Background Data storage tags (DSTs) record and store information about animals and their environment, and can provide important data relevant to fish culture, ecology and conservation. A DST has recently been developed that records heart rate (fH), electrocardiograms (ECGs), tri-axial acceleration and temperature. However, at the time of this study, no research using these tags had been performed on fish or determined the quality of the data collected. Thus, our research asked: do these DSTs provide reliable and meaningful data? To examine this question, Atlantic salmon (1.4 ± 0.7 kg) were implanted with DSTs, then swam at increasing speeds in a swim tunnel after 1 week of recovery. Further, in two separate experiments, salmon (2.4 ± 0.1 kg) were implanted with DSTs and held in a large tank with conspecifics for 1 week at 11 °C or 6 weeks at 8–12 °C. Results External acceleration (EA) and variation in EA (VAR) increased exponentially with swimming speed and tail beat frequency. The quality index (QI) assigned to ECG recordings (where QI0 means very good quality, and QI1, QI2 and QI3 are of reduced quality) did not change significantly with increasing swimming speed (QI0 ~ 60–80%). However, we found that the accuracy of the tag algorithm in estimating fH from ECGs was reduced when QI>0. Diurnal patterns of fH and EA were evident from the time the salmon were placed in the tank. Heart rate appeared to stabilize by ~ 4 days post-surgery in the first experiment, but extended holding showed that fH declined for 2–3 weeks. During extended holding, the tag had difficulty recording low fH values fH, it is highly recommended that ECGs be saved when possible for quality control and so that fH values with QI>0 can be manually calculated. Conclusions With these DSTs, parameters of acceleration can be used to monitor the activity of free-swimming salmon. Further, changes in fH and heart rate variability (HRV) due to diurnal rhythms, and in response to temperature, activity and stressors, can be recorded.

Background Behaviour of potential host fish during chemical treatment against the ectoparasite Gyrodactylus salaris is a vital factor in designing treatment strategies, evaluating risk factors and establishing insights into previously failed treatments. The effectiveness of any chemical treatment may be compromised if fish either are forced to, or seek out actively, areas of the river where the water quality is less affected by the chemicals. The aim of this study was to develop and apply an acoustic fish tag for fish localization with sensors for in situ measurement of water conductivity and temperature to investigate fish behaviour before, during and after an aluminium (Al) treatment. The sensor tag allowed discrimination between water qualities, and thereby quantification of exposure to treatment water. Findings Adult Atlantic salmon and anadromous brown trout from river Lærdalselva were tagged with external conductivity transmitters and followed daily by a network of passive receivers and by manual tracking 1 week ahead of treatment, during a 2-week aluminium (Al) treatment period and one week after an Al treatment. The results show no avoidance behaviour related to the Al treatment and most of the fish exhibited a behaviour during the treatment that did not differ significantly from the behaviour observed before or after the treatment. Data collected from the tags showed that the fish experienced increased conductivity during Al administration, suggesting successful exposure to treatment water. The tag gave verifiable environmental information and functioned well in the turbulent and acoustically demanding river environment, albeit with variable detection range. Conclusions The conductivity and temperature tag provided novel data on fish behaviour and exposure during the Al treatment period. Results show that fish exhibit normal behaviour during this period and no avoidance response can be detected in the collected data.

BackgroundThe establishment of Burmese pythons (Python bivittatus)in Everglades National Park, Florida, USA, has been connected to a > 90% decline in the mesomammal population in the park and is a major threat to native reptile and bird populations. Efforts to control this population are underway, but are hampered by a lack of information about fine-scale activity cycles and ecology of these cryptic animals in the wild. We aimed to establish a technique for monitoring the activity of Burmese pythons in the wild using acceleration data loggers (ADLs), while attempting to identify any behavioral patterns that could be used to help manage this invasive species in the Greater Everglades and South Florida.ResultsWe obtained continuous acceleration and temperature data from four wild snakes over periods of 19 to 95 days (mean 54 ± 33 days). Snakes spent 86% of their time at rest and 14% of their time active, including transiting between locations. All snakes showed at least one period of continuous transiting lasting 10 h or more, with one animal transiting continuously for a period of 58.5 h. Acceleration data logger-derived transiting bout duration was significantly correlated with the distance snakes traveled per hour for two snakes that also carried GPS loggers. Snakes were most active in midday or early-night depending on individual and time of year, but all snakes were least likely to be active in the early mornings (0400–0700 h local time). Very little movement took place at temperatures below 14 °C or above 24 °C, with most movement taking place between 15° and 20 °C. One animal showed a highly unusual rolling event that may be indicative of a predation attempt, but this could not be confirmed.ConclusionsFine-scale activity and some behaviors were apparent from ADL data, making ADLs a potentially valuable, unbiased tool for monitoring large-bodied snakes in the wild. Snakes spent the majority of their time resting, but also moved continuously for several hours at a time during bouts of transiting. Results suggest that individuals may shift their diel activity pattern based on season. Understanding seasonal differences in activity levels can improve the accuracy of population estimates, help detect range expansion, and improve managers’ ability to find and capture individuals.

Recent advances in tracking systems have revolutionized our ability to study animal movement in the wild. In aquatic environments, high-resolution acoustic telemetry systems make it technically possible to simultaneously monitor large amounts of individuals at unprecedented spatial and temporal resolutions, providing a unique opportunity to study the behaviour and social interactions using a reality mining approach. Despite the potential, high-resolution telemetry systems have had very limited use in coastal marine areas due to the limitations that these environments pose to the transmission of acoustic signals. This study aims at designing and testing a high-resolution acoustic telemetry system to monitor, for the first time, a natural fish population in an open marine area. First, we conducted preliminary range tests and a computer simulation study to identify the optimal design of the telemetry system. Then, we performed a series of stationary and moving tests to characterize the performance of the system in terms of positioning efficiency and precision. Finally, we obtained a dataset corresponding to the movements of 170 concurrently tagged individuals to demonstrate the overall functioning of the system with a real study case of the behaviour of a small-bodied coastal species. Our results show that high-resolution acoustic telemetry systems efficiently generate positional data in marine systems, providing a precision of few meters, a temporal resolution of few seconds, and the possibility of tracking hundreds of individuals simultaneously. Data post-processing using a trajectory filter and movement models proved to be key to achieve a sub-meter positioning precision. The main limitation detected for our system was the restricted detection range, which was negatively affected by the stratification of the water column. Our work demonstrates that high-resolution acoustic telemetry systems are an effective method to monitor the movements of free-ranging individuals at the population level in coastal sites. By providing highly precise positioning estimates of large amounts of individuals, these systems represent a powerful tool to study key ecological processes regarding the social interactions of individuals, including social dynamics, collective movements, or responses to environmental perturbations, and to extend the studies to poorly studied small-sized species or life-stages., The telemetry system was financed by the German Federal Ministry of Education and Research (grant no. #033W024A). Background Fish telemetry using electronic transmitter or data storage tags has become a common method for studying free-swimming fish both in the wild and in aquaculture. However, fish used in telemetry studies must be handled, anaesthetised and often subjected to surgical procedures to be equipped with tags, processes that will shift the fish from their normal physiological and behavioural states. In many projects, information is needed on when the fish has recovered after handling and tagging so that only the data recorded after the fish has fully recovered are used in analyses. We aimed to establish recovery times of adult Atlantic salmon (Salmo salar) after an intraperitoneal tagging procedure featuring handling, anaesthesia and surgery. Results Based on ECG and accelerometer data collected with telemetry from nine individual Atlantic salmon during the first period after tagging, we found that heart rate was initially elevated in all fish and that it took an average of ≈ 4 days and a maximum of 6 days for heart rate to return to an assumed baseline level. One activity tag showed no consistent decline in activity, and two others did not show strong evidence of complete recovery by the end of the experiment: baseline levels of the remaining tags were on average reached after ≈ 3.3 days. Conclusion Our findings showed that the Atlantic salmon used in this study required an average of ≈ 4 days, with a maximum interval of 6 days, of recovery after tagging before tag data could be considered valid. Moreover, the differences between recovery times for heart rate and activity imply that recovery time recommendations should be developed based on a combination of indicators and not just on e.g. behavioural observations. © The Author(s) 2021. Background With the increase in telemetry studies over the past decade, improving understanding of how different tagging methods influence the probability of presence in a receiver array is important in maximizing the resulting data. Disappearance from the array may be due to mortality from surgery complications, tag loss, predation, or emigration. Internally tagging fish on a boat can cause barotrauma injuries, increased stress from prolonged handling times, or predation after fish have been released back into the water. Conducting in situ internal acoustic tagging at depth of capture removes barotrauma stresses and simplifies the release method, which may improve fish survival and decrease risk of disappearance from the array. In this study, we used 8 years of acoustic tagging data to determine if the tagging method (in situ versus on the boat) influenced the likelihood of a fish being detected 4 and 6 days after the tagging event. Results At 6 days after tagging, Kaplan–Meier survival curves revealed that the probability of presence for fish tagged on the boat was 66% compared to 90% for fish tagged in situ. Tagging method was the only variable to significantly affect probability of presence based on Cox proportional hazards models, with fish tagged in situ ~ 75% less likely to disappear from the array compared to fish tagged on the boat at both 4 and 6 days after tagging. Examining tagging methods separately, handling time only marginally influenced probability of presence of boat-tagged fish and no variables had a significant effect on probability of presence of in situ tagged fish. Conclusions In this study, tagging method was the only variable to significantly affect the probability of presence for internally tagged fish. Other factors had little to no influence, but correlation of variables limited what factors could be included in the models. Implanting internal acoustic tags in situ is not a practical method for every species and for every environment, but given the increased probability of presence demonstrated here, we strongly suggest it be considered where applicable.

