Multiple stressors linked to anthropogenic activities can influence how organisms adapt and evolve. So far, a consensus on how multiple stressors drive adaptive trajectories in natural populations has not been reached. Some meta‐analysis reports show predominance of additive effects of stressors on ecological endpoints (e.g., fecundity, mortality), whereas others show synergistic effects more frequently. Moreover, it is unclear what mechanisms of adaptation underpin responses to complex environments. Here, we use populations of Daphnia magna resurrected from different times in the past to investigate mechanisms of adaptation to multiple stressors and to understand how historical exposure to environmental stress shapes adaptive responses of modern populations. Using common garden experiments on resurrected modern and historical populations, we investigate (1) whether exposure to one stress results in higher tolerance to a second stressor; (2) the mechanisms of adaptation underpinning long‐term evolution to multistress (genetic evolution, plasticity, evolution of plasticity); and (3) the interaction effects of multiple stressors on fitness (synergism, antagonism, additivity). We measure the combined impact of different levels of resource availability (algae) and biocides on fitness‐linked life‐history traits and interpret these results in light of historical environmental exposures. We show that exposure to one stressor can alter tolerance to second stressors and that the interaction effect depends on the severity of either stressor. We also show that mechanisms of adaptation underpinning phenotypic evolution significantly differ in single‐stress and multistress scenarios. These adaptive responses are driven largely by synergistic effects on fecundity and size at maturity, and additive effects on age at maturity. Exposure to multiple stressors shifts the trade‐offs among fitness‐linked life‐history traits, with a stronger effect on Daphnia populations when low‐resource availability and high biocide levels are experienced. Our study indicates that mitigation interventions based on single‐stress analysis may not capture realistic threats.

The use of adapted, efficient and reliable tools is fundamental to measure and control the environmental impact surrounding industrial installations. Robust, autonomous and working by remote control, HFNI (High Frequency Non Invasive) Valvometry is one possible solution for the biomonitoring of aquatic ecosystems. Based on valve activity scanning, it enables the study of behavior and different life traits of bivalve mollusks in the natural environment. Animals are used as biosensors representative of water quality and capable of reflecting the occurrence of environmental alteration. Detecting a disturbance and identifying its nature are key elements in the process of reducing environmental risk. As such, the first objective of this work was to analyze the behavioral response to crude oil of bivalve mollusks instrumented with HFNI Valvometry in a multistress context. The second objective was to evaluate the underlying disturbances occurring in their milieu intérieur when exposed to crude oil with the ultimate goal of associating behavioral changes (symptoms) and physiological disorders. To do so, studies were conducted on two species, the freshwater clam Corbicula fluminea and the marine oyster Magallana gigas, under controlled laboratory or semi-natural conditions, with crude oil at environmentally realistic concentrations, constant or variable and in one case lethal. We used various stress factors: water temperature, turbidity, presence of barium, noise pollution and hypoxia. The behavioral response to crude oil was found to be (i) clear and distinguishable in the majority of situations and (ii) concentration-dependent, allowing to propose a global relationship between a valvometric index and crude oil concentration in the range 0 - 60 mg·L-1. Disturbances in the milieu intérieur were assessed using a proteomic approach on tissues directly involved in behavioral activity (gills, muscles, nervous ganglia). Whatever the tissue studied, the species or the exposure conditions, the results reflect a disruption of metabolic and cellular processes with a dose-effect toxicological impact quantified by PAH analyses in tissues or nominal exposure values. As a whole, this new set of analyses (i), confirms the capacities of the valvometry concept in the oil and gas field, (ii) make it possible to approach the relationship between behavior and underlying physiological changes in bivalves and (iii) pave the way to new perspectives for online and remote impact assessment., L’utilisation d’outils adaptés, efficaces et fiables est fondamentale pour mesurer et contrôler l’impact environnemental autour des installations industrielles. Robuste, autonome et permettant un suivi à distance et en ligne, la Valvométrie HFNI (Haute Fréquence Non Invasive) est une solution possible pour le biomonitoring des écosystèmes aquatiques. Elle permet l’étude à distance et en temps quasi-réel du comportement et de différents traits de vie de groupes de mollusques bivalves dans le milieu naturel par numérisation de leur activité valvaire. Les animaux sont ainsi utilisés comme des témoins représentatifs de la qualité de l’eau, capables de traduire la survenue d’évènements modifiant leur environnement. Détecter rapidement le début d’une perturbation et identifier