Abstract Objective To investigate the efficacy of Endobutton plate combined with high-strength suture Nice knot fixation in the treatment of distal clavicular fractures with coracoclavicular ligament injuries. Methods A retrospective analysis was performed on 43 patients who sustained distal clavicular fractures along with injuries to the coracoclavicular ligament. These patients were treated between January 2017 and December 2023. The fractures were classified according to the fixation method: high-strength Nice knot suture fixation (experimental group, n = 23) and acromioclavicular Kirschner wire fixation (control group, n = 20). The basic information of the two groups of patients, including age, gender, cause of injury, fracture classification, hospitalization duration, fracture healing time and complications, was collected and analyzed. The increase rate of coracoclavicular space on the affected side was collected and analyzed. The pain level of the affected shoulder was assessed using the visual analog scale (VAS). The shoulder joint function was assessed using the American Shoulder and Elbow Surgeons (ASES) scores and Constant-Murley scores before and after surgery. Results No significant differences were observed in the general demographic data, including age, gender, injury etiology, Craig classification, and hospitalization duration between the two groups (p > 0.05). Both groups were followed for a period ranging from 12 to 33 months, with an average follow-up of 20.53 ± 5.16 months. The bone healing time in the experimental group was significantly shorter than in the control group (12.82 ± 1.12 weeks vs. 17.25 ± 1.71 weeks, p 0.05). Both groups demonstrated significant improvements in VAS scores, Constant-Murley scores, and ASES scores post-operatively compared to pre-operative values (p 0.05). The control group reported one case of infection related to the Kirschner wire and one case of Kirschner wire displacement postoperatively. Conversely, no significant complications were reported in the experimental group. Conclusion In the management of distal clavicle fractures accompanied by coracoclavicular ligament injuries, particularly oblique fractures or those with butterfly-shaped fragments, the application of a high-strength Nice knot suture in conjunction with Endobutton plate fixation can effectively stabilize the fracture site. This approach not only mitigates complications associated with Kirschner wire fixation but also enhances fracture healing, leading to favorable postoperative outcomes.

Abstract The study aims to develop and validate a predictive model for effectively predicting in-hospital mortality in patients admitted to the intensive care unit due to Gastrointestinal bleeding (GIB). A retrospective cohort study was conducted, including data from patients in the Electronic Intensive Care Unit Collaborative Research Database (eICU-CRD) and Medical Information Mart for Intensive Care-IV Database (MIMIC-IV) with a diagnosis of GIB. Patients from the eICU-CRD were randomly allocated into both development and validation sets, and those from MIMIC-IV were assigned as an external validation group. Multivariate logistic regression was employed to create a predictive model, which was depicted as a nomogram. This study included a total of 2929 patients with GIB from the eICU-CRD and 718 patients from the MIMIC-IV. The area under the receiver operating characteristic curve for the nomogram was 0.893 in the development set, 0.860 in the internal validation set, and 0.781 in the external validation set. The mortality rate was 25.7% in the high-risk group (nomogram scores > 101.974) and 2.8% in the low-risk group (nomogram scores ≤ 101.974). The nomogram exhibited excellent discrimination, calibration, and clinical utility in predicting in-hospital mortality among patients admitted to the intensive care unit with GIB.

Abstract Despite antiretroviral therapy (ART), HIV persists in latently-infected cells (the HIV reservoir) which decay slowly over time. Here, leveraging >500 longitudinal samples from 67 people living with HIV (PLWH) treated during acute infection, we developed a mathematical model to predict reservoir decay from peripheral CD4 + T cells. Nonlinear generalized additive models demonstrated rapid biphasic decay of intact DNA (week 0-5: t1/2 ~ 2.83 weeks; week 5-24: t1/2 ~ 15.4 weeks) that extended out to 1 year. These estimates were ~5-fold faster than prior decay estimates among chronic treated PLWH. Defective DNA had a similar biphasic pattern, but data were more variable. Predicted intact and defective decay rates were faster for PLWH with earlier timing of ART initiation, higher initial CD4 + T cell count, and lower pre-ART viral load. In this study, we advanced our limited understanding of HIV reservoir decay at the time of ART initiation, informing future curative strategies targeting this critical time.

Abstract Background Debriefing is the essential element of simulation teaching. Peer-led structured debriefing simulations could be a suitable approach because of the peers’ similarity in age and experience to the students. The purpose of this study was to compare the teaching effectiveness of peer-led debriefing versus instructor-led debriefing in high-fidelity simulation scenarios. Methods The study used a mixed-method approach, integrating quasi-experimental and qualitative components. A total of 88 third-year nursing students were randomly distributed into the instructor-led or peer-led group, with equal sample sizes in both cohorts. The study compared knowledge acquisition, simulation performance, student satisfaction and self-confidence in learning, and debriefing experience between the two groups. In addition, interviews were conducted with students in the peer-led group after the course. Results The scores of students’ simulation performance scale were higher in the peer-led group than in the instructor-led group, and both groups made significant gains in knowledge. The peer-led group and instructor-led group demonstrated similar performance in terms of knowledge, simulated performance, reporting experience, student satisfaction, and learning confidence. The semi-structured interview results indicated that students in the peer-led group expressed approval of peer facilitators. Conclusions Our study demonstrated that students in the peer-led group showed better performance in the simulation, and both groups demonstrated improved knowledge. Thus, with the premise of peers receiving training, the peer-led simulation teaching method can be regarded as a supplementary strategy for simulation teaching to enrich the form of simulation teaching and improve the effectiveness of simulation teaching.

Abstract Seawater electrolysis using renewable electricity offers an attractive route to sustainable hydrogen production, but the sluggish electrode kinetics and poor durability are two major challenges. We report a molybdenum nitride (Mo2N) catalyst for the hydrogen evolution reaction with activity comparable to commercial platinum on carbon (Pt/C) catalyst in natural seawater. The catalyst operates more than 1000 hours of continuous testing at 100 mA cm−2 without degradation, whereas massive precipitate (mainly magnesium hydroxide) forms on the Pt/C counterpart after 36 hours of operation at 10 mA cm−2. Our investigation reveals that ammonium groups generate in situ at the catalyst surface, which not only improve the connectivity of hydrogen-bond networks but also suppress the local pH increase, enabling the enhanced performances. Moreover, a zero-gap membrane flow electrolyser assembled by this catalyst exhibits a current density of 1 A cm−2 at 1.87 V and 60 oC in simulated seawater and runs steadily over 900 hours.

When underwater acoustic homing weapons identify underwater scale targets, it is necessary to extract different dimensional features from underwater target echoes and combine the features to form a complementary feature set to improve the recognition accuracy. However, due to the different application scenarios of different features, introducing all features leads to a high dimension of the feature set and may contain redundant information among each other, which will increase the difficulty of recognition. In the active recognition problem of underwater acoustic homing weapons, the feature set has a high dimension and needs to be selected. To solve these problems, a feature selection algorithm based on random forest(RF)was proposed in this paper. At the same time, to solve the problem of small amounts and unbalanced types of active echo data of underwater acoustic homing weapons, the synthetic minority oversampling technique was adopted in the feature domain. The feature subsets selected by the proposed method were put into the classifiers for testing by using real data. The results show that the proposed method can obtain better feature subsets and effectively improve the recognition accuracy.

Abstract Long non-coding RNAs (lncRNAs) are typically described as RNA transcripts exceeding 200 nucleotides in length, which do not code for proteins. Recent advancements in technology, including ribosome RNA sequencing and ribosome nascent-chain complex sequencing, have demonstrated that many lncRNAs retain small open reading frames and can potentially encode micropeptides. Emerging studies have revealed that these micropeptides, rather than lncRNAs themselves, are responsible for vital functions, including but not limited to regulating homeostasis, managing inflammation and the immune system, moderating metabolism, and influencing tumor progression. In this review, we initially outline the rapidly advancing computational analytical methods and public tools to predict and validate the potential encoding of lncRNAs. We then focus on the diverse functions of micropeptides and their underlying mechanisms in the pathogenesis of disease. This review aims to elucidate the functions of lncRNA-encoded micropeptides and explore their potential applications as therapeutic targets in cancer.

