Meshless Shepard interpolation method (MSIM) is used to simulate the destruction process within the arch reinforcement area of tunnel lining. In this method, the shape functions are formed by the partition of unity and the finite cover technology, which is not affected by discontinuous domains, with delta property at any desired node, and applies boundary conditions in an easy and correct way. The MSIM method combines the advantages of both the conventional meshless method and the numerical manifold method. The finite cover technology enables the shape functions to not be affected by the discontinuities in the solution domain, which overcomes the difficulty resulting from the conventional meshless method. The finite covers and the partition of unity functions are formed using the influence domains of a series of nodes, which removes the obstacle from conventional numerical manifold method and has simpler formation of finite covers than numerical manifold method. Virtual crack closure technique is used to calculate the intensity factor of crack-tip stress. The results of progressive destruction process simulation on the cracking patterns within the arch reinforcement area indicate that the method is suitable for tracking the crack propagation in complex stress conditions. Fluctuations in groundwater levels along the coast have a significant impact on the extent of saltwater intrusion into freshwater aquifers. This study aims to simulate the groundwater flow and solute transport in the region by using the mSim toolbox in the MATLAB and COMSOL Multiphysics. The investigation is focussed on a micro-basin of Pavanje river located along the west coast of India. The model results are calibrated and validated against the field observations. The results show that the variation of the water table over the year is significant and range from about 3–14 m. There exists a reasonable correlation between the simulated and observed values of groundwater level and salinity. The wells that are most vulnerable to seawater intrusion in the region are identified. The COMSOL model estimated a salinity range of 0–20 mol/m3. Additionally, the model is used to understand the response of coastal aquifer to various stress scenarios. Furthermore, it demonstrates the use of an adapted direct search algorithm for calculating the holograms that allows for imaging across an extended field of view. The system was tested on fixed targets and live cells yielding a two times resolution increase over conventional diffraction‐limited imaging. It provides a significant improvement in the efficiency of pattern projection and more importantly it allows for the testing of more diverse excitation patterns than possible with amplitude‐only projection. For example, PSF engineering using phase modulation can be easily incorporated into the calculated holograms, potentially generating subdiffraction structures in the excitation pattern. The ability to incorporate PSF engineering into SIM opens up holographic MSIM as a potential method for further increasing resolution with little or no change to the imaging system. Structured illumination microscopy is a super-resolution technique used extensively in biological research. However, this technique is limited in the maximum possible resolution increase. Here we report the results of simulations of a novel enhanced multi-spot structured illumination technique. This method combines the super-resolution technique of difference microscopy with structured illumination deconvolution. Initial results give at minimum a 1.4-fold increase in resolution over conventional structured illumination in a low-noise environment. This new technique also has the potential to be expanded to further enhance axial resolution with three-dimensional difference microscopy. The requirement for precise pattern determination in this technique also led to the development of a new pattern estimation algorithm which proved more efficient and reliable than other methods tested.

The human cytomegalovirus (HCMV) viral mitochondria-localized inhibitor of apoptosis (vMIA) protein, traffics to mitochondria-associated membranes (MAM), where the endoplasmic reticulum (ER) contacts the outer mitochondrial membrane (OMM). vMIA association with the MAM has not been visualized by imaging. Here, we have visualized this by using a combination of confocal and superresolution imaging. Deconvolution of confocal microscopy images shows vMIA localizes away from mitochondrial matrix at the Mitochondria-ER interface. By gated stimulated emission depletion (GSTED) imaging, we show that along this interface vMIA is distributed in clusters. Through multicolor, multifocal structured illumination microscopy (MSIM), we find vMIA clusters localize away from MitoTracker Red, indicating its OMM localization. GSTED and MSIM imaging show vMIA exists in clusters of ~100–150 nm, which is consistent with the cluster size determined by Photoactivated Localization Microscopy (PALM). With these diverse superresolution approaches, we have imaged the clustered distribution of vMIA at the OMM adjacent to the ER. Our findings directly compare the relative advantages of each of these superresolution imaging modalities for imaging components of the MAM and sub-mitochondrial compartments. These studies establish the ability of superresolution imaging to provide valuable insight into viral protein location, particularly in the sub-mitochondrial compartments, and into their clustered organization.

The endoplasmic reticulum (ER) membrane is closely apposed to the outer mitochondrial membrane (OMM), which facilitates communication between these organelles. These contacts, known as mitochondria-associated membranes (MAM), facilitate calcium signaling, lipid transfer, as well as antiviral and stress responses. How cellular proteins traffic to the MAM, are distributed therein, and interact with ER and mitochondrial proteins are subject of great interest. The human cytomegalovirus UL37 exon 1 protein or viral mitochondria-localized inhibitor of apoptosis (vMIA) is crucial for viral growth. Upon synthesis at the ER, vMIA traffics to the MAM and OMM, where it reprograms the organization and function of these compartments. vMIA significantly changes the abundance of cellular proteins at the MAM and OMM, including proteins that regulate calcium homeostasis and cell death. Through the use of superresolution imaging, we have shown that vMIA is distributed at the OMM in nanometer scale clusters. This is similar to the clusters reported for the mitochondrial calcium channel, VDAC, as well as electron transport chain, translocase of the OMM complex, and mitochondrial inner membrane organizing system components. Thus, aside from addressing how vMIA targets the MAM and regulates survival of infected cells, biochemical studies and superresolution imaging of vMIA offer insights into the formation, organization, and functioning of MAM. Here, we discuss these insights into trafficking, function, and organization of vMIA at the MAM and OMM and discuss how the use of superresolution imaging is contributing to the study of the formation and trafficking of viruses. In this paper, we report on electronic and field emission properties of metal–semiconductor–insulator–metal (MSIM) heterostructures employing semiconducting and insulating hydrogenated amorphous carbon (a-C:H) films. The a-C:H layers were deposited from plasma polymerisation of methane in a 20 kHz discharge. The current as a function of voltage (I–V) characteristic is consistent with Fowler–Nordheim (F–N) type tunnelling of carriers from semiconductor cathode into polymer-like a-C:H. The onset field for emission is about ∼6 V/μm. The transition barrier is found to be ∼0.05 eV, which is close to that obtained with a-C:H:N/Si heterojunctions. capacitance–voltage (C–V) measurements show negligible hysteresis and zero flat band voltage, and the interface states density, estimated from capacitance–frequency plot, is very low (NSS∼4×1010 cm−2).
