by CurNPs supplementation. The serum enzyme activity, cholesterol and triglyceride reduced and protein increased by CurNPs supplementation. Also, The CurNPs, increased relative weight of spleen and immune indices concentrations in serum. Moreover, The CurNPs addition improved gut morphological traits in the Jejunum. In this study, the effects of nanocurcumin on acetaminophen-induced acute hepatorenal toxicity in domestic pigeons (Columba livia) were investigated. Fifteen pigeons were randomly assigned into three groups. Group I was served as a negative control group and received tap water as a placebo. Pigeons in groups II and III were administered acetaminophen at the beginning of the experiment (hr 0). Group III was further treated with nanocurcumin, at 12 hr after acetaminophen administration, being continued every 12 hr for two days. The birds were observed for clinical signs of acute drug toxicity. Blood samples were collected from the pigeons at hr 0, 12, 24 and 48 of the experiment for biochemical analysis of the serum. The results showed that acetaminophen toxicity increased the serum levels of aspartate aminotransferase, alanine aminotransferase, urea and uric acid in the pigeons. Nanocurcumin treatment of acetaminophen intoxicated pigeons attenuated increases in biomarkers of the liver and kidney functions towards control levels. Also, the consumption of nanocurcumin minimized histopathological changes in the liver and kidney. A mortality of 60.00% was seen in the acetaminophen-induced toxicity group; while, none of the birds treated with nanocurcumin died. The precise pathophysiology of polycystic ovary syndrome (PCOS) is not well-founded. In an attempt to fill this gap, the current study was executed to probe the effect of nanocurcumin (NCC) on ovarian tissue, in vitro fertilization (IVF) and pre-implantation embryo development in a mouse model of PCOS. Fifty adult female mice were randomly categorized into five equal groups including non-treated control and PCOS (receiving 0.20 mg estradiol valerate (EV) intraperitoneally once a day for 21 days) as well as NCC12.50 + PCOS, NCC25 + PCOS and NCC50 + PCOS (receiving respectively 12.50, 25.00 and 50.00 mg kg-1 NCC daily along with EV injection through oral gavages for 21 days) groups. Subsequently, ovarian histo-architecture and total anti-oxidant capacity, and malonaldehyde and catalase levels as well as in vitro fertilizing potential, early embryonic development and serum testosterone concentration were analyzed. Results showed that NCC in a dose-dependent manner improved ovarian cyto-architectural organization and oxidant/anti-oxidant balance along with IVF rate and pre-implantation embryo development in PCOS mice. Exposure to a hypobaric hypoxic environment at high altitudes can lead to liver injury, and mounting evidence indicates that pyroptosis and inflammation play important roles in liver injury. Curcumin (Cur) can inhibit pyroptosis and inflammation. Therefore, our purpose here was to clarify the mechanism underlying the protective effect of nanocurcumin (Ncur) and Cur in a rat model of high altitude‐associated acute liver injury. Eighty healthy rats were selected and exposed to different altitudes (6000 or 7000 m) for 0, 24, 48, or 72 h. Fifty normal healthy rats were divided into normal control, high‐altitude control, salidroside (40 mg/kg [Sal‐40]), Cur (200 mg/kg [Cur‐200]), and Ncur (25 mg/kg [Ncur‐25]) groups and exposed to a high‐altitude hypobaric hypoxic environment (48 h, 7000 m). Serum‐liver enzyme activities (alanine transaminase, aspartate transaminase, and lactate dehydrogenase were detected and histopathology of liver injury was evaluated by hematoxylin and eosin staining, and inflammatory factors were detected in liver tissues by enzyme‐linked immunosorbent assays. Pyroptosis‐associated proteins (gasdermin D, gasdermin D N‐terminal [GSDMD‐N], pro‐Caspase‐1, and cleaved‐Caspase‐1 [cleaved‐Casp1]) and inflammation‐associated proteins (nuclear factor‐κB [NF‐κB], phospho‐NF‐κB [P‐NF‐κB], and high‐mobility group protein B1 [HMGB1]) levels were analyzed by immunoblotting. Ncur and Cur inhibited increased serum‐liver enzyme activities, alleviated liver injury in rats caused by high‐altitude hypobaric hypoxic exposure, and downregulated inflammatory factors, including tumor necrosis factor‐α, interleukin (IL)‐1β, IL‐6, and IL‐18, in rat liver tissues. The level of P‐NF‐κB, GSDMD‐N, cleaved‐Casp1, and HMGB1 in rat liver tissues increased significantly after high‐altitude exposure. Background: Cyclophosphamide (CYP) is a cardiotoxic agent with antineoplastic and immunosuppressive properties. Objectives: To detect the histological, immunohistochemical, biochemical cardiac toxicity of CYP and determine the protective effectiveness of nanocurcumin on the cardiac muscle of albino rats. Material and methods: The current work used forty Wistar rats allocated into four groups; normal control (I), sham control (II), CYP-treated (III) and concomitant protective CYP + nanocurcumin (IV). Results: CYPtreated group (III) showed fragmented, disrupted cardiac myocytes with cellular infiltrates and interstitial edema. Blood vessels were dilated congested. Cardiac myocytes showed pyknosis, vacuolations and some showed karyolysis. Much improvement was observed in group IV. The area percentage of fibrous tissue, TNF-α and iNOS immunoreactivity in CYP-treated group III showed statistically significant increase in contrast to that of the control group. Use of nanocurcumin in groups IV ameliorate these changes. Biochemically, there were significant increase in means of CK-MB, cTn-1, MDA, levels and decrease of GPx activity in group III (CYP -treated group) compared to the control group. Uses of nanocurcumin has been observed to improve these changes. Conclusion: Cyclophosphamide (CYP) had deleterious effects on the histological structure of the heart, cardiac enzymes, collagen fibers deposition and myofibroblast proliferation in albino rats. Cancer has recently become one of the leading causes of mortality across the globe and has persisted as a severe health concern for the past several years. The present study is focused on the fabrication of nanocurcumin–nano iron oxide composites to improve its solubility without compromising the structural features of curcumin which would eventually enhance the targeted drug administration. The superparamagnetic iron oxide nanoparticles used for the synthesis of nanocomposite were produced via green route using Pimenta dioica leaf extract as reducing agent. Various characterization techniques such as UV–Visible spectroscopy, FTIR, XRD, TG, SEM-EDS and TEM analysis have been adopted to characterize the synthesized nanocomposites and found to be spherical in shape with size ranging from 10 to 25 nm. In vitro cytotoxic studies of nano iron oxide, nanocurcumin and the nanocomposite were conducted against DLD-1 (colorectal cancer) cell lines and normal L929 (Fibroblast) cell lines. From the results, it was found that the IC50 value of the curcumin/iron oxide nanocomposites against DLD-1 cells is 37.96 µg/mL whereas that against L929 cells is 247.4 µg/mL which implies very high toxicity towards cancer cells and less toxic towards normal cells. The novel curcumin/iron oxide nanocomposites developed in the present research is highly effective in exterminating colorectal cancer cells with minimum side effects. [ABSTRACT FROM AUTHOR]

