Background: Insights into (poly)phenol exposure represent a modifiable factor that may modulate inflammation in chronic pancreatitis (CP), yet intake is poorly characterized and methods for assessment are underdeveloped. Aims: The aims are to develop and test a method for estimating (poly)phenol intake from a 90-day food frequency questionnaire (FFQ) using the Phenol-Explorer database and determine associations with dietary patterns in CP patients versus controls via analysis of previously collected cross-sectional data. Methods: Fifty-two CP patients and 48 controls were recruited from an ambulatory clinic at a large, academic institution. To assess the feasibility of the proposed methodology for estimating dietary (poly)phenol exposure, a retrospective analysis of FFQ data was completed. Mann–Whitney U tests were used to compare (poly)phenol intake by group; Spearman correlations and multivariable-adjusted log-linear associations were used to compare (poly)phenol intakes with dietary scores within the sample. Results: Estimation of (poly)phenol intake from FFQs was feasible and produced estimates within a range of intake previously reported. Total (poly)phenol intake was significantly lower in CP vs controls (463 vs. 567mg/1000kcal; p = 0.041). In adjusted analyses, higher total (poly)phenol intake was associated with higher HEI-2015 (r = 0.34, p < 0.001), aMED (r = 0.22, p = 0.007), EDIH (r = 0.29, p < 0.001), and EDIP scores (r = 0.35, p < 0.001), representing higher overall diet quality and lower insulinemic and anti-inflammatory dietary potentials, respectively. Conclusions: Using enhanced methods to derive total (poly)phenol intake from an FFQ is feasible. Two new aryltetralin-type lignans (1-2) were isolated from the dichloromethane fraction of 95% ethanol extract of Camellia oleifera fruit husk. Their structures were elucidated on the basis of spectroscopic analysis, and the absolute configurations of compounds 1-2 were determined by the comparison of measured ECD curves with the quantum chemical calculated ones. The new compounds were tested for their antioxidant activities and cytotoxicity against three human cancer cell lines (Huh-7, H460 and MCF-7). While compounds 1 and 2 only showed slight DPPH radical scavenging activities with the IC50 values of 38.68 ± 5.02 and 56.62 ± 1.49 μM, respectively. [ABSTRACT FROM AUTHOR]

Main conclusion: The secondary metabolic conversion of monolignans to sesquilignans/dilignans was closely related to seed germination and seedling establishment in Arctium lappa. Arctium lappa plants are used as a kind of traditional Chinese medicines for nearly 1500 years, and so far, only a few studies have put focus on the key secondary metabolic changes during seed germination and seedling establishment. In the current study, a combined approach was used to investigate the correlation among secondary metabolites, plant hormone signaling, and transcriptional profiles at the early critical stages of A. lappa seed germination and seedling establishment. Of 50 metabolites in methonolic extracts of A. lappa samples, 35 metabolites were identified with LC–MS/MS and 15 metabolites were identified with GC–MS. Their qualitative properties were examined according to the predicted chemical structures. The quantitative analysis was performed for deciphering their metabolic profiles, discovering that the secondary metabolic conversion from monolignans to sesquilignans/dilignans was closely correlated to the initiation of A. lappa seed germination and seedling establishment. Furthermore, the critical transcriptional changes in primary metabolisms, translational regulation at different cellular compartments, and multiple plant hormone signaling pathways were revealed. In addition, the combined approach provides unprecedented insights into key regulatory mechanisms in both gene transcription and secondary metabolites besides many known primary metabolites during seed germination of an important traditional Chinese medicinal plant species. The results not only provide new insights to understand the regulation of key medicinal components of 'ARCTII FRUCTUS', arctiin and arctigenin at the stages of seed germination and seedling establishment, but also potentially spur the development of seed-based cultivation in A. lappa plants. The identification of antiplasmodial lead compounds from the dry seeds of Piper guineense has been performed using a bioassay-directed protocol. Briefly, its methanolic seeds extract has been suspended in water and partitioned using n -hexane, dichloromethane, ethyl acetate and n -butanol to afford four main fractions A−D which were further submitted to antiplasmodial assay against Plasmodium falciparum 3D7. The bioassay results showed that its dichloromethane-soluble fraction has the highest potency with 31.8 % of the parasite P. falciparum 3D7 survival. The purification of the active fraction with successive column chromatography led to the isolation and identification of eleven compounds including six piperine-type amides (1 − 6), two lignans (7 and 8), one fatty acid (9), and two steroids (10 and 11). The antiplasmodial assay of compounds 1 − 8 against the two strains Pf 3D7 and Pf Dd2 showed that only the alkaloids 1 − 6 displayed moderate potencies with the highest IC 50 values of 17.4 ± 0.7 µ g/mL (for 6 against Pf 3D7) and 14.6 ± 0.3 µ g/mL (for 2 against Pf Dd2). Furthermore, the cytotoxicity of the tested compounds against Vero cell lines showed that only compounds 5 and 6 displayed a cytotoxicity with CC 50 values of 335.4 ± 7.6 µ g/mL and 151.8 ± 2.4 µ g/mL, respectively, while their selectivity index limits an acceptable range. The results support the use of the plant in folk medicine for handling malaria, and demonstrate that piperine-type amides are responsible for the antiplasmodial activity of P. guineense and are good leads in the development of safe and potent antimalarial drugs. Lignan is a class of diphenolic compounds that arise from the condensation of two phenylpropanoid moieties. Oilseed and cereal crops (e.g., flaxseed, sesame seed, wheat, barley, oats, rye, etc.) are major sources of plant lignan. Methods for commercial isolation of the lignan secoisolariciresinol diglucoside (SDG) are not well reported, as most publications describing the detection, extraction, and enrichment of SDG use methods that have not been optimized for commercial scale lignan recovery. Simply scaling up laboratory methods would require expensive infrastructure to achieve a marketable yield and reproducible product quality. Therefore, establishing standard protocols to produce SDG and its derivatives on an industrial scale is critical to decrease lignan cost and increase market opportunities. This review summarizes the human health benefits of flaxseed lignan consumption, lignan physicochemical properties, and mammalian lignan metabolism, and describes methods for detecting, extracting, and enriching flaxseed lignan. Refining and optimization of these methods could lead to the development of inexpensive lignan sources for