Meadowsweet (Filipendula ulmaria) is an essential oil-bearing, pharmacologically valuable medicinal plant growing wild in floodlands that are either not usually used for agriculture or have low economic value. The aim of this work was to understand the relationship between the yield of F. ulmaria inflorescences and the qualitative and quantitative composition of the essential oil during flowering stages in different habitats. Five different habitats of F. ulmaria were investigated for evaluation of inflorescence characteristics (length and weight) and the quantitative and qualitative composition of essential oils in early and late blooming stages (growing stage BBCH 62–63 and BBCH 65–67/72–73, respectively). The results showed significant (p < 0.05) positive correlations between the length and weight of inflorescences both in early and in late blooming stages (r = 0.73 and r = 0.72, respectively). The early blooming stage was observed to have 1.2–2 times greater quantity of volatile oils in comparison with the late blooming stage. Salicylaldehyde and methylsalicylate were the main salicylates in both blooming stages. Spearman's correlation showed statistically significant relationship between percentages of salicylaldehyde and methylsalicylate (r = −0.94, p < 0.05). An increase in salicylaldehyde was accompanied by a decrease in methylsalicylate in essential oils. Statistically significant relations between the investigated parameters of plant and the parameters of habitat were not detected. The dried raw material yield of F. ulmaria in 1 ha in the late blooming stage was 18–56% higher than in the early blooming stage; however, the yield of essential oil in the early flowering stage was 1.5–1.6 times higher than in the late flowering stage. The obtained research data suggest that the low economic value of floodlands—the habitats of F. ulmaria—can be increased by using these areas as natural industrial plots. [ABSTRACT FROM AUTHOR]

Many over-the-counter products contain methylsalicylate. Topical salicylates rarely produce systemic toxicity when used appropriately; however, methylsaliclyate can be absorbed through intact skin. Scrotal skin can have up to 40-fold greater absorption compared to other dermal regions. We report a unique case of salicylate poisoning resulting from the use of a methylsalicylate-containing rubefacient to facilitate masturbation in a male teenager. Saliclyate toxicity has not previously been reported from the genital exposure to methylsaliclyate.

Inducing resistance in plants by application of elicitors of defense reactions is an attractive plant protection strategy, especially for grapevine (Vitis vinifera) which is susceptible to severe fungal diseases. Though induced resistance (IR) can be successful in controlled conditions, under outdoor conditions IR is in most cases not effective enough for practical disease control. Progress in the application of IR requires a better understanding of grapevine defense mechanisms and the ability to monitor defense markers in order to identify factors (physiological, environmental…) that can impact IR in the vineyard.Volatile organic compounds (VOCs) are well-known plant defenses compounds that have only received little or no attention in the case of grape-pathogen interactions to date. This prompted us to investigate whether an elicitor, the sulfated laminarin (PS3), actually induces the production of VOCs in grapevine. Online analysis (PTR-QMS) of VOC emissions in dynamic cuvettes and passive sampling in gas tight bags with solid phase micro extraction (SPME-GC-MS) under greenhouse conditions showed that PS3 elicited emission of VOCs. Some of them (as (E,E)-α-farnesene) might be good candidates as biomarkers of elicitor-IR whereas methyl salicylate appears to be rather a biomarker of downy mildew infection. A negative correlation between VOC emission and disease severity suggests a positive role of VOCs in grape defense against diseases.