Abstract Trapping electromagnetic waves within the radiation continuum holds significant implications in the field of optical science and technology. Photonic bound states in the continuum (BICs) present a distinctive approach for achieving this functionality, offering potential applications in laser systems, sensing technologies, and other domains. However, the simultaneous achievement of high Q-factors, flat-band dispersions, and wide-angle responses in photonic BICs has not yet been reported, thereby impeding their practical performance due to laser direction deviation or sample disorder. Here, we theoretically demonstrate the construction of moiré BICs in one-dimensional photonic crystal (PhC) slabs, where high-Q resonances in the entire moiré flat band are achieved. Specifically, we numerically validate that the radiation loss of moiré BICs can be eliminated by aligning multiple topological polarization charges with all diffraction channels, enabling the strong suppression of far-field radiation from the entire moiré band. This leads to a slow decay of Q-factors away from moiré BICs in the momentum space. Moreover, it is found that Q-factors of the moiré flat band can still maintain at a high level with structural disorder. In experiments, we fabricate the designed 1D moiré PhC slab and observe both high-Q resonances and a slow decrease of Q-factors for moiré flat-band Bloch modes. Our findings hold promising implications for designing highly efficient optical devices with wide-angle responses and introduce a novel avenue for exploring BICs in moiré superlattices.

Abstract Background Gastric cancer is one of the most common malignant tumors worldwide, with high incidence and mortality rates, and it has a complex etiology and complex pathological features. Depending on the tumor type, gastric cancer can be classified as intestinal-type and diffuse-type gastric cancer, each with distinct pathogenic mechanisms and clinical presentations. In recent years, machine learning techniques have been widely applied in the medical field, offering new perspectives for the diagnosis, treatment, and prognosis of gastric cancer patients. Methods This study recruited 2158 gastric cancer patients and constructed prognostic prediction models for both intestinal-type and diffuse-type gastric cancer. Clinical pathological data were collected from patients, and machine learning algorithms were used for feature selection and model construction. The performance of the models was validated with training and testing datasets. The Shapley additive explanations (SHAP) values were used to interpret the model predictions and identify the main factors that influence patient survival. Results In the prognostic model for intestinal-type gastric cancer, the gradient boosting decision tree (GBDT) model demonstrated the best performance, with key features including pTNM, CA125, tumor size, CA199, and PALB. Similarly, in the prognostic model for diffuse-type gastric cancer, the GBDT model was utilized, with key features comprising pTNM, Borrmann type IV disease, lymphocyte (LYM), lactate dehydrogenase (LDH), potassium (K), perineural invasion (PNI), tumor size, and whole stomach location. Risk stratification analysis revealed that the prognosis of high-risk patients was significantly worse than that of low-risk patients. Conclusion Machine learning shows great potential in predicting survival outcomes of gastric cancer patients, providing strong support for the development of personalized treatment plans.

Abstract In response to the challenge of telomere attrition during DNA replication, cancer cells predominantly employ telomerase or, in 10–15% of cases, the alternative lengthening of telomeres (ALT). The intricate details of ALT, however, remain elusive. In this study, we unveil that the knockdown of lamina-associated polypeptide 2 alpha (LAP2α) in ALT cells results in telomere dysfunction, triggering a notable increase in ALT-associated hallmarks, including high frequencies of PML bodies (APBs), C-rich extrachromosomal circles (C-circles), and telomere sister chromatid exchange (T-SCE). Furthermore, LAP2α emerges as a crucial player in break-induced telomere replication for telomerase-positive cells following telomeric double-strand breaks. Mechanistically, our investigation suggests that LAP2α may influence the regulation of the heterochromatic state of telomeres, thereby affecting telomeric accessibility. In line with our findings, LAP2α expression is markedly reduced in ALT-positive osteosarcoma. And the use of methotrexate (MTX) can restore the heterochromatin state altered by LAP2α depletion. This is evidenced by a significant inhibition of tumor proliferation in ALT-positive patient-derived xenograft (PDX) mouse models. These results indicate the important role of LAP2α in regulating ALT activity and offer insights into the interplay between lamina-associated proteins and telomeres in maintaining telomere length. Importantly, our findings may help identify a more appropriate target population for the osteosarcoma therapeutic drug, MTX.

Taking the No.4 coal seam in the return air roadway in the upper formation No.1 coal mining area of a mine as the background, based on the energy dissipation theory, the energy dissipation mechanical model of arch roadway was established based on the energy balance equation and the energy analysis theory of surrounding rock loose circle, and the failure range of surrounding rock loose circle was positively correlated with the energy dissipation. Based on the Moore-Coulomb strain softening constitutive model, the energy dissipation fish program based on FLAC3D software was developed and numerical simulation was carried out, and the average energy dissipation range of the roadway in the model was determined to be 5.9 m. Combined with the field drilling imaging test, the failure range of the loose ring of the surrounding rock of the roadway was determined to be 5.9 m and 6.04 m respectively. The above content proves that the energy dissipation theory can be well applied to the analysis of the loose zone of surrounding rock.

Abstract Background Despite significant progress in the prognosis of pediatric T-cell acute lymphoblastic leukemia (T-ALL) in recent decades, a notable portion of children still confronts challenges such as treatment resistance and recurrence, leading to limited options and a poor prognosis. LIM domain-binding protein 1 (LDB1) has been confirmed to exert a crucial role in various physiological and pathological processes. In our research, we aim to elucidate the underlying function and mechanisms of LDB1 within the background of T-ALL. Methods Employing short hairpin RNA (shRNA) techniques, we delineated the functional impact of LDB1 in T-ALL cell lines. Through the application of RNA-Seq, CUT&Tag, and immunoprecipitation assays, we scrutinized master transcription factors cooperating with LDB1 and identified downstream targets under LDB1 regulation. Results LDB1 emerges as a critical transcription factor co-activator in cell lines derived from T-ALL. It primarily collaborates with master transcription factors (ERG, ETV6, IRF1) to cooperatively regulate the transcription of downstream target genes. Both in vitro and in vivo experiments affirm the essential fuction of LDB1 in the proliferation and survival of cell lines derived from T-ALL, with MYB identified as a significant downstream target of LDB1. Conclusions To sum up, our research establishes the pivotal fuction of LDB1 in the tumorigenesis and progression of T-ALL cell lines. Mechanistic insights reveal that LDB1 cooperates with ERG, ETV6, and IRF1 to modulate the expression of downstream effector genes. Furthermore, LDB1 controls MYB through remote enhancer modulation, providing valuable mechanistic insights into its involvement in the progression of T-ALL.

Abstract Exploring land use/cover (LULC) change is essential for the sustainable development of ecologically fragile areas. The main objective of this study was to clarify the characteristics and differences in the spatiotemporal changes of LULC on the Loess Plateau (LP) based on the transfer matrix and land use dynamics and to quantitatively describe the impact of natural factors on LULC using a geodetector. The results indicated that the overall LULC change in the LP was characterized by a decrease in the area of cropland, grassland, and bare land, and an increase in the area of woodland and built-up land. This trend shows a clear phase-change characteristic around 2000. LULC changes were primarily affected by human activities in the southeastern agricultural region. The project of returning farmland to forest and grassland had a great impact on LULC change in the central region. Vegetation was sensitive to temperature and precipitation, and the impact of LULC change was significantly higher than that in the humid region in the northwest arid region. NDVI, PRE, and TEM were determined to be the main contributors to LULC changes in the LP. These results provide a scientific basis for the sustainable development of LP.

Abstract Immune evasion represents a crucial milestone in the progression of cancer and serves as the theoretical foundation for tumor immunotherapy. In this study, we reveal a negative association between Human Papillomavirus (HPV)-encoded circular RNA, circE7, and the infiltration of CD8+ T cells in head and neck squamous cell carcinoma (HNSCC). Both in vitro and in vivo experiments demonstrate that circE7 suppresses the function and activity of T cells by downregulating the transcription of LGALS9, which encodes the galectin-9 protein. The molecular mechanism involves circE7 binding to acetyl-CoA carboxylase 1 (ACC1), promoting its dephosphorylation and thereby activating ACC1. Activated ACC1 reduces H3K27 acetylation at the LGALS9 gene promoter, leading to decreased galectin-9 expression. Notably, galectin-9 interacts with immune checkpoint molecules TIM-3 and PD-1, inhibiting the secretion of cytotoxic cytokines by T cells and promoting T cell apoptosis. Here, we demonstrate a mechanism by which HPV promotes immune evasion in HNSCC through a circE7-driven epigenetic modification and propose a potential immunotherapy strategy for HNSCC that involves the combined use of anti-PD-1 and anti-TIM-3 inhibitors.