application as an ingredient in medicines, dietary supplements, and other healthy ingredients. Extra virgin olive oil (EVOO) is a symbol of the Mediterranean diet, constituting its primary source of fat. The beneficial effect of EVOO is strictly related to the presence of fatty acids and polyphenols, bioactive compounds endowed with nutraceutical properties. Among EVOO polyphenols, lignans possess a steroid-like chemical structure and are part of the phytoestrogen family, which is renowned for its health properties. The natural lignans (+)-pinoresinol and 1-acetoxypinoresinol (1-AP) are commonly present in olives and in EVOO. Although (+)-pinoresinol is found in different edible plants, such as flaxseed, beans, whole-grain cereals, sesame seeds, and certain vegetables and fruit, 1-AP was exclusively identified in olives in 2000. So far, the scientific literature has extensively covered different aspects of (+)-pinoresinol, including its isolation and nutraceutical properties. In contrast, less is known about the olive lignan 1-AP. Therefore, this review aimed to comprehensively evaluate the more important aspects of 1-AP, collecting all the literature from 2016 to the present, exploring its distribution in different cultivars, analytical isolation and purification, and nutraceutical properties. Polyphenols are ubiquitous plant metabolites that demonstrate biological activities essential to plant–environment interactions. They are of interest to plant food consumers, as well as to the food, pharmaceutical and cosmetic industry. The class of the plant metabolites comprises both widespread (chlorogenic acids, luteolin, quercetin) and unique compounds of diverse chemical structures but of the common biosynthetic origin. Polyphenols next to sesquiterpenoids are regarded as the major class of the Inuleae-Inulinae metabolites responsible for the pharmacological activity of medicinal plants from the subtribe (Blumea spp., Dittrichia spp., Inula spp., Pulicaria spp. and others). Recent decades have brought a rapid development of molecular and analytical techniques which resulted in better understanding of the taxonomic relationships within the Inuleae tribe and in a plethora of data concerning the chemical constituents of the Inuleae-Inulinae. The current taxonomical classification has introduced changes in the well-established botanical names and rearranged the genera based on molecular plant genetic studies. The newly created chemical data together with the earlier phytochemical studies may provide some complementary information on biochemical relationships within the subtribe. Moreover, they may at least partly explain pharmacological activities of the plant preparations traditionally used in therapy. Breast cancer is recognized as one of the most common cancers worldwide that can lead to death. Alternative treatment options are needed due to drug resistance caused by current treatment methods such as chemotherapy, inclusion of healthy cells in the target, and possible side effects. In this context, there is great interest in natural compounds and their active metabolites. One of these is flaxseed (FS), which is one of the most studied foods to be associated with breast cancer. FS is a functional food with high nutritional value. FS components (fatty acids, fiber, and lignans) and especially the basic lignan structure in FS content, secoisolariciresinol diglucoside (SDG) and its metabolites enterolactone (ENL) and enterodiol (END) have beneficial effects on breast cancer progression. This review aimed to develop a perspective for further research on this type of cancer in the future by giving some general information about FS and its components and evaluating some studies showing potential effects on breast cancer. Objective/Background: This study aims to explore the Kadsura coccinea extract (KCE)'s effect on lipid accumulation in vitro and its chemical components characterizations, aiming at developing a new alternative plant medicinal resource to fight against non-alcoholic fatty liver disease (NAFLD). Methods: After toxicological evaluation of KCE on HepG2 cells, Oil red O staining model and intracellular TGs quantification kit were used to examine the effects of KCE on lipid accumulation in vitro. The chemical components characterizations and potential active chemical constituents of the bioactive KCE were analyzed using ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) technology. Results: Among the chosen non-toxic concentrations of KCE (5, 10, 20 μg/mL), KCE can reduce the number and volume of lipid droplets in the oleic acid induced HepG2 cells in a dose-dependent manner, and the results of TGs quantification were almost consistent with the results of the oil red O staining experiment. These data indicate the KCE has ameliorative effect on lipid accumulation in vitro. In addition, a total of 26 compounds from the KCE were tentatively identified, dibenzocyclooctadiene lignans (DCLs) including Kadsulignan L and Gomisin J could be the main supposed components. Schisandra chinensis (Schisandraceae) is a medicinal plant widely used in traditional Chinese medicine. Under the name Wu Wei Zi, it is used to treat many diseases, especially as a stimulant, adaptogen, and hepatoprotective. Dibenzocyclooctadiene lignans are the main compounds responsible for the effect of S. chinensis. As a part of ongoing studies to identify and evaluate anti-inflammatory natural compounds, we isolated a series of dibenzocyclooctadiene lignans and evaluated their biological activity. Furthermore, we isolated new sesquiterpene 7,7-dimethyl-11-methylidenespiro[5.5]undec-2-ene-3-carboxylic acid. Selected dibenzocyclooctadiene lignans were tested to assess their anti-inflammatory potential in LPS-stimulated monocytes by monitoring their anti-NF-κB activity, antioxidant activity in CAA assay, and their effect on gap junction intercellular communication in WB-ras cells. Some S. chinensis lignans showed antioxidant activity in CAA mode and affected the gap junction intercellular communication. Secondary metabolites are specialized metabolic products synthesized by plants, insects, and bacteria, some of which exhibit significant physiological activities against other organisms. Plants containing bioactive secondary metabolites have been used in traditional medicine for centuries. In developed countries, one-fourth of medicines directly contain plant-derived compounds or indirectly contain them via semi-synthesis. These compounds have contributed considerably to the development of not only medicine but also molecular biology. Moreover, the biosynthesis of these physiologically active secondary metabolites has attracted substantial interest and has been extensively studied. However, in many cases, the degradation mechanisms of these secondary metabolites remain unclear. The biosynthesis and distribution of lignans in medicinal plants, particularly in Schisandra sphenanthera, hold significant pharmacological importance. This study bridges the knowledge gap in understanding the tissue-specific