Camel products are receiving great interest worldwide because of their high functional properties and nutritive values. Therefore, this study was focused on the variation of copper [Cu], zinc [Zn], manganese [Mn], selenium [Se], iron [Fe], iodine [I], and some heavy metals, cobalt [Co], lead [Pb], and cadmium [Cd], in the blood and tissues of slaughtered camels from five regions in Saudi Arabia [SA] during the summer and winter seasons, because environmental factors vary from region to region. Whole blood, meat, liver, rumen fluid, and rumen tissues were collected from the slaughterhouse in each region during the two seasons. Moreover, samples were prepared and analyzed for trace mineral and heavy metal concentrations using ICP-MS. The data were statistically analyzed as part of a complete randomized design and correlation analysis for season and location using SAS. The findings revealed a pattern in the minerals, with Ca being the only mineral that was unrelated to other minerals in the liver. For lead and cadmium, our mean value in liver [0.40 µg/g] was below the limit of the EU standard for cadmium [0.50 µg/g], while in meat and liver, lead contents [1.62 µg/g and 2.57 µg/g, respectively] were above the limit of the EU standard [0.10 and 0.20 µg/g, respectively]. For meat, the significantly highest positive correlations were observed between P and Mg [R2 = 0.928], Fe and Mn [R2 = 0.860], and Co and Mn [R2 = 0.821]. For rumen tissues, P and Mg were highly correlated [R2 = 0.958] as well as Zn and Mg [R2 = 0.857], Zn and P [R2 = 0.836], and Fe and Ca [R2 = 0.802]. As a result, a region and season reflect variations in mineral concentrations in SA during the summer and winter seasons. Further intensive research is needed to investigate the minerals’ biological mechanisms in camels under different environmental conditions.

Gold-mining activities have been demonstrated to result in significant environmental pollution by Hg, Pb, and Mn, causing serious concerns regarding the potential threat to the public health of neighboring populations around the world. The present study focused on heavy-metal contamination in the eggs, blood, feed, soil, and drinking water on chicken farms, duck farms, and free-grazing duck farms located in areas < 25 km and > 25 km away from a gold mine in northern Thailand. In an area < 25 km away, Hg, Pb, and Mn concentrations in the eggs of free-grazing ducks were significantly higher than > 25 km away (p < 0.05). In blood, Hg concentration in free-grazing ducks was also significantly higher than those in an area > 25 km away (p < 0.05). Furthermore, the Pb concentration in the blood of farm ducks was significantly higher than in an area > 25 km away (p < 0.05). The concentration of Cd in drinking water on chicken farms was significantly higher for farms located within 25 km of the gold mine (p < 0.05). Furthermore, a high correlation was shown between the Pb (r2 = 0.84) and Cd (r2 = 0.42) found between drinking water and blood in free-grazing ducks in the area < 25 km away. Therefore, health risk from heavy-metal contamination was inevitably avoided in free-grazing activity near the gold mine. The incremental lifetime cancer risk (ILCR) in the population of both Pb and Cd exceeded the cancer limit (10−4) for all age groups in both areas, which was particularly high in the area < 25 km for chicken-egg consumption, especially among people aged 13–18 and 18–35 years old. Based on these findings, long-term surveillance regarding human and animal health risk must be strictly operated through food chains and an appropriate control plan for poultry businesses roaming around the gold mine.

Freshwater fishes from the Tonle Sap Lake are used in several programs to improve the nutritional status of children and pregnant women. Our aim was to characterize the overall nutritional profile and heavy metal contents of ten freshwater fish species. The lipid contents ranged from 1.4 to 10.0 g/100 g and fish can be considered rich in omega 3. The vitamin A content was ten times higher in small fish eaten whole than in fillets of large fish; the same applies to the Fe and Zn contents, but to a lesser extent. Mn, As and Pb contents were over the maximum permissible levels in several fishes. According to SAIN and LIM classification, the ten fish species belong to the food groups recommended for health. However, when heavy metals were integrated in LIM, seven species were ranked into the food to consume in small quantities. Globally, the most interesting fish species was Henicorhynchus siamensis.

The OCBIL theory comprises a set of hypotheses to comprehend the biota of old, climatically buffered, infertile landscapes (OCBILs). Here, we review evidence from the literature to evaluate the extent to which this theory could apply to the biodiversity hotspot of New Caledonia. We present geological, pedological and climatic evidence suggesting how the island might qualify as an OCBIL. The predictions of OCBIL theory are then reviewed in the context of New Caledonia. There is evidence for a high rate of micro-endemism, accumulation of relict lineages, a high incidence of dioecy, myrmecochory and nutritional specializations in plants. New Caledonian vegetation also exhibits several types of monodominant formations that reveal the importance of disturbances on the island. Fires and tropical storms are likely to be important factors that contribute to the dynamic of New Caledonian ecosystems. Although naturally infertile, there is archaeological evidence that humans developed specific horticultural practices in the ultramafic landscapes of New Caledonia. Further comparisons between New Caledonia and other areas of the world, such as South Africa and Southwest Australia, are desirable, to develop the OCBIL theory into a more robust and generalized, testable framework and to determine the most efficient strategies to preserve their outstanding biodiversity.