Objective: This study was conducted to investigate the differences in several serum adipokines in perinatal dairy cows with type I and II ketosis, and the correlations between these adipokines and the two types of ketosis.Methods: Serum adiponectin (ADP), leptin (LEP), resistin, tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) levels, and energy balance indicators related to ketosis were measured. Type I and II ketosis were distinguished by serum glucose (Glu) and Y values and the correlations between adipokines in the two types of ketosis were analyzed.Results: β-Hydroxybutyric acid of type I ketosis cows was significantly negatively correlated with insulin (INS) and LEP and had a significant positive correlation with serum ADP. In type II ketosis cows, ADP and LEP were significantly negatively correlated, and INS and resistin were significantly positively correlated. Revised quantitative INS sensitivity check index (RQUICKI) values had a significantly positive correlation with ADP and had a very significant and significant negative correlation with resistin, TNF-α, and IL-6. ADP was significantly negatively correlated with resistin and TNF-α, LEP had a significantly positive correlation with TNF-α, and a significantly positive correlation was shown among resistin, IL-6, and TNF-α. There was also a significant positive correlation between IL-6 and TNF-α.Conclusion: INS, ADP, and LEP might exert biological influences to help the body recover from negative energy balance, whereas resistin, TNF-α, and IL-6 in type II ketosis cows exacerbated INS resistance and inhibited the production and secretion of ADP, weakened INS sensitivity, and liver protection function, and aggravated ketosis.

Objective: Tibetan pigs, an excellent species unique to China, face serious threats, which in turn affects the development and utilization of the outstanding advantages of plateau hypoxia adaptability and reduces their genetic diversity. Therefore, a discussion of measures to conserve this genetic resource is necessary. The method, based on genetic diversity, genetic divergence and total genetic contribution rate of population, reflects the priority conservation order and varies depending on the three different purposes of conservation.Methods: We analyzed mitochondrial DNA control region (D-loop) variation in 1,201 individuals from nine Tibetan pig populations across five provinces and downloaded 564 mtDNA D-loop sequences from three indigenous pig breeds in Qinghai, Sichuan, and Yunnan Provinces distributed near the Tibetan pigs.Results: We analyzed three different aspects: Changdu Tibetan pigs have the highest genetic diversity, and from the perspective of genetic diversity, the priority conservation is Changdu Tibetan pigs. Hezuo Tibetan pigs have the highest genetic contribution, so the priority conservation is Hezuo Tibetan pigs in the genetic contribution aspect. Rkaze Tibetan pigs were severely affected by indigenous pig breeds, so if considering from the perspective of introgression, the priority conservation is Rkaze Tibetan pigs.Conclusion: This study evaluated genetic diversity and comprehensively assessed conservation priority from three different aspects in nine Tibetan pig populations.

Objective: The present study aimed to survey seasonal changes in reproductive performance of local cows receiving artificial insemination (AI) in the Pursat province of Cambodia, a tropical country, to investigate if ambient conditions affect the reproductive performance of cows as to better understand the major problems regarding cattle production.Methods: The number of cows receiving AI, resultant number of calving, and calving rate were analyzed for those receiving the first AI from 2016 to 2017. The year was divided into three seasons: cool/dry (from November to February), hot/dry (from March to June), and wet (from July to October), based on the maximal temperature and rainfall in Pursat, to analyze the relationship between ambient conditions and the reproductive performance of cows. Body condition scores (BCS) and feeding schemes were also analyzed in these seasons.Results: The number of cows receiving AI was significantly higher in the cool/dry season than the wet season. The number of calving and calving rate were significantly higher in cows receiving AI in the cool/dry season compared with the hot/dry and wet seasons. The cows showed higher BCSs in the cool/dry season compared to the hot/dry and wet seasons probably due to the seasonal changes in the feeding schemes: these cows grazed on wild grasses in the cool/dry season but fed with a limited amount of grasses and straw in the hot/dry and wet seasons.Conclusion: The present study suggests that the low number of cows receiving AI, low number of calving, and low calving rate could be mainly due to poor body condition as a result of the poor feeding schemes during the hot/dry and wet seasons. The improvement of body condition by the refinement of feeding schemes may contribute to an increase in the reproductive performance in cows during the hot/dry and wet seasons in Cambodia.

Objective: This study aimed to determine the effect of intermittent feeding and cold water on performance and carcass traits in broiler chickens between 4 to 6 wk of age exposed to daily high temperature.Methods: Broilers were assigned to four treatment groups according to a 2×2 factorial design between 22 to 42 d of age (80 broilers per treatment, 4 replications). Broilers were divided into two main groups as feeding type (ad-libitum [AL] and intermittent [IF] for 6 h daily) and sub-groups as water temperature (normal [NW], 24.9°C and cold [CW], 16.4°C). Heat treatment was applied between 11.00 to 17.00 h daily between 22 to 42 d of age.Results: Live weight at 6th wk was not affected by feeding type and water temperature, but the live weight was significantly higher in IF chickens at the 5th wk (p0.05). Water temperature had no significant effect on heart, liver, gizzard, and abdominal fat percentages (p>0.05).Conclusion: It is concluded that IF increased the average weekly gain in chickens reared under daily heat stress for 6 h between 22 to 42 d of age. IF in hot environmental conditions slightly increased performance without adversely affecting health, welfare, and physiological traits, whereas CW implementation had no significant effect on performance. It can also be said that IF suppresses a sudden increase in body temperature depending on age and live weight.

Vitamins and minerals categorized as micronutrients are the essential components of animal feed for maintaining health and improving immunity. Micronutrients are important bioactive molecules and cofactors of enzymes as well. Besides being cofactors for enzymes, some vitamins such as the fat-soluble vitamins, vitamin A and D have been shown to exhibit hormone-like functions. Although they are required in small amount, they play an influential role in the proper functioning of a number of enzymes which are involved in many metabolic, biochemical and physiological processes that contribute to growth, production and health. Micronutrients can potentially have a positive impact on bone health, preventing bone loss and fractures, decreasing bone resorption and increasing bone formation. Thus, micronutrients must be provided to livestock in optimal concentrations and according to requirements that change during the rapid growth and development of the animal and the production cycle. The supply of nutrients to the animal body not only depends on the amount of the nutrient in a food, but also on its bioavailability. The bioavailability of these micronutrients is affected by several factors. Therefore, several technologies such as nanoparticle, encapsulation, and chelation have been developed to improve the bioavailability of micronutrients associated with bone health. The intention of this review is to provide an updated overview of the importance of micronutrients on bone health and methods applied to improve their bioavailability.