biosynthesis and distribution of these compounds, with a focus on Gomisin J. Our phytochemical analysis revealed a distinct accumulation pattern of Gomisin J, predominantly in the roots, contrasting with the distribution of Pregomisin and Dihydroguaiaretic acid. This finding highlights the roots' unique role in lignan storage and biosynthesis. Further, differential gene expression analysis across various tissues illuminated the biosynthetic pathways and regulatory mechanisms of these lignans. Utilizing Weighted Gene Co-expression Network Analysis (WGCNA), we identified the MEtan module as a key player, strongly correlated with Gomisin J levels. This module's in-depth examination revealed the crucial involvement of four cytochrome P450 (CYP) enzymes and eight transcription factors. Notably, the CYP genes DN6828 and DN2874-i3 exhibited up-regulation in roots across both male and female plants, while DN51746 was specifically up-regulated in male roots, indicating a potential gender-specific aspect in Gomisin J biosynthesis. Comparative analysis with functionally characterized CYP71A homologs suggests these CYP genes might be involved in distinct biosynthetic pathways, including terpenoids, alkaloids, and phenylpropanoids, and potentially in lignan biosynthesis. This hypothesis, supported by their more than 55% identity with CYP71As and strong correlation with Gomisin J concentration, opens avenues for novel discoveries in lignan biosynthesis, pending further functional characterization. Our research provides a comprehensive understanding of the genetic and metabolic mechanisms underlying the tissue-specific distribution of lignans in Schisandra sphenanthera, offering valuable insights for their pharmacological exploitation. Linum album Kotschy ex Boiss. as one of the endemic perennial plants in Iran is a natural source of lignan compounds. In the present study, the seeds of L. album were collected from its natural habitats in four regions of Iran (Aliabad, Jowkar, Dasht-e Arzhan, Taleqan). The effects of ecotype and gibberellic acid (GA) pretreatment were evaluated on germination traits of L. album seeds in a factorial based on completely randomized design with four replications. Also, the capacity of studied ecotypes in regard to secoisolariciresinol (SECO), podophyllotoxin (PTOX) and 6-methoxy-podophyllotoxin (6-MPTOX) production was assayed under in vitro condition by high performance liquid chromatography (HPLC) based on completely randomized design with four replications. In all studied ecotypes of L. album, seed germination percentage, germination rate and seedling growth was significantly improved in response to GA pretreatment. The obtained sterilized seedlings were used as in vitro explants that were successfully propagated in MS medium without any growth regulators. The in vitro proliferation rate in ecotype of Jowkar in terms of shoot number and plantlet biomass was significantly higher than the other ecotypes. The highest SECO and PTOX contents were extracted in the Dasht-e Arzhan plantlets which were higher (up to four times) than the other ecotypes. Also, the plantlets of Dasht-e Arzhan, Taleqan and Jowkar ecotypes showed the highest content of 6-MPTOX. According to germination, morphological and phytochemicals traits, the ecotype of Dasht-e Arzhan can be considered as a good candidate for breeding programs of improving PTOX production in this plant. The fruit of Forsythia suspensa (Thunb.) Vahl has been used in traditional Chinese medicine as "Forsythiae fructus". The species is also grown in parks and gardens, and on streets and building lots, as an ornamental plant, but it requires pruning. In this study, the allelopathic activity and allelopathic substances in the leaves of pruned branches of F. suspensa were investigated to determine any potential application. The leaf extracts of F. suspensa showed growth inhibitory activity against three weed species; Echinochloa crus-galli, Lolium multiflorum, and Vulpia myuros. Two allelopathic substances in the extracts were isolated through the bioassay-guided purification process, and identified as (-)-matairesinol and (-)-arctigenin. (-)-Matairesinol and (-)-arctigenin, which showed significant growth inhibitory activity at concentrations greater than 0.3 mM in vitro. The inhibitory activity of (-)-arctigenin was greater than that of (-)-matairesinol. However, both compounds were more active than (+)-pinolesinol which is their precursor in the biosynthetic pathway. The investigation suggests that F. suspensa leaves are allelopathic, and (-)-matairesinol and (-)-arctigenin may contribute to the growth inhibitory activities. Therefore, the leaves of the pruned branches can be applied as a weed management strategy in some agricultural practices such as using the leaf extracts in a foliar spray and the leaves in a soil mixture, thereby reducing the dependency on synthetic herbicides in the crop cultivation and contributing to developing eco-friendly agriculture. Protein, oil content, linoleic acid, and lignan are several key indicators for evaluating the quality of flaxseed. In order to optimize the testing methods for flaxseed's nutritional quality and enhance the efficiency of screening high-quality flax germplasm resources, we selected 30 flaxseed species widely cultivated in Northwest China as the subjects of our study. Firstly, we gathered hyperspectral information regarding the seeds, along with data on protein, oil content, linoleic acid, and lignan, and utilized the SPXY algorithm to classify the sample set. Subsequently, the spectral data underwent seven distinct preprocessing methods, revealing that the PLSR model exhibited superior performance after being processed with the SG smoothing method. Feature wavelength extraction was carried out using the Successive Projections Algorithm (SPA) and the Competitive Adaptive Reweighted Sampling (CARS). Finally, four quantitative analysis models, namely Partial Least Squares Regression (PLSR), Support Vector Regression (SVR), Multiple Linear Regression (MLR), and Principal Component Regression (PCR), were individually established. Experimental results demonstrated that among all the models for predicting protein content, the SG-CARS-MLR model predicted the best, with and of 0.9563 and 0.9336, with the corresponding Root Mean Square Error Correction (RMSEC) and Root Mean Square Error Prediction (RMSEP) of 0.4892 and 0.5616, respectively. In the optimal prediction models for oil content, linoleic acid and lignan, the R2 p was 0.8565, 0.8028, 0.9343, and the RMSEP was 0.8682, 0.5404, 0.5384, respectively. The study results show that hyperspectral imaging technology has excellent potential for application in the detection of quality characteristics of flaxseed and provides a new option for the future non-destructive testing of the nutritional quality of flaxseed. One new iridoid glucoside, 7- O - E -feruloylgardoside (1) and two new lignan glycosides, 8′-hydroxyphillyrin (2) and 6''-(p -hydroxybenzyl) phillyrin (3), together with three known compounds, khainaoside