International audience; Mining activity may cause heavy metal accumulation, which threatens human and animal health by their long-term persistence in the environment. This study aims to assess the impact of polymetallic pollution on chicken (Gallus domesticus) from old lead mining sites in northeast of Tunisia: Jebel Ressas (JR). Samples of soil and chickens were collected from five sites being ranked along a gradient of heavy metal contamination. Heavy metal loads were evaluated in soil samples and in chicken liver and kidney. Biochemical evaluation of oxidative stress parameters termed as Catalase (CAT), Glutathione-S-Transferase (GST), and Malondialdehydes (MDA) accumulation was monitored. Metallothionein protein level was assessed as a specific response to heavy metals. DNA alteration was achieved using MNi frequency in the investigated tissues. Finally, the evaluation of gene expression levels of CAT, GST, mt1, mt4, P53, bcl2, caspase3 and DNA-ligase was performed. Our data showed the highest loads of Cd, Cu, Zn and Pb in tissues of animals from site 3, being more pronounced in kidney. Biochemical data suggested a significant increase in antioxidant enzymes activities in all sites respect to control except in site 3 were CAT and GST were inhibited. DNA alteration was observed in all tissues being very pronounced in animals from site 3. Overall, transcriptomic data showed that genes involved in apoptosis were up-regulated in animals exposed to the most contaminated soils. Our data suggest that chicken and selected biomarkers offer a suitable model for biomonitoring assessment of heavy metals transfer through the food web in mining sites. Finally, the obtained results of heavy metals accumulation and related alterations should be carefully considered in view of the controversial relationship between distribution and toxicology of contaminants in exposed organisms.

New Caledonia is a global biodiversity hotspot known for its metal hyperaccumulator plants. X-ray fluorescence technology (XRF) has enabled non-destructive and quantitative determination of elemental concentrations in herbarium specimens from the ultramafic flora of the island. Specimens belonging to six major hyperaccumulator families (Cunoniaceae, Phyllanthaceae, Salicaceae, Sapotaceae, Oncothecaceae and Violaceae) and one to four specimens per species of the remaining ultramafic taxa in the herbarium were measured. XRF scanning included a total of c. 11 200 specimens from 35 orders, 96 families, 281 genera and 1484 species (1620 taxa) and covered 88.5% of the ultramafic flora. The study revealed the existence of 99 nickel hyperaccumulator taxa (65 known previously), 74 manganese hyperaccumulator taxa (11 known previously), eight cobalt hyperaccumulator taxa (two known previously) and four zinc hyperaccumulator taxa (none known previously). These results offer new insights into the phylogenetic diversity of hyperaccumulators in New Caledonia. The greatest diversity of nickel hyperaccumulators occur in a few major clades (Malphigiales and Oxalidales) and families (Phyllanthaceae, Salicaceae, Cunoniaceae). In contrast, manganese hyperaccumulation is phylogenetically scattered in the New Caledonian flora.

[Departement_IRSTEA]Eaux [TR1_IRSTEA]ARCEAU [ADD1_IRSTEA]Hydrosystèmes et risques naturels; International audience; RECOTOX is a cross-cutting initiative promoting an integrated research to respond to the challenges of monitoring, understanding, and mitigating environmental and health impacts of pesticides in agroecosystems. The added value of RECOTOX is to develop a common culture around spatial ecotoxicology including the whole chain of pressure-exposure-impact, while strengthening an integrated network of in natura specifically equipped sites. In particular, it promotes transversal approaches at relevant socioecological system scales, to capitalize knowledge, expertise, and ongoing research in ecotoxicology and, to a lesser extent, environmental toxicology. Thus, it will open existing research infrastructures in environmental sciences to research programs in ecotoxicology of pesticides.

Environmental pollution through heavy metals is an upcoming universal problem that relentlessly endangers human health, biodiversity and ecosystems. Hence remediating these heavy metal pollutants from the environment by engineering soil microbiome through metatranscriptomics is befitting reply. In the present investigation, we have constructed size fractionated cDNA libraries from eukaryotic mRNA of cadmium (Cd) contaminated soil and screened for Cd tolerant genes by yeast complementation system by using Cd sensitive ycf1Δ mutant. We are reporting one of the transformants PLCe10 (from library C, 1–4 kb) with potential tolerance towards Cd toxicity (40 μM–80 μM). Sequence analysis of PLCe10 transcript showed homology to von Willebrand factor type D domain (VWD) of vitellogenin-6 of Ascaris suum encoding 338 amino acids peptide. qPCR analysis revealed that PLCe10 induced in presence of Cd (32 fold) and also accumulated maximum amount of Cd at 60 μM Cd. This cDNA was further tested for its tolerance against other heavy metals like copper (Cu), zinc (Zn) and cobalt (Co). Heterologous complementation assays of cDNA PLCe10 showed a range of tolerance to Cu (150 μM–500 μM), Zn (10 mM–12 mM) and Co (2–4 mM). Results of the present study suggest that cDNA PLCe10 is one of the functional eukaryotic heavy metal tolerant genes present among the soil microbial community and could be exploited to rehabilitate metal contaminated sites.