sa nature sont des éléments clés du processus de réduction du risque environnemental. A ce titre, le premier objectif de cette thèse était d’analyser la capacité de réponse comportementale face à un pétrole brut de mollusques bivalves instrumentés avec la Valvométrie HFNI en situation multistress, représentative d’un contexte environnemental réaliste. Le second objectif était d’évaluer les perturbations sous-jacentes de leur milieu intérieur lors de l’exposition au pétrole brut et mieux comprendre le mécanisme des changements de comportement. Pour ce faire, des études ont été réalisées chez deux espèces de mollusques bivalves, la palourde d’eau douce Corbicula fluminea et l’huitre marine Magallana gigas, en conditions semi-naturelles ou contrôlées de laboratoire, avec des concentrations de pétrole brut constantes ou variables, environnementales (et dans un cas létales), et en présence de différents facteurs de stress additionnels : la température de l’eau, la turbidité, la présence de baryum, de pollution sonore ou d’hypoxie. La réponse comportementale au pétrole brut s’est avérée (i) claire et discernable dans la majorité des situations créées et (ii) fonction de la concentration, ce qui permet de proposer un schéma global d’évolution d’indices valvométriques dans la gamme 0 – 60 mg·L-1. Les perturbations du milieu intérieur ont été évaluées par une approche de protéomique sur des tissus en relation avec l’activité comportementale (branchies, muscles, ganglions nerveux). Quel que soit le tissu étudié, l’espèce ou les conditions d’exposition, les résultats traduisent une modification des processus métaboliques et cellulaires avec un impact toxicologique fonction de la contamination quantifiée par des dosages de HAP dans les tissus ou des valeurs nominales d’exposition. Ces analyses permettent d’aborder les bases explicatives du comportement chez les bivalves, de corréler comportement et changements du milieu intérieur et ouvrent la voie à des perspectives d’évaluation d’impacts en ligne et à distance.

Surface waters worldwide are threatened by anthropogenic influences. Many of these waters gain part of their flow from groundwater and it has been noted that surface water and groundwater are effectively a single resource. However, the importance of this connection between surface- and groundwaters in multi-stressed catchments is not yet well understood. The research presented in this thesis therefore aimed to quantify the impact and importance of the groundwater contribution to streams and aimed to improve understanding of how the dynamics of the groundwater affect the propagation of stressors to streams. A conceptual framework is presented for the analysis of linked multiply-stressed GW-SW systems. This framework is illustrated by applying it on four European catchments to analyse the propagation and buffering of multi-stressors through groundwater to surface waters. The importance of the groundwater system varies between the catchments due to differences in catchment characteristics and stressor combinations. Results from these catchments show that the effect of stressors can propagate through the groundwater system towards surface waters, and groundwater thus functions as a connection between a catchment and its stream(s). In addition to this, the flow of groundwater over potentially long distances and over longer time scales creates a buffer capacity to the effect of stressors by adding time lags and attenuating stressor signals, although this strongly dependents on groundwater flow paths and the shape and dynamics of groundwater travel time distributions. Travel time distributions are studied by calculating them dynamically for three Dutch lowland catchments using a high-resolution spatially-distributed 3D groundwater flow model in a novel way. These detailed distributions showed that the streams consist of younger water in the wet periods and older water in dry periods due to the interplay between the activation of shallow short flow paths contributing young water towards for instance shallow drains and the intensification of fluxes through all flow paths when groundwater levels rise. The calculated TTDs were combined with chemical input curves to reconstruct concentrations in the stream water. We were able to explain the short-term variability of nitrate and chloride in the stream with the variable contribution of different groundwater flow paths and the time-variable part of the catchment that is contributing to streamflow. We used the model conceptually to advance our understanding of time lags in the breakthrough of agricultural nitrate. Parameters such as the thickness of the unsaturated zone, mean travel time, input patterns and the distance of agricultural fields determine time lags between peak inputs and maximum concentrations in the stream. The importance of groundwater for stream temperature is investigated by locating seepage in two of the study streams by measuring temperatures using long fibre-optic cables in combination with measurements of the groundwater tracer Radon-222. Using the field measurements, a stream temperature model was made to separate the different energy fluxes which showed that the cooling effect of groundwater. Scenario modelling showed that groundwater in groundwater-fed catchments may play an important role in the mitigation of climate warming because of its capacity of reducing summer maximum temperatures.