B (4), phillyrin (5), and lantibeside (6), were isolated from the whole plants of Lancea tibetica. Their structures were elucidated on the basis of spectroscopic methods and X-ray crystallographic analysis. A DPPH free radical scavenging assay showed that 1 and 4 exhibited moderate antioxidant activity (IC 50 45.05 ± 3.04 and 48.00 ± 3.29 μ g/ml) compared with vitamin C (IC 50 3.31 ± 0.16 μ g/ml). A functional set dahi (fermented milk analogous to yoghurt) with a desirable probiotic (Lactiplantibacillus plantarum A5) count of 9.36 log CFU/mL and excellent techno-functional attributes (DPPH: 41.95% RSA, firmness: 485.49 g, sensory overall acceptability: 8.51) was developed to contain 260 mg of SDG in 20 g of dahi. Twenty-four female Albino Wistar rats (3 months old, >180 g) were ovariectomized (OVX) and divided into three groups: OVX control, OVX and control dahi, and OVX and SDG-enriched dahi. The animal study found that ovariectomy decreased serum calcium, oestrogen, and bone ash calcium levels by 32.27, 30.95, and 48.46 percent, respectively, compared to the sham group (n = 8), while daily administration of SDG-enriched dahi (20 g) for eight weeks restored them. The proximal tibial metaphysis and distal femoral epiphysis micro-CT study showed that the ovariectomy lowered bone mineral density (BMD) by 11.06% and 9.18%, respectively, and lowered Trabecular thickness (Tb. Th) by 12.66% and 11.86%, respectively, while increasing Trabecular separation (Tb. Sp.) by 90.69% and 87.70%, respectively, compared to the sham control-group rats. SDG-enriched dahi improved BMD by 16.06 and 12.24% and Tb. Th by 35.32 and 19.62%, respectively, and decreased Tb. Sp by 47.04 and 47.22%, respectively, in OVX rats. The Phyllanthus genus exhibits a broad distribution spanning across the majority of tropical and subtropical regions. Due to their ability to synthesize medicinal bioactive compounds such as lignans, they have been utilized historically in traditional medicine to treat a wide range of ailments. This review discusses the current knowledge on the potency of lignans for medicinal purposes, the benefit of lignans for plants, various lignans produced by Phyllanthus, and how lignan synthesis could be increased through biotic and abiotic elicitation. Finally, we present a set of connected hypotheses to explain how signaling crosstalk between endophytic microbes and drought stress responses regulates lignan production. Although the mechanisms of lignan synthesis in Phyllanthus are not fully explored, this review strongly supports the view that endophytic fungi and drought stress can increase lignan production in plants belonging to the genus Phyllanthus. The medicinal plant–endophyte–drought stress relationship helps to improve the lignan yield of Phyllanthus, which is crucial for human health and can be optimized under in vitro and in vivo conditions. Background: There has been an increasing global prevalence of depression and other psychiatric diseases in recent years. Perceived stress has been proven to be associated with psychiatric and somatic symptoms. Some animal and human studies have suggested that consuming foods abundant in lignans and phytosterols may be associated with lower levels of stress, depression, and anxiety. Still, the evidence is not yet strong enough to draw firm conclusions. Thus, we investigated the association between dietary intake of these phytochemicals and the level of stress experienced by adult individuals. Methods: Diet was assessed using self-reported 7-day dietary records. The intakes of lignans and phytosterols were estimated using databases with their content in various food products. The Perceived Stress Scale (PSS) was implemented to measure the level of perceived stress. A logistic regression analysis was used to test for associations. Results: The odds of elevated PSS were negatively associated with dietary intake of total phytosterols, stigmasterol, and β-sitosterol, with evidence of a decreasing trend across tertiles of phytochemicals. The analysis for doubling the intake reinforced the aforementioned relationships and found protective effects against PSS for total lignans, pinoresinol, and campesterol. Conclusions: Habitual inclusion of lignans and phytosterols in the diet may play a role in psychological health. To address the global outbreak of depression and other mental health issues triggered by stress, it is important to take a holistic approach. There is a need to develop effective strategies for prevention and treatment, among which certain dietary interventions such as consumption of products abundant in lignans and phytosterols may play a substantial role. Two lignans (lariciresinol (1) and dehydrodiconiferyl alcohol (2)), and four monoterpenoids (vitexoid (3), 6-hydroxy-2,6-dimethyl-2,7-octadienoic acid (4), 6-hydroxy-2,6-dimethyloct-7-enoic acid (5), 2,6-dimethylocta-2,7-diene-1,6-diol (6)) from the male Carica papaya flowers are conducted a cytotoxic activity for lung cancer cells (A549), liver cancer cells (Hep3B), and breast cancer cells (MCF-7). The results, lignan (1), and monoterpenoid (3), (6) exhibited medium cytotoxic activity on these cancer cells with IC50 from 34.73±1.55 to 66.12±5.60 µg/mL. Lignan (2) displayed medium cytotoxic activity on MCF-7, Hep3B with IC50 60.85±4.43, 62.32± 5.03 µg/mL, respectively, and weak cytotoxic activity on A549 with IC50 70.99±7.26 µg/mL. Monoterpenoids (4), and (5) showed medium cytotoxic activity on MCF-7 with IC50 54.15±5.89, 67.49±2.41 µg/mL, respectively, and weak cytotoxic activity on A549, and Hep3B with IC50 from 72.25±3.13 to 86.03±7.57 µg/mL. ABSTRACTThis study was designed to investigate the amino acid profile, vitamin B complex, and bioactive properties present in recently cultivated rye variants in Pakistan. The four rye variants, named as Gp 1, Gp-2, Gp-3, and Gp-4, were procured from the Forage and Pulses section of AARI Faisalabad. The rye samples were ground into flour using a hammer-type 120-ton laboratory mill. For the analysis of amino acids, vitamin B complex, phenolic constituents, alkylresorcinols, and lignin, standard HPLC and GC-MS methods were employed following the extraction with appropriate reagents. The results revealed that among the rye variants, Gp-2 showed the highest content of vitamin B complex, with thiamin at 252.33 μg/100 g, niacin at 397 μg/100 g, and pyridoxine at 99 μg/100 g. Pantothenic acid (246.70 μg/100 g) and ascorbic acid (470 μg/100 g) were found in the highest concentrations in Gp-3. The riboflavin content of Gp-1 was 84.40 g/100 g, while the folate content of Gp-4 was 24.40 g/100 g. In terms of phenolic compounds, the range of chlorogenic acid content ranged from 86.70 to 92.43 mg/kg, p-coumaric acid from 2.70 to 5.40 mg/kg, gallic acid from 64.70 to 97.10 mg/kg, caffeic acid from 16.90 to 25.90 mg/kg, and coumaric acid from 218.10 to 228.40 mg/kg. In conclusion, Gp-2 observed as the most nutritionally rich and bioactive variant, distinguished by its high essential amino acids, vitamins, and other bioactive components.