Abstract Background X-linked intellectual disability-hypotonic facies syndrome-1 (MRXHF1) and Alpha-thalassemia X-linked intellectual disability (ATR-X) syndrome are caused by pathogenic variant in the ATRX gene, a member of the switch/sucrose non-fermentable (SWI-SNF) protein family that exhibits chromatin remodeling activity. These syndromes show a wide spectrum of clinical manifestations, such as distinctive dysmorphic features, mild-to-profound intellectual disability, motor development delay, seizures, urogenital abnormalities, and gastrointestinal disorders. Case presentation and literature review A 3-year-old boy from a Chinese non-consanguineous family was diagnosed with MRXHF1 by whole-exome sequencing. Comprehensive family history information was obtained. The Medline database was searched until 1st Aug 2023 for articles related to ATRX pathogenic variant. Data on gene/protein mutations and clinical symptoms were extracted. The proband showed intellectual disability, motor development delay, typical facial abnormalities, urogenital defect, behavior problems, and optical nerve dysplasia. A novel frameshift mutation c.399_400dup, (p.Leu134Cysfs*2) in the ATRX gene was the primary cause, which occurs right before the ATRXDNMT3-DNMT3L (ADD) domain of ATRX protein. Missense mutation is the most common variation type. The ADD and helicase-like domains are the most frequently affected domains. Epilepsy, congenital heart disease, urogenital defect, acoustic defect, and optical defect are more prevalent in patients with frameshift mutations compared to those with missense mutations. There are more urogenital defects with C-terminal frameshift mutations than with N-terminal frameshift mutations. Conclusion We described a novel frameshift mutation in the ATRX gene in a patient with MRXHF1 syndrome and summarized the genotype–phenotype relationship of ATRX pathogenic variant by variation type and affected protein domain. The regulatory mechanism underlying ATRX variant requires comprehensive analysis in future studies.

Abstract. Background:. China is one of the countries with the largest burden of gastrointestinal and liver diseases (GILD) in the world. The GILD constitutes various causes of mortality and disability. The study aimed to investigate the trend of GILD in China using the Global Burden of Diseases Study 2019 (GBD 2019) data resources from 1990 to 2019. Methods:. The data on the age-standardized mortality rates (ASMR) and disability-adjusted life years (DALYs) for GILD in China from 1990 to 2019 were collected from the GBD 2019 data resources. Furthermore, the ranking of the main causes of deaths and DALYs, as well as the trends of ASMR, DALYs, years of life lost (YLLs), and years of life lost due to disability (YLDs) per 1,000,000 in GILD were reported. Results:. The ASMR and DALYs for stomach cancer, liver cancer, and esophageal cancer, which ranked top three among the GILDs from 1990 to 2019, were gradually decreasing. Significant decreases in the ASMR and DALYs were found in diarrheal diseases and acute hepatitis (A, E, and C). However, noteworthy increases were found in those of colon and rectum cancer (CRC) and pancreatic cancer. Trend of DALYs, mortality, and YLLs rates for most of GILD were decreasing from 1990 to 2019, except the burden of CRC and pancreatic cancer with an increasing trend. The DALYs, mortality and YLLs of most GILD diseases showed decreasing trends from 1990 to 2019, except the burden of CRC and pancreatic cancer with an increasing trends. Conclusions:. The result of the GBD 2019 showed that the rates of most GILDs decreased in China; however, gastrointestinal and liver cancer, such as stomach cancer still held the top ranking. Furthermore, the shift from infectious diseases to non-communicable causes among GILD burden is occurring.

Implant-associated Staphylococcus aureus (S. aureus) osteomyelitis is a severe challenge in orthopedics. While antibiotic-loaded bone cement is a standardized therapeutic approach for S. aureus osteomyelitis, it falls short in eradicating Staphylococcus abscess communities (SACs) and bacteria within osteocyte-lacuna canalicular network (OLCN) and repairing bone defects. To address limitations, we developed a borosilicate bioactive glass (BSG) combined with ferroferric oxide (Fe3O4) magnetic scaffold to enhance antibacterial efficacy and bone repair capabilities. We conducted comprehensive assessments of the osteoinductive, immunomodulatory, antibacterial properties, and thermal response of this scaffold, with or without an alternating magnetic field (AMF). Utilizing a well-established implant-related S. aureus tibial infection rabbit model, we evaluated its antibacterial performance in vivo. RNA transcriptome sequencing demonstrated that BSG + 5%Fe3O4 enhanced the immune response to bacteria and promoted osteogenic differentiation and mineralization of MSCs. Notably, BSG + 5%Fe3O4 upregulated gene expression of NOD-like receptor and TNF pathway in MSCs, alongside increased the expression of osteogenic factors (RUNX2, ALP and OCN) in vitro. Flow cytometry on macrophage exhibited a polarization effect towards M2, accompanied by upregulation of anti-inflammatory genes (TGF-β1 and IL-1Ra) and downregulation of pro-inflammatory genes (IL-6 and IL-1β) among macrophages. In vivo CT imaging revealed the absence of osteolysis and periosteal response in rabbits treated with BSG + 5%Fe3O4 + AMF at 42 days. Histological analysis indicated complete controls of SACs and bacteria within OLCN by day 42, along with new bone formation, signifying effective control of S. aureus osteomyelitis. Further investigations will focus on the in vivo biosafety and biological mechanism of this scaffold within infectious microenvironment.

Objective The principles of limb reconstruction are crucial for treatment success, but there is no unified standard for complex limb deformities. The aim of this study was to analyze the characteristics of the cases of post‐traumatic lower limb deformity and explore the new principle of limb reconstruction. Method A retrospective analysis was conducted of 148 patients with post‐traumatic lower limb deformity who underwent surgery from May 1978 to December 2023; 85 were males (57.4%) and 63 were females (42.6%); 65 cases of left side (43.9%), 79 cases of right side(53.4%), and 4 cases were on both sides (2.7%), the average age was 24.64 years (5–69). There were 4 cases suffering hip deformities, 40 cases of femoral deformities, 18 cases from knee, 40 cases from tibiofibular, 93 cases of foot and ankle deformities, and some patients also had two or more types. All patients underwent surgical intervention in an average of 40.5 months (12–96) after injury. According to the evaluation of limb deformities, deformity correction and functional reconstruction with external fixation were implemented, following the principle of “one walking, two lines, and three balances.” The clinical evaluation adopts the criteria of Qinsihe lower limb deformity correction and functional reconstruction. Result 148 patients with post‐traumatic lower limb deformities were followed up for 40.9 (12–356) months. The main surgical procedures implemented were tendon lengthening and soft tissue release (84 cases), osteotomy (93 cases), joint fusion (30 cases), and tendon transposition (16 cases); there were multiple surgical procedures in some patients. Among them, 124 cases used external fixators for stress control and 27 cases used internal fixation, while 3 cases used plaster or brace. There were 5 wire reactions postoperatively, which improved after dressing change and oral antibiotics. There were 2 pin infections, which improved by pin removing. No surgical related deep infections occurred, and no surgical related neurovascular damage occurred. At the last follow‐up, all limb deformities were corrected, limb function improved, and the results of treatment was very satisfactory. According to Qinsihe evaluation criteria for lower limb deformities, 74 cases were excellent, 56 cases good, and 18 cases fair, with an excellent and good rate of 87.84%. Conclusion Stress control with external fixation is effective, safe, and controllable in correcting and reconstructing post‐traumatic lower limb deformities. The principle of “one walking, two lines, and three balances” plays an important role in the entire process of stress control limb reconstruction.