The vulnerability level of natural flounder populations was assessed, integrating responses at the molecular, individual and population levels. A multi-estuary approach was carried out on the fish responses in contrasted environments (thermal stress: Mondego estuary; chemical stress: Seine estuary; moderately stressed systems: Bay of Douarnenez, Vilaine and Canche estuaries).A demo-genetic approach, considering the variability of microsatellites, underlined a reduced effective size for the southern peripheral population of the Mondego estuary, thus characterized by a high ecological risk.In the Seine population, a reduced interindividual variability was observed considering the expression levels of genes involved in bioenergetics (COII, 12S); this pattern could explain the reduced ability of this population to cope with another stress (ie thermal stress).Signatures of adaptation to pollutants (high level of energetic metabolism, management of oxidative stress, modification of membrane phopholipids) were observed in the Seine, but could be very costly (reduced muscle energetic reserve). Thus, we consider that the Seine population is displaying a high ecological risk.A one month fish caging experiment was conducted in the Seine estuary. Fish responses were analyzed by proteomic; they underlined a decreasing pollution gradient from upstream to downstream, and thus allowed to characterize the ecological status of the estuary atmicroscale.This study highlighted pertinent tools for the assessment of flounder population vulnerability and for the exploration of the ecological status of estuarine systems., Le niveau de vulnérabilité de populations naturelles de flet a été abordé dans cette étude, par l’intégration des réponses du poisson aux niveaux moléculaire, individuel et populationnel. Une approche multi-estuaire est développée ; les réponses du flet sont ainsi mesurées dans des conditions contrastées (stress thermique : estuaire du Mondego ; stress chimique : estuaire de Seine ; faibles niveaux de stress : Baie de Douarnenez, estuaires de Vilaine et Canche). Par une approche démo-génétique basée sur la variabilité des marqueurs microsatellites, nous avons mis en évidence une faible taille efficace de la population du Mondego située en limite Sud de l’aire de distribution, et donc caractérisée par un risque écologique élevé. La population de Seine présente une faible variabilité interindividuelle dans l’expression de différents gènes impliqués dans le métabolisme énergétique (COII, 12S), qui pourrait expliquer son faible potentiel à résister au réchauffement climatique. Cette population montre des signatures d’adaptation face au stress chimique (métabolisme énergétique élevé, gestion du stress oxydant, modification des phospholipides membranaires), engendrant probablement un coût physiologique fort (réduction des réserves énergétiques au niveau musculaire). La population de Seine présente donc un risque écologique élevé. Enfin, un encagement de flet a été mené sur un mois en estuaire de Seine; les réponses du poisson analysées par protéomique shot-gun ont mis en évidence un gradient de pollution décroissant amont-aval, soit une estimation de l’état écologique du système à micro-échelle. Cette thèse a identifié différents outils pertinents pour estimer le niveau de vulnérabilité de populations de flet, et pour explorer l’état écologique des écosystèmes estuariens.

In their natural environment, plants are generally confronted with multiple co-occurring stresses. However, the interaction between stresses is not well known and transcriptomic data in response to combined stresses remain scarce. This study aims at characterizing the interaction between transcriptomic responses to biotic stress and nitrogen (N) limitation. Plants were grown in low or full N, infected or not with Erwinia amylovora (Ea) and plant gene expression was analyzed through microarray and qRT-PCR. Most Ea-responsive genes had the same profile (induced/repressed) in response to Ea in low and full N. In response to stress combination, one third of modulated transcripts responded in a manner that could not be deduced from their response to each individual stress. Many defense-related genes showed a prioritization of their response to biotic stress over their response to N limitation, which was also observed using Pseudomonas syringae as a second pathosystem. Our results indicate an interaction between transcriptomic responses to N and biotic stress. A small fraction of transcripts was prioritized between antagonistic responses, reflecting a preservation of the plant defense program under N limitation. Furthermore, this interaction also led to a complex and specific response in terms of metabolism and cellular homeostasis-associated genes.