Backgrounds and Objectives: The use of anti-inflammatory supplements is important in improving the executive function of obese people. This research aims to investigate the effect of 6 weeks of aerobic exercise with moderate intensity as well as the consumption of nanocurcumin on IL1β, nitric oxide (NO), and depression in women aged 60–65 with metabolic syndrome. Materials and Methods: Fourty-four women with metabolic syndrome were randomly selected and divided into four groups of 10 based on their use of nanocurcumin supplement. The treatment included the training (T), nanocurcumin (N), training + nanocurcumin (TN), and the control groups. The groups exposed to training performed aerobic exercise for 6 weeks (three sessions per week). Blood samples were obtained before and after the training period for antioxidant indicators and lipid degradation measurement. Also, the Beck anxiety questionnaire was used for evaluating levels of anxiety. T-test and one-way analysis of variance (ANOVA) tests were used for the assessment of within-group and between-group differences, respectively. Results: There was a significant difference in IL1β, NO, and depression before and after exercise in all three experimental groups (p ≤ 0.05). Also, the results showed a significant difference in the level of NO and depression in the experimental groups. The highest decrease in these variables was observed in the T and TN groups (p ≤ 0.05). Conclusion: These findings indicated that 6-week nanocurcumin supplementation with aerobic training is a suitable method for reducing IL1β, NO, and depression, preventing metabolic, cardiovascular, and inflammatory diseases in women with metabolic syndrome. Chronic obstructive pulmonary disease (COPD) is a chronic inflammatory process in the airways that results in airflow obstruction. It is mainly linked to cigarette smoke exposure. Th17 cells have a role in the pathogenesis of COPD by secreting pro-inflammatory cytokines, which cause hyperinflammation and progression of the disease.This study aimed to assess the potential therapeutic effects of nanocurcumin on the Th17 cell frequency and its responses in moderate and severe COPD patients. This study included 20 patients with severe COPD hospitalized in an intensive care unit (ICU) and 20 patients with moderate COPD. Th17 cell frequency, Th17-related factors gene expression (RAR-related orphan receptor t (RORγt), IL-17, IL-21, IL-23, and granulocyte-macrophage colony-stimulating factor), and serum levels of Th17-related cytokines were assessed before and after treatment in both placebo and nanocurcumin-treated groups using flow cytometry, real-time PCR, and ELISA, respectively.According to our findings, in moderate and severe nanocurcumin-treated COPD patients, there was a substantial reduction in the frequency of Th17 cells, mRNA expression, and cytokines secretion level of Th17-related factors compared to the placebo group. Furthermore, after treatment, the metrics mentioned above were considerably lower in the nanocurcumin-treated group compared to before treatment.Nanocurcumin has been shown to decrease the number of Th17 cells and their related inflammatory cytokines in moderate and severe COPD patients. As a result, it might be used as an immune-modulatory agent to alleviate the patient's inflammatory state.

Background: One of the most efficient treatments for dry eye syndrome (DES) is to use nanocarriers as a potential delivery system. We aim to evaluate curcumin in a nano emulsion formulation. Methods: A new formulation containing 5.5% curcuminoid was used. DLS, Zeta potential, TEM, and HPLC tests were performed to determine the size and morphology. First, 30 mice were selected as atropine-induced dry eye models. Next, 25 mice in 5 groups were treated with the nano emulsion at different doses, and corneal tissues were separated for evaluation. Results: The DLS test results were indicative of the particles' stability. Nano curcumin appeared to be thoroughly effective in all groups, with the highest dose showing the most similarity to the healthy control group. Conclusions: Curcumin-based nano emulsion eye drop is a promising candidate for DES management. However, further investigation is required to evaluate the possible risks in humans.

The concern over agricultural waste disposal can be resolved by converting them into value-added products. Here, sustainable fruit coatings and films have been developed via. wealth from waste concept. The cellulose nanofibers (CNF) have been extracted from the waste onion skin. The isolated CNF has been characterised using scanning electron microscopy (SEM), transmission electron microscopy (TEM), optical microscopy (OM), dynamic light scattering (DLS) and solid-state 13C NMR spectroscopy. A combination of cellulose nanofiber (CNF) and nanocurcumin in polyvinyl alcohol (PVA) matrix has been used for the fabrication of coating over fresh mandarin oranges. The effect of nanocurcumin in PVA-CNF nanocomposite on the post-harvest maintenance of mandarin oranges has been investigated. The morphology of the prepared films is studied by SEM and atomic force microscopy (AFM). The quality of the orange fruits coated using the above formulation by dip coating method was analysed via. weight loss analysis, total soluble solid (TSS), pH, titrable acidity (TA) and antioxidant activities. It was found that the nanocurcumin incorporated PVA-CNF coating was effective in reducing mass loss and maintaining the physiochemical properties of oranges in comparison with other formulations. The developed method could be extended to use as food packaging films, which would be a sustainable solution for agricultural waste valorisation and plastic pollution. Through this research work, we propose an environment friendly approach for the preservation of fruits and vegetables.