Abstract Introduction In congenital aniridia caused by mutations in paired box 6 (PAX6), PAX6 influences the migration and differentiation of limbal epithelial cells (LECs), thereby playing a pivotal role in aniridia-associated keratopathy. The antidepressants ritanserin and duloxetine affect PAX6 expression in LECs. Limbal stromal cells, which support limbal epithelial stem cells, are crucial in the limbal stem cell niche. This study explores how ritanserin and duloxetine influence gene expression in primary human limbal stromal cells from subjects with congenital aniridia and from healthy subjects, in vitro. Methods Primary human limbal stromal cells from corneas affected by aniridia (AN-LSCs) (n = 8) and from healthy corneas (LSCs) (n = 8) were isolated and cultured in either low-glucose serum-free (LGSF) or normal-glucose serum-containing (NGSC) media. Cells were treated with 4 µM ritanserin or duloxetine for 24 h. Quantitative PCR (qPCR) and western blot were used to assess the expression of PAX6, FOSL2, TGF-β1, ACTA2A1, LUM, COL1A1, COL5A1, DSG1, FABP5 and ADH7. Results In AN-LSCs with LGSF-medium, ritanserin increased PAX6 messenger RNA (mRNA) (p = 0.007) and decreased TGF-β1 and FOSL2 mRNA levels (P = 0.005, P = 0.038). In addition, TGF-β1 protein levels decreased with both treatments (P = 0.02, P = 0.007), and FABP5 protein level increased, using ritanserin (P = 0.019). In LSCs with LGSF-medium, ACTA2A1 mRNA levels decreased using ritanserin and duloxetine (P = 0.028; P = 0.031), while FABP5 mRNA levels increased with ritanserin treatment (P = 0.003). Also, duloxetine use reduced α-SMA protein (P = 0.013) and increased FABP5 protein levels (P = 0.029). In LSCs with NGSC-medium, ritanserin elevated LUM, FABP5 and ADH7 mRNA and protein levels (P = 0.025, P = 0.003, P = 0.047, P = 0.024, P = 0.013, P = 0.039). Conclusions The results of our study confirmed that the antipsychotropic drugs ritanserin and duloxetine alter PAX6 and TGF-β1 gene expression in AN-LSCs cultured in LGSF-medium. These drugs were found to have an impact on retinoic acid signaling pathways and keratocyte characteristic markers both in LSCs and AN-LSCs, using different culture media.

Abstract Toe fringes are a key innovation for sand dwelling lizards, and the relationship between toe fringe function and substrate properties is helpful in understanding the adaptation of lizards to sand dune environments. We tested the sand burial performance of Phrynocephalus mystaceus on different sand substrates with toe fringe manipulation, with the aim of assessing whether the function of the toe fringes shifts under different substrate properties, especially in highly mobile substrates. The sand burial performance of P. mystaceus was influenced by substrate properties in relation to the toe fringe states of the lizard. After removal of the bilateral toe fringes, the sand burial ability score of P. mystaceus was significantly higher on sand substrates below 100 mesh than on native sand substrates. As the angle of stability of the substrate properties decreased, the sand burial performance of the lizard was even better after the bilateral toe fringes were removed. The results of the LASSO model and the path analysis model showed that the stability angle provided the opposite effect on sand burial performance in different toe fringe states. These results further suggest that the sand burial function of toe fringes may not be suitable for highly mobile sand substrates. It remains to be tested further whether the function of toe fringes is more important for running on sand.

Aiming at the problem of large deformation and instability control of narrow coal pillar in the vertical stratification section of extra-thick coal seam, the stability and control technology of the surrounding rock of narrow coal pillar in extra-thick coal seam are studied by combining theoretical analysis, numerical simulation and field test. Based on the calculation of the triaxial stress in the narrow coal pillar, the yield criterion of the plane strain of the narrow coal pillar is obtained based on the Mohr-Coulomb failure strength, the failure characteristics and the evolution law of the damage degree of the narrow coal pillar are qualitatively analyzed, and the instability mechanism and size effect of the narrow coal pillar are further analyzed. Finally, the key technologies of surrounding rock control of the narrow coal pillar are given. It is verified by numerical simulation and field test. The research results show that ① The damage degree of the middle part of the narrow coal pillar is greater than that of the two sides, and the damage degree of the middle part from the top to the bottom develops from serious to slight. With the increase of width-height ratio, the damage degree and the proportion of serious damage area in the narrow coal pillar gradually decrease. When the width-height ratio is greater than 1∶1, a large range of slight damage area begins to appear in the middle and lower part of the narrow coal pillar. When the width-height ratio is greater than 5∶3, the proportion of slight damage area exceeds 50%. When C≥3 MPa, or φ≥20°, the damage degree of the two sides of the narrow coal pillar becomes slight. ② The side of the mining roadway in the working face is a low-stress bearing area of the narrow coal pillar. Under the action of high stress in the upper part, the coal body in the large-scale low-stress bearing area moves to the roadway, resulting in a continuous large deformation of the two sides, which in turn affects the stability of the roof. The height of coal pillar is the main controlling factor for the stability of the narrow coal pillar in the middle and bottom layered sections. The increase of width-height ratio of the narrow coal pillar has little influence on the bearing strength of high strength bearing area, while the bearing strength of low strength bearing area increases obviously. Reasonable width-height ratio of the narrow coal pillar can balance the proportion of high and low strength bearing area in coal pillar. ③ The stability of surrounding rock should be considered in the design of layered narrow coal pillars when both sides of the working face are mined out, so as to ensure that the range of high-strength bearing area inside the coal pillar is greater than half of the width and height of the coal pillar. Through the joint strengthening support measures, multiple joint control areas from shallow to deep are formed in the coal pillar to jointly maintain the self-stability of the coal pillar.

Abstract The prevalence of autism spectrum disorders (ASDs) differs substantially between males and females, suggesting that sex-related neurodevelopmental factors are central to ASD pathogenesis. Numerous studies have suggested that abnormal brain specialization patterns and poor regional cooperation contribute to ASD pathogenesis, but relatively little is known about the related sex differences. Therefore, this study examined sex differences in brain functional specialization and cooperation among children with ASD. The autonomy index (AI) and connectivity between functionally homotopic voxels (CFH) derived from resting-state functional magnetic resonance imaging (rs-fMRI) were compared between 58 male and 13 female children with ASD. In addition, correlations were examined between regional CFH values showing significant sex differences and symptom scores on the autism behavior checklist (ABC) and childhood autism rating scale (CARS). Male children with ASD demonstrated significantly greater CFH in the left fusiform gyrus (FG) and right opercular part of the inferior frontal gyrus (IFGoperc) than female children with ASD. In addition, the CFH value of the left FG in male children with ASD was negatively correlated with total ABC score and subscale scores for sensory and social abilities. In contrast, no sex differences were detected in brain specialization. These regional abnormalities in interhemispheric cooperation among male children with ASD may provide clues to the neural mechanisms underlying sex differences in ASD symptomatology and prevalence. Abstract Background Klotho plays a pivotal role in human aging. Metabolic syndrome (MetS) is composed of multiple conditions that are also risk factors for cardiovascular disease and diabetes. We try to discuss gender-specific differences in Klotho and the associations between Klotho and MetS components. Materials and methods The National Health and Nutrition Examination Survey database from cycle 2015–2016 was analyzed. MetS was defined according to the 2005 updated criteria by the American Heart Association and National Heart Lung and Blood Institute. Gender-specific differences in serum Klotho, and associations between Klotho level and MetS components were examined. Results A total of 2475 participants (40–79 years old) with comprehensive data were included (52% women). In general, lower Klotho was associated with advanced age, male sex, tobacco use, elevated triglycerides, renal insufficiency, inflammation, low estradiol, and low sex hormone-binding globulin (SHBG). The correlation between MetS and Klotho was more obvious in women, mainly in waist circumference and triglyceride. There were no gender-specific differences in the associations between Klotho and renal dysfunction, but multivariate linear regression analysis showed gender differences in other factors associated with Klotho. Estradiol, SHBG, high-density lipoprotein cholesterol (HDL), and high-sensitivity C-reactive protein (CRP) were associated with Klotho levels independent of age and renal function in men, whereas in women, Klotho was independently associated with triglycerides and white blood cell count. Conclusion Klotho levels had gender disparities regardless of age, renal function, and sex hormones. In the current cohort, triglycerides were the major component of MetS that was independently associated with serum Klotho levels, and the association was particularly seen in women. However, HDL was found to be the male-specific MetS component independently associated with Klotho.