Regulations, such as the EU Water Framework Directive, for the protection of the aquatic environment, encourage the development of new sensitive and predictive tools to evaluate the effects of contaminants on aquatic living organisms. In this context, energy metabolism biomarkers are of interest given the central place of energy in all physiological processes (i.e. defense, growth and reproduction). In this study, we addressed for the first time three digestive enzymes (amylase, trypsin and intestinal alkaline phosphatase) and evaluated their interest as novel biomarkers in the threespine stickleback (Gasterosteus aculeatus), a model species in multidisciplinary fields of biology, and recently in ecotoxicology. The interest of the two digestive enzymes as biomarkers was evaluated in comparison with other parameters, usually applied in ecotoxicology studies: biometric parameters, reproduction parameters, antioxidant parameters, and other energy metabolism parameters. For that, juvenile sticklebacks were exposed for 3 months in semi-static conditions to cadmium at 0.5 μg.L-1 (chemical stress), and two water temperatures 16°C and 21°C (physical stress). Results showed that the high temperature (21 °C) was the major stress factor on stickleback physiology. The very low cadmium concentration used in this study had no effect on most of the studied parameters (i.e. growth parameters, antioxidant parameters and parents' reproduction parameters). However, digestive enzymes showed an interesting sensitivity to cadmium exposure which was emphasis at 21 °C (significant decrease). Although cadmium had no effect on parental reproduction, negative consequences on their offspring survival were observed after in vitro fertilization trials (decreased survival capacity). Compared to conventional measurements, digestive enzymes responded rapidly, which could be an important advantage for their use as early warning tool, to reflect organism’s health.

La production d’antibiotiques et la résistance aux antibiotiques est un des mécanismes d’interaction entre microorganismes des sols. L’antibiose par production d’antibiotique est un mécanisme très ancien utilisé par les bactéries et les champignons pour limiter le développement de bactéries compétitrices. Les sols sont donc un réservoir potentiel de bactéries résistantes aux antibiotiques (BRA) et de gènes de résistance aux antibiotiques (GRA). Ce pool de gènes de résistance des sols est appelé le résistome des sols, il constitue un réservoir de GRA potentiellement transmissibles aux agents pathogènes de l’Homme et des animaux. Plusieurs pratiques agricoles peuvent conduire à l’introduction de BRA et de GRA, mais aussi de résidus d’antibiotiques (fluoroquinolones, sulfamides, cyclines …) dans les sols. D’abord, l’amendement des sols avec des produits résiduaires organiques (PRO) qu’ils soient d’origine urbaine (boues issues du traitement des eaux usées, compost d’ordures ménagères …) ou agricole (fumiers, lisiers, digestats …) peut conduire à la contamination des sols par des BRA ou des GRA. Une deuxième pratique agricole pouvant conduire à la contamination des sols est l’irrigation des cultures ou des parcs avec des effluents de station d’épuration des eaux usées. Cette pratique déjà utilisée couramment dans de nombreux pays arides risque de se développer en France en lien avec le changement climatique. Ces PRO ou ces effluents liquides contiennent de plus des résidus d’antibiotiques (et d’autres produits pharmaceutiques) non dégradés lors des traitements de ces déchets. Ces résidus d’antibiotiques peuvent induire l’émergence ou sélectionner de nouveaux gènes ou mécanismes de résistance dans les sols, mais également modifier la structure des communautés microbiennes des sols. L’enrichissement des sols agricoles ou des sols urbains (utilisés pour des activités récréatives) en bactéries résistantes aux antibiotiques et en GRA peut induire des risques pour la santé : i) risque de contamination des productions végétales ou des animaux, ii) risque de contamination des ressources en eau (par ruissellement ou lessivage des sols contaminés). L’atténuation de la contamination des sols requiert donc des traitements supplémentaires (compostage, ozonation, désinfection par les UV ou désinfection thermique) des PRO ou des effluents avant rejet dans l ‘environnement. La persistance de BRA et de GRA dans les sols cultivés peut augmenter le risque de contamination des productions végétales et donc de nos aliments (en particulier ceux consommés à l’état frais). Le transfert de GRA, ayant émergés dans les sols, vers des pathogènes humains par transfert horizontal de gènes (HGT) sera augmenté. De tels évènements se sont déjà produits en particulier pour l’émergence des souches résistantes aux céphalosporines de troisième génération et aux beta-lactamines en général, cette famille comprenant de nombreux antibiotiques de dernier recours. L’OMS parle de pandémie mondiale pour ces souches dont les gènes ancestraux de résistance proviennent de souches environnementales, que l’on retrouve en particulier dans les sols. L’exposé visera donc à déterminer l’impact des BRA et des résidus d’antibiotiques sur le résistome des sols et aussi à définir les paramètres qui vont influencer la survie des BRA et la persistance des GRA dans les sols.