Objective: Quantitative polymerase chain reaction (qPCR) has been extensively used in the field of mesenchymal stem cell (MSC) research to elucidate their characteristics and clinical potential by normalization of target genes against reference genes (RGs), which are believed to be stably expressed irrespective of various experimental conditions. However, the expression of RGs is also variable depending on the experimental conditions, which may lead to false or contradictory conclusions upon normalization. Due to the current lack of information for a clear list of stable RGs in bovine MSCs, we conducted this study to identify suitable RGs in bovine MSCs.Methods: The cycle threshold values of ten traditionally used RGs (18S ribosomal RNA [18S], beta-2-microglobulin [B2M], H2A histone family, member Z [H2A], peptidylprolyl isomerase A [PPIA], ribosomal protein 4 [RPL4], succinate dehydrogenase complex, subunit A [SDHA], beta actin [ACTB], glyceraldehyde-3-phosphate dehydrogenase [GAPDH], TATA box binding protein [TBP], and hypoxanthine phosphoribosyltrasnfrase1 [HPRT1]) in bovine bone marrow-derived MSCs (bBMMSCs) were validated for their stabilities using three types of RG evaluation algorithms (geNorm, Normfinder, and Bestkeeper). The effect of validated RGs was then verified by normalization of lineage-specific genes (fatty acid binding protein 4 [FABP4] and osteonectin [ON]) expressions during differentiations of bBMMSCs or POU class 5 homeobox 1 (OCT4) expression between bBMMSCs and dermal skins.Results: Based on the results obtained for the three most stable RGs from geNorm (TBP, RPL4, and H2A), Normfinder (TBP, RPL4, and SDHA), and Bestkeeper (TBP, RPL4, and SDHA), it was comprehensively determined that TBP and RPL4 were the most stable RGs in bBMMSCs. However, traditional RGs were suggested to be the least stable (18S) or moderately stable (GAPDH and ACTB) in bBMMSCs. Normalization of FABP4 or ON against TBP, RPL4, and 18S presented significant differences during differentiation of bBMMSCs. However, although significantly low expression of OCT4 was detected in dermal skins compared to that in bBMMSCs when TBP and RPL4 were used in normalization, normalization against 18S exhibited no significance.Conclusion: This study proposes that TBP and RPL4 were suitable as stable RGs for qPCR study in bovine MSCs.

Objective: The aim of this study was to compare overfeeding performance, fatty acid composition, blood chemistry, enzymes and genes expression overfed Xupu and Landes geese.Methods: Sixty male Xupu geese (80 d) and Landes geese (80 d) were selected. After a period of one-week of pre-overfeeding, Xupu and Landes geese were overfed three meals of 550 and 350 g/d, respectively, of a high-carbohydrate diet in the first week of the overfeeding period. The next week, geese were given four meals of 1,200 and 850 g/d, respectively, over 8 to 14 d. Background The blue shark Prionace glauca is a highly migratory species with a circumglobal distribution. Mean movement rate, defined by the horizontal tracking distance between two data points over the duration of time, is commonly used to understand the horizontal displacement of highly migratory species across a wide range. However, the estimation of mean movement rates for blue sharks has never been conducted using a statistical model. We therefore investigated the mean movement rates using a generalized linear mixed model with data from satellite tags to estimate the range of mean movement rates for 10 blue sharks in the northwestern Pacific Ocean and to reveal the interaction of mean movement rate with several factors. Results (1) Estimations of mean movement rates for the 10 blue sharks were significantly influenced by behavioral differences among individuals; (2) uncertainty in the estimation (i.e., predictive and confidence intervals) of mean movement rates for these blue sharks was larger over shorter time periods, and (3) the predictive intervals of mean movement rates for the sharks ranged widely from 0.33 to 5.02 km/h. Conclusion Blue sharks are considered to opportunistically change their mean movement rates regardless of differences in sex, movement direction, or season.

Objective: Estimating the genetic diversity and structures, both within and among chicken breeds, is critical for the identification and conservation of valuable genetic resources. In chickens, microsatellite (MS) marker polymorphisms have previously been widely used to evaluate these distinctions. Our objective was to analyze the genetic diversity and relationships among 22 chicken breeds in Asia based on allelic frequencies.Methods: We used 469 genomic DNA samples from 22 chicken breeds from eight Asian countries (South Korea, KNG, KNB, KNR, KNW, KNY, KNO; Laos, LYO, LCH, LBB, LOU; Indonesia, INK, INS, ING; Vietnam, VTN, VNH; Mongolia, MGN; Kyrgyzstan, KGPS; Nepal, NPS; Sri Lanka, SBC) and three imported breeds (RIR, Rhode Island Red; WLG, White Leghorn; CON, Cornish). Their genetic diversity and phylogenetic relationships were analyzed using 20 MS markers.Results: In total, 193 alleles were observed across all 20 MS markers, and the number of alleles ranged from 3 (MCW0103) to 20 (LEI0192) with a mean of 9.7 overall. The NPS breed had the highest expected heterozygosity (Hexp, 0.718±0.027) and polymorphism information content (PIC, 0.663±0.030). Additionally, the observed heterozygosity (Hobs) was highest in LCH (0.690±0.039), whereas WLG showed the lowest Hexp (0.372±0.055), Hobs (0.384±0.019), and PIC (0.325±0.049). Nei’s DA genetic distance was the closest between VTN and VNH (0.086), and farthest between KNG and MGN (0.503). Principal coordinate analysis showed similar results to the phylogenetic analysis, and three axes explained 56.2% of the variance (axis 1, 19.17%; 2, 18.92%; 3, 18.11%). STRUCTURE analysis revealed that the 22 chicken breeds should be divided into 20 clusters, based on the highest ΔK value (46.92).Conclusion: This study provides a basis for future genetic variation studies and the development of conservation strategies for 22 chicken breeds in Asia.

Objective: The study was conducted to determine the effects of body weight (BW) and fiber sources on nutrient digestibility, fiber fermentation and short chain fatty acids (SCFA) concentration in different intestinal segments of growing pigs fed high-fiber diets.Methods: Nine barrows with initial BW of 25.17±0.73 kg and 9 barrows with initial BW of 63.47±2.18 kg were allotted to a duplicate 9×2 Youden Square design with 3 dietary treatments and 2 periods. The dietary treatments were formulated with 3 different high-fiber ingredients: corn bran, sugar beet pulp, and soybean hulls, respectively. Objective: This study assessed genomic prediction accuracies based on different selection methods, evaluation procedures, training population (TP) sizes, heritability (h2) levels, marker densities and pedigree error (PE) rates in a simulated Korean beef cattle population.Methods: A simulation was performed using two different selection methods, phenotypic and estimated breeding value (EBV), with an h2 of 0.1, 0.3, or 0.5 and marker densities of 10, 50, or 777K. A total of 275 males and 2,475 females were randomly selected from the last generation to simulate ten recent generations. The simulation of the PE dataset was modified using only the EBV method of selection with a marker density of 50K and a heritability of 0.3. The proportions of errors substituted were 10%, 20%, 30%, and 40%, respectively. Genetic evaluations were performed using genomic best linear unbiased prediction (GBLUP) and single-step GBLUP (ssGBLUP) with different weighted values. The accuracies of the predictions were determined.Results: Compared with phenotypic selection, the results revealed that the prediction accuracies obtained using GBLUP and ssGBLUP increased across heritability levels and TP sizes during EBV selection. However, an increase in the marker density did not yield higher accuracy in either method except when the h2 was 0.3 under the EBV selection method. Based on EBV selection with a heritability of 0.1 and a marker density of 10K, GBLUP and ssGBLUP_0.95 prediction accuracy was higher than that obtained by phenotypic selection. The prediction accuracies from ssGBLUP_0.95 outperformed those from the GBLUP method across all scenarios. When errors were introduced into the pedigree dataset, the prediction accuracies were only minimally influenced across all scenarios.Conclusion: Our study suggests that the use of ssGBLUP_0.95, EBV selection, and low marker density could help improve genetic gains in beef cattle.

Objective: The purpose of this study was to reveal the metabolic shift in the fungus cocultured with the methanogen (Methanobrevibacter thaueri).Methods: Gas chromatography-mass spectrometry was used to investigate the metabolites in anaerobic fungal (Pecoramyces sp. F1) cells and the supernatant.Results: A total of 104 and 102 metabolites were detected in the fungal cells and the supernatant, respectively. The partial least squares-discriminant analysis showed that the metabolite profiles in both the fungal cell and the supernatant were distinctly shifted when co-cultured with methanogen. Statistically, 16 and 30 metabolites were significantly (p2-producing and H2-utilizing microorganisms. However, metabolism in hydrogenosome needs to be further studied to gain better insight in the hydrogen transfer among microorganisms.

Objective: The objective of this experiment was to investigate the effects of fat-soluble vitamin injection on plasma and tissue vitamin status in nursery pigs.Methods: A total of 16 pigs (initial body weight: 7.15±1.1 kg) were allotted to 2 treatments at d 7 post-weaning. Pigs were fed a corn-soybean meal-based basal diet with no supplemental vitamin A and i.m. injected with 300,000 IU of retinyl palmitate, 900 IU of d-α-tocopherol and 30,000 IU of vitamin D3 with control pigs having no vitamin injection. Blood (d 0, 3, 7, and 14 post-injection) and tissue samples (liver, brain, heart, lung, and muscle; d 7 and 14 post-injection) were collected from pigs. Retinyl palmitate, retinol, and α-tocopherol concentrations were analyzed in plasma and tissues, while plasma was assayed for 25-hydroxycholecalciferol (25-OHD3).Results: Plasma retinol and 25-OHD3 concentrations increased by the vitamin injection from d 3 to 14 post-injection (p3. As plasma levels decreased post-injection, vitamin A level in liver and vitamin E level in muscle, heart and liver increased. The α-tocopherol found in plasma after injection was distributed to various tissues but retinyl palmitate only to the liver.

