Both Maqui and Calafate possess high antioxidant activity, which grants them medicinal power and makes them an excellent alternative for improving health. The aim of this study is to create a functional food with therapeutic properties capable of counteracting oxidative stress and thereby contributing to improving people's health. To achieve this, a muffin with inulin, Maqui, and Calafate has been developed. The results show that the incorporation of inulin alone increases the fiber content and antioxidant capacity of the muffins; however, Maqui and Calafate contribute significantly more. Furthermore, contents of phenolic compounds are elevated, and an increase in the folic acid content is observed in the samples compared to the control. We can conclude that producing products with inulin, Maqui, and Calafate can be used to enhance the nutritional value and increase the antioxidant activity of bakery products, providing nutrients while also delivering unique characteristics in color, aroma, and flavor, making them appealing to consumers. Metabolic syndrome (MetS) is a complex disease that includes metabolic and physiological alterations in various organs such as the heart, pancreas, liver, and brain. Reports indicate that blackberry consumption, such as maqui berry, has a beneficial effect on chronic diseases such as cardiovascular disease, obesity, and diabetes. In the present study, in vivo and in silico studies have been performed to evaluate the molecular mechanisms implied to improve the metabolic parameters of MetS. Fourteen-day administration of maqui berry reduces weight gain, blood fasting glucose, total blood cholesterol, triacylglycerides, insulin resistance, and blood pressure impairment in the diet-induced MetS model in male and female rats. In addition, in the serum of male and female rats, the administration of maqui berry (MB) improved the concentration of MDA, the activity of SOD, and the formation of carbonyls in the group subjected to the diet-induced MetS model. In silico studies revealed that delphinidin and its glycosylated derivatives could be ligands of some metabolic targets such as α-glucosidase, PPAR-α, and PPAR-γ, which are related to MetS parameters. The experimental results obtained in the study suggest that even at low systemic concentrations, anthocyanin glycosides and aglycones could simultaneously act on different targets related to MetS. Smoking is a major risk factor for delayed fracture healing, affecting several aspects of early fracture repair, including inflammation, osteogenesis, and angiogenesis. Panax ginseng (GE) and maqui berry extract (MBE) were shown in our previous studies to reduce smoke-induced cellular damage in late bone-healing in vitro models. We aimed here to analyze their effects on the early fracture repair of smokers in a 3D co-culture model of fracture hematomas and endothelial cells. Both extracts did not alter the cellular viability at concentrations of up to 100 µg/mL. In early fracture repair in vitro, they were unable to reduce smoking-induced inflammation and induce osteo- or chondrogenicity. Regarding angiogenesis, smoking-induced stress in HUVECs could not be counteracted by both extracts. Furthermore, smoking-impaired tube formation was not restored by GE but was harmed by MBE. However, GE promoted angiogenesis initiation under smoking conditions via the Angpt/Tie2 axis. To summarize, cigarette smoking strikingly affected early fracture healing processes in vitro, but herbal extracts at the applied doses had only a limited effect. Since both extracts were shown before to be very effective in later stages of fracture healing, our data suggest that their early use immediately after fracture does not appear to negatively impact later beneficial effects. Methods: Twenty patients were randomly assigned to a MBE or a placebo group (PLC). DED parameters including Schirmer's test 1 (ST1), tear film break-up time (TBUT), ocular surface disease index (OSDI), and corneal staining were assessed before treatment and 2 months post-treatment. No significant change in TBUT and corneal staining was observed between the study groups. Levels of proinflammatory factors such as IL-1β, IL-6, IL-17A, TNFα, and MMP9 were observed to be significantly reduced, along with a significant increase in IL-10 levels following treatment in the MBE group compared with the PLC group. Conclusion: Consumption of MBE resulted in the resolution of DED signs and symptoms, along with a reduction in ocular surface inflammation.