Plants are immobile organisms which have to adapt to abiotic and biotic constraints. Among bioticstress, established or emerging viral diseases, may be responsible for major yield losses withsignificant consequences. Genetic control is the most effective, environmentally and consumerfriendlyway to control viral infections. Understanding plant/virus interactions remains essential tosearch for new sources of resistance. This work, focuses on the study of the natural pathosystemArabidopsis thaliana/Turnip mosaic virus (TuMV). Most of the trials were conducted in commongarden conditions allowing the analysis of the interaction in a multistress environment. A. thaliana’sresponse was explored through the study of disease-related traits and the variations in primary andsecondary metabolites. This work allows i) the fine characterization of A. thaliana’s response toTuMV in multistress conditions through the exploration of the natural diversity of a world and Frenchpopulation ii) to determine the genetic architecture of this interaction by genome wide associationsand QTL mapping. Several new loci potentially involved in the response have been identified iii) tohighlight the interest of metabolic phenotyping to discriminate accessions according to theirsusceptibility to TuMV. The multidisciplinary approaches contribute to a better characterization andunderstanding of plant-virus interaction., Les plantes sont des organismes immobiles qui doivent répondre et s’adapter à des contraintes abiotiques et biotiques. Parmi les stress biotiques, les maladies virales, établies ou émergentes, peuvent être responsables de pertes de rendement majeures aux conséquences économiques importantes. Face aux phytovirus la lutte génétique constitue le moyen de lutte le plus efficace, le plus respectueux de l’environnement et du consommateur. Comprendre l’interaction entre les plantes et les virus reste indispensable pour rechercher de nouvelles sources de résistances. Ce travail de thèse s’intéresse à l’étude du pathosystème naturel Arabidopis thaliana/Turnip mosaic virus (TuMV). Les essais ont été menés majoritairement en conditions extérieures permettant une analyse de l’interaction dans un environnement multistress. La réponse d’A. thaliana a été explorée par l’étude de traits liés à la maladie et par la variation en métabolites primaires et secondaires. Ce travail a permis i) de caractériser de façon fine la réponse d’A. thaliana au TuMV en conditionsmultistress en exploitant la diversité naturelle d’une population mondiale et française ii) de déterminer l’architecture génétique de cette interaction par des approches de génétique d’association et de QTL mapping. Plusieurs nouveaux loci potentiellement impliqués dans la réponse ont été identifiés iii) de montrer l’intérêt du phénotypage métabolique pour discriminer les accessions en fonction de leur sensibilité au TuMV. La multidisciplinarité des approches constitue la richesse de ce travail de thèse qui contribue à une meilleure caractérisation et compréhension de la réponse des plantes lors d’une infection virale.

National audience; Stenotrophomonas maltophilia is a ubiquitous bacterial species known to be a soil and rhi- zospheric inhabitant as well as a human opportunistic pathogen. Multi-drug resistant strains are frequently found in hospitals because of its ability to acquire resistance genes through horizontal gene transfer and its possession of efflux pumps. Recent data also showed the link between antibiotic resistance and metal resistance among both clinical and environmen- tal strains. In this study, we evaluated over a 3 months study the impact of a cocktail of antibiotics (ciprofloxacin and sulfamethoxazole) and metals (Cu and Zn) on indigenous S. maltophilia in soil using an experimental design of soil columns exposed to manure contain- ing various concentrations of the cocktail. In parallel, the toxicant adsorption, distribution, and migration in the soil matrix were measured to interpret the observed biological response. Data showed that regardless of the concentration of the cocktail i.e. from low (1X PEC) to high (40X PEC) doses, no changes in population abundance were observed after 7 and 100 days of exposure. Genetic diversity (REP-profile) analysis among more than 650 isolates showed that metals were the dominant stressors. Antibiotic resistance gene abundance (sulfonamide and quinolone) was unaltered and no resistance gene transfer was observed. Finally, no changes were observed on the phenotypic (antibiotic resistance) properties at low or high antibiotic doses. These data suggest the lack of a dose/response effect as well as the lack of synergic effects between metals and antibiotics on the genotypic and phenotypic evolution of S. maltophilia populations.

