Every fungal cell is encapsulated in a cell wall, essential for cell viability, morphogenesis, and pathogenesis. Most knowledge of the cell wall composition in fungi has focused on ascomycetes, especially human pathogens, but considerably less is known about early divergent fungal groups, such as species in the Zoopagomycota and Mucoromycota phyla. To shed light on evolutionary changes in the fungal cell wall, we studied the monosaccharide composition of the cell wall of 18 species including early diverging fungi and species in the Basidiomycota and Ascomycota phyla with a focus on those with pathogenic lifestyles and interactions with plants. Our data revealed that chitin is the most characteristic component of the fungal cell wall, and was found to be in a higher proportion in the early divergent groups. The Mucoromycota species possess few glucans, but instead have other monosaccharides such as fucose and glucuronic acid that are almost exclusively found in their cell walls. Additionally, we observed that hexoses (glucose, mannose and galactose) accumulate in much higher proportions in species belonging to Dikarya. Our data demonstrate a clear relationship between phylogenetic position and fungal cell wall carbohydrate composition and lay the foundation for a better understanding of their evolution and their role in plant interactions.

Fucoidan, a sulfated polysaccharide found primarily in brown algae, is known for exhibiting various biological activities, many of which have been attributed to its sulfate content. However, recent advancements in techniques for analyzing polysaccharide structures have highlighted that not only the sulfate groups but also the composition, molecular weight, and structures of the polysaccharides and their monomers play a crucial role in modulating biological effects. This review comprehensively provides the monosaccharide composition, degree of sulfation, molecular weight distribution, and linkage of glycosidic bonds of fucoidan, focusing on the diversity of its biological activities based on various characteristics. [Display omitted] As different monosaccharides exhibit different redox characteristics, this paper presented a novel colorimetric sensor array based on the glucose oxidase-like (GOx-like) activity of Au nanoparticles (NPs) for monosaccharides identification. AuNPs can use O 2 , ABTS+•, or [Ag(NH 3) 2 ]+ as an electron acceptor to catalyze the oxidation of monosaccharides in different velocity, resulting in cross-responsive signals. The current sensor array can distinguish between different monosaccharides or their mixtures through linear discriminant analysis (LDA) and hierarchical clustering analysis (HCA). Moreover, the glucose and fructose concentrations can be estimated simultaneously using a neural network regression model based on the sensor array. Given the limited specificity and accuracy observed in the current official colorimetric quantification of polysaccharide in Lycium barbarum, our study aims to establish a novel, specific, accurate, and economic pre‐column derivatization ultra‐high‐performance liquid chromatography (UHPLC) method for determining the monosaccharide and polysaccharide content in L. barbarum. The optimization of extraction, hydrolysis, and derivatization (using 1‐phenyl‐3‐methyl‐5‐pyrazolone) processes for polysaccharide from L. barbarum was conducted initially, followed by separation of nine monosaccharides within 20 min using UHPLC with a C18 column. Subsequently, a novel method known as quantitative analysis of multiple components by single marker was developed, utilizing either additive 2‐deoxy‐D‐ribose or any monosaccharide present in the sample as a single reference standard to simultaneously detect the contents of polysaccharide and nine monosaccharides in L. barbarum. To validate the accuracy of the established method, the quantitative results of our approach were compared to both external and internal standard method methods. The minimal relative errors in the quantitative determination of monosaccharides among the three methods confirmed the dependability of the method. By analyzing 20 batches of L. barbarum samples, D‐galacturonic acid exhibited the highest content and the polysaccharide levels ranged from 3.02 to 13.04 mg/g. Polygonati Rhizoma, as a traditional medicinal herb, possesses pharmacological effects enhancing physical strength and immunity. In this study, a systematic analysis of the monosaccharide and non-polysaccharides components in Polygonati Rhizoma was conducted using pre-column derivatization high-performance liquid chromatography (HPLC–DAD) and liquid chromatography coupled to electrostatic orbitrap high-resolution mass spectrometry (LC–Orbitrap-MS) techniques. The polysaccharides from Polygonati Rhizoma were initially extracted, hydrolyzed, and derivatized with 1-phenyl-3-methyl-5-pyrazolone (PMP), resulting in the successful detection of five monosaccharides. The high sensitivity and specificity of the HPLC–DAD method were confirmed. Furthermore, by comparing the external standard method (ESM) and the quantitative analysis of multi-components by single-marker (QAMS) revealed that D-mannose is the most abundant monosaccharide in Polygonati Rhizoma. The LC–Orbitrap-MS analysis of Polygonati Rhizoma led to the identification of 53 compounds, including organic acids, amino acids, amides, saponins, alkaloids, esters, and others. The secondary sludge of wastewater treatment plants is an abundant and problematic bacterial biomass that accumulates nutrients from wastewater, mainly as proteins and carbohydrates. Recent studies have focused on energy recovery of this biomass by anaerobic digestion to produce biogas. However, fractional recovery of the sludge components could increase its value and provide the basis for a biorefinery based on this waste. Since ≈ 40-60% of the bacterial dry weight is protein, this biomass could be an important source of functional peptides or amino acids, and the carbohydrates could be used to produce bioplastics or biofuels. This study compares chemical, physical and biological hydrolysis methods and their sequential and assisted combinations to recover proteins and carbohydrates from sludge. Ultrasound-assisted alkaline treatment provided the highest protein solubilization yield (97.2%) with low degradation, resulting in peptide recovery yields of 75.1% with sizes from 70-215 kDa, 40% of essential amino acids and purity of 35.3% with NaOH 1M. The hydrothermal-alkaline combination almost completely solubilized the proteins but not the carbohydrates (77.4%) with high degradation (52.6%). The hydrothermal-acidic combination achieved high carbohydrate solubilization (94%) and recoveries of glucose (63.6%) and xylose (12.6%) but low protein recovery (43.7%) as small size peptides.