In Chilean traditional medicine, Aristotelia chilensis (Mol) Stuntz is commonly utilized for a variety of conditions (particularly digestive problems). We investigated the effects of an aqueous extract from the fruit pulp of A. chilensis (Maqui berry: M2) on endoplasmic reticulum stress and mitochondrial pathway-induced early apoptosis in Ishikawa cells. Using the MTT assay, the aqueous extract was found to have an inhibitory impact on human endometrial cancer Ishikawa cells, with an IC 50 of 5.36 μg/mL. The changes in Ishikawa cells under the effect of M2 were detected by flow cytometry with cycle arrest in S phase, reduced mitochondrial membrane potential, ROS and calcium ion aggregation. Western blotting and RT-qPCR were used to identify the intracellular mRNA and protein alterations. The presence of M2 resulted in the up-regulation of mitochondrial pathway-related proteins Bad, Bax, and Cyto C, down-regulation of Bcl-2, and up-regulation of endoplasmic reticulum stress-related proteins IRE1α, p -eif2α, ATF4, and CHOP. Cigarette smoking-induced oxidative stress has harmful effects on bone metabolism. Maqui berry extract (MBE) and ginseng extract (GE) are two naturally occurring antioxidants that have been shown to reduce oxidative stress. By using an osteoblast and osteoclast three-dimensional co-culture system, we investigated the effects of MBE and GE on bone cells exposed to cigarette smoke extract (CSE). The cell viability and function of the co-culture system were measured on day 14. Markers of bone cell differentiation and oxidative stress were evaluated at gene and protein levels on day 7. The results showed that exposure to CSE induced osteoporotic-like alterations in the co-culture system, while 1.5 µg/mL MBE and 50 µg/mL GE improved CSE-impaired osteoblast function and decreased CSE-induced osteoclast function. The molecular mechanism of MBE and GE in preventing CSE-induced bone cell damage is linked with the inhibition of the NF-κB signaling pathway and the activation of the Nrf2 signaling pathway. Therefore, MBE and GE can reduce CSE-induced detrimental effects on bone cells and, thus, prevent smoking-induced alterations in bone cell homeostasis. Aristotelia chilensis is a plant whose fruit is considered a powerful natural antioxidant. During the last years, some investigations of the fruit have been carried out, finding antioxidant properties in the juice or the phenolic fraction. The antioxidant properties of the plant are useful in the inhibition of enzymes related to diabetes such as pancreatic aldose reductase and alpha-amylase. Because many synthetic drugs used today have limitations and potentially harmful side effects, the use of naturally occurring compounds, such as flavonoids, is clinically attractive. In this study, the characterization of aqueous extracts of fruits and in vitro plants of A. chilensis was carried out based on their content of anthocyanins and total phenols, the antioxidant capacity by the antiradical activity 2,2-diphenyl-1-picrilhydrazil (DPPH), and the profile of anthocyanins and other phenolic compounds by liquid chromatography coupled to mass spectrometry (LC-MS/MS). Subsequently, the effect of these extracts on the inhibition of bovine aldose reductase and pancreatic alpha-amylase enzymes was determined. According to our results, extracts of fruits and in vitro plants of A. chilensis achieved inhibition of the bovine aldose reductase enzyme of 85.54 ± 1.86% and 75.67 ± 1.21%, respectively. Likewise, the percentage of inhibition of the pancreatic alpha-amylase enzyme for fruit extracts was 29.64 ± 0.63%, while for in vitro plant extracts it was 47.66 ± 0.66%. The antioxidant and enzymatic inhibition activity of the extracts were related to the content of anthocyanins, such as delphinidin and cyanidin glycosides as well as the phenols derived from quercetin, myricetin, and kaempferol. The results obtained allow us to suggest that the in vitro culture of plants of A. chilensis represents a viable biotechnological alternative to obtain phenolic compounds for the inhibition of aldose reductase and pancreatic alpha-amylase enzymes. Pasta, a staple food worldwide consumed, was fortified with maqui (Aristotelia chilensis) berry powder (MBP) and the effect of MBP inclusion was evaluated concerning technological, nutritional, and sensory properties. Fresh pasta samples were formulated by replacing 0, 7.5, and 15 g 100 g−1 of durum wheat semolina with MBP. The inclusion of MBP did not affect the moisture content, but the water activity decreased in the fortified samples, while pH values decreased with increasing MBP levels in the recipe. The pasta fully cooking time and the swelling index were reduced, while the cooking loss and the firmness increased with increasing MBP levels. In addition, MBP increased the total dietary fiber, ash, and phenol contents, along with the in vitro antioxidant activities. The starch hydrolysis index and the predicted glycemic index of cooked fresh pasta decreased along with the increase of MBP addition. The MPB addition to fresh pasta could represent a valuable strategy for increasing its nutritional value, maintaining pasta's technological properties without affecting the sensory acceptability. Introduction: Prolonged ozone exposure can produce a state of oxidative stress, which in turn causes alterations in the dynamics of the brain and affects memory and learning. Moreover, different investigations have shown that high flavonoid content berries show a great antioxidant activity. The relationship between the protective effect of the maqui berry extract and its antioxidant properties in the brain has not been studied in depth. Objectives: The present study evaluated whether the protection exerted by the aqueous extract of maqui berry in brain regions associated with cognitive performance is due to its antioxidant capacity. Methods: Sprague Dawley rats were exposed to 0.25 ppm ozone and administered with maqui berry extracts. At the end of the treatments, spatial learning and short- and long-term memory were evaluated, as well as oxidative stress markers. Results: The administration of 50 and 100 mg/kg of the aqueous extract of maqui berry was effective in preventing the cognitive deficit caused by chronic exposure to ozone. The antioxidant effect of the administration of maqui berry was analyzed in the prefrontal cortex, hippocampus, and amygdala. Oxidative stress markers levels decreased and the enzymatic activity of superoxide dismutase diminished in animals exposed to ozone treated with the 50 mg/kg dose of maqui berry. Discussion: These results show a relationship between protection at the cognitive level and a decrease in oxidative stress markers, which suggests that the prevention of cognitive damage is due to the antioxidant activity of the maqui berry. Maqui (Aristotelia chilensis [Molina] Stuntz) is a dioecious small tree native to Chile and southwestern Argentina. This species has gained attention due to its high polyphenol content and anti-oxidant capacity. Nevertheless, genetics studies and information about A. chilensis population genetics are scarce and even contradictory. In fact, the available species-specific simple sequence repeat (SSR) markers are not informative at all, and so we decided to identify and characterize new ones able to trace individual genotypes, a basic tool intended for different genetic studies. We identified and characterized 15 new polymorphic SSR markers for A. chilensis. These markers were evaluated in three populations distributed along 1000 km of Central Chile, exhibiting up to 10 alleles per locus and a combined expected heterozygosity of 0.858. Markers were also informative in two related species, Aristotelia peduncularis (Labill.) Hook. f. and Crinodendron patagua Molina (Elaeocarpaceae), with 13 and six SSRs showing clear amplification patterns, respectively. This new set of SSR markers are highly polymorphic and informative, being the first ones available for the effective fingerprinting of maqui genotypes. A proof of concept of that was the differentiation of six maqui accessions that are under domestication for productive purposes, based on a subset of the polymorphic SSR markers. The aim of the present work was the development of a nutraceutical product from maqui fruits rich in antioxidants. Based on an aqueous fruit extract with high levels of polyphenols and anthocyanins, maqui powder was obtained by spray drying using SiO2 as drying adjuvant. Using a factorial statistical design, optimal operating conditions were found to obtain powders with high content of antioxidants and physical properties suitable for direct compression. The atomization flowrate was the factor that most affected the studied responses. At low atomization flowrates a powder of intense purple color, good flowability (Carr index = 13.4 ± 2.5%; angle of repose = 22.3 ± 1.6°), appropriate physical stability (Tg = 55.6 °C; hygroscopicity = 16.5 ± 0.4 g/100 g), high anthocyanin content (1573 ± 68 mg cyanidin-3-glucoside/100 g), and high polyphenol content (4365 ± 57 mg gallic acid equivalent/100 g) were obtained. The liver is one of the first organs affected by accumulated ectopic lipids. Increased de novo lipogenesis and excessive triglyceride accumulation in the liver are hallmarks of nonalcoholic fatty liver disease (NAFLD) and are strongly associated with obesity, insulin resistance, and type 2 diabetes. Maqui dietary supplemented diet-induced obese mice showed better insulin response and decreased weight gain. We previously described that these positive effects of maqui are partially due to an induction of a brown-like phenotype in subcutaneous white adipose tissue that correlated with a differential expression of Chrebp target genes. In this work, we aimed to deepen the molecular mechanisms underlying the impact of maqui on the onset and development of the obese phenotype and insulin resistance focusing on liver metabolism. Our results showed that maqui supplementation decreased hepatic steatosis caused by a high-fat diet. Changes in the metabolic profile include a downregulation of the lipogenic liver X receptor (LXR) target genes and of fatty acid oxidation gene expression together with an increase in the expression of small heterodimer partner interacting leucine zipper protein (Smile), a corepressor of the nuclear receptor family. Our data suggest that maqui supplementation regulates lipid handling in liver to counteract the metabolic impact of a high-fat diet.