Objective: This study was aimed to investigate the muscle-specific beef color stability at normal and high ultimate pHs.Methods: The impact of muscle (Longissimus lumborum [LL] vs psoas major [PM]) and pH (normal ultimate pH [Np] vs high pH dark cutting beef [Hp]) on color stability, indicated by basic color traits, metmyoglobin reducing activity (MRA) and oxygen consumption (OC), as well as the lipid oxidation, were determined over 7 days of display at 4°C.Results: Hp-LL had the highest pH (6.92), followed by Hp-PM (6.01), Np-PM (5.76), and Np-LL (5.52). Hp-LL had increased (p*, chroma and % oxymyoglobin during display. Hp-LL also had the highest metmyoglobin (MMb) reducing activity and OC among all the samples, thus, the greatest color stability, although very dark throughout storage, with lowest values for lightness (L*) and yellowness (b*). Np-LL also exhibited relatively high color stability, as a result of its lower % MMb and OC and higher MRA than psoas muscle samples. The 0.2 unit difference of the pH between Hp and Np psoas muscle, resulted in the difference of the color intensity, not the color stability. Interestingly, high pH psoas muscle (Hp-PM) did not have better color stability than Np-PM, and in fact had lower color stability than even Np-LL. The similar level of OC and lipid oxidation cannot explain the difference in color stability between Hp-PM and Np-LL.Conclusion: The Hp does not always show better color stability compared with Np beef, which depends on the muscle type. The balance of MRA and OC is important to keep the color in great intensity and stability in the meantime.

Objective: This study was conducted to investigate the effects of dietary corn resistant starch (RS) on the intestinal morphology and barrier functions of broilers.Methods: A total of 320 one-day-old broilers were randomly allocated to 5 dietary treatments: one normal corn–soybean (NC) diet, one corn–soybean–based diet supplementation with 20% corn starch (CS), and 3 corn–soybean–based diets supplementation with 4%, 8%, and 12% corn resistant starch (RS) (identified as 4% RS, 8% RS, and 12% RS, respectively). Each group had eight replicates with eight broilers per replicate. After 21 days feeding, one bird with a body weight (BW) close to the average BW of their replicate was selected and slaughtered. The samples of duodenum, jejunum, ileum, caecum digesta, and blood were collected.Results: Birds fed 4% RS, 8% RS and 12% RS diets showed lower feed intake, BW gain, jejunal villus height (VH), duodenal crypt depth (CD), jejunal VH/CD ratio, duodenal goblet cell density as well as mucin1 mRNA expressions compared to the NC group, but showed higher concentrations of cecal acetic acid and butyric acid, percentage of jejunal proliferating cell nuclear antigen-positive cells and delta like canonical Notch ligand 4 (Dll4), and hes family bHLH transcription factor 1 mRNA expressions. However, there were no differences on the plasma diamine oxidase activity and D-lactic acid concentration among all groups.Conclusion: These findings suggested that RS could suppress intestinal morphology and barrier functions by activating Notch pathway and inhibiting the development of goblet cells, resulting in decreased mucins and tight junction mRNA expression.

The demand for food is increasing day by day because of the increasing global population. Therefore, meat, the easiest and largely available source of protein, needs to be produced in large amounts with good quality. The pork industry is a significant shareholder in fulfilling the global meat demands. Notably, myogenesis- development of muscles during embryogenesis- is a complex mechanism which culminates in meat production. But the molecular mechanisms which govern the myogenesis are less known. The involvement of miRNAs in myogenesis and meat quality, which depends on factors such as myofiber composition and intramuscular fat contents which determine the meat color, flavor, juiciness, and water holding capacity, are being extrapolated to increase both the quantity and quality of pork. Various kinds of microRNAs (miRNAs), miR-1, miR-21, miR22, miR-27, miR-34, miR-127, miR-133, miR-143, miR-155, miR-199, miR-206, miR-208, miR-378, and miR-432 play important roles in pig skeletal muscle development. Further, the quality of meat also depends upon myofiber which is developed through the expression of different kinds of miRNAs at different stages. This review will focus on the mechanism of myogenesis, the role of miRNAs in myogenesis, and meat quality with a focus on the pig.

Objective: To evaluate the pancreatic differentiation potential of α-1,3-galactosyltransferase knockout (GalTKO) pig-derived bone marrow-derived mesenchymal stem cells (BM-MSCs) using epigenetic modifiers with different pancreatic induction media.Methods: The BM-MSCs have been differentiated into pancreatic β-like cells by inducing the overexpression of key transcription regulatory factors or by exposure to specific soluble inducers/small molecules. In this study, we evaluated the pancreatic differentiation of GalTKO pig-derived BM-MSCs using epigenetic modifiers, 5-azacytidine (5-Aza) and valproic acid (VPA), and two types of pancreatic induction media – advanced Dulbecco's modified Eagle's medium (ADMEM)-based and N2B27-based media. GalTKO BM-MSCs were treated with pancreatic induction media and the expression of pancreas-islets-specific markers was evaluated by real-time quantitative polymerase chain reaction, Western blotting, and immunofluorescence. Morphological changes and changes in the 5'-C-phosphate-G-3' (CpG) island methylation patterns were also evaluated.Results: The expression of the pluripotent marker (POU class 5 homeobox 1 [OCT4]) was upregulated upon exposure to 5-Aza and/or VPA. GalTKO BM-MSCs showed increased expression of neurogenic differentiation 1 in the ADMEM-based (5-Aza) media, while the expression of NK6 homeobox 1 was elevated in cells induced with the N2B27-based (5-Aza) media. Moreover, the morphological transition and formation of islets-like cellular clusters were also prominent in the cells induced with the N2B27-based media with 5-Aza. The higher insulin expression revealed the augmented trans-differentiation ability of GalTKO BM-MSCs into pancreatic β-like cells in the N2B27-based media than in the ADMEM-based media.Conclusion: 5-Aza treated GalTKO BM-MSCs showed an enhanced demethylation pattern in the second CpG island of the OCT4 promoter region compared to that in the GalTKO BM-MSCs. The exposure of GalTKO pig-derived BM-MSCs to the N2B27-based microenvironment can significantly enhance their trans-differentiation ability into pancreatic β-like cells.

Objective: An initial RNA-Sequencing study revealed that UDP-galactose-4-epimerase (GALE) was one of the most promising candidates for milk protein concentration in Chinese Holstein cattle. This enzyme catalyzes the interconversion of UDP-galactose and UDP-glucose, an important step in galactose catabolism. To further validate the genetic effect of GALE on milk protein traits, genetic variations were identified, and genotypes-phenotypes associations were performed.Methods: The entire coding region and the 5’-regulatory region (5’-UTR) of GALE were re-sequenced using pooled DNA of 17 unrelated sires. Association studies for five milk production traits were performed using a mixed linear animal model with a population encompassing 1,027 Chinese Holstein cows.Results: A total of three variants in GALE were identified, including two novel variants (g.2114 A>G and g.2037 G>A) in the 5’-UTR and one previously reported variant (g.3836 G>C) in an intron. All three single nucleotide polymorphisms (SNPs) were associated with milk yield (pC.Conclusion: Overall, our findings provided new insights into the polymorphic variations in bovine GALE gene and their associations with milk protein concentration. The data indicate their potential uses for marker-assisted breeding or genetic selection schemes.