This study was performed in order to explore the correlation between the monosaccharide composition and hypoglycemic activity of Berberi dasystachya polysaccharides (BDPs). BDPs were extracted with hot water from B. dasystachya fruit from five regions of Qinghai province (I–V). The chemical compositions and primary structures of BDPs were determined, and their in vitro hypoglycemic activity was evaluated in terms of α-glucosidase and α-amylase inhibitory activities, and their protective effect against apoptosis induced by glucolipotoxicity (GLTy) in RIN-m5F cells was assessed. The results showed that BDPs were heteropolysaccharides connected by β-glycosidic bonds that had apyranose backbone without a triple-helix conformation, and exhibited excellent thermal stability at temperatures below 200 ℃. In addition, the monosaccharide composition and molecular mass of BDPs varied significantly depending on geographical origin. BDP-I showed good hypoglycemic activity in vitro, and the inhibitory rate of BDP-I on α-amylase was as high as 67.41% (P < 0.05). BDPs exhibited good protective effects on GLTY-induced apoptosis in RIN-m5F cells, and BDP-I had the best cell proliferation-promoting activity, which effectively decreased the levels of reactive oxygen species (ROS) and tumor necrosis factor-α (TNF-α). Finally, correlation analysis showed that monosaccharides such as glucose (Glc), galactose (Gal), rhamnose (Rha) exhibited a strongly positive correlation with hypoglycemic activity in vitro, and there was a strongly negative correlation between mannose (Man) and ROS levels. This study provides a theoretical basis for the utilization and development of B. dasystachya Maxim. berry in terms of hypoglycemic activity.

BackgroundCurrent assessment of dietary carbohydrates does not adequately reflect the nutritional properties and effects on gut microbial structure and function. Deeper characterization of food carbohydrate composition can serve to strengthen the link between diet and gastrointestinal health outcomes.ObjectivesThe present study aims to characterize the monosaccharide composition of diets in a healthy US adult cohort and use these features to assess the relationship between monosaccharide intake, diet quality, characteristics of the gut microbiota, and gastrointestinal inflammation.MethodsThis observational, cross-sectional study enrolled males and females across age (18-33 y, 34-49 y, and 50-65 y) and body mass index (normal, 18.5-24.99 kg/m2; overweight, 25-29.99 kg/m2; and obese, 30-44 kg/m2) categories. Recent dietary intake was assessed by the automated self-administered 24-h dietary recall system, and gut microbiota were assessed with shotgun metagenome sequencing. Dietary recalls were mapped to the Davis Food Glycopedia to estimate monosaccharide intake. Participants with >75% of carbohydrate intake mappable to the glycopedia were included (N = 180).ResultsDiversity of monosaccharide intake was positively associated with the total Healthy Eating Index score (Pearson's r = 0.520, P = 1.2 × 10-13) and negatively associated with fecal neopterin (Pearson's r = -0.247, P = 3.0 × 10-3). Comparing high with low intake of specific monosaccharides revealed differentially abundant taxa (Wald test, P < 0.05), which was associated with the functional capacity to break down these monomers (Wilcoxon rank-sum test, P < 0.05).ConclusionsMonosaccharide intake was associated with diet quality, gut microbial diversity, microbial metabolism, and gastrointestinal inflammation in healthy adults. As specific food sources were rich in particular monosaccharides, it may be possible in the future to tailor diets to fine-tune the gut microbiota and gastrointestinal function. Ulva rigida green macroalgae contain a variety of polysaccharides. A recent study investigated the optimum concentration and yield of polysaccharide extraction from oven-dried U. rigida biomass using a water-soluble polysaccharide extraction method that adhered to safety standards. This study utilised complete factorial experiments to examine the effects of varying factors on polysaccharide extraction. Results showed a positive correlation between increased levels of all factors and higher polysaccharide extraction yield. This study also found that the main factors and their interaction had a significant impact on the extracted polysaccharides from U. rigida. The highest polysaccharide content and yield were 9.5 mg/mL and 189 mg/g, respectively. Water-soluble polysaccharides demonstrated the presence of reducing sugar (8 mg/g), phenolics (0.69 mg/g) and flavonoids (1.42 mg/g) and exhibited antioxidant properties. Results revealed that freeze-dried polysaccharide powders were primarily composed of the monosaccharide rhamnose. Preliminary results on the effect of these powders on probiotics demonstrated that supplementation of polysaccharides from U. rigida promoted viable Lactobacillus rhamnosus ATCC 53103 growth during cultivation. This discovery has the potential to revolutionise the human food industry and promote the development of functional ingredients for novel and future food products, with numerous applications in the nutraceutical and pharmaceutical industries. Monosaccharides are vital building blocks in bioengineering applications; however, their extraction from intricate mixtures is challenging and uses substantial amounts of energy. Polymers of intrinsic microporosity (PIMs) offer an innovative avenue for separating monosaccharides. We modified PIM‐1membranes to improve the glucose/xylose separation by incorporating polyethylene glycol monomethyl ether (mPEG). The optimal mPEG (molecular weight: 1000 Da; mass fraction: 2.5 %; solvent: methanol) delivered a xylose separation coefficient of 2.62. With the hybrid membrane of PIM‐1‐mPEG (50 w.t.%) and hydrophilic Tröger's base polymerer (DMBP‐TB, 50 w.t.