The aim of this work was to obtain powders rich in bioactive compounds from maqui berry aqueous extracts by spray drying. First, the process parameters of the maqui aqueous extraction were optimized. The optimal operating conditions were found using an experimental Box–Behnken design with three factors: solvent/fruit ratio (2:1, 3.5:1 and 5:1), extraction temperature (25, 50 and 75 °C) and extraction time (30, 75 and 120 min). Soluble solids content, monomeric anthocyanin content (ACY), total polyphenol content (TPC) and antioxidant capacity in the liquid extracts were analyzed as key responses to find the optimal extraction conditions. Secondly, the best aqueous extract (solvent/fruit ratio = 2:1; extraction temperature = 75 °C and extraction time = 75 min) was subjected to spray drying. The effects of different drying adjuvants (maltodextrin, colloidal silicon dioxide, arabic gum, and microcrystalline cellulose) on the powders flow properties, the process yield (PY), the bioactive compounds content and the superficial color were studied. The product based on colloidal silicon dioxide presented the best powder properties: excellent flowability (α: 30.4 ± 0.7°, CI: 8.0 ± 1.7%), adequate moisture content (4.9 ± 0.3%), very good PY (70 ± 1%), high ACY (1528 ± 41 mg cy-3glu/100 g of powder) and TPC (3936 ± 132 mg GAE/100 g of powder), and a purple hue. Here, we report the genome sequence and evidence for transcriptional activity of a virus-like element in the native Chilean berry tree Aristotelia chilensis. We propose to name the endogenous sequence as Aristotelia chilensis Virus 1 (AcV1). High-throughput sequencing of the genome of this tree uncovered an endogenous viral element, with a size of 7122 bp, corresponding to the complete genome of AcV1. Its sequence contains three open reading frames (ORFs): ORFs 1 and 2 shares 66%–73% amino acid similarity with members of the Caulimoviridae virus family, especially the Petunia vein clearing virus (PVCV), Petuvirus genus. ORF1 encodes a movement protein (MP); ORF2 a Reverse Transcriptase (RT) and a Ribonuclease H (RNase H) domain; and ORF3 showed no amino acid sequence similarity with any other known virus proteins. Analogous to other known endogenous pararetrovirus sequences (EPRVs), AcV1 is integrated in the genome of Maqui Berry and showed low viral transcriptional activity, which was detected by deep sequencing technology (DNA and RNA-seq). Phylogenetic analysis of AcV1 and other pararetroviruses revealed a closer resemblance with Petuvirus. Overall, our data suggests that AcV1 could be a new member of Caulimoviridae family, genus Petuvirus, and the first evidence of this kind of virus in a fruit plant.

Various natural sources of antioxidants, which may help defy oxidative stress and thereby reduce the risk of many diseases, are being searched for. Exotic fruits and good quality exotic fruit juices can be an alternative to many berries grown in Europe. This paper presents the antioxidant properties and composition of polyphenols of acai, maqui berry and noni juices. Then they were compared to local juices rich in polyphenols, such as elderberry, raspberry and blueberry juices. The research has shown that the highest antioxidant capacity possessed acai juice among the exotic juices and elderberry juice among the local ones. The same two juices had the highest content of polyphenolic compounds, including flavonoids. Noni and maqui berry juices were inferior to acai and elderberry juices in this respect, and did not surpass considerably the qualities of raspberry and blueberry juices. The highest content of total anthocyanins was detected in elderberry juice. Exotic acai juice contains the highest levels of flavonols (mainly catechins) as well as ferulic and chlorogenic acids. Concluding, juices from exotic acai, noni and maqui berry fruits, because of their antioxidant properties, can be an interesting alternative to local juices. BACKGROUND: Anthocyanins, flavonoids and organic acids widely occurring in extracts of the fruits of Aristotelia chilensis ("Maqui"), concertedly acts on the expression of cyclooxygenase-2 (COX-2), NF-κB, HT-29 and Caco-2 colon cancer cell growth inhibition and on the production of inflammatory mediators. OBJECTIVE: To assess the anti-inflammatory effects of extracts from fruits of "Maqui Berry", on the HT-29 and Caco-2 human colorectal cancer cell lines by measuring COX-2 and NF-κB as well as their antioxidant activities. MATERIAL AND METHODS: Methanol/water extracts