International audience; Although game meat consumption is widespread across the globe, involving a great diversity of species in very different ecological and sociocultural environments, the safety of wild meat products is rarely assessed routinely. We conducted a survey to evaluate the safety of two rodents products (Rattus tanezumi and Bandicota indica) purchased from roadside markets in central Thailand. Total aerobic bacteria and Escherichia coli counts measured were above Thai poultry product standards for 83.3% and 100% of the samples, respectively (n = 108), although there was no difference between rat species (analysis of variance [ANOVA], p > 0.05). Salmonella spp. were isolated from 32% of the swabs collected (67/208), including 19 different serovars. All strains were resistant or partially resistant to at least 2 of the 16 antibiotics tested, with levels of resistance varying greatly among antibiotics (e.g., 97% of strains sensitive to ciprofloxacin and 98.5% of strains resistant to cloxacillin). Detectable traces of Cd and Pb were found in 98% and 46% of the samples analyzed, respectively (n = 61). Pb and Cd concentrations measured in the kidneys of R. tanezumi were significantly higher than in the liver and muscles and significantly higher than B. indica muscles and kidneys but not liver (ANOVA, p < 0.05). These results highlight potential health hazards that may be associated with rodent-meat consumption, including contamination by coliform bacteria, multiresistant Salmonella spp. strains, and heavy metals. The significance of these results for public health cannot be determined precisely in the absence of appropriate standards, and information gaps remain regarding the frequency of rodent-meat consumption and the origin of bacterial and heavy metal contaminations (i.e., capture environment or during carcass handling and processing). We suggest that appropriate information and training on best hygienic practices for preparing, cooking, and preserving rodent meat should be provided to the producers and to the consumers.

Purpose: Non-sanitary landfill or open dumping stored both non-hazardous and hazardous waste disposed from households, restaurants, agricultural activities, and industrial sectors that could contaminated to the environment. This study aimed to assess the environmental status and its impact of opened solid waste landfill located nearby agriculture and natural resources in municipal area of central Thailand. Methods & Materials: Soil and water samples were collected from six locations of landfill and surrounding areas such as paddy field and water reservoir at two-month interval for six consecutive months. Heavy metal contamination and quality of soil and water including physical, chemical, biological parameters were analyzed by using standard methods. Agricultural activities of the land nearby the landfill and practice behavior of workers in the landfill have been observed. Results: We found Salmonella spp. and Clostridium perfringens contaminated in soil samples collected from both inside and outside of landfill. Heavy metals such as Iron (Fe), Magnesium (Mg) and Aluminum (Al) were found in high level, additionally, contamination of Copper (Cu) in soil collected inside the landfill was presented. Water collected from leachate and natural resource was contaminated by coliform bacteria and Salmonella spp. Level of Fe in all water samples was high, however this amount was not above limited value. Inappropriate self-protective and non-hygienic behavior of workers in the landfill was also found. Conclusion: Soil and water samples collected from the opened landfill were contaminated with bacteria. Occupational hazard, protective awareness and protective measure of people who exposed to the landfill such as garbage collectors and farmers should be concerned. Level of heavy metals in soil and water, especially in paddy fields around the landfill should be periodically monitored.

Background: Nickel (Ni) phytomining operations cultivate hyperaccumulator plants (‘metal crops’) on Ni-rich (ultramafic) soils, followed by harvesting and incineration of the biomass to produce a high-grade ‘bio-ore’ from which Ni metal or pure Ni salts are recovered. Scope: This review examines the current status, progress and challenges in the development of Ni phytomining agronomy since the first field trial over two decades ago. To date, the agronomy of less than 10 species has been tested, while most research focussed on Alyssum murale and A. corsicum. Nickel phytomining trials have so far been undertaken in Albania, Canada, France, Italy, New Zealand, Spain and USA using ultramafic or Ni-contaminated soils with 0.05–1 % total Ni. Conclusions: N, P and K fertilisation significantly increases the biomass of Ni hyperaccumulator plants, and causes negligible dilution in shoot Ni concentration. Organic matter additions have pronounced positive effects on the biomass of Ni hyperaccumulator plants, but may reduce shoot Ni concentration. Soil pH adjustments, S additions, N fertilisation, and bacterial inoculation generally increase Ni phytoavailability, and consequently, Ni yield in ‘metal crops’. Calcium soil amendments are necessary because substantial amounts of Ca are removed through the harvesting of ‘bio-ore’. Organic amendments generally improve the physical properties of ultramafic soil, and soil moisture has a pronounced positive effect on Ni yield. Repeated ‘metal crop’ harvesting depletes soil phytoavailable Ni, but also promotes transfer of non-labile soil Ni to phytoavailable forms. Traditional chemical soil extractants used to estimate phytoavailability of trace elements are of limited use to predict Ni phytoavailability to ‘metal crop’ species and hence Ni uptake.