Sesame (Sesamum indicum L.), an oilseed crop, is gaining worldwide recognition for its healthy functional ingredients as consumption increases. The content of lignans, known for their antioxidant and anti-inflammatory effects, is a key agronomic trait that determines the industrialization of sesame. However, the study of the genetics and physiology of lignans in sesame is challenging, as they are influenced by multiple genes and environmental factors, therefore, the understanding of gene function and synthetic pathways related to lignan in sesame is still limited. To address these knowledge gaps, we conducted genetic analyses using F7 recombinant inbred line (RIL) populations derived from Goenbaek and Gomazou as low and high lignin content variants, respectively. Using the QTL-seq approach, we identified three loci, qLignan1-1, qLignan6-1, and qLignan11-1, that control lignan content, specifically sesamin and sesamolin. The allelic effect between loci was evaluated using the RIL population. qLignan6-1 had an additive effect that increased lignan content when combined with the other two loci, suggesting that it could be an important factor in gene pyramiding for the development of high-lignan varieties. This study not only highlights the value of sesame lignan, but also provides valuable insights for the development of high-lignan varieties through the use of DNA markers in breeding strategies. Lignans are a large category of polyphenolic compounds that have low molecular weight and are widely distributed in the plant kingdom. They have been recognized for their potential antioxidizing and antiproliferative action. One of the most important lignans is cubebin which is abundantly isolated from the leaves and seeds of Piper cubeba and Piper nigrum. Cubebin possesses numerous biological actions such as antileukemic, trypanocidal, antimycobacterial, analgesic, anti-inflammatory, histamine antagonist, antifungal, and antispasmodic. This review discusses the in vitro and in vivo pharmacological studies on cubebin related to biochemistry and pharmacological applications and it ensures that it widely shows therapeutic potential. We expect that these therapeutic actions will set a new track in the formation of novel biological agents by the derivatization of cubebin. Lindera obtusiloba Blume has several activities, such as anti-inflammatory, anti-allergic, anti-tumor, anti-wrinkle, and antioxidant activities. Interleukin-6 (IL-6) is a classic pro-inflammatory cytokine that is associated with various functions, such as proliferation, invasion, inflammatory responses and functions within antioxidant defense systems. In this study, we investigated IL-6-induced STAT3 activation of lignan compounds isolated from L. obtusiloba. The structures of the isolated compounds were elucidated via spectroscopic nuclear magnetic resonance (NMR) and electrospray ionization mass spectrometry (ESI-MS). As a result, seven lignans were identified from L. obtusiloba. All the isolated compounds (1–7) were evaluated for their IL-6-induced STAT3 inhibitory effects in Hep3B cells using a luciferase reporter assay. Of the isolates, compounds 1 and 5 showed inhibitory effects against IL-6-stimulated STAT3 activation. Furthermore, the mRNA expression levels of inflammation-related genes such as CRP, IL-1b, and SOCS3 were significantly reduced by exposure to compounds 1 and 5. The protein levels of p-STAT3 and p-JAK2 in IL-6-induced U266 cells were regulated in the presence of lignans derived from Lindera obtusiloba by Western blot assay. Based on the results, this study of L. obtusiloba demonstrates that the species has promise as a bioactive candidate for the treatment of IL-6-induced STAT3-related disease. [ABSTRACT FROM AUTHOR]

Secoisolariciresinol diglucoside (SDG) and tracheloside (TCL) are the main lignan components of flaxseed cake and safflower seed cake, which are by‐products of oil extraction. Both SDG and TCL are metabolized into mammalian lignan enterolactone (EL) with the involvement of intestinal bacteria. In this research, we evaluated the anti‐osteoporosis effects of SDG and the in vivo metabolites EL and enterodiol (ED) prepared in our previous work, as well as the newly isolated chemical constituents from safflower seed, including TCL, the lactone ring opening product of TCL (OTCL) and two alkaloids on the alloxan‐induced zebrafish model. All the compounds showed significant anti‐osteoporosis effects at 80 µM, with p < 0.05 for EL and p < 0.001 for other compounds compared with the model. SDG and TCL showed the most significant and concentration‐dependent effects, with p < 0.001 compared with model at 20 µM. The alkaloids, N‐coumaroylserotonin glucoside and N‐feruloylserotonin glucoside, also showed anti‐osteoporosis at 20 µM with p < 0.01, whereas EL, ED, and OTCL showed no significant effects. Quantitative real‐time polymerase chain reaction revealed that SDG and TCL upregulated the expression of osteogenic genes Runx2, SP7, OPG, Col1a1a, Alp, ON, OPN, and OCN in alloxan‐treated zebrafish. The in vivo metabolite of lignans, EL, showed significant anti‐inflammatory effect (p < 0.01) at 20 µM, which might also help to combat osteoporosis and other complications caused by excessive immune response in the body. The results provided scientific data for using the oil extraction by‐products as sources of anti‐osteoporosis compounds. Tetrahydrofuran ring formation from dibenzylbutyrolactone lignans is a key step in the biosynthesis of aryltetralin lignans including deoxypodophyllotoxin and podophyllotoxin. Previously, Fe(II)- and 2-oxoglutarate-dependent dioxygenase (2-ODD) from Podophyllum hexandrum (Himalayan mayapple, Berberidaceae) was found to catalyze the cyclization of a dibenzylbutyrolactone lignan, yatein, to give deoxypodophyllotoxin and designated as deoxypodophyllotoxin synthase (DPS). Recently, we reported that the biosynthesis of deoxypodophyllotoxin and podophyllotoxin evolved in a lineage-specific manner in phylogenetically unrelated plant species such as P. hexandrum and Anthriscus sylvestris (cow parsley, Apiaceae). Therefore, a comprehensive understanding of the characteristics of DPSs that catalyze the cyclization of yatein to deoxypodophyllotoxin in various plant species is important. However, for plant species other than P. hexandrum , the isolation of the DPS enzyme gene and the type of the enzyme, e.g. whether it is 2-ODD or another type of enzyme such as cytochrome P-450, have not been reported. In this study, we