and its partitions (acetone and ethyl acetate) from three varieties of "Maqui" were used to access their effects on growth of HT-29 and Caco-2 colon cancer cells, COX-2, NF-κB, NO formation, oxidations by DPPH, TBARS, FRAP and ORAC. RESULTS: Fractions rich in anthocyanins, exhibited potent chemoprotective abilities on decreasing growth of HT-29 and Caco-2 colon cancer cells. The results indicated that the extracts suppressed the production of nitric oxide (NO), through the down-regulation of inducible nitric oxide synthases (iNOS). The structural features of the extracts correlate with the inhibition of NF-κB and COX-2 protein expressions and a potent antioxidant activity by assays of DPPH, FRAP, TBARS and ORAC. The inhibition of cell growth and NO production by selected extracts was dose-dependent with significant effects seen at concentration as low as 25.0 and 10.0 ppm, respectively. CONCLUSIONS: The phenolics (anthocyanins, flavonoids, and organic acids) that occur in extracts from Maqui berry Aristotelia chilensis may provide therapeutic potential against colon cancer. Maqui (Aristotelia Chilensis) berry features a unique profile of anthocyanidins that includes high amounts of delphinidin-3-O-sambubioside-5-O-glucoside and delphinidin-3-O-sambubioside and has shown positive effects on fasting glucose and insulin levels in humans and murine models of type 2 diabetes and obesity. The molecular mechanisms underlying the impact of maqui on the onset and development of the obese phenotype and insulin resistance was investigated in high fat diet-induced obese mice supplemented with a lyophilized maqui berry. Maqui-dietary supplemented animals showed better insulin response and decreased weight gain but also a differential expression of genes involved in de novo lipogenesis, fatty acid oxidation, multilocular lipid droplet formation and thermogenesis in subcutaneous white adipose tissue (scWAT). These changes correlated with an increased expression of the carbohydrate response element binding protein b (Chrebpb), the sterol regulatory binding protein 1c (Srebp1c) and Cellular repressor of adenovirus early region 1A–stimulated genes 1 (Creg1) and an improvement in the fibroblast growth factor 21 (FGF21) signaling. Our evidence suggests that maqui dietary supplementation activates the induction of fuel storage and thermogenesis characteristic of a brown-like phenotype in scWAT and counteracts the unhealthy metabolic impact of an HFD. This induction constitutes a putative strategy to prevent/treat diet-induced obesity and its associated comorbidities.

In our previous investigation, delphinidin, one of the most abundant anthocyanins found in vegetables and berry fruits, had been shown to inhibit osteoclasts and prevent bone loss in mouse models of osteoporosis. In the present study, we investigated whether a delphinidin glycoside-enriched maqui berry extract (MBE, Delphinol®) exhibits beneficial effects on bone metabolism both in vitro and in vivo. MBE stimulated the osteoblastic differentiation of MC3T3-E1 cells, as indicated by enhanced mineralized nodule formation, and increased alkaline phosphatase activity, through the upregulation of bone morphogenetic protein 2 (Bmp2), runt-related transcription factor 2 (Runx2), Osterix (Osx), osteocalcin (Ocn), and matrix extracellular phosphoglycoprotein (Mepe) mRNA expression. Immunostaining and immunoprecipitation assays demonstrated that MBE suppressed NF-κB transnucleation through acting as a superoxide anion/peroxynitrite scavenger in MC3T3-E1 cells. Simultaneously, MBE inhibited both osteoclastogenesis in primary bone marrow macrophages and pit formation by maturated osteoclasts on dentine slices. Microcomputed tomography (micro-CT) and bone histomorphometry analyses of femurs demonstrated that the daily ingestion of MBE significantly increased BV/TV (ratio of bone volume to tissue volume), Tb.Th (trabecular thickness), Tb.N (trabecular number), N.Nd/N.Tm (node to terminus ratio), OV/TV (ratio of osteoid volume to tissue volume), BFR/TV (bone formation rate per tissue volume), and significantly decreased Tb.Sp (trabecular separation), ES/BS (ratio of eroded surface to bone surface) and N.Oc/BS (number of osteoclast per unit of bone surface), compared to vehicle controls in osteopenic mouse models. These findings suggest that MBE can be a promising natural agent for the prevention of bone loss in osteopenic conditions by not only inhibiting bone resorption, but also stimulating bone formation.