A post Ferro-nickel mining land management needs to recognize the physical, chemical characteristics of soil and plants that grow naturally by tolerating the levels of heavy metals. The objective of this study were to: i) investigate and evaluate the natural process of plant colonization on abandoned ferro-nickel mining sites located on ultramafic substrate; to ii) understand the relationship between soil properties and species distribution and to iii) assess the potential of hyperaccumulator plants for the soil phytoremediation. This area was divided into five 100 m(2) plots that were representatives of the variability of the site, based on localization, land use and total vegetative cover. For each plot, the physicochemical characteristics of the soils, the inventory of plant species and the accumulation of Ni by all plant taxa were determined. The soils in the ex-mining area of Perrenjas appeared highly polluted by Ni, Co and Cr compared to the natural levels of the surrounding ultramafic soils. The toxic levels of trace metals and the deficiency of organic matter appeared as major constraints for plant colonization. In total, 96 plant species belonging to 26 families in the studied mining sites of Perrenjas were recorded. Several plants show the ability to tolerate and accumulate metals and may be useful for phyto-stabilization. Alyssum murale was the best candidate for Ni phyto-extraction technologies, with concentrations above 1000 mg/kg Ni in plant tissues.

Background: Hyperaccumulator plants are unusual plants that accumulate particular metals or metalloids, such as nickel, zinc, cadmium and arsenic, in their living tissues to concentrations that are hundreds to thousands of times greater than what is normal for most plants. The hyperaccumulation phenomenon is rare (exhibited by less than 0.2% of all angiosperms), with most of the ~500 hyperaccumulator species known globally for nickel. Scope: This review highlights the contemporary understanding of nickel hyperaccumulation processes, which include root uptake and sequestration, xylem loading and transport, leaf compartmentation and phloem translocation processes. Conclusions: Hyperaccumulator plants have evolved highly efficient physiological mechanisms for taking up nickel in their roots followed by rapid translocation and sequestration into the aerial shoots. The uptake of nickel is mainly involved with low affinity transport systems, presumably from the ZIP family. The presence of high concentrations of histidine prevents nickel sequestration in roots. Nickel is efficiently loaded into the xylem, where it mainly presents as Ni2+. The leaf is the main storage organ, which sequestrates nickel in non-active sites, e.g. vacuoles and apoplast. Recent studies show that phloem translocates high levels of nickel, which has a strong impact on nickel accumulation in young growing tissues.

Rapid heavy metal soil surveys at large scale with high sampling density could not be con- ducted with traditional laboratory physical and chemical analyses because of the high cost, low efficiency and heavy workload involved. This study explored a rapid approach to assess heavy metals contamination in 301 farmland soils from Fuyang in Zhejiang Province, in the southern Yangtze River Delta, China, using portable proximal soil sensors. Portable X-ray fluorescence spectroscopy (PXRF) was used to determine soil heavy metals total concentrations while soil pH was predicted by portable visible-near infrared spectroscopy (PVNIR). In Morocco, pollution caused by closed mines continues to be a serious threat to the environment, like the generation of acid mine drainage. Mine drainage is produced by environmental and microbial oxidation of sulfur minerals originating from mine wastes. The fundamental role of microbial communities is well known, like implication of Fe-oxidizing and to a lesser extent S-oxidizing microorganism in bioleaching. However, the structure of the microbial communities varies a lot from one site to another, like diversity depends on many factors such as mineralogy, concentration of metals and metalloids or pH, etc. In this study, prokaryotic communities in the pyrrhotite-rich tailings of Kettara mine were characterized using the Illumina sequencing. In-depth phylogenetic analysis revealed a total of 12 phyla of bacteria and 1 phyla of Archaea. The majority of sequences belonged to the phylum of Proteobacteria and Firmicutes with a predominance of Bacillus, Pseudomonas or Corynebacterium genera. Many microbial populations are implicated in the iron, sulfur and arsenic cycles, like Acidiferrobacter, Leptospirillum, or Alicyclobacillus in Fe; Acidiferrobacter and Sulfobacillus in S; and Bacillus or Pseudomonas in As. This is one of the first description of prokaryotic communities in pyrrhotite-rich mine tailings using high-throughput sequencing.