Objective: The objective of the present experiment was to construct self-draining beds to keep surface bedding materials clean and dry for beef cattle comfort in a temperate climate.Methods: In Experiment 1, a self-draining bed was covered with sand at depths of 10 cm (S-10a), 15 cm (S-15), and 20 cm (S-20) respectively. In Experiment 2, self-draining beds of different sizes were covered with 10 cm of sand (S-10b) and wood shavings (WS) at depths of 15 cm and 20 cm (WS-15 and WS-20). Fifteen cattle were engaged to evaluate the comfort of self-draining beds covered with different bedding materials.Results: No cattle lay in the feed alley and cattle spent more time lying on S-10a than S-15 or S-20 in Experiment 1 (p0.05). Accordingly, the aim of this study was to investigate the effects of chicken genotype on the macronutrient content, bioactive peptide content, and antioxidant capacity within different breast meat.Methods: In this experiment, three genotypes of chicken, Thai indigenous, black-boned, and broiler (control), were reared with commercial feed under the same conditions. Thirty chickens were slaughtered at typical market age and the breasts were separated from the carcass to determine macronutrient content using the AOAC method. The antioxidant capacities of the chicken breasts were evaluated by in vitro antioxidant assays and the protein pattern was investigated using gel electrophoresis. Objective: The objective of this study was to determine the digestible energy (DE) and metabolizable energy (ME) of yellow dent corn sourced from different meteorological origins fed to growing pigs and develop equations to predict the DE and ME of yellow dent corn from southwestern China.Methods: Sixty crossbred barrows were allotted to 20 treatments in a triplicate 20×2 incomplete Latin square design with 3 replicated pigs per dietary treatment during 2 consecutive periods. Each period lasted for 12 days, and total feces and urine during the last 5 days of each period were collected to calculate the energy contents.Results: On dry matter (DM) basis, the DE and ME in 20 corn grain samples ranged from 15.38 to 16.78 MJ/kg and from 14.93 to 16.16 MJ/kg, respectively. Selected best-fit prediction equations for DE and ME (MJ/kg DM basis) for yellow dent corn (n = 16) sourced from southwestern China were as follows: DE = 28.58–(0.12×% hemicellulose)+(0.35×% ether extract)–(0.83×MJ/kg gross energy)+(0.20×% crude protein)+(0.49×% ash); ME = 30.42– (0.11×% hemicellulose)+(0.31×% ether extract)–(0.81×MJ/kg gross energy).Conclusion: Our results indicated that the chemical compositions, but not the meteorological conditions or physical characteristics could explain the variation of energy contents in yellow dent corn sourced from southwestern China fed to growing pigs.

Objective MicroRNAs (miRNAs) are the most abundant small RNAs. Approximately 2000 annotated miRNAs genes have been found to be differentially expressed in ovarian follicles during the follicular development. Many miRNAs exert their regulatory effects on the apoptosis of follicular granulosa cells (FGCs) and follicular development (FD). However, accurate roles and mechanism of miRNAs regulating apoptosis of FGCs remain undetermined. Methods In this review, we summarized the regulatory role of each miRNA or miRNA cluster on FGCs apoptosis and FD on the bases of 41 academic articles retrieved from PubMed and web of science and other databases. Results Total of 30 miRNAs and 4 miRNAs clusters in 41 articles were reviewed and summarized in the present article. 29 documents indicated explicitly that 24 miRNAs and miRNAs clusters in 29 articles promoted or induced FGCs apoptosis through their distinctive target genes. The remaining 10 miRNAs and miRNAs of 12 articles inhibited FGCs apoptosis. were reported in all articles. MiRNAs exerted modulation actions by at least 77 signal pathways during FGCs apoptosis and FD. Conclusion We concluded that miRNAs or miRNAs clusters could modulate the apoptosis of GCs (including follicular GCs, mural GCs and cumulus cells) by targeting their specific genes. A great majority of miRNAs show promoting role on apoptosis of FGCs in mammals. But the accurate mechanism of miRNAs and miRNA clusters has not been well understood. It is extremely necessary to ascertain clearly the role and mechanism of each miRNA or miRNA cluster in the future. Understanding precise functions and mechanisms of miRNAs in FGCs apoptosis and FD will be beneficial for finding new diagnostic and treatment strategy or scheme for infertility and ovarian diseases in humans and animals.

Objective: On the hypothesis that grazing of cattle prompts organs to secrete or internalize circulating microRNAs (c-miRNAs) in parallel with changes in energy metabolism, we aimed to clarify biological events in adipose, skeletal muscle, and liver tissues in grazing Japanese Shorthorn (JSH) steers by a transcriptomic approach.Methods: The subcutaneous fat (SCF), biceps femoris muscle (BFM), and liver in JSH steers after three months of grazing or housing were analyzed using microarray and quantitative polymerase chain reaction (qPCR), followed by gene ontology (GO) and functional annotation analyses.Results: The results of transcriptomics indicated that SCF was highly responsive to grazing compared to BFM and liver tissues. The ‘Exosome’, ‘Carbohydrate metabolism’ and ‘Lipid metabolism’ were extracted as the relevant GO terms in SCF and BFM, and/or liver from the >1.5-fold-altered mRNAs in grazing steers. The qPCR analyses showed a trend of upregulated gene expression related to exosome secretion and internalization (charged multivesicular body protein 4A, vacuolar protein sorting-associated protein 4B, vesicle associated membrane protein 7, caveolin 1) in the BFM and SCF, as well as upregulation of lipolysisassociated mRNAs (carnitine palmitoyltransferase 1A, hormone-sensitive lipase, perilipin 1, adipose triglyceride lipase, fatty acid binding protein 4) and most of the microRNAs (miRNAs) in SCF. Moreover, gene expression related to fatty acid uptake and inter-organ signaling (solute carrier family 27 member 4 and angiopoietin-like 4) was upregulated in BFM, suggesting activation of SCF-BFM organ crosstalk for energy metabolism. Meanwhile, expression of plasma exosomal miR-16a, miR-19b, miR-21-5p, and miR-142-5p was reduced. According to bioinformatic analyses, the c-miRNA target genes are associated with the terms ‘Endosome’, ‘Caveola’, ‘Endocytosis’, ‘Carbohydrate metabolism’, and with pathways related to environmental information processing and the endocrine system.Conclusion: Exosome and fatty acid metabolism-related gene expression was altered in SCF of grazing cattle, which could be regulated by miRNA such as miR-142-5p. These changes occurred coordinately in both the SCF and BFM, suggesting involvement of exosome in the SCF-BFM organ crosstalk to modulate energy metabolism.

Objective: Thyroid hormone responsive spot 14 alpha (THRSP) has been used to investigate the regulation of de novo lipogenesis because the variation of THRSP mRNA content in the tissue affects directly the ability of that tissue to synthetize lipids. Also, this gene responds to thyroid hormone stimulation and high level of carbohydrate feeding or insulin-injection. This study was carried out to investigate variations within THRSP and their effects on body and carcass weights in Korean native chicken (KNC).Methods: A total of 585 chickens which represent the five lines of KNC (Black, Gray-Brown, Red-Brown, White, and Yellow-Brown) were reared and body weight data were recorded every two weeks from hatch until 20 weeks of age. Polymerase chain reaction- restriction fragment length polymorphism, DNA chips for Agilent 2100 Bioanalyzer, and Fluidigm Genotyping Technology, were applied to genotype selected markers. A linear mixed-effect model was used to access association between these single nucleotide polymorphism (SNP) markers and growth-related traits.Results: A total of 30 polymorphisms were investigated in THRSP. Of these, nine SNPs for loci were selected to perform association analyses. Significant associations were detected between g.-49G>T SNP with body weight at 20 weeks of age (BW20), g.451T>C SNP with growth at 10 to 12 weeks of age (GR10-12), and g.1432A>C SNP with growth at 14 to 16 weeks trait (GR14-16) and body weight at 18 weeks of age (BW18). Moreover, diplotype of the THRSP gene significantly affected body weight at 12 weeks of age (BW12) and GR10-12 traits. Diplotype of ht1/ht2 was favorable for BW12 and GR10-12 traits.Conclusion: These results suggest that THRSP can be regarded as a candidate gene for growth traits in KNC.

Objective: This study was conducted to estimate genetic parameters for milk yield traits using daily milk yield records from parlour data generated in an intensively managed commercial dairy farm with Jersey and Jersey-Friesian cows in Sri Lanka.Methods: Genetic parameters were estimated for first and second lactation predicted and realized 305-day milk yield using univariate animal models. Genetic parameters were also estimated for total milk yield for each 30-day intervals of the first lactation using univariate animal models and for daily milk yield using random regression models fitting second-order Legendre polynomials and assuming heterogeneous residual variances. Breeding values for predicted 305-day milk yield were estimated using an animal model.Results: For the first lactation, the heritability of predicted 305-day milk yield in Jersey cows (0.08±0.03) was higher than that of Jersey-Friesian cows (0.02±0.01). The second lactation heritability estimates were similar to that of first lactation. The repeatability of the daily milk records was 0.28±0.01 and the heritability ranged from 0.002±0.05 to 0.19±0.02 depending on day of milk. Pearson product-moment correlations between the bull estimated breeding values (EBVs) in Australia and bull EBVs in Sri Lanka for 305-day milk yield were 0.39 in Jersey cows and –0.35 in Jersey-Friesian cows.Conclusion: The heritabilities estimated for milk yield in Jersey and Jersey-Friesian cows in Sri Lanka were low, and were associated with low additive genetic variances for the traits. Sire differences in Australia were not expressed in the tropical low-country of Sri Lanka. Therefore, genetic progress achieved by importing genetic material from Australia can be expected to be slow. This emphasizes the need for a within-country evaluation of bulls to produce locally adapted dairy cows.