Research interest regarding maqui (Aristotelia chilensis) has increased over the last years due to its potential health benefits as one of the most antioxidant-rich berries. Ultrasound-assisted extraction (UAE) is an advanced green, fast, and ecological extraction technique for the production of high quality extracts from natural products, so it has been proposed in this work as an ideal alternative extraction technique for obtaining extracts of high bioactivity from maqui berries. In order to determine the optimal conditions, the extraction variables (percentage of methanol, pH, temperature, ratio “sample mass/volume of solvent„, amplitude, and cycle) were analyzed by a Box-Behnken design, in conjunction with the response surface method. The statistical analysis revealed that the temperature and the percentage of methanol were the most influential variables on the extraction of the total phenolic compounds and total anthocyanins, respectively. The optimal extraction time was determined at 15 min for total phenolic compounds, while it was only 5 min for anthocyanins. The developed methods showed a high precision level with a coefficient of variation of less than 5%. Finally, the new methods were successfully applied to several real samples. Subsequently, the results were compared to those that were obtained in previous experiments by means of microwave assisted extraction (MAE). Similar extraction yields were obtained for phenolic compounds under optimized conditions. However, UAE proved to be slightly more efficient than MAE in the extraction of anthocyanins.

Aristotelia chilensis (Mol.) Stuntz, commonly called Maqui, is known for its pharmacological uses in traditional medicine of native Mapuche communities in Chile. It is reported to have excellent medicinal properties that help to cure chronic diarrhea, dysentery, throat diseases, intestinal tumors, fever, wounds healing, and scars. Pharmacologically, fruits and leaves of Maqui have been investigated for antioxidant, antidiabetic, and antiviral activities. The current high demand of natural antioxidants has placed Maqui as one of the most over-exploited species in Chile. This review analyzes the information collected from the published research articles, and highlights the phytochemical and pharmacological aspects of A. chilensis. BACKGROUND: Fruits like blackcurrant, blueberry and maqui berry stand out for their high content of anthocyanins that give products an attractive red colour, in addition to their rich composition of other polyphenols and vitamins. Comprehensive understanding of thermal properties of foods and quantitative changes in quality attributes are necessary for the proper design of thermal processes. OBJECTIVE: The aim of the present study was to evaluate the kinetics of monomeric anthocyanin (MAcy) and colour degradation due to thermal processing of pulps of blackcurrant, blueberry and maqui berry produced in El Bolsón, Argentina. METHODS: Anthocyanins content was measured with pH-differential method. CIELab parameters were obtained with a Minolta Spectrophotometer, total phenolics were determined by Folin-Ciocalteu method. MAcy degradation and evolution of colour parameter a* were fitted to a first-order model. RESULTS: Activation energies for anthocyanins losses were between 81 and 94 kJ/mol, and for the deterioration of colour between 85 and 89 kJ/mol. In the case of deterioration of anthocyanins, maqui berry presented the highest values of half-life times especially at 90°C. In the case of colour, blackcurrant showed greater stability showing average colour degradation half-life times between 5 and 11 times higher than those of anthocyanins loss. CONCLUSIONS: Kinetic parameters calculated for different berry juices can be used to design a thermal treatment to obtain a high retention of colour and bioactive compounds. [ABSTRACT FROM AUTHOR]

Maqui berry (Aristotelia chilensis) is being explored in the food industry, and is considered to be one of the healthiest berries due to its bioactive components and high commercial value. Microwave-assisted extraction (MAE) was developed for the determination of total phenolic compounds and anthocyanins from maqui. A Box⁻Behnken experimental design was employed in conjunction with a response surface methodology to optimize the conditions based on 27 different experiments. The extractions were carried out with four factors (i.e., methanol percentage, pH, temperature, and solvent volume:sample mass ratio), and two responses—total phenolics and anthocyanins. Temperature and methanol percentage were found to be the most influential parameters for total phenolic compounds and anthocyanins, respectively. The optimum MAE conditions were: 65% MeOH in water at pH 2, temperature of 100 °C, and a ratio of 10:0.5 for total phenolics; and 60% MeOH in water at pH 2, temperature of 50 °C, and a ratio of 14:0.5 for anthocyanins. Kinetics assays were carried out and an optimum time of only 2 min was identified for the extractions. Repeatability and intermediate precision were also evaluated, and coefficients of variation below 5% were obtained. The new methods were successfully applied to a foodstuff made with maqui.