New Caledonian serpentine (ultramafic) soils contain high levels of toxic heavy metals, in particular nickel, (up to 20 g kg−1) and are deficient in essential elements like carbon, nitrogen and phosphorus while having a high magnesium/calcium ratio. Although previous studies showed that ectomycorrhizal symbioses could play an important role in the adaptation of the endemic plants to ultramafic soils (FEMS Microbiol Ecol 72:238–49, 2010), none of them have compared the diversity of microbial communities from ultramafic vs non-ultramafic soils in New Caledonia. We explored the impact of edaphic characteristics on the diversity of ectomycorrhizal (ECM) fungi associated with different endemic species of Tristaniopsis (Myrtaceae) growing under contrasting soil conditions in the natural ecosystems of New Caledonia. ECM root tips were thus sampled from two different ultramafic sites (Koniambo massif and Desmazures forest) vs two volcano-sedimentary ones (Arama and Mont Ninndo). The molecular characterization of the ECM fungi through partial sequencing of the ITS rRNA gene revealed the presence of different dominant fungal genera including, both soil types combined, Cortinarius (36.1%), Pisolithus (18.5%), Russula (13.4%), Heliotales (8.2%) and Boletellus (7.2%). A high diversity of ECM taxa associated with Tristaniopsis species was found in both ultramafic and volcano-sedimentary soils but no significant differences in ECM genera distribution were observed between both soil types. No link could be established between the phylogenetic clustering of ECM taxa and their soil type origin, thus suggesting a possible functional—rather than taxonomical—adaptation of ECM fungal communities to ultramafic soils.

International audience; Nickel (Ni), an essential micronutrient, is toxic to plants and animals at elevated levels. Fewer data are available on the dynamics of Ni than for trace metals such as Cu and Zn. Dietary intake is the major source of Ni, but almost no information is available on dietary intake for Asian populations whose diet contains much less animal products and processed foods than in Western countries. In south-eastern China, most food is grown on peri-urban agricultural land and thus, food crops from these areas should be the major source of dietary Ni. A large-scale survey of Ni in soils and edible crops, including 515 sites and 24 crop species from 10 crop groups, was conducted in Fujian Province, SE China, to assess Ni intake for a typical SE Asian diet. Correlation, principal component analysis (PCA) and data mining were used to identify soil and plant factors that determine Ni accumulation in crops. Both soil Ni content and plant species contributed to Ni accumulation. Crop Ni (Ni-crop) was positively correlated with soil Ni DTPA-extractible (diethylenetriaminepentaacetic acid) (Ni-DTPA) for many crops. PCA did not identify strong relations between soil-to-crop transfer factors and any soil properties. Stepwise linear correlations found positive correlations between Ni-crop and Ni-DTPA; however, relationships between Ni-crop and both silt content and pH varied between crops. Dietary intake of foods containing mean Ni content could lead to health risks, especially for children. It was concluded that sandy soils, rich in organic matter with a large Ni content, are unsuitable for the production of rice and legumes.

Soils contaminated with heavy metals represent a serious threat for humans and all ecosystems. In some cases, they can be remediated by metal phytoextraction, using accumulator or hyperaccumulator plants, however the fate of harvested metal-enriched biomass has to be addressed. Cadmium (Cd) and zinc (Zn) are very common contaminants of urban, industrial and agricultural soils. To date, no process has been developed to recover these metals from plant biomass. This contribution presents a novel hydrometallurgical process designed to recover Cd and Zn from the biomass of a Zn/Cd hyperaccumulator plant Noccaea caerulescens . Dried plants are first ashed at 620 °C, a process which has been carefully investigated to avoid metal loss, and then the ash is acid leached. Following this, the process comprises two main steps: one to extract Cd and Zn and the other for Cd cementation using Zn powder and selective precipitation of Zn. Optimal cementation conditions have been determined with synthetic solutions using a Box-Behnken experimental design. In this context, a mole ratio Zn:Cd of 2:1, a temperature of 25 °C and a duration of 50 min proved optimal. The full process was tested on a sample of plant biomass. It demonstrated that Cd and Zn recovery was possible by cementation and precipitation. This new process still has to be optimized and up-scaled but it paves the way to Cd and Zn recovery from diverse types of soils, and even to ‘metal cultivation’, an emergent phytotechnology now referred to as agromining.