Floral scent composed of low molecular weight volatile organic compounds. The sweet fragrance of any evening blooming flower is dominated by benzenoid and terpenoid volatile compounds. Floral scent of Jasminum sambac (Oleaceae) includes three major benzenoid esters – benzylacetate, methylbenzoate, and methylsalicylate and three major terpene compounds viz . ( E )- β -ocimene, linalool and α -farnesene. We analyzed concentrations and emission rates of benzenoids and terpenoids during the developmental stages of J. sambac flower. In addition to spatial emission from different floral parts, we studied the time-course mRNA accumulations of phenylalanine ammonia-lyase (PAL) and the two representative genes of terpenoid pathway, namely 1-deoxy- d -xylulose 5-phosphate reductoisomerase (DXR) and terpene synthase (TPS). Further, in vitro activities of several enzymes of phenylpropanoid/benzenoid pathway viz., PAL and acetyl-coenzyme A: benzylalcohol acetyltransferase (BEAT), S -adenosyl- l -methionine: benzoic acid carboxyl methyl transferase (BAMT) and S -adenosyl- l -methionine: salicylic acid carboxyl methyltransferase (SAMT) were studied. All the above enzyme activities along with the in vitro activities of DXR and TPS were found to follow a certain rhythm as observed in the emission of different benzenoid and terpenoid compounds. Linalool emission peaked after petal opening and coincided with maximal expression of JsTPS gene as evidenced from RT-PCR analyses (semi-quantitative). The maximum transcript accumulation of this gene was observed in flower petals, indicating that the petals of J. sambac flower play an important role as a major contributor of volatile precursors. The transcripts accumulation of JsDXR and JsTPS in different developmental stages and in different floral part showed that emissions of terpenoid volatiles in J. sambac flower are partially regulated at transcription levels. Background: Salicylic acid and its derivatives are widely used drugs with potential toxicity. The main areas of salicylate derivatives determination are biological liquids and pharmaceuticals analysis. Results: Silica-titania xerogel has been used for solid phase spectrophotometric determination of various salicylate derivatives (salicylate, salicylamide, methylsalicylate). The reaction conditions influence on the interaction of salicylate derivatives with silica-titania xerogels has been investigated; the characteristics of titanium(IV)-salicylate derivatives complexes in solid phase have been described. The simple solid phase spectrophotometric procedures are based on the formation of xerogel incorporated titanium(IV) colored complexes with salicylate derivatives. A linear response has been observed in the following concentration ranges 0.1-5, 0.5-10 and 0.05-4.7 mM for salicylate, salicylamide, and methylsalicylate, respectively. The proposed procedures have been applied to the analysis of human urine, synthetic serum, and pharmaceuticals. Conclusions: The simple solid phase spectrophotometric procedures of salicylate derivatives determination based on the new sensor materials have been proposed for biological liquids and pharmaceuticals analysis. Systemic acquired resistance (SAR) is an inducible defense mechanism in plants that confers enhanced resistance against a variety of pathogens. SAR is activated in the uninfected systemic (distal) organs in response to a prior (primary) infection elsewhere in the plant. SAR is associated with the activation of salicylic acid (SA) signaling and the priming of defense responses for robust activation in response to subsequent infections. The activation of SAR requires communication by the primary infected tissues with the distal organs. The vasculature functions as a conduit for the translocation of factors that facilitate long-distance intra-plant communication. In recent years, several metabolites putatively involved in long-distance signaling have been identified. These include the methyl ester of SA (MeSA), the abietane diterpenoid dehydroabietinal (DA), the dicarboxylic acid azelaic acid (AzA), and a glycerol-3-phosphate (G3P)-dependent factor. Long-distance signaling by some of these metabolites also requires the lipid-transfer protein DIR1 (DEFECTIVE IN INDUCED RESISTANCE 1). The relative contribution of these factors in long-distance signaling is likely influenced by environmental conditions, for example light. In the systemic leaves, the AGD2-LIKE DEFENSE RESPONSE PROTEIN1 (ALD1)-dependent production of the lysine catabolite pipecolic acid (Pip), FLAVIN-DEPENDENT MONOOXYGENASE1 (FMO1) signaling, as well as SA synthesis and downstream signaling are required for the activation of SAR. This review summarizes the involvement and interaction between long-distance SAR signals and details the recently discovered role of Pip in defense amplification and priming that allows plants to acquire immunity at the systemic level. Commercial formulations of non-steroidal anti-inflammatory