Maqui berry ( Aristotelia chilensis (Molina) Stuntz) has demonstrated a great potential as functional ingredient. However, determining the effect to which the maqui extract or its phytochemicals will benefit the consumers requires further knowledge. Thus, the aim of the present study was to determine the effect of in vitro gastrointestinal digestion (GID) on (i) the recovery and bioaccessibility indexes, (ii) the stability of polyphenolic compounds (phenolic acids, flavonoids and anthocyanins) and (iii) the changes in antioxidant activity of maqui berry grown in Chile. The extracts obtained in each phase (oral, gastric and intestinal) of GID were used to analyse the stability of polyphenolic compounds by means of HPLC whereas the antioxidant activity was determined using four different methodologies. All polyphenolic compounds decreased their concentration after GID and principally the anthocyanins content which was severely affected. The GID process decreased the scavenging properties in 89.9% and 84.2% with DPPH and ABTS assays, respectively, as well as the reducing power 74.1% with respect to non-digested sample. On the other hand, the chelating activity was increased (126.8%). At the end of GID process, the bioaccessibility of phenolic and flavonoid compounds was 78.19 and 14.20%, respectively. The results obtained suggest that although a great amount of maqui berry polyphenolic compounds are lost during digestion process they still have a great potential as antioxidants agents. BACKGROUND The aim of the present study was to determine (1) the polyphenolic profile (phenolic acids, flavonoids and anthocyanins), (2) the antioxidant using four different methodologies ( DPPH, ABTS, FRAP and FIC) and (3) the antibacterial properties of maqui berry [ Aristotelia chilensis (Molina) Stuntz] ( MB) grown in Chile. RESULTS The HPLC analysis of MB showed a total of 19 polyphenolic compounds identified as anthocyanins (eight compounds), flavonols (10 compounds) and ellagic acid. Delphinidin derivatives were the predominant anthocyanins while quercetin derivatives were the predominant flavonols. MB showed an antioxidant activity measured with DPPH, ABTS, FRAP and FIC methods of 28.18, 18.66, 25.22 g Trolox equivalent kg−1 and 0.12 g ethylenediaminetetraacetic acid equivalent kg−1, respectively. With regard to the antibacterial activity, all strains tested were affected by MB. Aeromonas hydrophila and Listeria innocua showed the highest sensitivity to maqui berry extracts with MIC values of 40 and a 50 mg mL−1, respectively. The purpose of this work was to evaluate chitosan edible films (CH) incorporated with maqui berry extracts (MB) for (i) the inhibition growth of some bacterial strains (ii) their total phenolic (TPC) and total flavonoid content and (iii) their antioxidant activity to define if the chitosan edible films incorporated with maqui berry extract (CH + MB) could be used as natural active films for food use. The antioxidant activity was determined with three different analytical assays: DPPH radical scavenging ability assay, Ferrous chelating capacity (FIC) and ferric reducing activity power (FRAP). The agar disc diffusion method was used to determine the antibacterial activity against Listeria innocua , Serratia marcescens , Aeromonas hydrophila , Achromobacter denitrificans , Alcaligenes faecalis , Pseudomonas fluorescens , Citrobacter freundii and Shewanella putrefaciens . CH + MB showed higher antioxidant activity, at all concentrations (0.5 and 1%) and with all methods assayed than CH. Furthermore, the antioxidant activity of CH + MB occurs in a concentration-dependent manner. Regarding the antimicrobial activity, CH + MB were effective against seven of the eight tested bacteria. This antibacterial activity takes place in a concentration-dependent manner. CH were not active, against six of the eight bacteria strain tested. Here, we report the genome sequence and evidence for transcriptional activity of a virus-like element in the native Chilean berry tree Aristotelia chilensis. We propose to name the endogenous sequence as Aristotelia chilensis Virus 1 (AcV1). High-throughput sequencing of the genome of this tree uncovered an endogenous viral element, with a size of 7122 bp, corresponding to the