International audience; Ectomycorrhizal fungi (ECM) isolates of Pisolithus albus (Cooke and Massee) from nickel-rich ultramafic topsoils in New Caledonia were inoculated onto Acacia spirorbis Labill. (an endemic Fabaceae) and Eucalyptus globulus Labill. (used as a Myrtaceae plant host model). The aim of the study was to analyze the growth of symbiotic ECM plants growing on the ultramafic substrate that is characterized by high and toxic metal concentrations i.e. Co, Cr, Fe, Mn and Ni, deficient concentrations of plant essential nutrients such as N, P, K, and that presents an unbalanced Ca/Mg ratio (1/19). ECM inoculation was successful with a plant level of root mycorrhization up to 6.7%. ECM symbiosis enhanced plant growth as indicated by significant increases in shoot and root biomass. Presence of ECM enhanced uptake of major elements that are deficient in ultramafic substrates; in particular P, K and Ca. On the contrary, the ECM symbioses strongly reduced transfer to plants of element in excess in soils; in particular all metals. ECM-inoculated plants released metal complexing molecules as free thiols and oxalic acid mostly at lower concentrations than in controls. Data showed that ECM symbiosis helped plant growth by supplying uptake of deficient elements while acting as a protective barrier to toxic metals, in particular for plants growing on ultramafic substrate with extreme soil conditions. Isolation of indigenous and stress-adapted beneficial ECM fungi could serve as a potential tool for inoculation of ECM endemic plants for the successful restoration of ultramafic ecosystems degraded by mining activities.

International audience; Due to diffuse atmospheric fallouts of process particles enriched by metals and metalloids, polluted soils concern large areas at the global scale. Useful tools to assess ecotoxicity induced by these polluted soils are therefore needed. Earthworms are currently used as biotest, however the influence of specie and earthworm behaviour, soil characteristics are poorly highlighted. Our aim was therefore to assess the toxicity of various polluted soils with process particles enriches by metals and metalloids (Pb, Cd, Cu, Zn, As and Sb) collected from a lead recycling facility on two earthworm species belonging to different ecological types and thus likely to have contrasted behavioural responses (Eiseina hortensis and Lum-bricus terrestris). The combination of behavioural factors measurements (cast production and biomass) and physico-chemical parameters such as metal absorption, bioaccumulation by earthworms and their localization in invertebrate tissues provided a valuable indication of pollutant bioavailability and ecotoxicity. Soil characteristics influenced ecotoxicity and metal uptake by earthworms, as well as their soil bioturbation.

International audience; The acid mine drainage (AMD) impacted creek of the Carnoulès mine (Southern France) is characterized by acid waters with a high heavy metal content. The microbial community inhabiting this AMD was extensively studied using isolation, metagenomic and metaproteomic methods, and the results showed that a natural arsenic (and iron) attenuation process involving the arsenite oxidase activity of several Thiomonas strains occurs at this site. A sensitive quantitative Selected Reaction Monitoring (SRM)-based proteomic approach was developed for detecting and quantifying the two subunits of the arsenite oxidase and RpoA of two different Thiomonas groups. Using this approach combined with 16S rRNA gene sequence analysis based on pyrosequencing and FISH, it was established here for the first time that these Thiomonas strains are ubiquitously present in minor proportions in this AMD and that they express the key enzymes involved in natural remediation processes at various locations and time points. In addition to these findings, this study also confirms that targeted proteomics applied at the community level can be used to detect weakly abundant proteins in situ.

Des stratégies sont ainsi mises en oeuvre pour contrôler l’homéostasie minérale, limiter les carences et maintenir les éléments essentiels et contaminants en deçà de leurs seuils de toxicité. Comprendre ces processus fondamentaux constitue un enjeu majeur pour l’agriculture, l’alimentation et l’environnement.

Background: Dietary guidelines are designed to help meet nutritional requirements, but they do not explicitly or quantitatively account for food contaminant exposures.Objective: In this study, we aimed to test whether dietary changes needed to achieve nutritional adequacy were compatible with acceptable exposure to food contaminants.Methods: Data from the French national dietary survey were linked with food contaminant data from the French Total Diet Study to estimate the mean intake of 204 representative food items and mean exposure to 27 contaminants, including pesticides, heavy metals, mycotoxins, nondioxin-like polychlorinated biphenyls (NDL-PCBs) and dioxin-like compounds. For each sex, 2 modeled diets that departed the least from the observed diet were designed: 1) a diet respecting only nutritional recommendations (NUT model), and 2) a diet that met nutritional recommendations without exceeding Toxicological Reference Values (TRVs) and observed contaminant exposures (NUTOX model). Food, nutrient, and contaminant contents in observed diets and NUT and NUTOX diets were compared with the use of paired t tests.Results: Mean observed diets did not meet all nutritional recommendations, but no contaminant was over 48% of its TRV. Achieving all the nutrient recommendations through the NUT model mainly required increases in fruit, vegetable, and fish intake and decreases in meat, cheese, and animal fat intake. These changes were associated with significantly increased dietary exposure to some contaminants, but without exceeding 57% of TRVs. The highest increases were found for NDL-PCBs (from 26% to 57% of TRV for women). Reaching nutritional adequacy without exceeding observed contaminant exposure (NUTOX model) was possible but required further departure from observed food quantities.Conclusions: Based on a broad range of nutrients and contaminants, this first assessment of compatibility between nutritional adequacy and toxicological exposure showed that reaching nutritional adequacy might increase exposure to food contaminants, but within tolerable levels. However, there are some food combinations that can meet nutritional recommendations without exceeding observed exposures.