%), the separation factor for xylose/glucose in an aqueous solution was 2.51 for single‐stage running and 11.32 after five‐stage running. There are large fractions of micropores for PIM‐1‐mPEG, and there is difference on solute‐membrane interactions for pentose/hexose, which are regarded to be the main driving force for the high pentose/hexose selectivity in methanol. The blending of PIM‐1‐mPEG and DMBP‐TB, integrates the microporosity and hydrophilicity, finally endues the high pentose/hexose selectivity in aqueous solution. These microporous membranes are promising materials for efficiently separating monosaccharides and jnl> small organic molecules while minimizing energy consumption. It was found by the photocolorimetry method that binary compositions of protocatechic acid with monosaccharides (galactose, mannose) exhibit a pronounced antiradical synergistic effect in reaction with 2,2ʹ-diphenyl-1-picrylhydrazyl in acidic media. The synergistic properties of phenol-saccharide mixtures decrease with increasing pH of the medium. Using NMR spectroscopy and density functional theory, it was shown that the mechanism of synergism consists in the formation of intermolecular hydrogen–bonded phenol-monosaccharide complexes. The ionization energies of the donor and donor-acceptor complexes are lower than those of protocatechuic acid and hence the complexes react more actively with the radical compared to the initial antioxidant. It is important to recognize the great diversity of monosaccharides commonly encountered in animals, plants, and microbes, as well as to organize them in a visually interesting style that also emphasizes their similarities and relatedness. This article discusses the nature of building blocks, monosaccharides, and monosaccharide derivatives—terms commonly used in discussing "glycomolecules" found in nature. To aid in awareness of monosaccharide diversity, here is presented a Periodic Table of Monosaccharides. The rationale is given for construction of the Table and the selection of 103 monosaccharides, which is largely based on those presented in the KEGG and SNFG websites of monosaccharides, and includes room to enlarge as new discoveries are made. The Table should have educational value and is intended to capture the attention and foster imagination of those not very familiar with glycosciences, and encourage researchers to delve deeper into this fascinating area. Background: Cellulose is an organic carbon source that can be used as a sustainable alternative for energy, materials, and chemicals. However, the substantial challenge of converting it into soluble sugars remains a major obstacle in its use as a biofuel and chemical feedstock. A new class of enzymes knowns as copper-dependent polysaccharide monooxygenases (PMOs) or lytic polysaccharide monooxygenases (LPMOs) can break down polysaccharides such as cellulose, chitin, and starch through oxidation. This process enhances the efficiency of cellulose degradation by cellulase. Results: The genome of the fungus Magnaporthe oryzae, the causal agent of rice blast disease, contains the MGG_00245 gene, which encodes a putative PMO referred to as MoAA16. MoAA16 has been found to be highly expressed in planta during the early stages of fungal infection. The gene was optimized for heterologous expression in Pichia pastoris, and its oxidative cleavage activity on cellulose was characterized by analyzing soluble oligosaccharide products using highperformance anion exchange chromatography (HPAEC-PAD). The reaction catalyzed by MoAA16 requires 2 electrons from an electron donor, such as ascorbic acid, and aerobic conditions. It primarily produces Glc1 to Glc4 oligosaccharides, as well as oxidized cellobionic and cellotrionic acids. MoAA16 has been observed to enhance cellulase hydrolysis on phosphoric acid swollen cellulose (PASC) substrate, resulting in the production of more monosaccharide products. Conclusions: Our findings reveal the successful heterologous expression of MoAA16 in P. pastoris and its cellulose-active PMO properties. The molecular complexity of the carbohydrates consumed by humans has been deceptively oversimplified due to a lack of analytical methods that possess the throughput, sensitivity, and resolution required to provide quantitative structural information. However, such information is becoming an integral part of understanding how specific glycan structures impact health through their interaction with the gut microbiome and host physiology. This work presents a detailed catalogue of the glycans present in complementary foods commonly consumed by toddlers during weaning and foods commonly consumed by American adults. The monosaccharide compositions of over 800 foods from diverse food groups including Fruits, Vegetables, Grain Products, Beans, Peas, Other Legumes, Nuts, Seeds; Sugars, Sweets and Beverages; Animal Products, and more were obtained and used to construct the "Davis Food Glycopedia" (DFG), an open-access database that provides quantitative structural information on the carbohydrates in food. While many foods within the same group possessed similar compositions, hierarchical clustering analysis revealed similarities between different groups as well. Such a Glycopedia can be used to formulate diets rich in specific monosaccharide residues to provide a more targeted modulation of the gut microbiome, thereby opening the door for a new class of prophylactic or therapeutic diets.

Polysaccharides are the main group of macromolecules in wines. Climate change is a major problem for viticulturists as it leads to the production of unbalanced grapes. This is attributed to a mismatch between the technological maturity and phenolic maturity of grapes, which can negatively impact the production of high quality wines. To mitigate this effect, biostimulants can be applied to grapevines. For the first time in the literature, this work studied the foliar application of methyl jasmonate plus urea (MeJ + Ur) on the vineyard and its effect on the monosaccharide and polysaccharide composition of Tempranillo grapes and wines over two consecutive seasons. To achieve this, the extraction and precipitation of polysaccharides was conducted, and the identification and quantitation of monosaccharides was performed via GC–MS. The effect of MeJ + Ur foliar treatment in both the grapes and wines was season-dependent. The MeJ + Ur treatment had a slight impact on the monosaccharide composition of the grapes and also demonstrated a small effect on the wines. Multifactor and discriminant analysis revealed that the season had a greater influence on the monosaccharide and polysaccharide composition of grapes and wines compared to the influence of MeJ + Ur treatment. Interestingly, the MeJ + Ur-treated wines exhibited a higher sensory evaluation than the control wines in the second vintage. To gain further insights into the effect of MeJ + Ur foliar application on the monosaccharide and polysaccharide composition of grapes and wines, further investigations should be conducted.