complete genome of AcV1. Its sequence contains three open reading frames (ORFs): ORFs 1 and 2 shares 66%-73% amino acid similarity with members of the Caulimoviridae virus family, especially the Petunia vein clearing virus (PVCV), Petuvirus genus. ORF1 encodes a movement protein (MP); ORF2 a Reverse Transcriptase (RT) and a Ribonuclease H (RNase H) domain; and ORF3 showed no amino acid sequence similarity with any other known virus proteins. Abstract: We used a murine model of type II diabetes, which reproduces the major features of the human disease, and a number of cellular models to study the antidiabetic effect of ANC, a standardised anthocyanin-rich formulation from Maqui Berry (Aristotelia chilensis). We also isolated delphinidin 3-sambubioside-5-glucoside (D3S5G), a characteristic anthocyanin from Maqui Berry, and studied its antidiabetic properties. We observed that oral administration of ANC improved fasting blood glucose levels and glucose tolerance in hyperglycaemic obese C57BL/6J mice fed a high fat diet. In H4IIE rat liver cells, ANC decreased glucose production and enhanced the insulin-stimulated down regulation of the gluconeogenic enzyme, glucose-6-phosphatase. In L6 myotubes ANC treatment increased both insulin and non-insulin mediated glucose uptake. As with the ACN, oral administration of pure D3S5G dose-dependently decreased fasting blood glucose levels in obese C57BL/6J mice, and decreased glucose production in rat liver cells. D3S5G also increased glucose uptake in L6 myotubes and is at least partially responsible for ANC’s anti-diabetic properties. [Copyright &y& Elsevier]

• Plasma concentration of bioavailable (poly)phenols was assessed upon a longitudinal intervention. • Influence of sweetener on intestinal absorption of maqui-citrus (poly)phenols was evaluated. • Sucralose and stevia allowed the highest bioavailability in the majority of cases. • Sucralose and stevia were found as suitable alternative to sucrose in term of (poly)phenols bioavailability. The comparison of non-caloric sweeteners (stevia and sucralose) and sucrose, on the plasma concentration and cumulative effects of phenolic compounds, was achieved. A long-term intervention, consisting of the daily intake of 330 mL of healthy citrus-maqui soft drinks, for 60 days, by 138 healthy overweight adults, was followed. A total of 24 bioavailable metabolites derived from caffeic acid, 3,4-di-hydroxyphenylacetic acid, eriodictyol, homoeriodictyol, hippuric acid, naringenin, 2,4,6-tri-hydroxybenzaldehyde, and vanillic acid were detected in peripheral blood plasma. A similar augment of bioactive compounds in plasma concentrations were found for the three beverages, in the range 12.3% (day 0)- 85.3% (day 60), depending on the analyte considered. Due to this, the present study highlights sucralose and stevia as valuable alternatives to sucrose, providing and non-significantly different plasma concentration and cumulative effect in the plasma, thus contributing to prevent a diversity of metabolic disorders and health constraints. Maqui is a wild native berry from the Chilean and Argentinean Patagonia that is considered an important source of bioactive compounds for its high antioxidant activity and anthocyanin concentration. Processing methods that improve shelf-life can affect stability of antioxidant compounds. Furthermore, there is limited information of the processing effect on non-anthocyanin phenolic compounds, antioxidant capacity and quality of maqui-derived products. The aim of this work was to those characteristics on processed fruit using conventional canning (CC), convective forced hot air drying (CFHAD), osmotic drying (OD), and freeze-drying (FD). Ten non-anthocyanin phenolics were quantified using HPLC-MS. Luteolin-7- O -glucoside and hydroxycinnamic acid concentration showed a significant reduction by 45% when processed using CFHAD and OD. Rutin and hyperoside concentrations were significantly higher (24.76 μg/g and 100.20 μg/g) in FD maqui when compared to CC and OD treatments. CFHAD achieved the lowest water activity (0.41) and CC the highest total soluble solids content 28.00 °Brix. Antioxidant capacity of CC maqui samples showed a 17.6% decrease, while FD, CFHAD, and OD samples showed an average 21.8% increase.