The taxonomic status of eleven rhizospheric bacterial strains belonging to the genus Burkholderia and isolated from roots of Costularia (Cyperaceae), tropical herbaceous pioneer plants growing on ultramafic soils in New Caledonia, was investigated using a polyphasic taxonomic approach. The genetic analyses (16S rRNA genes, gyrB, recA, nreB and cnr) confirmed that all strains are Burkholderia and cluster into two separated groups. The DNA hybridization results showed low relatedness values to the closest relatives Burkholderia species. The phenotypic analyses confirmed that the two groups of strains could be differentiated from each other and from other known Burkholderia species. This polyphasic study revealed that these two groups of strains represent each a novel species of Burkholderia, for which the names Burkholderia novacaledonica sp. nov. (type strain STM10272(T)=LMG28615(T)=CIP110887(T)) and B. ultramafica sp. nov. (type strain STM10279(T)=LMG28614(T)=CIP110886(T)) are proposed, respectively. These strains of Burkholderia presented specific ecological traits such as the tolerance to the extreme edaphic constraints of ultramafic soils: they grew at pH between 4 and 8 and tolerate the strong unbalanced Ca/Mg ratio (1/19) and the high concentrations of heavy metals i.e. Co, Cr, Mn and Ni. Noteworthy B. ultramafica tolerated nickel until 10mM and B. novacaledonica up to 5mM. The presence of the nickel (nreB) and cobalt/nickel (cnr) resistance determinants encoding for protein involved in metal tolerance was found in all strains of both groups. Moreover, most of the strains were able to produce plant growth promoting molecules (ACC, IAA, NH3 and siderophores). Such ecological traits suggest that these new species of Burkholderia might be environmentally adaptable plant-associated bacteria and beneficial to plants.

Waste dumps generated by mining activities contain heavy metals that are dispersed into areas leading to significant environmental contamination. The objectives of this study were (i) to survey native plants and their associated AM fungal communities from waste soils in a Moroccan mine site and (ii) to follow Eucalyptus growth in soil collected from the waste-mine. AM spores from native plant species were collected from the mining site and the surrounding uncontaminated areas were multiplied and inoculated onto Eucalyptus camaldulensis. The results showed that (i) the native plant species recorded in the waste did not show an active metal uptake, (ii) the selected native plant species are associated with AM mycorrhizal fungi and (iii) the use of AM fungi adapted to these drastic conditions can improve the growth of the fast-growing tree, E. camaldulensis and its tolerance to high soil Cu content. In conclusion, it is suggested that in order to define efficient low-cost phytostabilization processes, the use of native resources (i.e., mixtures of native mycorrhizal fungi) in combination with fast-growing tree species such as Eucalyptus, could be used to optimize the establishment of a permanent cover plant in contaminated areas.

Microbial species richness and assemblages across ultramafic ecosystems were investigated to assess the relationship between their distributional patterns and environmental traits. The structure of microorganism communities in the Koniambo massif, New Caledonia, was investigated using a metagenetic approach correlated with edaphic and floristic factors. Vegetation cover and soil properties significantly shaped the large phylogenetic distribution of operational taxonomic unit within microbial populations, with a mean per habitat of 3,477 (± 317) for bacteria and 712 (± 43) for fungi. Using variance partitioning, we showed that the effect of aboveground vegetation was the most significant descriptor for both bacterial and fungal communities. The floristic significant predictors explained 43% of the variation both for the bacterial and fungal community structures, while the edaphic significant predictors explained only 32% and 31% of these variations, respectively. These results confirm the previous hypothesis, that the distribution of microorganisms was more structured by the vegetation cover rather than the edaphic characteristics and that microbial diversity is not limited in ultramafic ecosystems.

International audience; The aim of this study was to identify bacteria present in the nodules of the legume Hedysarum spinosissimum growing in metal-contaminated soils; and to test whether these root-nodule bacteria are able to promote host plant growth and enhance their phytostabilization potential. Seventy-four bacteria were isolated from nodules of H. spinosissimum growing in 3 different mining sites in Morocco and were identified by 16S rDNA gene sequencing. They belonged to 8 genera affiliated to Pseudomonas (49 strains), Pantoea (11), Rhizobium (6), Herbaspirillum (3), Bacillus (2), and one strain of Serratia, Agrobacterium and Azospirillum. Seven strains, presenting a high tolerance to Zn and Pb, exhibited capacity of inorganic phosphate solubilization and ammonia production from peptone degradation. The inoculation of H. spinosissimum, growing in 2000 μM of Zn by Pseudomonas putida and Herbaspirillum huttiense suppresses Zn toxicity symptoms in plants and also enhances plant growth by significantly increasing plant shoot and root fresh weights. The maximal Zn accumulation was observed in roots of plants inoculated by Pseudomonas putida with a translocation factor of 0.05(\textpm 0.006). Our results, evidence that the selection of metal-resistant bacteria is a key step in polluted soils for the use of plants like Hedysarum spinosissimum for in situ phytoremediation.

