Introduction: Although international clinical practice guidelines recognize a continued role for menopausal hormone therapy (HT), particularly for symptomatic women

Reproduction is essential for perpetuation of the species and, hence, is controlled by a sophisticated network of regulatory factors of central and peripheral origin that integrate at the hypothalamic-pituitary-gonadal (HPG) axis. Among the central regulators of reproduction, kisspeptins, as major stimulatory drivers of gonadotropin-releasing hormone (GnRH) neurosecretion, have drawn considerable interest in the last decade. However, the dynamic, if not cyclic (in the female), nature of reproductive function and the potency of kisspeptins and other stimulatory signals of the HPG axis make tenable the existence of counterbalance inhibitory mechanisms, which may keep stimulation at check and would allow adaptation of reproductive maturation and function to different endogenous and environmental conditions. In this context, discovery of the gonadotropin-inhibitory hormone (GnIH) in birds, and its mammalian homolog, RFRP, opened up the exciting possibility that this inhibitory signal might operate centrally to suppress, directly or indirectly, GnRH/gonadotropin secretion, thus reciprocally cooperating with other stimulatory inputs in the dynamic regulation of the reproductive hypothalamic pituitary unit. After more than 15 years of active research, the role of GnIH/RFRP in the control of the HPG axis has been documented in different species. Yet, important aspects of the physiology of this system, especially regarding its relative importance and actual roles in the control of key facets of reproductive function, remain controversial. In the present work, we aim to provide a critical review of recent developments in this area, with special attention to studies in rodent models, using pharmacological tools and functional genomics. In doing so, we intend to endow the reader with an updated view of what is known (and what is not known) about the physiological role of GnIH/RFRP signaling in the control of mammalian reproduction.

An experiment was conducted using indigenous Kivircik ewes to evaluate the effect of intravaginal progestagen sponges, containing 30 mg of fluorogestone acetate (FGA), followed by administration of pregnant mare serum gonadotrophin (PMSG) on inducing synchronized oestrus in the season and fertility. Three times of PMSG administration relative to sponge withdrawal (24 h before (n = 30), at (n = 29) or 24 h after (n = 29)) and two routes of PMSG administration (intramuscular (n = 46) and subcutaneous (n = 42) were compared for estrous response, number of multiple births and fecundity rates. There were no significant differences in terms of estrous response, due to differences in the time and route of PMSG administration. Background In mammals, pheromones play an important role in social and innate reproductive behavior within species. In rodents, vomeronasal receptor type 1 (V1R), which is specifically expressed in the vomeronasal organ, is thought to detect pheromones. The V1R gene repertoire differs dramatically between mammalian species, and the presence of species-specific V1R subfamilies in mouse and rat suggests that V1R plays a profound role in species-specific recognition of pheromones. In ruminants, however, the molecular mechanism(s) for pheromone perception is not well understood. Interestingly, goat male pheromone, which can induce out-of-season ovulation in anestrous females, causes the same pheromone response in sheep, and vice versa, suggesting that there may be mechanisms for detecting "inter-species" pheromones among ruminant species. Results We isolated 23 goat and 21 sheep intact V1R genes based on sequence similarity with 32 cow V1R genes in the cow genome database. We found that all of the goat and sheep V1R genes have orthologs in their cross-species counterparts among these three ruminant species and that the sequence identity of V1R orthologous pairs among these ruminants is much higher than that of mouse-rat V1R orthologous pairs. Furthermore, all goat V1Rs examined thus far are expressed not only in the vomeronasal organ but also in the main olfactory epithelium. Conclusion Our results suggest that, compared with rodents, the repertoire of orthologous V1R genes is remarkably conserved among the ruminants cow, sheep and goat. We predict that these orthologous V1Rs can detect the same or closely related chemical compound(s) within each orthologous set/pair. Furthermore, all identified goat V1Rs are expressed in the vomeronasal organ and the main olfactory epithelium, suggesting that V1R-mediated ligand information can be detected and processed by both the main and accessory olfactory systems. The fact that ruminant and rodent V1Rs have distinct features suggests that ruminant and rodent V1Rs have evolved distinct functions.

International audience; We determined whether kisspeptin could be used to manipulate the gonadotropin axis and ovulation in sheep. First, a series of experiments was performed to determine the gonadotropic responses to different modes and doses of kisspeptin administration during the anestrous season using estradiol-treated ovariectomized ewes. We found that: 1) injections (iv) of doses as low as 6 nmol human C-terminal Kiss1 decapeptide elevate plasma LH and FSH levels, 2) murine C-terminal Kiss1 decapeptide was equipotent to human C-terminal Kiss1 decapeptide in terms of the release of LH or FSH, and 3) constant iv infusion of kisspeptin induced a sustained release of LH and FSH over a number of hours. During the breeding season and in progesterone-synchronized cyclical ewes, constant iv infusion of murine C-terminal Kiss1 decapeptide-10 (0.48 mu mol/h over8h) was administered 30h after withdrawal of a progesterone priming period, and surge responses in LH occurred within 2 h. Thus, the treatment synchronized preovulatory LH surges, whereas the surges in vehicle-infused controls were later and more widely dispersed. During the anestrous season, we conducted experiments to determine whether kisspeptin treatment could cause ovulation. Infusion (iv) of 12.4 nmol/h kisspeptin for either 30 or 48 h caused ovulation in more than 80% of kisspeptin-treated animals, whereas less than 20% of control animals ovulated. Our results indicate that systemic delivery of kisspeptin provides new strategies for the manipulation of the gonadotropin secretion and can cause ovulation in noncyclical females.