The objective of this study is the development of innovative nanocurcumin-based formulations designed for the treatment and prevention of oxidative stress and diabetes. Nanocurcumin was obtained through a micronization process and subsequently encapsulated within biopolymers derived from corn starch and fenugreek mucilage, achieving encapsulation rates of 75% and 85%, respectively. Subsequently, the encapsulated nanocurcumin was utilized in the formulation of sugar-free syrups based on Stevia rebaudiana Bertoni. The stability of the resulting formulations was assessed by monitoring particle size distribution and zeta potential over a 25-day period. Dynamic light scattering (DLS) revealed a particle size of 119.9 nm for the fenugreek mucilage-based syrup (CURF) and 117 nm for the corn starch-based syrup (CURA), with polydispersity indices PDIs of 0.509 and 0.495, respectively. The dissolution rates of the encapsulated nanocurcumin were significantly enhanced, showing a 67% improvement in CURA and a 70% enhancement in CURF compared with crude curcumin (12.82%). Both formulations demonstrated excellent antioxidant activity, as evidenced by polyphenol quantification using the 2.2-diphenyl 1-pycrilhydrazyl (DPPH) assay. In the evaluation of antidiabetic activity conducted on Wistar rats, a substantial reduction in fasting blood sugar levels from 392 to 187 mg/mL was observed. The antioxidant properties of CURF in reducing oxidative stress were clearly demonstrated by a macroscopic observation of the rats’ livers, including their color and appearance.

The use of natural compounds, such as curcumin, to treat infections caused by bacteria, viruses, fungi, parasites, inflammatory diseases, and various types of cancer is an active and dynamic area of research. Curcumin has a long history of use in the food industry, and there is currently a growing interest in its therapeutic applications. Numerous clinical trials have consistently shown that curcumin, a polyphenolic compound, is safe and well-tolerated even at high doses. There is no toxicity limit. However, the clinical efficacy of curcumin has been limited by its constraints. However, scientific evidence indicates that the use of adjuvants and carriers, such as nanoparticles, exosomes, micelles, and liposomes, can help overcome this limitation. The properties, functions, and human benefits of using nanocurcumin are well-supported by scientific research. Recent evidence suggests that nanocurcumin may be a beneficial therapeutic modality due to its potential to decrease gene expression and secretion of specific inflammatory biomarkers involved in the cytokinestorm seen in severe COVID-19, as well as increase lymphocyte counts. Nanocurcumin has demonstrated the ability to improve clinical manifestations and modulate immune response and inflammation in various autoinflammatory diseases. Additionally, its efficacy, affordability, and safety make it a promising replacement for residual cancer cells after tumor removal. However, further studies are necessary to evaluate the safety and efficacy of nanocurcumin as a new therapeutic in clinical trials, including appropriate dosage, frequency, and duration.

This article discusses a study that evaluated the antifungal activity of nano-curcumin against Candida species, which cause otomycosis, a fungal infection of the ear. The study found that nano-curcumin showed some antifungal activity against Candida, but it was less effective than commonly used antifungal drugs. However, due to its safety profile, nano-curcumin could be considered as a potential treatment option for otomycosis with further research. The objective of the present study was to investigate the effects of in ovo injection of nanocurcumin on intestinal histomorphology and some serum components of the hatched chicks. The chicken eggs were injected with 1, 10, 100, 1000, and 10000 ppm of nanocurcumin, and the other two groups were used as control and injected control (saline). At the end of the experiment, blood and tissue samples were collected for biochemical and histological examination. For histological examination, small intestine was sampled and routine histological methods containing fixation, dehydration, clearing and paraffin embedding were used. Sections were stained with hematoxylin & eosin for light microscopy evaluation. Blood biochemical parameters were analyzed by an automatic analyzer. Results indicated that in ovo injection of saline significantly reduced albumin, globulin, total protein and AST (aspartat aminotransferase) compared to the control group, so that injection of nanocurcumin corrected the reduction of these factors. Uric acid was increased by injection of 1, 10 and 100 ppm nanocurcumin compared to the other groups. Blood glucose levels decreased slightly with increasing the dose of nanocurcumin, and in the 1000 ppm nanocurcumin group, the levels were significantly lower than those of the control. In ovo injection of 1000 ppm nanocurcumin significantly increased the villi height and villus height/crypt depth ratio compared to the saline and 1 ppm nanocurcumin groups. Although the primary purpose of this study was to evaluate the effects of in ovo administration of nanocurcumin, it was demonstrated that the use of saline in the in ovo injection was inappropriate. Due to the diverse medicinal and pharmacokinetic properties of turmeric, it is well-known in the therapeutic, pharmaceutic, nutraceutical, cosmetic, and dietary industries. It gained importance due to its multitude of properties, such as wound-healing, anti-inflammatory, anti-oxidant, anti-microbial, cytoprotective, anti-aging, anti-cancer, and immunomodulatory effects. Even though the natural healing effect of turmeric has been known to Indians as early as 2500 BCE, the global demand for turmeric has increased only recently. A major reason for the beneficiary activities of turmeric is the presence of the yellow-colored polyphenolic compound called curcumin. Many studies have been carried out on the various properties of curcumin and its derivatives. Despite its low bioavailability, curcumin has been effectively used for the treatment of many diseases, such as cardiovascular and neurological diseases, diabetes, arthritis, and