Ethylenediurea (EDU) is a synthetic chemical known to protect plants from the phytotoxic effects of tropospheric ozone (O3). Although many studies have proposed the use of EDU for studying the O3 effects under field conditions, its mechanism of action is not fully understood, and it is unclear whether it exerts a specific antiozonant action, or if it may also interact with other oxidative stresses. The aim of this work was to evaluate the effect of EDU on forest species in a Mediterranean environment where, during summer, vegetation is exposed to multiple oxidative stresses, such as O3 and drought. The experiment was conducted on Fraxinus ornus L. (Manna ash) plants growing in six mesocosms, three maintained under full irrigation, while the other three were subjected to drought for 84 days. In each mesocosm, three plants were sprayed every 15 days with 450 ppm EDU. Gas exchange and chlorophyll “a” fluorescence measurements carried out through the experimental period highlighted that EDU did not affect stomatal conductance and had an ameliorative effect on the functionality of drought-stressed plants, thus suggesting that it may act as a generic antioxidant. The implications of these findings for the applicability of EDU in field studies are discussed.

In dit rapport worden de graadmeters voorgesteld voor het compartiment water ten behoeve van de Milieubalans/Milieuverkenningen. Graadmeters worden behandeld voor zowel zoute als zoete oppervlaktewateren. Het zoete oppervlaktewater is onderverdeeld in Rijkswateren (rivieren, grote kanalen en meren in beheer bij Rijkswaterstaat) en regionale wateren (beheerd door lokale waterbeheerders). De regionale wateren zijn onderverdeeld in meren, sloten, kanalen en vaarten, en stromende wateren. De graadmeters voor de Rijkswateren en een aantal regionale wateren worden behandeld volgens de thema's verzuring, vermesting en verspreiding. M.b.t. alle regionale wateren wordt een classificering naar ecologisch niveau gebruikt. De nadruk bij de keuze van de graadmeters ligt op de chemische randvoorwaarden voor een duurzame ecosysteem ontwikkeling. Enige zeer kenmerkende biologische graadmeters voor de genoemde thema's worden eveneens gebruikt. Een speciaal hoofdstuk is gewijd aan waterbodems. Sommige effecten worden niet veroorzaakt door een bepaald thema, maar komen tot stand door meerdere actoren. Zij worden besproken in het hoofdstuk over multiple stress. Volksgezondheidseffecten van het oppervlaktewater worden als apart thema behandeld.

Dit rapport is een verantwoording achteraf van de werkwijzen, berekeningen, keuzen en aannamen bij de paragraaf over "Milieukwaliteit en natuur" in de vijfde Nationale Milieuverkenning (MV5). Deze MV5-paragraaf behandelt de invloed van de milieukwaliteit op de ecologische kwaliteit van de levende natuur in Nederland.Om deze invloed aan te geven is de indicator 'milieustress' geintroduceerd. Milieustress wordt gebruikt om uit te drukken hoe soorten reageren op veranderingen in de milieukwaliteit. Het wordt berekend uit de afname in voorkomen van soorten t.o.v. een referentiesituatie (1950) als gevolg van veranderingen in milieukwaliteit. Ook zijn de aandelen van de verschillende milieufactoren in de totale milieustress aangegeven.Voor een aantal soortgroepen en milieuthema's is de omvang van de milieustress vastgesteld op basis van kwantitatieve modelberekeningen. Voor andere soortgroepen en milieuthema's waar geen modellen voor beschikbaar waren zijn semikwantitatieve inschattingen door deskundigen gemaakt. Deze twee schattingswijzen worden aanvullend en naast elkaar gebruikt.De in de MV5 gepresenteerde methode voor het bepalen van de milieustress geeft de huidige stand van kennis weer. Met de gevolgde werkwijze en indicator is het mogelijk gebleken aan te geven wat de invloed is van de milieukwaliteit op de natuur voor de huidige situatie en 2030 (volgens scenario's). De invloeden van enkele milieuthema's blijven echter onduidelijk en de onzekerheden zijn soms groot.