report the identification and characterization of A. sylvestris DPS (AsDPS). Phylogenetic analysis showed that AsDPS belonged to the 2-ODD superfamily and shared moderate amino acid sequence identity (40.8%) with P. hexandrum deoxypodophyllotoxin synthase (PhDPS). Recombinant protein assay indicated that AsDPS and PhDPS differ in terms of the selectivity of substrate enantiomers. Protein modeling using AlphaFold2 and site-directed mutagenesis indicated that the Tyr305 residue of AsDPS probably contributes to substrate recognition. This study advances our understanding of the podophyllotoxin biosynthetic pathway in A. sylvestris and provides new insight into 2-ODD involved in plant secondary (specialized) metabolism. Multiple varieties of flaxseeds have been identified in the world, yet the relationship between these varieties, their agronomic traits, and their seeds' quality remains unclear. This study aimed to determine the level of lignan, vitamins and carotenoids in 40 selected flaxseed varieties, and the relationship between varieties, agronomic traits, and seed quality was investigated. In this study, notably, fiber flax variety No. 225 exhibited the highest lignan content among all tested seeds. Additionally, oil variety No. 167 demonstrated the highest level of α-tocotrienol (α-T3), β-tocopherol (β-T), γ-tocotrienol (γ-T3), and β-carotene (β-Car.). Conversely, intermediate flax variety No. 16 displayed the highest content of α-tocopherol (α-T), but lowest content of lutein (Lut.), zeaxanthin (Zea.), β-carotene (β-Car.), and total carotenoids (Total Car.). Furthermore, a correlation was observed between petal color with the lignan, while a strong correlation has been explored in seed yield, seed type, plant natural height, and fiber content in straw. Fruits of Schisandrachinensis, an East Asian liana plant, are currently more and more used to produce nutrient supplements that positively affect human health due to the content of various secondary metabolites. On the other hand, these substances because of their bioactivity can cause possible allelopathic or toxic effects concerning other organisms (algae, plants, animals). But the ecotoxicological properties of S. chinensis outside its area of origin have yet to be sufficiently verified. Two crustaceans, Daphnia magna and Thamnocephalus platyurus, were selected as model aquatic organisms to test the potential impact of S. chinensis active compounds on the aquatic environment. Crude water extract from S. chinensis fruits, simulating the natural leakage of active substances in water, was tested in treatments from 0.0045 to 45 mg/L (according to the content of schisandrin as the dominating lignan). Effective concentration (EC50) causing 50% lethal effect for D. magna was established to 0.0448 mg/L after 24 h and 0.0152 mg/L after 48 h. EC50 for T. platyurus reached 0.4572 mg/L after 24 h, i.e. more than ten times higher than for D. magna. This study showed that the potential environmentally relevant concentrations of S. chinensis bioactive compounds could represent a severe risk to aquatic ecosystems. [ABSTRACT FROM AUTHOR]

Major listeriosis outbreaks have been associated with fresh produce contaminated with Listeria monocytogenes. Strains that synthesize the Pss exopolysaccharide (EPS) have an estimated 102 to 104-fold advantage over nonsynthesizing strains in causing listeriosis. They more readily attach to the surfaces of fruit and vegetables forming EPS-biofilms that better withstand stresses associated with produce storage and consumption. Here, we show that the threat to fresh produce safety posed by the listerial EPS-biofilms may be countered by broadly available maple products. We serendipitously discovered that aqueous extracts of wood from several Acer (maple) and Carya (pecan, hickory) species inhibit the formation of listerial EPS-biofilms without affecting bacterial viability. One active ingredient in maple wood was identified as nortrachelogenin-8'-O-β-D-glucopyranoside (NTG). At 120 μM, this lignan decreased colonization of the EPS-synthesizing L. monocytogenes on cantaloupe pieces by approximately 150-fold, and on cut celery and lettuce by 10 to 11-fold. Another lignan, lariciresinol, which is abundant in a common food sweetener, maple syrup, had antibiofilm activity comparable to that of NTG. Diluted in the range of 1:200 to 1:800 maple syrup from two random manufacturers prevented formation of listeiral EPS-biofilms. Importantly, not only did maple products drastically decrease colonization of fresh produce by the EPS-synthesizing strains, they also decreased, by 6 to 30-fold, colonization by the L. monocytogenes strains that do not synthesize measurable EPS, including strains from the infamous 2011 cantaloupe listeriosis outbreak. Inhibition of surface colonization by various listerial strains, broad availability of maple sap and syrup as well as maple lumber processing waste position maple products as potential antibiofilm agents for protecting fresh produce from L. monocytogenes. Ferula sinkiangensis K. M. Shen (Apiaceae) is distributed in arid desert areas of Xinjiang, and its resin is a traditional Chinese medicine to treat gastrointestinal digestive diseases. To explore bioactive components from F. sinkiangensis, three new lignans and thirteen known components were isolated. The structural elucidation of the components was established utilizing spectroscopic analyses together with ECD calculations. Griess reaction results indicated new compounds 1 and 2 significantly decreased NO production in LPS-stimulated RAW 264.7 macrophages, and ELISA results indicated that they effectively attenuated LPS-induced inflammation by inhibiting TNF-α, IL-1β, and IL-6 expressions. The in silico approach confirmed that compound 1 docked into the receptors with strong binding energies of −5.84~−10.79 kcal/mol. In addition, compound 6 inhibited the proliferation of AGS gastric cancer cells with IC50 values of 15.2 μM by suppressing the cell migration and invasion. The investigation on the fruits of Pittosporum elevaticostatum Chang et Yan led to the isolation of two undescribed metabolites (1 – 2, named pitelevatin A and B) and twelve known ones (3 – 14), including one phthalide (1), four monoquiterpenoids (2 – 5), two sesquiterpenoids (6 – 7), two diterpenoids (8 – 9), and five phenolic analogues (10 – 14). The structures of 1 and 2 were elucidated based on spectroscopic methods, including high resolution electrospray ionization mass spectroscopy (HR-ESI-MS), one