The problem solved is to determine safer and more effective technologies for post-harvest processing of fruits. To achieve the goal, lemon fruits (of different degrees of wilting) were cut crosswise, each piece was treated with miramistin and deсasan in concentrations of 0.1 %, 0.3 %, and 0.5 %. The samples were placed on agar blocks with pure cultures of pathogens and placed in wet Petri dishes, kept at a temperature of 25 °C in a thermostat for 6–10 days; the experiment was repeated three times. The research aimed to establish the concentration of antimicrobial drugs for treating lemons; determine the weight loss of lemon fruits depending on the storage temperature. The object of the study is the process of preserving the quality of lemon fruits during post-harvest treatment with antiseptics during cold storage. It is proposed to treat lemon fruits with miramistin and deсasan in concentrations of 0.3 % and 0.5 % in order to inhibit the development of blue and green mold during storage. Damage to turgor fruits is two times less than that of weakly wilted ones. At a storage temperature of 10 °C, fruits are stored for 40–45 days. At the same time, daily losses are 0.49 %. Lowering the storage temperature to 4 °C extends the storage period to 90–100 days, with a daily mass loss of 0.08 %. A curvilinear correlation dependence of the second order of weight loss of lemon fruits on the duration of storage was established (R2=0.9758–0.9903). Treatment with antiseptics makes it possible to implement environmentally friendly solutions that will make it possible to exclude chemically synthesized fungicides and preserve the natural properties of fresh citrus fruits after harvesting, as well as extend their shelf life. Organic farming is growing rapidly worldwide since it is perceived as more respectful of the environment than conventional farming. In this sense, organic agriculture is highly appreciated by consumers since consumers around the world believe that organic food has a higher content of beneficial compounds for health and consider it of higher quality. For that reason, the objective of this research was to evaluate the nutritional, sensorial, and functional quality of the 'Fino 49' lemon grafted on Citrus macrophylla in conventional and organic cultivation. Fatty acids, amino acids, total phenol, and polyphenols were quantified, antioxidant activity was measured, and sensory descriptive analysis was performed. Conventional farming led to an increase in amino acid content (641 mg L−1) and an increase in polyunsaturated fatty acids (254 mg 100 g−1) and monounsaturated fatty acids (37.61 mg 100 g−1). On the other hand, organically produced lemon fruits had better sensory profile (highlighting overall aroma (6.5), lemon odor (6.8), sourness (5.8), floral (0.6), and fresh lemon flavor (9.8)), and lower thrombogenicity index (0.15). The type of cultivation (organic and conventional) had no influence on the antioxidant activity (~1.60, ~3.08, and ~4.16 mmol Trolox L−1 for ABTS+, DPPH•, and FRAP, respectively) and polyphenols content (85.51 and 86.69 conventional and organic, respectively). Together with lime, lemon is recognized as the third most extensively cultivated citrus species worldwide, trailing only behind orange and mandarin. The enhancement in the economic value of lemons as a valuable resource in Southern Italy depends by the competitiveness of local lemon production in both domestic and international markets. This necessitates conducting a comprehensive study that thoroughly explores the available indigenous germplasm at the local and national levels. This study aims to identify the most promising Sicilian and Campanian cultivars that are well suited for both fresh fruit consumption and the production of processed goods. This re-study assumes even greater importance considering the ongoing climate changes, as environmental stresses significantly impact the ripening process and the timing of fruit development. Our study has highlighted a notable diversity among the 18 investigated lemon cultivars, particularly highlighting specific cultivars that possess desirable attributes for fresh consumption. The cultivars that showed the greatest cumulative production over the 10 years of the study were Erice with 467.89 kg/tree and Femminello Siracusano 2KR with 408.44 kg. Notably, cultivars like Segesta, Erice, and Kamarina have exhibited higher percentages of juice content ranging from 27.30% to 31.08%. These cultivars show great potential for abundant juice content and optimal acidity levels for direct consumption. On the other hand, cultivars characterized by enhanced yield, such as Femminello Siracusano 2KR, Femminello Fior d'Arancio m79, and Erice, may prove to be particularly well suited to produce processed goods. The market sales of symptomless lemons in 2020 revealed a persistent and severe internal black rot with browning of the membrane wall and juice sac. To investigate the causal agent, externally symptom-free lemons from Mediterranean locations were sampled from supermarkets, local markets, and bazaars in three southeastern Turkish provinces. Internal black rot symptoms were observed in 13.69% of the 1300 externally symptomless fruits, and Alternaria was isolated from 96.63% of those exhibiting these symptoms. Morphological analysis confirmed the identity of Alternaria alternata in 26 representative isolates collected from various markets and bazaars. To examine genetic diversity or polymorphisms, start codon targeted (SCoT) markers were used. Species