In 2002, Wolf described a new species of Stenotrophomonas, Stenotrophomonas rhizophila, which is a non-pathogenic plant inhabitant. The defining characteristics of the new species, in contrast to S. maltophilia, were the following: growth at 4 °C, but its absence at 37 °C; the use of xylose as a carbon source; low osmolytic tolerance. S. rhizophila is isolated exclusively from the rhizosphere or from the internal tissues of plants, in particular from the vascular tissues of the root and stem. In 2014, a number of authors, studying cultivated heterotrophic bacteria and yeasts, which dominate in plant samples collected from various terrestrial biotopes near the Ukrainian Antarctic station on Galindez Island in Marine Antarctica, isolated a number of bacterial isolates. The authors' phylogenetic analysis using only BLAST analysis made it possible to determine the approximate phylogenetic affiliation of the bacterial isolate to the family Gammaproteobacteria (genus Stenotrophomonas, species S. rhizophila). It is known that many processes that determine the biological features of microorganisms, the nature of their relationships between them, as well as micro- and macroorganisms in biocenoses, are carried out with the participation of the surface structures of the bacterial cell, which are in direct contact with the environment. Among them, of particular interest are lipopolysaccharides (LPS), components of the outer membrane of Gram-negative bacteria. However, to date, we have not found any works devoted to the study of S. rhizophila lipopolysaccharides in the literature available to us. The aim of the work was to clarify the phylogenetic position of the 6p5m bacterial strain isolated from the Antarctic region by constructing dendrograms, to study some of its phenotypic properties, to isolate lipopolysaccharides and study chemical and component composition, serological, and biological properties. Methods. Identification was carried out via ribosomal phylogeny. A fragment of the 16S rRNA gene of bacterial strain 6p5m was amplified by PCR using primers 8F and 1492R. The phylogenetic position of the strains was determined by construction of dendrograms, which show the position of the investigated strains among closely related and typical species and type species (programs ClustalX 2.1, Mega v. 6.00). LPS was obtained from cells by water-phenol extraction, electrophoresis was carried out in polyacrylamide gel, monosaccharide and fatty acid composition was determined by chromato-mass spectrometry, antigenic activity was studied by immunodiffusion in agar. Results. Phylogenetic analysis of the nucleotide sequence of the 16S rRNA gene revealed a high level of homology (99.8%) of the bacterial strain 6p5m with typical strain S. rhizophila DSM 14405T from the GenBank database, which allows us to classify it. Comparing phylogenetic analysis with phenotypic data, we can assert that strain 6p5m belongs to the S. rhizophila species. When studying the biopolymer composition of LPS by specific reactions to each component, it was found that the carbohydrate content was relatively low and amounted to 27.42% and 13.34% for LPS1 and LPS2, respectively. The content of characteristic LPS components: heptose and 2-keto-3-deoxyoctonic (KDO) acid was 1.78% and 0.034%, as well as 5.38% and 0.09%, respectively, for LPS1 and LPS2. Analysis of the monosaccharide composition of LPS preparations showed that fucose (72.57%) is present as a dominant monosaccharide in LPS1 and rhamnose (65.04%) in LPS2. The main monosaccharides of the extracellular component were galactose (39.92%) and rhamnose (24.36%). Analysis of the lipid part of the studied LPS indicates significant differences between them. In the composition of lipid A LPS1 and LPS2 of S. rhizophila 6p5m, anti-iso-pentadecanoic acid (a-i-C15:0) was predominant (25.58% and 39.24%, respectively), and 3-hydroxydodecanoic acid (3-OH-C12:0) in LPS2 (26.21%), hexadecanoic acid (С16:0) (27.06% and 10.51%, respectively) for LPS1 and LPS2. Cyclic acids were present only in LPS1, while i-C15:0 and 9-C16:1 only in LPS2. Electrophoretic analysis showed heterogeneity characteristic of an LPS molecule, manifested by a plurality of bands in the form of a «ladder» when the gel was stained with silver ions, which is associated with the presence in the composition of LPS preparations of Ospecific polysaccharide structures with different lengths of oligosaccharide chains, which determine their different molecular weight. On the electropherogram of LPS1 and LPS2, bands were not visualized in the upper part of the electrophoretic track, which indicates the absence of S-forms of LPS with long O-specific chains, but a significant predominance of SR-form molecules in the membrane LPS pool. S. rhizophila 6p5m LPS showed significantly higher pyrogenic activity compared to pyrogenal, a pharmaceutical preparation, an active ingredient of which is Shigella typhi LPS. The extracellular polymer did not exhibit pyrogenic activity. In the double immunodiffusion reaction in agar according to Ouchterlony, it was found that the studied LPSs in the homologous system exhibit antigen activity. The antigens present in both LPS1and LPS2 and the extracellular component are identical: the antigen present in each of the wells binds to all antibodies that can interact with the antigen from the other well. It was shown that pre-sowing treatment of seeds with LPS preparations of S. rhizophila 6p5m stimulates the energy of seed germination. LPS1 (+2.82 cm) showed the greatest impact in comparison with the control. A significant stimulatory effect of LPS1, LPS2, and extracellular polymer was observed when studying their effects on mustard seedlings. Conclusions. The taxonomic position of bacterial strain 6r5m of the ecosystems of the polar region (Antarctica) was determined via the phylogenetic analysis taking into account the phenotypic features of the strain. The bacterial strain is represented in the phylum Proteobacteria, class Gammaproteobacteria, family Xanthomonadaceae, genus Stenotrophomonas, species S. rhizophila. From S. rhizophila cells, LPS preparations were obtained, which differed in monosaccharide and fatty acid composition, but showed high pyrogenicity. S. rhizophila represents a promising alternative to S. maltophilia for applications in agricultural biotechnology and biological control due to its ability to both stimulate plant growth and protect roots from biotic and abiotic stresses. The infrared (IR) and vibrational circular dichroism (VCD) spectra of methyl β‐D‐glucopyranose in water were measured. Both implicit and explicit solvation models were utilized to explain the observed spectra. The vast body of existing experimental