Abstract Risk cascading propagation research mostly focuses on complex theoretical networks. Recently, the vulnerability of international chip supply has increased notably, and it is strategically important to explore how shortage risk affects the emergence dynamics of the real chip trade systems. This study abstracts the global chip trade relationship data for 2009–2021 into multiple asymmetrically weighted networks. Using macro-network and micro-node indicators, we explore the topological traits of international chip trade networks and their evolutionary laws. Accordingly, we propose risk cascading propagation models driven by node failure and edge restraint and further innovate to open up the research paradigm of focused-edge networks. Furthermore, a community infection-driven risk cascading propagation mechanism that incorporates community risk absorption capacity is introduced, considering both the propagation attenuation effects and the trade dependency degree. A multi-dimensional dynamic perspective reveals the hidden systemic risks in international chip trade. The main results are as follows: first, international chip trade networks are highly connected and cohesive, consistent with small-world characteristics. Second, the proportion of economies that collapse because of supply shortage risk shocks increases with the impact coefficient $$\alpha /\beta$$ α / β . The dominant power in chip crisis propagation has shifted from Europe and America to Asia, and mainland China’s risk penetration capacity has enhanced significantly. Third, focused-edge networks conform to a multi-hub radiation pattern. Before the COVID-19 pandemic, the degree of control and spillover effects of chip supply shortages in hub economies on the international trade was increasing progressively. Fourth, an increase in absorption capacity $$\lambda$$ λ or attenuation factor $$\gamma$$ γ consistently leads to a decline in avalanche scale, with $$\lambda$$ λ exhibiting critical thresholds. These findings will help policymakers pursue efficient management strategies for chip trade, thereby improving supply stability and security.

Radioisotopes tend to play important roles in many fields, such as industry, healthcare, agriculture, aerospace, etc. Radioisotope production is mainly through accelerators or research reactors, and high flux research reactor is one of the most effective approaches for radioisotope production. The physical basis of preparing radioisotope relies on nuclear reactions occurring in the reactor core, which includes fission, (n,γ), (n,α), and (n,p) reaction, etc. Among them, fission and (n,γ) reaction are most important in the nuclear reactor. For example, the 99Mo could be generated by uranium fission and extracting from the fission products, or through the radiative capture reaction from enriched 98Mo. As for the fission method, the irradiation target is gradually transitioning from high enriched uranium (HEU) target to low enriched uranium (LEU) target due to the requirement of non-proliferation. In this paper, studies on the impacts of different fission targets on radioisotope productions are conducted. Moreover, an optimized study on the radiative capture method is performed to improve the production efficiency. It is concluded that it is advantageous to use radiative capture method to generate radioisotopes in high flux reactor, which helps to improve the specific activity with environmental friendliness.

It is essential to understand the formation and evolution mechanism of intermetallic compounds (IMCs) in friction stir welding (FSW) of dissimilar Al/Mg alloys to mitigate the deleterious effect of hard and brittle IMCs on the performance of dissimilar joints. To this end, a multiphase-field model with input variables of both temperature and strain rate is developed to study the whole IMCs formation and growth process at the Al/Mg bonding interface in weld nugget zone. The effects of diffusion, elastic deformation, and large plastic deformation on the generation and growth of IMCs are considered. The numerical simulation results indicate that due to the influence of high dislocation density, the higher diffusion coefficient of the Mg matrix leads to the nucleation of Al12Mg17 first. When IMCs grow into layers, the higher diffusion coefficient of Al3Mg2 leads to a faster growth rate of Al3Mg2 than that of Al12Mg17. The elastic energy caused by lattice mismatch between IMCs and matrix plays an inhibitory role in the growth of IMCs. Large plastic deformation in FSW causes an increase in both the stored energy which promotes the IMCs growth and the dislocation density which intensifies the diffusion process, causing IMCs to form at the interface in a short time. With the model, the IMCs evolution from generation to growth is quantitatively analyzed. The predicted thickness of IMCs matches well with the experimental measurements.

Background: Drug-induced liver injury (DILI) is a leading cause of drug development failures during clinical trials and post-market introduction. Current biomarkers, such as ALT and AST, lack the necessary specificity and sensitivity needed for accurate detection. Exosomes, which protect LncRNAs from RNase degradation, could provide reliable and easily accessible options for biomarkers. Materials and methods: RNA-sequencing was used to identify differentially expressed LncRNAs (DE-LncRNAs), followed by isolation of LncRNAs from plasma exosomes in this study. Exosome characterization was conducted by transmission electron microscopy (TEM), nanoparticle tracking analysis (NTA), and Western blot (WB). Bioinformatics analysis included functional enrichment and co-expression network analysis. Five rat models were established, and quantitative real-time PCR was used to verify the specificity and sensitivity of two candidate exosomal LncRNAs. Results: The APAP-induced hepatocellular injury model was successfully established for RNA-sequencing, leading to the identification of several differentially expressed exosomal LncRNAs. Abstract Background Middle-aged and elderly individuals are the most susceptible groups for metabolic diseases, with their dietary behaviors being significant influencing factors. Exploring the association between overall dietary behaviors and obesity metabolic phenotypes is crucial for early prevention and control of chronic diseases, precision treatment and personalized interventions. Methods We conducted a cross-sectional study of 15,160 middle-aged and older adults between June 2019 and August 2021 to collect information on their body mass index (BMI), biochemical indices and disease history. The population was classified into four categories by the criteria of obesity metabolic phenotypes: metabolically healthy non-obesity (MHNO), metabolically unhealthy non-obesity (MUNO), metabolically healthy obesity (MHO) and metabolically unhealthy obesity (MUO). Scores were calculated based on compliance with healthy eating behavior patterns (appropriate or light dietary taste, moderately soft and hard food, slightly hot food temperature, medium or slow eating speed, daily intake of dietary supplements and eating with others), and the population was categorized into subgroups 0–2 (did not meet and met only 1 or 2), 3–4 (met 3 or 4), 5–6 (met 5 or 6). Abstract To evaluate whether radiomics models based on unenhanced paranasal sinuses CT images could be a useful tool for differentiating inverted papilloma (IP) from chronic rhinosinusitis with polyps (CRSwNP). This retrospective study recruited 240 patients with CRSwNP and 106 patients with IP from three centers. 253 patients from Qilu Hospital were randomly divided into the training set (n = 151) and the internal validation set (n = 102) with a ratio of 6:4. 93 patients from the other two centers were used as the external validation set. The patients with the unilateral disease (n = 115) from Qilu Hospital were selected to further develop a subgroup analysis. Lesion segmentation was manually delineated in CT images. Least absolute shrinkage and selection operator algorithm was performed for feature reduction and selection. Decision tree, support vector machine, random forest, and adaptive boosting regressor were employed to establish the differential diagnosis models. 43 radiomic features were selected for modeling. Among the models, RF achieved the best results, with an AUC of 0.998, 0.943, and 0.934 in the training set, the internal validation set, and the external validation set, respectively. In the subgroup analysis, RF achieved an AUC of 0.999 in the training set and 0.963 in the internal validation set. The proposed radiomics models offered a non-invasion and accurate differential approach between IP and CRSwNP and has some significance in guiding clinicians determining the best treatment plans, as well as predicting the prognosis.

Abstract Approximately 75% of stroke survivors have movement dysfunction. Rehabilitation exercises are capable of improving physical coordination. They are mostly conducted in the home environment without guidance from therapists. It is impossible to provide timely feedback on exercises without suitable devices or therapists. Human action quality assessment in the home setting is a challenging topic for current research. In this paper, a low-cost HREA system in which wearable sensors are used to collect upper limb exercise data and a multichannel 1D-CNN framework is used to automatically assess action quality. The proposed 1D-CNN model is first pretrained on the UCI-HAR dataset, and it achieves a performance of 91.96%. Then, five typical actions were selected from the Fugl-Meyer Assessment Scale for the experiment, wearable sensors were used to collect the participants’ exercise data, and experienced therapists were employed to assess participants’ exercise at the same time. Following the above process, a dataset was built based on the Fugl-Meyer scale. Based on the 1D-CNN model, a multichannel 1D-CNN model was built, and the model using the Naive Bayes fusion had the best performance (precision: 97.26%, recall: 97.22%, F1-score: 97.23%) on the dataset. This shows that the HREA system provides accurate and timely assessment, which can provide real-time feedback for stroke survivors’ home rehabilitation.