identification was based on the sequences of translation elongation factor 1-alpha (TEF 1-α), RNA polymerase second largest subunit (RPB2), and glyceraldehyde-3-phosphate dehydrogenase (GAPDH). SCoT 29 exhibited the highest amount of polymorphic amplicons, with 18 polymorphic bands scored when compared to the other six scoreable markers. The 26 representative isolates from various markets and bazaars were divided into four clades by SCoT 29. However, multilocus sequence analyses confirmed the identification of a single species, A. alternata. In terms of virulence and the impact of temperature on growth, representative isolates from four SCoT clusters were evaluated. Data suggests that SCoT markers can be used to evaluate virulence-based polymorphism in A. alternata ; nonetheless, all clades showed comparable temperature responses, with 25 °C being the optimum. There was also no association between individual morphotypes and SCoT clade membership. Due to the lack of obvious signs on lemon fruits, producers, merchants, and consumers are unable to identify black rot, and the widespread presence of A. alternata in lemons may pose a significant risk that must be avoided at all costs. • In 2020, market sales of symptomless lemons displayed severe internal black rot, accompanied by browning of the membrane wall and juice sac. • Alternaria alternata was isolated from 96.63% of 13.69% of 1300 outwardly symptomless fruits with internal black rot. • The SCoT marker 29 could assess virulence-based polymorphism in A. alternata , however all clades responded similarly to temperature, with 25 °C optimal, and SCoT clade membership was unrelated to morphotypes. • This is the first investigation into the existence and incidence of lemon fruit black rot in Turkey, as well as the genetic diversity of the causal agent, A. alternata , using SCoT markers from around the globe. • While shopping, producers, merchants, and customers cannot spot black rot in lemon fruits, and the pervasive incidence of A. alternata in lemons may pose a significant risk that must be avoided at all costs. An extracellular polygalacturonase [EC 3.2.1.15], named PG-543, was purified from the culture of Geotrichum candidum IEV 543, a phytopathogenic fungi isolated from infected lemon. The molecular mass of the enzyme was estimated to be 26 kDa by SDS-PAGE. The pH optimum determined was 5.5; the enzyme showed high stability in the pH range between 4.0 and 6.0. The temperature optimum was 37 ºC. Furthermore, a proteinaceous inhibitor of the polygalacturonase (PG) named PGIP was isolated from lemon albedo. This protein was able to maintain the inhibitory activity in a wide range of pH and temperature. The maximal interaction between PG-PGIP was reached after 20 min of contact. PGIP did not show toxic effect on Artemia salina in the concentration range that was active on PG enzyme. Also, the proteinaceous inhibitor did not show mutagenic nor phytotoxic effects. Our results suggest that, the PGIP isolated from lemon could be used as a natural product to decrease the propagation of pathogenic fungi responsible for postharvest diseases in citrus by the inhibition of hydrolytic enzymes secreted by pathogens. In order to investigate the seasonal variations of antimicrobial properties and chemical composition of essential oils (EOs), three different cultivars of Citrus limon L. Burm. spp. (Femminello Santa Teresa, Monachello and Femminello Continella) were collected at 6-week intervals, from December 2012 to April 2013, for a total of four harvests. The EOs were extracted from lemon peel by hydro-distillation. The antimicrobial activity, tested by paper disc diffusion method, was evaluated against common food-related pathogenic bacteria (Listeria monocytogenes, Staphylococcus aureus, Salmonella enterica and Enterobacter spp.). EOs were more effective against Gram-positive than Gram-negative bacteria at each collection time, but a strong strain dependence was evidenced. Monachello EOs showed the highest inhibition power. The chemical characterisation of the EOs performed by gas chromatography/mass spectrometry identified from 36 to 42 molecules. The chemical difference registered among samples and seasons may explain the different antimicrobial efficacies recorded. [ABSTRACT FROM AUTHOR]

Bacillus sp strain IBA 33 metabolites, isolated from decaying lemon fruits, were evaluated for the control of pathogenic and non-pathogenic fungi (Penicillium digitatum, Geotrichum candidum, Penicillium expansum, Aspergillus clavatus, Aspergillus flavus, Aspergillus niger, and Fusarium moniliforme). These metabolites were recovered from Landy medium (LM) without aminoacids. In order to optimize metabolites production the LM was modified by adding different concentrations and sources of amino acids and carbohydrates at different culture conditions. Bacillus sp strain IBA 33 metabolites efficacy to control fungi were evaluated with in vitro and in vivo assays. A. flavus growth inhibition was 52% with the metabolites of Bacillus sp strain IBA 33 recovered from LM (MBLM) in vitro assays. MBLM supplemented with 0.5% glutamic acid, inhibited the growth of P. digitatum, G. candidum, A. clavatus, A. niger and F. moniliforme by 65%, 88.44%, 84%, 34% and 92% respectively. The highest inhibition of P. expansum was 45% with MBLM supplemented with 0.5% aspartic acid. Similar results were obtained in vivo assays. These results showed that Bacillus sp strain IBA 33 metabolites specificity against fungi depended on the composition of the LM. [ABSTRACT FROM AUTHOR]