Objective: This study was conducted to reveal potential metabolic differences of dairy cows fed corn stover (CS) and rice straw (RS) instead of alfalfa hay (AH) as main forage source.Methods: Thirty multiparous mid-late lactation Holstein dairy cows were selected and randomly assigned to three diets, AH, CS, or RS (n = 10). After 13 weeks of the feeding trial, coccygeal arterial and superficial epigastric venous plasma samples were collected before morning feeding for gas chromatography time-of-flight/mass spectrometry analyses.Results: In the artery, 8 and 13 metabolites were detected as differential metabolites between AH and CS, and between AH and RS, respectively. The relative abundance of phenylpropanoate (log2fold change [FC]) = 1.30, 1.09), panthenol (log2FC = 2.36, 2.20), threitol (log2FC = 1.00, 1.07), and 3,7,12-trihydroxycoprostane (log2FC = 0.79, 0.78) were greater in both CS and RS than in AH, and tyrosine (log2FC = –0.32), phenylalanine (log2FC = –0.30), and pyruvic acid (log2FC = –0.30) were lower in RS than in AH. In the vein, 1 and 7 metabolites were detected as differential metabolites between AH and CS, and between AH and RS, respectively. By comparing AH and RS, we found that metabolic pathways of phenylalanine, tyrosine, and tryptophan biosynthesis and phenylalanine metabolism were enriched by integrative artery and vein analysis. Furthermore, AH and RS, arterial phenylpropanoate and 4-hydroxyproline were positively, and phenylalanine was negatively correlated with milk urea nitrogen. Finally, in AH and CS, arterial panthenol was negatively correlated with feed efficiency.Conclusion: Arterial metabolic profiles changed more than those in the veins from animals on three forage diets, differing in amino acids. We found that phenylalanine, tyrosine, and tryptophan biosynthesis and phenylalanine metabolism were restricted when cows were fed low-quality cereal straw diets.

Objective: This study was conducted to evaluate the effects of inclusion level and amino acid (AA) supplementation on energy values of soybean oil (SO) as determined by difference method or regression method when fed to growing pigs.Methods: Thirty-six barrows (initial body weight: 28.0±1.3 kg) were randomly assigned to one of 6 dietary treatments, which included 2 control diets formulated using a basal diet with or without AA supplementation, and 4 experimental diets with 5% or 10% SO addition in the 2 control diets, respectively. All pigs were individually housed in metabolism crates for 19 d, and during the last 5 d, total urine and feces production were collected. The nutrient digestibility in diets and the digestible energy (DE) and metabolizable energy (ME) values of SO were determined using the difference method and the regression method, respectively.Results: Our results showed that there were no interaction effects (p>0.05) between AA supplementation and SO inclusion levels on energy values of SO and dietary nutrient digestibility. Objective: The rapid and reliable detection of the African swine fever virus (ASFV) plays an important role in emergency control and preventive measures of ASF. Some methods have been recommended by FAO/OIE to detect ASFV in clinical samples, including realtime polymerase chain reaction (PCR). However, mismatches in primer and probe binding regions may cause a false-negative result. Here, a slight modification in probe sequence has been conducted to improve the qualification of real-time PCR based on World Organization for Animal Health (OIE) protocol for accurate detection of ASFV in field samples in Vietnam.Methods: Seven positive confirmed samples (four samples have no mismatch, and three samples contained one mutation in probe binding sites) were used to establish novel real-time PCR with slightly modified probe (Y = C or T) in comparison with original probe recommended by OIE.Results: Both real-time PCRs using the OIE-recommended probe and novel modified probe can detect ASFV in clinical samples without mismatch in probe binding site. A high correlation of cycle quantification (Cq) values was observed in which Cq values obtained from both probes arranged from 22 to 25, suggesting that modified probe sequence does not impede the qualification of real-time PCR to detect ASFV in clinical samples. However, the samples with one mutation in probe binding sites were ASFV negative with OIE recommended probe but positive with our modified probe (Cq value ranked between 33.12-35.78).Conclusion: We demonstrated for the first time that a mismatch in probe binding regions caused a false negative result by OIE recommended real-time PCR, and a slightly modified probe is required to enhance the sensitivity and obtain an ASF accurate diagnosis in field samples in Vietnam.

Objective: The aim of this study is to characterize the impact of additional electrical stimulation (AES) and various pre-rigor holding temperatures (for 3 h) on the ageing-potential of hot boned bovine M. longissimus lumborum (LL).Methods: Paired LL loins from 12 bulls were hot-boned within 40 min of slaughter, immediate AES applied and subjected to various holding temperatures (5°C, 15°C, 25°C, and 35°C) for 3 h.Results: AES did not accelerate the rate of rigor attainment, but the 3 h pre-rigor holding temperature did. Shear force values decreased as the pre-rigor holding temperatures increased. AES and holding for 3 h (at 25°C) resulted in higher water-holding capacity.Conclusion: Data confirmed that AES did not influence the various meat quality parameters in the present study, but pre-rigor holding temperature (25°C) alone or in combination with AES resulted in superior meat quality.

Objective: This study was designed to determine how feeding diets differing in crude protein (CP) and undegraded intake protein (UIP) levels affected productivity, blood metabolites, carcass characteristics, and the production economics of Hanwoo steers.Methods: Thirty-six Hanwoo steers (age = 8.2±0.5 mo; body weight = 254±16.1 kg) were assigned at random to one of three treatments (4 steers/pen; 3 pens/treatment): i) a low-CP diet (LP; control) containing 12.1% CP with 35.1% UIP, 12.0% CP with 36.8% UIP, and 12.9% CP with 48.8% UIP, in the growing, fattening, and finishing periods, respectively; ii) a high-CP, low-UIP diet (HPLU) containing 15.0% CP with 33.7% UIP, 14.0% CP with 35.7% UIP, and 13.1% CP with 46.7% UIP, respectively; and iii) a high-CP, high-UIP diet (HPHU) containing 15.0% CP with 45.8% UIP, 14.0% CP with 44.6% UIP, and 13.0% CP with 51.1% UIP, respectively.Results: The treatments did not affect feed intake and growth performance, except for average daily gain during the fattening period that tended to be the lowest (p = 0.08) in the HPLUfed steers. The feed CP conversion ratio over the entire feeding period was higher with high-CP diets. The treatments did not affect most blood metabolites; however, blood cholesterol and low-density lipoprotein concentrations during the fattening and finishing periods were the lowest in steers fed a HPLU diet. The treatments had negligible effects on cold carcass weight, yield traits including longissimus muscle area, backfat thickness, yield index, and yield grade, plus quality traits including meat color, fat color, texture, and maturity. However, marbling score and frequency of carcass quality grade 1++ were greater in HPHU-fed steers.Conclusion: Feeding diets with higher CP and UIP levels did not affect growth performance but tended to improve the carcass quality of Hanwoo steers, resulting in greater economic return.

Objective: This study was conducted to investigate the roles of LIM kinases (LIMK1 and LIMK2) during porcine early embryo development. We checked the mRNA expression patterns and localization of LIMK1/2 to evaluate their characterization. We further explored the function of LIMK1/2 in developmental competence and their relationship between actin assembly and cell junction integrity, specifically during the first cleavage and compaction.Methods: Pig ovaries were transferred from a local slaughterhouse within 1 h and cumulus oocyte complexes (COCs) were collected. COCs were matured in in vitro maturation medium in a CO2 incubator. Metaphase II oocytes were activated using an Electro Cell Manipulator 2001 and microinjected to insert LIMK1/2 dsRNA into the cytoplasm. To confirm the roles of LIMK1/2 during compaction and subsequent blastocyst formation, we employed a LIMK inhibitor (LIMKi3).Results: LIMK1/2 was localized in cytoplasm in embryos and co-localized with actin in cell-to-cell boundaries after the morula stage. LIMK1/2 knockdown using LIMK1/2 dsRNA significantly decreased the cleavage rate, compared to the control group. Protein levels of E-cadherin and β-catenin, present in adherens junctions, were reduced at the cell-to-cell boundaries in the LIMK1/2 knockdown embryos. Embryos treated with LIMKi3 at the morula stage failed to undergo compaction and could not develop into blastocysts. Actin intensity at the cortical region was considerably reduced in LIMKi3-treated embryos. LIMKi3-induced decrease in cortical actin levels was attributed to the disruption of adherens junction and tight junction assembly. Phosphorylation of cofilin was also reduced in LIMKi3-treated embryos.Conclusion: The above results suggest that LIMK1/2 is crucial for cleavage and compaction through regulation of actin organization and cell junction assembly.