Although exocytosis is the major mechanism by which cells secrete products into their environment, little is known about the mechanism of this fundamental process. Previous studies on the regulation of luteinizing hormone (LH) exocytosis have used intact cells exclusively. It is not possible, however, to determine the precise requirements for exocytosis in intact cells since the cytosol is not directly accessible. Permeabilization of the plasma membrane allows experimental manipulation of the intracellular milieu while preserving the exocytic apparatus. The diameter of the atoxin pores (2-3 nm) allowed the exchange of small molecules such as ATP while larger cytosolic proteins such as lactate dehydrogenase were retained. Because of the slow exchange of small molecules through a-toxin pores a protocol was developed which combines prolonged pre-equilibration of the permeabilized cells at 0°C before stimulation with strong Ca²⁺ buffering. Under these conditions an increase in the [Ca²⁺]free stimulated a 15-20 fold increase in LH exocytosis (EC₅₀ pCa 5.5). After 12-15 minutes the rate of exocytosis declined and the cells became refractory to Ca²⁺. At resting [Ca²⁺]free (pea 7), cAMP stimulated a rapid, 2 - 3 fold, increase in LH exocytosis. cAMP caused a modest enhancement of Ca²⁺-stimulated LH exocytosis by causing a left shift in the EC₅₀ for Ca²⁺ from pCa 5.6 to pCa 5.9. Activation of protein kinase C (PKC) with phorbol 12-myristate 13-acetate (PMA) synergistically enhanced cAMP-stimulated LH exocytosis, an effect which was further augmented by increasing the [Ca²⁺]free· Gonadotrophin-releasing hormone (GnRH) was found to stimulate cAMP production in intact pituitary cells. Since previous studies have shown that GnRH activates PKC and stimulates a rise in cytosolic [Ca²⁺]free, these results suggest that a synergistic interaction of the cAMP, PKC and Ca²⁺ second messenger systems is of importance in the mechanism of GnRH-stimulated LH exocytosis. When permeabilized cells were equilibrated for prolonged periods in the absence of MgATP, Ca²⁺-stimulated LH exocytosis declined. The time course of the decline closely followed the leakage of intracellular ¹⁴C-ATP. Addition of MgATP rapidly restored full Ca²⁺-stimulated LH exocytosis. Ca²⁺-, cAMP-, and PMA-stimulated LH exocytosis were all dependent on millimolar MgATP concentrations (EC₅₀ 1 .5-3 mM). It has been postulated that PKC is a mediator of Ca²⁺- stimulated exocytosis. Several findings in the present study argue against this hypothesis. Firstly, PMA and Ca²⁺ had additive effects on LH exocytosis at all [Ca²⁺]free· Secondly, PMA was able to stimulate further LH release from cells made refractory to high [Ca²⁺]free· Thirdly, the PKC inhibitor staurosporine did not inhibit Ca²⁺-stimulated LH exocytosis under conditions in which it inhibited PMAstimulated exocytosis. Fourthly, in cells desensitized to PMA by prolonged exposure to a high PMA concentrations, Ca²⁺-stimulated LH exocytosis was not inhibited. And finally, Ba²⁺+ was able to stimulate LH exocytosis to a maximal extent similar to Ca²⁺ despite the fact that Ba²⁺+ is an extremely poor activator of PKC. Since Ba²⁺+ is also a poor activator of calmodulin, this latter result implies that calmodulin does not mediate the effect of Ca²⁺. In agreement with this, the calmodulin inhibitor calmidazolium did not inhibit Ca²⁺-stimulated LH exocytosis. Since GTP-binding proteins have been implicated in regulated exocytosis in other cell systems, the effects of guanine nucleotides on LH exocytosis were examined. At resting cytosolic [Ca²⁺]free (pea 7), the GTP analogues GTPyS and GMPPNP stimulated LH exocytosis with similar potencies (EC₅₀ 20-50 μM). Additional experiments indicated that the effects of these GTP analogues could not be explained by activation of either PKC alone or cAMP-dependent protein kinase alone. In the presence of both PMA and cAMP, GMPPNP did not stimulate a further increase in the rate of LH exocytosis, suggesting that the stimulatory actions of guanine nucleotides may be mediated by the combined activation of PKC and generation of cAMP, as a result of activation of signal-transducing G proteins. In contrast, pretreatment of cells with GTPyS at low [Ca²⁺]free markedly inhibited subsequent responses to Ca²⁺, cAMP, PMA, and cAMP plus PMA. This inhibitory effect required lower GTPyS concentrations than the stimulatory effect (IC₅₀ 1-10 μM), and was not observed with GMPPNP. These findings indicate the involvement of a distinct guanine nucleotide-binding protein in exocytosis at a site distal to second messenger generation.