and theoretical data suggested that about eight explicit water molecules are needed to account for the solvent effects, supported by the current Quantum Cluster Growth (QCG) analysis. Extensive manual and systematic conformational searches of the molecular target and its water clusters were carried out by using a recently developed conformational searching tool, conformer‐rotamer ensemble sampling tool (CREST), and the microsolvation model in the associated QCG code. The Boltzmann averaged IR and VCD spectra of the methyl β‐D‐glucopyranose‐(water)n (n = 8) conformers in the PCM of water provide better agreement with the experimental ones than those with n = 0, 1, and 2. The explicit solvation with eight water molecules was shown to greatly modify the conformational preference of methyl β‐D‐glucopyranose from its monomeric form. Further analyses show that the result is consistent with the existence of long‐lived methyl β‐D‐glucopyranose monohydrates with the additional explicit water effects being accounted for with the quantum mechanical treatment of the other seven close‐by water molecules in the PCM of water. Objectives The incidence of inflammatory bowel disease (IBD) is increasing globally, and excessive added sugar consumption has been identified as one of the contributing factors. In the context of IBD, it is essential to explore functional sweeteners that can improve metabolic health and minimize the risk of IBD-related symptoms. This review article aims to shed light on the effects of natural mono- and di-saccharides as alternative sweeteners, specifically focusing on potential benefits for IBD. Methods A comprehensive literature review was performed using PubMed and Google Scholar databases with articles published after the year 2000. The search terms ‘IBD’, ‘added sugar’, ‘sweeteners’, ‘mono-saccharide’, and ‘di-saccharide’ were combined to retrieve relevant articles. A total of 21 manuscripts, aligning with the objectives of the study, were selected. Papers focusing on artificial or high-intensity sweeteners were excluded to ensure relevant literature selection. Results Multiple studies have emphasized the association between the high consumption of added sugars such as simple sugars and the increased risk of developing IBD. This is suggested to be attributed to the induction of pro-inflammatory cytokine productions and dysbiosis of the gut microbiota. Consequently, there is a growing demand for safe and functional sweeteners, in particular mono- and di-saccharides, that can serve as alternatives for IBD patients. Those functional sweeteners regulate inflammation, oxidative stress, and Intestinal barrier protection, and restore microbiome profiles in various IBD models including cells, animals, and humans. Conclusions Understanding these mechanisms resolves the link between how sugar consumption and IBD, and highlights the beneficial effects of natural alternative sweeteners on IBD when they were administered by itself or as a replacement for simple sugar. Further, exploration of this relationship leads us to recognize the necessity of natural alternative sweeteners in dietary planning. This knowledge could potentially lead to more effective dietary strategies for individuals with IBD.

The simultaneous determination of rhamnose, ribose, arabinose, fucose, galactose, xylose, galacturonic acid, mannitol, glucuronic acid and mannose in whey protein sports nutrition powder polysaccharide hydrolyzate was established. An ultra-high performance liquid chromatography-tandem mass spectrometry method was used for the analysis of 12 sugar compounds. The samples of whey protein sports nutrition powder were extracted by ultrasound, and the protein was removed by the Sevag method until no protein was precipitated, and then the monosaccharide composition was determined by acid hydrolysis. Using 5 mmol/L ammonium acetate+0.02% formic acid aqueous solution as mobile phase A, methanol as mobile phase B, in the electrospray ion source negative ion (electrospray ion source negative ion, ESI−), in the multiple reaction monitoring mode (multiple reaction monitoring, MRM) analysis, the detection was conducted by external standard method for quantification. The results showed that in the mass concentration range of 0.10~6.00 μg/mL, the 12 sugar compounds had good linear relationship with each monosaccharide, the correlation coefficients (r) were all greater than 0.995. The detection limit was 0.0037~0.0326 mg/g. The quantitative limit was 0.0123~0.1087 mg/g. 12 sugar compounds were added to the samples at three levels with concentrations of 40, 100 and 160 mg/g, and the average recovery was 76.12%~99.12%, relative standard deviations (RSDs) 0.12%~10.01% (n=6). This method was used to analyze the samples of whey protein sports nutrition powder, and the results showed that 12 carbohydrate compounds were detected in different degrees in 15 samples, and the content of various monosaccharides in different samples was quite different. The establishment of this method could provide technical support and basic data for the structural composition and activity of whey protein sports nutrition powder polysaccharide.

In order to obtain high-quality Auricularia auricula glycoprotein and clarify its monosaccharide and amino acid composition, the process conditions of ultrasonic assisted alkaline extraction of Auricularia auricula glycoprotein were optimized by single factor experiment and response surface methodology, and the glycoprotein was separated and purified by AKTAgo purifier and DEAE GE Healthcare chromatographic column, and its monosaccharide and amino acid composition were analyzed by high performance liquid chromatography (HPLC). The optimum extraction conditions of Auricularia auricula glycoprotein were: ultrasonic time 90 min, material liquid ratio 1:90 g/L, ultrasonic power 200 W, pH8.4, extraction temperature 45 ℃, and the comprehensive score of glycoprotein extraction was 55.93. After separation and purification, the purity of the glycoprotein was 74.1% and the protein recovery rate was 37.5%. The monosaccharide composition of the glycoprotein was mainly glucose (9.95 mg/g) and mannose (5.24 mg/g), and the amino acids were mainly L-aspartic acid/L-asparagine (1.16 mg/g), L-phenylalanine (1.14 mg/g) and L-glutamic acid/L-glutamine (1.10 mg/g). Therefore, the ultrasonic alkali method can effectively improve the extraction efficiency of Auricularia auricula glycoprotein, and the Auricularia auricula glycoprotein contains acidic heteropolysaccharides and the necessary amino acids are complete, which provides a scientific reference for the in-depth research and application development of Auricularia auricula glycoprotein.