demension (1D) and 2D nuclear magnetic resonance (NMR), ultraviloet (UV), electronic circular dichroism (ECD) spectra and experimental and calculated specific optical rotation (SOR). Compounds 1 – 3 , 7 and 11 showed significant nitric oxide (NO) production inhibitory activity in lipopolysaccharide (LPS)-induced BV-2 microglial cells by inhibiting the expression of nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), with IC 50 values ranging from 6.35 to 25.75 μ M. ABSTRACT The gut microbial ecosystem underlies physiological relationships between the gut and distal organs. Mechanisms remain elusive but rely at least partially on the production of a diverse set of absorbable metabolites and host gene expression regulation. Here we show that in female mice, gut cecal microbiota profiles are related to microRNAs (miRNAs) expressed in the mammary gland. A subset of these miRNAs were found to regulate genes involved in breast cancer-related processes, such as cell proliferation and migration. To determine if these relationships could be exploited toward the reduction of breast cancer risk, we studied if they are modifiable by dietary flaxseed (FS), a source of lignan secoisolariciresinol diglucoside (SDG) and alpha-linolenic acid (ALA)-rich oil (FSO), both with antitumor effects. Importantly, SDG, but not ALA, needs microbial processing to release bioactive metabolites. We found that the microbiota and mammary gland miRNA are related, and FS modifies these relationships toward an antioncogenic phenotype. FSO- and SDG-related miRNAs were found to be involved in different pathways and neither FSO nor SDG alone could recapitulate the effects of whole FS, affecting unique pathways related to extracellular matrix processing. These findings highlight the existence of inter-organ microbiota-miRNA relationships, show that dietary interventions interact to affect them, and suggest a novel route for breast cancer prevention. IMPORTANCE Breast cancer is a leading cause of cancer mortality worldwide. There is a growing interest in using dietary approaches, including flaxseed (FS) and its oil and lignan components, to mitigate breast cancer risk. Importantly, there is recognition that pubertal processes and lifestyle, including diet, are important for breast health throughout life. Mechanisms remain incompletely understood. Our research uncovers a link between mammary gland miRNA expression and the gut microbiota in young female mice. We found that this relationship is modifiable via a dietary intervention. Using data from The Cancer Genome Atlas, we also show that the expression of miRNAs involved in these relationships is altered in breast cancer in humans. These findings highlight a role for the gut microbiome as a modulator, and thus a target, of interventions aiming at reducing breast cancer risk. They also provide foundational knowledge to explore the effects of early life interventions and mechanisms programming breast health.

Extra virgin olive oil (EVOO) is a symbol of the Mediterranean diet, constituting its primary source of fat. The beneficial effect of EVOO is strictly related to the presence of fatty acids and polyphenols, bioactive compounds endowed with nutraceutical properties. Among EVOO polyphenols, lignans possess a steroid-like chemical structure and are part of the phytoestrogen family, which is renowned for its health properties. The natural lignans (+)-pinoresinol and 1-acetoxypinoresinol (1-AP) are commonly present in olives and in EVOO. Although (+)-pinoresinol is found in different edible plants, such as flaxseed, beans, whole-grain cereals, sesame seeds, and certain vegetables and fruit, 1-AP was exclusively identified in olives in 2000. So far, the scientific literature has extensively covered different aspects of (+)-pinoresinol, including its isolation and nutraceutical properties. In contrast, less is known about the olive lignan 1-AP. Therefore, this review aimed to comprehensively evaluate the more important aspects of 1-AP, collecting all the literature from 2016 to the present, exploring its distribution in different cultivars, analytical isolation and purification, and nutraceutical properties.

The effect of microwave pretreatment of sesame seeds on the flavor, sensory quality, nutrition, and safety of sesame oil was systematically studied to provide a theoretical foundation for the application of microwave for aroma enhancement in sesame oil. Sesame oil samples were hydraulically pressed from sesame seeds microwaved at different powers (0, 180, 360, 540, 720 and 900 W) for 6 min. A total of 91 aroma active compounds were identified in sesame oil by solvent-assisted flavor evaporation and headspace solid-phase microextraction combined with gas chromatography-olfactometry-mass spectrometry. The total concentration of heterocyclic compounds including pyrazines, pyrroles, pyridines, thiazoles, thiophenes, and furans in sesame oil were positively correlated with microwave power, while the total concentration of carbonyl compounds including aldehydes, ketones, and esters increased initially and decreased later with an increase in microwave power. The intensities of roasted sesame-like, nutty, burnt and smoky aromas, and the bitter taste of sesame oil gradually increased, while the intensities of earthy, woody, and green odors decreased. The oil yield of microwave-pretreated sesame seeds (30.02%–31.93%), which was higher than that of raw sesame seeds (24.89%), was positively correlated with microwave power. Microwave pretreatment increased the nitrogen solubility index (NSI, from 11.7% to 18.07%) of sesame meal and the concentration of tocopherol in sesame oil (from 392.18 to 462.92 mg/kg). The highest NSI and tocopherol content appeared after 540 and 700 W microwave pretreatment, respectively. The concentrations of sesamin (13.01–15.78 mg/100 g), heterocyclic amines (HCAs, 33.19–143.88 ng/g), and polycyclic aromatic hydrocarbons (PAHs, 0.40–9.03 ng/g) in sesame oil were positively correlated with microwave power. The concentrations of PAHs and HCAs in sesame oil did not exceed the Chinese national standard limit. In summary, appropriate microwave pretreatment can provide sesame oil with a rich flavor, improve its nutritional value and oxidation stability, and increase the utilization value of sesame meal protein. Therefore, microwave pretreatment is highly advantageous and promising for application in aroma enhancement in sesame oil l and high-value utilization of sesame meal protein.