cancer. Introduction: Retina is an organ that is very sensitive to getting hurt because of its high oxygen requirement and high polyunsaturated fatty acid content. Cisplatin is used as an effective chemotherapy in different type of cancer but it reduced the cellular viability and increased the reactive oxygen species (ROS) generation. Aim of Work: The goal of this work was to investigate the role of nano curcumin particles in preventing the Cisplatin (CP) - induced retinotoxicity in adult male albino rats. Material and Methods: 30 adult male albino rats divided into three groups as following; group I was the control group, group II was the Cisplatin-treated group (single intraperitoneal injection of CP 7 mg/kg), and group III was the nano curcumin -treated group (single intraperitoneal of CP 7 mg/kg + oral nano curcumin 50 mg/kg/14 d). Results: Cisplatin (CP) single injection caused histological changes in the retinal tissue and decreased the expression of eNOS level and elevated the caspase-3 as well as the TNF-α in retinal tissue of albino rats. By colorimetric analysis, reduction in the total antioxidant and elevation in MDA levels were also seen. Co-administration of nano curcumin particles with CP restored the normal histological structure of the retina of rats and significantly improved the cellular antioxidant capacity of the retina resulting in retinal protection against the retinotoxicity and apoptotic cell death caused by the CP therapy. Conclusion: Cisplatin has severe destructive impacts on the retina and co- administration of nano curcumin particles have a protective role on Cisplatin- induced retinotoxicity. Diabetes is predicted to affect 700 million people by the year 2045. Despite the potential benefits for diabetics, curcumin's low bioavailability significantly reduces its utility. However, newer formulation methods of decreasing particle size, such as through nanotechnological advances, may improve curcumin's bioavailability and cell-absorption properties. Various curcumin nanoformulations such as nanofibers, nanoparticles-like nanostructured lipid carriers (NLCs), Solid Self-Nanoemulsifying Drug Delivery Systems (S-SNEDDS) and nanohydrogels have been evaluated. These studies reported increased bioavailability of nanoformulated curcumin compared to free curcumin. Here, we provide a detailed review of the antidiabetic effects of nanocurcumin compounds and subsequent effects on diabetic complications. Overall, various nanocurcumin formulations highly increase curcumin water-solubility and bioavailability and these safe formulations can positively affect managing some diabetes-related manifestations and complications. Moreover, nanocurcumin efficacy in various diabetes complications is discussed. These complications included inflammation, neuropathy, depression, anxiety, keratopathy, cataract, cardiomyopathy, myocardial infarction (MI), nephropathy, erectile dysfunction and diabetic wound. Moreover, several nanocurcumin formulations improved wound healing in the diabetic. However, few studies have been performed in humans, and most results have been reported from cellular and animal studies. Introduction: Previous studies have shown that converting curcumin into nanoparticles can enhance its therapeutic effect in models of convulsion. Changes in calmodulin-dependent Ca2+/protein kinase (CaMKII) are associated with some experimental models of epilepsy. This study aimed to evaluate the hippocampal CaMKII phosphorylation following the therapeutic effect of nanocurcumin in a model of pentylenetetrazole (PTZ)-induced convulsion. Methods: Nanocurcumin (with an average particle diameter of 150 nm) was produced using bovine serum albumin and was administered orally (gavage) to male NMRI mice weighing 25-30 g at doses of 50 and 100 mg/kg. One hour later, PTZ at a dose of 85 mg/kg was administered intraperitoneally to the animal. Following intraperitoneal PTZ injection, the latency of myoclonus, clonus, general tonic convulsion, and mortality rate were recorded in male mice. After behavioral testing, the anesthetized animals were euthanized, and the levels of phosphorylated and total forms of hippocampal CaMKII were determined by Western blotting technique. Results: The results showed that oral administration of nanocurcumin at doses of 50 and 100 mg/kg significantly improved convulsion induced by PTZ compared with natural curcumin. In all groups, convulsion increased hippocampal CaMKII phosphorylation, and nanocurcumin did not modulate it. Conclusion: The findings of this study showed that the therapeutic effect of nanocurcumin in a model of pentylenetetrazoleinduced convulsion was not associated with a change in hippocampal CaMKII activity pattern. The present study investigated the effects of dietary supplementation of nanocurcumin (NCur = 150 ppm) in African catfish feed for 15 days along with two doses of florfenicol—a therapeutic dose (FFC1 = 10 mg/kg.BW) and a high dose (FFC2 = 30 mg/kg.BW). Serum biochemical parameters and histopathological changes in liver and kidney tissues were determined post-feeding. Additionally, fish were experimentally challenged with a virulent strain of Aeromonas veronii to explore the fish's capacity to resist infections. Results indicated that the serum level of total protein was significantly elevated in the FFC2 + Ncur group compared with control, Ncur, and FFC2 groups. Total protein level was also higher in FFC1 + Ncur compared with that of the FFC1 group. Moreover, feeding NCur, FFC1 + Ncur, and FFC2 + Ncur resulted in a significant increase in globulin levels compared to control, FFC1, and FFC2-Fed groups. However, the albumin level was unaffected. Serum levels of AST, ALT, ALP, and urea significantly increased in FFC2-fed fish compared to controls indicating liver damage. The higher dosage of FFC2 induced histological alterations in the liver and kidney architecture, represented by a rupture of the central vein, cytoplasmic vacuolation, hepatocytes necrosis, and severe inflammation. Surprisingly, NCur inclusion improved most of the altered biochemical parameters and mitigated FFC-induced histological damage in the hepatic and renal tissues. Similarly, fish that were fed a combination of both FFC and Ncur showed a higher