Environmentally-mediated drug resistance in B-cell precursor acute lymphoblastic leukemia (BCP-ALL) significantly contributes to relapse. Stromal cells in the bone marrow environment protect leukemia cells by secretion of chemokines as cues for BCP-ALL migration towards, and adhesion to, stroma. Stromal cells and BCP-ALL cells communicate through stromal galectin-3. Here, we investigated the significance of stromal galectin-3 to BCP-ALL cells. We used CRISPR/Cas9 genome editing to ablate galectin-3 in stromal cells and found that galectin-3 is dispensable for steady-state BCP-ALL proliferation and viability. However, efficient leukemia migration and adhesion to stromal cells are significantly dependent on stromal galectin-3. Importantly, the loss of stromal galectin-3 production sensitized BCP-ALL cells to conventional chemotherapy. We therefore tested novel carbohydrate-based small molecule compounds (Cpd14 and Cpd17) with high specificity for galectin-3. Consistent with results obtained using galectin-3-knockout stromal cells, treatment of stromal-BCP-ALL co-cultures inhibited BCP-ALL migration and adhesion. Moreover, these compounds induced anti-leukemic responses in BCP-ALL cells, including a dose-dependent reduction of viability and proliferation, the induction of apoptosis and, importantly, the inhibition of drug resistance. Collectively, these findings indicate galectin-3 regulates BCP-ALL cell responses to chemotherapy through the interactions between leukemia cells and the stroma, and show that a combination of galectin-3 inhibition with conventional drugs can sensitize the leukemia cells to chemotherapy.

Ulva rigida green macroalgae contain a variety of polysaccharides. A recent study investigated the optimum concentration and yield of polysaccharide extraction from oven-dried U. rigida biomass using a water-soluble polysaccharide extraction method that adhered to safety standards. This study utilised complete factorial experiments to examine the effects of varying factors on polysaccharide extraction. Results showed a positive correlation between increased levels of all factors and higher polysaccharide extraction yield. This study also found that the main factors and their interaction had a significant impact on the extracted polysaccharides from U. rigida. The highest polysaccharide content and yield were 9.5 mg/mL and 189 mg/g, respectively. Water-soluble polysaccharides demonstrated the presence of reducing sugar (8 mg/g), phenolics (0.69 mg/g) and flavonoids (1.42 mg/g) and exhibited antioxidant properties. Results revealed that freeze-dried polysaccharide powders were primarily composed of the monosaccharide rhamnose. Preliminary results on the effect of these powders on probiotics demonstrated that supplementation of polysaccharides from U. rigida promoted viable Lactobacillus rhamnosus ATCC 53103 growth during cultivation. This discovery has the potential to revolutionise the human food industry and promote the development of functional ingredients for novel and future food products, with numerous applications in the nutraceutical and pharmaceutical industries.

Entamoeba histolytica is a parasitic protozoan with roles in pathogenicity of intestinal amoebiasis. E. histolytica trophozoites lack functional mitochondria and their energy production depends mostly on glycolysis. D-Glucose has a pivotal role in this process and trophozoites store this sugar as glycogen in glycogen granules. Rare sugars, which are defined as sugars present in nature in limited amounts, are of interest as natural low-calorie sweeteners for improving physical conditions of humans. One such rare sugar, D-allose, can be absorbed by a sodium-dependent glucose cotransporter as a substitute for D-glucose, and some rare sugars are known to inhibit growth of cancer cells, Caenorhabditis elegans and Tritrichomonas foetus. Based on these observations, we examined the effects of rare sugars on growth of E. histolytica trophozoites, together with those of D-galactose and D-fructose. The results indicate that treatment with D-allose or D-psicose (D-allulose) alone inhibits proliferation of E. histolytica trophozoites, but that these sugars enhance proliferation of trophozoites in the presence of D-glucose or D-galactose. The trophozoites could take up D-glucose and D-galactose, but not D-fructose, D-allose or D-psicose. Cell sizes of the trophozoites also differed depending on the culture medium.

Polysaccharides are the main group of macromolecules in wines. Climate change is a major problem for viticulturists as it leads to the production of unbalanced grapes. This is attributed to a mismatch between the technological maturity and phenolic maturity of grapes, which can negatively impact the production of high quality wines. To mitigate this effect, biostimulants can be applied to grapevines. For the first time in the literature, this work studied the foliar application of methyl jasmonate plus urea (MeJ + Ur) on the vineyard and its effect on the monosaccharide and polysaccharide composition of Tempranillo grapes and wines over two consecutive seasons. To achieve this, the extraction and precipitation of polysaccharides was conducted, and the identification and quantitation of monosaccharides was performed via GC–MS. The effect of MeJ + Ur foliar treatment in both the grapes and wines was season-dependent. The MeJ + Ur treatment had a slight impact on the monosaccharide composition of the grapes and also demonstrated a small effect on the wines. Multifactor and discriminant analysis revealed that the season had a greater influence on the monosaccharide and polysaccharide composition of grapes and wines compared to the influence of MeJ + Ur treatment. Interestingly, the MeJ + Ur-treated wines exhibited a higher sensory evaluation than the control wines in the second vintage. To gain further insights into the effect of MeJ + Ur foliar application on the monosaccharide and polysaccharide composition of grapes and wines, further investigations should be conducted. Acid hydrolysis of concob by wet process phosphoric acid was proposed, and the effects of pretreatment temperature and time on the content of each component in corncob acid hydrolysis solution were systematically