Abstract Purpose This study aimed to investigate the effects of different shrub plants on the structure and diversity of inter-root soil bacterial communities, in order to provide scientific support for ecological restoration and revegetation of the Mu Us Desert. Methods Three major shrub plants (Artemisia ordosica, Salix psammophila, Caragana microphylla) in the Mu Us Desert were selected for this study. Using high-throughput sequencing technology, the bacterial community structure and diversity in the inter-root soils of these plants were analysed in depth, and combined with the determination of soil physicochemical and microbiological properties, the response characteristics of the bacterial diversity in the inter-root soils of the different plants were assessed comprehensively. Results It was found that although the soil pH did not show significant differences among different plant growths, the SOC, TN and TP contents were higher in Salix psammophila sample plot and Artemisia ordosica sample plot, which indicated that the plant growths had a positive effect on the soil nutrient contents. Through Venn diagram analysis, it was observed that the number of OTUs of bacteria in the soils of different shrubland sites varied, and all of them were higher than those in the soils of the sample sites where no plants grew, which indicated that plants had an effect on soil bacterial diversity. The bacterial Chao1 index were higher in the Artemisia ordosica sample plot sample site, suggesting that the growth of Artemisia ordosica contributes to the enhancement of soil bacterial richness. Soil bacterial communities showed compositional differences among different sample plots, especially the higher relative abundance of Betaproteobacteria in the Artemisia ordosica sample plot sample plot, which may be related to the increase of soil organic matter content. Conclusion The results of the study revealed that specific plants, such as Artemisia ordosica, can significantly improve the soil nutrient status of windy sandy soils, increase soil organic matter and nitrogen content, and thus enhance the diversity and abundance of soil microorganisms. The bacterial community structure in the inter-root soils of different plants differed significantly, with changes in the relative abundance of the dominant phyla, such as Alphaproteobacteria, Betaproteobacteria and Actinobacteria, reflecting the differences in soil nutrient status. These findings emphasise the important role of plants on soil chemical properties and microbial community structure, providing an important basis for soil management and ecological restoration.

Abstract Metalens, characterized by their unique functions and distinctive physical properties, have gained significant attention for their potential applications. To further optimize the performance of metalens, it is necessary to characterize the phase modulation of the metalens. In this study, we present a multi-distance phase retrieval system based on optical field scanning and discuss its convergence and robustness. Our findings indicate that the system is capable of retrieving the phase distribution of the metalens as long as the measurement noise is low and the total length of the scanned light field is sufficiently long. This system enables the analysis of focal length and aberration by utilizing the computed phase distribution. We extend our investigation to measure the phase distribution of the metalens operating in the near-infrared (NIR) spectrum and identify the impact of defects in the sample on the phase. Additionally, we conduct a comparative analysis of the phase distribution of the metalens in air and ethanol and observe the variations in the phase modulation of the metalens in different working mediums. Our system provides a straightforward method for the phase characterization of metalens, aiding in optimizing the metalens design and functionality.

Abstract The impact of efferocytosis-related genes (ERGs) on the diagnosis of colorectal cancer (CRC) remains unclear. In this study, efferocytosis-associated biomarkers for the diagnosis of CRC were identified by integrating data from transcriptome sequencing and public databases. Finally, the expression of biomarkers was validated by real-time quantitative polymerase chain reaction (RT-qPCR). Our study may provide a reference for CRC diagnosis. Background It has been shown that some efferocytosis related genes (ERGs) are associated with the development of cancer. However, it is still uncertain how ERGs may influence the diagnosis of colorectal cancer (CRC). Methods In our study, the CRC cohorts were gained from transcriptome sequencing and the gene expression omnibus (GEO) database (GSE71187). Efferocytosis related biomarkers with diagnostic utility for CRC were identified through combining differentially expressed analysis, machine learning algorithms, and receiver operating characteristic (ROC) analysis. Then, infiltration abundance of immune cells between CRC and control was evaluated. The regulatory networks (including mRNA-miRNA-lncRNA and miRNA/transcription factors (TF)-mRNA networks) were created. Finally, the expression of biomarkers was validated via real-time quantitative polymerase chain reaction (RT-qPCR). Results There were 3 biomarkers (ELMO3, P2RY12, and PDK4) related diagnosis for CRC patients gained. ELMO3 was highly expressed in CRC group, while P2RY12 and PDK4 was lowly expressed. Besides, the infiltrating abundance of 3 immune cells between CRC and control groups was significantly differential, namely activated CD4 memory T cells, macrophages M0, and resting mast cells. We then constructed a mRNA-miRNA-lncRNA network containing 3 mRNAs, 33 miRNAs, and 22 lncRNAs, and a miRNA/TF-mRNA network including 3 mRNAs, 33 miRNAs, and 7 TFs. Additionally, RT-qPCR results revealed that the expression trends of all biomarkers were consistent with the transcriptome sequencing data and GSE71187. Conclusion Taken together, this study provides three efferocytosis related biomarkers (ELMO3, P2RY12, and PDK4) for diagnosis of CRC, providing a scientific reference for further studies of CRC.

Abstract With the rapid development of Chinese transportation networks, such as the Sichuan-Tibet railway, numerous tunnels are under construction or planned in mountainous regions. Some of these tunnels must traverse or be situated near active fault zones, which could suffer damage from fault slip. In this study, the seismic response of a mountain tunnel subjected to coseismic faulting was analyzed using a fault-structure system in a two-step process. Firstly, a nonuniform slip model was proposed to calculate the ground deformations and internal displacements induced by a specific active fault on a geological scale, considering nonuniform slips on the fault plane. The 1989 Loma Prieta and 2022 Menyuan earthquakes were chosen as case studies to validate the proposed slip model. Secondly, the calculated displacement of the Menyuan earthquake was used as the input load for the discrete–continuous coupling analysis of the Daliang tunnel on an engineering scale. The simulated deformation of the Daliang tunnel aligned with the on-site damage observations following the Menyuan earthquake. Lastly, the effects of different fault conditions on the tunnel seismic response were investigated. The results indicate that the distribution of the peak longitudinal strain of the lining is governed by fault mechanisms, and the degree of fault slip significantly influences the response of the tunnel. A tunnel passing through an active fault with a wider fault fracture zone and smaller dip angle experience less damage.

Abstract Background Synovial fibrosis is a common complication of knee osteoarthritis (KOA), a pathological process characterized by myofibroblast activation and excessive extracellular matrix (ECM) deposition. Fibroblast-like synoviocytes (FLSs) are implicated in KOA pathogenesis, contributing to synovial fibrosis through diverse mechanisms. Nuclear protein 1 (NUPR1) is a recently identified transcription factor with crucial roles in various fibrotic diseases. However, its molecular determinants in KOA synovial fibrosis remain unknown. This study aims to investigate the role of NUPR1 in KOA synovial fibrosis through in vivo and in vitro experiments. Methods We examined NUPR1 expression in the murine synovium and determined the impact of NUPR1 on synovial fibrosis by knockdown models in the destabilization of the medial meniscus (DMM)-induced KOA mouse model. TGF-β was employed to induce fibrotic response and myofibroblast activation in mouse FLSs, and the role and molecular mechanisms in synovial fibrosis were evaluated under conditions of NUPR1 downexpression. Additionally, the pharmacological effect of NUPR1 inhibitor in synovial fibrosis was assessed using a surgically induced mouse KOA model. Results We found that NUPR1 expression increased in the murine synovium after DMM surgical operation. The adeno-associated virus (AAV)-NUPR1 shRNA promoted NUPR1 deficiency, attenuating synovial fibrosis, inhibiting synovial hyperplasia, and significantly reducing the expression of pro-fibrotic molecules. Moreover, the lentivirus-mediated NUPR1 deficiency alleviated synoviocyte proliferation and inhibited fibroblast to myofibroblast transition. It also decreased the expression of fibrosis markers α-SMA, COL1A1, CTGF, Vimentin and promoted the activation of the SMAD family member 3 (SMAD3) pathway. Importantly, trifluoperazine (TFP), a NUPR1 inhibitor, attenuated synovial fibrosis in DMM mice. Conclusions These findings indicate that NUPR1 is an antifibrotic modulator in KOA, and its effect on anti-synovial fibrosis is partially mediated by SMAD3 signaling. This study reveals a promising target for developing novel antifibrotic treatment.

The integration of unmanned aerial vehicle (UAV) networks with intelligent reflecting surface (IRS) technology offers a promising solution to enhance wireless communication by dynamically altering signal propagation. This study addresses the challenge of maximizing system energy efficiency (EE) in IRS-assisted UAV networks. The primary objective is to optimize power allocation and IRS reflection design to achieve this goal. To tackle the optimization problem, we employ a block coordinate descent (BCD) method, decomposing it into three subproblems: phase shift optimization, power allocation, and trajectory planning. These subproblems are iteratively solved using an improved particle swarm optimization (PSO) algorithm. Simulation results demonstrate that the proposed PSO algorithm effectively plans high-quality UAV trajectories in complex environments, significantly enhancing EE. The findings suggest that the IRS-assisted UAV model outperforms traditional UAV models, offering substantial improvements in wireless communication quality and EE.