resistance when exposed to A. veronii infection, as the mortality rates were significantly lower than those of the control group. Our findings indicated that incorporating NCur into the diet can have multiple benefits for the sustainable aquaculture of African catfish. These advantages encompass mitigating drug-induced damage to the liver and kidney tissues, augmenting the therapeutic efficacy of FFC in a synergistic manner and providing protection against A. veronii infections. Chronic inflammation and tissue damage can result from uncontrolled inflammation during SARS-CoV-2 or COVID-19 infections, leading to post-acute COVID conditions or long COVID. Curcumin, found in turmeric, has potent anti-inflammatory properties but limited effectiveness. This study developed nanocurcumin, a curcumin nanoparticle, to enhance its physical and chemical stability and investigate its in vitro anti-inflammatory properties upon CoV2-SP induction in lung epithelial cells. Nanocurcumin was prepared by encapsulating curcumin extract in phospholipids. The particle size, polydispersity index, and zeta potential of nanocurcumin were measured using dynamic light scattering. The encapsulated curcumin content was determined using HPLC analysis. The encapsulation efficiency of curcumin was 90.74 ± 5.35% as determined by HPLC. Regarding the in vitro release of curcumin, nanocurcumin displayed a higher release content than non-nanoparticle curcumin. Nanocurcumin was further investigated for its anti-inflammatory properties using A549 lung epithelial cell line. As determined by ELISA, nanocurcumin showed inhibitory effects on inflammatory cytokine releases in CoV2-SP-stimulated conditions, as evidenced by a significant decrease in IL-6, IL-1β and IL-18 cytokine secretions compared with the spike-stimulated control group (p < 0.05). Additionally, as determined by RT-PCR, nanocurcumin significantly inhibited the CoV2-SP-stimulated expression of inflammatory genes (IL-6, IL-1β, IL-18, and NLRP3) compared with the spike-stimulated control group (p < 0.05). Regarding the inhibition of NLRP3 inflammasome machinery proteins by Western blot, nanocurcumin decreased the expressions of inflammasome machinery proteins including NLRP3, ASC, pro-caspase-1, and the active form of caspase-1 in CoV2-SP-stimulated A549 cells compared with the spike-stimulated control group (p < 0.05). Overall, the nanoparticle formulation of curcumin improved its solubility and bioavailability, demonstrating anti-inflammatory effects in a CoV2-SP-induced scenario by inhibiting inflammatory mediators and the NLRP3 inflammasome machinery. Abstract Background Oral cancer, one of the most common cancers, has unimproved 5-years survival rate in the last 30 years and the chemo/radiotherapy-associated morbidity. Therefore, intervention strategies that evade harmful side effects of the conventional treatment modalities are of need. Herbal therapy as a complementary preventive/therapeutic modality has gained attention. Curcumin is one of the herbal compounds possessing unique anticancer activity and luminescent optical properties. However, its low water solubility limits its efficacy. In contrast, curcumin at the nanoscale shows altered physical properties with enhancing bioavailability. Methods The current study evaluated the impact of nanocurcumin as an anti-oral cancer herbal remedy, comparing its efficacy against the native curcumin complement and conventional chemotherapeutic. An optimized polymeric-stabilized nanocurcumin was synthesized using the solvent-antisolvent precipitation technique. After assuring the solubility and biocompatibility of nanocurcumin, we determined its cytotoxic dose in treating the squamous cell carcinoma cell line. We then evaluated the anti-tumorigenic activity of the nano-herb in inhibiting wound closure and the cytological alterations of the treated cancer cells. Furthermore, the cellular uptake of the nanocurcumin was assessed depending on its autofluorescence. Results The hydrophilic optimized nanocurcumin has a potent cancerous cytotoxicity at a lower dose (60.8 µg/mL) than the native curcumin particles (212.4 µg/mL) that precipitated on high doses hindering their cellular uptake. Moreover, the nanocurcumin showed differential targeting of the cancer cells over the normal fibroblasts with a selectivity index of 4.5. With the confocal microscopy, the luminescent nanoparticles showed gradual nuclear and cytoplasmic uptake with apparent apoptotic cell death, over the fluorescent doxorubicin with its necrotic effect. Furthermore, the nanocurcumin superiorly inhibited the migration of cancer cells by -25%. Conclusions The bioavailable nanocurcumin has better apoptotic cytotoxicity. Moreover, its superior luminescence promotes the theranostic potentialities of the nano-herb combating oral cancer.

Objective: Postoperative atrial fibrillation (POAF) is the most frequent dysrhythmias observed following coronary artery bypass graft (CABG) surgery. Several studies have shown the beneficial effects of curcumin on cardiovascular diseases; however, there is no clinical trial to examine its effect on POAF. This randomized, double‐blind, placebo‐controlled clinical study was designed to evaluate the prophylactic effects of a nano-formulation of curcumin (SinaCurcumin™) versus placebo on POAF and levels of biomarkers of inflammation and oxidative stress in patients undergoing CABG surgery.Materials and Methods: A total of 234 eligible patients were randomized to receive 240 mg curcumin nano-formulation or placebo three days prior to the surgery and on the first four postoperative days. The occurrence of POAF was monitored for at least 96 hr after the surgery. Also, C-reactive protein (hs-CRP), malondialdehyde (MDA) and glutathione (GSH) levels were assessed at baseline and the end of the study.Results: Analyses were done in the intention-to-treat population. No significant difference was observed in the occurrence of POAF between the treatment (9.5%) and placebo (11.5%) groups. Also, curcumin intervention did not alter serum concentration of the hs-CRP, MDA, or GSH in comparison with placebo.Conclusion: In conclusion, it seems that perioperative treatment with SinaCurcumin™ did not prevent POAF after CABG surgery.