A multipolluted soil sampled from a former coking plant in Lorraine (France) was evaluated for its genotoxic effects on coelomocytes of the Eisenia fetida earthworm using the comet assay. The biological efficiency of thermal desorption of the contaminated soil was also investigated. The untreated polluted soil was shown to be genotoxic to earthworms. Although thermal desorption reduced the concentration of PAHs by 94% ([summation operator]16 PAHs=1846 and 101 mg/kg before and after thermal desorption, respectively), the treatment did not eliminate the genotoxicity of soil pollutants to earthworms but increased it. The concentration of non-volatile metals did not change after thermal desorption. Among metals found in the treated soil, cadmium, chromium and nickel could explain the genotoxicity of the contaminated soil after thermal desorption. The treatment could increase the bioavailability and genotoxicity of heavy metals, through a modification of the soil's organic matter, the speciation of heavy metals and their binding to organic matter. This study underlines the importance of measuring biological effects, in order to evaluate the risk associated with formerly contaminated soils and the efficiency of remediation.

Six soil/waste samples were incubated at 28 °C under standard conditions to assess the impact of organic waste applications on heavy metal (HM) exchangeable fractions. The three studied soils are representative of the pedological diversity that prevails in Reunion and characterized by high natural total Cr, Cu, Ni and Zn concentrations. For organic waste, we tested two products that differed in their chemical composition, physical state and different HM contents, i.e. pig slurry (PS) and green waste compost (CT), which were incorporated at dosages consistent with the actual agricultural setting: 350 and 1000 kgN ha − 1 (subsequently called 350 and 1000). Variations in pH and electrical conductivity (EC) were monitored during incubation, while also measuring carbon mineralization and HM exchangeable fractions (Cr, Cu, Ni and Zn). The physicochemical parameter variation patterns were not identical in the three soils. The greatest variations were noted in the cambisol: pH reduction and EC increase. Conversely, in the andosol, there were only slight variations in pH and EC during incubation. The C–CO 2 quantities detected were derived from PS mineralization, and were always higher than those from the mineralization of CT, irrespective of the soil considered, and C mineralization was proportional to the initial quantity of incorporated waste (350 vs 1000). We explained the difference in the andosol mineralization coefficient between the 350 and 1000 dosages as being the result of an interaction between the organic constituents and the pseudo-cristallized aluminosilicates from the soil (allophane and/or imogolite), which limited degradation of the incorporated organic matter (OM). Despite organic waste mineralization, the pH variations recorded during the incubations and the increased salinity, we did not detect any increase in the HM exchangeable fractions, but instead noted a decrease in the Cu exchangeable fraction (soil + PS incubation), which could be explained by complexation of this element by the OM.

International audience; The comprehension of metal homeostasis in plants requires the identification of molecular markers linked to stress tolerance. Proteomic changes in leaves and cambial zone of Populus tremula×P. alba (717-1B4 genotype) were analyzed after 61 days of exposure to cadmium (Cd) 360 mg/kg soil dry weight in pot-soil cultures. The treatment led to an acute Cd stress with a reduction of growth and photosynthesis. Cd stress induced changes in the display of 120 spots for leaf tissue and 153 spots for the cambial zone. It involved a reduced photosynthesis, resulting in a profound reorganisation of carbon and carbohydrate metabolisms in both tissues. Cambial cells underwent stress from the Cd actually present inside the tissue but also a deprivation of photosynthates caused by leaf stress. An important tissue specificity of the response was observed, according to the differences in cell structures and functions.

A-09-41; International audience; We analysed variation in microbial community richness and function in soils associated with a fire-induced vegetation successional gradient from low maquis (shrubland) through tall maquis to rainforest on metal-rich ultramafic soils at Mt Do, New Caledonia. Random amplified polymorphic DNA fingerprinting was used to determine the extent of genetic relatedness among the microbial communities and indicated that the open and tall maquis microbial communities were more similar to each other than they were to the rainforest community. Sole-source carbon utilization indicated variation in the microbial communities, again with greater diversity in rainforest soils. Plate counts showed that both rainforest and maquis soils contained bacteria that can grow in the presence of up to 20 mmol L−1 nickel and 10 mmol L−1 chromium. Understanding microbial community composition and dynamics in these ultramafic soils may lead to a better understanding of the processes facilitating vegetation succession from shrubland to forest on these high-metal substrates, and of approaches to successful revegetation following mining for metals including nickel, chromium and cobalt.