【Objective】In October 2023, skin ulceration, subcutaneous hemorrhage and enteritis appeared in a breeding base of Epinephelus coioides in Shenzhen, China. We clarified the causative organisms that triggered the sudden death of E. coioides and their biological characteristics, and screened the disinfectant that effectively inhibited the causative organisms, with an aim to provide theoretical guidance for the scientific prevention and control of E. coioides infected with Vibrio alginolyticus.【Method】The pathogen of V. alginolyticus was isolated from the diseased tissues of the dying E. coioides by streak plate method, the morphological characteristics, physiological and biochemical properties, phylogenetic tree, virulence genes and resistance genes as well as drug sensitivity of the pathogen were investigated, and regression infection tests were carried out.【Result】A dominant strain was isolated from the diseased tissues of E. coioides and named as strain SZGP226. The results of phylogenetic analysis showed that, the sequence of strain SZGP226 was homologous to V. alginolyticus HH101307, IB1 and Xmb015, with a similarity of 99.72%. The strain was identified as positive in physiological and biochemical indexes such as sucrose, mannitol, glucose, urea, oxidase activity and V-P, and it also carried five virulence genes, namely, tlh, toxR, ctsA, sto and vpi. The results of drug sensitivity showed that strain SZGP226 was resistant to 9 antibiotics like ampicillin and benzoxacillin, and carried 9 resistance genes including qnrB, floR, tetC, tetM, qnrD, gyrA, Sul1, SHV, and Oxa-24. The screening results of disinfectants showed that, sodium hydroxide had the best bactericidal effect, and its minimal inhibition concentration (MIC) was 8 mg/mL. The regression infection test showed that strain SZGP226 was strongly pathogenic to the E. coioides, with a LC50 of 1.63×107 CFU/mL, and it could cause obvious pathological damage to the liver, kidney and spleen of the E. coioides.【Conclusion】V. alginolyticus was the main pathogenic bacterium for the sudden death of the E. coioides, and its causative genes included tlh, toxR, ctsA, sto and vpi, and the LC50 was 1.63×107 CFU/mL. Sodium hydroxide (3 mg/mL) showing the best bactericidal effect on strain bactericidal effect could be selected to clean up the culture environment before aquaculture was carried out.

【Objective】The study was conducted to investigate the causes of sudden death of largemouth bass (Micropterus salmoides) in a bass culture base in Shenzhen-Shanwei Special Cooperation Zone, and reveal the biological characteristics of the Pathogen, with an aim to provide theoretical reference for disease prevention, treatment and prevention in breeding largemouth bass.【Method】A dominant bacterial strain, named SZNH-S20230817, was isolated from the liver, spleen, intestine and lesion tissues of the diseased largemouth bass, and the isolate was subsequently subjected to morphological characteristics observation, physiological and biochemical characteristics identification, artificial infection experiment, drug sensitivity test, MIC and MBC determination, 16S rRNA sequencing, identification of resistance genes and virulence genes to analyse its drug resistance and pathology.【Result】The results showed that the extended 16S rRNA sequence of SZNH-S20230817 had a 99.38% homology with Aeromonas caviae AP019195, and also carried four virulence genes, including aer, act, lip and fla. The results of physiological and biochemical identification showed that, indole, oxidase, phenylalanine, mannitol, xylose, arginine dihydrolysate and glucose acid production of the strain were positive, and the strain SZNH-S20230817 was identified as Aeromonas caviae. Regression infection revealed that the strain SZNH-S20230817 was highly pathogenic, with an LC50 of 1.39×107 CFU/mL for largemouth bass. The results of drug sensitivity tests showed that the bacterium was resistant to 12 antibiotics such as cefazolin, kanamycin, tetracycline and vancomycin, ampicillin, and clindamycin, and also carried 13 resistance genes such as qnrS, catI, emrB, Sul1, rmtB and etc. Histopathological analysis showed that infection with the strain SZNH-S20230817 could cause swelling and freeing of splenocytes, fracture and rupture of intestinal mucosa, shedding of villi, necrosis of renal cells, infiltration of inflammatory cells and vacuolisation of hepatic cells in the fish. The results of in vitro bacterial inhibition tests showed that the bacterium had low MIC and MBC against rheum officinale and gallnut, and the remaining seven herbs had weak inhibition against the bacterium.【Conclusion】Aeromonas caviae is the pathogen that causes the sudden death of largemouth bass, which carries four virulence genes of aer, act, lip and fla, and has an LC50 of 1.39×107 CFU/mL against largemouth bass. It is resistant to antibiotics such as cefazolin, kanamycin and tetracycline. Low dose of rheum officinale and gallnut can inhibit the growth of the bacterium effectively, with good preventive and inhibitory effects.

In order to improve the problem of serious dust pollution in the fully-mechanized mining face, a new air distribution dust control method with air separator installed was proposed for the fully-mechanized mining face of Daliuta Coal Mine in Yulin, Shaanxi Province. Fluent was used to simulate the dust diffusion rule of the new-type air distribution dust control method, and the influences of different distances between the air pressure outlet and the working face and different pressure-pumping ratios on the change of dust diffusion rule were analyzed. When the air pressure outlet is 10-20 m away from the working face, dust accumulates and settles in the bottom of the roadway in the working face area, and the dust concentration is low at the driver’s side, and the dust control effect is better. When the distance between the air pressure outlet and the working face is 10 m and the pressure-pumping ratio is 1.0, the high concentration of dust accumulates in the working face area, and the dust concentration at the driver is controlled below 50 mg/m3, and the dust control effect is better. Through field measurement, the error between dust concentration and simulation results is less than 15.2%.

Abstract Fractional flow reserve (FFR) has become the gold standard for evaluating coronary lesion-specific ischemia. However, FFR is an invasive method that may cause possible complications in the coronary artery and requires expensive equipment, which limits its use. Promising noninvasive diagnostic methods, such as computed tomography angiography–derived FFR (CT-FFR) and the quantitative flow ratio (QFR), have been proposed. In this study, we evaluated the diagnostic performance of the QFR and CT-FFR in predicting coronary lesion-specific ischemia, with the FFR serving as the reference standard. Patients with suspected or known coronary artery disease who underwent coronary CT angiography revealing 30–90% diameter stenosis in the main coronary artery (≥ 2.0 mm reference diameter) were enrolled. The FFR was measured during invasive coronary angiography (within 15 days after coronary CT angiography). An FFR ≤ 0.8 was the reference standard for coronary lesion-specific ischemia. A total of 103 vessels from 92 consecutive patients (aged 59.8 ± 9.2 years; 60.9% were men) were evaluated. The diagnostic performance of a QFR ≤ 0.80 for predicting coronary lesion-specific ischemia demonstrated good diagnostic accuracy, sensitivity, and specificity (92.2%, 87.2%, and 96.4%, respectively), with an area under the receiver operating characteristic curve (AUC) of 0.987 (P

Abstract Background Despite the use of targeted therapeutic approaches, T-cell acute lymphoblastic leukemia (T-ALL) is still associated with a high incidence of complications and a poor prognosis. Indisulam (also known as E7070), a newly identified molecular glue compound, has demonstrated increased therapeutic efficacy in several types of cancer through the rapid degradation of RBM39. This study aimed to evaluate the therapeutic potential of indisulam in T-ALL, elucidate its underlying mechanisms and explore the role of the RBM39 gene. Methods We verified the anticancer effects of indisulam in both in vivo and in vitro models. Additionally, the construction of RBM39-knockdown cell lines using shRNA confirmed that the malignant phenotype of T-ALL cells was dependent on RBM39. Through RNA sequencing, we identified indisulam-induced splicing anomalies, and proteomic analysis helped pinpoint protein changes caused by the drug. Comprehensive cross-analysis of these findings facilitated the identification of downstream effectors and subsequent validation of their functional roles. Results Indisulam has significant antineoplastic effects on T-ALL. It attenuates cell proliferation, promotes apoptosis and interferes with cell cycle progression in vitro while facilitating tumor remission in T-ALL in vivo models. This investigation provides evidence that the downregulation of RBM39 results in the restricted proliferation of T-ALL cells both in vitro and in vivo, suggesting that RBM39 is a potential target for T-ALL treatment. Indisulam’s efficacy is attributed to its ability to induce RBM39 degradation, causing widespread aberrant splicing and abnormal translation of the critical downstream effector protein, THOC1, ultimately leading to protein depletion. Moreover, the presence of DCAF15 is regarded as critical for the effectiveness of indisulam, and its absence negates the ability of indisulam to induce the desired functional alterations. Conclusion Our study revealed that indisulam, which targets RBM39 to induce tumor cell apoptosis, is an effective drug for treating T-ALL. Targeting RBM39 through indisulam leads to mis-splicing of pre-mRNAs, resulting in the loss of key effectors such as THOC1.