The quality of Schisandra chinensis (Turcz.) Baill. (S. chinensis) is principally attributed to lignan compounds. In this paper, a simple and rapid strategy for simultaneous extraction and determination of 10 lignans from S. chinensis was established through matrix solid-phase dispersion (MSPD) assisted by diol-functionalized silica (Diol). The experimental parameters for MSPD extraction were screened using the response surface methodology (RSM). Diol (800 mg) was used as a dispersant and methanol (MeOH, 85%, v/v) as an eluting solvent (10 mL), resulting in a high extraction efficiency. MSPD extraction facilitated the combination of extraction and purification in a single step, which was less time-consuming than and avoided the thermal treatment involved in traditional methods. The simultaneous qualification and quantification of 10 lignans was achieved by combining MSPD and high-performance liquid chromatography (HPLC). The proposed method offered good linearity and a low limit of detection starting from 0.04 (schisandrin C) to 0.43 μg/mL (schisantherin B) for lignans, and the relative standard deviation (RSD, %) values of precision were acceptable, with a maximum value of 1.15% (schisantherin B and schisanhenol). The methodology was successfully utilized to analyze 13 batches of S. chinensis from different cultivated areas of China, which proved its accuracy and practicability in the quantitative analysis of the quality control of S. chinensis. A new lignan, named 4'-methoxy-neotaiwanensol (1), together with five known compounds (2−6) were isolated from the leaves of the plant Piper sarmentosum. The known compounds consist of a phenylpropanoid (2), a flavonoid (3), two acyclic monoterpenoids (4 and 5), and a phenylacetic acid derivative (6). Structure elucidation of these compounds was conducted by detailed interpretation of the spectral data including NMR and HRESIMS data. Compounds 4 and 5 are new natural products and the NMR data in methanol-d4 were reported for the first time. The other known compounds were elucidated to be hydroxychavicol (2), hispidulin (3), 4-hydroxy-benzeneacetic acid (6). The extractive contents of three softwood species largely used in the wood industry, namely Abies alba (Silver fir), Picea abies (spruce) and Pseudotsuga menziesii (Douglas fir), have been determined quantitatively for knots and at different points chosen along their branches, before analysis using high-performance liquid chromatography coupled with Mass Spectrometry (HPLC-MS). The results indicated that branchwood samples located in close proximity to the stem present high contents of extractives similar to those recorded for the knots. HPLC analysis showed quite similar chemical compositions, indicating that first cm of the branches could be considered as an additional source of knotwood. The antibacterial, antifungal and antioxidant activities of knot's extractives have been investigated with the dual objective of better understanding the role of high levels of secondary metabolites present in the knot and evaluating their potential for biorefinery applications. The antioxidant activity study showed that crude extracts of Douglas fir knotwood presented higher radical scavenging activity levels than the extracts of Silver fir and spruce, which presented more or less the same activities. Silver fir and spruce knotwood extracts presented higher antibacterial activity levels than the Douglas fir knotwood extracts did, while Douglas fir knotwood extracts presented more fungal growth inhibition than the spruce and fir knotwood extracts did. The structure–activity relationships indicate that radical scavenging and antifungal activities are associated with a higher relative quantity of flavonoids in the crude extracts, while higher relative quantities of lignans are associated with antibacterial activity. One new lignan, julibrissinoside II, along with thirteen known compounds, was isolated from the stem bark of Albizia julibrissin. The structure of julibrissinoside II was determined on the basis of extensive spectroscopic methods, including NMR and CD spectroscopic data. The isolated compounds were tested for their SREBP-1c inhibitory activity at different concentrations using mouse hepatocyte AML12 cell lines. Most phenylpropanoid pathway flux is directed toward the production of monolignols, but this pathway also generates multiple bioactive metabolites. The monolignols coniferyl and sinapyl alcohol polymerize to form guaiacyl (G) and syringyl (S) units in lignin, components that are characteristic of plant secondary cell walls. Lignin negatively impacts the saccharification potential of lignocellulosic biomass. Although manipulation of its content and composition through genetic engineering has reduced biomass recalcitrance, in some cases, these genetic manipulations lead to impaired growth. The reduced-growth phenotype is often attributed to poor water transport due to xylem collapse in low-lignin mutants, but alternative models suggest that it could be caused by the hyper-or hypoaccumulation of phenylpropanoid intermediates. In Arabidopsis thaliana, overexpression of FERULATE 5-HYDROXYLASE (F5H) shifts the normal G/S lignin ratio to nearly pure S lignin and does not result in substantial changes to plant growth. In contrast, when we overexpressed F5H in the low-lignin mutants cinnamyl dehydrogenase c and d (cadc cadd), cinnamoyl-CoA reductase 1, and reduced epidermal fluorescence 3, plant growth was severely compromised. In addition, cadc cadd plants overexpressing F5H exhibited defects in lateral root development. Exogenous coniferyl alcohol (CA) and its dimeric coupling product, pinoresinol, rescue these phenotypes. These data suggest that mutations in the phenylpropanoid pathway limit the biosynthesis of pinoresinol, and this effect is exacerbated by overexpression of F5H, which further draws down cellular pools of its precursor, CA. Overall, these genetic manipulations appear to restrict the synthesis of pinoresinol or a downstream metabolite that is necessary for plant growth. [ABSTRACT FROM AUTHOR]

Investigation on the chemical components of Valeriana jatamansi Jones (Caprifoliaceae), a new lignan with pyran-ring, dipsalignan G (1), along with eight known compounds (2–9) were isolated. Their structures were elucidated by extensive analysis of 1D, 2D NMR and HR-ESI-MS spectroscopic data. Additionally, possible biosynthetic pathway of 1 was proposed. Finally, biological evaluation results showed that 8 had significant scavenging ability to ABTS and DPPH free radicals, with IC50 values of 1.35 ± 0.01 and 2.94 ± 0.01 μg/ml, respectively.