Background Autonomous underwater vehicles (AUVs) and animal telemetry have become important tools for understanding the relationships between aquatic organisms and their environment, but more information is needed to guide the development and use of AUVs as effective animal tracking platforms. A forward-facing acoustic telemetry receiver (VR2Tx 69 kHz; VEMCO, Bedford, Nova Scotia) attached to a novel AUV (gliding robotic fish) was tested in a freshwater lake to (1) compare its detection efficiency (i.e., the probability of detecting an acoustic signal emitted by a tag) of acoustic tags (VEMCO model V8-4H 69 kHz) to stationary receivers and (2) determine if detection efficiency was related to distance between tag and receiver, direction of movement (toward or away from transmitter), depth, or pitch. Results Detection efficiency for mobile (robot-mounted) and stationary receivers were similar at ranges less than 300 m, on average across all tests, but detection efficiency for the mobile receiver decreased faster than for stationary receivers at distances greater than 300 m. Detection efficiency was higher when the robot was moving toward the transmitter than when moving away from the transmitter. Detection efficiency decreased with depth (surface to 4 m) when the robot was moving away from the transmitter, but depth had no significant effect on detection efficiency when the robot was moving toward the transmitter. Detection efficiency was higher when the robot was descending (pitched downward) than ascending (pitched upward) when moving toward the transmitter, but pitch had no significant effect when moving away from the transmitter. Conclusion Results suggested that much of the observed variation in detection efficiency is related to shielding of the acoustic signal by the robot body depending on the positions and orientation of the hydrophone relative to the transmitter. Results are expected to inform hardware, software, and operational changes to gliding robotic fish that will improve detection efficiency. Regardless, data on the size and shape of detection efficiency curves for gliding robotic fish will be useful for planning future missions and should be relevant to other AUVs for telemetry. With refinements, gliding robotic fish could be a useful platform for active tracking of acoustic tags in certain environments.

Objective: The objective of this study was to characterize the number of loci affecting growth traits and the distribution of single nucleotide polymorphism (SNP) effects on growth traits, and to understand the genetic architecture for growth traits in Hanwoo (Korean cattle) using genome-wide association study (GWAS), genomic partitioning, and hierarchical Bayesian mixture models.Methods: GWAS: A single-marker regression-based mixed model was used to test the association between SNPs and causal variants. A genotype relationship matrix was fitted as a random effect in this linear mixed model to correct the genetic structure of a sire family. Genomic restricted maximum likelihood and BayesR: A priori information included setting the fixed additive genetic variance to a pre-specified value; the first mixture component was set to zero, the second to 0.0001×σ_g^2, the third 0.001 × σ_g^2, d the fourth to 0.01 × σ_g^2. BayesR fixed a priori information was not more than 1% of the genetic variance for each of the SNPs affecting the mixed distribution.Results: The GWAS revealed common genomic regions of 2 Mb on bovine chromosome 14 (BTA14) and 3 had a moderate effect that may contain causal variants for body weight at 6, 12, 18, and 24 months. This genomic region explained approximately 10% of the variance against total additive genetic variance and body weight heritability at 12, 18, and 24 months. BayesR identified the exact genomic region containing causal SNPs on BTA14, 3, and 22. However, the genetic variance explained by each chromosome or SNP was estimated to be very small compared to the total additive genetic variance. Causal SNPs for growth trait on BTA14 explained only 0.04% to 0.5% of the genetic varianceConclusion: Segregating mutations have a moderate effect on BTA14, 3, and 19; many other loci with small effects on growth traits at different ages were also identified.

Objective: This study aimed to determine the effects of stress during slaughter of beef cattle on physiological parameters, carcass, and meat quality at a Federal Inspection Type slaughterhouse located in the southeast of Mexico.Methods: A total of 448 carcasses of male Zebu×European steers with an average age of 36 months were included. Carcass assessment of presence of bruises and bruise characteristics was carried out on each half-carcass. Blood variable indicators of stress (packed cell volume, neutrophil to lymphocyte ratio, glucose, cortisol concentration) and meat quality parameters (pH, color, shear force, drip loss) were evaluated.Results: Of the 448 carcasses evaluated, 81% of the carcasses showed at least one bruise; one bruise was detected in 36.6% and two bruises in 27.0% of animals. Of the 775 bruises found, 69.2% of the bruises were grade 1 in region 3. Of the 448 carcasses studied, 69.6% showed hyperglycemia (6.91 mmol/L); 44.3% and 22.7% showed high (74.7 ng/mL) and extremely high (108.8 ng/mL) cortisol levels, respectively, indicative of inadequate handling of animals during preslaughter and slaughter. Of the carcasses evaluated, 90.4% had a pH ≥5.8 with an average of pH 6.3. In both pH groups, meat samples showed L* values >37.0 (81.6%) and a shear force >54.3 N; meat pH≥5.8 group showed a drip loss of 2.5%. These findings were indicative of dark, firm, and dry (DFD) meat. According to principal component analysis, grades 1 and 2 bruises in region 3 and grade 1 bruises in region 5 were highly associated with cortisol, drip loss, and color parameters b* and h* and were negatively associated with L*, a*, and C*.Conclusion: The bruises probably caused by stress-inducing situations triggered DFD meat. Appropriate changes in handling routines in operating conditions should be made to minimize stress to animals during the slaughter process to improve animal welfare and meat quality.

Objective: The objective of this study was to evaluate the effects of lysophospholipids (LPL) supplementation on rumen fermentation, degradability, and microbial diversity in forage with high oil diet in an in vitro system.Methods: Four experimental treatments were used: i) annual ryegrass (CON), ii) 93% annual ryegrass +7% corn oil on a dry matter (DM) basis (OiL), iii) OiL with a low level (0.08% of dietary DM) of LPL (LLPL), and iv) OiL with a high level (0.16% of dietary DM) of LPL (HLPL). An in vitro fermentation experiment was performed using strained rumen fluid for 48 h incubations. In vitro DM degradability (IVDMD), in vitro neutral detergent fiber degradability, pH, ammonia nitrogen (NH3-N), volatile fatty acid (VFA), and microbial diversity were estimated.Results: There was no significant change in IVDMD, pH, NH3-N, and total VFA production among treatments. Objective: This study was to evaluate the effect of husbandry systems and strains on cecum microbial diversity of Jingyang chickens under the same dietary conditions.Methods: A total of 320 laying hens (body weight, 1.70±0.15 kg; 47 weeks old) were randomly allocated to one of the four treatments: i) Silver-feathered hens in enrichment cages (SEC) with an individual cage (70×60×75 cm), ii) Silver-feathered hens in free range (SFR) with the stocking density of 1.5 chickens per ten square meters, iii) Gold-feathered hens in enrichment cages (GEC), iv) Gold-feathered hens in free range (GFR). The experiment lasted 8 weeks and the cecum fecal samples were collected for 16S rDNA high throughput sequencing at the end of experiment.Results: i) The core microbiota was composed of Bacteroidetes (49% to 60%), Firmicutes (21% to 32%) and Proteobacteria (2% to 4%) at the phylum level. ii) The core bacteria were Bacteroides (26% to 31%), Rikenellaceae (9% to 16%), Parabacteroides (2% to 5%) and Lachnoclostridium (2% to 6%) at the genus level. iii) The indexes of operational taxonomic unit, Shannon, Simpson and observed species were all higher in SFR group than in SEC group while in GEC group than in GFR group, with SFR group showing the greatest diversity of cecum microorganisms among the four groups. iv) The clustering result was consistent with the strain classification, with a similar composition of cecum bacteria in the two strains of laying hens.Conclusion: The core microbiota were not altered by husbandry systems or strains. The free-range system increased the diversity of cecal microbes only for silver feathered hens. However, the cecum microbial composition was similar in two strain treatments under the same dietary conditions.