A controlled experiment was conducted in order to understand how functional and structural traits of species with different leaf habits (Fraxinus ornus and Quercus ilex) shift as a consequence of nitrogen (N) addition (30 kg ha yr−1) and to explore the effect that N has on the water stress response. The experiment was divided in two stages: stage I, N addition under well-watered condition; stage II, N addition under drought. Functionality of the photosynthetic machinery, growth and biomass partitioning were assessed. The N content at leaf level increases in F. ornus only, which invests resources on photosynthetic machinery, whereas Q. ilex tends to store N in non-photosynthetic biomass, increasing relative growth rate and biomass, resulting in different allometric ratio. This effect may play a role in water stress response. Stomatal conductance of Q. ilex treated with N and subjected to water stress is lower relative to drought treatment. In a warming climate, higher temperatures are likely to modulate positively or negatively the effect of other environmental factors on biota, although such interactions are poorly documented. Here, we explore under controlled conditions the combined effects of two common stressors in freshwater ecosystems, higher temperature and sediment load, on the embryonic development of arctic charr ( Salvelinus alpinus L.). In the warm treatment, embryos had a lower survival, a longer incubation period and a smaller body size with a bigger yolk sac volume. Our data show a significant interaction between temperature and sediment load with temperature increasing dramatically the negative effects of sediment load on fitness-related traits. In the climate change context, these findings highlight the importance of taking into account different thermal scenarios when examining the effect of environmental or anthropogenic stressors.

The originality of this project consists in the interdisciplinary approach gathering the impact of toxic phytoplankton blooms and metallic pollution on health (behaviour, stress, immunity, genetic) and disease development of the Manila clam, an economieally important species. The Manila clam Ruditapes philippinarum is the second exploited bivalve in the world. It was introduced in Arcachon bay during the 80's and is today exploited by professional fishermen (57 licences). A 50% decrease of the stock was observed during the last five years. Beyond fishing pressure, a PhD thesis identified several diseases as potential threats, like perkinsosis (=Dermo) and brown muscle disease (BMD). The project confirms the settlement of these pathologies with high prevalence and intensity, impacting clams growth and condition index. Perkinsosis and BMD rather develop in elevated mudflats with lower water turnover (north-western area). The hypothesis that the BMD agent would be a virus is still valid although it had not been yet demonstrated. Phytoplanctonic related toxins in Manila clams remain very low but the occurrence of spirolids justifies our project to experiment multi-contamination. Beyond the technological challenges (toxic algae culture, multitreatments and multi-responses protocol), this project highlighted effects of "toxie algae" and/or "metal" contamination on histologieal, immune, behavioural and molecular parameters. Our expertise was partly acquired with the assistance of the Fishing Committee and, in return, provided help this Committee for their own fishing strategies, L'originalité de ce projet réside dans le développement d'une approche interdisciplinaire intégrant l'impact des efflorescences d'algues toxiques et la pollution métallique sur la biologie (comportement, stress, immunité, génétique) et le développement des maladies d'un bivalve fouisseur d'intérêt économique, la palourde. La palourde japonaise Ruditapes philippinarum est la seconde espèce de bivalve exploitée dans le monde. Espèce introduite dans le bassin d'Arcachon dans les années 1980, elle soutient aujourd'hui une pêche professionnelle forte de 57 licences. Cependant, ces cinq dernières années ont vu le stock chuter. Outre la pêche, un certain nombre de maladies se présentent comme menace potentielle, notamment la perkinsose et la maladie du muscle marron (BMD). Le projet confirme l'installation de ces pathologies à des niveaux élevés de prévalence et d'intensité, impactant la croissance et l'indice de condition des palourdes. Perkinsose comme BMD sont favorisés sur les estrans les plus émergeants et sur les zones à plus faible renouvellement d'eau (secteur nord-ouest). L'hypothèse d'un agent pathogène de la BMD de nature virale reste forte, même si la démonstration n'a encore pu être faite. Si les palourdes du bassin d'Arcachon concentrent peu de toxines d'origine phytoplanctoniques (2010-11), la présence de spirolides suffit à justifier les expériences de contamination croisées menées dans ce projet. Au-delà des défis techniques (cultures d'algues toxiques, multiplicité des traitements et des mesures), ce projet a mis en évidence des atteintes histologiques, immunitaires, comportementales et moléculaires liées à la contamination «algues toxiques» et/ou «métaux». L'expertise acquise a été en partie possible grâce à une aide technique du Comité des Pêches et, en retour, a permis d'orienter le Comité dans certaines de ses stratégies.