Abstract Objectives The geographical environment and military activities in the plateau area pose potential work-related stressors for military personnel, leading to burnout which is an external manifestation of internal energy exhaustion caused by stress. Without countermeasures, this can result in serious military problems. This study aims to examine the association between burnout and occupational stressors among military personnel stationed in the plateau area of China. Material and methods A stratified randomized cluster sampling survey was conducted among 2026 military personnel from 6 different troops stationed in the plateau area of China. The Chinese Maslach Burnout Inventory-General Survey(MBI-GS in Chinese) was administered from March 2022 to December 2023, and data were analyzed using SPSS version 25. Results A total of 2026 military personnel participated in the survey. The mean overall burnout score was 3.37 ± 0.73, with emotional exhaustion at 2.69 ± 0.89, depersonalization at 3.58 ± 0.92, and professional achievement at 3.81 ± 0.85 levels respectively reported by participants on average scale scores ranging from zero to six. Severe level of burnout was reported by 43.2% of participants while medium level of burnout was reported by 54 .3%. Age, education level, length of military service, and household income were identified as important factors influencing burnout. Conclusion This study highlights a relatively high prevalence of burnout among military personnel stationed in plateau areas necessitating attention towards their occupational health particularly focusing on working hours and economic aspects so as to formulate effective policies and implement intervention measures that strengthen career development for soldiers deployed in such regions.

Al–Li alloy has been widely used in the manufacture of aerospace aircraft structural parts due to its high specific strength and excellent comprehensive performance. Friction stir welding (FSW), as a solid-state joining technique, generates comparatively lower heat generation in contrast to fusion welding. However, even with this reduced heat input, the welded joints of 2195 Al–Li alloy still exhibit noticeable thermal softening. FSW is very sensitive to welding heat input, and the thermal cycle of high peak temperature coarsens the grain in the weld zone, thereby reducing joint properties. To overcome these problems, external cooling can be applied to reduce peak temperature and increase cooling speed, improving the performance of FSW joints. In order to study the mechanism of external cooling on the microstructure evolution of joints, this work established a multi-scale simulation of splat cooling assisted friction stir welding (SCaFSW) of 2195 Al–Li alloy. The temperature change and material deformation data in SCaFSW were calculated in the macroscopic finite element model and used for further Monte Carlo microstructure evolution simulation, and compared with the conventional FSW process. Results reveal that the splat cooling significantly reduces the area of the high temperature region of the workpiece, decreasing the temperature at the same location 10 mm behind the tool by approximately 50 °C compared to FSW. While the local temperature field around the tool pin is not affected by the splat cooling, which mainly accelerates the cooling rate of the welded joint and reduces the plastic strain. In addition, the dislocation density in the weld nugget zone (WNZ) of SCaFSW joint is higher than that in conventional FSW, which promotes the process of grain nucleation and finally refines the grain in the WNZ. The simulated average grain size and thermal cycling results are in good agreement with the experimental results.

Abstract Background Recently, the application of deep learning (DL) has made great progress in various fields, especially in cancer research. However, to date, the bibliometric analysis of the application of DL in cancer is scarce. Therefore, this study aimed to explore the research status and hotspots of the application of DL in cancer. Methods We retrieved all articles on the application of DL in cancer from the Web of Science database Core Collection database. Biblioshiny, VOSviewer and CiteSpace were used to perform the bibliometric analysis through analyzing the numbers, citations, countries, institutions, authors, journals, references, and keywords. Results We found 6,016 original articles on the application of DL in cancer. The number of annual publications and total citations were uptrend in general. China published the greatest number of articles, USA had the highest total citations, and Saudi Arabia had the highest centrality. Chinese Academy of Sciences was the most productive institution. Tian, Jie published the greatest number of articles, while He Kaiming was the most co-cited author. IEEE Access was the most popular journal. The analysis of references and keywords showed that DL was mainly used for the prediction, detection, classification and diagnosis of breast cancer, lung cancer, and skin cancer. Conclusions Overall, the number of articles on the application of DL in cancer is gradually increasing. In the future, further expanding and improving the application scope and accuracy of DL applications, and integrating DL with protein prediction, genomics and cancer research may be the research trends.

In order to study the effects of reverse transcriptase inhibitors, protease inhibitors and flavonoids on two target cells infected by HIV in a heterogeneous environment, an HIV mathematical model at the cellular level was established. Research shows that infected cells can be categorized into immature infected cells, latent infected cells, and mature infected cells based on the infection process. The basic reproduction number $ R_{0} $ was established, and it is proved that $ R_{0} $ serves as a threshold parameter: When $ R_{0} < 1 $, the disease-free steady state is globally asymptotically stable, and the disease is extinct; when $ R_{0} > 1 $, the solution of the system is uniformly persistent, and the virus exists. Considering the huge advantages of drug intervention in controlling HIV infection, the optimal control problem was proposed under the condition that the constant diffusion coefficient is positive, so as to minimize the total number of HIV-infected cells and the cost of drug treatment. To illustrate our theoretical results, we performed numerical simulations in which the model parameters were obtained with reference to some medical studies. The results showed that: (1) as $ R_{0} $ increases, the risk of HIV transmission increases; (2) pharmacological interventions are important in early treatment of HIV spread and control of viral load in the body; (3) the treatment process must consider the heterogeneity of medication, otherwise it will not be conducive to suppressing the spread of the virus and will increase costs.

To address air pollution and offer a convenient and comfortable living environment, the Chinese government launched a smart city pilot (SCP) project in 2012, accompanied by a comprehensive set of environmental and energy-related laws and regulations. Although academic interest in smart cities has surged, there remains a notable gap in empirical research exploring the economic, environmental, and energy effects of such initiatives. Taking 232 prefecture-level cities from 2003 to 2017 as research subjects, this study measures energy efficiency by using energy consumption per unit of GDP and adopts a difference-in-differences (DID) analysis to investigate the impact of SCPs on energy efficiency. The empirical results indicate that SCPs improved energy efficiency by promoting urban technological innovation capabilities and green total factor productivity, and this effect was more pronounced in cities that were more dependent on traditional fossil fuel energy sources and had more developed fiscal and financial levels. Studying the impact of smart city construction on energy utilization efficiency in developing countries, such as China, is not only significantly enlightening for China’s green and low-carbon transition but also provides reference opinions for constructing smart cities and the path to enhancing energy efficiency in other developing countries. The findings provide valuable insights into the global development of smart cities, urban sustainability, and high-quality economic growth.

Abstract Background Flavivirus is a challenge all over the world. The replication of flavivirus takes place within membranous replication compartments (RCs) derived from endoplasmic reticulum (ER). Flavivirus NS1 proteins have been proven essential for the formation of viral RCs by remodeling the ER. The glycosylation of flavivirus NS1 proteins is important for viral replication, yet the underlying mechanism remains unclear. Methods HeLa cells were used to visualize the ER remodeling effects induced by NS1 expression. ZIKV replicon luciferase assay was performed with BHK-21 cells. rZIKV was generated from BHK-21 cells and the plaque assay was done with Vero Cells. Liposome co-floating assay was performed with purified NS1 proteins from 293T cells. Results We found that the glycosylation of flavivirus NS1 contributes to its ER remodeling activity. Glycosylation deficiency of NS1, either through N-glycosylation sites mutations or tunicamycin treatment, compromises its ER remodeling activity and interferes with viral RCs formation. Disruption of NS1 glycosylation results in abnormal aggregation of NS1, rather than reducing its membrane-binding activity. Consequently, deficiency in NS1 glycosylation impairs virus replication. Conclusions In summary, our results highlight the significance of NS1 glycosylation in flavivirus replication and elucidate the underlying mechanism. This provides a new strategy for combating flavivirus infections.

This study sought to determine the effects of rosemary leaf powder (RP) on laying performance, egg quality, serum indices, gut barrier function, and cecal microbiota and metabolites of late-phase laying hens. A total of 84 “Jing Tint 6” laying hens at 65-week old were randomly divided into 2 groups and fed either a basal diet (CON) or a basal diet supplemented with 0.3% RP.