Objective The study sought to estimate the relationship between body surface temperature (BST) and temperature humidity index (THI) and to present the validity of THI as a heat stress index in the field. Methods Eight Hanwoo heifers (20 to 32 month) were examined in a field trial, with a space allowance of 10 m2 per head. The BST was measured using an infrared thermographic camera. The BST of five body regions (eyes, hindquarters, nose, part of horns, and ears), ambient temperature (AT), and relative humidity (RH) were measured 7 times daily (07, 09, 11, 13, 15, 17, and 19 h) during each season with three replicates. Results The THI ranged 34.0 to 56.9 during spring (AT, −1.0°C to 13.4°C), 75.1 to 84.7 during summer (AT, 24.9°C to 33.6°C), 55.8 to 70.9 during autumn (AT, 13.0°C to 26.0°C) and 17.5 to 39.2 during winter (AT, −10.4°C to 1.0°C). In the regression analysis, the coefficient of determination (R2) between THI and BST was 0.88, 0.72, 0.83, 0.86, and 0.85 for the eyes, hindquarters, nose, part of horn, and ears area, respectively. This indicates that BST has a strong correlation with AT and RH. Expression equations were estimated as Y (THI) = 31.54+0.1085X (BST of eyes) and Y (THI) = 30.48+0.1147X (BST of hindquarters) by simple linear regression analysis in this experiment. Conclusion Consequently, the upper bound for heat stress estimation can be specified ranging from THI of 65 (eyes) to 70 (hindquarters). From this we can expect a precise feeding system for Korean native cattle in the field.

Background Advances in biologging technology allow researchers access to previously unobservable behavioral states and movement patterns of marine animals. To relate behaviors with environmental variables, features must be evaluated at scales relevant to the animal or behavior. Remotely sensed environmental data, collected via satellites, often suffers from the effects of cloud cover and lacks the spatial or temporal resolution to adequately link with individual animal behaviors or behavioral bouts. This study establishes a new method for remotely and continuously quantifying surface ice concentration (SIC) at a scale relevant to individual whales using on-animal tag video data. Results Motion-sensing and video-recording suction cup tags were deployed on 7 Antarctic minke whales (Balaenoptera bonaerensis) around the Antarctic Peninsula in February and March of 2018. To compare the scale of camera-tag observations with satellite imagery, the area of view was simulated using camera-tag parameters. For expected conditions, we found the visible area maximum to be ~ 100m2 which indicates that observations occur at an equivalent or finer scale than a single pixel of high-resolution visible spectrum satellite imagery. SIC was classified into one of six bins (0%, 1–20%, 21–40%, 41–60%, 61–80%, 81–100%) by two independent observers for the initial and final surfacing between dives. In the event of a disagreement, a third independent observer was introduced, and the median of the three observer’s values was used. Initial results (n = 6) show that Antarctic minke whales in the coastal bays of the Antarctic Peninsula spend 52% of their time in open water, and only 15% of their time in water with SIC greater than 20%. Over time, we find significant variation in observed SIC, indicating that Antarctic minke occupy an extremely dynamic environment. Sentinel-2 satellite-based approaches of sea ice assessment were not possible because of persistent cloud cover during the study period. Conclusion Tag-video offers a means to evaluate ice concentration at spatial and temporal scales relevant to the individual. Combined with information on underwater behavior, our ability to quantify SIC continuously at the scale of the animal will improve upon current remote sensing methods to understand the link between animal behavior and these dynamic environmental variables.

Objective: This study aimed to elucidate the effect of miR-140 on the proliferation of porcine fetal fibroblasts (PFFs) and identify the target genes of miR-140 in PFFs.Methods: In this study, bioinformatics software was used to predict and verify target genes of miR-140. Quantitative polymerase chain reaction and western blot were used to detect the relationship between miR-140 and its target genes in PFFs. Dual luciferase reporter gene assays were performed to assess the interactions among miR-140, type 1 insulinlike growth factor receptor (IGF1R), and SRY-box 4 (SOX4). The effect of miR-140 on the proliferation of PFFs was measured by CCK-8 when PFFs were transfected with a miR-140 mimic or inhibitor. The transcription factor SOX4 binding to promoter of IGF1R was detected by chromatin immunoprecipitation assay (ChIP).Results: miR-140 directly targeted IGF1R and inhibited proliferation of PFFs. Meanwhile, miR-140 targeted transcription factor SOX4 that binds to promoter of porcine IGF1R to indirectly inhibit the expression of IGF1R. In addition, miR-140 inhibitor promoted PFFs proliferation, which is abrogated by SOX4 or IGF1R knockdown.Conclusion: miR-140 inhibited PFFs proliferation by directly targeting IGF1R and indirectly inhibiting IGF1R expression via SOX4, which play an important role in the development of porcine fetal.

Objective: The aim of study was to investigate the effects of season and single layer centrifugation (SLC) before cryopreservation on post-thaw bull sperm quality in Thailand.Methods: Semen was collected from 6 bulls (Bos indicus) in summer, rainy season and winter 2014 through 2016. Semen characteristics, sperm morphology, sperm kinematics, viability, chromatin structure and mitochondrial membrane were evaluated. Meteorological data were available from the local meteorological station;Results: Season had an effect on semen characteristics in the raw ejaculate, with higher proportions of normal spermatozoa and lower abnormalities in winter than in the other two seasons. Sperm kinematics, viability, DNA fragmentation index, and mitochondrial membrane potential were not different between seasons. Sperm samples selected by SLC had greater normal morphology and a lower proportion with bent tails than controls and higher values of progressive motility (PRO), beat cross frequency, linearity, straightness, wobble (WOB), and lower values of slow motility, velocity average path (VAP), velocity curved line, and amplitude of lateral head displacement than controls. In addition, SLCselection had a favorable effect on PRO, VAP, and WOB that differed among seasons.Conclusion: Our results suggested that these bulls were well adapted to their location, with season having an effect on sperm morphology. Moreover, SLC could be used prior to cryopreservation, regardless of season, to enhance normal morphology and kinematics of bull sperm samples without adversely affecting other parameters of sperm quality. However, there was considerable variation among bulls in DNA fragmentation index, mitochondrial membrane potential and sperm viability. In addition, SLC had a positive effect on sperm morphology and sperm kinematics, which could be expected to influence fertility.

Objective: The objective of this study was to estimate the genetic parameters and trends for milk, fat, and protein yields in the first three lactations of Thai dairy cattle using a 3-trait,- 3-lactation random regression test-day model.Methods: Data included 168,996, 63,388, and 27,145 test-day records from the first, second, and third lactations, respectively. Records were from 19,068 cows calving from 1993 to 2013 in 124 herds. (Co) variance components were estimated by Bayesian methods. Gibbs sampling was used to obtain posterior distributions. The model included herd-year-month of testing, breed group-season of calving-month in tested milk group, linear and quadratic age at calving as fixed effects, and random regression coefficients for additive genetic and permanent environmental effects, which were defined as modified constant, linear, quadratic, cubic and quartic Legendre coefficients.Results: Average daily heritabilities ranged from 0.36 to 0.48 for milk, 0.33 to 0.44 for fat and 0.37 to 0.48 for protein yields; they were higher in the third lactation for all traits. Heritabilities of test-day milk and protein yields for selected days in milk were higher in the middle than at the beginning or end of lactation, whereas those for test-day fat yields were high at the beginning and end of lactation. Genetics correlations (305-d yield) among production yields within lactations (0.44 to 0.69) were higher than those across lactations (0.36 to 0.68). The largest genetic correlation was observed between the first and second lactation. The genetic trends of 305-d milk, fat and protein yields were 230 to 250, 25 to 29, and 30 to 35 kg per year, respectively.Conclusion: A random regression model seems to be a flexible and reliable procedure for the genetic evaluation of production yields. It can be used to perform breeding value estimation for national genetic evaluation in the Thai dairy cattle population.

Objective: This study was carried out to investigate physicochemical properties, and antioxidant and antimicrobial activities of low-fat sausages (LFSs) covered with sodium alginate (SA) film alone and with powder film (TSA-film) formed by cross-linking cherry tomato powder (CTP) and SA with calcium chloride (CaCl2).Methods: Sausages covered with the biodegradable film were assessed based on the measurement of pH, color (L*, a*, b*), proximate analysis, expressive moisture (EM), texture profile analysis, total plate counts (TPC), violet red bile, and 2-Thiobarbituric acid reactive substances (TBARS) during storage under refrigeration. LFSs wrapped with TSA-film were compared with those wrapped with SA-film and without film (control) during storage at 10°C for 35 days.Results: The LFSs covered with the mixed film had lower pH, lightness (L*), EM%, TBARS, and TPC, but lower yellowness (b*) and hardness values than those wrapped with TSA-film alone.Conclusion: Lipid oxidation and microbial growth was retarded in sausages covered with biodegradable films, especially multiple films as compared to single film, thereby resulting in extended shelf-life of the LFSs.