drugs (NSAIDs) are developed for human use but the extent to which they will pass through equine skin is unknown. Skin was harvested from five Thoroughbred geldings from the thorax, groin and leg (dorsal metacarpal) regions and frozen (-20 °C} until required. Two grams of methylsalicylate (MeSa) gel was applied to defrosted full-thickness samples in diffusion cells and the penetration of MeSa and its active metabolite, salicylate (Sa), through skin samples were measured over 24 h. Significantly higher (P ≤ 0.02) total salicylate (AUC; MeSa + Sa) penetrated through skin from the leg region (5491.3 h mg/L) compared to thorax (3710.7 h mg/L) and groin (3571.5 h mg/L) In addition, there was a significantly higher (P ≤ 0.01) rate of penetration of total Sa through leg skin in the first 6 h after application. It was concluded that the commercial formulation of MeSa would achieve therapeutic levels of total salicylate beneath sites of topical application, with a faster and more pronounced response through the leg region, compared to the upper body. Some silyl ethers of acetaminophen and methylsalicylate of general formula ArOSiRR'R'' ArOH = parahydroxyacetanilide or 2-hydroxybenzoic acid methyl ester; SiRR'R'' = trimethylsilyl, triethylsilyl, dimethyltert(Bu)silyl, triphenylsilyl, dimethylsilyl, tris(trimethylsilyl)silyl (sisyl) or dimethylvinylsilyl have been prepared and fully characterized. The compounds were prepared from the combination of either acetaminophen or methylsalicylate with the silyl chloride of these groups. Two methods were employed. In the first Et3N was used in THF to remove HCl in coupling the silyl group to the organic substrate. In the other method imidazole as a base in dimethylformamide (DMF) were used. The effect of the silyl groups in providing stability to the compounds preventing or enhancing hydrolysis of the silicon-containing group was determined. Sequential injection chromatography (SIC) system was created by incorporating a monolithic chromatography column into a standard sequential injection analysis (SIA) manifold. The system was successfully used for separation of salicylic acid (SA) and its ester methylsalicylate (MS) and for determination of these compounds in a topical pharmaceutical preparation. The separation was performed on Chromolith® SpeedROD RP-18e, 50–4.6 mm column (Merck, Germany) placed between the selection valve and the detection cell with acetonitrile–water (35/60 (v/v)), pH 2.5 as the mobile phase. The eluted compounds were detected by diode array Ocean Optics spectrophotometer at 240 nm. Propylparaben (PP) was chosen as a suitable internal standard for quantification of the desired substances. The relation between the absorbance signal and concentration was linear in the range 0.17–43.2 μg ml−1 for SA, 0.24–60.2 μg ml−1 for MS and 0.24–58.8 μg ml−1 for PP. The detection limits were 0.04 μg ml−1 (SA) and 0.15 μg ml−1 (MS), respectively. The recoveries of the determined compounds in the pharmaceutical paste were found above 96%. The aim of the present work was to verify the possibility of detecting various tissue antigens using standard immunocytochemical methods in paraffin sections of specimens stored in methylsalicylate for two years and longer before embedding in paraffin. Using the immunocytochemical reaction for the intermediate filament proteins nestin, vimentin, and GFAP, the neuron marker NeuN, neuron-specific enolase, and doublecortin as examples, these experiments showed that prolonged storage of dehydrated specimens in methylsalicylate does not prevent detection of antigens using immunocytochemical reactions. Biotic stresses on forest trees are caused by various pest insects and plant pathogens. Attack by these parasites is known to induce the emissions of various biogenic volatile organic compounds (BVOCs), and the profile of these emissions often differs between infested and healthy plants. This difference in emission profile can be used for the non-destructive early-stage diagnosis of the stressor organism. We studied how phloem feeding by a large pine aphid (Cinara pinea Mordvilko) on the branch bark of Scots pine (Pinus sylvestris L.) affects BVOC emissions compared to those of healthy plants in two experiments. We found that in aphid-infested plants, methyl salicylate (MeSA) emissions significantly increased, and the emission rates were dependent on aphid density on the studied branch. Aphid infestation did not significantly affect total monoterpene emission, while the emissions of total sesquiterpenes were substantially higher in aphid-infested saplings than in uninfested plants. Sesquiterpene (E, E)-α-farnesene was emitted at increased rates in both experiments, and the aphid alarm pheromone sesquiterpene (E)-β-farnesene, only in the experiment with higher aphid pressure. We conclude that the rapid increase in MeSA emissions is the most reliable indicator of aphid infestation in pine trees together with (E, E)-α-farnesene. [ABSTRACT FROM AUTHOR]