discussed. The Saeman model and two-phase model were used for analysis，and the reaction kinetics equations applicable to different products were established. The results showed that with the increase of acid hydrolysis temperature and time, corncob is easier to be degraded，and the organic content in acid hydrolysis solution is increasing，the monomer product was mainly arabinose within 30 min and xylose over 30 min. The inhibitory products, furfural and 5-hydroxy methylfurfural (5-HMF) were degraded by xylose and galactose. The change trend of oligomer and monomer content with temperature and time is different. BackgroundThe stable isotope deuterium dose-to-mother (DTM) technique to estimate nonbreast milk water intake demonstrates that maternal self-report methods of infant feeding overestimate the true prevalence of exclusively breastfeeding practices.ObjectiveWe aimed to determine potential monosaccharide and oligosaccharide markers that distinguish between exclusively breastfed (EBF) versus nonexclusively breastfed (non-EBF) infants utilizing LC-MS-based methods.MethodsData for the analysis were collected as part of a larger, longitudinal study of 192 breastfed Indonesian infants aged 2 mo and followed up at 5 mo. Feces samples were collected from infants aged 2 mo (n = 188) and 5 mo (n = 184). EBF and non-EBF strata at each time point were determined via the DTM technique. The affinity interactions between boronic acids and sugars have been successfully exploited in many fields, such as the sensing of saccharides, selective enrichment of glycoconjugates, and drug delivery. However, despite multiple techniques having been adopted to investigate the reaction of boronate affinity, the pathway of boronate esters formation under aqueous conditions remains controversial. We report a MALDI-MS approach to investigate the interactions between phenylboronic acid and monosaccharides in neutral aqueous solution by using polylevodopa as an innovative substrate instead of conventional matrix. A series of unusual tri-benzeneboronic esters were then revealed. The mass spectrometry data indicate that they bear a dibenzenepyroboronate cyclic ester moiety with seven-membered ring or eight-membered ring. With the aid of theoretical computations, their most likely geometrical structures are elucidated, and these tri-benzeneboronic esters are proposed to be formed via a boroxine binding monosaccharide pathway. This work provides more insight into the mechanism of boronate affinity interaction between boronic acid and sugars and proves the developed MALDI-MS approach is promising for studying interactions between small molecules. The great flexibility of direct laser writing (DLW) arises from the possibility to fabricate precise three‐dimensional structures on very small scales as well as the broad range of applicable materials. However, there is still a vast number of promising materials, which are currently inaccessible requiring the continuous development of novel photoresists. Herein, a new bio‐sourced resist is reported that uses the monomeric unit of chitin, N‐acetyl‐D‐glucosamine, paving the way from existing hydrogel resists based on animal carbohydrates to a new class of non‐hydrogel ones. In addition, it is shown that the combined use of two photoinitiators is advantageous over the use of a single one. In this approach, the first photoinitiator is a good two‐photon absorber at the applied wavelength, while the second photoinitiator exhibits poor two‐photon absorbtion abilities, but is better suited for cross‐linking of the monomer. The first photoinitiator absorbs the light acting as a sensitizer and transfers the energy to the second initiator, which subsequently forms a radical and initializes the polymerization. This sensitization effect enables a new route to utilize reactive photointiators with a small two‐photon absorption cross section for DLW without changing their chemical structure. Algae living inside corals provide sugars for their host by digesting their own cell walls.

Reef-building corals thrive in oligotrophic environments due to their possession of endosymbiotic algae. Confined to the low pH interior of the symbiosome within the cell, the algal symbiont provides the coral host with photosynthetically fixed carbon. However, it remains unknown how carbon is released from the algal symbiont for uptake by the host. Here we show, using cultured symbiotic dinoflagellate, Breviolum sp., that decreases in pH directly accelerates the release of monosaccharides, that is, glucose and galactose, into the ambient environment. Under low pH conditions, the cell surface structures were deformed and genes related to cellulase were significantly upregulated in Breviolum. Importantly, the release of monosaccharides was suppressed by the cellulase inhibitor, glucopyranoside, linking the release of carbon to degradation of the agal cell wall. Our results suggest that the low pH signals the cellulase-mediated release of monosaccharides from the algal cell wall as an environmental response in coral reef ecosystems.

The purpose of the study was to improve the extraction of polysaccharide from the leaves of Cercis chinensis Bunge using ultrasound, and compare the difference between boiling and ultrasound extraction in terms of polysaccharide content, monosaccharide compounds, and evaluate how the factors affected the bioactivity. The best conditions, according to the single factor experiments and the Box-Bohnken design (BBD), were an intensity of ultrasound of 180 W, duration of extraction of 40 min, proportion of water to material of 15:1 (g/g), and a higher polysaccharide yield of 20.02 ± 0.55 (mg/g) than in boiling extraction (16.09 ± 0.82 mg/g). The antioxidative experiment suggested the polysaccharide by ultrasound exhibited higher DPPH, hydroxyl radical scavenging capacities, and reducing power at 1.2–1.4 mg/mL, which was superior to the boiling polysaccharide. Further analysis showed that the ultrasonic purified polysaccharides like Gla, N-Glu, and GluA contained more total sugar and uronic acids than the boiling method did. This may indicate that the ultrasonic isolation of the polysaccharides increase the antioxidant activity of the polysaccharides.