Phytotherapeutics is widely used nowadays as an alternative to the current antifungal drugs to reduce their side effects. Curcumin, with its wide therapeutic array as antioxidant and anti-inflammatory agent, is one of the natural compounds that ha..s an antifungal effect, especially when being used at nanoscale to increase its bioavailability. Our research aimed to evaluate clinically and microbiologically the effect of using topical nanocurcumin suspension to treat oral candidiasis. After 4 days from induction of oral candidiasis (baseline), we randomly divided 39 female BALB/c mice into three groups of 13 animals; nanocurcumin, nystatin, and sham groups. All animals in nanocurcumin and nystatin groups received topical treatment twice daily for 10 days. Then, we performed clinical and microbiological evaluations at baseline, day 5, and day 10. By the end of treatment, our results revealed that nanocurcumin promoted a significant reduction in the number of candida colonies. There was no statistically significant difference neither clinically nor microbiologically between nanocurcumin and nystatin groups. In conclusion, nanocurcumin has a good antifungal effect as nystatin, however, its therapeutic efficacy takes a longer time to appear than nystatin. The enhanced bioavailability of curcumin at the nanoscale qualifies this nano-herb as a promising alternative therapy for oral candidiasis, evading nystatin-associated morbidity. Background: Nowadays, attention is directed to herbal treatments in an attempt to lessen the adverse effects of diabetes. Nanoformulation of curcumin (NC) was shown to enhance stability and water solubility compared to native curcumin. Objective: To examine the effect of different NC concentrations on the histopathological structure of the submandibular salivary gland of diabetic rats. Methods: 28 rats were divided equally into 4 groups. Group I: Control group, Group II (diabetic), III (diabetic + nanocurcumin low dose), and IV (diabetic + nanocurcumin high dose): Rats of groups II, III and IV were injected with a single dose of alloxan (140 mg/kg) to induce diabetes. After 7 days, groups III and IV were treated for 6 weeks with NC (100 mg/kg/day) for group III, and (200 mg/kg/day) for group IV. Submandibular salivary glands were assessed histologically, immunohistochemically using α smooth muscle actin (α SMA) and ultrastructurally. Results: Diabetic samples showed destruction of parenchymal elements of the gland, with thick fiber bundles encircling the excretory ducts and minimal reaction for α SMA. Amelioration of the gland's architecture was detected in groups III and IV with reduction of collagen deposition and elevation of positive immunoreactivity to α SMA. Conclusion: NC profoundly repaired the induced diabetic histopathological and ultrastructural alterations of the gland in a dose dependent manner.

Background and Objective: Oxidative stress is one of the causes of tinnitus. The therapeutic effect of nanocurcumin on the antioxidant processes of various diseases has been confirmed; however, no study, to the best of our knowledge, has been conducted on its therapeutic effect on tinnitus. Therefore, the present study aimed to determine the effect of nanocurcumin on treating tinnitus. Materials and Methods: This study was a randomized controlled trial (triple-blind intervention). A total of 62 patients with tinnitus were selected by randomized block design with observance of concealment. Then, the participants were divided into two groups of 31 and were treated with nanocurcumin and a placebo. Both groups were similar in terms of age and gender with frequency matching methods. Hearing tests were performed for all patients. Patients received 80 mg/day of nanocurcumin and similar placebo capsules for 21 days. Results: The mean and standard deviation of the age in the nanocurcumin and placebo groups were 44.77 ± 15.2 and 46.32 ± 15.2 years, respectively (P>0.05). The difference between the mean duration of tinnitus was not significant in both groups (P>0.05). The results indicated that the antioxidant nanocurcumin reduced the tinnitus score (P=0.02). However, nanocurcumin failed to affect the frequency, high score, degree of annoyance, mean score of speech discrimination, and speech reception threshold (P>0.05). Also, no side effect was reported from using nanocurcumin. Conclusion: Nanocurcumin is not effective in the treatment of tinnitus.

Curcumin is widely known for its antibacterial, antioxidant and anti inflammatory effects and has been reported to possess anticancerous activity as well. However, its medical application is limited because of poor bioavailability and rapid metabolism. In this study, we encapsulated curcumin in solid lipid nanoparticles and studied its anticancerous effect in Dalton's Ascites Lymphoma (DAL) mice model. The physicochemical characteristics of curcumin solid lipid nanoparticles (CUR-SLN) were assessed and the anticancer efficacy was determined by in vivo studies. The curcumin solid lipid nanoparticles were synthesized by solvent emulsification evaporation method with particle size less than 100 nm. Antitumor effect of nanocurcumin (50 mg/kg) and curcumin (100 mg/kg) was evaluated in Dalton's Ascites Lymphoma bearing mice. Pathological and immunohistochemical parameters were studied. Mean survival time and percentage increase in lifespan were assessed. Nanocurcumin group showed more significant influence in reducing tumor volume and weight, inducing apoptosis, reducing angiogenesis and invasion restoring antioxidant parameters and increased mean survival time. Curcumin and nanocurcumin inhibited the activation of nuclear factor-kappa B (Nf-kB), and thereby proved the pathway by which it induced anti-angiogenic and anti-invasive property. Background: Megakaryopoiesis is characterized by progressive polyploidization and the expression of megakaryocytic markers. Numerous transcription factors and physiological signaling pathways regulate this phenomenon. Megakaryocyte differentiation induction in the K562 cell line and hematopoietic stem cells via nanocurcumin drug has been identified in our previous study. K562 cells are typical Chronic Myelogenous Leukemia (CML) cells that are resistant to apoptosis and express the bcr-abl fusion gene. These cells have the potential to differentiate into erythrocytes and megakaryocytes. Curcumin is well known as a component with strong potential to alter NFκB activity in various cells. NFκB pathway regulates various genes such as apoptotic and immune response genes. The aim of the current study is to evaluate the possible role of nanocurcumin in NFκB pathway regulation during the megakaryopoiesis process in the K562 cell line. Materials and Methods: Megakaryocyte markers expression and phenotype alteration of nanocurcumintreated K562 cells have been detected by flow cytometry and microscopy imaging. The nuclear level of the RelA (p65) subunit of NFκB was determined by western blot test in K562 cells during megakaryopoiesis induction via nanocurcumin treatment at different times. The expression of NFκB target genes including c-MYC, BAX, and NQO1 was also analyzed in nanocurcumin-treated K562 cells by quantitative RT-PCR assay at different times. Results: It was demonstrated that nanocurcumin leads to an increase in NFκB activity transiently during megakaryocyte differentiation, which is followed by a change in the expression of c-MYC, BAX, and NQO1 target genes. Conclusion: The NFκB pathway can be considered