Abstract

Antimicrobial peptides in skin secretions of toads is a promising methods to combat a wide spectrum of bacteria. Histone H2A is a type of DNA-binding protein that acts as a precursor for several antimicrobial peptides. In toads (family Bufonidae) buforin I and buforin II are examples of antimicrobial peptides that derived from histone H2A. This study investigated the genetic diversity and phylogenetic analysis and in silico prediction of antimicrobial peptides derived from histone H2A of Duttaphrynus melanostictus and Phyrinoidis asper, which were collected from Bogor Agricultural University’s campus area. A new set of primers (Buf_fwd and Buf_rev) were designed by using PrimerBLAST, to amplify 393 nucleotides of the histone H2A gene that codes 131 amino acids. Haplotype diversity of both species are very low. Phylogenetic analysis shows the sample D. melanostictus and P. asper are separated to each other in two different clades. Several short predicted peptides from histone H2A show a potential as an antimicrobial peptides based on in silico prediction. Psychochemical characteristics and 3D structure of potent antimicrobial peptides are described.

Abstract

Aggregatibacter actinomycetemcomitant (Aa) is gram-negative bacteria composed of lipopolysaccharides (LPS) in its outer membrane. This bacteria is normally found in dental plaque and periodontal pockets causing parotid gland damage when it is present in excess amounts. LPS is an endotoxin and a complex glycolipid composed of polysaccharides (hydrophilic) and lipid A (hydrophobic) which can induces Reactive Oxygen Species (ROS), and increase the levels of malonaldialdehyde (MDA) and tissue damage. The aims of this research are to find the expression of iNOS parotid gland cell and to determine levels of MDA using thiobarbituric acid (TBA) spectrophotometric test at 533 nm. The results showed a significantly difference (PAa rats (6.307 ± 0.0473 ppm). iNOS expression is confirmed by the brown spots formed on the parotid gland, affording the enhancements of iNOS expression in Aa rats and control rats of 2.292 ± 0.038 and 0.166 ± 0.003, respectively (P Keywords : Aggregatibacter actinomycetemcomitans, Reactive Oxygen Species, malondialdehyde, iNOS

3. Characterization of Polyclonal Antibody Induced by Autoantibody TPO (Thyroidperoxidase) From Autoimmune Thyroid Disease (AITD) Serum with ELISA and Western Blotting

Abstract

Autoantibody TPO is a potential marker for early detection of autoimmune thyroid disease (AITD). Autoantibody TPO has a specifity and a sensitivity ranging from 82% to100% in comparison to other AITD serology markers. Concentration of autoantibody TPO in sera had a positive correlation with activities of chronic AITD. This research have been conducted to investigate the characteristic of polyclonal antibody TPO induced by autoantibody TPO from serum of AITD patients. The autoantibody TPO was isolated from serum of AITD patients and its band proteins was confirmed using western blotting, acquiring molecular weight of 51.81 kDa. The protein was induced to a rabbit (Oryctolagus cuniculus) and recognized as an antigen. Serum of rabbit produced polyclonal antibody which was characterized using ELISA and Western blotting. The result showed that autoantibody TPO isolated from serum of AITD patients was immunogenic, which induced the polyclonal antibody anti-TPO (anti-anti-TPO). It was found that the highest polyclonal antibody was obtained on 3rd serum bleeding with the absorbance of 0.3225 and a molecular weight of 51.81 kDa, which is able to specifically recognize autoantibody TPO. Abstract

Anionic and neutral fullerene derivatives were dissolved in water by β-(1,3-1,6)-D-glucan (β-1,3-glucan) as a solubilizing agent. In the water-solubilized complexes, the concentrations of fullerene derivatives were ca. 0.30 mM and the average particle sizes were ca. 90 nm. The β-1,3-glucan complexed fullerene derivative with a carboxylic acid was found to have higher photodynamic activity toward macrophages under visible-light irradiation (λ > 610 nm) when compared with that of other β-1,3-glucan-complexed fullerene derivatives. This result suggests that carboxylic acid moieties in the complex enhance the binding affinity with β-1,3-glucan-receptors on the surface of macrophages when β-1,3-glucan is recognized.