Een van de aandachtspunten in het milieubeleid is de voorspelling van effecten van milieuveranderingen op ecosystemen. Het Multistress mOdel voor de VEgetatie (MOVE) beantwoordt aan deze behoefte. Binnen dit model wordt een relatie gelegd tussen veranderingen van bodemeigenschappen, zoals zuurgraad, bemestingsgraad en vochttoestand en de effecten daarvan op de vegetatie (de vegetatiemodule). Deze studie is bedoeld om de mogelijkheden na te gaan van het ontwikkelen van een bodemfauna-module (BOEF) analoog aan MOVE. Deze bodemfauna-module kan de vegetatie-module aanvullen in gebieden waar de natuurlijke vegetatie nagenoeg is verdwenen, zoals in landbouwgebieden, en kan wellicht effecten van sommige stress-factoren beter modelleren. Hierbij zijn 3 vragen van belang:1 Zijn er voldoende gegevens over het voorkomen van bodemfauna en bijbehorende milieufactoren beschikbaar ? 2 Bestaan er relaties tussen het voorkomen van bodemfauna- soorten en relevante milieufactoren ? 3 Zijn deze gevonden relaties te gebruiken in het kader van voorspelling en eventueel normstelling ? Aangezien de bodemfauna uit zeer veel verschillende diergroepen bestaat, die vrijwel nergens in onderlinge samenhang worden bestudeerd, is er in deze studie een diergroep uitgekozen waarvan redelijk veel bekend is. De diergroep die gekozen is, is de klasse der nematoden. Zoveel mogelijk gegevens, afkomstig van verschillende onderzoeksgroepen in Nederland, over deze diergroep zijn bijeengebracht. Vervolgens is nagegaan welke milieufactor het meest bepaald was op de locaties waar nematodengegevens van beschikbaar waren. Dit bleek pH te zijn. Vervolgens werden met behulp van logistische regressie relaties berekend tussen het voorkomen van de taxa en de pH. Om de bruikbaarheid van de modellen na te gaan is voorbeeldsgewijs een aantal toepassingen uitgewerkt. In totaal bleken ruim 700 punten bemonsterd te zijn, verspreid over 207 locaties. Hiervan was van 657 punten de pH bekend. In totaal werden ruim 200 verschillende taxa gevonden. Van 87 taxa konden responsiecurven berekend worden. Van deze taxa kon voor 15 taxa geen significant model worden gevonden, terwijl 34 een unimodale responsie vertoonden met pH en 38 een lineaire responsie. Een mogelijke toepassing van de modellen in het kader van normstelling is onderzocht door het aantal taxa wat potentieel kan voorkomen te relateren aan de pH. Het blijkt dat bij pH lager dan 5 het aantal potentiele taxa drastisch afneemt, bij een pH lager dan 4 kunnen nog slechts 50% van de soorten zich handhaven. De modellen kunnen ook gebruikt worden om het voorkomen van de taxa te voorspellen op plaatsen waar geen gegevens van bekend zijn. Dit is gebeurd aan de hand van een willekeurig gekozen genus Choaonolaimus. Het daadwerkelijk voorkomen van het genus en het voorspelde voorkomen bleek redelijk overeen te komen, maar ook is het duidelijk dat pH niet de enige factor is die het voorkomen bepaald. Een derde toepassing is de vergelijking tussen verschillende typen van stress in een bepaald gebied. Er wordt in dit rapport niet naar meerdere factoren gekeken, dus die vergelijking is hier niet mogelijk. Wel is het mogelijk om met de modellen indices te berekenen die verschillende typen van stress aangeven. In dit verband zijn de Maturity Index en de Plante-Parasieten Index uitgerekend. De conclusies van deze studie zijn als volgt samen te vatten. Met de beschikbare gegevens is het mogelijk om voor bodemfauna-taxa responsiecurven af te leiden voor zuurgraad. Andere factoren zouden eveneens meegenomen kunnen worden, zij het tot op minder uitgebreid niveau. De berekende curven zijn op verschillende manieren toe te passen en zijn bruikbaar voor het voorspellen van effecten van milieuveranderingen op ecosystemen. Een vergroting van de gegevensset zou een verbetering van de modellen opleveren en zou kunnene leiden tot een groter aantal taxa bruikbaar voor toepassingen.