a new pathway for inducing megakaryocyte differentiation by nanocurcumin for the purposes of in vitro and in vivo megakaryopoiesis experiments. Background: Oral squamous cell carcinoma (OSCC) is the sixth most common mouth cancer in the world. The aim of the present study is comparing the effects of using Nanocurcumin, and photodynamic therapy (PDT), alone or together in treatment of OSCC in rats. Methods: Forty Wister male rats were divided into Control (group 1), 650 nm diode Laser only (group 2), Nanocurcumin alone (group 3), and PDT with a combination of laser with Nanocurcumin (group 4). Then, OSCC in the tongue induced by dimethylbenz anthracene (DMBA). The treatments were evaluated clinically, histopathologically, and immunohistochemically through BCL2 and Caspase-3 genes expression. Results: Positive control with OSCC displayed significant weight loss, while PDT group gained more than nanocurcumin treated groups as well as laser groups comparing with control positive group. The histological examination of the tongue in PDT group showed improvement. In laser group, there were partial loss of surface epithelium with various ulcers and dysplasia and partial improvement by this type of treatment. The tongue in the positive control group showed ulcer in the dorsum surface with inflammatory cells, hyperplasia of the mucosa membrane around the ulcer (acanthosis) with increase of dentition, vacuolar degeneration of prickle cell layer and increase mitotic activity of basal cell layer together with dermal proliferation. Conclusions: Under the condition of the present study, PDT using nanocurcumin photosensitizer was effective in the treatment of OSCC regarding clinical, histological and gene expression of BCL2 and Caspase-3. The study aims to investigate the potential effect of nanocurcumin as feed additive in the diet of Oreochromis niloticus to improve its growth performance, health status and resistance against Aspergillus flavus. The control group was fed on a basal diet without nanocurcumin, and four diets T1, T2, T3 and T4 were supplemented with 10, 25, 40 and 55 mg/kg of nanocurcumin, respectively, in triplicate (20 fish/replicate). The duration of the feeding trial was 60 days. The final body weight, weight gain, specific growth rate and survival rate showed significantly (p < 0.05) increased values in the nanocurcumin groups than the control. Fish fed with nanocurcumin supplementation showed improvement in RBCs, haemoglobin, total protein, albumin and globulin while there was a decrease in the liver enzymes (AST and ALT), glucose and alkaline phosphatase. The creatinine was also decreased in fish fed nanocurcumin. The digestive enzymes amylase and lipase increased in the nanocurcumin‐treated groups, and the triglycerides values showed non‐significant increase, whereas the cholesterol values showed non‐significant decrease in T1 and T4. Meanwhile, the cortisol was nearly the same in all groups. At the end of the trial, the fish were challenged with Aspergillus flavus for 15 days. Aspergillus flavus resulted in the mortality of 100% of the control group and the groups with lower doses of nanocurcumin (T1 and T2) within the first week and second week post challenge respectively. In the treatments with high doses (T3 and T4), higher survival rates were recorded in a dose‐dependent manner. The pathogenicity of Aspergillus flavus was confirmed histopathologically. It was concluded that the dietary supplementation of nanocurcumin enhanced the health status of O. niloticus by improving the haemato‐immunological response and body composition parameters of the fish, and protected it from the Aspergillus flavus infection with optimum inclusion levels of 25–40 mg/kg diet. Lipopolysaccharide (LPS) proved to be an important tool, not only in the induction of neuroinflammatory models, but also in demonstrating the behavioral and cognitive consequences of endotoxemia. Curcumin, in its native form, has proven to be a worthy candidate for further development as it protects the dopaminergic neurons against LPS-induced neurotoxicity. However, it remains hindered by its poor bioavailability. In this study we aim to explore the possible molecular mechanism of LPS-induced neurotoxicity and the possible protective effects of orally supplemented nanocurcumin. Thirty-six adult male Wistar rats weighing 170–175 g were divided into six groups and treated with single I.P. (intra-peritoneal) dose of LPS (sigma and extracted; separately) (5 mg/kg BW) plus daily oral nanocurcumin (15 mg/kg BW). The rats were followed for 7 days after the LPS injection and nanocurcumin supplementations daily via oral gavage. After scarification, the levels of neurotransmitters, antioxidants, and amyloidogenesis markers were assessed in brain tissues. Nanocurcumin showed adequate antioxidant and neuroprotective effects, rescuing the rats which had been injected intraperitoneally with LPS endotoxin. [ABSTRACT FROM AUTHOR]

Abstract Background The adverse effect of aflatoxin in broilers is well known. However, dietary supplementation of Saccharomyces cell wall and/or Nanocurcumin may decrease the negative effect of aflatoxin B1 because of the bio-adsorbing feature of the functional ingredients in Yeast Cell Wall and the detoxification effect of curcumin nanoparticles. The goal of this study was to see how Saccharomyces cell wall/Nanocurcumin alone or in combination with the aflatoxin-contaminated diet ameliorated the toxic effects of aflatoxin B1 on broiler development, blood and serum parameters, carcass traits, histology, immune histochemistry, liver gene expression, and aflatoxin residue in the liver and muscle tissue of broilers for 35 days. Moreover, the withdrawal time of aflatoxin was measured after feeding the aflatoxicated group an aflatoxin-free diet. Broiler chicks one day old were distributed into five groups according to Saccharomyces cell wall and/or nanocurcumin with aflatoxin supplementation. The G1 group was given a formulated diet without any supplements. The G2 group was supplemented with aflatoxin (0.25 mg/kg diet) in the formulated diet. The G3 group was supplemented with aflatoxin (0.25 mg/kg diet) and Saccharomyces cell wall (1 kg/ton diet) in the formulated diet. The G4 group was supplemented with aflatoxin (0.25 mg/kg diet) and nanocurcumin (400 mg/kg) in the formulated diet. The G5 group was supplemented with aflatoxin (0.25 mg/kg diet) and Saccharomyces cell wall (1 kg/ton diet) in combination with nanocurcumin (200 mg/kg) in the formulated diet. Results According to the results of this study, aflatoxin supplementation had a detrimental impact on the growth performance, blood and serum parameters, carcass traits, and aflatoxin residue in the liver and muscle tissue of broilers. In addition, aflatoxin supplementation led to a liver injury that was indicated by serum biochemistry and pathological lesions in the liver tissue. Moreover, the shortening of villi length in aflatoxicated birds resulted in a decrease in both the crypt depth ratio and the villi length ratio. The expression of CYP1A1 and Nrf2 genes in the liver tissue increased and decreased, respectively, in the aflatoxicated group. In addition, the aflatoxin residue was significantly (P ≤ 0.05) decreased in the liver tissue of the aflatoxicated group after 2 weeks from the end of the experiment. Conclusion Saccharomyces cell wall alone or with nanocurcumin attenuated these negative effects and anomalies and improved all of the above-mentioned metrics.