Aluminum (Al) toxicity is the primary factor limiting agricultural productivity in acid soils. The cell wall is mainly composed of saccharides and the first barrier for aluminum (Al) to enter plant root cells, but it is unknown whether and how root saccharide components are involved in regulating the Al tolerance of Lespedeza that is well adapted to acid soils. Here, we used Al-tolerant Lespedeza bicolor and Al-sensitive Lespedeza cuneata to examine the association of root saccharide components with Lespedeza Al tolerance through analyzing the saccharide changes of roots exposed to Al toxicity. Al-sensitive Lespedeza accumulated more Al and pectin but less hemicellulose in the root cell walls than Al-tolerant Lespedeza. Al treatment decreased the amounts of total sugar secreted from and within the roots of only Al-tolerant Lespedeza. Al treatment decreased the content of root monosaccharides including glucose and mannose in both Lespedeza species, but increased the xylose contents in only Al-sensitive Lespedeza. Taken together, less cell wall pectin rather than hemicellulose is responsible for less root Al accumulation, and Al-decreased root saccharide contents may enhance root organic-acid secretion to chelate toxic Al, both of which contribute to Lespedeza Al tolerance. Mayonnaise is an oil-in-water emulsion containing 70–80% finely dispersed droplets of oil in a continuous phase of water. Since mayonnaise has a sour and acidic taste, its sugar profile is barely noticed and thus often disregarded. However, today, there are various variants of mayonnaise available on the market; hence, it is crucial to understand their mono- and disaccharide profile, in order to determine the precise total sugar composition. The traditional methods of sugar analysis available, such as titration, can only quantify sucrose and are unable to differentiate between mono- and disaccharides. The aim of this study was to develop and validate a method for the quantification of total sugars, including fructose, glucose, sucrose, and lactose, in eggless mayonnaise, using a high performance liquid chromatography refractive index detector (HPLC-RID). Sugars were separated on an amino column with an oven temperature of 35 °C, using an isocratic solvent system consisting of a 75:25 v/v mixture of acetonitrile and HPLC water, at a 0.9 mL/min flow rate with RID. Method validation was performed for the linearity, specificity, precision, accuracy, LOD, LOQ, and robustness. A linearity for total sugars, with a regression coefficient of 0.9998, was obtained within the range of 0.05024 to 10.048 mg/mL. The relative standard deviation was less than 2.0% for the intra-day and inter-day precision. The accuracy was found to be 96.78–108.88% using a three-level recovery method. The LOD and LOQ were also found to be suitable. The samples used in this study contained 0.24–10.32% total sugars. The sucrose value obtained matched the label claim of the products and no significant differences were observed between results in a paired sample t-test. This showed the applicability of the proposed method for analyzing the sugar profile in a finished product. Routine analysis of total sugars in eggless mayonnaise and similar finished products can thus be performed using this technique, which was found to be simple, rapid, and reproducible. The mixed enzymes-assisted mechanical bio-extraction method was first used to extract polysaccharides from Dendrobium officinale. Different parameters including the ratio of enzyme, the amount of enzyme, the grinding time, the extraction time and the solid/liquid ratio were investigated by single factor experiments and multifactorial experiments. Through the response surface methodology the optimal extraction conditions were obtained with the ratio of cellulase to pectinase was 2: 1 and total amount of enzyme was 0.23 mg, the grinding time of 11.48 min, the extraction time of 5.99 min. The obtained polysaccharide extracts were hydrolyzed and derivatized and then injected into high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (HPLC-Q-TOF/MS) for monosaccharide composition analysis. After optimization of the chromatographic conditions (including mobile phase and column), twelve monosaccharides were successfully determined within 20 min. The proposed method provided satisfactory linearity with the correlation coefficients higher than 0.99, suitable recoveries (81.46–114.92 %), acceptable reproducibility ranging from 0.06 % to 4.77 %, low limits of detection (0.70–45.45 ng/mL). Compared with other methods, this method makes the extraction efficiency much higher and has the advantages of simple operation, environmental friendliness and mild extraction conditions. The excessive consumption of dietary sugar induces changes in gut microbiota, which is associated with obesity and metabolic dysregulation. This study investigated the effects of monosaccharide and fructooligosaccharide (FOS) intake on metabolic function and intestinal environment in germ-free (GF) mice lacking gut microbiota. GF mice were provided with a chow diet and administered a water solution containing 15 % glucose, fructose, or FOS for 4 weeks. Compared with FOS, glucose, and fructose induced increased hepatic lipid accumulation, increased adipocyte size in white adipose tissue, and upregulated hepatic lipogenic gene expression. FOS exhibited notably higher activation of hepatic AMP-activated protein kinase compared with those consuming glucose or fructose. Moreover, the number of goblet cells in the intestinal mucosa increased significantly with FOS consumption. Collectively, these findings indicate that while monosaccharides caused metabolic disorders in GF mice, FOS alleviated these disorders and increased the number of goblet cells in the intestinal mucosa. These results provide evidence for the occurrence of these effects independently of the gut microbiota. Pectic polysaccharides are considered the highly complex natural plant polysaccharides which plays a vital role in plant tissue structure and human health. Detailed characterization of the monosaccharide composition can provide insights into the pectic polysaccharide structure. Nevertheless, when analyzing the monosaccharides of pectic polysaccharide, it is crucial to address the issue of incomplete hydrolysis that can occur due to the formation of acid-induced precipitates. Based on above, the main purpose of this article is to provide an optimized method for monosaccharide analysis of pectic polysaccharides through acid hydrolysis optimization using high-performance anion exchange chromatography (HPAEC) The results indicate that reducing the sample concentration to 0.5 mg/mL effectively reduces the acid gelling phenomenon and promotes the complete hydrolysis of pectin polysaccharides. The optimized parameters for acid hydrolysis involve 110 °C for 6 h in 2 M TFA. Furthermore, the consistency of this method is assessed, along with its ability to analyze pectin polysaccharides from various fruits. During mitosis, phosphorylation and dephosphorylation of lamins triggers the nuclear envelope disassembly/assembly. However, it hasn't been known whether lamin proteins undergo any modification other than phosphorylation during the cell cycle. Glycosylation of lamin proteins is one of the less studied post-translational modification. Glycosylation and phosphorylation compete for the same positions and interplay between two modifications generate a post-translational code in the cell. Based on this, we hypothesized that glycosylation of lamin A/C protein may be important in the regulation of the structural organization of the nuclear lamina during interphase and mitosis. We analysed the glycan units of lamin A/C protein in lung carcinoma cells synchronized at G2/M and S phases via CapLC-ESI-MS/MS. Besides, the outermost glycan units were determined using lectin blotting and gold-conjugated antibody and lectin staining. TEM studies also allowed us to observe the localization of glycosylated lamin A/C protein. With this study, we determined that lamin A/C protein shows O-glycosylation at G2/M and S phases of the cell cycle. In addition to O-GlcNAcylation and O-GalNAcylation, lamin A/C is found to be contain Gal, Fuc, Man, and Sia sugars at G2/M and S phases for the first time. Quorum sensing (QS) is an important microbial signaling system that controls the expression of many virulence genes. Combination treatment with QS inhibitors and antibiotics may be the new treatment option for infectious diseases in the future. Therefore, there is an increasing need to discover anti-QS compounds to prevent antimicrobial resistance. We isolated methyl-β-D-glucopyranoside from Scabiosa comosa and found that it exerts an anti-QS activity against Chromobacterium violaceum CV026 without exhibiting any antimicrobial activity.