This study aimed to assess the effects of nanocurcumin as a phyto-additive on red tilapia foundation stock under different salinities. Seven hundred and twenty Florida red tilapia (Oreochromis sp.), equally divided by sex, participated in a 72-day experiment. Initially weighed (males: 46.7 ± 0.37 g; females: 33.2 ± 0.51 g), they were acclimated in 36 concrete tanks separately (triplicates per treatment). Twelve treatments (six for each sex) evaluated nanocurcumin's impact (100 mg/kg diet) at salinities of 2 (control), 18 and 28‰. Nanocurcumin and salinity interaction significantly influenced various parameters. Fish fed the nanocurcumin diet exhibited significantly enhanced growth parameters across all salinities and sexes, along with improved feed utilization indicated by lower feed conversion ratios. However, a slight decrease in survival rate was observed at the highest salinity. The nanocurcumin diet altered whole-body composition in males, with decreased moisture and increased protein, lipid, and ash content compared to the control diet, whereas females showed sex-specific responses. Fish fed nanocurcumin diet modulated the internal organ indices (HSI, VSI, TSI and GSI). Both diet and salinity significantly influenced digestive enzyme activities, with nanocurcumin consistently increasing protease, amylase, and lipase activities. Additionally, both factors significantly impacted reproductive performance, sex hormone, blood parameters, stress indicators (glucose and cortisol), growth hormone levels, as well as antioxidants and non-specific immunity (SOD, CAT, and LYZ). Abstract Curcumin is a polyphenolic molecule with antibacterial, antioxidant, anti‐inflammatory, and antimicrobial properties. This study aimed to prepare nanocurcumin by encapsulating in biopolymers to improve its stability, bioavailability, water‐solubility, antibacterial efficiency against methicillin‐resistant Staphylococcus aureus. Three effective variables of curcumin concentration, polymer concentration, and water volume on curcumin‐loaded polymer nanoparticles, were optimized. The average size of polyacrylic acid (PAA), polyvinyl alcohol (PVA), and polyethyleneimine (PEI) nanoparticles were obtained 75.2, 77.1, 86.4 nm, respectively. The nanoparticles had a spherical shape, a smooth and uniform surface morphology. The MIC of PAA, PVA, and PEI nanoparticles was 0.480, 0.390, and 0.340 mg/mL, respectively and the MIC of PAA, PVA, and PEI combined with methicillin was 0.330, 0.260, and 0.200 mg/mL, respectively. According to the results, curcumin‐loaded PEI nanoparticles had the highest inhibitory effect against methicillin‐resistant S. aureus among the synthesized nanoparticles. The results showed that solvent volume, polymer concentration and curcumin concentration had a significant effect on particle size. The inhibitory properties of curcumin nanoparticles significantly increased due to the smaller particle size and increased penetration into the bacterium. Curcumin‐loaded nanoparticles can be promising drug carriers for the treatment of infections, cancer, and other diseases.

The therapeutic activities of curcumin have long been investigated in some chronic and inflammatory diseases. This study was designed to investigate the protective effects of nanocurcumin on intestinal barrier function, apoptosis, and oxidative stress in rats exposed to traffic noise. Forty rats were divided into four groups: two traffic noise-exposed groups of animals that received either vehicle (NOISE) or nanocurcumin (NCUR + NOISE) and two control groups that either remained intact (CON) or received nanocurcumin (NCUR). Nanocurcumin injection (15 mg/Kg/ip) and traffic noise exposure were administered daily for two weeks. The relative protein expression of intestinal tight junctions, occludin, and ZO-1 and Bax/Bcl-2 ratio was measured to evaluate barrier integrity and apoptosis in intestinal samples, respectively. Plasma D-lactate concentration was examined as a criterion of intestinal permeability. Corticosterone, superoxide dismutase (SOD) activity, glutathione (GSH), total antioxidant capacity (TAC), and nitrite were measured in serum. The noise exposure increased Bax/Bcl-2 ratio, corticosterone, and oxidative stress in the NOISE animals. Nanocurcumin treatment improved the Bax/Bcl-2 ratio and reduced corticosterone and oxidative stress in the NCUR + NOISE animals. The expression of tight junction proteins was decreased while the concentration of D-lactate was increased in the NOISE animals. Nanocurcumin did not efficiently impact the expression of tight junction proteins and the D-lactate level in the NCUR + NOISE group. Nanocurcumin administration displayed antioxidant and anti-apoptotic roles in the noise-exposed rats, however, it did not affect the intestinal barrier integrity. We concluded that reduced apoptosis in the intestine might be related to the antioxidant activity of nanocurcumin and its modulatory effects on the HPA axis in the nanocurcumin-treated animals.

Abstract We report a facile room temperature telescoping synthesis of a nanocurcumin complex with 17.5‐fold permeation enhancement as determined by comparative in vitro permeation study with raw curcumin. The permeation results were further validated with in silico drug absorption prediction using ADMET predictors.
